Assisted sexual coral propagation, resulting in greater genet diversity via genetic recombination, has been hypothesized to lead to more adaptable and, hence, resilient restored populations compared to more common clonal techniques. Coral restoration efforts have resulted in substantial populations of 'Assisted sexual Recruits' (i.e., juvenile corals derived from assisted sexual reproduction; AR) of multiple species outplanted to reefs or held in in situ nurseries across many locations in the Caribbean. These AR populations provided context to evaluate their relative resilience compared to co-occurring coral populations during the 2023 marine heat wave of unprecedented duration and intensity that affected the entire Caribbean. Populations of six species of AR, most ranging in age from 1–4 years, were surveyed across five regions during the mass bleaching season in 2023 (Aug-Dec), alongside co-occurring groups of corals to compare prevalence of bleaching and related mortality. Comparison groups included conspecific adult colonies as available, but also the extant co-occurring coral assemblages in which conspecifics were rare or lacking, as well as small, propagated coral fragments. Assisted sexual recruits had significantly lower prevalence of bleaching impacts (overall pooled ~ 10%) than conspecific coral populations typically comprised of larger colonies (~ 60–100% depending on species). In addition, small corals derived from fragmentation (rather than sexual propagation) in two regions showed bleaching susceptibility intermediate between AR and wild adults. Overall, AR exhibited high bleaching resistance under heat stress exposure up to and exceeding Degree Heating Weeks of 20°C-weeks. As coral reefs throughout the globe are subject to increasingly frequent and intense marine heatwaves, restoration activities that include sexual reproduction and seeding can make an important contribution to sustain coral populations. The effects of turbidity and sedimentation stress on early life stages of corals are poorly understood, particularly in Atlantic species. Dredging operations, beach nourishment, and other coastal construction activities can increase sedimentation and turbidity in nearby coral reef habitats and have the potential to negatively affect coral larval development and metamorphosis, reducing sexual reproduction success. In this study, we investigated the performance of larvae of the threatened Caribbean coral species Orbicella faveolata exposed to suspended sediments collected from a reef site in southeast Florida recently impacted by dredging (Port of Miami), and compared it to the performance of larvae exposed to sediments collected from the offshore, natal reef of the parent colonies. In a laboratory experiment, we tested whether low and high doses of each of these sediment types affected the survival, settlement, and respiration of coral larvae compared to a no-sediment control treatment. In addition, we analyzed the sediments used in the experiments with 16S rRNA gene amplicon sequencing to assess differences in the microbial communities present in the Port versus Reef sediments, and their potential impact on coral performance. Overall, only O. faveolata larvae exposed to the high-dose Port sediment treatment had significantly lower survival rates compared to the control treatment, suggesting an initial tolerance to elevated suspended sediments. However, significantly lower settlement rates were observed in both Port treatments (low- and high-dose) compared to the control treatment one week after exposure, suggesting strong latent effects. Sediments collected near the Port also contained different microbial communities than Reef sediments, and higher relative abundances of the bacteria Desulfobacterales, which has been associated with coral disease. We hypothesize that differences in microbial communities between the two sediments may be a contributing factor in explaining the observed differences in larval performance. Together, these results suggest that the settlement success and survival of O. faveolata larvae are more readily compromised by encountering port inlet sediments compared to reef sediments, with potentially important consequences for the recruitment success of this species in affected areas. Tetrabromopyrrole (TBP) is a readily biosynthesized marine proteobacterial compound that induces coral settlement (attachment and metamorphosis) at concentrations ranging from 50 - 250 ng ml-1 (0.13 - 0.65 μM). This suggests a great potential for the use of this compound as a settlement inducer for restoration purposes. However, the applicability and optimal concentration of TBP for many coral species is not yet known. Furthermore, TBP is an unstable compound, which may present both challenges and benefits to its potential use for restoration purposes. In order to assess the utility of this compound for restoration, settlement induction by TBP was assessed among a wide range of Caribbean coral species. Additionally, a suite of halogenated compounds (tribromopyrrole, pentabromopseudolin, dibromophenol, tribromophenol, bromophene, n-methyl tetrabromopyrrole, tetrachloropyrrole, dibromoindole, n-methyl tetrachloropyrrole and dibromopyridine) that are related to TBP, some of which have similar antibiotic and antialgal properties, were also tested for settlement induction activity. These compounds were chosen based on their structural similarity to TBP or their identity as a product within the bacterial TBP biosynthetic pathway. TBP induced settlement in nine of ten coral species tested including seven not previously reported (Dendrogyra cylindrus, Orbicella faveolata, Colpophyllia natans, Diploria labyrinthiformis, Pseudodiploria clivosa, Acropora cervicornis, Montastraea cavernosa) at concentrations ranging from 0.375 - 1.5 μM. The dire state of coral reefs demands a rapid increase in the scale and efficiency of coral restoration methods in addition to mitigating local and global stressors. Larval propagation can provide vast numbers of coral propagules from an individual spawning event and increased genetic diversity in restored populations. The conversion of embryos collected from wild, broadcast‐spawning populations to settlers that can be outplanted to the reef is a key component of this production process. We present preliminary results on settlement yield (i.e. % embryos converted to settled polyps on outplantable substrates) following in situ mass culture in floating mesocosms (Coral Rearing In‐situ Basins, or CRIBs; 5.6 m3 volume, 5.4 m2 surface area) that can be implemented independent of land‐based facilities. Ten trials over 2 years were conducted in three locations using five Caribbean broadcast‐spawning species. Embryos were added at different stocking densities and settlement was scored 2–4 weeks after fertilization. Two trials failed, resulting in no effective settlement, but the remaining eight trials resulted in between 1% and 11% settlement yield (overall mean 5.3%) and 77–100% of substrates exposed to larvae acquired settlers (average production: 700 substrates trial−1). Parallel land‐based trials showed a similar range (<1–14%) and mean (3.6%) settlement yield over nine trials. These values are also similar to the previously published lab and field‐based trials using Pacific Acropora spp. Continued optimization of CRIB design and execution is expected to improve consistency, overall yield, and efficiency in the production of sexual propagules for restoration. As coral reefs continue to decline due to climate change and other stressors, scientists have proposed adopting genomic tools, such as biomarkers, to aid in the conservation and restoration of these threatened ecosystems. Biomarkers are easily measured indicators of biological processes that can be used to predict or diagnose health, resilience, and other key performance metrics. The ultimate goal of developing biomarkers is to determine the conservation value and utility of a given coral colony, including the host animal, its algal symbionts, and their microbial partners. However, this goal remains distant because most efforts have not yet moved beyond the initial discovery phase. We review recent progress in the development of coral molecular biomarkers from a practical standpoint and consider the many challenges that remain as roadblocks to large‐scale implementation. We caution practitioners that, while biomarkers are a promising technology, they are unlikely to be available for field application in the near future barring a rapid shift in research focus from discovery to subsequent validation and field trials. To facilitate such a shift, we propose a stepwise framework to guide additional study in this area, with the aim of accelerating practical molecular biomarker development to enhance coral restoration practice. The effects of multiple stressors on the early life stages of reef-building corals are poorly understood. Elevated temperature is the main physiological driver of mass coral bleaching events, but increasing evidence suggests that other stressors, including elevated dissolved inorganic nitrogen (DIN), may exacerbate the negative effects of thermal stress. To test this hypothesis, we investigated the performance of larvae of Orbicella faveolata and Porites astreoides, two important Caribbean reef coral species with contrasting reproductive and algal transmission modes, under increased temperature and/or elevated DIN. We used a fluorescence-based microplate respirometer to measure the oxygen consumption of coral larvae from both species, and also assessed the effects of these stressors on P. astreoides larval settlement and mortality. Overall, we found that (1) larvae increased their respiration in response to different factors (O. faveolata in response to elevated temperature and P. astreoides in response to elevated nitrate) and (2) P. astreoides larvae showed a significant increase in settlement as a result of elevated nitrate, but higher mortality under elevated temperature. This study shows how microplate respirometry can be successfully used to assess changes in respiration of coral larvae, and our findings suggest that the effects of thermal stress and nitrate enrichment in coral larvae may be species specific and are neither additive nor synergistic for O. faveolata or P. astreoides. These findings may have important consequences for the recruitment and community reassembly of corals to nutrient-polluted reefs that have been impacted by climate change. [ABSTRACT FROM AUTHOR]
