Abstract Background Health-related quality of life (HRQOL) in cancer patients has attracted increasing attention, which may be associated with self-rated health (SRH), anxiety, and depression. However, limited studies have focused on the mediating role of anxiety and depression in the relationship between SRH and HRQOL among cancer patients. Therefore, this study aims to explore the serial multiple mediating effects of anxiety and depression between SRH and HRQOL in cancer patients. Methods This cross-sectional study investigated a total of 565 hospitalized cancer patients in Anhui province in China from November 2020 to October 2021. SRH was assessed using a single-item measure, anxiety and depression were assessed using the Hospital Anxiety and Depression Scale (HADS) and HRQOL was assessed using the EuroQol-5 Dimension (EQ-5D, three-level version). Socio-demographic and clinical characteristics were analyzed using descriptive statistics. The relationships between SRH, anxiety, depression, and HRQOL were evaluated by Pearson correlation analysis. DNA and RNA methylation are essential epigenetic modifications that play a pivotal role in various physiological processes by modulating gene expression at the post-transcriptional level. Abnormal DNA/RNA methylation is closely linked to a range of human diseases, including cancer and neurological disorders. Therefore, the accurate and sensitive detection of DNA/RNA methylation is of great importance in both epigenetic biology research and disease diagnosis. Technological advancements have led to the comprehensive development of methylation detection methods. Among these, electrochemical techniques have garnered significant attention owing to their simplicity and effectiveness in detection. This review provides an overview of methylation detection technologies based on electrochemical principles developed over the past five years, including electrochemical, photoelectrochemical, electrochemiluminescence, and fluorescence methodologies. Additionally, it discusses the advantages and disadvantages of these approaches, challenges and advancements in methylation detection.

Abstract Background Extracellular vesicles (EVs) derived from various cell sources exert cardioprotective effects during cardiac ischemic injury. Our previous study confirmed that EVs derived from ischemic-reperfusion injured heart tissue aggravated cardiac inflammation and dysfunction. However, the role of EVs derived from normal cardiac tissue in myocardial ischemic injury remains elusive. Results In the present study, normal heart-derived EVs (cEVs) and kidney-derived EVs (nEVs) were isolated and intramyocardially injected into mice after myocardial infarction (MI). We demonstrated that administration of both cEVs and nEVs significantly improved cardiac function, reduced the scar size, and alleviated inflammatory infiltration into the heart. In addition, cardiomyocyte apoptosis was inhibited, whereas angiogenesis was enhanced in the hearts receiving cEVs or nEVs treatment. Moreover, intramyocardial injection of cEVs displayed much better cardiac protective efficacy than nEVs in murine MI models. RNA-seq and protein-protein interaction (PPI) network analysis revealed the protective mRNA clusters in both cEVs and nEVs. These mRNAs were involved in multiple signaling pathways, which may synergistically orchestrate to prevent the heart from further damage post MI. Conclusions Collectively, our results indicated that EVs derived from normal heart tissue may represent a promising strategy for cardiac protection in ischemic heart diseases.

Castanopsis hystrix Hook. f. & Thomson ex A. DC. 1863 (Fagaceae) is an evergreen broad-leaved tree with high economic and ecological value. In this study, the complete chloroplast genome of C. hystrix was sequenced, assembled and annotated. The plastome (plastid genome) of C. hystrix was 160,624 bp in size, consisting of a pair of inverted repeats (IRs, 25,699 bp), a large-single-copy (LSC, 90,276 bp) region, and a small-single-copy (SSC, 18,950 bp). The overall GC content of C. hystrix was 36.8%. A total of 133 genes were annotated, including 88 protein-coding genes (PCG), 37 transfer RNA genes (tRNA), and eight ribosomal RNA genes (rRNA). A maximum likelihood analysis showed that the Castanopsis species form a monophyletic clade. C. hystrix is most closely related to C. tibetana with 100% bootstrap support value. The result enriches the genomic data for the genus Castanopsis, which will contribute to future studies in phylogenetics and evolution.

Abstract Extracellular vesicles (EVs) exist throughout our bodies. We recently revealed the important role of intracardiac EVs induced by myocardial ischemia/reperfusion on cardiac injury and dysfunction. However, the role of EVs isolated from normal tissues remains unclear. Here we found that EVs, derived from murine heart, lung, liver and kidney have similar effects on macrophages and regulate the inflammation, chemotaxis, and phagocytosis of macrophages. Interestingly, EV‐treated macrophages showed LPS resistance with reduced expressions of inflammatory cytokines and enhanced phagocytic activity. Furthermore, we demonstrated that the protein content in EVs contributed to the activation of inflammation, while the RNA component mainly limited the excessive inflammatory response of macrophages to LPS. The enrichment of miRNAs, including miR‐148a‐3p, miR‐1a‐3p and miR‐143‐3p was confirmed in tissue EVs. These EV‐enriched miRNAs contributed to the inflammation remission in LPS induced macrophages through multiple pathways, including STAT3, P65 and SAPK/JNK. Moreover, administration of both EVs and EV‐educated macrophages attenuated septic injury and cytokine storm in murine CLP models. Taken together, the present study disclosed that EVs from normal tissues can orchestrate the homeostasis of macrophages and attenuate inflammatory injury of sepsis. Therefore, tissue derived EVs or their derivatives may serve as potential therapeutic strategies in inflammatory diseases.

Vehicle electrification has emerged as a global strategy to address climate change and emissions externalities from the transportation sector. Deployment of charging infrastructure is needed to accelerate technology adoption; however, managers and policymakers have had limited evidence on the use of public charging stations due to poor data sharing and decentralized ownership across regions. In this article, we use machine learning based classifiers to reveal insights about consumer charging behavior in 72 detected languages including Chinese. We investigate 10 years of consumer reviews in East and Southeast Asia from 2011 to 2021 to enable infrastructure evaluation at a larger geographic scale than previously available. We find evidence that charging stations at government locations result in higher failure rates with consumers compared to charging stations at private points of interest. This evidence contrasts with predictions in the U.S. and European markets, where the performance is closer to parity. We also find that networked stations with communication protocols provide a relatively higher quality of charging services, which favors policy support for connectivity, particularly for underserved or remote areas.

Abstract Background and objectives Research suggests that fibrinogen (Fib) concentrations are used to assess the occurrence and severity of diabetic foot (DF) and to monitor the progression of diabetic foot in patients. However, its correlation with Fib function has not been reported. Here, angle α and k value, reflecting the Fib function, were used to analyse its correlation with DF, and their potential as biological indicators for evaluating the occurrence and severity of DF was explored. Subjects and methods This clinical study enrolled 163 type 2 diabetes mellitus (T2DM) patients, who were divided into the diabetes with DF (84 cases) group, diabetes with no DF (79 cases) group. Meanwhile, 90 healthy unrelated subjects were enrolled as controls. Results Angle α and fibrinogen levels increased greatly in subjects with DF compared with those without. Abstract Background Human umbilical cord-derived mesenchymal stem cells (HucMSCs) have been recognized as a promising cell for treating myocardial infarction (MI). Inflammatory response post MI is critical in determining the cardiac function and subsequent adverse left ventricular remodeling. However, the local inflammatory effect of HucMSCs after intramyocardial injection in murine remains unclear. Methods HucMSCs were cultured and transplanted into the mice after MI surgery. Cardiac function of mice were analyzed among MI-N.S, MI-HucMSC and MI-HucMSC-C–C Motif Chemokine receptor 5 (CCR5) antagonist groups, and angiogenesis, fibrosis and hypertrophy, and immune cells infiltration of murine hearts were evaluated between MI-N.S and MI-HucMSC groups. We detected the expression of inflammatory cytokines and their effects on CD4+ T cells migration. Results HucMSCs treatment can significantly improve the cardiac function and some cells can survive at least 28 days after MI. Intramyocardial administration of HucMSCs also improved angiogenesis and alleviated cardiac fibrosis and hypertrophy. Moreover, we found the much higher numbers of CD4+ T cells and CD4+FoxP3+ regulatory T cells (Tregs) in the heart with HucMSCs than that with N.S treatment on day 7 post MI. In addition, the protein level of C–C Motif Chemokine Ligand 5 (CCL5) greatly increased in HucMSCs treated heart compared to MI-N.S group. In vitro, HucMSCs inhibited CD4+ T cells migration and addition of CCL5 antibody or CCR5 antagonist significantly reversed this effect. In vivo results further showed that addition of CCR5 antagonist can reduce the cardioprotective effect of HucMSCs administration on day 7 post MI injury. Conclusion These findings indicated that HucMSCs contributed to cardiac functional recovery and attenuated cardiac remodeling post MI. Intramyocardial injection of HucMSCs upregulated the CD4+FoxP3+ Tregs and contributed to the migration of CD4+ T cells into the injured heart via CCL5/CCR5 pathway.

In large-scale wind turbines, the force state of the pitch system greatly influences safe operation and service life. This paper provides a novel method to estimate blade pitch load, bearing friction torque, and motor pitch torque. In this method, the force equilibrium equations are established by investigating the force of the pitch system under multiple operating conditions. The multidimensional BIN method is employed to classify the supervisory control and data acquisition (SCADA) data of wind turbines into several intervals. The multidimensional scatter data is processed in a single-valued way. Then, the estimating model of the pitch system forces is established by combining the obtained data and the equilibrium equations. Taking a 2 MW wind turbine as an example, the variation characteristics of blade pitch load, bearing friction torque, and motor pitch torque under multiple operating conditions are analyzed. Some interesting and valuable conclusions are obtained. For example, when the wind speed increases, the blade pitch load increases significantly in the maximum wind energy tracking region, but there is no obvious change in the observed constant power output region. The wind speed and azimuth have little effect on the bearing friction torque. The variation trend of motor pitch torque is consistent with that of blade pitch load in the maximum wind energy tracking region.

Excessive and chronic inflammation post myocardial infarction (MI) causes cardiac fibrosis and progressive ventricular remodeling, which leads to heart failure. We previously found high levels of IL-27 in the heart and serum until day 14 in murine cardiac ischemia‒reperfusion injury models. However, whether IL-27 is involved in chronic inflammation-mediated ventricular remodeling remains unclear. In the present study, we found that MI triggered high IL-27 expression in murine cardiac macrophages. The increased expression of IL-27 in serum is correlated with cardiac dysfunction and aggravated fibrosis after MI. Furthermore, the addition of IL-27 significantly activated the JAK/STAT signaling pathway in cardiac fibroblasts (CFs). Meanwhile, IL-27 treatment promoted the proliferation, migration and extracellular matrix (ECM) production of CFs induced by angiotensin II (Ang II). Collectively, high levels of IL-27 mainly produced by cardiac macrophages post MI contribute to the activation of CFs and aggravate cardiac fibrosis.

Abstract Mitochondrial dysfunction is considered to be a key contributor to the development of heart failure. Replacing injured mitochondria with healthy mitochondria to restore mitochondrial bioenergy in myocardium holds great promise for cardioprotection after infarction. This study aimed to investigate whether direct transplantation of exogenous mitochondria derived from mesenchymal stem cells (MSC‐mt) is beneficial and superior in protecting cardiac function in a mouse model of myocardial infarction (MI) compared to mitochondria derived from skin fibroblast (FB‐mt) and to explore the underlying mechanisms from their effects on the endothelial cells. The isolated MSC‐mt presented intact mitochondrial morphology and activity, as determined by electron microscopy, JC‐1 mitochondrial membrane potential assay, and seahorse assay. Direct injection of MSC‐mt into the peri‐infarct region in a mouse MI model enhanced blood vessel density, inhibited cardiac remodeling and apoptosis, thus improving heart function compared with FB‐mt group. The injected MSC‐mt can be tracked in the endothelial cells. In vitro, the fluorescence signal of MSC‐mt can be detected in human umbilical vein endothelial cells (HUVECs) by confocal microscopy and flow cytometry after coculture. Compared to FB‐mt, MSC‐mt more effectively protected the HUVECs from oxidative stress‐induced apoptosis and reduced mitochondrial production of reactive oxygen species. MSC‐mt presented superior capacity in inducing tube formation, enhancing SCF secretion, ATP content and cell proliferation in HUVECs compared to FB‐mt. Mechanistically, MSC‐mt administration alleviated oxidative stress‐induced endothelial senescence via activation of ERK pathway. These findings suggest that using MSCs as sources of mitochondria is feasible and that proangiogenesis could be the mechanism by which MSC‐mt transplantation attenuates MI. MSC‐mt transplantation might serve as a new therapeutic strategy for treating MI.

Overactive inflammatory responses contribute to progressive cardiac dysfunction after myocardial infarction (MI). Mesenchymal stem cell (MSC) has generated significant interest as potent immune modulators that can regulate excessive immune responses. We hypothesized that intravenous (iv) administration of human umbilical cord-derived MSC (HucMSC) exerts systemic and local anti-inflammation effects, leading to improved heart function after MI. In murine MI models, we confirmed that single iv administration of HucMSC (30×104) improved cardiac performance and prevented adverse remodeling after MI. A small proportion of HucMSC is trafficked to the heart, preferentially in the infarcted region. HucMSC administration increased CD3+ T cell proportion in the periphery while decreased T cell proportion in both infarcted heart and mediastinal lymph nodes (med-LN) at 7-day post-MI, indicating a systematic and local T cell interchange mediated by HucMSC. The inhibitory effects of HucMSC on T cell infiltration in the infarcted heart and med-LN sustained to 21-day post-MI. Our findings suggested that iv administration of HucMSC fostered systemic and local immunomodulatory effects that contributed to the improvement of cardiac performance after MI.

Gynura japonica (Asteraceae) is a folk herbal medicine with multi-pharmacological functions involving analgesic, hemostatic and antiangiogenic activities. The study was conducted to assemble the complete chloroplast (cp) genome of G. japonica through a genome-skimming approach. The assembled cp genome was 151,023 bp in size, with 62.8% AT content, consisting of a large single copy (LSC) of 83,185 bp, two copies of inverted repeat (IRa and IRb) of 24,847 bp, and a small single copy (SSC) of 18,144 bp. The cp genome of G. japonica contained 133 genes, including eight ribosomal RNA genes (rRNAs), 37 transfer RNA genes (tRNAs), 86 protein-coding genes (PCGs), and two pseudogenes (ψycf1 and ψrps19). Our phylogenomic analysis based on whole plastid genomes strongly supports G. japonica is a sister to the clade including Crassocephalum crepidioides and Jacobaea vulgaris.

Abstract Background Application of mesenchymal stem cell-derived exosomes (MSC-EXO) has emerged as a novel therapeutic strategy for myocardial infarction (MI). Our previous study showed that pretreatment with hemin, a potent heme oxygenase-1 (HO-1) inducer, enhanced the cardioprotective effects of MSCs in a mouse model of MI. This study aimed to investigate the therapeutic effects of EXO derived from hemin-pretreated MSCs (Hemin-MSC-EXO) in MI and explore the potential mechanisms. Methods MSC-EXO and Hemin-MSC-EXO were collected and characterized. MSC-EXO and Hemin-MSC-EXO were intramuscularly injected into the peri-infarct region in a mouse model of MI. Heart function of mice was assessed by echocardiography. The mitochondrial morphology of neonatal mice cardiomyocytes (NMCMs) under serum deprivation and hypoxic (SD/H) conditions was examined by Mitotracker staining. The cellular senescence of NMCMs was determined by senescence-associated-β-galactosidase assay. A loss-of-function approach was adopted to determine the role of Hemin-MSC-exosomal-miR-183-5p in the regulation of cardiomyocyte senescence Results EXO were successfully isolated from the supernatant of MSCs and Hemin-pretreated MSCs. Compared with MSC-EXO, injection of Hemin-MSC-EXO significantly improved cardiac function and reduced fibrosis. Both MSC-EXO and Hemin-MSC-EXO ameliorated cardiomyocyte senescence and mitochondrial fission in vitro and in vivo, and the latter exhibited better protective effects. MicroRNA sequencing revealed a higher level of miR-183-5p in Hemin-MSC-EXO than in MSC-EXO. MiR-183-5p knockdown partially abrogated the protective effects of Hemin-MSC-EXO in attenuating mitochondrial fission and cellular senescence of cardiomyocytes induced by SD/H. High mobility group box-1 (HMGB1) abundance was lower in Hemin-MSC-EXO-treated than MSC-EXO-treated mouse hearts, and HMGB1 was identified as one of the potential target genes of miR-183-5p. Mechanistically, Hemin-MSC-EXO inhibited SD/H-induced cardiomyocyte senescence partially by delivering miR-183-5p into recipient cardiomyocytes via regulation of the HMGB1/ERK pathway. Furthermore, knockdown of miR-183-5p reduced the Hemin-MSC-EXO-mediated cardioprotective effects in a mouse model of MI. Conclusion Our results reveal that Hemin-MSC-EXO are superior to MSC-EXO in treating MI. Exosomal miR-183-5p mediates, at least partially, the cardioprotective effects of Hemin-MSC-EXO by inhibiting cardiomyocyte senescence via regulation of the HMGB1/ERK pathway. This study highlights that MSC-EXO have high translational value in repairing cardiac dysfunction following infarction. Graphic abstract

Nanhaia speciosa (Fabaceae) is a woody perennial vine used as an important traditionally Chinese medicine. In this study, the complete chloroplast genome of Nanhaia speciosa was sequenced and assembled. The chloroplast genome of N. speciosa was 132,551bp in length including only one copy of the inverted repeat (IR). It encoded a total of 110 genes, containing 76 protein-coding genes, 30 tRNA and 4 rRNA. The overall GC content was 34.1%. Phylogenetic analysis using a matrix of 69 protein-coding genes illustrated that N. speciosa is most closely related to Wisteriopsis reticulata of tribe Wisterieae.

Abstract Background As China’s aging trend intensifies and the proportion of empty nests increases, the health-related quality of life of the elderly is the focus of social attention. Previous studies focused on the quality of life of the elderly, rather than empty nesters, and lacked the discussion of the mechanism of influencing factors. Thus, this study aimed to explore the influencing factors of the quality of life (QOL) and interaction mechanisms among empty nesters in Shanxi Province, China, so as to provide practical reference for improving the QOL of empty nesters. Methods A total of 4901 empty nesters in Shanxi Province, China, were selected using multi-stage random cluster sampling method in this cross-sectional study. The quality of life was assessed with the Short Form 36 Health Survey (SF-36). Multiple linear regression analysis (stepwise) was performed to examine the factor associated with QOL. Structural equation model (SEM) approach was conducted to test the direct and indirect association between QOL influencing factors with QOL. Results The average QOL score of empty nesters was 63.34 (SD = 17.23). The multiple linear regression revealed that gender, age, education, monthly income, drinking status, exercise frequency, physical examination frequency, attention to daily nutritional health, relationship with a spouse and relationship with children were significant predictors of the empty nester’s QOL (P

Abstract Background Mesenchymal stem cells (MSCs) can improve cutaneous wound healing via the secretion of growth factors. However, the therapeutic efficacy of MSCs varies depending upon their source. Induced pluripotent stem cells are emerging as a promising source of MSCs with the potential to overcome several limitations of adult MSCs. This study compared the effectiveness of conditioned medium of MSCs derived from induced pluripotent stem cells (iMSC-CdM) with that derived from umbilical cord MSCs (uMSC-CdM) in a mouse cutaneous wound healing model. We also investigated the mechanisms of protection. Methods The iMSC-CdM or uMSC-CdM were topically applied to mice cutaneous wound model. The recovery rate, scar formation, inflammation and angiogenesis were measured. We compared angiogenesis cytokine expression between iMSC-CdM and uMSC-CdM and their protective effects on human umbilical vein endothelial cells (HUVECs) under H2O2-induced injury. The effects of iMSC-CdM on energy metabolism, mitochondria fragmentation and apoptosis were measured. Results Topical application of iMSC-CdM was superior to the uMSC-CdM in accelerating wound closure and enhancing angiogenesis. Expression levels of angiogenetic cytokines were higher in iMSC-CdM than they were in uMSC-CdM. The iMSC-CdM protected HUVECs from H2O2 induced injury more effectively than uMSC-CdM did. Administration of iMSC-CdM stimulated HUVEC proliferation, tube formation and energy metabolism via the ERK pathway. Mechanistically, iMSC-CdM inhibited H2O2-induced mitochondrial fragmentation and apoptosis of HUVECs. Conclusion Collectively, these findings indicate that iMSC-CdM is more effective than uMSC-CdM in treating cutaneous wounds, and in this way, iMSC-CdM may serve as a more constant and sustainable source for cell-free therapeutic approach. The key problem to be solved in the process of wind turbine (WT) operation and maintenance is to obtain the wind turbine performance accurately. The power curve is an important indicator to evaluate the performance of wind turbines. How to model and obtain the power curve of wind turbines has always been one of the hot topics in research. This paper proposes a novel idea to get the actual power curve of wind turbines. Firstly, the basic data preprocessing algorithm is designed to process the zero value and null value in the original supervisory control and data acquisition (SCADA) data. The moving average filtering (MAF) method is employed to deal with the wind speed, the purpose of which is to consider the comprehensive result of wind on the wind turbine power in a certain period. According to the momentum theory of the ideal wind turbine and combined with the characteristics of the anemometer installation position, the deviation between the measured wind speed and the actual wind speed is approximately corrected. Here, the influence of dynamic changes in air density is also considered. Then, the Gaussian fitting algorithm is used to fit the wind-power curve. The characteristics of the power curve before and after wind speed correction are compared and analyzed. At the same time, the influence of the parameter uncertainty on the reliability of the power curve is considered and investigated. Finally, the characteristics of the power curves of four wind turbines are compared and analyzed. The research results show that among these power curves, WT3 and WT4 are the closest, WT2 is the next, and WT1 has the farthest deviation from the others. The research work provides a valuable basis for on-site performance evaluation, overhaul, and maintenance of wind turbines.

Abstract Background Apocynum venetum L. is an important medicinal plant that is mainly distributed in the coastal areas and northwest of China. In addition to its high medical and economic value, its adaptation to saline-alkali and coastal saline lands makes A. venetum an ideal candidate for use in vegetation restoration. To date, the study of A. venetum has been limited in the northwest region of China, little attention has been paid to the genetic diversity and population structure of A. venetum populations in the coastal region. Here, we performed transcriptome sequencing of total RNA from A. venetum leaves and developed efficient expressed sequence tag-simple sequence repeat (EST-SSR) markers for analyzing the genetic diversity and population structure of A. venetum in the coastal region. Results A total of 86,890 unigenes were generated after de novo assembly, and 68,751 of which were successfully annotated by searching against seven protein databases. Furthermore, 14,072 EST-SSR loci were detected and 10,243 primer pairs were successfully designed from these loci. One hundred primer pairs were randomly selected and synthesized, twelve primer pairs were identified as highly polymorphic and further used for population genetic analysis. Population genetic analyses showed that A. venetum exhibited low level of genetic diversity (mean alleles per locus, N A = 3.3; mean expected heterozygosity, H E = 0.342) and moderate level of genetic differentiation among the populations (genetic differentiation index, F ST = 0.032–0.220) in the coastal region. Although the contemporary (mean m c = 0.056) and historical (mean m h = 0.106) migration rates among the six A. venetum populations were moderate, a decreasing trend over the last few generations was detected. Bayesian structure analysis clustered six populations into two major groups, and genetic bottlenecks were found to have occurred in two populations (QG, BH). Conclusions Using novel EST-SSR markers, we evaluated the genetic variation of A. venetum in the coastal region and determined conservation priorities based on these findings. The large dataset of unigenes and SSRs identified in our study, combining samples from a broader range, will support further research on the conservation and evolution of this important coastal plant and its related species.

Although mesenchymal stem cell- (MSC-) based therapy has shown promising results for myocardial infarction (MI), low cell survival heavily limits its beneficial effects. Apelin plays an essential regulatory role in cell proliferation. This study was aimed at determining whether Apelin-13 pretreatment could improve the survival of MSCs in the ischemic heart and enhance their cardioprotective efficacy against MI. MSCs were pretreated with or without Apelin-13 for 24 hours and then exposed to serum deprivation and hypoxia (SD/H) for 48 hours. The mitochondrial morphology of MSCs was assessed by MitoTracker staining. The apoptosis of MSCs was determined by TUNEL staining. The level of mitochondrial reactive oxygen species (ROS) of MSCs was detected by Mito-Sox staining. MSCs and Apelin-13-pretreated MSCs were transplanted into the peri-infarct region in a mouse MI model. Apelin-13 pretreatment protected MSCs against SD/H-induced mitochondrial fragmentation and apoptosis. Apelin-13 pretreatment reduced ROS generation induced by SD/H in MSCs. Furthermore, Apelin-13 pretreatment enhanced the angiogenesis of MSCs under SD/H conditions. Mechanistically, Apelin-13 pretreatment inhibited SD/H-induced MSC apoptosis by downregulating mitochondrial fission via activation of the ERK pathway, and these effects were partially abrogated by ERK inhibitor U0126. Apelin-13 pretreatment promoted the survival of MSCs in the ischemic heart. Moreover, transplantation with Apelin-13-pretreated MSCs improved heart function and increased angiogenesis accompanied by decreased fibrosis compared with MSC transplantation at 28 days following MI. These findings reveal that pretreatment with Apelin-13 improves MSCs survival and enhances their therapeutic efficacy for MI. Our study provides a novel approach to improve MSC-based therapy for cardiovascular disease.

Data pre-processing is the first step of using SCADA data to study the performance of wind turbines. However, there is a lack of knowledge of how to obtain more effective data pre-processing algorithms. This paper fully explores multiple data pre-processing algorithms for power curve modeling. A three-stage data processing mode is proposed, namely, preliminary data filtering and compensation (Stage I), secondary data filtering (Stage II), and single-valued processing (Stage Ⅲ). Different data processing algorithms are selected at different stages and are finally merged into nine data processing algorithms. A novel evaluation method based on energy characteristic consistency (ECC) is proposed to evaluate the reliability of various algorithms. The influence of sliding mode and benchmark of Binning on data processing has been fully investigated through indicators. Four wind turbines are selected to verify the advantages and disadvantages of the nine data processing methods. The result shows that at the same wind speed, the rotational speed and power values obtained by MLE (maximum likelihood estimation) are relatively high among the three single-valued methods. Among the three outlier filtering methods, the power value obtained by KDE (kernel density estimation) is relatively large. In general, KDE-LSM (least square method) has good performance in general. The sum of four evaluating index values obtained by KDE-LSM from four wind turbines is the smallest.

This article reports an empirical study on embedding digital literacies in the language teacher education curriculum with pre-service and in-service teachers enrolled in a graduate-level language teaching method course. Two different types of technology-mediated collaborative tasks were implemented, one being collaborative reading for writing using Perusall and Google Docs, and the other being collaborative infographic composing using Canva. The analyses of the participants' semi-structured individual interviews revealed multiple benefits of digital practices that the pre-service and in-service teachers perceived, including enhancing content learning, providing cognitive scaffolding, fostering communication/collaboration skills, fostering creativity, enhancing learning motivation, improving pedagogic skills, and developing digital literacy skills. They also reported detailed plans or practices for implementing digital pedagogy using the same technology tools, adopting similar tasks, and extending the digital tasks. This study contributes to the underrepresented research domain on digital pedagogy for language teacher training.

Abstract Background Some Gynura species have been reported to be natural anti-diabetic plants. Improvement of their traits towards application relies on hybridization. Clearly, phylogenetic relationships could optimize compatible hybridizations. For flowerings plants, chloroplast genomes have been used to solve many phylogenetic relationships. To date, the chloroplast genome sequences of 4 genera of the tribe Senecioneae have been uploaded to GenBank. The internal relationships within the genus Gynura and the relationship of the genus Gynura with other genera in the tribe Senecioneae need further research. Results The chloroplast genomes of 4 Gynura species were sequenced, assembled and annotated. In comparison with those of 12 other Senecioneae species, the Gynura chloroplast genome features were analysed in detail. Subsequently, differences in the microsatellite and repeat types in the tribe were found. From the comparison, it was found that IR expansion and contraction are conserved in the genera Gynura, Dendrosenecio and Ligularia. Compared to other regions on the chloroplast genome, the region from 25,000 to 50,000 bp was not conserved. Seven ndh genes in this region are under purifying selection, with small changes in amino acids. The whole chloroplast genome sequences of 16 Senecioneae species were used to build a phylogenetic tree. Based on the oldest Artemisia pollen fossil, the divergence time was estimated. Conclusions Sequencing the chloroplast genomes of 4 Gynura species helps us to solve many problems. The phylogenetic relationships and divergence time among 4 Gynura and 16 Senecioneae species were evaluated by comparing their chloroplast genomes. The phylogenetic relationship of the genera Gynura and Ligularia was different from that observed previous work. In a previous phylogenetic tree, the genus Ligularia belonged to the Tussilagininae subtribe, which was in a lineage that diverged earlier than other genera. Further morphology and genome-wide analyses are needed to clarify the genus relationships.

Cnidoscolus aconitifolius is a leafy green heathy vegetable and medicinal plant belongs to the family Euphorbiaceae. In the present study, we sequenced the complete chloroplast genome of C. aconitifolius, which is 158,658 bp in length and consisted of two copies of inverted repeat (IR) of 26,982 bp separated by a large single copy (LSC) of 87,022 bp and a small single copy (SSC) of 17,672 bp. The GC content of C. aconitifolius was 36.3%. A total of 130 genes were predicted, including 86 protein-coding genes, 36 tRNAs and 8 rRNAs. The plastid phylogenomic analysis support C. aconitifolius is closely related to Manihot esculenta.

Profiling bodily fluids is crucial for monitoring and discovering metabolic markers of disease. In this study, a comprehensive analysis approach based on 1D-LC-MS/MS and 2D-LC-MS/MS was applied to profile normal human urine metabolites from 348 children and 315 adults. A total of 2357 metabolites were identified, including 1831 endogenous metabolites and 526 exogenous ones. In total, 1005 metabolites were identified in urine for the first time. The urinary metabolites were mainly involved in amino acid metabolism, small molecule biochemistry, lipid metabolism and cellular compromise. The comparison of adult’s and children’s urine metabolomes showed adults urine had more metabolites involved in immune response than children’s, but the function of binding of melatonin, which belongs to the endocrine system, showed a higher expression in children. The urine metabolites detected by the 1D-LC-MS/MS method were mainly related to amino acid metabolism and lipid metabolism, and the 2D-LC-MS/MS method not only explored metabolites from 1D-LC-MS/MS but also metabolites related to cell signaling, cell function and maintenance, etc. Our analysis comprehensively profiled and functionally annotated the metabolome of normal human urine, which would benefit the application of urinary metabolome to clinical research.

The primary wind turbines’ in-service performance evaluation method is mining and analyzing the SCADA data. However, there are complex mathematical and physical relationships between multiple operating parameters, and so far, there is a lack of systematic understanding. To solve this issue, the distribution of wind turbines’ operating parameters was first analyzed according to the characteristics of the energy flow of wind turbines. Then, the correlation calculation was performed using the Spearman correlation coefficient method based on the minute-level data and second-level data. According to the numerical characteristics of the nacelle vibration acceleration, the data preprocessing technology sliding window maximum (SWM) was proposed during the calculation. In addition, taking temperature correlation as an example, two-dimensional scatter (including single-valued scatter) and three-dimensional scatter features were combined with numerical analysis and physical mechanism analysis to understand the correlation characteristics better. On this basis, a quantitative description model of the temperature characteristics of the gearbox oil pool was constructed. Through this research work, the complex mathematical and physical relationships among the multi-parameters of the wind turbines were comprehensively obtained, which provides data and theoretical support for the design, operation, and maintenance.

Alnus formosana (Betulaceae) is an important ecological and economic deciduous tree species widely distributed throughout subtropical regions of Taiwan province, China. At the present study, the complete chloroplast genome of A. formosana was assumbled using next-generation sequencing technology. The complete chloroplast sequence is 161,029 bp in length, which consisted of a large single copy (LSC, 89,720 bp) and a small single copy (SSC; 19,205 bp) separated a pair of inverted repeats (IRs; 26,052 bp). The overall guanine-cytosine (GC) content was 36.4%. A total of 131 genes were annotated, including 85 protein-coding genes, 37 tRNAs, eight rRNAs and one pseudogene (ψycf1). The phylogenetic analysis fully resolved A. formosana in a clade with A. japonica. The plastome of A. formosana will provide informative genomic resources for further phylogenetic application and genetic improvement.

The protective effects of mesenchymal stem cell (MSC)-based therapy for myocardial infarction (MI) are largely hampered as they age. Apelin is an endogenous ligand of its receptor APJ and plays an essential role in regulating multiple biological activities including MSC proliferation and survival. In this study, we investigated whether Apelin regulates MSC senescence and whether its overexpression could rejuvenate aged MSCs (AMSCs) to improve cardiac protection following infarction in mice. MSC senescence was evaluated by senescence-associated β-galactosidase assays. Apelin level was examined by western blotting. Autophagy was determined by transmission electron microscopy. The cardioprotective effect of AMSCs with Apelin overexpression (Apelin-AMSCs) was assessed in a mouse MI model. Apelin expression was dramatically reduced in AMSCs. Interestingly, knockdown of Apelin induced young MSCs (YMSC) senescence, whereas overexpression rescued AMSC senescence. Apelin overexpression also increased AMSC angiogenic capacity. Mechanistically, Apelin overexpression upregulated the autophagy level of AMSCs by activating AMP-activated protein kinase (AMPK) signaling, thereby rejuvenating AMSCs. Compared with AMSCs, transplantation of Apelin-AMSCs achieved better therapeutic efficacy for MI by enhancing cell survival and angiogenesis. In conclusion, our results reveal that Apelin activates AMPK to rejuvenate AMSCs by increasing autophagy and promotes cardioprotection following infarction in mice. This study identified a novel target to rejuvenate AMSCs and enhance their therapeutic efficacy.

Abstract Extracellular vesicles (EVs) curb important biological functions. We previously disclosed that ischemia‐reperfusion (IR) induces increased release of EVs (IR‐EVs) in the heart. However, the role of IR‐EVs in IR pathological process remains poorly understood. Here we found that adoptive transfer of IR‐EVs aggravated IR induced heart injury, and EV inhibition by GW4869 reduced the IR injury. Our in vivo and in vitro investigations substantiated that IR‐EVs facilitated M1‐like polarization of macrophages with increased expression of proinflammatory cytokines. Further, we disclosed the miRNA profile in cardiac EVs and confirmed the enrichment of miRNAs, such as miR‐155‐5p in IR‐EVs compared to EVs from the sham heart (S‐EVs). In particular, IR‐EVs transferred miR‐155‐5p to macrophages and enhanced the inflammatory response through activating JAK2/STAT1 pathway. Interestingly, IR‐EVs not only boosted the local inflammation in the heart, but even triggered systemic inflammation in distant organs. Taken together, we newly identify an IR‐EVs–miR‐155‐5p–M1 polarization axis in the heart post IR. The EVs derived from IR‐injured heart contribute to both local and systemic inflammation. Importantly, EV inhibition by GW4869 is supposed to be a promising therapeutic strategy for IR injury.

Abstract Background In China, the problems of population aging and empty nesting have become important issues which will affect the social stability and economic development. The aim of this study was to explore the health promoting lifestyles and influencing factors among empty nesters and compare with non-empty nesters to find out their differences, so as to provide a scientific evidence for people to formulate health management strategies for elderly. Methods A cross-sectional survey which used a stratified random cluster sampling method, was conducted among 500 elders in six districts of Taiyuan, China, there were 288 empty nesters and 212 non-empty nesters. The general information and health- promoting lifestyles were investigated by using the self-made General Information Questionnaire and Health Promoting Lifestyle Scale(HPLP). Two-sample t-test and Chi-square test were used to compare the sociodemographic factors, HPLP scores of empty nesters to non-empty nesters; Multiple stepwise linear regression was performed to estimate influencing factors related to the HPLP of empty nesters and non-empty nesters. Castanea sativa Mill. is mainly grown in the temperate regions of continental Europe, and it has a considerable economic value. In this study, the complete chloroplast genome sequence of Castanea sativa was characterized. Leaves were collected from the National Botanical Garden of Latvia. The chloroplast genome was determined to be 160,938 bp in length. It contained large single-copy (LSC) and small single-copy (SSC) regions of 90,519 and 18,967 bp, respectively, which were separated by a pair of 25,726 bp inverted repeat (IR) regions. The genome is predicted to contain 130 genes, including 83 protein-coding genes, 37 tRNA genes, eight rRNA genes, and two pseudo genes. The overall GC content of the genome is 36.8%. A phylogenetic tree reconstructed by 34 chloroplast genomes reveals that C. sativa is most closely related to the clade including C. henryi, C. seguinii and C. mollissima.

The Castanea mollissima ‘Chuizhili’ is an important variety of C. mollissima in breeding for dwarf chestnut and ornamental trees due to the weeping characteristic in China. In this study, the complete chloroplast genome of C. mollissima ‘Chuizhili’ was identified and sequenced by using Illumina sequencing data. The genome size is 160,799 bp, with a large single-copy (LSC, 90,430 bp) region, a small single-copy (SSC, 18,997 bp) region, and separated by a pair of 25,686 bp inverted repeat (IR) regions. A total of 130 genes are successfully annotated, including 83 protein-coding genes, 37 tRNA genes, 8 rRNA genes, and 2 pseudo-genes. The phylogenetic relationships revealed that C. mollissima ‘Chuizhili’ is closely related to Castanea henryi in Fagaceae.

The complete chloroplast genome sequence of the Tertiary relict tree Zelkova serrata was reported in this study. The chloroplast genome is 158,875 bp in length with a typical angiosperm quantitative structure consisting of a large single copy (87,412 bp) and a small single copy (18,683 bp) separated by a pair of inverted repeat (26,390 bp). Genome annotation revealed a total of 129 genes comprising 84 protein-coding genes, 37 tRNA genes, and eight rRNA genes. Phylogenomic analysis based on the whole plastomes indicated that Z. serrata and Z. schneideriana formed a well-supported monophyletic clade sister to genus Ulmus in Ulmaceae.

Hosta plantaginea is an important ornamental and horticultural plant endemic to China. In this study, we generated complete chloroplast genome of H. plantaginea using high-throughput sequencing. The complete chloroplast sequence is a circular molecule of 157,091 bp in size, consisting of a large single copy (LSC, 86,061 bp) and a small single copy (SSC; 18,282 bp) separated by a pair of inverted repeats (IRs; 26,374 bp), The total GC content is 37.8%, with 35.9, 31.7, and 43.0% in LSC, SSC, and IRs, respectively. A total of 132 genes are annotated, including 84 protein-coding genes, 38 tRNAs, 8 rRNAs, and 2 pseudogenes. The phylogenetic analysis revealed that H. plantaginea was formed to be the early diverging species within Hosta.

Philadelphus pekinensis Rupr. is a common perennial deciduous shrub distributed in temperate China. Here, we report the complete chloroplast genome of P. pekinensis. The cp genome is totally 157,308 bp in length, including large single-copy (LSC) region of 86,457 bp, small single-copy (SSC) region of 18,735 bp, and two separated inverted regions (IRs) of 26,058 bp, respectively. It contains 134 genes, including 85 protein-coding genes, 8 rRNA genes, and 37 tRNA genes. The overall GC content is 43.1%. The phylogenetic analysis reveals the monophyly of P. pekinensis and P. calvescens, which is more related with Carpenteria californica than other species in the Hydrangeaceae family.

The tourists’ value of traditional village representing personal values, influences the tourists’ behavior deeply. This paper, with the soft ladder method of MEC theory from the perspective of the tourist, studies the value of tourists born in the 60s, 70s, 80s, and 90s of the traditional villages in Hongcun and Xidi, which indicates 39 MEC value chains, and reveals 11 important attributes of Huizhou traditional villages, 16 tourism results, and 9 types of tourists’ values. With constructing a sustainable development model of Huizhou traditional villages based on tourists’ value, it shows an inherent interaction between tourists’ value and traditional village attributes subdividing the tourism products and marketing channels of Huizhou traditional villages, which is of great significance to the sustainable development of traditional villages in Huizhou.

Saposhnikovia divaricata is traditional herbal medicine with a long history in China. We reported the complete chloroplast genome of S. divaricate using the next generation sequencing. A total of 115 unique genes were annotated, consisting of 81 protein coding genes, 30 tRNA and 4 rRNA. The overall AT content was 69.2%. The molecular phylogenetic tree reveals that S. divaricate is closely related to Peucedanum in tribe Selineae.

Soil salinity is one of the major environmental stresses affecting plant growth, development, and reproduction. Salt stress also affects the accumulation of some secondary metabolites in plants. Glehnia littoralis is an endangered medicinal halophyte that grows in coastal habitats. Peeled and dried Glehnia littoralis roots, named Radix Glehniae, have been used traditionally as a Chinese herbal medicine. Although Glehnia littoralis has great ecological and commercial value, salt-related mechanisms in Glehnia littoralis remain largely unknown. In this study, we analysed the transcriptome of Glehnia littoralis in response to salt stress by RNA-sequencing to identify potential salt tolerance gene networks. The complete chloroplast (cp) genome sequence is 160,104 bp in length, with a quantitative molecule structure comprising two copies of inverted repeats (IRa and IRb) of 26,737 bp separated by a large single copy (LSC) of 88,151bp, a small single copy (SSC) of 18,479 bp. A total of 131 genes were identified including 84 protein-coding genes, 37 tRNA genes, eight rRNA genes and two pseudogene (Ψycf1, ΨinfA), respectively. Phylogenomic analysis suggests that M. alternifolia is closely related to the rest species of Myrtaceae with strong bootstrap values.

Peucedanum praeruptorum is an important traditional herbal medicine unique to China. The complete chloroplast genome of P. praeruptorum was generated here using high-throughput sequencing. The plastome was 147,197 bp in size, which consisted of a pair of inverted repeats (IRs; 18,713 bp), a large single copy (LSC; 92,161 bp) and a small single copy (SSC; 17,610 bp). The GC content of the plastome was 37.6%, with 44.5%, 36.0%, and 31.1% in IRs, LSC, and SSC, respectively. A total of 128 genes were annotated, including 84 protein-coding genes, 35 tRNAs, eight rRNAs, and one pseudogene (Ψycf1). The phylogenomic analysis indicated that P. praeruptorum formed a monophyletic clade with Peucedanum japonicum.

Active wake management (AWM) via yaw control has been discussed in recent years as a potential way to improve the power production of a wind farm. In such a technique, the wind turbines will be required to work frequently at misaligned yaw angles in order to reduce the vortices in the wake area behind the turbines. However, today, it is still not very clear about how yaw operation affects the dynamics and power generation performance of the wind turbines. To further understand the effects of yaw operation, numerical research is conducted in this paper. In the study, the optimal size of the flow field used in the computational fluid dynamics (CFD) calculation was specifically discussed in order to obtain an efficient numerical model to quickly and accurately predict the dynamics and the performance of the turbines. Through this research, the correlation between the blade loads during yaw and non-yaw operations is established for aiding yaw control, and the blade loads and power generation performances of the wind turbine during yaw operation under different wind shear and blade deflection conditions are analyzed for understanding the effects of yaw operation. It is found that the optimal size of the flow field for performing efficient and accurate CFD calculations does exist. The misaligned yaw operation generally tends to decrease the loads acting on the blade. However, the aerodynamic energy captured by the turbine rotor and blade loads during yaw operation is not only dependent on the yaw angle of the rotor but is also affected by wind speed, rotor speed, the pitch angle of the blades, blade deflection, and wind shear. Particularly, it is interestingly found that wind shear can cause undesirable fluctuation of the power, which will challenge the power quality of the wind farm if no measures are taken.

Glehnia littoralis is an important medicinal herb disjunctly distributed at sandy beaches in eastern Asia and western North America. The complete chloroplast genome sequence of G. littoralis is 147,552 bp in length and structurally divided into four distinct regions: two copies of inverted repeat of 18,365 bp separated by a large single copy (LSC) of 93,277 bp and a small single copy (SSC) of 17,545 bp. This paper discusses the increasingly prominent role of ChatGPT in providing feedback and assessment for L2 writing in the digital age. It reviews representative studies that address five research strands about the use of ChatGPT in L2 writing contexts. After a critical evaluation of the existing literature, the author extensively explains four innovative sub-topics on leveraging ChatGPT for L2 writing feedback and assessment, drawing on multiple creative projects undertaken by her research team. These include: 1) ChatGPT-student collaboration in the L2 writing process; 2) ChatGPT-supported teacher feedback; 3) collaborative processing of ChatGPT feedback; and 4) the potential of ChatGPT for L2 writing assessment. This paper also addresses the importance of teacher training on the use of GenAI in language and writing instruction.

The fatty acid elongase 1 (FAE1) gene catalyzes the initial condensation step in the elongation pathway of VLCFA (very long chain fatty acid) biosynthesis and is thus a key gene in erucic acid biosynthesis. Based on a worldwide collection of 62 accessions representing 14 tribes, 31 genera, 51 species, 4 subspecies and 7 varieties, we conducted a phylogenetic reconstruction and correlation analysis between genetic variations in the FAE1 gene and the erucic acid trait, attempting to gain insight into the evolutionary patterns and the correlations between genetic variations in FAE1 and trait variations. As multimodal texts become ubiquitous in the digital age, analyzing how writers interact with readers via visuals is becoming increasingly important. Enlightened by Kress and van Leeuwen's (1996, 2001) semiotic work, D'Angelo (2010) extended Hyland's (2000) text-based metadiscourse model and proposed a new framework for analyzing the visual metadiscourse of multimodal texts. We used D'Angelo's (2016) visual metadiscourse framework to evaluate the comprehensibility and organization of the EFL students' infographics in this study. Two research questions guided our inquiry: (1) How do EFL students use diverse visual metadiscourse resources in their infographic posters? (2) In what ways, if any, does the use of visual interactive resources relate to the assessed quality of the infographics? We collected a total of 122 infographic posters and coded 5 categories of interactive resources for each poster (i.e., information value, framing, connective elements, graphic elements, and fonts), and then carried out frequency counts for each category and subcategories, taking all the posters into account. Moreover, we graded the EFL students' infographics according to a multi-traits grading rubric, and then conducted multiple regression analyses of their use of visual interactive resources and the scores for their infographics in the aspect of visual effects. The results show that the students used diverse visual interactive resources to guide their audience through the text. The assessed quality of the infographics was found to be significantly correlated with the use of graphic elements and fonts. This study sheds new light on digital multimodal pedagogy, particularly on the role of visual metadiscourse in teaching and assessing multimodal texts in language/writing classrooms.
