Stroke and traumatic injury in brain or spinal cord are often life-threating conditions and major causes of death or permanent disability with high impact in the health care system. There are several stages of intervention to improve the neurological outcome. Acutely, fast interventions aiming to reestablish cerebral blood flow in ischemic stroke, to stop bleeding after brain hemorrhage, and to reduce edema after contusions are amongst mandatory actions. Current studies aim to develop accompanying strategies for brain cell protection based on enhancing endogenous protective mechanism, blocking cell death pathways, or through immunomodulation. After the acute phase, interventions are intended to promote recovery of function using rehabilitation with state-of-the-art technologies enabled by robotics. Other advanced strategies include cell, gene, and immune therapies, and brain function modulation with the aid of smart nanotechnologies. There is great expectation in the fast evolving novel approaches for improvement of neurological deficits in these unpredictable and devastating condition

The release of RNA-containing extracellular vesicles (EV) into the extracellular milieu has been demonstrated in a multitude of different in vitro cell systems and in a variety of body fluids. RNA-containing EV are in the limelight for their capacity to communicate genetically encoded messages to other cells, their suitability as candidate biomarkers for diseases, and their use as therapeutic agents. Although EV-RNA has attracted enormous interest from basic researchers, clinicians, and industry, we currently have limited knowledge on which mechanisms drive and regulate RNA incorporation into EV and on how RNA-encoded messages affect signalling processes in EV-targeted cells. Moreover, EV-RNA research faces various technical challenges, such as standardisation of EV isolation methods, optimisation of methodologies to isolate and characterise minute quantities of RNA found in EV, and development of approaches to demonstrate functional transfer of EV-RNA in vivo. These topics were discussed at the 2015 EV-RNA workshop of the International Society for Extracellular Vesicles. This position paper was written by the participants of the workshop not only to give an overview of the current state of knowledge in the field, but also to clarify that our incomplete knowledge – of the nature of EV(-RNA)s and of how to effectively and reliably study them – currently prohibits the implementation of gold standards in EV-RNA research. During immunologic synapse (IS) formation, human CD38 redistributes to the contact area of T cell-antigen-presenting cell (APC) conjugates in an antigen- dependent manner. Confocal microscopy showed that CD38 preferentially accumulated along the contact zone, whereas CD3-ζ redistributed toward the central zone of the IS. APC conjugates with human T cells or B cells transiently expressing CD38-green fluorescent protein revealed the presence of 2 distinct pools of CD38, one localized at the cell membrane and the other in recycling endosomes. Both pools were recruited to the T/APC contact sites and required antigen-pulsed APCs. The process appeared more efficient in T cells than in APCs. CD38 was actively recruited at the IS of T cells by means of Lck-mediated signals. Overex- pression of CD38 in T cells increased the levels of antigen-induced intracellular calcium release. Opposite results were obtained by down-regulating surface CD38 expression by means of CD38 siRNA. CD38 blockade in influenza HA-specific T cells inhibited IL-2 and IFN-γ production, PKCe phosphorylation at Thr538, and PKC6 recruitment to the IS induced by antigen-pulsed APCs. The vertical muon intensity measured with MACRO at the Gran Sasso Laboratory, in the depth range 3200-7000 hg cm(-2) of standard rock, corresponding to 2-200 TeV/nucleon primary energy range, is presented. An estimation of the muon energy spectrum at the surface will also be shown. The results will be compared with those of other experiments and with the computation of the intensities obtained using a full Monte Carlo calculation with different hypotheses for the primary cosmic ray composition.

We describe the techniques chosen to search tor magnetic monopoles using the MACRO streamer tube sub-system. The hardware and the details of the analysis procedures will be discussed also. We report on the first measurement of the flux of upgoing muons resulting from interactions of atmospheric neutrinos in the rock below MACRO. The ratio of the observed to the expected number of events integrated over all nadir angles is 0.73 ± .09 stat. ± .06 sys. ± .12 theor. . The flux of upgoing muons as a function of nadir angle is presented and compared to Monte Carlo expectations. At the 90% confidence level, the data are consistent with no neutrino oscillations or some possible oscillation hypothese with the parameters suggested by the Kamiokande contained-event analysis.

The vertical underground muon intensity has been measured in the slant depth range 3200-7000 hg cm-2 (standard rock) with the completed lower part of the MACRO detector at the Gran Sasso laboratory, using a large sample of data. These observations are used to compute the surface muon flux and the primary ''all-nucleon'' spectrum. An analysis of systematic uncertainties introduced by the interaction models in the atmosphere and the underground propagation of muons is presented. A comparison of our results with published data is also presented. © 1995 The American Physical Society.

A search for slowly moving magnetic monopoles in the cosmic radiation was conducted from October 1989 to November 1991 using the large liquid scintillator detector subsystem of the first supermodule of the MACRO detector at the Gran Sasso underground laboratory. The absence of candidates established an upper limit on the monopole flux of 5.6×10-15 cm-2 sr-1 s-1 at 90% confidence level in the velocity range of 10-4

In the past decade, extracellular vesicles (EVs) have been recognized as potent vehicles of intercellular communication, both in prokaryotes and eukaryotes. This is due to their capacity to transfer proteins, lipids and nucleic acids, thereby influencing various physiological and pathological functions of both recipient and parent cells. While intensive investigation has targeted the role of EVs in different pathological processes, for example, in cancer and autoimmune diseases, the EV-mediated maintenance of homeostasis and the regulation of physiological functions have remained less explored. Here, we provide a comprehensive overview of the current understanding of the physiological roles of EVs, which has been written by crowd-sourcing, drawing on the unique EV expertise of academia-based scientists, clinicians and industry based in 27 European countries, the United States and Australia. This review is intended to be of relevance to both researchers already working on EV biology and to newcomers who will encounter this universal cell biological system. Therefore, here we address the molecular contents and functions of EVs in various tissues and body fluids from cell systems to organs. We also review the physiological mechanisms of EVs in bacteria, lower eukaryotes and plants to highlight the functional uniformity of this emerging communication system.

Multimuon data from the MACRO experiment at Gran Sasso have been analysed using a new method which allows one to estimate the primary cosmic ray fluxes. The estimated all-particle spectrum is higher and flatter than the one obtained from direct measurements but is consistent with EAS measurements. The average mass number shows a weak dependence on the primary energy below 10^{6} GeV. Even within large uncertainties our data support a possible increase of the average mass number at higher energies.

The MACRO underground experiment has started a systematic experimental study of the structure of a very high multiplicity event sample, with the aim of gaining a better understanding of the features of the high energy showers. We present here two new analyses: a) the analysis of the distribution of the separation between pairs of high multiplicity muons (''decoherence'' function); b) the search for clusters of muons within high multiplicity events. The first results are discussed together with a comparison between the real data and the Monte Carlo predictions.
