Background and objectives: Soil contamination with Toxocara spp. eggs is the main source of human toxocariasis. The aim of this study was to determine rate of soil contamination with Toxocara spp. eggs in Yazd, central Iran. Methods: During September 2014 to February 2015, a total of 220 soil samples were collected from public parks, primary schools, green areas, kindergartens, suburban areas, streets, residential complexes, backyards and a bus terminal in the city of Yazd, Yazd Province. Soil samples were investigated for the presences of Toxocara eggs by flotation technique using sucrose solution (1.2 g/cm3) and using light microscopy. Data analysis was carried out by SPSS (version 23) software package. Results: A total of 220 soil samples from nine different places were examined. Of these samples, 51 (23.2%) were contaminated with Toxocara spp. eggs. The contamination rate was 26.3% in public parks, 19.2% in primary schools, 28.6% in green areas, 22.2% in kindergartens, 24% in suburban areas, 8.3% in streets, 18.8% in residential complexes, 19% in backyards and 28.6% in the bus terminal. There was no significant difference in the soil contamination rate with Toxocara spp. eggs between different sampling places (χ2 = 3.096, d.f = 8 and P= 0.92) and sampling areas (χ2= 2.289, d.f = 2 and P= 0.318). Conclusion: The high rate of soil contamination with Toxocara spp. eggs in Yazd emphasize on the need for preventive measures, such as improving sanitation, providing public health education and controlling stray dogs and cats near children play areas. Background: This study investigates the scolicidal effects of Cinnamomum zeylanicum essential oil against the protoscoleces of hydatid cysts and its toxicity in the mice model. Materials and Methods: Gas chromatography/mass spectroscopy analyses were used to identify the constituents of essential oil. Protoscoleces were treated with different concentrations of the essential oil (6.25-100 µL/mL) in each test tube for 5-30 min. The viability of protoscoleces was confirmed using eosin exclusion test (0.1% eosin staining). Forty-eight male NMRI mice were also used to determine the toxicity of C. zeylanicum essential oil (0.5-4 mL/kg). Results: The main components were found to be cinnamaldehyde (91.8%), ρ metoxicinamate (1.57%), and a pinene (1.25%). Findings indicate that C. zeylanicum essential oil with the concentrations of 100 and 50 µL/mL killed 100% of protoscoleces after 5 min of exposure. Also, the lower concentrations of C. zeylanicum essential oil motivated a late protoscolicidal effect. The LD50 value of intraperitoneal injection of C. zeylanicum essential oil was 2.07 mL/kg body weight after 48 h, and the maximum nonfatal dose was 1.52 mL/kg body weight. The results also showed that there was no significant toxicity following oral administration of C. zeylanicum essential oil for 2 weeks. Conclusion: The results exhibited the favorable scolicidal activity of C. zeylanicum, which could be applied as a natural scolicidal agent in hydatid cyst surgery. [ABSTRACT FROM AUTHOR]

Background: The present study aimed to evaluate the effect of methotrexate (MTX) alone and in combination with meglumine antimoniate (MA, Glucantime) against sensitive and MA-resistant Leishmania tropica stages in vitro. Methods: The present study was carried out in 2014 in Leishmaniasis Research Center at School of Medicine, Kerman University of Medical sciences, Kerman, Iran. The effects of MTX alone and along with MA on promastigote and amastigote stages of sensitive (SS) and MA-resistant (RS) L. tropica strains have been evaluated using a colorimetric MTT assay and in a macrophage model, respectively. In addition, the inhibitory effect of MTX on the Leishmania invasion of murine macrophages was assessed in promastigotes of both strains of L. tropica. Sensitive and MA resistant L. tropica are referred to those isolates that are responsive or non-responsive to one or two courses of treatment by MA systemically and/or intralesionally, respectively. Results: The findings of OD and IC50 showed that MTX plus MA (SS: 16.1 µg/ml, RS: 39.8 µg/ml) had a higher anti-leishmanial effect than MA (SS: 52.2 µg/ml, RS: 170 µg/ml) or MTX alone (SS: 22.2 µg/ml, RS: 51.4 µg/ml) on promastigotes of both strains of L. tropica. The MTX plus MA caused a significant decrease (P<0.05) in the mean infection rate (MIR) and the mean number of amastigotes in each macrophage compared with positive control. Infectivity of promastigotes is significantly (P<0.05) reduced when it was preincubated with MTX. Conclusion: This study indicated high potency and a synergistic effect of MTX on MA in inhibiting the growth rateof promastigote and amastigote stages of sensitive and meglumine antimoniate-resistant L. tropica. Background: Echinococcus granulosus, Taenia multiceps, Taenia ovis and Taenia hydatigena are among the most prevalent taeniid species of dogs. These tapeworms infect ruminant and humans as intermediate hosts and domestic/wild carnivores as the definitive hosts. Molecular tools using hypervariable microsatellite regions might provide more information about parasite variation. Highly variable and specific tools are needed for transmission tracking studies of canine echinococcosis as an essential element for implementation of hydatid control programs. Suitable microsatellite markers used so far are EmsJ, EmsK, EmsB, EMms1, Egmsca1, Egmsga1, U1 snRNA. The purpose of the present study was to determine the microsatellite variability of EmsB as well as six other microsatellites in major taeniid species infecting dogs in Iran. Methods: Twenty isolates of each of the four Taeniidae tapeworms were collected from sheep during routine veterinary inspection in Tehran, Alborz and Kerman provinces from October 2010 to May 2011. After DNA extraction, PCR was set up with optimum conditions using specific primers for each individual microsatellite marker. All the PCR products were evaluated by agarose gel electrophoresis. We used SDSPAGE for evaluating patterns of PCR products in the tapeworms. Results: E. granulosus as well as Taenia species could be differentiated based on EmsB microsatellite patterns. The electrophoresis patterns of two taeniid genera were readily distinguishable. EmsB could be specifically used in epidemiological studies of canine echinococcosis. Zataria multifloraBoiss (Lamiaceae) commonly grows in Iran is a popular medicinal plant with various pharmacological activities mentioned in traditional Iranian medicine and modern phytotherapy. This study was designed to evaluate the chemical composition and scolicidal effects ofZ. multifloraessential oil on the protoscoleces of hydatid cysts on anin vitromodel. The components of theZ. multifloraessential oil were identified by GC/MS analysis. Protoscoleces were aseptically aspirated from the livers of naturally infected sheep. Various concentrations of essential oil, thymol and carvacrol were used for 5–30 minutes. Eosin exclusion test was used to determine the viability of protoscoleces. The main components were thymol (41.8%), carvacrol (28.8%), andp-cymene (8.4%). Findings showed that essential oil at the concentrations of 12.5 and 6.25 μL/mL killed 100% protoscoleces after 5 and 20 minutes of exposure, respectively. In addition, thymol and carvacrol at the concentrations of 100 μg/mL and 100 μL/mL killed 100% protoscoleces after 10 minutes incubation, respectively. Obtained results in this investigation for the first time demonstrated thatZ. multifloraessential oil and its main components might be a natural source for the production of new scolicidal agents. This study aims to evaluate the in vitro scolicidal effects of Nectaroscordum tripedale L. leave extract against protoscoleces of hydatid cysts and its acute toxicity in mice model. Various concentrations of the extract (12.5-100 mg/mL) were used for 5 to 30 min. Viability of protoscoleces was confirmed using eosin exclusion test (0.1% eosin staining). In addition, the acute toxicity of N. tripedale extract was determined for 2 days in mice model. The results showed that the N. tripedale extract at the concentration of 100 mg/mL after 5 min of exposure killed 100% protoscoleces. Similarly, the mean of mortality rate of protoscoleces after 10 min of exposure to concentration of 50 mg/mL was 100%. The LD50 values of intraperitoneal injection of the N. tripedale extract was 3.36 g/kg body wt. and the maximum nonfatal doses were 2.98 g/kg body wt. Background: This investigation aimed to evaluate the in vitro scolicidal effects of Pistacia khinjuk methanolic extract against protoscoleces of hydatid cysts and its acute toxicity in mice NMRI model. Methods: Protoscoleces were aseptically extracted from sheep livers having hydatid cysts. Various concentrations of the essential oil (12.5- 100 mg/mL) were used for 10 to 60 min. Viability of protoscoleces was confirmed using eosin exclusion test (0.1% eosin staining). Twenty-four male NMRI mice were used to assess the acute toxicity of P. khinjuk. Results: P. khinjuk extract at the concentrations of 100 mg/mL after 10 min of exposure killed 100% of protoscoleces. Similarly, the mean of mortality rate of protoscoleces after 20 min of exposure to the concentration of 50 mg/mL was 100%. The LD50 of the intraperitoneal injection of the P. khinjuk methanolic extract was 2.8 g/kg and the maximum non-fatal dose was 1.7 g/kg. Conclusion: The findings demonstrated effective scolicidal effects of P. khinjuk extract with no considerable toxicity that might be a natural source for the producing of new scolicidal agent. Extremely low frequency electromagnetic fields have been classified as a possible human carcinogen by the International Agency for Research on Cancer and this has raised some concern about its health effects on employees extensively exposed to these fields at thermal power plants. In this study, the effect of using vitamin E and C supplements have been examined on employees working at a thermal power plant. In this randomized controlled, double-blind clinical trial, 81 employees from different parts of the thermal power plant were enrolled between July and November 2017, and divided into four groups: Group 1 received vitamin E (400 units/day), Group 2: vitamin C (1000 mg/day), Group 3: vitamin E + C and Group 4: no intervention. DNA damage was measured in peripheral blood lymphocytes using comet assay and apoptosis, using flow cytometry. Based on the results, tail intensity and tail length in the vitamin E group, and all comet assay indices in the vitamin E + C and vitamin C groups (except DNA damage index) significantly decreased after the intervention, while the comet assay indices did not change significantly in the control group. None of the flow cytometry indices including early apoptosis, late apoptosis and necrosis changed after intervention in either group. The use of antioxidant vitamins such as E and C, can increase the activity of the non-enzymatic antioxidant defense system, and protect DNA from damage caused by exposure to extremely low frequency magnetic fields. But, taking these vitamins has no effect on apoptosis.
