Yellowfin tuna (Thunnus albacares) is popular with consumers due to its delicious meat and high nutritional value. It is a globally recognized high-end marine economic fish. With the advancement of fishing equipment and the increase in fishing efforts, the amount of tuna resources has decreased significantly. To make up for the insufficient supply of tuna in the market and protect wild populations, it is essential to carry out research on artificial aquaculture technology for tuna and to gradually establish full-cycle cultures of tuna. In order to study the differences in the nutritional components and quality of yellowfin tuna muscles of different sizes, three different-sized yellowfin tuna, J1 (4.2±1.2) kg, J2 (22.5± 2.5) kg, and J3 (50.8±3.9) kg, were used as the research subjects, and conventional biochemical analysis methods were used to compare and analyze the proximate compositions, amino acids, fatty acids, and mineral elements of tuna muscle. The results showed that (1) the moisture level of the J1 group was significantly higher than that of the J2 and J3 groups (PJ2>J1. The content of nonessential amino acids was J3>J2>J1 (P0.05). The fatty acids in yellowfin tuna muscle were mainly PUFAs, and the content of DHA accounted for 37.46%~39.18% of the total fatty acid content. Abstract Background Intracerebral hemorrhage (ICH) is a common stroke type with high morbidity and mortality. There are mainly three surgical methods for treating ICH. Unfortunately, thus far, no specific surgical method has been proven to be the most effective. We carried out this study to investigate whether minimally invasive surgeries with endoscopic surgery or stereotactic aspiration (frameless navigated aspiration) will improve functional outcomes in patients with supratentorial ICH compared with small-bone flap craniotomy. Methods In this parallel-group multicenter randomized controlled trial conducted at 16 centers, patients with supratentorial hypertensive ICH were randomized to receive endoscopic surgery, stereotactic aspiration, or craniotomy at a 1:1:1 ratio from July 2016 to June 2022. The follow-up duration was 6 months. Patients were randomized to receive endoscopic evacuation, stereotactic aspiration, or small-bone flap craniotomy. The primary outcome was favorable functional outcome, defined as the proportion of patients who achieved a modified Rankin scale (mRS) score of 0–2 at the 6-month follow-up. Results A total of 733 patients were randomly allocated to three groups: 243 to the endoscopy group, 247 to the aspiration group, and 243 to the craniotomy group. Finally, 721 patients (239 in the endoscopy group, 246 in the aspiration group, and 236 in the craniotomy group) received treatment and were included in the intention-to-treat analysis. Primary efficacy analysis revealed that 73 of 219 (33.3%) in the endoscopy group, 72 of 220 (32.7%) in the aspiration group, and 47 of 212 (22.2%) in the craniotomy group achieved favorable functional outcome at the 6-month follow-up (P = .017). We got similar results in subgroup analysis of deep hemorrhages, while in lobar hemorrhages the prognostic outcome was similar among three groups. Old age, deep hematoma location, large hematoma volume, low preoperative GCS score, craniotomy, and intracranial infection were associated with greater odds of unfavorable outcomes. The mean hospitalization expenses were ¥92,420 in the endoscopy group, ¥77,351 in the aspiration group, and ¥100,947 in the craniotomy group (P = .000). Conclusions Compared with small bone flap craniotomy, endoscopic surgery and stereotactic aspiration improved the long-term outcome of hypertensive ICH, especially deep hemorrhages. Abstract Near-surface atmospheric moisture is a key environmental and hydro-climatic variable that has significant implications for the natural and human systems. However, high-resolution moisture data are severely lacking for fine-scale studies. Here, we develop the first 1 km high spatial resolution dataset of monthly moisture index collection in China (HiMIC-Monthly) over a long period of 2003~2020. HiMIC-Monthly is generated by the light gradient boosting machine algorithm (LightGBM) based on observations at 2,419 weather stations and multiple covariates, including land surface temperature, vapor pressure, land cover, impervious surface proportion, population density, and topography. This collection includes six commonly used moisture indices, enabling fine-scale assessment of moisture conditions from different perspectives. Results show that the HiMIC-Monthly dataset has a good performance, with R2 values for all six moisture indices exceeding 0.96 and root mean square error and mean absolute error values within a reasonable range. The dataset exhibits high consistency with in situ observations over various spatial and temporal regimes, demonstrating broad applicability and strong reliability.

Abstract The intensification of global heat wave events is seriously affecting residents' emotional health. Based on social media big data, our research explored the spatial pattern of residents' sentiments during heat waves (SDHW). Besides, their association with urban functional areas (UFAs) was analyzed using the Apriori algorithm of association rule mining. It was found that SDHW in Beijing were characterized by obvious spatial clustering, with hot spots predominately dispersed in urban areas and far suburbs, and cold spots mainly clustered in near suburbs. As for the associations with urban function areas, green space and park areas had significant effects on the positive sentiment in the study area, while a higher percentage of industrial areas had a greater impact on negative SDHW. When it comes to combined UFAs, our results revealed that the green space and park area combined with other functional areas was more closely related to positive SDHW, indicating the significance of promoting positive sentiment. Subdistricts with a lower percentage of residential and traffic areas may have a more negative sentiment. There were two main combined UFAs that have greater impacts on SDHW: the combination of residential and industrial areas, and the combination of residential and public areas. This study contributes to the understanding of improving community planning and governance when heat waves increase, building healthy cities, and enhancing urban emergency management.

Abstract Surface, aircraft, and satellite measurements indicate pervasive early cold season (Augut–September) CO2 emissions across Arctic regions, consistent with increased ecosystem metabolism in plants and soils. A key remaining question is whether cold season sources will become large enough to permanently shift the Arctic into a net carbon source. Polar orbiting GHG satellites provide robust estimation of regional carbon budgets but lack sufficient spatial coverage and repeat frequency to track sink‐to‐source transitions in the early cold season. Mission concepts such as the Arctic Observing Mission (AOM) advocate for flying imaging spectrometers in highly elliptical orbits (HEO) over the Arctic to address sampling limitations. We perform retrieval and flux inversion simulation experiments using the AURORA mission concept, leveraging a Panchromatic imaging Fourier Transform Spectrometer (PanFTS) in HEO. Our simulations demonstrate the potential benefits of increased CO2 sampling for detecting emissions during the early cold season.

Abstract Although it is known that the frequency and intensity of heatwaves are affected by the El Niño–Southern Oscillation (ENSO), unknown are the ENSO modulations on the moving properties (e.g., moving distance and speed) of spatiotemporally contiguous heatwaves. Here, we investigate the relationship between ENSO and the moving patterns of contiguous heatwaves. We show that contiguous heatwaves are likely more frequent, more persistent, and longer‐traveling, but slower‐moving during El Niño than La Niña episodes. The differences in the tropical contiguous heatwaves between El Niño and La Niña are influenced by persistent high‐pressure anomalies. During the following summers, El Niño can induce anomalous atmospheric circulation characterized by an intensified subsidence over the western North Pacific and ascending motion over the tropical Indian and Pacific Oceans. These features provide favorable conditions for the occurrence and maintenance of contiguous heatwaves.

Nature-based Solutions (NbS) is a concept aims to address challenges related to human well-being, including human health, socio-economic development, and ecological security on both national and global scales. This concept was proposed against the backdrop of global climate change, ecological crises affecting humanity, and threats to sustainable development and it has been increasingly recognized and applied internationally over the past decade. The principle of NbS is reciprocity between humans and ecosystems, which is completely in line with the goals, systems, practices, and concepts guiding China's construction of ecological civilization. This paper clarifies concepts, evaluation criteria, and comprehensive benefits related to NbS, to trace the connection between NbS and both traditional and contemporary ecological civilization principles in China. By drawing valuable experience from NbS practices in China, it will focus on proposing policy and implementation suggestions to address challenges currently faced in NbS applications in China. This research will promote the research and practice of nature-based solutions in China. Meanwhile, provide novel perspectives and technologies for urban ecological design as well as sustainable development.

Abstract For 800 Gbps/λ and beyond optical transmission systems, frequency-dependent impairments (FDI) degrade coherent optical transceiver (CO-TRx) performance severely. Calibration and compensation of such FDI in factories includes amplitude/phase frequency response (AFR/PFR), skew, and ripple for both transmitters(Tx) and receivers (Rx). However, due to the polarization rotation and phase rotation effects in optical link, the separation and extraction of FDI from different polarization or I/Q tributaries is challenging. Here we report a FDI calibration method based on orthogonal separation scheme and frequency domain analysis. The proposal can simultaneously characterize and separate the Tx/Rx sides FDI including AFR, PFR, and time skew of four tributaries. Finally, the effectiveness is demonstrated by transmitting a 96 GBuad Nyquist-16QAM signal on a 64 GBaud-class CO-TRx.

AbstractContext Dolichos trilobus Linn (Leguminosae) is often used in Yi ethnic medicine to treat pain, fracture, and rheumatism.Objective To explore the therapeutic potential of doliroside B (DB) from D. trilobus and its disodium salt (DBDS) and the underlying mechanism in pain.Materials and methods In the writhing test, Kunming mice were orally treated with DB and DBDS at doses of 0.31, 0.62, 1.25, 2.5, and 5 mg/kg. Vehicle, morphine, indomethacin, and acetylsalicylic acid were used as negative and positive control on the nociception-induced models, respectively. In the hot plate test, mice were orally treated with DB and DBDS at doses of 2.5, 5, 10, and 20 mg/kg. In the formalin test, mice were orally treated with DB and DBDS at doses of 2.5, 5, 10, and 20 mg/kg. In the meanwhile, lipopolysaccharide-induced inflammatory model in RAW264.7 macrophages was adopted to study the mechanism of pain alleviation for DBDS.Results DBDS (5 mg/kg) inhibited the writhing number by 80.2%, which exhibited the highest antinociceptive activity in pain models. DBDS could selectively inhibite the activity of COX-1. Meanwhile, it also reduced the production of NO, iNOS, and IL-6 by 55.8%, 69.0%, and 49.9% inhibition, respectively. It was found that DBDS also positively modulated the function of GABAA1 receptor.Discussion and conclusions DBDS displayed antinociceptive activity by acting on both the peripheral and central nervous systems, which may act on multitargets. Further work is warranted for developing DBDS into a potential drug for the treatment of pain.

Ratiometric DNA biosensors, which utilize DNA as the recognition element and integrate dual signals from diverse sensing platforms including fluorescence, electrochemistry, and surface-enhanced Raman scattering (SERS), have demonstrated remarkable proficiency in detecting a wide range of targets. In this review, we showcase the significant progress achieved by ratiometric biosensors. Firstly, ratiometric biosensors have made notable advancements in analyzing real samples. These include the analysis of pH values near cancer cells, quantification of miRNA in human cell lysates, detection of human telomerase RNA in cell extracts, and performing DNA logic-gated in situ bioimaging on cell membranes. Secondly, excellent sensitivity has been attained through the utilization of effective amplification methods such as RCA, HCR, and CHA, among others. Thirdly, the construction of stable reference signals has resulted in significantly improved precision for ratiometric biosensors. This breakthrough has overcome matrix effects, enabling reliable detection in real samples with high selectivity. This review provides a comprehensive overview of recent advancements in strategies employed by ratiometric DNA biosensors. We present three types of biosensors based on distinct sensing platforms: fluorescent, electrochemical, and SERS biosensors. Additionally, we discuss future directions and primary challenges in the development of ratiometric DNA biosensors.

Abstract Precipitation exhibits a pronounced seasonal cycle, of which the phase and amplitude are closely associated with water resource management. While previous studies suggested an emerged delaying phase in the past decades, whether the amplified amplitude has emerged is controversial. Using multiple observational data sets and climate simulations, here we show that the amplification of precipitation annual cycle has emerged in most global land areas since the 1980s, especially in the tropics. These amplifications are mainly driven by anthropogenic emissions, and will be further intensified by 17.6% in the future (2081–2100) under high emission scenario (Shared Socioeconomic Pathways, SSP585), and limited to 7.2% under SSP126 scenario, relative to the historical period (1982–2014). Precipitation seasonality will be amplified by 4.2% for each 1°C of global warming, which is seen in all emission scenarios. The mitigation of lower emissions is helpful for alleviating the amplification of precipitation seasonality in a warming world.

The increasing incidence and mortality of prostate cancer worldwide significantly impact the life span of male patients, emphasizing the urgency of understanding its pathogenic mechanism and associated molecular changes that regulate tumor progression for effective prevention and treatment. RNA modification, an important post-transcriptional regulatory process, profoundly influences tumor cell growth and metabolism, shaping cell fate. Over 170 RNA modification methods are known, with prominent research focusing on N6-methyladenosine, N7-methylguanosine, N1-methyladenosine, 5-methylcytidine, pseudouridine, and N4-acetylcytidine modifications. These alterations intricately regulate coding and non-coding RNA post-transcriptionally, affecting the stability of RNA and protein expression levels. This article delves into the latest advancements and challenges associated with various RNA modifications in prostate cancer tumor cells, tumor microenvironment, and core signaling molecule androgen receptors. It aims to provide new research targets and avenues for molecular diagnosis, treatment strategies, and improvement of the prognosis in prostate cancer.

The formed surface quality of metallic sealing ring of aero-engine affects the aircraft service performance directly. However, existing inspection methods, such as the final destructive inspection and the line laser scanner section profile measurement, only evaluate the formed quality from a 2-D view, that is, single or multiple radial formed section profiles. The lack of geometric information of 3-D surface is not conducive to the comprehensive monitoring of forming quality and process planning. Therefore, based on the line laser scanners, this paper mainly proposes a vibration errors compensation method based on self-feature registration. Aiming at the problem of rigid transformation of the measurement profile caused by random vibration during the rotary motion of metallic sealing ring, the feature of measurement profile in stationary scene (MPSS), that is, the medial axis, is used as the reference for the correct pose of measurement profile. The principle of finding the correct pose of measurement profile in rotary motion scene (MPRMS) is to minimize the distance between the medial axes. Next, based on the rotary motion information of metallic sealing ring and the geometric information of measurement system, a 3-D reconstruction matrix is built, so as to convert each measurement profile to the base coordinate system in turn, and finally a 3-D dynamic measurement method for the metallic sealing ring forming surface is built. The effectiveness of the proposed method is verified through simulation experiment and real measurement.

Fire blight, caused by the Gram-negative bacterium Erwinia amylovora, poses a substantial threat to pome fruit production worldwide. Despite existing control strategies, a pressing need remains for sustainable and environmentally friendly fire blight management. Myxobacteria, renowned for their predatory behavior and potent enzymes, emerge as a groundbreaking biocontrol approach with significant potential. Here, we report the biocontrol potential of a novel Myxococcus fulvus WCH05, against E. amylovora. Using various in vitro and planta assays, we demonstrated the multifaceted biocontrol abilities of strain WCH05. In plate predation assays, strain WCH05 exhibited not only strong predation against E. amylovora but also broad-spectrum activities against other plant pathogenic bacteria. Pre-treatment with strain WCH05 significantly decreased pear blossom blight incidence in detached inflorescence assays, achieving a controlled efficacy of 76.02% that rivaled the antibiotic streptomycin (79.79%). In greenhouse trials, strain WCH05 effectively reduced the wilting rate and disease index in young pear seedlings, exhibiting both protective (73.68%) and curative (68.66%) control. Further investigation revealed that the biocontrol activity of strain WCH05 relies on both direct contact and extracellular enzyme secretion. While cell extracts lacked inhibitory activity, ammonium sulfate-precipitated secreted proteins displayed potent lytic activity against E. amylovora. Substrate spectrum analysis identified peptidases, lipases, and glycosidases among the secreted enzymes, suggesting their potential roles in pathogen degradation and biocontrol efficacy. This study presents the first evidence of Myxococcus fulvus WCH05 as a biocontrol agent against fire blight. Its potent predatory abilities and enzymatic arsenal highlight its potential for sustainable disease management in pome fruit production. Future research will focus on identifying and characterizing specific lytic enzymes and optimizing strain WCH05 application strategies for field efficacy.

Software Cost Estimation (SCE) is one of the research priorities and challenges in the construction of cyber-physical-social systems (CPSSs). In CPSS, it is urge to process environmental and social information accurately and use it to guide social practice. Thus, in response to the problems of low prediction accuracy, poor robustness, and poor interpretability in SCE, this paper proposes a SCE model based on Autoencoder and Random Forest. First, preprocess the project data, remove outliers, and build regression trees to fill in missing attributes in the data. Second, construct a Autoencoder to reduce the dimensionality of factors that affect software cost. Subsequently, the performance of the model was trained and validated using the XGBoost framework on three datasets: COCOMO81, Albrecht, and Desharnais, and compared with common cost prediction models. The experimental results show that the MMRE, MdMRE, and PRED (0.25) values of the proposed model on the COCOMO81 dataset reached 0.21, 0.16, and 0.71, respectively. Compared with other models, the proposed model achieved significant improvements in accuracy and robustness.

Satellite-derived bathymetry plays a significant role in characterizing river systems, furnishing invaluable insights for applications such as flood risk management. In this paper, we introduce a bathymetry inversion framework, namely reflectance transformation - convolutional neural network (RT-CNN), geared towards long river segments, exemplified by a 210-kilometer stretch of the Bei River's in South China. The framework hinges on a neural network driven by a reflectance transformation model, an optical construct that harnesses radiance data from visible, near-infrared, and shortwave infrared bands. This modified input undergoes spatial–temporal fusion and convolutional neural network processing, culminating in high-resolution (10-meter) bathymetric estimations. The RT-CNN approach adeptly blends Landsat and Sentinel imagery with a substantial dataset of over 18,000 SONAR measurements, integrating convolutional neural network regressions for bathymetric inference. Comparative analysis against established machine learning methodologies, including random forest, gradient boosting decision tree, support vector machines, and multiple linear regression, underscores the supremacy of the proposed RT-CNN framework. Despite the Bei River's changing hydrodynamic attributes along its length, the RT-CNN delivers accurate bathymetric estimates. Specifically, across 2012, 2016, and 2019, the convolutional neural network consistently outperforms other algorithms, achieving an accuracy range of 0.43 to 1.63 m (with a 18% accuracy improvement compared to the existing methods) within an approximate elevation difference of 60 m from upstream to downstream. Temporal trend analyses indicate that heightened precipitation volumes and vegetation loss correlate with downstream shallowing in the Bei River during recent years.

Abstract Aims Cardiomyocyte apoptosis is an important factor leading to the occurrence and development of heart failure (HF), which is associated with high mortality of patients with cardiovascular diseases. This study aims to investigate the underlying mechanisms of HF in terms of expression and regulation patterns using bioinformatics and experimental validation. Methods and results Two HF datasets were collected: a dataset GSE112056 downloaded from the GEO database (including mRNA and miRNA sequencing data) and another is the laboratory‐owned mRNA dataset. Differential mRNAs and miRNAs in the two datasets were screened using the raw Bayesian approach method. Gene Ontology was used to perform functional enrichment analysis of the differential mRNAs and co‐expression network analysis of the differential mRNAs, combined with nuclear transcription factors in the differential miRNAs and mRNAs for target gene prediction. A HF cell model was constructed using mouse cardiomyocytes (HL‐1), and the role and mechanism of miRNA‐103‐3p‐Hlf (hepatic leukaemia factor) in the process of HF was verified by cell transfection, luciferase reporter gene, WB, and qPCR. We found that Hlf gene expression was decreased in the HF model group and strongly correlated with FYCO1 (FYVE and coiled‐coil domain‐containing protein 1) gene, a phenomenon enriched in apoptotic autophagy‐related pathways. MiR‐103‐3p expression was up‐regulated in the HF model group, and its targeting correlation with Hlf was confirmed by luciferase activity assay. Abstract Human-perceived temperature (HPT) describes the joint effects of multiple climatic factors such as temperature and humidity. Extreme HPT events may reduce labor capacity and cause thermal discomfort and even mortality. These events are becoming more frequent and more intense under global warming, posing severe threats to human and natural systems worldwide, particularly in populated areas with intensive human activities, e.g., the North China Plain (NCP). Therefore, a fine-scale HPT dataset in both spatial and temporal dimensions is urgently needed. Here we construct a daily high-resolution (~1 km) human thermal index collection over NCP from 2003 to 2020 (HiTIC-NCP). This dataset contains 12 HPT indices and has high accuracy with averaged determination coefficient, mean absolute error, and root mean squared error of 0.987, 0.970 °C, and 1.292 °C, respectively. Moreover, it exhibits high spatiotemporal consistency with ground-level observations. The dataset provides a reference for human thermal environment and could facilitate studies such as natural hazards, regional climate change, and urban planning.

Abstract With the accelerated operation of subway networks, the increasing number of subway transfer stations results in inefficient passenger travel. The target of this paper is to solve the research question of how to reduce transfer waiting time (TWTT) for heterogeneous passengers. The key problem is to determine the optimal concerted train timetable considering the transfer walking time (TWKT) of the passengers. On the basis of field survey data, the regression method was used to establish a TWKT prediction model for general passengers (G) and vulnerable passengers (V), including the elderly, passengers traveling with children, and those carrying large luggage. Afterward, a two-objective integer programming model was formulated to minimize the subway operating costs and TWTT for each group, in which V is given the priority weight to ensure social equity. The headway, loading capacity, and TWKT of heterogeneous passengers were set as optimization model constraints. A genetic algorithm (GA) was designed to find the optimal solution. A case study in which the Beijing Jianguomen Station was selected as the key transfer station was conducted to verify the performance of the proposed model. Key results show that the total TWTT for V and G can be reduced by 18.6% and 27.2%, respectively, with one train saved. Results of the parameter sensitivity analysis reveal the interconnection between the operating cost, heterogeneous passenger proportion, and transfer time. The proposed model can be used for improving transfer efficiency for passengers while considering the enterprise operating costs.

Sphingolipids play an important role in cotton fiber development, but the regulatory mechanism is largely unclear. We found that serine palmitoyltransferase (SPT) enzyme inhibitors, myriocin and sphingosine (dihydrosphingosine (DHS) and phytosphingosine (PHS)), affected early fiber elongation in cotton, and we performed a sphingolipidomic and transcriptomic analysis of control and PHS-treated fibers. Myriocin inhibited fiber elongation, while DHS and PHS promoted it in a dose–effect manner. Using liquid chromatography–tandem mass spectrometry (LC–MS/MS), we found that contents of 22 sphingolipids in the PHS-treated fibers for 10 days were changed, of which the contents of 4 sphingolipids increased and 18 sphingolipids decreased. The transcriptome analysis identified 432 differentially expressed genes (238 up-regulated and 194 down-regulated) in the PHS-treated fibers. Among them, the phenylpropanoid biosynthesis pathway is the most significant enrichment. The expression levels of transcription factors such as MYB, ERF, LBD, and bHLH in the fibers also changed, and most of MYB and ERF were up-regulated. Auxin-related genes IAA, GH3 and BIG GRAIN 1 were up-regulated, while ABPs were down-regulated, and the contents of 3 auxin metabolites were decreased. Our results provide important sphingolipid metabolites and regulatory pathways that influence fiber elongation.

At present, the main methods of breeding Dendrobium officinale (D. officinale) include introduction and domestication, selective breeding, hybrid breeding, and mutation breeding. In the process, traditional methods of field investigation of agronomic traits are often used to select resistant varieties. Although these breeding methods are effective, they have a certain degree of subjectivity and empiricism, and the breeding cycle is long. Electrophysiological instruments were used in this experiment to test the material transport and metabolic capacity of D. officinale in a karst forest epiphytic environment and to quickly evaluate the suitability of different strains of D. officinale in a drought environment. These instruments detected the data on electrophysiological information of leaves of different strains of D. officinale under long-term drought conditions, providing immediate access to the inherent electrophysiological information of the leaves of these strains. Based on the electrophysiological parameters of D. officinale leaves as defined by the inherent electrophysiological information of plants, the water metabolism, nutrient transport, and metabolic capacity in different leaves were evaluated. The key electrophysiological indexes were verified by combining the results of chlorophyll fluorescence and chlorophyll content. The results indicate the following: (1) Parameters defined based on electrophysiological information effectively characterized the differences in intracellular water utilization, the nutrient transport status, and the metabolic capacity of different D. officinale strains. (2) The intrinsic physiological resistance, intrinsic physiological reactance, active nutrient transport capacity, and passive nutrient transport capacity were closely related to the growth status and chlorophyll function of D. officinale leaves. These electrophysiological parameters could serve as critical indicators for evaluating the drought resistance of D. officinale. (3) Under severe drought stress, strain LH1 exhibited less leaf damage, adequate water/nutrient supply, vigorous life activities, and excellent drought resistance. We found that strain LH1 demonstrates better adaptation to the arid environment of karst forest lands. The electrophysiological detection method employed in this study offers a new technique for screening wild-cultivated D. officinale resistance strains. The results indicate that the real-time online leaf electrophysiological information measured by the method in this study can characterize the energy and material metabolism of crops, greatly improving the efficiency of crop-variety selection and reducing costs. These conclusions can be used to obtain real-time information on the transmission of water and nutrients within plant cells and can provide theoretical support for studying the adaptation mechanisms of crops to adverse environments.

In mammals, specificity protein 1 (SP1) was the first Cys2-His2 zinc finger transcription factor to be isolated within the specificity protein and Krüppel-like factor (Sp/KLF) gene family. SP1 regulates gene expression by binding to Guanine–Cytosine (GC)-rich sequences on promoter regions of target genes, affecting various cellular processes. Additionally, the activity of SP1 is markedly influenced by posttranslational modifications, such as phosphorylation, acetylation, glycosylation, and proteolysis. SP1 is implicated in the regulation of apoptosis, cell hypertrophy, inflammation, oxidative stress, lipid metabolism, plaque stabilization, endothelial dysfunction, fibrosis, calcification, and other pathological processes. These processes impact the onset and progression of numerous cardiovascular disorders, including coronary heart disease, ischemia-reperfusion injury, cardiomyopathy, arrhythmia, and vascular disease. SP1 emerges as a potential target for the prevention and therapeutic intervention of cardiac ailments. In this review, we delve into the biological functions, pathophysiological mechanisms, and potential clinical implications of SP1 in cardiac pathology to offer valuable insights into the regulatory functions of SP1 in heart diseases and unveil novel avenues for the prevention and treatment of cardiovascular conditions.

Driven by the demands of ocean exploration, an increasing number of manta ray-inspired robots have been designed and manufactured, primarily utilizing flexible skeletons combined with motor-driven mechanisms. However, the mechanical analysis of these designs remains underdeveloped, often relying on simplistic imitation of biological prototypes and typically neglecting the vertical motion induced by pectoral fin flapping. This paper presents a fluid–structure interaction analysis framework that couples rigid body motion with elastic deformation using flexible multibody dynamics and the vortex particle method. An implicit iterative algorithm with Aitken relaxation is employed to address added-mass instability, and the framework has been validated against experimental data. An analysis of a representative manta-bot model shows that self-induced vertical undulations reduce the thrust coefficient by approximately 40% compared to fixed vertical degrees of freedom, while slightly improving overall propulsive efficiency. The study also highlights the critical role of mass distribution in manta-bots, noting that excessive focus on complex pectoral fin movements and large fin mass can significantly reduce thrust by increasing vertical displacement, ultimately proving counterproductive.

BackgroundIt has been discovered that Janus kinase 2 (JAK2) exon12 mutations lead to the polycythemia vera (PV) phenotype, while somatic mutations of calreticulin (CALR) are associated with essential thrombocythemia (ET) or primary myelofibrosis. In this article, we report a case of ET with coexistence of JAK2 exon12 and CALR mutations. The objective of this study was to elucidate the pathogenicity mechanism of a JAK2 exon12 mutation (JAK2N533S) and the role of the coexistence of mutations on the hematological phenotype.MethodsWe designed a colony analysis of tumor cells obtained from this patient, and attempted to identify mutant genes using DNA from hair follicles. Mutation impairment prediction and conservative analysis were conducted to predict the mutation impairment and structure of JAK2N533S. In addition, we conducted a functional analysis of JAK2N533S by constructing Ba/F3 cell models.ResultsThree distinct tumor subclones, namely JAK2N533Shet+/CALRtype1het+, JAK2N533Shet+/CALRwt, and JAK2N533Shet+/CALRtype1hom+, were identified from the 17 selected erythroid and 21 selected granulocyte colonies. The analysis of hair follicles yielded positive results for JAK2N533S. According to the bioinformatics analysis, JAK2N533S may exert only a minor effect on protein function. Functional studies showed that JAK2N533S did not have a significant effect on the proliferation of Ba/F3 cells in the absence of interleukin-3 (IL-3), similar to wild-type JAK2. Notably, there were no increased phosphorylation levels of JAK2-downstream signaling proteins, including signal transducer and activator of transcription 3 (STAT3) and STAT5, in Ba/F3 cells harboring the JAK2N533S.ConclusionOur study revealed that the JAK2N533Shet+/CALRtype1het+ subclone was linked to a significant expansion advantage in this patient, indicating that it may contribute to the development of the ET phenotype. We further demonstrated that JAK2N533S, as a noncanonical JAK2 exon12 mutation, is a germline mutation that may not exert an effect on cell proliferation and protein function. These results and the present body of available data imply that certain noncanonical JAK2 mutations are not gain-of-function mutations leading to the development of myeloproliferative neoplasms.

ABSTRACTTimely and accurate wetland information is necessary for wetland resource management. Recent advances in machine learning and remote sensing have facilitated cost-effective monitoring of wetlands. However, reliable methods for fine-grained and rapid wetland mapping are still lacking. To address the issue, a wetland sample set with 20 categories for China was collected based on a sampling strategy that combines automatic sample generation and visual interpretation. Simultaneously, a novel multi-stage method for fine-grained wetland classification was proposed, which integrates pixel-based and object-based strategies using ensemble learning algorithms and multi-source remote sensing data. First, a pixel-based ensemble learning algorithm was implemented to classify five rough wetland categories and six non-wetland categories. Second, an object-based ensemble learning approach was designed to separate the water cover in the pixel-based classification results into eight detailed categories. Third, the merged pixel-based and object-based classification results were refined with knowledge-based post-processing procedures to identify 14 fine-grained wetland categories. Results using the Pixel Information Expert Engine (PIE-Engine) cloud platform proved the effectiveness of the proposed wetland classification method. The overall accuracy, kappa, and weighted F1 reached 87.39%, 82.80%, and 86.02%, respectively. The adopted ensemble learning algorithm yielded better performance than classifiers such as CatBoost, random forest, and XGBoost. The incorporation of spectral, texture, shape, topographic, and geographic features from multi-source data contributed to differentiating wetland categories. According to the relative contribution, spectral indexes (NDVI and NDWI), texture features (sum average and contrast), and topographic features (slope and elevation) were identified as important leading predictors for the first-stage pixel-based classification. Shape features (shape index and compactness) and auxiliary features (geographic location) were crucial predictors for the second-stage object-based classification. Compared with other products, our 10-m wetland mapping results for national wetland reserves were rich in detail and fine in categories. Overall, the constructed sample set and developed classification method show promise in laying a foundation for large-scale wetland mapping. The derived wetland maps can provide support for wetland protection and restoration.

Heatwaves are among the deadliest weather-related disasters. They are not only seen in summer but also may appear in spring and autumn, especially under climate warming. Although the mechanisms of summertime heatwaves have been extensively studied, how they differ from spring and autumn seasons and possibly season-dependent mechanisms of heatwaves are poorly understood. Here we examine the specific processes associated with spring, summer, and autumn heatwaves in southern China during 1961–2020. We show that heatwaves in all three seasons are intensifying in frequency, duration, and intensity, and they exhibit distinctly season-dependent circulation changes. Summer heatwaves are characterized by a warm-core high-pressure structure, while spring and autumn heatwaves exhibit a warm low pattern. Summer heatwaves are mainly triggered by the westward (eastward) extension of the western North Pacific subtropical high (South Asian high), and are accompanied by anomalous lower-level anticyclone and positive pressure anomalies. In contrast, spring and autumn heatwaves coincide with lower-than-normal pressure and anomalous southwesterly over East Asia, indicative of a weakening of the East Asian winter monsoon circulation and the Siberian high. Their developments are linked to the eastward movement of an upper-tropospheric wave-like pattern over the mid-latitudes. It is also found that autumn heatwaves are largely contributed by local preceding soil moisture deficit, in comparison to spring and summer heatwaves. Our findings contribute to a better understanding of the heatwave characteristics in different seasons and provide insights into their mitigation strategies.

Biodiversity loss is one of the major crises facing humanity today, and with none of the biodiversity targets represented by the Aichi Biodiversity Targets being met, there is an urgent need to promote transformative change to halt and reverse the trend of biodiversity loss. Nature-based solutions (NbS) have emerged as an important way to address the global crisis due to its inherent principles involving holistic and systemicness, diversity, stability, sustainability, trade-offs, and compatibility. The relationship between biodiversity and NbS is analyzed from a mechanistic and functional perspective, and the pathway of NbS as a proxy for biodiversity conservation is revealed by restoring of ecosystem complexity and trophic levels to put nature on the path. We here propose the dual intensions of promoting NbS for biodiversity conservation, as (1) the goal aimed at enhancing diversity, stability, and sustainability of ecosystem, and (2) the tool involves use natural processes. Based on the above understanding of the link between NbS and biodiversity, this paper further compares the conceptual connotation of NbS with the consistency of biodiversity conservation, as well as the NbS methods for biodiversity conservation including natural, agricultural and urban area. Practical cases of NbS for biodiversity conservation or synergy of NbS for biodiversity are summarized in align with differentiated typical ecosystems. The multiple benefits of NbS for biodiversity conservation, climate change and sustainable development are discussed. The vision of integrating NbS into strategic planning for biodiversity conservation is presented to provide a reference for promoting the implementation of Kunming-Montreal Global Biodiversity Framework and the mainstreaming of NbS in biodiversity conservation.

Kunming-Montreal Global Biodiversity Framework sets out key actions for biodiversity conservation worldwide in the coming period, including integrating biodiversity and its multiple values into economic and social activities. As a party to the United Nations Convention on Biological Diversity (CBD), China has made unremitting efforts to promote the biodiversity mainstreaming, integrating biodiversity conservation into top-level national decision-making and major strategic planning, and integrating it into the policies, norms and assessment mechanisms of ecological environments, natural resources and other relevant industries. Different functional departments, scientific research institutions, enterprises, social organizations and other organizations have carried out extensive and in-depth researches based on multiple aspects of biodiversity conservation and management. Benchmark the objectives of Kunming-Montreal Global Biodiversity Framework and refer the advanced experience of biodiversity mainstreaming practices in other countries, we discussed and analyzed the conceptual connotation of biodiversity mainstreaming, summarized the practice and stage results of biodiversity mainstreaming in China. Then focusing on different actors including governments, enterprises and the public, we propose the implementation path of promoting biodiversity mainstreaming in an all-round way for China in the new era, including: (1) To import a consistent action framework which is mitigation protection hierarchy with four steps, contains avoidance, mitigation, recovery, and offset; (2) To give play to the leading role of government governance, and to integrate biodiversity conservation into government governance systems and industrial development layout with the help of various planning and policy tools; (3) To support enterprises to take joint actions and internalize ecological and environmental hazards into business operations which can promote sustainable production and consumption; (4) To raise public awareness to promote broad participation in biodiversity conservation, establish a system of public participation in biodiversity conservation through various publicity and education actions, to guide the public to practice a green and low-carbon lifestyle, and to translate into real benefits of biodiversity and climate mitigation and improvement. By taking the above actions, we will strive to integrate biodiversity into the policy mechanisms of government departments at all levels and social production and life practices, so as to provide references for improving biodiversity governance decisions.

Abstract Mountain rivers are characterized by wide grain size distributions and complex bed surface structures, which significantly affect bedload transport. Owing to the lack of a clear understanding of the quantitative influence of the bed surface structure on the bedload transport rate, existing methods for estimating the bedload transport rate in mountain rivers produce large errors. Based on theoretical analysis and flume experiments, this study reveals the influence of bed surface structure on nonuniform bedload transport and proposes a method for estimating bedload transport rate considering the quantitative influence of bed surface structure. The findings of the present study provide theoretical methodological support for predicting the sediment transport and bed evolution in mountain rivers.

Abstract The use of a stent to coil an aneurysm can alter the position of the main blood vessel and affect blood flow within the sac. This study thoroughly examines the impact of stent-induced changes on the risk of MCA aneurysm rupture. The research aims to assess the effects of coiling and vessel deformation on blood flow dynamics by comparing the OSI, WSS, and blood structure of two distinct MCA aneurysms to identify high-risk areas for hemorrhage. Computational fluid dynamics is used to model blood flow. The results indicate that aneurysm deformation does not always decrease the risk of rupture, and coiling is more effective in occluding blood flow than aneurysm deformation.

Abstract Stevia rebaudiana Bertoni is a valuable medicinal plant and an essential source of natural sweetener, steviol glycosides (SGs), with rebaudioside A (RA) being one of the main components of SGs. bHLH transcription factors play a crucial role in plant development and secondary metabolism. In this study, 159 SrbHLH genes were identified from the S. rebaudiana genome, and each gene was named based on its chromosome location. The SrbHLH proteins were then clustered into 18 subfamilies through phylogenetic analysis. The analysis of conserved motifs and gene structure further supported the classification of the SrbHLH family. Chromosomal location and gene duplication events of SrbHLH genes were also studied. Moreover, based on the RNA-Seq data of different tissues of S. rebaudiana, 28 SrbHLHs were co-expressed with structural genes involved in RA biosynthesis. The expression pattern of candidate SrbHLH genes were confirmed by qPCR. Finally, dual luciferase reporter assays (DLAs) and subcellular localization analysis verified SrbHLH22, SrbHLH111, SrbHLH126, SrbHLH142, and SrbHLH152 are critical regulators of RA biosynthesis. This study provides new insights into the function of SrbHLHs in regulating SGs biosynthesis and lays the foundation for future applications of SrbHLH genes in molecular breeding of S. rebaudiana.

Abstract Background Adolescent idiopathic scoliosis (AIS) is characterized by low lean mass without vertebral deformity. The cause-and-effect relationship between scoliosis and paraspinal muscle imbalance has long puzzled researchers. Although FTO has been identified as a susceptibility gene for AIS, its potential role in the asymmetry of paraspinal muscles has not been fully elucidated. Methods We investigated the role of Fto in murine myoblast proliferation, migration, and myogenic differentiation. We examined its precise regulatory influence on murine muscle fiber remodeling in vitro and in vivo. We identified the downstream target gene of Fto by screening key regulators of murine muscle fiber remodeling and identified its m6A reader. Deep paraspinal muscle samples were obtained from the concave and convex sides of AIS patients with or without Schroth exercises, and congenital scoliosis served as a control group. We compared the content of type I fibers, expression patterns of fast- and slow-type genes, and levels of FTO expression. Results FTO contributed to maintain the formation of murine slow-twitch fibers both in vitro and in vivo. These effects were mediated by the demethylation activity of FTO, which specifically demethylated NFATC1 and prevented YTHDF2 from degrading it. We found a significant reduction in type I fibers, mRNA levels of MYH7 and MYH7B, and expression of FTO on the concave side of AIS. The percentage of type I fibers showed a positive correlation with the expression level of FTO. The asymmetric patterns observed in AIS were consistent with those seen in congenital scoliosis, and the asymmetry of FTO expression and fiber type in AIS was largely restored by Schroth exercises. Conclusions FTO supports the formation of murine slow-twitch fibers in an NFATC1–YTHDF2 dependent manner. The consistent paraspinal muscle features seen in AIS and congenital scoliosis, as well as the reversible pattern of muscle fibers and expression of FTO in AIS suggest that FTO may contribute to the muscle fiber remodeling secondary to scoliosis.

Cotton fiber is a raw material for the global textile industry and fiber quality is essential to its industrial application. Carotenoids are plant secondary metabolites that may serve as dietary components, regulate light harvesting, and scavenge reactive oxygen species. Although carotenoids accumulate predominantly in rapidly elongating cotton fibers, their roles in cotton fiber development remain poorly understood. In this study, a fiber-specific promoter proSCFP was applied to drive the expression of GhOR1Del, a positive regulator of carotenoid accumulation, to upregulate the carotenoid level in cotton fiber in planta. Fiber length, strength, and fineness were increased in proSCFP:GhOR1Del transgenic cotton and abscisic acid (ABA) and ethylene contents were increased in elongating fibers. The ABA downstream regulator GhbZIP27a stimulated the expression of the ethylene synthase gene GhACO3 by binding to its promoter, suggesting that ABA promoted fiber elongation by increasing ethylene production. These findings suggest the involvement of carotenoids and ABA signaling in promoting cotton fiber elongation and provide a strategy for improving cotton fiber quality.

Fire blight, caused by the bacterium Erwinia amylovora, is a major threat to pear production worldwide. Bacteriophages, viruses that infect bacteria, are a promising alternative to antibiotics for controlling fire blight. In this study, we isolated a novel bacteriophage, RH-42-1, from Xinjiang, China. We characterized its biological properties, including host range, plaque morphology, infection dynamics, stability, and sensitivity to various chemicals. RH-42-1 infected several E. amylovora strains but not all. It produced clear, uniform plaques and exhibited optimal infectivity at a multiplicity of infection (MOI) of 1, reaching a high titer of 9.6 × 109 plaque-forming units (PFU)/mL. The bacteriophage had a short latent period (10 min), a burst size of 207 PFU/cell, and followed a sigmoidal one-step growth curve. It was stable at temperatures up to 60 °C but declined rapidly at higher temperatures. RH-42-1 remained viable within a pH range of 5 to 9 and was sensitive to extreme pH values. The bacteriophage demonstrates sustained activity upon exposure to ultraviolet radiation for 60 min, albeit with a marginal reduction. In our assays, it exhibited a certain level of resistance to 5% chloroform (CHCl3), 5% isopropanol (C3H8O), and 3% hydrogen peroxide (H2O2), which had little effect on its activity, whereas it showed sensitivity to 75% ethanol (C2H5OH). Electron microscopy revealed that RH-42-1 has a tadpole-shaped morphology. Its genome size is 14,942 bp with a GC content of 48.19%. Based on these characteristics, RH-42-1 was identified as a member of the Tectiviridae family, Alphatectivirus genus. This is the first report of a bacteriophage in this genus with activity against E. amylovora.

Organic acids are a key active component of Pinellia ternata (Thunb.) Breit, and their concentration is closely associated with the quality of P. ternata. Developing an efficient and rapid method for detecting organic acids can offer a valuable technology for real-time assessment of P. ternata quality. In this work, a high-performance liquid chromatography coupled to triple quadrupole mass spectrometry (HPLC-QqQ-MS/MS) approach, combining the optimization of extraction conditions using response surface methodology (RSM), was developed for the simultaneous quantitative analysis of six organic acids in P. ternata. The optimal extraction conditions for organic acids in P. ternata were ultrasonic extraction with a solid–liquid ratio of 1:50, ultrasonic time of 60 min, and extraction temperature of 55 °C. Multiple-reaction monitoring (MRM) scanning was employed for quantification using HPLC-QqQ-MS/MS in negative ionization mode through a single run of 10 min, and the limit of detection (LLOD) and limit of quantification (LLOQ) for organic acids were as low as 0.138 ng/mL and 0.614 ng/mL, respectively. The relative standard deviations (RSDs) of reproducibility, precision, and stability were all lower than 5.0%, and recovery rates were 97.75–107.14%, with RSDs < 5.0%. Finally, this method was successfully applied to determine the investigated organic acids in 12 production regions of P. ternata, revealing significant differences between different production areas. This indicates that the optimized method is suitable for further accurate investigations of organic acids of P. ternata.

Abstract Osteosarcoma (OS) is the most common primary malignant pediatric bone tumor and is characterized by high heterogeneity. Studies have revealed a wide range of phenotypic differences among OS cell lines in terms of their in vivo tumorigenicity and in vitro colony-forming abilities. However, the underlying molecular mechanism of these discrepancies remains unclear. The potential role of mechanotransduction in tumorigenicity is of particular interest. To this end, we tested the tumorigenicity and anoikis resistance of OS cell lines both in vitro and in vivo. We utilized a sphere culture model, a soft agar assay, and soft and rigid hydrogel surface culture models to investigate the function of rigidity sensing in the tumorigenicity of OS cells. Additionally, we quantified the expression of sensor proteins, including four kinases and seven cytoskeletal proteins, in OS cell lines. The upstream core transcription factors of rigidity-sensing proteins were further investigated. We detected anoikis resistance in transformed OS cells. The mechanosensing function of transformed OS cells was also impaired, with general downregulation of rigidity-sensing components. We identified toggling between normal and transformed growth based on the expression pattern of rigidity-sensing proteins in OS cells. We further uncovered a novel TP53 mutation (R156P) in transformed OS cells, which acquired gain of function to inhibit rigidity sensing, thus sustaining transformed growth. Our findings suggest a fundamental role of rigidity-sensing components in OS tumorigenicity as mechanotransduction elements through which cells can sense their physical microenvironment. In addition, the gain of function of mutant TP53 appears to serve as an executor for such malignant programs.

Abstract Heatwaves impose serious impacts on ecosystems, human health, agriculture, and energy consumption. Previous studies have classified heatwaves into independent daytime, independent nighttime, and compound daytime-nighttime types, and examined the long-term changes in the three types. However, the underlying mechanisms associated with the variations in different heatwave types remain poorly understood. Here we present the first investigation of the local physical processes associated with the daytime, nighttime, and compound heatwaves over the global land during 1979–2020. The results show that three heatwave types occur frequently and increasingly in most regions worldwide. Nighttime and compound heatwaves exhibit stronger increases in both frequency (the yearly number of the events) and fraction (the ratio of the yearly number of one heatwave type to the total yearly number of all types) than daytime heatwaves. Composite diagnostic analyses of local meteorological variables suggest that daytime heatwaves are associated with increased solar radiation under dry conditions and reduced cloud cover and humidity under a clear sky. In contrast, nighttime heatwaves are typically accompanied by moist conditions with increases in cloud fraction, humidity, and longwave radiation at night. These synoptic conditions for daytime and nighttime heatwaves are combined to contribute to compound heatwaves. Local divergences and moisture fluxes responsible for different heatwaves are further revealed. Positive moisture divergence anomalies are seen in most land areas for daytime and compound heatwaves, while they mainly appear in low latitudes for nighttime heatwaves. Our research provides a comprehensive understanding of the local mechanisms of different heatwave types, informing future risks and impact assessments.

IQ67-domain (IQD) proteins function in plant defense and in organ development. The mechanisms by which they influence cotton fiber development are unknown. In the present study, GhIQD10 was expressed mainly in the transition period of cotton fiber development, and GhIQD10-overexpression lines showed shorter fibers. GhIQD10 interacted with GhCaM7 and the interaction was inhibited by Ca2+. In in vitro ovule culture, Ca2+ rescued the shorter-fiber phenotype of GhIQD10-overexpression lines, which were insensitive to the Ca2+ channel inhibitor verapamil and the Ca2+ pool release channel blocker 2-aminoethoxydiphenyl borate. We conclude that GhIQD10 affects cotton fiber elongation via Ca2+ signaling by interacting with GhCaM7. Brassinosteroid (BR) biosynthesis and signaling genes were up-regulated in GhIQD10-overexpression lines. Fiber development in these lines was not affected by epibrassinolide or the BR biosynthesis inhibitor brassinozole, indicating that the influence of GhIQD10 on fiber elongation was not associated with BR.

Under global warming and rapid urbanization, heat extremes, ozone pollution, and their co-occurrences are emerging and posing severe risks to human health. However, possibly different characteristics of independent heat days (IHD), independent ozone pollution (IOP) and compound heat-ozone pollution (CHOP) events are unclear. In this study, we present an investigation of the spatial distribution and mechanisms associated with IHD, IOP and CHOP events during May–October in 2014–2022 by taking the Guangdong-Hong Kong-Macao Greater Bay Area (GBA) of China as an example. The results show that central GBA faces higher compound risk than northwestern and southeastern regions. IHD events are primarily driven by extremely high temperatures and accompanied by anomalous anticyclone and high pressure at both lower and upper troposphere levels, under the joint effects of the South Asian high and western North Pacific subtropical high. IOP events are predominantly accompanied by decreased cloud cover, air humidity and enhanced downward solar radiation. CHOP events are influenced by increases in both temperature and downward solar radiation. The circulation patterns of CHOP events are somewhat similar to IOP but with much stronger magnitude and faster developing process, and both are likely related to tropical cyclone activities. Our findings can strengthen the early forecasting of these extreme events and mitigate their negative impacts.

Background: The plasma concentrations of the four most commonly used first-line anti-tuberculosis (TB) drugs, isoniazid (INH), rifampicin (RMP), ethambutol (EMB), and pyrazinamide (PZA), are often not within the therapeutic range. Insufficient drug exposure could lead to drug resistance and treatment failure, while excessive drug levels may lead to adverse reactions. The purpose of this study was to identify the physiological parameters influencing anti-TB drug concentrations.Methods: A retrospective cohort study was conducted. The 2-h plasma concentrations of the four drugs were measured by using the high-performance liquid chromatography-tandem mass spectrometry method.Results: A total of 317 patients were included in the study. The proportions of patients with INH, RMP, EMB, and PZA concentrations within the therapeutic range were 24.3%, 31.5%, 27.8%, and 18.6%, respectively. There were positive associations between the concentrations of INH and PZA and RMP and EMB, but negative associations were observed between the concentrations of INH and RMP, INH and EMB, RMP and PZA, and EMB and PZA. In the multivariate analysis, the influencing factors of the INH concentration were the PZA concentration, total bile acid (TBA), serum potassium, dose, direct bilirubin, prealbumin (PA), and albumin; those of the RMP concentration were PZA and EMB concentrations, weight, α-l-fucosidase (AFU), drinking, and dose; those of the EMB concentration were the RMP and PZA concentrations, creatinine, TBA and indirect bilirubin; and those of the PZA concentration were INH, RMP and EMB concentrations, sex, weight, uric acid and drinking.Conclusion: The complex correlations between the concentrations of the four first-line anti-TB drugs lead to a major challenge in dose adjustment to maintain all drugs within the therapeutic window. Levels of TBA, PA, AFU, and serum potassium should also be considered when adjusting the dose of the four drugs.

High‐performance beam‐steering devices play a crucial role for optical wireless communication (OWC), which can meet the demand for increasing number of wireless mobile devices and emerging high‐speed multimedia applications. Conventional beam‐steering optical components like spatial light modulator and digital micromirror device are limited to realize a small steering angle (typ. several degrees), thus dramatically limits the spatial scope of OWC. Herein, a beam‐steering metadevice utilizing an ultracompact metasurface assisted with a spatial light modulator is presented, which can significantly increase the beam‐steering angle without at the cost of complicated optical setup like conventionally used angle magnifier. Based on the actively tunable beam‐steering metadevice, an intelligent bidirectional optical broadcasting communication system is designed and experimentally demonstrated, which exhibits nine broadcasting areas covering a large field‐of‐view of 20° × 20°, with user‐defined dynamic beam‐steering ability in each area, and each user has a data rate of 10 Gbps for both upstream and downstream transmission. The proposed metadevices can meet the demand of intelligent optical wireless communication which requires both high‐performance and low‐cost for practical purpose, under currently available technical architectures.

Background2’,5’-oligoadenylate synthetase 1 (OAS1), has been reported as a tumor driver gene in breast carcinoma and pancreatic carcinoma. However, the role of OAS1 in most tumors has not been reported.MethodsThe original data of 35 tumor types were down load from the TCGA (The Cancer Genome Atlas) database and Human Protein Atlas (HPA) database. TIMER2, Kmplot, UALCAN, and TISIDB tools were used to investigate the expression and function of OAS1, and the role of OAS1 in prognosis, diagnostic value, and immune characteristics of pan-cancer. LUAD and PRAD cell lines, A549, H1975, PC-3 and C4-2 were utilized to perform cell function tests.ResultsOAS1 expression was up-regulated in 12 tumor types and down-regulated in 2 tumor types. High OAS1 expression was correlated with poor prognosis in 6 tumor types, while high OAS1 expression was correlated with good prognosis in 2 tumor types. OAS1 was correlated with molecular subtypes in 8 tumor types and immune subtypes in 12 tumor types. OAS1 was positively associated with the expression of numerous immune checkpoint genes and tumor mutational burden (TMB). OAS1 had potential diagnostic value in 15 tumor types. Silence of OAS1 significantly inhibited the cell proliferation ability, and promoted G2/M cell cycle arrest of LUAD and PRAD cells. Meanwhile, silence of OAS1 enhanced cisplatin-induced apoptosis of LUAD and PRAD cells, but weakened cell migration.ConclusionThis pan-cancer study suggests that OAS1can be used as a molecular biomarker for prognosis in pan-cancer and may play an important role in tumor immune response.

The deployment of disease resistance genes is currently the most economical and environmentally sustainable method of crop protection. However, disease resistance genes can rapidly break down because of constant pathogen evolution, particularly when they are deployed singularly. Polygenic resistance is, therefore, considered the most durable, but combining and maintaining these genes by breeding is a laborious process as effective genes are usually unlinked. The deployment of polygenic resistance with single-locus inheritance is a promising innovation that overcomes these difficulties while enhancing resistance durability. Because of major advances in genomic technologies, increasing numbers of plant resistance genes have been cloned, enabling the development of resistance transgene stacks (RTGSs) that encode multiple genes all located at a single genetic locus. Gene stacks encoding five stem rust resistance genes have now been developed in transgenic wheat and offer both breeding simplicity and potential resistance durability. The development of similar genomic resources in phytopathogens has advanced effector gene isolation and, in some instances, enabled functional validation of individual resistance genes in RTGS. Here, the wheat stem rust pathosystem is used as an illustrative example of how host and pathogen genomic advances have been instrumental in the development of RTGS, which is a strategy applicable to many other agricultural crop species.

Pinellia ternata (Thunb.) Breit. is a traditional Chinese medicinal plant that has been widely used in China, Japan and Korea to relieve cough, vomiting, and inflammation. This review summarizes the recent research focus on germplasm resources, genetic diversity, functional genes, genetic breeding and prospects of P. ternata. The impact of germplasm resources and biogeography are the key factors of the effects of traditional Chinese medicinal materials, avoiding the medical negligence caused by using the confused medicine. Genetic diversity and genetic breeding are the basis of germplasm improvement. A virus-free technique of tissue culture is used to rapid propagation of P. ternata, promoting the production of seedlings without season restrictions. Functional gene research is the theoretical basis and target of germplasm improvement. Many genes, such as PtsHSP17.2 and PtSAD have been confirmed to play an important role in heat stress, guiding the selection and breeding of heat-resistant and drought-resistant P. ternata resources. Still, some problems exist in the production of P. ternata, presenting a challenge in breeding and cultivation. We summarize previous studies here and propose directions for further study to advance the research in the production of P. ternata.

Choosing the appropriate antibiotics to treat bacterial infections has grown more challenging as a result of the emergence of antibiotic-resistant bacteria. Aminoglycosides, as broad-spectrum antibiotics, are increasingly being used clinically; however, for most effective employment of aminoglycosides, a comprehensive understanding of aminoglycoside resistance genes’ prevalence and dissemination is required. Therefore, to better understand the global resistance status of aminoglycoside antibiotics and the prevalence of antibiotic-resistance genes (ARGs) in various bacterial species, this systematic review gathered relevant data from multiple studies. Two primary resistance mechanisms—aminoglycoside enzymatic modification and 16S rRNA methylation—were assessed, and the prevalence of the corresponding ARGs was described. The coexistence of aminoglycoside ARGs with other ARGs was also demonstrated, as was the relationship between aminoglycoside ARGs and resistant phenotypes. The lack of effective therapeutic agents to combat resistant pathogens presents a real threat to public health. The combination of aminoglycosides with other antibiotics may provide a novel treatment strategy.

Colon cancer is a disease with high prevalence worldwide. This study sought to investigate Kruppel-like factor 17 (KLF17) mechanism in the development of colon cancer through four-and-a-half-LIM domain protein 1 (FHL1). In colon cancer cells, KLF17 and FHL1 expression was detected by reverse transcription-quantitative polymerase chain reaction (RT-qPCR) and Western blot. After gain- and loss-of-function experiments in colon cancer cells, cell proliferative, invasive, and migrating abilities were tested by cell counting kit-8, transwell, and scratch assays, respectively. The expression of epithelial–mesenchymal transition (EMT)-related genes, E-cadherin, N-cadherin, and Vimentin, was measured by RT-qPCR and Western blot. Chromatin immunoprecipitation and dual-luciferase reporter gene assays were performed to detect the binding of KLF17 and the FHL1 promoter. Finally, a transplantation tumor model in nude mice was established for in vivo validation. Mechanistically, KLF17 facilitated FHL1 transcription by binding to the FHL1 promoter. KLF17 or FHL1 upregulation suppressed the colon cancer cell proliferative, invasive, and migrating capacities, accompanied by elevated E-cadherin expression and diminished N-cadherin and Vimentin expression. Furthermore, FHL1 silencing abrogated the repressive impacts of KLF17 upregulation on colon cancer cell EMT, proliferative, invasive, and migrating capabilities. Furthermore, KLF17 augmented FHL1 expression and curtailed the growth of transplanted tumors in nude mice. Conclusively, KLF17 promoted FHL1 transcription, thereby impeding the invasion, migration, and EMT of colon cancer cells.

A detailed theoretical study is conducted on the nonlinear interference in the same-wavelength bidirectional coherent optical fiber communication systems. The Gaussian noise (GN) model used to evaluate nonlinear interference (NLI) in unidirectional systems is applied and extended to bidirectional transmission scenarios. The extended NLI model shows that in a bidirectional transmission communication system, the backward signal almost does not introduce additional nonlinear crosstalk to the forward signal due to the strong walk-off effect between forward and backward transmitted signals. Specifically, the ratio of the nonlinear crosstalk introduced by the forward and backward signals is about 21 dB, which means that the traditional GN model is also applicable in the bidirectional scenario. This conclusion is validated on the platform of a same-wavelength bidirectional coherent optical communication system based on Optisystem software.

In this paper, an interference fading mitigation method based on subband phase-shift transform in coherent phase-sensitive optical time-domain reflectometer ($\Phi$-OTDR) is proposed. The operation principle of the subband phase-shift transform with complementary amplitude characteristic is presented. The phase shift transform is performed in the digital domain without requiring complex frequency modulation. After subband phase-shift transform, the interference fading can be effectively mitigated through optimal tracking and rotated vector sum (RVS) methods. The experimental results show that the fluctuation of phase intensity is reduced from $\sim$70 dB to $\sim$20 dB and the average standard deviation of the recovered phase is also reduced from 0.223 rad to 0.046 rad. In addition, different disturbance signals are successfully recovered, verifying the effectiveness of the proposed method.

Downscaling Chlorophyll-a (Chl-a) concentration derived from satellite image is crucial for refined applications such as water quality monitoring. However, the precision of downscaling is usually constrained by various environmental factors. In this paper, we develop a downscaling method for Chl-a concentration to improve precision, especially for inland lakes with different surrounding environment. The method downscales the Sentinel-3 Chl-a concentration from 300 m to 30 m, based on the multivariate analysis (MVA) and the gradient boosting decision tree (GBDT) model. Firstly, we analyzed 21 Chl-a concentration related indices to identify optimal factors for Chl-a concentration variability. Secondly, a GBDT model is constructed to convey the non-linear relationship between the optimal factors and Chl-a concentration at coarse resolution. Finally, fine-resolution Chl-a concentrations were produced by employing the model to refine cofactors for 12 distinct lakes. The results indicated that the proposed MVA-GBDT method effectively inferred the variability of Chl-a concentration with a mean RMSE of 4.505 mg/m3, an improvement of 5%–39% over other methods. Furthermore, for lakes with large water quality heterogeneity, the method led to a cross validation RMSE and a difference in accuracy of 5.371 mg/m3 and 0.866 mg/m3, respectively. In addition, this study examined the significance of the auxiliary factors and found that the NDCI and WST were the two most important factors for MVA-GBDT to detect Chl-a concentration distributions, particularly for NDCI in lakes with high nutrient contrasts. These findings contribute to the generation of fine-scale Chl-a concentrations in lakes and support related applications.
