Abstract

Accumulation of amyloid fibrils in the viscera and connective tissues causes systemic amyloidosis, which is responsible for about one in a thousand deaths in developed countries (1). Localized amyloid can also have serious consequences; for example, cerebral amyloid angiopathy is an important cause of haemorrhagic stroke. The clinical presentations of amyloidosis are extremely diverse and the diagnosis is rarely made before significant organ damage is present (1). There is therefore a major unmet need for therapy that safely promotes the clearance of established amyloid deposits. Over 20 different amyloid fibril proteins are responsible for different forms of clinically significant amyloidosis and treatments that substantially reduce the abundance of the respective amyloid fibril precursor proteins can arrest amyloid accumulation (1). Unfortunately, control of fibril-protein production is not possible in some forms of amyloidosis and in others it is often slow and hazardous (1). There is no therapy that directly targets amyloid deposits for enhanced clearance. However, all amyloid deposits contain the normal, non-fibrillar plasma glycoprotein, serum amyloid P component (SAP) (2,3). Here we show that administration of anti-human-SAP antibodies to mice with amyloid deposits containing human SAP triggers a potent, complement-dependent, macrophage-derived giant cell reaction that swiftly removes massive visceral amyloid deposits without adverse effects. Anti-SAP-antibody treatment is clinically feasible because circulating human SAP can be depleted in patients by the bis-D-proline compound CPHPC4, thereby enabling injected anti-SAP antibodies to reach residual SAP in the amyloid deposits. The unprecedented capacity of this novel combined therapy to eliminate amyloid deposits should be applicable to all forms of systemic and local amyloidosis., SAP is selectively concentrated in amyloid deposits by its avid binding to all amyloid fibril types (2,3). Includes bibliographical references., The purpose of this research is to examine the extent of instructional leadership of principals in high performing secondary schools in the metropolitan area of Cape Town, Western Cape. A mixed methods design of quantitative and qualitative research was undertaken. For the quantitative phase, the Principal Instructional Management Rating Scale (PIMRS) was administered to 5 principals and 136 teachers. One sample t-tests found statistically significant differences between the mean scores of principals and those of teachers. Data analysis of the PIMRS indicated that principals were most active in protecting instructional time, promoting professional development, providing incentives for learning and framing the school’s goals and least active in supervising and evaluating instruction, maintaining a high visibility and providing incentives for teachers. In the qualitative phase, interviews with the five principals yielded a result that underscores the value which principals place on professional accountability, trusting teachers to deliver the curriculum, building coherence, promoting professional development, giving professional autonomy and fostering relationships. The study found that the principal’s role is multi-faceted and complex and is neither limited to the instructional leadership behaviours of the PIMRS nor to the job description of the Personnel Administration Measures (PAM). Instructional leadership functions not measured by the PIMRS, such as the appointment of teachers, selection of pupils, engaging with stakeholder groups, establishing internal coherence and building trust by sharing instructional leadership practises with senior teachers were very important. School leaders internalize the expectations embedded in accountability systems and have woven these into an internal set of expectations and responsibilities that represent the school’s internal accountability systems. The thesis concludes with the view that both shared leadership and instructional leadership are important as they are indirectly related to pupil achievement.

28. A major stress-inducible Mr-42000 wall glycoprotein of French bean (Phaseolus vulgaris L.)

Abstract

A major wall protein of suspension-cultured cells of French bean has been isolated and characterised. It can be prepared from walls or the culture filtrate and in composition it is particularly rich in proline, valine and glutamic acid/glutamine and contains appreciable amounts of hydroxyproline. The N-terminus shows some glycosylation, while following chemical deglycosylation the first 38 residues were found to be identical to those of proline-rich proteins from soybean. However, the composition of the highly purified Mr-42000 bean protein differs considerably from the soybean proteins and must contain its own specific domains. An antibody was raised and used to demonstrate the inducibility of the Mr-42000 bean protein in response to elicitor action. The protein was found to be mainly localised in the intercellular spaces of the cortical cells of bean hypocotyls and at the wall-plasmalemma interface of xylem vessels, another potentially accessible compartment for pathogens. Following wounding, the protein was found to be generally distributed in the wall of epidermal and cortical cells of the hypocotyls. The Mr-42000 protein is cross reactive with antibodies raised to glycoproteins of the Rhizobium infection thread and the chitin-binding hydroxyproline-rich glycoprotein, potato lectin. These common epitopes together with the previously demonstrated chitin-binding properties of the bean protein indicate a role in host-microbial interactions. Furthermore, the Mr-42000 protein itself bound to the growing hyphal tips of the bean pathogen, Colletotrichum lindemuthianum.
