BACKGROUND: The oleaginous yeast Rhodotorula toruloides is a promising chassis organism for the biomanufacturing of value-added bioproducts. It can accumulate lipids at a high fraction of biomass. However, metabolic engineering efforts in this organism have progressed at a slower pace than those in more extensively studied yeasts. Few studies have investigated the lipid accumulation phenotype exhibited by R. toruloides under nitrogen limitation conditions. Consequently, there have been only a few studies exploiting the lipid metabolism for higher product titers. RESULTS: We performed a multi-omic investigation of the lipid accumulation phenotype under nitrogen limitation. Specifically, we performed comparative transcriptomic and lipidomic analysis of the oleaginous yeast under nitrogen-sufficient and nitrogen deficient conditions. Clustering analysis of transcriptomic data was used to identify the growth phase where nitrogen-deficient cultures diverged from the baseline conditions. Independently, lipidomic data was used to identify that lipid fractions shifted from mostly phospholipids to mostly storage lipids under the nitrogen-deficient phenotype. Through an integrative lens of transcriptomic and lipidomic analysis, we discovered that R. toruloides undergoes lipid remodeling during nitrogen limitation, wherein the pool of phospholipids gets remodeled to mostly storage lipids. We identify specific mRNAs and pathways that are strongly correlated with an increase in lipid levels, thus identifying putative targets for engineering greater lipid accumulation in R. toruloides. One surprising pathway identified was related to inositol phosphate metabolism, suggesting further inquiry into its role in lipid accumulation. CONCLUSIONS: Integrative analysis identified the specific biosynthetic pathways that are differentially regulated during lipid remodeling. This insight into the mechanisms of lipid accumulation can lead to the success of future metabolic engineering strategies for overproduction of oleochemicals.

LOCO is a code for analysis of the linear optics in a storage ring based on the closed orbit response to steering magnets. The analysis provides information on focusing errors, BPM gain and rotation errors,and local coupling. Here, we report the results of an application of LOCO to the KEK-ATF. Although the analysis appears to have provided useful information on the optics of the machine, it appears that one of the main aims of the study to reduce the vertical emittance by correcting the local coupling was not successful, and we discuss some possible reasons for this.

Over the span of 40 years, a substantial number of conceptual and empirical studies have been conducted on financial vulnerability (henceforth FV). These studies primarily covered socioeconomics, finance, management, and medicine. However, there is a paucity of comprehensive reviews and scientific mapping of the extant literature in the FV domain. Bibliometric analysis attempts to provide quantitative and qualitative knowledge in this area. This study was based on a review of 475 articles published in Scopus-indexed journals from 1990 to 2023. The present study employed the Biblioshiny R studio Bibliometrix package for data extraction and analysis. Our analysis provides information on recent publication trends; prominent authors, institutes, and countries; citations; thematic groups; keyword analysis; and social network analysis to identify influential work in this research domain and future gaps. The present analysis contributes to consolidating the existing fragmented literature on FV and highlights its significance during the current pandemic. Additionally, the study would be useful for researchers, practitioners, and academicians to proceed to further explore the area and outline the trends and their empirical investigation. Thus, this paper provides a comprehensive overview of the evolution of financial vulnerability research, utilizing both systematic review and bibliometric analysis to map trends, key themes, and influential works. By synthesizing existing literature and visualizing the network of scholarly contributions, this study highlights the critical factors and providing the future research directions in the field. The findings offer valuable insights for researchers, policymakers, and practitioners, aiding in the formulation of strategies to mitigate financial vulnerability and promote financial resilience in diverse socioeconomic contexts. This study analyses the relationship between foreign direct investment, economic growth, urbanization, energy use, and carbon emissions in Brazil, Russia, India, China, and South Africa (BRICS countries) between 1990 and 2014. According to the empirical results, both environmental Kuznets curve (EKC) and pollution haven hypotheses (PHH) are confirmed. Urbanization contributes to reducing carbon emissions, while energy use is one of the main driving forces of ascending carbon emissions. The main advance of this paper lies in the moderating effect of foreign direct investment and energy usage on carbon emission in the case of the BRICS. The empirical results confirm a dampening impact of foreign direct investment on energy use, generating a correction in carbon emission. Thus, structural transformations are highlighted with a positive influence on energy efficiency and sustainable growth.
