Summary A library of well-characterized human induced pluripotent stem cell (hiPSC) lines from clinically healthy human subjects could serve as a useful resource of normal controls for in vitro human development, disease modeling, genotype-phenotype association studies, and drug response evaluation. We report generation and extensive characterization of a gender-balanced, racially/ethnically diverse library of hiPSC lines from 40 clinically healthy human individuals who range in age from 22 to 61 years. The hiPSCs match the karyotype and short tandem repeat identities of their parental fibroblasts, and have a transcription profile characteristic of pluripotent stem cells. We provide whole-genome sequencing data for one hiPSC clone from each individual, genomic ancestry determination, and analysis of mendelian disease genes and risks. We document similar transcriptomic profiles, single-cell RNA-sequencing-derived cell clusters, and physiology of cardiomyocytes differentiated from multiple independent hiPSC lines. The article describes an innovative shaping procedure for the Gregorian ring-focus reflector antenna with two swappable feed units (horn and subreflector) and a common reflector antenna for the flyaway terminal for communication at the C and Ku bands. Over such a multiband, the main challenge is to achieve a compact antenna structure that meets stringent radio-frequency (RF) specifications. The optimization is carried out to achieve enhanced peak gain and reduction of the sidelobe level through rotationally symmetric shaping of the main and subreflectors. Circular symmetric shaping provides ease in manufacturing of the subreflector and identical segments of the main reflector for portability. The measured results of a prototype model of the shaped ringfocus antenna with swappable feed units support the parallelshaping technique.

Application specific, lightweight yet high strength materials always remain in large demand to the aerospace engineers. Poly-ether-ether-ketone (PEEK) is a unique polymer which has low density, high glass transition temperature, and stability at an ultra-high vacuum condition. In addition, high compliance of PEEK and ease of vacuum forming facilitate the development of complex geometric shapes. In this research, we have investigated the suitability of PEEK material for parabolic antenna structure under space environment conditions. We have carried out rigorous space qualification tests on various categories of PEEK material. With the promising experimental results, we have developed a 450 mm diameter prime focal C-band Antenna. The parabolic antenna is made up of PEEK material, and it is supported on the curved ribs made up of PEEK CA30 material. The developed antenna setup is further installed for far-field radiation testing in a Compact Antenna Test Facility (CATF) in the anechoic chamber. Simultaneously, RF simulations are carried out using CST software. The simulated and measured far-field radiation patterns show close agreements. Thereafter, thermal analysis of the developed antenna and the effect of thermal expansion on radiation far-field are discussed. The developed antenna is very promising to be used in the future space missions.

ObjectiveIBD therapies and treatments are evolving to deeper levels of remission. Molecular measures of disease may augment current endpoints including the potential for less invasive assessments.DesignTranscriptome analysis on 712 endoscopically defined inflamed (Inf) and 1778 non-inflamed (Non-Inf) intestinal biopsies (n=498 Crohn’s disease, n=421 UC and 243 controls) in the Mount Sinai Crohn’s and Colitis Registry were used to identify genes differentially expressed between Inf and Non-Inf biopsies and to generate a molecular inflammation score (bMIS) via gene set variance analysis. A circulating MIS (cirMIS) score, reflecting intestinal molecular inflammation, was generated using blood transcriptome data. bMIS/cirMIS was validated as indicators of intestinal inflammation in four independent IBD cohorts.ResultsbMIS/cirMIS was strongly associated with clinical, endoscopic and histological disease activity indices. Patients with the same histologic score of inflammation had variable bMIS scores, indicating that bMIS describes a deeper range of inflammation. In available clinical trial data sets, both scores were responsive to IBD treatment. Despite similar baseline endoscopic and histologic activity, UC patients with lower baseline bMIS levels were more likely treatment responders compared with those with higher levels. Finally, among patients with UC in endoscopic and histologic remission, those with lower bMIS levels were less likely to have a disease flare over time.ConclusionTranscriptionally based scores provide an alternative objective and deeper quantification of intestinal inflammation, which could augment current clinical assessments used for disease monitoring and have potential for predicting therapeutic response and patients at higher risk of disease flares.

We present design, fabrication, and characterization results on a novel multi-spectral imaging lens. Our lens design combines a catadioptric geometry with a reflective tunable cholesteric liquid crystal cell. The lens images in a narrow (~50 nm) spectral band and can be continuously voltage-tuned over a broad wavelength range (>;400 nm). The lens works with circularly polarized light. A cholesteric liquid crystal cell reflects light if the incident circular polarization has the correct handedness and the wavelength is closely matched with the liquid crystal pitch. Otherwise, the light transmits unaffected. Our compact optical design converts this reflective mechanism into a transmissive one for the purpose of imaging. In addition, we use corrective refractive lenses to optimize imaging performance over wide field of view. The traditional approach would be to use tunable birefringent filter (e.g. Solc and Lyot filter) in conjunction with a broadband imaging lens, with the birefringent filter itself composed of multiple liquid crystal cells and polarizers. Tunability is provided by adjusting optical retardation of nematic liquid crystal cells through application of appropriate voltages. Our design uses a single liquid crystal cell and a single polarizer and is inherently high in optical transmission, significantly less complex and thus potentially low cost. The applications include forensic imaging, multispectral aerial surveys, dermatology, medical microscopy etc.

Kinase inhibitors (KIs) represent an important class of anti-cancer drugs. Although cardiotoxicity is a serious adverse event associated with several KIs, the reasons remain poorly understood, and its prediction remains challenging. We obtain transcriptional profiles of human heart-derived primary cardiomyocyte like cell lines treated with a panel of 26 FDA-approved KIs and classify their effects on subcellular pathways and processes. Individual cardiotoxicity patient reports for these KIs, obtained from the FDA Adverse Event Reporting System, are used to compute relative risk scores. These are then combined with the cell line-derived transcriptomic datasets through elastic net regression analysis to identify a gene signature that can predict risk of cardiotoxicity. We also identify relationships between cardiotoxicity risk and structural/binding profiles of individual KIs. We conclude that acute transcriptomic changes in cell-based assays combined with drug substructures are predictive of KI-induced cardiotoxicity risk, and that they can be informative for future drug discovery., Cardiotoxic adverse events associated with kinase inhibitors are a growing concern in clinical oncology. Here the authors use cellular transcriptomic responses of human cardiomyocytes treated with protein kinase inhibitors and the associated drug structural signatures to determine an integrated predictive signature of cardiotoxicity.

This paper presents the analysis of the multifaceted reflectarray (RFA) antenna for spatial bandwidth improvement. Geometries of faceted RFA have been designed by segmenting the phase correcting surface in tilted 3-panel and 5-panel configurations. Phase distribution over RFA surface and directivity variation with frequency have been computed using Geometric Optics (GO) based technique and achieved 6.5% and 7.4% 1-dB gain bandwidth in case of 3-panel and 5-panel faceted RFA, respectively as compared to a flat RFA having 1.8% 1-dB gain bandwidth.

This paper presents the analysis of phased array antenna where elements are clustered using Pinwheel type aperiodic tiling pattern, to improve side lobe level (SLL). Parametric analysis has been carried out to select optimum subsection of Pinwheel pattern for clustering of array which is excited with required phased excitation to achieve 10° and 20° beam scan. Worst SLL in 3D radiation pattern of an array with Pinwheel clustering is improved by 3.8dB and 6.6dB for 10° and 20° beam scan respectively. Moreover, no. of TRMs has also been reduced by 68% while maintaining significantly low SLL.

The growing interest in higher capacity communications satellite necessitates the development of compact and lightweight other antennas (such as beacon horn antenna) at Earth-viewing deck due to crowded accommodation and weight limitation. In this article, we describe an innovative and compact high-gain composite horn antenna for space-borne beacon application. We have designed, manufactured and measured beacon horn antenna operating from 10.7 GHz to 12.5 GHz. The beacon horn antenna is devised by combining stepped section at input, axial corrugated in mid and finally radial corrugated sections. This engineered multiple sections provide extra flexibility of optimisation, leading to high edge of coverage (EoC) gain for Indian mainland, Lakshadweep Islands and Andaman & Nicobar Islands.
