The human neocortex has a cytoarchitecture composed of six layers with an intrinsic organization that relates to afferent and efferent pathways for a high functional specialization. Various histological, neurochemical, and connectional techniques have been used to study these cortical layers. Here, we explore the additional possibilities of swift ion beam and synchrotron radiation techniques to distinguish cellular layers based on the elemental distributions and areal density pattern in the human neocortex. Breeding and engineering of biofortified crops will benefit from a better understanding of bottlenecks controlling micronutrient loading within the seeds. However, few studies have addressed the changes in micronutrient concentrations, localization, and speciation occurring over time. Therefore, we studied spatial patterns of zinc and iron accumulation during grain development in two barley lines with contrasting grain zinc concentrations. Microparticle-induced-X-ray emission and laser ablation-inductively coupled plasma mass spectrometry were used to determine tissue-specific accumulation of zinc, iron, phosphorus, and sulfur. Differences in zinc accumulation between the lines were most evident in the endosperm and aleurone. A gradual decrease in zinc concentrations from the aleurone to the underlying endosperm was observed, while iron and phosphorus concentrations decreased sharply. Iron co-localized with phosphorus in the aleurone, whereas zinc co-localized with sulfur in the sub-aleurone. We hypothesize that differences in grain zinc are largely explained by the endosperm storage capacity. Engineering attempts should be targeted accordingly.

Tartary buckwheat (Fagopyrum tataricum Gaertn.) is a gluten-free pseudo-cereal crop with a grain nutrient profile that makes it an excellent alternative foodstuff. The distribution of calcium (Ca), magnesium (Mg), phosphorus (P) and sulphur (S) was investigated by micro-PIXE (particle induced X-ray emission) to resolve allocation and concentration of the elements in nine distinct grain tissues. Magnesium, P and S were preferentially allocated to the cotyledons and the embryonic axis (both inner and outer tissues), and Ca was predominant in the pericarp where two Ca-rich layers were observed. Allocation of P and S to aleurone suggests that this layer of cells, although not as prominent as in cereal grain, is rich in phytate and proteins. Quantitative information on spatial distribution of mineral elements in the edible grain may be useful in the technological processing of the grain and particularly in reducing the amount of mineral-element loss during milling.

The existing external beamline at the Microanalytical Center of the Jo��ef Stefan Institute (JSI) in Ljubljana has been currently upgraded in order to extend the capabilities towards in-air PIXE imaging across relatively large sample surface area (~ 1 cm2) with lateral resolution of sub 100 ��m. For that purpose, a doublet of magnetic quadrupole lenses was installed on the beamline together with the newly designed exit nozzle with a 200 nm Si3N4 exit window resulting finally in ~ 50��50 mm2 lateral resolution. Additionally, an in-vacuum beam chopper was installed before the exit nozzle to measure the proton dose deposited on the sample. The beam characterization results are presented together with the first application from the field of biology, namely quantitative concentration maps of a longitudinal section of a whole thick wheat grain.

Human hair absorbs numerous biomolecules from the body during its growth. This can act as a fingerprint to determine substance intake of an individual, which can be useful in forensic studies. The cocaine concentration profile along the growth axis of hair indicates the time evolution of the metabolic incorporation of cocaine usage. It could be either assessed by chemical extraction and further analysis of hair bundels, or by direct single hair fibre analysis with mass spectroscopy imaging (MSI). Within this work, we analyzed the cocaine distribution in individual hair samples using MeV-SIMS. Unlike conventional surface analysis methods, we demonstrate high yields of nonfragmented molecular ions from the surface of biological materials, resulting in high chemical sensitivity and non-destructive characterisation. Hair samples were prepared by longitudinally cutting along the axis of growth, leaving half-cylindrical shape to access the interior structure of the hair by the probing ion beam, and attached to the silicon wafer. A focused 5.8 MeV 35Cl6+ beam was scanned across the intact, chemically pristine hair structure. A non-fragmented protonated [M+ H]+ cocaine molecular peak at m/z = 304 was detected and localized along the cross-section of the hair. Its intensity exhibits strong fluctuations along the direction of the hair’s growth, with pronounced peaks as narrow as 50 micrometres, corresponding to a metabolic incorporation time of approx. three hours.

International audience; Secondary ion mass spectrometry (SIMS), based on primary ions within the MeV energy domain, also known as MeV-SIMS, is a subject of increasing scientific interest. The main drive for the interest in the development of MeV-SIMS is the ability to desorb high yields of large non-fragmented organic molecular ions from the sample surface. This makes MeV-SIMS particulary useful in imaging of biological tissues.Imaging methods based on scanning a focused primary ion beam are associated with demanding focusing of the heavy energetic ions. As an alternative, stigmatic imaging mode has been studied here, applying point-to-point imaging characteristics of secondary ions in the linear Time-Of-Flight mass spectrometer. In stigmatic imaging approaches, spatial resolution is independent of the focussed spot size of the ionising primary ion beam, but instead dependant on the ability of the ion optics to project an image of the ion distributions removed from the surface onto a position sensitive ion detector.

Halophytes are able to tolerate relatively high concentrations of hazardous metals in a growing substrate, what makes them suitable candidates for phytoremediation of metal-contaminated soils. In this work, we aimed to study the physiological responses of the halophyte Sesuvium portulacastrum L. to Ni, with main focus on Ni localization, compartmentation and ligand environment, to decipher Ni tolerance and toxicity mechanisms. Seedlings were grown in hydroponic nutrient solution containing 0, 25, 50 and 100 μM Ni as NiCl2 for 3 weeks. Ni localization in leaves was assessed by micro-proton-induced X-ray emission (micro-PIXE). Ni ligand environment was studied by Ni K-edge X-ray absorption near edge structure (XANES). In addition, Ni-soluble, weakly bound/exchangeable and insoluble leaf tissue fractions were determined by sequential extraction. Results show that S. portulacastrum is able to tolerate up to ~ 500 μg g−1 dry weight (DW) of Ni in the shoots without significant growth reduction. At higher Ni concentrations (> 50 μM Ni in nutrient solution), chloroses were observed due to the accumulation of Ni in photosynthetically active chlorenchyma as revealed by micro-PIXE. Water storage tissue represented the main pool for Ni storage. Incorporation of Ni into Ca-oxalate crystals was also observed in some specimens, conferring tolerance to high leaf Ni concentrations. The majority of Ni (> 70%) was found in soluble tissue fraction. Ni K XANES revealed Ni bound mainly to O- (55%) and N-ligands (45%). Ni toxicity at higher Ni levels was associated with Ni binding to amino groups of proteins in cytosol of chlorenchyma and increased level of lipid peroxidation. Proline levels also increased at high Ni exposures and were associated with Ni-induced oxidative stress and alteration of water regime.

In this work the influence of surface roughness on the sputter yield of Mo under keV D ion bombardment was investigated for different impact angles. For this purpose, thin films of Mo (~ 120 nm) were deposited by pulsed laser deposition onto graphite substrates with varying surface roughness (Ra ranging from 5 nm to 2–3 µm). The as-deposited samples were irradiated at room temperature by 3 keV D3+ ions originating from an electron-cyclotron-resonance ion gun. Samples were exposed to D ions at angles between 0° and 70° and fluences in range of 1023 D/m2. The areal densities of the Mo marker layers were determined with Rutherford backscattering spectroscopy. For all the surfaces we observed a strong angular dependence of the sputter yield. For smooth and intermediate surface roughnesses, up to Ra ~ 280 nm, we obtained an increase of the sputter yield with the angle up to a factor of five compared to 0°. In contrast, at the highest surface roughness in the 2–3 µm range the sputtering yield decreases with increasing impact angle. The obtained data were compared to SDTrimSP-3D simulations. We obtained good agreement between the simulated and experimental sputter yield for surfaces for which we could provide high resolution atomic force microscopy (AFM) surface representations. As high-resolution surface mapping was not possible for surface roughness of 2–3 μm, we found large deviation between the calculation and the measured data. The combination of measured and simulated data represent important input for predicting the erosion rates of surfaces at inner walls of thermonuclear fusion devices, which are expected to change surface roughness over time by sustained plasma exposure.

Magnesium (Mg) and calcium (Ca) are essential mineral nutrients poorly supplied in many human food systems. In grazing livestock, Mg and Ca deficiencies are costly welfare issues. Here, we report a Brassica rapa loss-of-function schengen3 (sgn3) mutant, braA.sgn3.a-1, which accumulates twice as much Mg and a third more Ca in its leaves. We mapped braA.sgn3.a to a single recessive locus using a forward ionomic screen of chemically mutagenized lines with subsequent backcrossing and linked-read sequencing of second back-crossed, second filial generation (BC2F2) segregants. Confocal imaging revealed a disrupted root endodermal diffusion barrier, consistent with SGN3 encoding a receptor-like kinase required for normal formation of Casparian strips, as reported in thale cress (Arabidopsis thaliana). Analysis of the spatial distribution of elements showed elevated extracellular Mg concentrations in leaves of braA.sgn3.a-1, hypothesized to result from preferential export of excessive Mg from cells to ensure suitable cellular concentrations. This work confirms a conserved role of SGN3 in controlling nutrient homeostasis in B. rapa, and reveals mechanisms by which plants are able to deal with perturbed shoot element concentrations resulting from a “leaky” root endodermal barrier. Characterization of variation in leaf Mg and Ca accumulation across a mutagenized population of B. rapa shows promise for using such populations in breeding programs to increase edible concentrations of essential human and animal nutrients.

MeV-SIMS is an emerging mass spectrometry imaging method that employs fast, heavy ions to desorb secondary molecules from the analyzed sample. High yields and low fragmentation rates of large molecules, associated with the dominating electronic sputtering process, make it particularly useful in biomedical research, where insight into the distribution of organic molecules is vital. Both yield and fragmentation of desorbed molecules in MeV-SIMS rely on characteristics of the primary ion but may also be impaired by poor instrumental settings. After utilizing secondary ion optics in the linear mass spectrometer at the micro-analytical center of the Jožef Stefan Institute, we demonstrate very efficient detection of secondary ions. As a result, the secondary ion yield, using such settings, solely depends on the species and the characteristics of the primary ion. In order to analyze the yield dependence on the primary ion energy, and the corresponding stopping power within the electronic excitation regime, we used a continuous electron multiplier detector to measure the primary ion current during each measurement of the mass spectra. Secondary ion yield as a function of the primary ion energy and charge is presented as well as fragmentation rates of organic molecules arginine and leu-enkephalin. Other influential instrumental drawbacks are also studied, and their effect on the results is discussed.

In this work the synergism between displacement damage creation and presence of hydrogen isotopes was studied. Tungsten samples were irradiated by 10.8 MeV W ions with or without the presence of D ions with two different energies of 300 eV/D and 1000 eV/D and different temperatures. In order to compare the results obtained with different exposure parameters the samples were afterwards additionally exposed to D ions at 450 K to populate the created defects. By increasing the W irradiation time, ion flux and energy, the increase of D concentration and D retention was observed as measured by nuclear reaction analysis and thermal desorption spectroscopy. By fitting the D depth profiles and D desorption spectra by the rate equation code MHIMS-R we could see that additional fill-levels were populated with higher flux and ion energy which ends up in higher final D concentration and retention as compared to experiments with lower D flux and energy.

Element profile signatures of needle tissues differentiated four tissues: epidermis (main contributor: calcium), endodermis (main contributors: magnesium, sulphur and manganese), mesophyll (main contributor: potassium), and transfusion parenchyma (main contributor: zinc). Distribution of elements in cross-sections of Scots pine (Pinus sylvestris L.) needles was investigated using micro-proton-induced X-ray emission. Tissue-specific distributions of magnesium (Mg), sulphur (S), calcium (Ca), phosphorus (P), potassium (K), chlorine (Cl), manganese (Mn), iron (Fe), zinc (Zn), aluminium (Al) and silicon (Si) were resolved in a quantitative manner. Distribution maps and tissue-specific concentrations revealed the largest concentration of Ca in epidermis, of Mg, S and Mn in endodermis, of K in mesophyll and phloem and of Zn in transfusion parenchyma. Phosphorus, Cl, Fe, Al and Si did not exhibit apparent tissue-specific distribution. Inverse allocation of P and Ca was observed, a likely mechanism to prevent their precipitation. Taking the area of tissues into account, relative element distribution calculations indicated that mesophyll contained the majority of the elements studied, except Ca, which predominated in the epidermis (79% of total Ca concentration) and Mn, which predominated in the endodermis (40% of total Mn concentration). When considering a complete element profile of a particular tissue, four clusters were differentiated, which generally supported single-element observations. The first cluster differentiated mesophyll, xylem, phloem, transfusion tracheids and Strasburger cells with predominance of K, the second cluster differentiated epidermis on the basis of Ca, the third cluster differentiated endodermis with contributions from Mg, S and Mn, and the fourth cluster differentiated transfusion parenchyma with contribution from Zn. Information on tissue-specific-element allocations will complement structural and functional knowledge of needle tissues and advance our understanding of element/nutrient transfers in Scots pine.

Nano zero-valent iron (nZVI) has great potential in the remediation of metal(loid)-contaminated soils, but its efficiency in metal(loid) stabilization in the plant-microbe continuum is unclear. This study investigated nZVI-mediated metal(loid) behavior in the arbuscular mycorrhizal (AM) fungal-maize ( Zea mays L.) plant association. Plants with AM fungal inoculation were grown in metal(loid)- (mainly Zn and Pb) contaminated soils (Litavka River, Czech Republic) amended with/without 0.5% (w/w) nZVI. The results showed that nZVI decreased plant metal(loid) uptake but inhibited AM development and its function in metal(loid) stabilization in the rhizosphere. AM fungal inoculation alleviated the physiological stresses caused by nZVI and restrained nZVI efficiency in reducing plant metal(loid) uptake. Micro proton-induced X-ray emission (μ-PIXE) analysis revealed the sequestration of Zn (possibly through binding to thiols) by fungal structures in the roots and the precipitation of Pb and Cu in the mycorrhizal root rhizodermis (possibly by Fe compounds originated from nZVI). XRD analyses further indicated that Pb/Fe mineral transformations in the rhizosphere were influenced by AM and nZVI treatments. The study revealed the counteractive effects of AM and nZVI on plant metal(loid) uptake and uncovered details of metal(loid) behavior in the AM fungal-root-nZVI system, calling into question about nZVI implementation in mycorrhizospheric systems.

Cadmium (Cd) was shown to co-localise with calcium (Ca) in oxalate crystals in the stems and leaves of Cd tolerant Gomphrena claussenii, but Cd binding remained unresolved. Using synchrotron radiation X-ray absorption near edge spectroscopy we demonstrate that in oxalate crystals of hydroponically grown G. claussenii the vast majority of Cd is bound to oxygen ligands in oxalate crystals (>88%; Cd-O-C coordination) and the remaining Cd is bound to sulphur ligands (Cd-S-C coordination). Cadmium binding to oxalate does not depend on the amount of Ca supplied or from which organs the crystals originate (stems and mature leaves). By contrast, roots contain no oxalate crystals and therein Cd is bound predominantly by S ligands. The potential to remove Cd by extraction of Cd-rich oxalate crystals from plant material should be tested in phytoextraction or phytomining strategies.

A solid thick tritiated W sample was produced in order to probe the efficiency of detecting tritium via 3He ion beam using nuclear reactions analysis (NRA). Extensive literature search showed that there were only a few measurements done more than sixty years ago using the reaction with 3He. Their aim was mainly to measure the absolute cross section of the reaction. In order to use this reaction as an analysing tool for low level radioactive waste we need the differential cross section at certain angles. Using a thick tritiated W sample we measured the NRA signal in the 3He energy range from 0.7 MeV to 5.1 MeV and we have detected reaction products with energies between 6.5 MeV and 9.75 MeV that were not present on a deuterated W sample prepared in the same manner. This NRA signal proved to be due to deuterium and protons coming from the nuclear reaction between tritium and 3He. The detection signal increased with 3He energy up to 3.4 MeV and decreased with energy at the highest beam energies. At higher energies particles from W and target impurities start to disturb the measurement. Their origin might be due to other nuclear reactions, for example between 3He and W or 3He with 13C or 14N.

The performance of a nuclear microprobe is dominantly determined by the brightness of the injected ion beam. At Jožef Stefan Institute (JSI), negative hydrogen ion beams are created in a multicusp ion source and injected into a 2 MV tandetron accelerator. The output characteristics of the multicusp ion source were tuned in order to obtain matching proton beam intensities for the ion accelerator and for the object slits as well. For the optimal focusing of the proton beam in a high-current mode (I > 100 pA) to the sub-micrometer dimensions, dedicated thin nanostructures with sharp edges have been manufactured. Set of nanostructures was micromachined by focused ion beam (FIB) at film reference material, produced by Institute for Reference Materials and Measurements (IRMM) and constituted of 57 μg/cm 2 of titanium on vitreous carbon substrate. The proton beam profiles were measured by beam scans across the nanostructures over long measuring times, indicating eventual slow drifts of the sample from a reference beam direction. Overall, proton beam dimensions of 600 nm were obtained, demonstrating appropriate stability for micro-PIXE (micro-Particle Induced X-ray Emission) at sub-micrometer resolution for elemental analysis of biological tissue samples prepared in a freeze-dried state or in a frozen-hydrated state. The resulting performance required for micro-PIXE analysis in a high current mode with a 3 MeV proton beam is presented.

Imaging Mass Spectroscopy (IMS) is a unique research tool providing localization and identification of a wide range of biomolecules as essential data to understand biochemical processes in living organisms. Secondary Ion Mass Spectrometry with high-energy heavy ions (MeV-SIMS) is emerging as a promising IMS technique for chemical imaging of biological tissue.We measured the molecular mass spatial distributions in leaves of khat (Catha edulis). Khat is a natural drug plant, native to eastern Africa and the Arabian Peninsula. In these countries, fresh leaves are being chewed by significant part of population. It was reported that 80% of the adult population in Yemen chew the khat leaves. The main stimulating effects of khat are induced by a monoamine alkaloid called cathinone. During leaf ageing, cathinone is further metabolised to cathine and norephedrine. Earlier studies identified the alkaloids in khat, however little is known on their spatial distribution, reflecting the biosynthesis and accumulation in the tissue.Chemical mapping of alkaloids on cross-sections of khat leaves by MeV-SIMS was done at JSI by a pulsed 5.8 MeV Cl-35(6+) beam, focused to a diameter of 15 mu m, using a linear time-of-flight (TOF) spectrometer with a mass resolution of 500. In addition, measurements of MeV-SIMS mass spectra were performed at Kyoto University by a continuous broad beam of 6 MeV Cu-63(4+) ions at an orthogonal TOF spectrometer with a high mass resolution of 11,000. Sections of leaves were analysed and mass spectra obtained at both MeV-SIMS setups were compared. Tissue-level distributions of detected alkaloids are presented and discussed. (C) 2017 Elsevier B.V. A new parallel beam wavelength dispersive X-ray emission spectrometer for high energy resolution PIXE analysis using focused proton microbeam has been constructed and installed at the microprobe of the J. Stefan Institute. Polycapillary X-ray optics is used to enhance the solid angle of X-ray collection and to transform collected proton-induced X-rays into quasi parallel beam which is analyzed using diffraction on a flat crystal. The spectrometer is installed in a vacuum chamber and operates in the 2–10 keV energy range. The main characteristics and operational properties are presented together with the results of first characterization measurements. Finally, two selected experimental examples are given illustrating the capabilities of the spectrometer in PIXE analysis and fundamental research in atomic physics.

In this exploratory study, the application of secondary ion mass spectrometry with a heavy primary ion beam with an energy of several MeVs (MeV-SIMS) is presented for the simultaneous detection and spatial distribution mapping of lithium in a single, chemically unprocessed strand of scalp hair across a longitudinal profile.

Symmetrical protein cages have evolved to fulfil diverse roles in nature, including compartmentalization and cargo delivery(1), and have inspired synthetic biologists to create novel protein assemblies via the precise manipulation of protein-protein interfaces. Despite the impressive array of protein cages produced in the laboratory, the design of inducible assemblies remains challenging(2,3). Here we demonstrate an ultra-stable artificial protein cage, the assembly and disassembly of which can be controlled by metal coordination at the protein-protein interfaces. The addition of a gold (I)-triphenylphosphine compound to a cysteine-substituted, 11-mer protein ring triggers supramolecular self-assembly, which generates monodisperse cage structures with masses greater than 2 MDa. The geometry of these structures is based on the Archimedean snub cube and is, to our knowledge, unprecedented. Cryo-electron microscopy confirms that the assemblies are held together by 120 S-Au(i)-S staples between the protein oligomers, and exist in two chiral forms. The cage shows extreme chemical and thermal stability, yet it readily disassembles upon exposure to reducing agents. As well as gold, mercury(II) is also found to enable formation of the protein cage. This work establishes an approach for linking protein components into robust, higher-order structures, and expands the design space available for supramolecular assemblies to include previously unexplored geometries.

MeV-SIMS is an emerging mass spectrometry imaging method, which utilizes fast, heavy ions to desorb secondary molecules. High yields and low fragmentation rates of large molecules, associated with the electronic sputtering process, make it particularly useful in biomedical research, where insight into distribution of organic molecules is needed. Since the implementation of MeV-SIMS in to the micro-beam line at the tandem accelerator of Jožef Stefan Institute, MeV-SIMS provided some valuable observations on the distribution of biomolecules in plant tissue, as discussed by Jencic et al. (Nucl. Inst. Methods Phys. Res. B. 371, 205-210, 2016; Nucl. Inst. Methods Phys. Res. B. 404, 140-145, 2017). However, limited focusing ability of the chlorine ion beam only allowed imaging at the tissue level. In order to surpass shortcomings of the existing method, we introduced a new approach, where we employ a continuous, low-current primary beam. In this mode, we bombard thin samples with a steady chlorine ion flux of approx. 5000 ions/s. After desorbing molecules, chlorine ions penetrate through the thinly cut sample and trigger the time-of-flight "start" signal on a continuous electron multiplier detector, positioned behind the sample. Such bombardment is more effective than previously used pulsing-beam mode, which demanded several orders of magnitude higher primary ion beam currents. Sub-micrometer focusing of low-current primary ion beam allows imaging of biological tissue on a subcellular scale. Simultaneously, new time-of-flight acquisition approach also improves mass resolution by a factor of 5. Within the article, we compare the performance of both methods and demonstrate the application of continuous mode on biological tissue. We also describe the thin sample preparation protocol, necessary for measurements with low primary ion currents.

As sessile organisms, plants must constantly adapt to environmental changes. Plant performance is related to availability of light, water, and mineral nutrients, which is especially important in agricultural systems, where minerals are removed from the fields at harvest. Mineral nutrients are also essential for the normal functioning of animal and human body. Since plants are world’s primary producers representing the basis for animal and human diet, it is of extreme importance that they contain sufficient amounts of mineral nutrients and low amounts of antinutrients and noxious elements. To maintain homeostasis with optimal cell functioning and integrity, proper allocation of elements at organ, tissue, cellular, and subcellular level is essential. Studies of element localization are therefore crucial to uncover the mechanisms of element transport and accumulation, toxicity, and tolerance. In the last two decades, a remarkable progress has been made in the development and application of different 2D imaging techniques in complex biological systems, especially with regard to improved lateral resolution and sensitivity as well as sample preparation. In this chapter, the latest development and applications of focused beam techniques like microparticle-induced X-ray emission (μ-PIXE), micro-X-ray fluorescence spectrometry (μ-XRF), and laser ablation inductively coupled plasma-mass spectrometry (LA-ICPMS) in imaging the distributions of major and trace elements in plants are presented. These techniques are unique in providing in situ information and, with appropriate sample preparation, provide results true to biological condition of the living plants. For example the methods for sampling and sample preparation can in conjunction with measurement conditions compromise the element distribution: critical consideration is therefore required when reporting “in vivo” elemental distribution and concentration.

International audience; The possible synergistic effect and mutual influence of the defect production by W-ion damaging and presence of hydrogen isotopes in the crystal lattice of tungsten is studied. For this purpose we perform modelling of the experimental data where samples were in one case sequentially damaged by W ions followed by D-atom exposure and in the other case simultaneously damaged by W ions and exposed to D atoms. Modeling is performed by the MHIMS (migration of hydrogen isotopes in materials) code in which a model of trap creation due to W-ion irradiation during the D-atom exposures is implemented. With the help of the surface model and the experimental data the migration barrier from the surface to the bulk is determined as a function of exposure temperature. It is shown that there is a temperature dependence of the migration barrier which at high temperatures and low hydrogen atom surface coverages stabilizes at 2 eV being in good agreement with the first-principle calculations. The evolution of trap concentrations with temperature is obtained from fitting the deuterium (D) depth profiles and the thermo-desorption spectra of deuterium gas at different damaging temperatures. In both experiments, the desorption peaks corresponding to induced trap defects are described by two trapping types with energies of 1.83 eV and 2.10 eV attributed to dislocation loops and cavities, respectively. The concentration of the low energy trap is higher in the case of simultaneous W/D exposure as compared to sequential W/D exposure experiment. This gives an unambiguous proof that the presence of deuterium does have an influence of defect evolution in tungsten material and therefore prevents spontaneous annihilation the traps.

Within the EUROfusion MST1 work package, a series of experiments has been conducted on AUG and TCV devices to disentangle the role of plasma fueling and plasma shape for the onset of small ELM regimes. On both devices, small ELM regimes with high confinement are achieved if and only if two conditions are fulfilled at the same time. Firstly, the plasma density at the separatrix must be large enough (), leading to a pressure profile flattening at the separatrix, which stabilizes type-I ELMs. Secondly, the magnetic configuration has to be close to a double null (DN), leading to a reduction of the magnetic shear in the extreme vicinity of the separatrix. To broaden our analytical capabilities with molecular imaging in addition to the existing elemental imaging with micro-PIXE, a linear Time-Of-Flight mass spectrometer for MeV Secondary Ion Mass Spectrometry (MeV-SIMS) was constructed and added to the existing nuclear microprobe at the Jožef Stefan Institute. We measured absolute molecular yields and damage cross-section of reference materials, without significant alteration of the fragile biological samples during the duration of measurements in the mapping mode. We explored the analytical capability of the MeV-SIMS technique for chemical mapping of the plant tissue of medicinal cannabis leaves. A series of hand-cut plant tissue slices were prepared by standard shock-freezing and freeze-drying protocol and deposited on the Si wafer. We show the measured MeV-SIMS spectra showing a series of peaks in the mass area of cannabinoids, as well as their corresponding maps. The indicated molecular distributions at masses of 345.5 u and 359.4 u may be attributed to the protonated THCA and THCA-C4 acids, and show enhancement in the areas with opened trichome morphology.

Background Phosphorus (P) deficiency limits crop production worldwide. Crops differ in their ability to acquire and utilise the P available. The aim of this study was to determine root traits (root exudates, root system architecture (RSA), tissue-specific allocation of P, and gene expression in roots) that (a) play a role in P-use efficiency and (b) contribute to large shoot zinc (Zn) concentration in Brassica oleracea. Results Two B. oleracea accessions (var. sabellica C6, a kale, and var. italica F103, a broccoli) were grown in a hydroponic system or in a high-throughput-root phenotyping (HTRP) system where they received Low P (0.025 mM) or High P (0.25 mM) supply for 2 weeks. In hydroponics, root and shoot P and Zn concentrations were measured, root exudates were profiled using both Fourier-Transform-Infrared spectroscopy and gas-chromatography-mass spectrometry and previously published RNAseq data from roots was re-examined. In HTRP experiments, RSA (main and lateral root number and lateral root length) was assessed and the tissue-specific distribution of P was determined using micro-particle-induced-X-ray emission. The C6 accession had greater root and shoot biomass than the F103 accession, but the latter had a larger shoot P concentration than the C6 accession, regardless of the P supply in the hydroponic system. The F103 accession had a larger shoot Zn concentration than the C6 accession in the High P treatment. Although the F103 accession had a larger number of lateral roots, which were also longer than in the C6 accession, the C6 accession released a larger quantity and number of polar compounds than the F103 accession. A larger number of P-responsive genes were found in the Low P treatment in roots of the F103 accession than in roots of the C6 accession. Expression of genes linked with “phosphate starvation” was up-regulated, while those linked with iron homeostasis were down-regulated in the Low P treatment. Conclusions The results illustrate large within-species variability in root acclimatory responses to P supply in the composition of root exudates, RSA and gene expression, but not in P distribution in root cross sections, enabling P sufficiency in the two B. oleracea accessions studied.

During tea preparation mineral elements are extracted from the dried leaves of tea (Camellia sinensis (L.) Kuntze) plants into the solution. Micro-particle induced X-ray emission was employed to investigate the spatial distribution of magnesium (Mg), calcium (Ca) and manganese (Mn) in the young and old leaves of tea plants grown in the absence and presence of aluminium (Al) in the substrate. Results revealed that in tea leaves the largest concentrations of Mg occurred in the epidermis, of Ca in oxalate crystals and of Mn in epidermis and oxalate crystals; there was a leaf-age effect on tissue-specific concentrations of Mg, Ca and Mn with all tissues of old leaves containing larger concentrations of Mg, Ca and Mn than young leaves; supplementation of substrate with Al reduced concentrations of Mg, Ca and Mn in the old leaves, and a link between the distribution of Mg, Ca and Mn in the tea leaves with the extraction efficiencies of these elements into the tea was possible. We conclude that old leaves of tea plants cultivated under conditions of low Al availability will have the largest concentrations of Mg, Ca and Mn and may represent most acceptable ingredient for the preparation of tea.

Halophytes are able to tolerate relatively high concentrations of hazardous metals in a growing substrate, what makes them suitable candidates for phytoremediation of metal-contaminated soils. In this work, we aimed to study the physiological responses of the halophyte Sesuvium portulacastrum L. to Ni, with main focus on Ni localization, compartmentation and ligand environment, to decipher Ni tolerance and toxicity mechanisms. Seedlings were grown in hydroponic nutrient solution containing 0, 25, 50 and 100 μM Ni as NiCl

Sesuvium portulacastrum, a halophyte with high tolerance to heavy metals like Cd, Pb and Ni is considered for phytoremediation of metal contaminated saline soils. The tolerance to a selected metal ion could, by hypothesis, be stimulated through in vitro adaptation and regeneration of the plant. Seedlings obtained by in vitro micro-propagation, were exposed to 0, 25 and 50 μM Ni, as NiCl2, in agar-based medium for 30 days. Growth parameters, plant water content, the concentration of photosynthetic pigments, proline and malondialdehyde (MDA) concentrations were determined. Nickel and nutrients distribution in leaves was studied by micro-Proton-Induced-X-ray-Emission (μ-PIXE). The results showed that Ni was mainly accumulated in vascular bundles, next in water storage tissues and chlorenchyma. Ni concentrations in chlorenchyma increased with increasing Ni in culturing medium, in direct relation to decrease of photosynthetic pigments and increase of oxidative stress. As compared to control plants, Ni induced substantial increase in MDA and proline accumulation. Plants exposed to 50 μM Ni accumulated up to 650 μg g−1 of Ni in the shoots, exhibiting chlorosis and necrosis and a drastically reduced plant growth. Perturbations in uptake and distribution of nutrients were observed, inducing mineral deficiency, probably through membrane leakage. The mineral nutrient disturbances induced by Ni could be highly implicated in the restriction of S. portulacastrum development under the acute 50 μM Ni level.

In this paper, we present the use of focused MeV ion beams to study the distribution of deuterium (D), boron (B) and nitrogen (N) on tungsten (W) samples exposed in the divertor region of the ASDEX Upgrade tokamak during 15N-seeded L-mode discharges in deuterium and during non-seeded H-mode discharges in helium. In both experiment samples of various surface roughness were exposed and analyzed: W coatings on milled or polished graphite substrates and bulk W samples, ranging from the roughest (milled) to the smoothest (bulk W), to study the effects of surface roughness on deposition profiles of D, B and N. In the case of samples from the 15N experiment, we found that D, N and B are distributed quite homogeneously over the sample on the micrometer scale with some small variation inside the analysed area. The surface densities show strong variations in the poloidal direction, with maximum values slightly above the strike point. The amounts of the retained (D, B, N) were strongly correlated with the surface roughness of the samples, being the highest on the rough samples. Samples originating from the He campaign showed inhomogeneous distribution of impurities with a distinct micro-scale structure, which is most pronounced on pre-damaged W samples, where a rough fuzz-like surface is created during the exposure in the GLADIS machine. We observe a distinct difference in behavior of deposited B profiles from both experiments with higher retention of B in the He experiment.

Samples with tungsten (W) and tungsten-yttrium (Y) coatings on molybdenum were exposed to plasmas simulating the divertor conditions of ITER. The exposed surfaces were studied using in-situ laser induced breakdown spectroscopy diagnostics (LIBS) and the results were compared to those obtained by other surface characterization methods. Our results show that LIBS is a reliable technique for in situ monitoring of erosion, deposition, and fuel retention processes under reactor-relevant conditions. In the regions of the highest gross erosion the thickness of the remaining tungsten layer is thinnest and at the central part of samples both mixing and deposition of components took place. LIBS also allowed reliable recording of D retention. D accumulation is influenced by the surface temperature as well as by the surface irregularities.

Elemental concentrations in micro-PIXE (Particle Induced X-ray Emission) maps of elements in biological tissue slices have been determined using auxiliary information on the sample matrix composition from EBS (Elastic Backscattering Spectroscopy) and STIM (Scanning Transmission Ion Microscopy). The thin sample approximation may be used for evaluating micro-PIXE data in cases, where X-ray absorption in the sample can be neglected and the mass of elements in a selected area can be estimated. The resulting sensitivity amounts to an impressive 10−12 g of the selected elements. Two cases are presented as examples. In the first, we determined the total mass of gold nanoparticles internalized by human monocyte-derived dendritic cells (MDDC). In the second, an inventory of the mass of elements in the micro-particulate material adsorbed at the wall of the lorica of the microzooplankton species Tintinnopsis radix has been created.

Diverse physiological, biochemical and morphological adaptations enable plants to survive in extreme saline environments where osmotic and ionic stresses limit growth and development. Halophytes are salt-tolerant plants that can withstand extraordinarily high levels of Na and Cl in their leaves. The tissue and cellular distribution patterns of salt ions can be linked to the underlying mechanisms of salt tolerance. Application of fast, reliable, multi-elemental and quantitative techniques such as micro-proton-induced X-ray emission (micro-PIXE) will significantly contribute to and accelerate studies of plant salt tolerance, especially as micro-PIXE also provides spatially resolved quantitative data for light elements, such as Na and Mg. The spatial concentration distributions of Na, Mg, Cl, K, P and S in leaves of four halophytes (Bassia indica, Atriplex prostrata, Spartina maritima and Limonium angustifolium) were determined using micro-PIXE, to study the salt-tolerance strategies of the selected halophytes. Different distribution patterns of the studied elements were seen in the leaves; however, in all four of these plant species, Na was excluded from photosynthetically active chlorophyl tissues. With the exception of L. angustifolium, Cl, P and S contents (representing chloride, phosphate and sulphate ionic forms, respectively) did not ensure charge balance in the leaves, which suggests other anionic compounds, such as nitrate and organic anions, have crucial roles in maintaining electroneutrality in these halophytes. By increasing soil salinisation worldwide, the possibility to reliably complement spatial distributions of Na, Mg, Cl, K, P and S with plant structural morphology will contribute significantly to our understanding of plant tolerance mechanisms at the tissue and cell levels. In addition, these kinds of studies are of particular value for designing crop plants with high salt tolerance and for the development of phytoremediation technologies.

Deuterium retention was for the first time measured in tungsten samples simultaneously irradiated by W ions and exposed to D atoms at five different temperatures from 450K to 1000K. In order to obtain information on the defect concentration, samples were afterwards exposed to D atoms at 600K to populate the created defects. The results were compared to different sequential damaging/exposure experiments. Synergistic effects were observed, namely, higher D concentrations were found in the case of simultaneous damaging and D-atom loading as compared to sequential damaging at elevated temperatures and populating the defects afterwards. However, the deuterium retention is still lower as compared to sequential damaging at room temperature and post-damaging annealing. The observations are explained by stabilization of defects by the presence of solute hydrogen in the bulk that would annihilate at high temperatures without the presence of hydrogen. Results of simultaneous W-ion damaging and D exposure at elevated temperatures were also compared to a sequential experiment of W-ion damaging at room temperature and then D-atom loading at high temperatures showing that thermal D de-trapping dominates deuterium retention at high temperatures. In the last few decades, research has increasingly been aimed at clarifying how root system architecture, physiology and function are related to environmental drivers. ‘Negative halotropism’ has been defined as the alteration of root growth direction to avoid salinity. We suggested that ‘positive halotropism’ may be found in halophytes relying on salinity for optimal growth. Investigating root structure of the halophyte Bassia indica (Wight) A. J. Scott, we have shown that positive halotropism can explain the growth of horizontal roots towards optimal salt concentrations along a soil salinity gradient. Here we tested three hypotheses. First, that development of B. indica roots depends on a trade-off between optimal nutrient supply and saline concentrations: results of split-root-experiment showed a preference for sand enriched with nutrients and poor in salts. Second, that shallow horizontal roots enable B. indica to forage for nutrient-rich patches. Results demonstrated that bulk elemental analysis was not consistent with tissue-specific elemental analysis, and this can be explained by substantial variability of element composition of particular root segments. Third, we hypothesised that B. indica redistributes water horizontally through shallow horizontal roots. Results showed that back flow of water from the tap root towards tip root was possible in horizontal roots in saline microenvironment.

This study aimed to relate the properties of incrusted plant tissues and structures as well as biomineral concentrations and localization with leaf reflectance and transmittance spectra from 280 nm to 880 nm in the grasses Phragmites australis, Phalaris arundinacea, Molinia caerulea and Deschampsia cespitosa, and the sedge Carex elata. Redundancy analysis revealed that prickle-hair length on adaxial surface and thickness of lower epidermis exerted significant effects in P. australis; prickle-hair density at abaxial leaf surface and thickness of epidermis on adaxial leaf surface in P. arundinacea; thickness of epidermis on adaxial leaf in D. cespitosa; prickle-hair density on adaxial leaf surface and thickness of cuticle in M. caerulea; and prickle-hair density on adaxial leaf surface and cuticle thickness of the lower side in C. elata. Micro-PIXE and LEXRF elemental localization analysis show that all of these structures and tissues are encrusted by Si and/or by Ca. Reflectance spectra were significantly affected by the Ca concentrations, while Si and Mg concentrations and the Ca concentrations significantly affected transmittance spectra. High concentrations of Mg were detected in epidermal vacuoles of P. arundinacea, M. caerulea and D. cespitosa. Al co-localises with Si in the cuticle, epidermis and/or prickle hairs.

The existing external beamline at the Microanalytical Center of the Jožef Stefan Institute (JSI) in Ljubljana has been currently upgraded in order to extend the capabilities towards in-air PIXE imaging across relatively large sample surface area (~ 1 cm2) with lateral resolution of sub 100 µm. For that purpose, a doublet of magnetic quadrupole lenses was installed on the beamline together with the newly designed exit nozzle with a 200 nm Si3N4 exit window resulting finally in ~ 50×50 mm2 lateral resolution. Additionally, an in-vacuum beam chopper was installed before the exit nozzle to measure the proton dose deposited on the sample. The beam characterization results are presented together with the first application from the field of biology, namely quantitative concentration maps of a longitudinal section of a whole thick wheat grain.

The increased modularity in the total hip arthroplasty (THA) allows adapting the prosthesis to the specific anatomical characteristics of each patient. However, the advantages of the modular THA are shadowed by the increased number of prosthesis failures observed in patients. The presence of junction in modular THA may increase its risk of mechanical failure. Moreover, the micro movements between neck and stem could lead to the production of metallic debris which may cause tissue inflammation and unsealing of prosthesis due to osseous dissolution. It is necessary to understand the mechanism of dispersion of the metal particles from the prosthesis into the tissue. Techniques currently applied in hospitals, such as X-ray scans or optical tissue microscopies, are able to distinguish metal particles, but unable to identify their specific metallic origin. In this work, within the TissueMaps project, we have proved that Proton Induced X-ray Emission (PIXE) is able to provide the distribution and elemental composition of particles from the prosthesis into the pseudo capsular tissue samples (near the femoral head) and identify the features observed under optical microscopy, in a case of broken neck prosthesis.
