Background: Malnutrition and lack of micronutrients are seen in many patients with ulcerative colitis (UC). Considering that vitamins play an important role in preventing or protecting against inflammation at the metabolic, physiological, and pathological levels, this research was aimed to investigate the effect of a number of vitamin B groups in improving UC. Methods: Experimental colitis was induced by rectal administration of acetic acid (3%) in male Wistar rats, and mega doses of thiamine (20 mg/kg), riboflavin (15 mg/kg), pyridoxine (30 mg/kg), and cyanocobalamin (250 µg/kg) alone or in combination were administered intraperitoneally for 5 days. In another group, normal dose (1/10 of the above-mentioned doses) of four vitamins in combination was administered for 15 days (started 10 days before colitis induction). Colon tissues were weighted and evaluated in terms of macroscopic, microscopic, and biochemical markers. Results: Normal dose of four vitamins in combination form and mega dose of thiamine caused a significant increase in the body weight of animals. All treatments except for pyridoxine (mega dose) diminished ulceration index, total colitis index, and colon weight compared to the control group. Myeloperoxidase (MPO) activity and malondialdehyde (MDA) levels decreased significantly in all treatment groups. Conclusions: The anti-inflammatory and anti-ulcerative properties of selected B vitamins improved experimental colitis regardless of the dosage and duration of treatment. Despite its beneficial effect on biochemical markers, pyridoxine had the least effect in reducing the pathological features of colitis. Background and purpose: The new plan of using molecular targeted agents in combination with cytotoxic drugs may represent a promising strategy to increase the efficacy of chemotherapy. Hence, we examined whether α2β1 integrin-specific inhibitor, BTT-3033, could modulate the susceptibility of OVCAR3 and SKOV3 ovarian cancer cells to paclitaxel (PTX). Experimental approach: Ovarian cancer cell lines were treated with BTT-3033 and different concentrations of PTX. To determine the mechanisms involved in the PTX/BTT-3033 combination-induced cell death, cell viability, apoptosis, reactive oxygen species (ROS) production, mitochondrial membrane potential (MMP), and caspase-3 activity were evaluated. Findings/Results: Both BTT-3033 (≥ 1 µM) and PTX (≥ 0.01 µM) suppressed the proliferation of OVCAR3 and SKOV3 cells in a concentration-related manner. Pretreatment with BTT-3033 (1 µM), followed by PTX-induced synergistic antiproliferative effects, decreased the IC50 values of PTX from 0.45 to 0.03 µM in OVCAR3 and 0.35 to 0.02 µM in SKOV3 cells. All of the coefficients of drug interaction for various PTX and BTT-3033 combinations were found to be less than 1. Moreover, PTX/BTT-3033 combination induced more apoptotic cells (from 4.2% to 87.0% in OVCAR3 and 2.4% to 88.5% in SKOV3) than PTX alone. Combination therapy also decreased MMP and increased the caspase-3 activity. Additionally, we found that the PTX/BTT-3033 combination enhanced ROS production in OVCAR3 and SKOV3 cells. Conclusion and implications: BTT-3033 has demonstrated the ability to enhance the susceptibility of ovarian cancer cells to PTX by inducing MMP loss, ROS production, and mitochondrial apoptosis, therefore this combination therapy might represent a promising strategy for ovarian cancer treatment. Background and purpose: Tamarindus indica L. which has anti-inflammatory, radical scavenging, and ulcer healing effects can be useful for the alleviation of inflammatory bowel disease (IBD). Therefore, the effects of T. indica fruit pulp (TIPE) and seed extracts (TISE) were investigated on experimental colitis. Experimental approach: TIPE and TISE (125, 250, and 500 mg/kg) were made by maceration (ethanol/water: 80/30) and administered to male Wistar rats with acetic acid-induced colitis. Prednisolone (4 mg/kg) and mesalazine (100 mg/kg) were used as reference drugs. The colon tissues were examined for macroscopic and pathologic parameters and myeloperoxidase (MPO) and malondialdehyde (MDA) values. Findings/Results: The total phenols were 45.7 ± 1.1 and 453.0 ± 3.3 mg/g in terms of gallic acid for TIPE and TISE, respectively. Both of the extracts significantly improved most of the investigated parameters including body weight loss, the weight of colons, indices of ulcers, and total colitis. MPO activity and MDA in the treatment groups (except for TIPE at 125 mg/Kg) significantly decreased compared to the control. Conclusion and implications: Both TIPE and TISE were effective in the treatment of colitis however it seems that the effective ingredients were more concentrated in seeds rather than pulp extract so the highest dose of seed extract had a competitive effect with reference drugs. Introduction: Eryngium billardieri has been demonstrated in previous studies to possess anti-inflammatory, antioxidant, and wound-healing properties. Colitis, an inflammatory bowel disease with unknown causes, often leads to numerous side effects associated with current medications. Therefore, this study aimed to investigate the anti-ulcerative potential of E. billardieri extracts in experimental colitis. Methods: The hydroalcoholic extract of E. billardieri and its aqueous and ethyl acetate partitions were prepared using the maceration method, and the polyphenol content was determined for each extract. Male Wistar rats with acetic acid-induced colitis were orally administered three different doses (50, 100, 200 mg/kg) of the extract and each partition for 5 consecutive days. On the sixth day, the rats’ colons were removed and analyzed for macroscopic parameters (ulcer index), microscopic parameters (total colitis index), as well as inflammatory and oxidative stress markers, including myeloperoxidase and malondialdehyde, respectively. Results: The total phenol content for the dry extract and aqueous and ethyl acetate partitions were 6.51, 4.15, and 8.59 mg gallic acid equivalent/g, respectively. The hydroalcoholic extract and ethyl acetate partition at all three examined doses were able to significantly alleviate most parameters related to colitis. However, the aqueous partition did not improve most of the colitis features except for the tissue level of malondialdehyde. Conclusion: The study concludes that the total extract of E. billardieri, as well as the ethyl acetate partition, exhibited anti-colitis properties in a dose-related manner. It is suggested that the effective substances responsible for these properties are non-polar compounds that are not extracted by aqueous partitioning. Bakground: Ulcerative colitis (UC) is an inflammatory bowel disease that can be treated with many medications but they have various side effects and low cure rate. So, the need for finding novel drugs with better healing characters and less toxicity would be mandatory. Achillea millefolium (A. millefolium, Yarrow) has been traditionally used to treat bleeding, ulcers, wounds, liver, and bile disorders, and recently it has been shown to have anti-ulcer, analgesic, anti-inflammatory, antioxidant, and appetizing effects that make it as a good candidate for UC. Methods: UC was induced with intra-rectal instillation of acetic acid. A. millefolium hydroalcoholic extract (AMHE, 200, 400, and 600 mg/kg/day) and essential oil (AMEO, 62.5, 125, and 250 µl/kg/day) were given to six groups of male Wistar rats for 5 days. Dexamethasone (1 mg/kg/day, intra-peritoneal) and mesalazine (100 mg/kg/day, orally) were used as reference drugs. Colon tissue specimens were separated for assessing macroscopic, pathologic, and biochemical markers. Results: For AMHE, 77.2 mg/g equivalent to gallic acid was obtained for total phenols. Main assessed markers, including ulcer index, total colitis index, colon weight/length ratio, rats' weight gain, and malondialdehyde levels were significantly improved in AMHE (400 and 600 mg/kg/day) and AMEO (125 and 250 µl/kg/day) groups compared to controls. Myeloperoxidase activity was only attenuated in AMHE groups significantly. Conclusions: Both AMHE and AMEO were effective in healing experimental colitis. It seems antioxidant, anti-inflammatory, and anti-ulcer activities of Yarrow are responsible for these beneficial effects. Further studies are warranted to elucidate the exact mechanisms involved. Background and purpose: Inflammatory bowel disease (IBD) is a chronic and multifactorial disease with unknown etiology and a decisive cure. Salvia officinalis (sage) which has anti-inflammatory, anti-oxidative, and ulcer healing properties can be useful for the treatment of IBD. Therefore, the effect of S. officinalis ethanolic extract (SOEE) and methanolic partition (SOMP) was investigated on acetic acid-induced ulcerative colitis. Experimental approach: Male Wistar rats (180-220 g) were used. SOEE (30, 60, and 120 mg/kg) and SOMP (50, 100, and 150 mg/kg) were prepared through maceration method. Prepared extracts, dexamethasone (1 mg/kg, i.p.), and mesalamine (100 mg/kg) as reference drugs and normal saline as control were administered by gavage, 2 h before colitis induction and preserved for four further days to animals. The colon tissues were examined for macroscopic and pathologic parameters and myeloperoxidase (MPO) and malondialdehyde (MDA) levels. Findings/Results: SOEE (60 and 120 mg/kg) and SOMP at all doses alleviated colitis severity and indices both in macroscopic and microscopic views. MDA and MPO activities were also significantly declined in the extracts-treated groups compared to the controls. The lowest dose of SOEE couldn't meaningfully reduce any of the parameters compared to the control group. Conclusion and implications: Both extracts of S. officinalis exerted anti-colitis effects in rats, though methanolic partition was more effective, especially at the highest dose. It seems S. officinalis could exert protection against oxidative stress and inflammatory mediators in colitis tissue.
