Liquid water has been known to occur beneath the Antarctic ice sheet for more than 40 years (1), but only recently have these subglacial aqueous environments been recognized as microbial ecosystems that may influence biogeochemical transformations on a global scale (2-4). Here we present the first geomicrobiological description of water and surficial sediments obtained from direct sampling of a subglacial Antarctic lake. Subglacial Lake Whillans (SLW) lies beneath approximately 800 m of ice on the lower portion of the Whillans Ice Stream (WIS) in West Antarctica and is part of an extensive and evolving subglacial drainage network (5). The water column of SLW contained metabolically active microorganisms and was derived primarily from glacial ice melt with solute sources from lithogenic weathering and a minor seawater component. Heterotrophic and autotrophic production data together with small subunit ribosomal RNA gene sequencing and biogeochemical data indicate that SLW is a chemosynthetically driven ecosystem inhabited by a diverse assemblage of bacteria and archaea. Our results confirm that aquatic environments beneath the Antarctic ice sheet support viable microbial ecosystems, corroborating previous reports suggesting that they contain globally relevant pools of carbon and microbes (2,4) that can mobilize elements from the lithosphere (6) and influence Southern Ocean geochemical and biological systems (7)., Almost 400 subglacial lakes have been identified beneath the Antarctic ice sheet (8). Snow algae blooms are common occurrences in alpine systems and contribute to increasing snow and glacial ice melt rates. Despite the cosmopolitan distribution of snow algae, little is known about the role their life cycle plays in community composition and activity in snowpack. Mount Baker, in Washington, USA, is a rapidly changing alpine ecosystem. Here we present estimates of cellular biomass, microbial community composition, and primary productivity measurements associated with surficial snow algae blooms and a vertical profile within a snow pit to characterize the microbial ecology of this snowpack system. Chloromonas was the most abundant algae detected in our samples. Sphingobacteriaceae were the most dominant bacteria in samples where fungi were the most abundant eukaryote, whereas Chitinophagaceae dominated when Chlorophyceae were the most abundant eukaryote. In a snow pit vertical profile, we observed more algae in the green-pigmented, flagellated life cycle stage at depth (~30 cm below the surface) than at the surface where the astaxanthin-rich, aplanospore life cycle stage dominated. Higher resolution analyses revealed that Chloromonas-associated photosynthesis transcripts were more abundant at depth than at the surface of the snowpack. These results suggest that there is a photosynthetic niche within alpine snowpack that has unknown effects on the carbon budget and provides potentially undetected primary productivity resulting in a "cryptic" photosynthetic system. Bacterial diversity in Blood Falls, the surface manifestation of brine released from below Taylor Glacier, Antarctica was studied. The results suggested that the brine below the Taylor Glacier hosts a viable ecosystem with microorganisms capable of growth, surrounded by chemical energy present in reduced iron and sulfur compounds.

Evidence gathered from spacecraft orbiting Mars has shown that water ice exists at both poles and may form a large subsurface reservoir at lower latitudes. The recent exploration of the martian surface by unmanned landers and surface rovers, and the planned missions to eventually return samples to Earth have raised concerns regarding both forward and back contamination. Methods to search for life in these icy environments and adequate protocols to prevent contamination can be tested with earthly analogues. Studies of ice cores on Earth have established past climate changes and geological events, both globally and regionally, but only recently have these results been correlated with the biological materials (i.e., plant fragments, seeds, pollen grains, fungal spores, and microorganisms) that are entrapped and preserved within the ice. The inclusion of biology into ice coring research brings with it a whole new approach towards decontamination. Our investigations on ice from the Vostok core (Antarctica) have shown that the outer portion of the cores have up to 3 and 2 orders of magnitude higher bacterial density and dissolved organic carbon (DOC) than the inner portion of the cores, respectively, as a result of drilling and handling. The extreme gradients that exist between the outer and inner portion of these samples make contamination a very relevant aspect of geomicrobiological investigations with ice cores, particularly when the actual numbers of ambient bacterial cells are low. To address this issue and the inherent concern it raises for the integrity of future investigations with ice core materials from terrestrial and extraterrestrial environments, we employed a procedure to monitor the decontamination process in which ice core surfaces are painted with a solution containing a tracer microorganism, plasmid DNA, and fluorescent dye before sampling. Using this approach, a simple and direct method is proposed to verify the authenticity of geomicrobiological results obtained from ice core materials. Our protocol has important implications for the design of life detection experiments on Mars and the decontamination of samples that will eventually he returned to Earth. With the increase in intensive livestock operations between 1985 and 1995, concerns for water quality also increased. Thus, the amounts of phosphorus and nitrogen excreted by hogs, turkeys, chickens, and cattle were studied. Most of these nutrients were imported from outside the region. So, this data was also incorporated into the study. Locally grown feed crops were also tracked. Already several accidental releases from holding ponds have produced algae blooms. Yet, the data shows that current waste management practices has prevented most of the nutrient from entering the rivers. The study did not examine contamination of ground water by seepage from farms or volatilization of ammonia.

Geochemical characterisation of hypersaline waters is difficult as high concentrations of salts hinder the analysis of constituents at low concentrations, such as trace metals, and the collection of samples for trace metal analysis in natural waters can be easily contaminated. This is particularly the case if samples are collected by non-conventional techniques such as those required for aquatic subglacial environments. In this paper we present the first analysis of a subglacial brine from Taylor Valley, (~ 78°S), Antarctica for the trace metals: Ba, Co, Mo, Rb, Sr, V, and U. Samples were collected englacially using an electrothermal melting probe called the IceMole. This probe uses differential heating of a copper head as well as the probe's sidewalls and an ice screw at the melting head to move through glacier ice. Detailed blanks, meltwater, and subglacial brine samples were collected to evaluate the impact of the IceMole and the borehole pump, the melting and collection process, filtration, and storage on the geochemistry of the samples collected by this device. Comparisons between melt water profiles through the glacier ice and blank analysis, with published studies on ice geochemistry, suggest the potential for minor contributions of some species Rb, As, Co, Mn, Ni, NH4+, and NO2−+NO3− from the IceMole. The ability to conduct detailed chemical analyses of subglacial fluids collected with melting probes is critical for the future exploration of the hundreds of deep subglacial lakes in Antarctica.
