4. Simulation design for forthcoming high quality plasma wakefield acceleration experiment in linear regime at SPARC_LAB

Abstract

In the context of plasma wakefield acceleration beam driven, we exploit a high density charge trailing bunch whose self-fields act by mitigating the energy spread increase via beam loading compensation, together with bunch self-contain operated by the self-consistent transverse field. The work, that will be experimentally tested in the SPARC_LAB test facility, consists of a parametric scan that allows to find optimized parameters in order to preserve the high quality of the trailing bunch over the entire centimeters acceleration length, with a final energy spread increase of 0.1% and an emittance increase of 5 nm. The stability of trailing bunch parameters after acceleration is tested employing a systematic scan of the parameters of the bunches at the injection. The results show that the energy spread increase keeps lower than 1% and the emittance increase is lower than 0.02 mm mrad in all the simulations performed. The energy jitter is of the order of 5%.

Abstract

The present numerical investigation of a Plasma Wakefield Acceleration scenario in the weakly non linear regime with external injection is motivated by the upcoming campaigns at the SPARC\_LAB test facility where the final goal is to demonstrate modest gradient acceleration ($\sim$1 GV/m) with no quality loss. The accelerated bunch can be envisioned to seed a free electron laser. The numerical study has been conducted with the particle-in-cell code ${\tt ALaDyn}$, an exhaustive description of the plasma-acceleration version is provided. The configuration consider a two bunches setup with parameters in the facility range, the bunches are generated and pre-accelerated up to 100 MeV by a high brightness photo-injector prior plasma injection. To verify the working point robustness we have considered case scenario where the driver bunch reaches the plasma or with a larger dimension or with large emittance. We also present an analytical approach based on the envelope equation that allows to reduce the matching condition in the presence of a ramp. Here, we limit our interest to a simplified theoretical case with a linear plasma ramp. As a final aspect we propose to combine classical integrated bunch diagnostics with the test by Shapiro-Wilk, a mathematical test to diagnose bunch deviation from a Gaussian distribution.

Abstract

Beam driven ionisation injection is characterised for a variety of high-Z dopant. We discuss the region of extraction and why the position where electrons are captured influences the final quality of the internally-injected bunch. The beam driven ionisation injection relies on the capability to produce a high gradient fields at the bubble closure, with magnitudes high enough to ionise by tunnelling effect the still bounded electrons (of a high-Z dopant). The ionised electrons are captured by the nonlinear plasma wave at the accelerating and focusing wake phase leading to high-brightness trailing bunches. The high transformer ratio guarantees that the ionisation only occurs at the bubble closure. The quality of the ionisation-injected trailing bunches strongly and non-linearly depends on the properties of the dopant gas (density and initial ionisation state). We use the full 3D PIC code ${\tt ALaDyn}$ to consider the highly three-dimensional nature of the effect. By means of a systematic approach we have investigated the emittance and energy spread formation and the evolution for different dopant gases and configurations.

Abstract

Canine coronavirus (CCoV) is a common agent of gastroenteritis in dogs, although some variants have been found associated with systemic and often fatal diseases. Distinct genotypes (CCoV-I and CCoV-II) and subgenotypes (CCoV-IIa and CCoV-IIb) are worldwide distributed. In Italy, CCoV infections have been occasionally evaluated, but information about the molecular epidemiology and the genomic features of currently circulating strains is limited. This study reports the detection and molecular characterization of CCoV strains from samples collected from 284 dogs in Italy between 2019 and 2021. CCoV RNA was detected in 39 (13.7%) dogs, as a single viral agent (5 animals, 12.8%) or with other viral pathogens (canine parvovirus types 2a/2b/2c; canine adenovirus type 1; norovirus GIV.2) (34 animals, 87.2%). A total of 48 CCoV strains were detected either alone (CCoV-I: 51.3%, CCoV-IIa: 20.5%) or in copresence (CCoV-I and CCoV-IIa, 23.1%); surprisingly, CCoV-IIb was not identified in this study. Five clusters of CCoV-I were detected, and their spike gene sequences showed the highest nucleotide identities with CCoV-I strains collected from Greece in 2008/2009 and from China in 2021. CCoV-IIa spike gene sequences (three variants) had the highest nucleotide identities with CCoV-IIa strains collected in Greece in 2008/2009 and in Italy in 2009/2011. Given the high CCoV diversity and the variable pathogenicity potential, we underline the need of further surveillance studies to increase our understanding of the epidemiology and evolution of these viruses. Abstract

Canine parvovirus type-2 (CPV-2) is a DNA virus that causes a very common worldwide diffused infectious disease in dogs. Since its appearance, the CPV-2 evolved generating novel genetic and antigenic variants (CPV-2a/2b/2c) which are distributed throughout the world. In the present study, the frequency of CPV-2 in a canine population in Sicily (Italy) was investigated, using a polymerase chain reaction (PCR) for a fragment of the VP2 gene. Out of a total of 673 samples from 370 dogs, submitted to the laboratory from July 2009 to August 2015, 265 samples (39.38%) were positive and these were further analyzed by restriction fragment length polymorphism (RFLP) and DNA sequence analysis. A high prevalence of the CPV-2c variant (79.56% of CPV-2 positive dogs) was observed in this survey, underlining that this type of CPV field isolate is prevalent circulating in Sicily. Sequence and phylogenetic analysis showed a close relationship with CPV-2a and CPV-2c strains from Europe and non-European countries. Considering that CPV-2c is reaching a worldwide distribution and that this variant is also affecting vaccinated dogs, efforts should be made towards the development of new CPV vaccines.

Historical flora and/or vegetation surveys highlight how the small islands of the Ionian coast of Sicily (Bella, Ciclopi islands, Vendicari, Capo Passero), currently protected as Nature Reserves and/or part of the Natura 2000 network have undergone major landscape changes following alterations in anthropogenic pressure. Landscape changes have been more prominent for the islands located in the northern part of the Ionian coast, with the consequence of increasing populations of invasive alien species. Abstract

Canine parvovirus type 2 (CPV‐2) emerged suddenly in the late 1970s as pathogen of dogs, causing a severe and often fatal gastroenteric disease. The original CPV‐2 was replaced by three antigenic variants, CPV‐2a, CPV‐2b and CPV‐2c, which to date have gained a worldwide distribution with different relative proportions. All previous studies conducted in Africa were based on partial VP2 gene sequences. The aim of this study was to provide a genome analysis to characterize the CPV strains collected in Nigeria, Africa. Rectal swab samples (n = 320) were collected in 2018 and tested by means of an immunochromatographic assay. Among the 144 positive samples, 59 were selected for further analyses using different molecular assays. The results revealed a high prevalence of CPV‐2c (91.5%) compared to the CPV‐2a variant (8.5%). The VP2 gene sequences showed a divergence from the strains analysed in 2010 in Nigeria and a closer connection with CPV strains of Asian origin. The non‐structural gene analysis evidenced amino acid changes never previously reported. The molecular analysis based on genomic sequences evidenced a geographical pattern of distribution of the analysed strains, suggesting a potential common evolutionary origin with CPV of Asian origin. This study represents the first CPV molecular characterization including all the encoding gene sequences conducted in the African continent and contributes to define the current geographical spread of the CPV variants worldwide. Abstract

Background: Toxoplasmosis is one of the most frequent parasitic infections in animals causing reproductive disorders and thus notable economic losses in productivity. Among food animals, pigs along with sheep and goats possess the highest incidence of Toxoplasma gondii cysts in meat, and play a role as a source of human infection. Methods: The commercial ELISA kit (PrioCHECK® Toxoplasma Ab SR, Prionics Schlieren-Zurich, Switzerland) for the detection of specific antibodies against T. gondii in swine serum was compared with a commercial IFAT (indirect fluorescent antibody test) (Toxo-Spot IF, bioMérieux, France), used as the reference test. Results: The kappa value obtained comparing the results performed on sera by ELISA with the by IFAT was 1. By a receiver operating characteristics curve analysis, the commercial ELISA had a relative sensitivity of 100%, and a relative specificity of 100% respect to IFAT. Conclusions: The commercial ELISA showed a very good agreement with the commercial IFAT in the detection of serum antibodies to Toxoplasma in pigs. Our study confirmed the usefulness of the commercial ELISA kit for the detection of antibodies against T. gondii in pigs, representing a valuable tool to improve the diagnostic activity for T. gondii in swine populations at the farm level or at the slaughterhouse, contributing to the control of this widespread infection.
