A simple, sensitive optical analyzer for the rapid determination of cyanide in blood in point of care applications is described. HCN is liberated by the addition of 20% H(3)PO(4) and is absorbed by a paper filter impregnated with borate-buffered (pH 9.0) hydroxoaquocobinamide (hereinafter called cobinamide). Cobinamide on the filter changes color from orange (λ(max) = 510 nm) to violet (λ(max) = 583 nm) upon reaction with cyanide. This color change is monitored in the transmission mode by a light emitting diode (LED) with a 583 nm emission maximum and a photodiode detector. The observed rate of color change increases 10 times when the cobinamide solution for filter impregnation is prepared in borate-buffer rather than in water. The use of a second LED emitting at 653 nm and alternate pulsing of the LEDs improves the limit of detection by 4 times to ~0.5 μM for a 1 mL blood sample. Blood cyanide levels of imminent concern (≥10 μM) can be accurately measured in ~2 min. The response is proportional to the mass of cyanide in the sample: smaller sample volumes can be successfully used with proportionate change in the concentration LODs. Bubbling air through the blood-acid mixture was found effective for mixing of the acid with the sample and the liberation of HCN. A small amount of ethanol added to the top of the blood was found to be the most effective means to prevent frothing during aeration. The relative standard deviation (RSD) for repetitive determination of blood samples containing 9 μM CN was 1.09% (n = 5). The technique was compared blind with a standard microdiffusion-spectrophotometric method used for the determination of cyanide in rabbit blood. The results showed good correlation (slope 1.05, r(2) 0.9257); independent calibration standards were used.

Dimethyl sulfoxide solution of N‐(2‐hydroxynapthylidene)‐L‐isoleucinyl‐4,6‐O‐ethylidene‐β‐D‐glucopyranosylamine (L1) affords single crystals of its ketoenamine configuration. The noncovalent intermolecular interactions present in the molecules have been explored by Hirshfeld surface analysis and 2D fingerprint plots generated using crystallographic data. The structural parameters of L1 in both the tautomeric forms (ketoenamine and phenolimine) at ground states have been optimized using Hartree–Fock and density functional theory calculations in gaseous and solution phases. The Fourier transform infrared, nuclear magnetic resonance, and UV–visible spectra of this molecule have been compared with the theoretically calculated data of both the tautomeric forms. Additionally, Mulliken population analysis, molecular electrostatic potential, and natural bond order analysis provide a better understanding of charge distribution, presence of electrophilic/nucleophilic sites, and intramolecular charge transfer, respectively, in the molecule. We report a novel system to analyze atmospheric nitrophenols (NPs). Pain or air sample extracts (1 mL) are pre-concentrated on a narrow bore (2 mm) aliphatic anion exchanger. In the absence of strong retention of NPs exhibited by aromatic ion exchangers, retained NPs are eluted as a plug by injection of 100 [micro]L of 0.1 M [Na.sub.2]S[O.sub.4] on to a short (2 x 50 mm) reverse phase C-18 column packed with 2.2 [micro]m particles. The salt plug passes through the C-18 column unretained while the NPs are separated by an ammonium acetate buffered methanol--water eluent, compatible with mass spectrometry (MS). The eluted NPs are measured with a long path Teflon AF-based liquid core waveguide (0.15 x 1420 mm) illuminated by a 403 nm light emitting diode and detected by a monolithic photodiode-operational amplifier. The waveguide is rendered chemically active by suspending it over concentrated ammonia that permeates into the lumen. The NPs ionize to the yellow anion form ([[lambda].sub.max] ~ 400 nm). The separation of 4-nitrophenol, 2,4-dinitrophenol, 2-methyl-4-nitrophenol, 3-methyl-4-nitrophenol, and 2-nitrophenol (these are the dominant NPs, typically in that order, in both rain and air of Houston and Arlington, TX, confirmed by tandem MS) takes just over 5 min with respective S/N = 3 limits of detection (LODs) of 60, 12, 30, 67, and 23 pg/mL compared to MS/MS LODs of 20, 49, 11, 20, and 210 pg/mL. We report on electrodialytic introduction of ionizable molecules of significant size (e.g., 4-(2-pyridyl(azo) resorcinol, PAR)) in capillary scale flow systems. Such reagent introduction can be conducted without volumetric dilution, easily programmed through current control and with excellent mbdng characteristics. Electrodialytic transport of large hydrophobic aromatic ions through conventional aromatic ion exchangers is inefficient. Such ions are strongly retained by hydrophobic and [pi]-[pi] interactions. An external electric field cannot modulate this retention. We show that the electrodialytic introduction of aromatic dye anions is readily possible through both unmodified cellulose dialysis membranes and through cellulose membranes modified with methacrylate skeleton anion exchangers. The applied electrodialysis current conveniently controls the reagent flux. Although the applied voltage is sufficient to cause electrolytic production of hydrogen and oxygen; the gases are generated outside the flowstream of interest. The present device was constructed with a sub-microliter internal volume. We show capillary scale trace analysis of transition metals. A limit of detection of 0.5 fmol Zn (S/N = 3) is demonstrated with a capillary scale flow injection system with a simple light emitting diode based detector. 10.1021/ac100882a
