Precise mosquito identification is integral to effective arbovirus surveillance. Nonetheless, the conventional morphological approach to identifying mosquito species is laborious, demands expertise and presents challenges when specimens are damaged. DNA barcoding offers a promising alternative, surmounting challenges inherent in morphological identification. To integrate DNA barcoding into arbovirus surveillance effectively, a robust dataset of mosquito barcode sequences is required. This study established a comprehensive repository of Cytochrome Oxidase I (COI) barcodes, encompassing 177 samples representing 45 mosquito species from southern and northern Western Australia (WA), including 16 species which have not been previously barcoded. The average intraspecific and interspecific genetic distances were 1% and 6.8%, respectively. Anopheles annulipes sensu lato had the highest intraspecific distance at 9.1%, signifying a genetically diverse species. While validating the potential of COI barcodes to accurately differentiate mosquito species, we identified that some species pairs have low COI divergence. This includes Aedes clelandi and Ae. hesperonotius, Tripteroides atripes and Tp. punctolaeralis and Ae. turneri and Ae. stricklandi. In addition, we observed ambiguity in identification of the members of Culex sitiens subgroup (Cx. annulirostris, Cx. palpalis and Cx. sitiens) and three members of Cx. pipiens complex (Cx. australicus, Cx. globocoxitus, Cx. quinquefasciatus). In summary, despite presenting challenges in the identification of some mosquito species, the COI barcode accurately identified most of the species and generated a valuable resource that will support the WA arbovirus surveillance program and enhance public health intervention strategies for mosquito‐borne disease control. Tephritis angustipennis (Diptera: Tephritidae) and Campiglossa loewiana (Diptera: Tephritidae) are phytophagous pests in China. Their damage has significantly impacted the collection and cultivation of germplasm resources of native Asteraceae plants. However, the genetic characteristics and structure of their population are unclear. This study focused on the highly damaging species of T. angustipennis and C. loewiana collected from the three-river source region (TRSR). We amplified the mitochondrial cytochrome C oxidase subunit I (mt COI) gene sequences of these pests collected from this area and compared them with COI sequences from GenBank. We also analyzed their genetic diversity and structure. In T. angustipennis , 5 haplotypes were identified from 5 geographic locations; the genetic differentiation between France population FRPY (from Nylandia, Uusimaa) and China populations GLJZ (from Dehe Longwa Village, Maqin County), GLDR (from Zhique Village, Dari County), and GLMQ (from Rijin Village, Maqin County) was the strongest. GLJZ exhibited strong genetic differentiation from GLDR and GLMQ, with relatively low gene flow. For C. loewiana , 11 haplotypes were identified from 5 geographic locations; the genetic differentiation between the Chinese population GLMQ-YY (from Yangyu Forest Farm, Maqin County) and Finnish population FDNL (from Nylandia, Uusimaa) was the strongest, with relatively low gene flow, possibly due to geographical barriers in the Qinghai–Tibet plateau. Only 1 haplotype was identified across GLDR, GLMQ, and GLBM. High gene flow between distant locations indicates that human activities or wind dispersal may facilitate the dispersal of fruit flies and across different geographic. Geostatistical analysis suggested a recent population expansion of these 2 species in TRSR. Our findings provide technical references for identifying pests in the TRSR region and theoretical support for managing resistance, monitoring pest occurrences, analyzing environmental adaptability, and formulating biological control strategies for Tephritidae pests on Asteraceae plants. Thrips are devastating pests for various crops, and they can rasp tender leaves, terminal buds, and flowers, which specifically causing huge economic losses to cotton production. However, there is very little knowledge about the species composition of thrips in mainland China, as well as the genetic structure of the thrips populations, particularly in the cotton-producing regions. In this study, thrips were collected from 40 geographical locations at 8 different provinces which representing majority cotton-producing belts in China, and mitochondrial cytochrome oxidase subunit I sequence was used to identify species composition and evaluate the genetic diversity of collected thrips individuals. Based on experimental results proven that overall, 10 and 8 species of thrips were identified in seedling and flowering stage respectively, which is corresponding dominant species are Thrip tabaci (Lindeman) and Frankliniella intonsa (Trybom). Genetically, 24 haplotypes were identified in 310 T. tabaci individuals from 10 locations, and 263 haplotypes were detected in 1,861 F. intonsa individuals from 40 locations. Hap1 (T. tabaci) is the most widely distributed haplotype among all the T. tabaci samples. Likewise, Hap 2 is the most widely distributed and abundant haplotype among all samples of F. intonsa. The genetic differentiation degrees of T. tabaci between SXYC population and other 9 populations were high, but its gene flow in these 10 regions was relatively low, which might be due to geographical barriers. The Mantel tests showed no correlation between genetic distance and geographic distance of the 2 thrip species. Demographic analysis results showed that both T. tabaci and F. intonsa experienced population expansion in China. Taken together, this study identifies the species composition of thrips in major cotton-producing regions at different growth periods and evaluates effects of geomorphology on the geographical distribution of haplotypes of dominant thrips T. tabaci and F. intonsa. The occurrence of Xiphinema species in the rhizosphere of vineyards showing general decline phenotype in North Italy was investigated. Most of the collected soil samples contained two Xiphinema species belonging to X. americanum-group, along with X. index. Using the integrative taxonomy approach, combining morphological, molecular and multivariate morphological analyses, the two populations of the X. americanum-group were identified as X. rivesi and X. pachtaicum. Furthermore, one male specimen of X. rivesi was recovered and the presence of three juvenile developmental stages was confirmed based on the scatter diagram. D2-D3 expansion domains of LSU, ITS region and partial COI were used to determine the phylogenetic relationships of the two Xiphinema species from North Italy with other species belonging to X. americanum-group. Five other Xiphinema populations from different geographical areas were included in this study and only molecularly identified as X. rivesi, X. pachtaicum and X. incertum by sequencing the ITS and D2-D3 expansion domains of LSU rDNA. Abstract Morphologically cryptic and pseudo-cryptic species pose a challenge to taxonomic identification and assessments of species diversity and distributions. Such is the case for the sea cucumber Stichopus horrens, commonly confused with Stichopus monotuberculatus. Here, we used mitochondrial cytochrome oxidase subunit I (COI) and microsatellite markers to examine genetic diversity in Stichopus cf. horrens throughout the Philippine archipelago, to aid species identification and clarify species boundaries. Phylogenetic analysis reveals two recently diverged COI lineages (Clade A and Clade B; c. 1.35–2.54 Mya) corresponding to sequence records for specimens identified as S. monotuberculatus and S. horrens, respectively. Microsatellite markers reveal two significantly differentiated genotype clusters broadly concordant with COI lineages (Cluster 1, Cluster 2). A small proportion of individuals were identified as later-generation hybrids indicating limited contemporary gene flow between genotype clusters, thus confirming species boundaries. Morphological differences in papillae distribution and form are observed for the two species, however tack-like spicules from the dorsal papillae are not a reliable diagnostic character. An additional putative cryptic species was detected within Clade B-Cluster 2 specimens warranting further examination. We propose that these lineages revealed by COI and genotype data be referred to as Stichopus cf. horrens species complex.

The cryptic invasion of golden apple snails (Pomacea canaliculata and P. maculata) in Taiwan has caused significant ecological and economical damage over the last few decades, however, their management remains difficult due to inadequate taxonomic identification, complex phylogeny, and limited population genetic information. We aim to understand the current distribution, putative population of origin, genetic diversity, and potential path of cryptic invasion of Pomacea canaliculata and P. maculata across Taiwan to aid in improved mitigation approaches. The present investigation conducted a nationwide survey with 254 samples collected from 41 locations in 14 counties or cities across Taiwan. We identified P. canaliculata and P. maculata based on mitochondrial COI and compared their genetic diversity across Taiwan, as well as other introduced and native countries (based on publicly available COI data) to understand the possible paths of invasion to Taiwan. Based on mitochondrial COI barcoding, sympatric and heterogeneous distributions of invasive P. canaliculata and P. maculata were noted. Our haplotype analysis and mismatch distribution results suggested multiple introductions of P. canaliculata in Taiwan was likely originated directly from Argentina, whereas P. maculata was probably introduced from a single, or a few, introduction event(s) from Argentina and Brazil. Our population genetic data further demonstrated a higher haplotype and genetic diversity for P. canaliculata and P. maculata in Taiwan compared to other introduced regions. Based on our current understanding, the establishment of P. canaliculata and P. maculata is alarming and widespread beyond geopolitical borders, requiring a concerted and expedited national and international invasive species mitigation program. Simple Summary: The mitochondrial COI gene as a molecular marker is widely used in genetic diversity and population genetic diversity analysis. In this study, mitochondrial COI was used to analyze the genetic diversities of eight populations of Nerita yoldii along the coast of China. The results showed that N. yoldii has a high level of genetic diversity, with genetic structure differences observed between the Xiapu population and six other populations, excluding the Taizhou population. The results of neutral tests and nucleotide mismatch analyses indicate a historical demographic expansion in the N. yoldii population. Our study results provide foundational data for the conservation and sustainable utilization of N. yoldii resources. Nerita yoldii is a euryhaline species commonly found in the intertidal zone. To investigate the genetic diversity of 233 N. yoldii individuals from eight locations along the coast of China, we utilized the mitochondrial COI gene as a molecular marker. A total of 34 haplotypes were detected, exhibiting a mean haplotype diversity (Hd) of 0.5915 and a mean nucleotide diversity (Pi) of 0.0025, indicating high levels of genetic diversity among all populations. An analysis of molecular variance (AMOVA) indicated that the primary source of genetic variation occurs within populations. In addition, neutral tests and mismatch analyses suggested that N. yoldii populations may have experienced bottleneck events. Moderate genetic differentiation was observed between Xiapu and other populations, excluding the Taizhou population, and may be attributed to the ocean currents. Intensively studying the genetic variation and population structure of N. yoldii populations contributes to understanding the current population genetics of N. yoldii in the coastal regions of China. The Atlantic surfclam (Spisula solidissima solidissima) is an economically valuable clam species that supports a major US fishery. Until recently, fishery efforts along the southern edge of the surfclam range were low due to clam mortalities there in the 1990s. Recent surfclam fishing efforts off Virginia raised questions of whether the surfclam population has returned in the southern region or if a single cohort is supporting the fishery there. Questions have also arisen about whether S. s. similis is among the population fished off the coast of VA. Spisula solidissima similis is a warm-water cryptic subspecies of the Atlantic surfclam. Although morphologically indistinguishable, S. s. similis grows to a smaller size and is genetically distinct. Atlantic surfclams (n = 103) were collected from the fishing grounds off the coast of VA. Each surfclam was aged, and shell length and tissue weight recorded for comparison to surfclams of the same age from the center of the population. Analyses of mitochondrial (mtCOI) sequences suggests that the two groups sampled off VA are genetically homogeneous, both groups contain two divergent mitochondrial lineages, and one surfclam sampled shares the S. s. similis mtCOI sequence. There are multiple cohorts of surfclams, suggesting that environmental conditions may have improved for surfclams in the south, or that this population has acclimated to altered conditions. Simple Summary: Thrips, the notorious sap-sucking insects, serve as the vector of plant viruses. Their accurate species identification is essential for determining the vector species and implementing successful pest management techniques. Mitochondrial COI (DNA barcode) sequence variation has proven to be effective in identifying species in many insect pest groups. In this study, we identified 19 thrips species that were found on different host plants in Bangladesh. Among the 19 species, we identified four prominent vector species (Frankliniella intonsa, Thrips tabaci, Scirtothrips dorsalis and T. palmi) and one significant pollinator, Microcephalothrips abdominalis. The findings presented here emphasize the importance of conducting DNA barcoding, analyzing population structure, and assessing the genetic diversity of thrips species in this region. This research is crucial for comprehending their host preferences, potential for adaptation, and genetic variation at both local and regional levels. Accurate identification of pests and invasive species is essential for implementing effective control and quarantine measures. Although few studies on thrips species have been conducted in Bangladesh, the variation and genetic diversity of thrips species remain unknown. Propylea japonica (Coleoptera: Coccinellidae) is a natural enemy insect with a wide range of predation in Chinese mainland and is commonly used in pest management. However, its genetic pattern (i.e., genetic variation, genetic structure, and historical population dynamics) is still unclear, impeding the development of biological control of insect pests. Population genetic research has the potential to optimize strategies at different stages of the biological control processes. This study used 23 nuclear microsatellite sites and mitochondrial COI genes to investigate the population genetics of Propylea japonica based on 462 specimens collected from 30 sampling sites in China. The microsatellite dataset showed a moderate level of genetic diversity, but the mitochondrial genes showed a high level of genetic diversity. Populations from the Yellow River basin were more genetically diverse than those in the Yangtze River basin. Propylea japonica has not yet formed a significant genealogical structure in China, but there was a population structure signal to some extent, which may be caused by frequent gene flow between populations. The species has experienced population expansion after a bottleneck, potentially thanks to the tri-trophic plant–insect–natural enemy relationship. Knowledge of population genetics is of importance in using predators to control pests. Our study complements existing knowledge of an important natural predator in agroecosystems through estimating its genetic diversity and population differentiation and speculating about historical dynamics.

The Asian citrus psyllid (ACP) is the main vector of Citrus Huanglongbing, the most damaging citrus disease, causing significant financial losses in the citrus industry. Global warming has expanded the habitat of this pest, allowing it to continue its northward migration to China. Population genetic information of ACP is fundamentally essential for species management. This study investigated the genetic diversity and population structure of Chinese ACP using the mitochondrial cytochrome oxidase subunit I gene by dataset comprised 721 sequences from 27 geographic sites in China. Low haplotype diversity (0.323 ± 0.022) and low nucleotide diversity (0.00071 ± 0.00007) were observed in the entire population, which may indicate recent founder events. Twenty-three haplotypes were identified and clustered into 2 haplogroups: haplogroup I and haplogroup II. Haplogroup II included only 2 unique haplotypes, which occurred exclusively in the Southwest China ACP population. Genetic differentiation analyses were also indicative of Southwest China population was significantly differentiated from the remaining populations. Demographic history analysis showed that ACP population in China has experienced demographic expansion. Aim: To investigate the drivers of intra‐specific genetic diversity in centipedes, a group of ancient predatory soil arthropods. Location: Asia, Australasia and Europe. Time Period: Present. Major Taxa Studied: Centipedes (Class: Chilopoda). Methods: We assembled a database of 1245 mitochondrial cytochrome c oxidase subunit I sequences representing 128 centipede species from all five orders of Chilopoda. This sequence dataset was used to estimate genetic diversity for centipede species and compare its distribution with estimates from other arthropod groups. We studied the variation in centipede genetic diversity with species traits and biogeography using a beta regression framework, controlling for the effect of shared evolutionary history within a family. Results: A wide variation in genetic diversity across centipede species (0–0.1713) falls towards the higher end of values among arthropods. Overall, 27.57% of the variation in mitochondrial COI genetic diversity in centipedes was explained by a combination of predictors related to life history and biogeography. Genetic diversity decreased with body size and latitudinal position of sampled localities, was greater in species showing maternal care and increased with geographic distance among conspecifics. Main Conclusions: Centipedes fall towards the higher end of genetic diversity among arthropods, which may be related to their long evolutionary history and low dispersal ability. In centipedes, the negative association of body size with genetic diversity may be mediated by its influence on local abundance or the influence of ecological strategy on long‐term population history. Species with maternal care had higher genetic diversity, which goes against expectations and needs further scrutiny. Hemispheric differences in genetic diversity can be due to historic climatic stability and lower seasonality in the southern hemisphere. Overall, we find that despite the differences in mean genetic diversity among animals, similar processes related to life‐history strategy and biogeography are associated with the variation within them. Since the early 1990s in South Korea, climatic and anthropogenic factors have incurred the reduction of the wild seeds of the Pacific oyster, Crassostrea gigas, which raised concerns about losing genetic diversity and accelerating genetic deterioration. We assessed the genetic diversity of C. gigas populations from two farms (Tongyeong and Gadeokdo) on the southern coast, where about 80% of the cultivated oysters in Korea are produced. Tongyeong showed slightly higher diversity than Gadeokdo, but both populations had a similar genetic structure characterized by low nucleotide diversity. Comparative haplotype analyses provided data supporting genetic features of the populations that include (1) weak genotype-locality relationship, (2) low levels of gene flow between populations, and (3) possible seasonal fluctuation of genetic variation within a population. Furthermore, the highly alike haplotype network patterns were observed between the wild and farm populations as well as among the populations in neighboring countries, which suggests that the genetic structure is conserved between wild and hatchery populations, and geographic proximity has minimal influence on the genetic composition. Simple Summary: Blue oat mite species mainly include Penthaleus major, P. falcatus, P. tectus, and P. minor. Among them, P. major, P. falcatus and P. tectus are important pest mites on gramineous crops, which often occur in low-temperature and high-humidity environments. These three mites are difficult to distinguish in the field due to their small size and similar shape. Furthermore, they exhibit different resistance to chemical pesticides, making it challenging to control their outbreak. Worryingly, we only saw reports of frequent occurrence of P. major in winter wheat regions in China, and no other species of blue oat mites were found. Understanding the distribution pattern, genetic diversity, and genetic differentiation among populations of blue oat mite species in winter wheat regions of China is essential for developing effective control programs. In this study, we evaluated the distribution of blue oat mites in the major wheat regions in China and assessed the level of genetic diversity and genetic structure of blue oat mites from winter wheat regions of China using mitochondrial cytochrome c oxidase I (COI) sequences. Blue oat mite species, including Penthaleus major and P. tectus, are pests widely distributed across China that cause damage to winter wheat. This study evaluated the genetic diversity of P. major and P. tectus on Triticum hosts collected from 23 geographic locations based on mitochondrial cytochrome c oxidase subunit I (COI) sequences. We identified nine haplotypes in 438 P. major individuals from 21 geographic locations and five haplotypes in 139 P. tectus individuals from 11 geographic locations. Meanwhile, P. major exhibits high values of haplotype diversity (Hd) and nucleotide diversity (Pi) (Hd = 0.534 > 0.5 and Pi = 0.012 > 0.005), representing a large stable population with a long evolutionary history. P. tectus shows low values of Hd and Pi (Hd = 0.112 < 0.5 and Pi = 0 < 0.005), which suggest recent founder events. Moreover, demographic analysis suggested that P. major and P. tectus have not undergone a recent population expansion. The lowest genetic variation was observed in Xiangzhou (XZ-HB), Zaoyang (ZY-HB), Siyang (SY-JS), and Rongxian (RX-SC), with only one species and one haplotype identified in over 30 individuals. As a wild resource, the large yellow croaker Larimichthys crocea has been seriously threatened since the mid-1980s. Owing to the implementation of protection measures, such as the establishment of a protection zone, fishing prohibitions, restocking programs and successful mariculture, its resources have gradually recovered year by year. Limited by the low spatial resolution and incomplete spatial coverage of sampling stations, the spatial genetic structure and diversity of large yellow croakers from the southern Yellow Sea and north-central East China Sea remains unclear. In order to evaluate the genetic diversity status of this wild stock, 22 wild sites were collected from the southern Yellow Sea and north-central East China Sea and analyzed by investigating genetic variability and its population structure using mitochondrial COI sequence in this study. Among the 662 sequences, a total of 71 different haplotypes were defined. The haplotype diversity (h) and nucleotide diversity (π) values were 0.644~1.000 and 0.00220~0.00473 respectively. The highest h and π occurred in the southern Yellow Sea (YS). AMOVA analysis showed no genetic differentiation among those 22 sites after Bonferroni correction. By comparing with previous studies, the croaker has maintained relatively steady genetic diversity in recent years. Our result also suggested that the croakers in the South Yellow Sea and north-central East China Sea belonged to the same group. Thus, they can be released as a management unit without regard for heterogenicity among those in the sea area. Simple Summary: We studied small, dully coloured grass-miner moths (Elachistidae, Elachistinae). This is a large subfamily widely distributed throughout the world, but only a few species are known in the Neotropics. To facilitate further exploration, we reviewed one of nineteen species group within the largest genus, Elachista, along with identification keys of species. The E. praelineata species group now comprises of 37 described species, which are distributed in the Americas, Africa, Asia, and Australia. Neotropical species of the group are still underexplored, especially compared with that of Palearctic and Nearctic regions. So far only five species are known to belong to this group, mainly from the Ecuadorian Andes. In this study, we aimed to review and update information on the E. praelineata species group in the Neotropics. Here, we describe in detail a new species Elachista stonisi sp. nov. from western Bolivia. The female of E. albisquamella Zeller, 1877 is described for the first time and the female of E. lata Sruoga, 2010 is associated with the male. The description of new species and the association of females with males are supported morphologically and are confirmed by analysis of partial sequences of mitochondrial COI. Neotropical species of the Elachista praelineata species group are reviewed. Five species are recognized in the Neotropics. A new species, Elachista stonisi sp. nov., and female of E. albisquamella Zeller, 1877 are described and illustrated with photographs of the adults, some of the immature stages, female genitalia, and leaf mines. The present study aimed to investigate the prevalence and pathology of Sacculina beauforti infection on box crab Calappa lophos in the Southeast coast of India from January to December 2019-2021. About 340 Sacculina infested calappa lophos were collected and the overall prevalence was recorded as 13.08%. Among the four seasons, the maximal infection was recorded in summer 2020 (23.21%) and the minimum infection was noted in pre- monsoon (11.52%). Male and female crab was not significantly different in case of infection rate (p>0.05). The externae on crab was identified as Sacculina beauforti. Histological sections of S.beauforti infected hosts showed vacuolization, elongated haemocytes on hepatopancrease. Here, we report the first record of quagga mussel, Dreissena bugensis Andrusov, 1897, in Italy. This mollusc is native of the Dnieper River drainage of Ukraine and is one of the most aggressive invaders in freshwater ecosystems. The spread of quagga mussels throughout Europe and Northern America begun during the 1940s and 1989, respectively. Individuals of quagga mussel were identified in February and March 2022 in two stations located in the shallow (Bardolino) and deeper basins (Castelletto di Brenzone) of Lake Garda. Samples, collected with an Eckman grab, allowed identifying individuals of quagga mussel attached on macrophytes, stones and other mollusc shells. The identification of the individuals was carried out both through the analysis of morphological characters and genetic and phylogenetic analyses using the mitochondrial COI gene. The discovery of quagga mussel is the last of a long series of reports of non-indigenous species introduced into Lake Garda. Lacking direct connection by rivers or canals with other waterbodies colonized by D. bugensis at the northern side of the Alps, a more probable cause of introduction of this new species into Lake Garda can be due to unintentional overland transport through recreational boats and fishing gear. With this new discovery, Lake Garda has confirmed its pivotal role as a southern Alpine corridor for the introduction of non-indigenous species previously established at the northern border of the Alps. Considering its high colonization rate and compared with other invasion patterns observed in Europe and North America, it can be assumed that D. bugensis will soon establish itself with dominant populations throughout Lake Garda. During a nematode survey in Iran, an abundant population of sheathoid, migratory, root-ectoparasitic nematodes was recovered from a tea, Camellia sinensis (L.), Kuntze plantation for the first time. Morphological and molecular characterization identified the Iranian population as Hemicriconemoides kanayaensis . The morphometrics of H. kanayaensis agreed with the original description. Phylogenetic relationships within Hemicriconemoides -based on ITS region, D2 to D3 expansion regions of the 28S rRNA, and the partial 18S rRNA genes along with the partial mitochondrial COI gene-confirmed the occurrence of H. kanayaensis on the tea plantation in Iran. Melampus " sincaporensis ," Auriculastra " duplicata ", and Ellobium chinense are Red-Listed snail species of the family Ellobiidae that co-occur on salt marshes in mainland Japan. Here, we report the genetic diversity and population structures of the former two taxa in comparison with our previous data on E. chinense for the evaluation of connectivity and conservation values of their local populations. Analyses of 655-bp or 652-bp sequences of the mitochondrial cytochrome c oxidase subunit I gene showed the highest genetic diversity and panmictic structure for M. sp. cf. sincaporensis throughout its geographic range in Japan, whereas they showed the highest level of genetic subdivision for A. sp. cf. duplicata . Our laboratory observation of egg masses and planktotrophic larvae of A. " duplicata " and reference to previous ontogenetic data for the species of Melampus suggested differences in their fecundity and pelagic larval duration, which apparently have led to the contrasting levels of population differentiation in the study species. Particular need of conservation efforts was identified for the isolated population of A. " duplicata " in the Ise-Mikawa Bay area to avoid local extinction and shrinking of the species' geographic range. In addition, we present molecular and morphological evidence that individuals of A. " duplicata " from mangrove swamps in the northern part of Okinawa Island represent an independent, surviving lineage of a different species.

The brown planthopper, Nilaparvata lugens (Stål) is a major sucking insect pest of rice. This insect has long been considered as migratory; however, its route in India is still unknown. Hence, to find out its migration route genetic diversity, genetic structure and gene flow of 16 N. lugens populations from major rice growing regions of India was studied based on mitochondrial cytochrome oxidase I (COI). The results revealed a high genetic homogeneity among the populations on the basis of genetic diversity statistics and neutrality tests. There was a prevalence of a single major haplotype across the country. No spatial relevance was found with the genetic structure of the populations indicating presence of excessive gene flow among them. Extensive gene flow among populations was also confirmed with the presence of higher number of immigrants in North, Central, and East India. To further clarify the migration sources, 48 h air-mass reverse trajectory was performed for Varanasi just aftermath of cyclones Amphan and Yaas, which disclosed Eastern/Northeastern states along with Bangladesh and Myanmar as the possible source areas. Overall, the results revealed a single panmictic homogeneous population of N. lugens in India with extensive gene flow as a consequence of their migration. Heterodera avenae, a globally distributed plant-parasitic nematode, is one of the most significant pests on cereal crops. Iii China, it is widely distributed in cereal-growing areas of 16 provinces anc! causes serious Yelyield losses. In the present study, a total of 98 populations of H. avenae were collected from major wheat-growing regions in China and six other countries. The mitochondrial COI genes were amplified and analyzed. Forty-one mitochondrial COI haplotypes were identified, suggesting a prehigh genetic diversity and endemism level of H. avenae in China. Phylo-genetic analysis showed that H, avenae populations in China were divided into four clades. Significant evolutionary and genetic differences were found between Chinese (except Hubei) and foreign populations. Hap l, the most widely distributed haplotype, was considered to be a sep - arate evolutionary origin in China. The gene flow of H m'enae from the northwestern region to the north China region and Huang -Huai - Hai region was significant, so as the direction between north China and Huang-Huai-Hai region. We speculate that water flowing from the Yelyield low River and mechanical harvesters promoted gene exchange among these groups. A distance-based redundancy analysis showed that genetic distances observed among H avenae populations were explained foremost not only by geographic distance but also by temperature and prehigh cipitation. Stored grain insect pests are of economic importance, and they spread rapidly through the grain trade. Hence, it is important to identify these pests accurately. Although several morphological methods exist, it is tedious and timeconsuming. DNA barcoding using mitochondrial COI is an alternative approach that aids in precise species identification. In this study, 13 stored grain insect pests belonging to the order Coleoptera and Lepidoptera were collected from different storage structures in Coimbatore. A fragment of mt-COI was amplified and sequenced. Sequence analyses were carried out with BOLD (Barcode of Life Data System) and ABGD (Automatic Barcode Gap Discovery) tools. The barcoding gap analysis revealed that the inter-species genetic distance is greater than the intraspecies genetic distance. The ABGD analysis for species delimitation partitioned the Coleopteran and Lepidopteran datasets into 9 and 4 putative species, respectively. The barcode gap was absent in more closely related species. However, analysis of their sequences revealed significant variations. Our results showed the ability of the mt-COI to discriminate between the species, thus provide a complementary technique for the diagnosis of stored grain insect pests. [ABSTRACT FROM AUTHOR]

Grouper aquaculture is rapidly expanding in both tropical and subtropical regions. The presence of marine leeches (Pterobdella arugamensis; previously named Zeylanicobdella arugamensis) infesting cultured groupers, however, can have a fatal effect on grouper aquaculture production and cause significant economic loss. Understanding the marine leech's population structure is therefore important to determine its possible distributional origin and distributional mechanisms, which will help monitor and mitigate the infestation. In this study, a total of 84 marine leeches collected from cultured hybrid groupers Epinephelus spp. in Brunei Darussalam, Malaysia and Indonesia were identified as P. arugamensis, based on morphological and mitochondrial cytochrome c oxidase subunit I gene sequence analyses. These leech samples, together with additional sequences from the GenBank database, were grouped into four genetically distinct haplogroups: (1) Asia Pacific, (2) Borneo, (3) Surabaya and (4) Iran. The four populations were found to be highly diverged from each other. The results also suggested that the samples from the Asia Pacific population could be dispersed and transported from Indonesia. Simple Summary: The bioconversion of organic waste into valuable insect protein as an alternative animal feed ingredient has the potential to improve agricultural sustainability and may become a key element of future circular economy. However, while insects farmed for feed production are considered livestock from a regulatory perspective, systematic linking of genetic resource characterisations and fundamental phenotyping, crucial for precision breeding and feeding schemes, remains scarce even for prime insect candidates, such as the black soldier fly (BSF). The present study initiated to fill this knowledge gap by experimentally assessing BSF genotype-by-diet interactions for a number of economically and ecologically relevant larval phenotypic traits. Besides pervasive diet effects, strong impact of BSF genetic background and ubiquitous environment-mediated interactions were found. This implies some of the so-far unexplained response variation across global BSF studies could be driven by previously neglected mechanisms of genetic specificity, and thus that the concept of broad conspecific plasticity in this insect is likely too simplistic. Instead, it is emphasised that matching BSF genetics to dietary contexts is vital for purposive production optimisation, particularly when extrapolated to large-scale operations. These insights highlight that establishing tailored BSF breeding as an independent branch offers veritable opportunities to efficiently support this growing agricultural sector. Further advancing black soldier fly (BSF) farming for waste valorisation and more sustainable global protein supplies critically depends on targeted exploitation of genotype-phenotype associations in this insect, comparable to conventional livestock. This study used a fully crossed factorial design of rearing larvae of four genetically distinct BSF strains (FST: 0.11–0.35) on three nutritionally different diets (poultry feed, food waste, poultry manure) to investigate genotype-by-environment interactions. Phenotypic responses included larval growth dynamics over time, weight at harvest, mortality, biomass production with respective contents of ash, fat, and protein, including amino acid profiles, as well as bioconversion and nitrogen efficiency, reduction of dry matter and relevant fibre fractions, and dry matter loss (emissions). Virtually all larval performance and body composition traits were substantially influenced by diet but also characterised by ample BSF genetic variation and, most importantly, by pronounced interaction effects between the two. Across evaluated phenotypes, variable diet-dependent rankings and the lack of generally superior BSF strains indicate the involvement of trade-offs between traits, as their relationships may even change signs. Conflicting resource allocation in light of overall BSF fitness suggests anticipated breeding programs will require complex and differential selection strategies to account for pinpointed trait maximisation versus multi-purpose resilience. Abstract Background The white-backed planthopper (WBPH), Sogatella furcifera (Horváth) (Hemiptera, Delphacidae), is a migratory pest of rice in Asia. Shandong Province, in northern China, is located on the migration pathway of WBPH between southern and northeast China. The potential sources of WBPH in northern China are poorly understood. We studied the sources of WBPH in Shandong Province by determining the population genetic structure of WBPH in 18 sites distributed in Shandong and in six regions of the Greater Mekong Subregion (GMS). We used mitochondrial gene and single-nucleotide polymorphism (SNP) markers for analysis. Results All of the WBPH populations studied in the seven regions had low genetic diversity. Pairwise FST values based on mtDNA ranged from − 0.061 to 0.285, while FST based on SNP data ranged from − 0.007 to 0.009. These two molecular markers revealed that 4.40% (mtDNA) and 0.19% (SNP) genetic variation could be explained by the interpopulation variation, while the rest came from intrapopulation variation. The populations in the seven geographic regions comprised four hypothetical genetic clusters (K = 4) not associated with geographic location. Eighty-four of 129 individuals distributed across the given area were designated as recent migrants or of admixed ancestry. Although the substantial migration presented, a weak but significant correlation between genetic and geographic distances was found (r = 0.083, P = 0.004). Conclusion The Greater Mekong Subregion was the main genetic source of WBPH in Shandong, while other source populations may also exist. The genetic structure of WBPH is shaped by both migration and geographic barriers. These results help clarify the migration route and the source of WBPH in northern China.

Based on research carried out from 2016 to 2018 there are twenty-six species of aphids of the tribe Eulachnini in Lithuania. Seventeen are members of the subgenus Cinara of the genus Cinara, three of the subgenus Cupressobium and two of the subgenus Schizolachnus. In addition, there are four species of the genus Eulachnus. Summarized information on the distribution and host specificity of Eulachnini in Lithuania is presented. Nine species were in all climatic regions of Lithuania: C. (Cinara) brauni, C. (Cinara) hyperophila, C. (Cinara) neubergi, C. (Cinara) piceicola, C. (Cinara) pinea, C. (Cinara) pini, C. (Cinara) pruinosa, C. (Cupressobium) cupressi and C. (Cupressobium) juniperi. Five species of Lithuanian Eulachnini inhabit host plants of the genus Picea, three live on Larix, thirteen on Pinus, one on Abies, one on Thuja and three on Juniperus. Cinara (Cinara) piniphila was recorded on Pinus mugo and C. (Cinara) nuda on Pinus heldreichii for the first time. Partial COI and EF-1α sequences of C. (Cinara) hyperophila, C. (Cinara) pilosa and C. (Cinara) piceicola were obtained for the first time. Partial sequences of mitochondrial (COI) and nuclear (EF-1α) DNA of Lithuanian samples were used to explore molecular diversity using NJ trees, Automatic Barcode Gap Discovery (ABGD), General Mixed Yule Coalescent (GMYC) and Poisson Tree Processes (PTP). Species delimitation using GMYC (both on COI and EF-1α sequences), PTP (BI on COI) and ABGD (EF-1α) were the most consistent with traditional classifications. Pairwise between-species sample divergences (COI and EF-1α fragments) of the samples of the species complex C. (Cinara) pinea - C. (Cinara) piniphila indicate it is composed of a single species. Of the species of aphids that live on conifers, there are seven species of the tribe Eulachnini that are likely to shortly colonize Lithuania.

Blue oat mite species, including Penthaleus major and P. tectus, are pests widely distributed across China that cause damage to winter wheat. This study evaluated the genetic diversity of P. major and P. tectus on Triticum hosts collected from 23 geographic locations based on mitochondrial cytochrome c oxidase subunit I (COI) sequences. We identified nine haplotypes in 438 P. major individuals from 21 geographic locations and five haplotypes in 139 P. tectus individuals from 11 geographic locations. Meanwhile, P. major exhibits high values of haplotype diversity (Hd) and nucleotide diversity (Pi) (Hd = 0.534 > 0.5 and Pi = 0.012 > 0.005), representing a large stable population with a long evolutionary history. Aim: Effective control of invasive species and conservation of native biodiversity requires accurate species identification. Several species of apple snails (Ampullariidae: Pomacea) from South America have become widespread pests in Asia since their introduction in the early 1980s, but their taxonomic uncertainty has hindered our understanding of the invasive processes. We aim to determine the identity and distribution of Pomacea species in Hong Kong, which has been known as a stepping stone of species invasion. Location: Hong Kong. Methods: We collected 162 apple snails from five freshwater wetlands of Hong Kong, sequenced the mitochondrial COI, genotyped the nuclear EF1α and built a COI dataset of 1378 sequences of apple snails from mainland China, Malaysia, Argentina and Brazil. We identified the species, determine the introgression pattern and analysed their population structures. Results: We found the co‐occurrence of P. canaliculata, P. maculata and P. occulta in Hong Kong for the first time. Pomacea canaliculata represented the dominant species (85% of collected specimens). The three Pomacea species from Hong Kong had low genetic diversity. High genetic similarity existed between P. canaliculata populations of Hong Kong and Malaysia, P. occulta populations of Hong Kong and mainland China and P. maculata populations of mainland China and Malaysia. Our samples contained a high proportion of hybrids. Main conclusions: Apple snails may have been introduced into Hong Kong multiple times: both P. canaliculata and P. occulta likely came from Argentina; P. maculata came from both Argentina and Brazil. Genetic drift largely explains the differentiation of Pomacea spp. among regions. Introgressive hybridization may have contributed to population fitness by increasing genetic variation to neutralize the founder effect. The knowledge on species identity, distribution and genetic diversity of apple snails from this study will contribute to better understanding their invasive mechanisms and prevention of their continued spread in Asia. Several European freshwater crayfish species are currently included in one of the IUCN Red list categories. In the Iberian Peninsula, the native Austropotamobius pallipes species complex (the white clawed crayfish, WCC) has experienced a drastic decline since 1973. Implementing conservation management strategies for this species requires a better understanding of the patterns and structure of its genetic diversity. In this study, we assessed the levels and patterns of genetic variation in 71 populations along the whole distributional range of the WCC in the Iberian Peninsula. The two mitochondrial markers analyzed (Cytochrome Oxidase subunit I and 16S rRNA genes) indicated high levels of genetic diversity, which are significantly geographically structured in three main genetic groups, two corresponding to Northern and one to Central-Eastern and the westernmost Iberian Peninsula. The diversity found included new private haplotypes, and revealed the potential effect of paleogeographic barriers and last glaciations in the population structure observed. Current conservation and management programs for the WCC in the Iberian Peninsula should consider these three phylogeographic groups as essential management units in order to preserve the remaining genetic diversity in the species.

Competition between Kuril harbor seals (Phoca vitulina stejnegeri) and salmon set-net fishing industries has become a serious problem with the recent increase in the number of seals in Erimo, Hokkaido, Japan. We aimed to understand the detailed dietary structure of Kuril harbor seals focusing on intraspecific differences and verify whether "problem seals" who habitually use salmon set-nets could be characterized by intrinsic factors such as sex and maturity. We estimated the diet of Kuril harbor seals in two fishing seasons using DNA barcoding diet analysis on colon contents and verified intraspecific differences in their diet. In spring, their diets showed different tendencies between maturity stage; each adult seal fed on different prey items, suggesting that they avoid the intraspecies competition over food during the breeding season. Additionally, it was implied that some adult females habitually stole from salmon set-nets. Our dietary analysis showed dietary changes of Kuril harbor seals with different tendencies depending on maturity or sex, suggesting that problem individuals who habitually use set-nets can be characterized by intrinsic factors. This detailed dietary information can offer an accurate assessment of seal predation effects on fishing targets and the selective management of Kuril harbor seals, especially in mitigating seal–commercial fishery conflicts. The Clarion-Clipperton Zone (CCZ), located in the abyssal equatorial Pacific, has been subject to intensive international exploration for polymetallic nodule mining over the last four decades. Many studies have investigated the potential effects of mining on deep-sea ecosystems and highlighted the importance of defining environmental baseline conditions occurring at potential mining sites. However, current information on biodiversity and species distributions in the CCZ is still scarce and hampers the ability to effectively manage and reduce the potential impacts of mining activities. As part of the regulatory regimes adopted by the International Seabed Authority, concession holders are required to conduct an environmental impact assessment and gather baseline data on biodiversity and community structure in relation to their license areas. In the present study, we used an integrative molecular and morphological approach to assess species richness and genetic variation of deep-sea scavenging amphipods collected in two nodule-mining exploration areas (UK-1 and OMS-1 areas) and one Area of Particular Environmental Interest (APEI-6) in the eastern part of the CCZ. We analyzed the DNA sequences of the cytochrome c oxidase subunit I gene of 645 specimens belonging to ten distinct morphospecies. Molecular data uncover potential cryptic diversity in two investigated species, morphologically identified as Paralicella caperesca Shulenberger & Barnard, 1976 and Valettietta cf. anacantha (Birstein & Vinogradov, 1963). Our study highlights the importance of using molecular tools in conjunction with traditional morphological methods for modern biodiversity assessment studies, particularly to evaluate morphologically similar individuals and incomplete specimens. The bronze bug, Thaumastocoris peregrinus, is an endemic Australian eucalypt pest that has become a serious threat to the global forestry industry. Following a large outbreak within its native range in 2002, introduced populations of the bronze bug appeared in South Africa and Argentina. Since then, T. peregrinus has spread rapidly around the world, with the number of reported incursions increasing steadily. Despite the problems posed by the bronze bug, little is known about its global invasion pathways. In this study, we used multiple different population genetic methods to (1) gain new insight into aspects of bronze bug population history within Australia, and (2) characterise the spread of the bronze bug throughout South America, South Africa, New Zealand and the Mediterranean Basin. Taken together, the genetic data and the outbreak records suggest that, across the international regions surveyed, at least three separate introductions of T. peregrinus from Australia have occurred over the decade 2003–2012—one into South Africa, another into Argentina and a third into New Zealand—each of which is likely to have originated in Sydney. Populations in Europe and Israel appear to have become established from individuals introduced from South America, rather than from within the native Australian range, suggesting the existence of a bridgehead effect. These findings provide an important framework for understanding the global spread of invasive bronze bug populations. Revealing cryptic diversity constitutes the backbone of the future identification and description of a new lineage. For the genus Ophisops (Lacertidae), previous studies indicated that this genus is characterized by cryptic diversity as three clades were obtained for O. elegans and O. occidentalis that do not fit the classical systematics. Notably, we were interested to delineate the distribution range of the two clades of O. elegans described in Iran. We sequenced 65 individuals of Ophisops mainly from northwestern Iran for one mitochondrial (cytochrome c oxidase subunit 1 [COI]; 686 base pairs [bp]) and three nuclear genes (R35, MC1R, and PKM2; 1,857 bp). Phylogeographic analysis from mitochondrial and nuclear genes confirmed that Iranian samples belong to two major haplogroups (divergence of 13% for COI) that are also split into several subclades (divergence of 6%–10% for COI), revealing an unsuspected diversity within Iranian Ophisops. Divergence dating and biogeographical analysis indicated that most clades arose through vicariance and dispersal processes during the Mio‐Pliocene (between 7.2 and 1 Myr). However, a scenario of expansion/regression is also advocated for explaining the distribution and contact between three subclades in North Iran. We propose to recognize the three major clades as three potential candidate species. Moreover, we found some correspondence between several phylogenetic clades or subclades identified and six of the nine subspecies described for O. elegans. Both species and subspecies delimitation and identification would deserve additional investigations (including morphology, ecology, biogeography, and behavior) to fulfill the conditions of integrative taxonomy. Leafhoppers (Cicadellidae) occur in all biogeographical regions and feed on a wide range of host plants. Classic identification of leafhoppers is based only on the morphology of male genitalia, while DNA barcoding is an effective tool to identify different leafhopper species in any stage of their life cycle. Phylogenetic relationships among leafhoppers remain unexplored, but recent phylogenies have begun to interpret the evolutionary relationships of previously recognized tribes and subfamilies. Here we used mitochondrial Cytochrome Oxidase I sequences to barcode and determine the phylogenetic relationship among eight leafhopper genera collected from Egypt. Eight leafhopper samples from eight different genera were collected from various plants using a sweeping net and aspirator. Males of seven species were available while females only were available from the one genus (Austroagallia sp.). Novel leafhopper-specific mtDNA COI degenerate primers were designed. Our results showed that the mean of intraspecific COI sequence divergence in leafhoppers was 0.45%, with a range from 0.15% to 1.23%, and the divergences of most species were over 2%. For the first time, we were exploring the evolutionary relationships among surveyed genera (Austroagallia, Empoasca, Eupteryx, Exitianus, Macrosteles, Nephotettix, and Orosius) using a robust phylogenetic supertree. Based on the original research of 2017–2018, also reference data, four aphid species and five species complexes of the family Adelgidae listed for Lithuanian fauna. Pineus (Pineus) strobi (Hartig, 1839) is a new species for the fauna of Lithuania. Summarized information on the distribution and host specificity of adelgid species in Lithuania is presented. The most common species recorded from all geographical regions of Lithuania were the ones belonging to the species complexes Adelges laricis – A. tardus, Pineus orientalis – P. pini, Sacchiphantes viridis – S. abietis and Gilletteella cooleyi – G. coweni. Six species of Adelgidae inhabit host plants of the genus Picea, three live on Larix, three – on Pinus, two – on Abies, one – on Pseudotsuga. Partial sequences of mitochondrial (COI) and nuclear (EF-1α) DNA of Lithuanian samples were analysed in comparison with available data from GeneBank representing samples from Central Europe. Pairwise distances of COI and EF-1α fragments between samples belonging to species complexes Adelges laricis – A. tardus, Dreyfusia nordmannianae – D. piceae, Sacchiphantes viridis – S. abietis, Gilletteella cooleyi – G. coweni and Pineus orientalis – P. pini suggest each species complex being a single species. The Bemisia tabaci complex in Ecuador was studied with respect to phylogenetic relationships and eco-geographical distribution. Whitefly samples were collected from natural and agricultural environments in nine provinces of Ecuador (latitude, 2°N–5°S; longitude, 78°–81°W). Mitotypes were identified based on phylogenetic analysis of the 30-mtCOI-tRNAleuregion (832 bp) and corrected pairwise distance analysis. The distribution of mitotypes was modeled using MaxEnt, and their predicted niches were characterized according to environmental gradients. Four B. tabacimitotypes were identified, of which three are endemic, herein ECU1–3, and the other is the introduced B mitotype. Mitotypes ECU1 (44%), ECU2 (0.74%), and ECU3 (1.47%) grouped in the American Tropics (AMTROP) species and diverged by as much as 10%, which was higher than previous estimates for the AMTROP clade of 7–8.6%. Although haplotypes of ECU1 and ECU2 are known from the American Tropics, this is the first report of the ECU3 mitotype, which may possibly be restricted to southern Ecuador. The distribution of the three ECU-endemic mitotypes spanned the high-altitude niches of the western slope of the Andes, rich in microclimates with variable temperature and humidity conditions. The non-endemic B mitotype (47%) occurred only in the irrigated cropping systems located in hot and/or dry-tropical ecological niches. Of the endemic mitotypes, ECU1 occupied the most ecological niches. Among variables contributing to ECU1 and B mitotype niche range assignments, the most significant to influence ecological range was rainfall. The B. tabaciendemic to Ecuador were more diverse with respect to mtCOI-tRNAleu sequence than previously known, and occupied distinct microclimate niches suggestive of ecological resilience. Based on research carried out from 2016 to 2018 there are twenty-six species of aphids of the tribe Eulachnini in Lithuania. Seventeen are members of the subgenus Cinara of the genus Cinara, three of the subgenus Cupressobium and two of the subgenus Schizolachnus. In addition, there are four species of the genus Eulachnus. Summarized information on the distribution and host specificity of Eulachnini in Lithuania is presented. Nine species were in all climatic regions of Lithuania: C. (Cinara) brauni, C. (Cinara) hyperophila, C. (Cinara) neubergi, C. (Cinara) piceicola, C. (Cinara) pinea, C. (Cinara) pini, C. (Cinara) pruinosa, C. (Cupressobium) cupressi and C. (Cupressobium) juniperi. Five species of Lithuanian Eulachnini inhabit host plants of the genus Picea, three live on Larix, thirteen on Pinus, one on Abies, one on Thuja and three on Juniperus. Cinara (Cinara) piniphila was recorded on Pinus mugo and C. (Cinara) nuda on Pinus heldreichii for the fi rst time. Partial COI and EF-1a sequences of C. (Cinara) hyperophila, C. (Cinara) pilosa and C. (Cinara) piceicola were obtained for the fi rst time. Partial sequences of mitochondrial (COI) and nuclear (EF-1a) DNA of Lithuanian samples were used to explore molecular diversity using NJ trees, Automatic Barcode Gap Discovery (ABGD), General Mixed Yule Coalescent (GMYC) and Poisson Tree Processes (PTP). Species delimitation using GMYC (both on COI and EF-1a sequences), PTP (BI on COI) and ABGD (EF-1a) were the most consistent with traditional classifi cations. Pairwise between-species sample divergences (COI and EF-1a fragments) of the samples of the species complex C. (Cinara) pinea - C. (Cinara) piniphila indicate it is composed of a single species. Of the species of aphids that live on conifers, there are seven species of the tribe Eulachnini that are likely to shortly colonize Lithuania. [ABSTRACT FROM AUTHOR]

Domestication of animal species is often associated with a reduction in genetic diversity. The honey bee, Apis mellifera Linnaeus, 1758, has been managed by beekeepers for millennia for both honey and wax production and for crop pollination. Here we use both microsatellite markers and sequence data from the mitochondrial COI gene to evaluate genetic variation of managed A. mellifera in Thailand, where the species is introduced. Microsatellite analysis revealed high average genetic diversity with expected heterozygosities ranging from 0.620 ± 0.184 to 0.734 ± 0.071 per locus per province. Observed heterozygosities were generally lower than those expected under Hardy–Weinberg equilibrium, both locally and across the population as a whole. Mitochondrial sequencing revealed that the frequency of two evolutionary linages (C—Eastern European and O—Middle Eastern) are similar to those observed in a previous survey 10 yr ago. Our results suggest that Thai beekeepers are managing their A. mellifera in ways that retain overall genetic diversity, but reduce genetic diversity between apiaries. [ABSTRACT FROM AUTHOR]
