The purpose of this study was to study and analyze the composition and content of routine nutrients, amino acids, fatty acids, and mineral elements in muscle of three cultured populations of Cipangopaludina cathayensis, so as to provide a reference for the development and utilization of Cipangopaludina cathayensis germplasm resources. A total of 180 Cipangopaludina cathayensis with an average weight of (32.12±0.17) g were randomly divided into three groups with 60 Cipangopaludina cathayensis in each group. After shelling, the muscles were taken to test their nutritional components. The results showed that the crude protein, total amino acids, essential amino acids, and delicious amino acids in muscle of Luchuan County population were significantly higher than those of other populations (P<0.05). The amino acid score, chemical score, and essential amino acid index of Liujiang population were the highest. The contentrations of unsaturated fatty acids and polyunsaturated fatty acids in muscle of Luchuan County population were significantly higher than those of other populations, and the atherosclerosis index (IA) and thrombosis index (IT) were lower than those of other populations (P<0.05). The ratio of sigma-n-3/sigma-n-6PUFA of the three populations was in line with the recommended value of FAO/WHO for human diet (0.1~0.2). The contentrations of P and Ca in muscle of Rongshui County population were significantly higher than those of other populations (P<0.05). The study indicates that the muscle protein content of the three cultured populations is higher, the fat content is lower, and the delicious amino acids and unsaturated fatty acids are higher. The microbial priming effect—the decomposition of soil organic carbon (SOC) induced by plant inputs—has long been considered an important driver of SOC dynamics, yet we have limited understanding about the direction, intensity, and drivers of priming across ecosystem types and biomes. This gap hinders our ability to predict how shifts in litter inputs under global change can affect climate feedbacks. Here, we synthesized 18,919 observations of CO2 effluxes in 802 soils across the globe to test the relative effects (i.e., log response ratio [RR]) of litter additions on native SOC decomposition and identified the dominant environmental drivers in natural ecosystems and agricultural lands. Globally, litter additions enhanced native SOC decomposition (RR = 0.35, 95% CI: 0.32–0.38), with greater priming effects occurring with decreasing latitude and more in agricultural soils (RR = 0.43) than in uncultivated soils (RR = 0.28). In natural ecosystems, soil pH and microbial community composition (e.g., bacteria: fungi ratio) were the best predictors of priming, with greater effects occurring in acidic, bacteria‐dominated sandy soils. In contrast, the substrate properties of plant litter and soils were the most important drivers of priming in agricultural systems since soils with high C:N ratios and those receiving large inputs of low‐quality litter had the highest priming effects. Collectively, our results suggest that, though different factors may control priming effects, the ubiquitous nature of priming means that alterations of litter quality and quantity owing to global changes will likely have consequences for global C cycling and climate forcing. Aim: Fresh carbon (C) inputs to the soil can have important consequences for the decomposition rates of soil organic matter (priming effect), thereby impacting the delicate global C balance at the soil–atmosphere interface. Yet, the environmental factors that control soil priming effect intensity remain poorly understood at a global scale. Location: Global. Time period: 1980–2020. Methods: We conducted a global dataset of CO2 effluxes in 711 pairwise soils with 13C or 14C simple C sources inputs and without C inputs from incubation experiments in which isotope‐labelled C was used to quantify fresh C‐induced rather than exudate‐induced priming. Results: Soil priming effect intensity is predominantly positive. Soil texture and C content were identified as the most important factors associated with priming effects, with sandy soils from tropical and mid‐latitudes supporting the highest soil priming effect intensity, and soils with greater C content and fine textures from high latitudes maintaining the lowest soil priming effects. The negative association between C content and soil priming effect intensity was also indirectly driven by changing mean annual temperature, net primary productivity, and fungi : bacteria ratio. Using this information, we generated a global map of soil priming effect intensity, and found that the priming was lower at high latitudes and higher at lower latitudes. Main conclusions: Global patterns of soil priming effect intensity can be predicted using environmental data, with soil texture and C content playing a predominant role in explaining in priming effects. These effects were also indirectly driven by climate, vegetation and soil microbial properties. We present the first global atlas of soil priming effect intensity and advance our knowledge on the potential mechanisms underlying soil priming effect intensity, which are integral to improving the climate change and soil C dynamics components of Earth System models. Straw return has been widely used to increase SOC storage globally, but such C accrual is frequently constrained by other nutrients which are needed to maintain soil ecological stoichiometry balance. However, the impact of fertilization on SOC stocks under straw return remains unexplored at broad spatial scales. We constructed a national-scale dataset to explore the fertilization effect on SOC stocks (0–20 cm) under straw return and identified the dominant drivers and pathways in upland and paddy systems across China. On average, straw return enhanced SOC stocks and sequestration rate by 1.74 Mg C ha−1 and 0.27 Mg C ha−1 y−1 relative to straw removal (CK). Yet, straw co-applied with nutrient enrichment further increased SOC stock and sequestration rate by 0.81 Mg C ha−1 and 0.12 Mg C ha−1 y−1, compared to straw return only. These results indicated a general nutrient limitation on SOC sequestration under straw return. The fertilization effect on SOC stocks varied greatly across nutrient enrichment types and cropping systems, with greater soil C gain occurring under balanced fertilization (i.e., NPK) and more in uplands than in paddy fields. In the uplands, SOC sequestration with fertilization was positively correlated with increased belowground biomass. In paddy fields, however, improved biomass with fertilization did not translate into increased SOC accumulation, owing to the co-limitations of soil acidification and initial SOC. We further clarified the differential pathways by which chemical fertilization altered SOC stocks, i.e., productivity pathway in uplands but decomposition pathway in paddy fields. These mechanistic insights on the fertilization control over SOC can help us optimize the context-specific integrated straw and fertilization management strategies for greater SOC sequestration, and also inform the development of soil C projections by incorporating fertilization management into biogeochemical models in agroecosystems. Purpose. To study the corneal morphology and biomechanics in cases of small incision lenticule extraction with prophylactic accelerated collagen cross-linking (SMILE Xtra). Methods. This study was a retrospective study. 28 eyes of 14 patients with moderate-high risk of postoperative ectasia according to the Randleman scoring system underwent SMILE Xtra procedure. Outcome data were recorded including uncorrected distance visual acuity (UDVA), manifest refraction spherical equivalent (MRSE), surface regularity index (SRI), surface asymmetry index (SAI), simulated keratometry (SimK), posterior axial curvature (PAC), anterior and posterior corneal elevations (ACE and PCE), central corneal thickness (CCT), corneal resistance factor (CRF), corneal hysteresis (CH), and cornea-compensated intraocular pressure (IOPcc). The follow-up period was 12 months. Results. There were 28, 26, 22, 12, and 10 eyes enrolled at postoperative 1st day and 1st, 3rd, 6th, and 12th months, respectively. The UDVA improved from 1.27 ± 0.18 logMAR preoperatively to -0.06 ± 0.04 logMAR postoperatively (P < 0.05). The MRSE improved from -5.05 ± 1.15 D preoperatively to -0.14 ± 0.30 D postoperatively (P < 0.05). SAI, SimK, PAC, PCE, and CCT all changed significantly at 1st month postoperatively (P < 0.05) and stabilized during the remainder of the follow-up (P > 0.05). There was no significant change in SRI or ACE before and after surgery (P > 0.05). CRF, CH, and IOPcc all decreased significantly at 1st month postoperatively (P < 0.05) and remained stable afterwards (P > 0.05). Conclusions. The changes in the corneal morphology and biomechanics remained stable after SMILE Xtra, and there was no sign of postoperative ectasia or refractive regression. Single infrared (IR) image super‐resolution methods can help to reduce the cost and difficulty in manufacturing IR sensors for the imaging system. However, the deep learning‐based image SR methods need to build a complex network and thus consume a lot of computational power, which limits the application of SR technology on devices with low computing resources in practice. To solve this problem, the authors present a lightweight multi‐path feature fusion network (MFFN) for the single infrared (IR) image SR. A multi‐path feature fusion block (MFFB) is developed to extract and fuse multiple and discriminative features in a recursive feedback way. Specifically, the multiple features are refined via the linear feature extraction branch, shared‐source residual feature extraction branch, and channel attention branch in MFFB. Finally, the authors reconstruct the high‐resolution IR images from the low‐resolution counterpart based on the refined multiple features. The experimental results demonstrate that MFFN achieves high‐quality single infrared image SR and shows superiority over previous methods for several scale factors (e.g. ×2, ×3, and ×4). Background:Candida albicans catheter-related infection (CRI) is a great challenge in clinic now, mainly due to the difficulty in eradicating the biofilms. Purpose: In this study, the mechanism of the antibiofilm effect of myricetin (MY) on C. albicans was illustrated. A film forming system (FFS) containing MY and miconazole nitrate (MN) was developed, optimized, and evaluated. The anti-infection effect of MY+MN@FFS against C. albicans CRI was investigated in vivo. Study Design and Methods: To clarify the mechanism of the action of MY, the influence of MY on each key process of the formation of C. albicans biofilms was evaluated. To deliver MY and MN into the skin and form a drug reservoir on the surface of the skin, the FFS was used as a carrier and MY+MN@FFS was developed, optimized, and evaluated. After preliminary confirmation of drug safety, a percutaneously inserted C. albicans CRI mouse model was established to investigate the in vivo anti-infection effect of MY+MN@FFS by fluorescence microscopy and scanning electron microscopy on the outer surface of the catheters, hematoxylin/eosin staining, and periodic acid-Schiff staining of the mice skin tissues. Results: MY was found to inhibit the morphological transition of C. albicans and the secretion of exopolysaccharides, resulting in a reduction in biofilms. MY+MN@FFS exhibited excellent properties and no irritation to mice skin. In an in vivo anti-infection study, MY+MN@FFS exhibited an excellent preventive effect against percutaneously inserted C. albicans CRI. In this study, we studied the fraction of cooperators in the public goods game, taking into account the memory effect that affects the strategy updating. Unlike previous studies where an agent learned the opponent's last strategy based on their last payoffs, agents with memory in this study choose to cooperate according to opponents' effective strategies by comparing their effective payoffs based on payoffs and strategies in their memories. The effective payoff of an agent is the weighted average of previous strategies in the agent's memory. The weight is the decay measuring the significance of previous strategies and former payoffs are less significant than latter ones upon the future strategy. And it is the same with the effective strategy. By this means, when the effective payoff and the effective strategy share a same memory length and a same set of decay, the numerical simulation shows increasing the memory length or a homogeneous decay promotes cooperation among agents. However, it is a surprise that the effective payoff and the effective strategy have opposite effects. Homogeneous payoff weights lead to a higher fraction of cooperators, while heterogeneous strategy weights favors the cooperation, especially when agents only consider the latest strategy. Comparing the effect of memorizing payoffs and strategies, the effect of memorizing payoffs plays a dominant role. Furthermore, when the total memory length is limited, agents should memorize as many historical payoffs as possible. In addition the qualitative result above is independent of the rational noise. • Homogeneous weights on historic payoffs promote the cooperation level most. • Agents only need to remember components' latest strategy. • Long memory with rational agents promotes the cooperation. Purpose. The purpose of this study was to compare femtosecond laser-assisted in situ keratomileusis with prophylactic cross linking (FS-LASIK Xtra) and small-incision lenticule extraction with prophylactic cross linking (SMILE Xtra) in terms of their postoperative clinical outcomes. Methods. In this retrospective study, 24 patients (48 eyes) with myopia and myopia astigmatism were recruited from 2017 to 2018. All patients underwent comprehensive ophthalmic examinations preoperatively and follow-up visits at one and three months. Four patients (eight eyes) in each group were followed up for 12 months. The following were assessed at each visit: uncorrected distance visual acuity (UDVA), manifest refractive spherical equivalent (MRSE), keratometry values, biomechanical properties, anterior and posterior curvature, and corneal pachymetry. Results. The FS-LASIK Xtra and SMILE Xtra groups both included 24 eyes of 12 patients. At 1, 3, and 12 months after surgery, a UDVA of 20/20 or better was achieved for 91.7%, 91.7%, and 87.5% of individuals, respectively, in the FS-LASIK group and 95.8%, 100%, and 100% in the SMILE group, respectively. For 75% of eyes in the FS-LASIK Xtra group and 100% of eyes in the SMILE Xtra group, the achieved MRSE was within ±1.00D of attempted correction. The LASIK Xtra group had a significantly higher mean MRSE than the SMILE Xtra group at 3 and 12 months after surgery (P = 0.006 , 0.01), and the SMILE Xtra group had significantly higher K1 and K2 values than the FS-LASIK group at one month after surgery (P = 0.024 , 0.023). Corneal thickness decreased significantly at one month postoperatively and started to increase over the next 12 months in both groups (P = 0.001). The biomechanical properties showed no significant intergroup differences at the 12-month follow-up. Conclusions. FS-LASIK Xtra and SMILE Xtra are safe and effective in the correction of myopia and myopia astigmatism, and both procedures have the same effect on postoperative corneal morphology and biomechanics. This study assesses the sustainable development of countries worldwide and the impact of renewable energy on country-level sustainability for the period 1990–2014. The nonparametric method "data envelopment analysis" is employed to measure the performance of the countries by merging environmental and economic elements into unified sustainability measures. Reflecting the different attitudes with which the countries treat environmental and economic factors in pursuing sustainability, the assessment procedure assigns priority to either economic or environmental course. We find that if economic criterion is prioritized over environmental criterion, then the developed countries significantly outperform the developing countries until early 2000s. However, the gap quickly diminishes after the period. If environmental criterion takes precedence, then developed and developing countries have similar performance over the entire horizon. Moreover, no matter which criterion is prioritized, dispersion in sustainability levels for countries in different regions has shrunk over time. Panel analysis shows that the share of renewable energy in total energy consumption has positive and strongly significant impact on sustainability, no matter whether economic prosperity or environmental protection is the priority. Background: Drug-resistance and severe side effects of chemotherapeutic agents result in unsatisfied survival of patients with lung cancer. CXCLs/CXCR2 axis plays an important role in progression of cancer including lung cancer. However, the specific anti-cancer mechanism of targeting CXCR2 remains unclear. Methods: Immunohistochemical analysis of CXCR2 was performed on the microarray of tumor tissues of clinical lung adenocarcinoma and lung squamous cell carcinoma patients. CCK8 test, TUNEL immunofluorescence staining, PI-Annexin V staining, β-galactosidase staining, and Western blot were used to verify the role of CXCR2 in vitro. Animal models of tail vein and subcutaneous injection were applied to investigate the therapeutic role of targeting CXCR2. Flow cytometry, qRT-PCR, enzyme-linked immunosorbent assay (ELISA), and immunohistochemistry analysis were performed for further mechanistic investigation. Results: The expression of CXCR2 was elevated in both human lung cancer stroma and tumor cells, which was associated with patients' prognosis. Inhibition of CXCR2 promoted apoptosis, senescence, epithelial-to-mesenchymal transition (EMT), and anti-proliferation of lung cancer cells. In vivo study showed that tumor-associated neutrophils (TANs) were significantly infiltrate into tumor tissues of mouse model, with up-regulated CXCLs/CXCR2 signaling and suppressive molecules, including Arg-1 and TGF-β. SB225002, a selective inhibitor of CXCR2 showed promising therapeutic effect, and significantly reduced infiltration of neutrophils and enhanced anti-tumor T cell activity via promoting CD8+ T cell activation. Meanwhile, blockade of CXCR2 could enhance therapeutic effect of cisplatin via regulation of neutrophils infiltration. Conclusions: Our finds verify the therapeutic effects of targeting CXCR2 in lung cancer and uncover the potential mechanism for the increased sensitivity to chemotherapeutic agents by antagonists of CXCR2. We have developed and integrated a mobility-enhanced FET and a wakeup-free ferroelectric (FE) capacitor using Sn-doped InGaZnO (IGZTO) and demonstrated 1T1C FeRAM cell operation for 3-D embedded RAM application. IGZTO FET can achieve >20 cm2/V $\cdot $ s mobility with an ultrathin channel, which is $2\times $ higher than InGaZnO (IGZO) FET. The physics of mobility enhancement in oxide semiconductor FET with IGZTO channel is investigated by both experimental and simulation methods. The FE capacitor with large remanent polarization and low-temperature process at 400 °C is achieved. We have also studied the impact of thin-film access transistors on 1T1C cell operation with gate voltage dependence and capacitor size dependence. SPICE simulation is used for an extremely scaled device that achieves ~ns operation. BACKGROUND: Ridge–furrow mulching farming systems (RFMs) aim to increase field productivity and improve water use efficiency. To explore environment‐friendly and efficient farming systems is a central aspect of rainfed wheat field management in Kenya where rainfall utilization is at a low level. We introduced RFMs (including plastic film and grass straw mulching) to semiarid Kenya to evaluate the effects on field productivity, rainwater utilization, soil quality and economic profitability using old and modern wheat cultivars from 2012 to 2013. RESULTS: Across the cultivars, the RFMs increased grain yield, aboveground biomass and water use efficiency by 74–163%, 36–104% and 89–273%, respectively, compared with conventional flat planting (control). RFMs significantly shortened the vegetative period while prolonging the reproductive period. The net economic output under RFMs was 74–165% higher than that of the control. Grass straw mulching achieved the highest economic output to input ratio, almost 45% higher than plastic film mulching, despite the former harvested only 82% of the maximum field productivity of the latter. Compared with the control, grass straw mulching promoted the contents of soil organic carbon, total nitrogen and C:N ratio by 14%, 8% and 5%, respectively, while obviously decreased values of these parameters were observed under plastic mulching. CONCLUSIONS: Through reducing soil water loss, and improving rainwater use efficiency and soil quality, ridge–furrow grass straw mulching would be a sustainable option for boosting field productivity and thus ensuring local food security in rainfed agricultural areas of Kenya. © 2020 Society of Chemical Industry [ABSTRACT FROM AUTHOR]

Soil samples from an Alfisols with long-term (> 12 years) contrasting tillage intensities (i.e., no-till versus plow-till) were first divided into three distinct dry aggregate-size classes (i.e., mega-, macro-, and microaggregates) and then incubated with 13C-labeled wheat residue at three input levels of balanced nutrients supply under controlled laboratory conditions. Across all the treatments, from 55.1 to 83.4% of total straw-C was mineralized within the incubation period. The microaggregates significantly increased the straw-C mineralization rate by 3.6 and 3.1 mg C g−1 straw-C day−1 compared to the mega- and macroaggregates respectively, mainly due to the higher microbial biomass. The high input of balanced nutrients enhanced the straw-C mineralization. Moreover, the microbial C-use efficiency (CUE) of straw residue in the microaggregates was 25.1% and 7.3% higher than that in the mega- and macroaggregates, respectively. Legacy effect of the long-term no-till increased microbial CUE, mainly by reducing the C-nutrient stoichiometric imbalance. In contrast, straw addition to the microaggregates resulted in the lowest positive priming effect (PE) of SOC compared to that added to the mega- and macroaggregates. Both the nutrient input and no-till treatments positively contributed to the decrease in positive PE. The negative correlation between the microbial CUE of the straw residue and the positive PE was reported for the first time, and the inputs of straw residue and high-level nutrients to the microaggregates from the no-till treatment increased CUE but decreased positive PE. Baicalein is a traditional Chinese medicine with multiple pharmacological effects, including anti-inflammatory activity, anti-pathogen activity, antitumor activity and neuroprotective activity. However, due to the limited solubility and poor stability in aqueous solution, its clinical application is greatly compromised. Here, we investigated the solubility and thermodynamic properties of baicalein in a binary system of water and ethanol from 283.15 K to 328.15 K (common temperature in labs and pharmaceutical industry), in order to optimize the extraction and purification process of baicalein. The results showed that the experimental solubility of baicalein increased with the increased ethanol ratio in the binary system and with the rising temperature. Six models, the ideal mixture model, the polynomial empirical equation, the Apelblat equation, the Buchowski–Ksiazczak (λh) equation, the general single model and the combination of Jouyban–Acree and van't Hoff models, were chosen to correlate with the experimental data. The correlated results of the general single model were in good agreement with the experimentally measured solubility data with root mean square deviations (%) in the range of 0.22–1.06 × 10 − 3 . In addition, its thermodynamic property implied that the dissolution process of baicalein under the studied conditions is a non-spontaneous endothermic process. [ABSTRACT FROM AUTHOR]

We have investigated ferroelectric phase formation of Si-doped HfO2 through nucleation and phase transition in the thermal process by first-principles simulation. For the nucleation process from amorphous HfO2 in the thin film during thermal annealing, the tetragonal phase can be thermodynamically preferred with the effects of dopant and surface energy and temperature for which entropy is directly calculated from phonon spectrum calculations. During cooling down, the diffusionless phase transition from the tetragonal to ferroelectric phase takes place by atomic displacement and the transition to the monoclinic phase is suppressed by the high kinetic activation barrier. We have monolithically integrated resistance random access memory (RRAM) array with oxide semiconductor channel access transistor in 3-D stack, achieved uniform memory characteristics of 1T1R cells at each layer, and demonstrated basic functionality of XNOR operation as in-memory computing for binary neural network (BNN) artificial intelligence (AI) applications. The integrated TiN/Ti/HfO2/Ti stack RRAM shows high on/off ratio with stable retention and endurance properties. The indium gallium zinc oxide (IGZO) channel field-effect transistor (FET) works as an access transistor, which realized the low-temperature process, high mobility, and high drive current for RRAM. With the peripheral circuit, we demonstrate the XNOR operation using a pair of 1T1R cells. The impact of RRAM bit error rate on neural network is also investigated, which indicates the property of error-resilience in BNN. Cell surface engineering was proven as the efficient strategy for enhanced production of target metabolites. In this study, we want to improve the yield of target protein by engineering cell surface in Bacillus licheniformis. First, our results confirmed that deletions of d-alanyl-lipoteichoic acid synthetase gene dltD, cardiolipin synthase gene clsA and CDP-diacylglycerol-serine O-phosphatidyltransferase gene pssA were not conducive to cell growth, and the biomass of gene deletion strains were, respectively, decreased by 10.54 ± 1.43%, 14.17 ± 1.51%, and 17.55 ± 1.28%, while the concentrations of total extracellular proteins were improved, due to the increases of cell surface net negative charge and cell membrane permeability. In addition, the activities of target proteins, nattokinase, and α-amylase were also improved significantly in gene deletion strains. Furthermore, the triplicate gene (dltD, clsA, and pssA) deletion strain was constructed, which further led to the 45.71 ± 2.43% increase of cell surface net negative charge and 26.45 ± 2.31% increase of cell membrane permeability, and the activities of nattokinase and α-amylase reached 37.15 ± 0.89 FU/mL and 305.3 ± 8.4 U/mL, increased by 46.09 ± 3.51% and 96.34 ± 7.24%, respectively. Taken together, our results confirmed that cell surface engineering via deleting dltD, clsA, and pssA is an efficient strategy for enhanced production of target proteins, and this research provided a promising host strain of B. licheniformis for efficient protein expression. The outcome of patients with diffuse large B-cell lymphoma (DLBCL) with central nervous system (CNS) recurrence is poor. However, there is currently no consensus regarding diagnostic techniques. The aim of the present study was to investigate the cerebrospinal fluid (CSF) protein profile of DLBCL and identify a potential novel method for the early diagnosis of patients with DLBCL at high risk for subsequent CNS involvement. The CSF proteomic profiling of patients with DLBCL and a control group were compared using label-free liquid chromatography-tandem mass spectrometry. Gene Ontology and pathway analyses were conducted using the Database for Annotation, Visualization and Integrated Discovery. The protein interactions were analyzed using the Search Tool for the Retrieval of Interacting Genes/Proteins database. In the present study, a total of 53 differentially expressed proteins with >1 log2 fold change (false discovery rate <0.01, P<0.05) were identified and quantified. These proteins appeared to be involved in platelet degranulation, innate immune response and cell adhesion. Two hub gene network modules were obtained by protein-protein interaction network analysis. Of these proteins, secreted protein acidic and rich in cysteine (SPARC) and proenkephalin (PENK) were significantly decreased in the CSF of patients with DLBCL, which appeared to be correlated with CNS involvement. The findings of the present study indicate that decreased expression levels of SPARC and PENK in the CSF may serve as early-phase biomarkers to evaluate the risk of CNS involvement in patients with DLBCL, enabling clinicians to offer prophylactic therapy at the time of diagnosis. Background. For childhood acute lymphocytic leukemia (ALL), central nervous system leukemia (CNSL) is still the main reason of treatment failure. Changes of cerebrospinal fluid (CSF) proteome are deemed to occur after intrathecal chemotherapy. Objective. To find critical CSF biomarkers, which could be utilized to increase diagnostic and prognostic accuracy of CNSL. Methods. We performed proteomic profiling of CSF before and after the treatment of six sporadic paediatric patients diagnosed as ALL with central nervous system (CNS) involvement. CSF samples were properly processed and analyzed through the use of label-free liquid chromatography-tandem mass spectrometry (LC-MS/MS). Results. Among identified 428 unique proteins in all CSF samples, we quantified 10 altered proteins with diverse biological functions after induction chemotherapy. Conclusions. The levels of those 10 proteins change during the treatment of CNSL. Some of the proteins are likely to play a vital biological role as biomarkers for the development of ALL. To explore low–cost, high–yielding and environment–friendly rainfed farming technology is critical for sustainable agricultural development in semiarid east African Plateau (EAP). An innovative ridge–furrow mulching system (RFMS) was first introduced to EAP, with a two–year field experiment using maize cultivar KDVI in a typical semiarid site of Kenya from 2012 to 2013. Nine treatments were designed including ridge–furrow cultivation with black plastic mulching (RFB), straw mulching (RFS) and without mulching (RF) under high (375 kg ha −1 ), conventional (225 kg ha −1 ) and zero fertilization rates. We found RFB and RFS significantly increased soil water storage (0–40 cm) by 10.2% and 4.8%, while lowered evapotranspiration rate and improved thermal status, compared to RF across two growing seasons. Importantly, RFB and RFS increased soil organic carbon and C/N ratio (soil organic carbon/soil total nitrogen) by 7.8% and 4.9%, 7.2% and 5.1% respectively, following two growing seasons of farming practice. As expected, grain yield and water use efficiency (WUE) were averagely increased by 63.3% and 63.8%, 27.5% and 35.9% in RFB and RFS than those of RF respectively. Additionally, net income and economic output–to–input ratio were significantly greater in RFB and RFS than those of RF. On the other hand, conventional fertilization rate achieved similar grain yield and WUE as high fertilization did. Considering environmental and economic benefits, the RFMS farming system with conventional fertilization may act as a promising option to upgrade local dryland agricultural system in EAP. The farming practice of alternating ridges–furrows with plastic film mulching is widely used to improve field productivity of most staple crops such as maize and wheat in the semiarid Loess Plateau, but whether the technology has similar benefits on oil crops such as linseed flax ( Linum usitatissimum L.), still remains unknown. From 2012–2013, a 2–year field experiment was carried out to evaluate the effects of different ridges–furrows mulching patterns on soil hydrothermal parameters, linseed yield performance, water use efficiency and economic benefit at a rainfed semiarid site of the Loess Plateau. Four different treatments were designed as: i) alternating small and large ridges with full film mulching (RFM), ii) alternating ridges and furrows with plastic film mulching only on the ridges (RM), iii) flat planting with fully film mulching (FM) and iv) conventional flat planting without mulching (CK). Local widely cultivated variety Tianya 9 was used as testing cultivar. The results indicated that soil water storage during early growing stage was increased by 7.1%, 0.6% and 6.3% in RFM, RM and FM, respectively, compared with that of CK across two growing seasons. After harvesting, soil water storage in RFM was recovered to higher level than that of CK. Average soil temperature at seedling stage was 3.1 °C and 2.1 °C higher in RFM and FM than that of CK. In addition, film mulching generally accelerated crop development during early vegetative period, leading to a shorter growing season in RFM and RM. Particularly, both linseed yield and water use efficiency were enhanced by 116.2% and 154.8% in RFM, 50.3% and 72.4% in RM, 87.5% and 84.2% in FM across two growing seasons, respectively, compared with those of CK. Critically, the greatest net income and output to input ratio were achieved in RFM across both growing seasons. Present results clearly suggest that the practice of alternating small and large ridges with full film mulching provide an innovative option to optimize soil hydrothermal conditions, thereby boosting the productivity and profitability of dryland linseed flax in rainfed semiarid environment. In mammals, membrane-associated complement regulatory proteins (MCRP) can protect host cells from the damaging of the activated complement. In teleost, few studies on the function of MCRP have been documented. In the present report, we identified a MCRP (named CsMCRP) from the teleost fish tongue sole Cynoglossus semilaevis and examined its immune function. CsMCRP shares moderate sequence identities with fish DAF-like molecules. CsMCRP was predicted to be a transmembrane protein with three short consensus repeats located in the extracellular region. CsMCRP expression occurred in nine different tissues, especially blood, and in peripheral blood leukocytes (PBL). Recombinant CsMCRP inhibited complement activation and interacted with bacterial pathogen, the latter in a highly selective manner. Antibody blocking the CsMCRP on PBL significantly inhibited bacterial infection of PBL. Climate-related flowering phenology has attracted increasing concerns due to its irreplaceable contribution to innovative theory and application of plant phenology at global scale. Most of previous long-term observations were focused on interannual variations in flowering time in Europe and North America, however, very few of them in China were so far reported. Here, we present a meta-analysis through compiling the extended data set from 62 ground-based observations including 136 plant species from 217 observational sites across 8 climatic zones from 1963 to 2013 in China. Quantitative trends in 649 time series (1963–2013) of flowering phases and corresponding temperature changes were analyzed. The results indicated that the beginning, peak and ending of flowering were generally advanced by 2.2, 2.3 and 1.4 days decade −1 , respectively, as a result of temperature increase. Flowering duration tended to be prolonged, mainly due to higher temperature sensitivity and greater advancement in the beginning of flowering rather than the ending of flowering. Particularly, the most pronounced advancement in flowering time was observed in east China, up to 3.6 days decade −1 as a result of contemporaneous warming. Specifically, flowering time of herbaceous plants displayed an advancing trend by 0.7 day decade −1 per degree of increasing latitude. Furthermore, early-flowering species tended to flower earlier than late-flowering ones did. Wind-pollinated and herb species showed greater phenological advancement and temperature sensitivity than insect-pollinated species and other growth forms did. Our results were to some extent different from those in Europe and North America, but played a critical and complementary role at global scale. Edwardsiella piscicida is a significant pathogenic microorganism, and can lead to systematic infections in multifold hosts, including teleost, amphibians, and mammals. Ail is a surface protein involved in virulence. However, the underlining mechanism is still to be investigated in E. piscicida. This study examined the functions and immunoprotective properties of an Ail molecule from E. piscicida. Ail1 possessed a conserved Ail/Lom superfamily domain and was exposed to the bacterial surface. Mutation of ail1 significantly reduced the survival rate under acid and oxidizing environment, motility, biofilm formation, cellular infection, tissue dissemination, and causing mortality in infected fish. The loss abilities of TX01Δ ail1 were rescued by overexpression of the ail1 gene. Recombinant Ail1 (rAil1) could interact with peripheral blood leukocytes from Japanese flounder and affect E. piscicida infection in vitro. Vaccine trial showed that rAil1 was used as a potential subunit vaccine and could induce effective immune protection against E. piscicida infection. Fish vaccinated with rAil1 produced the specific antibody against rAil1 in serum, which increased the bactericidal activity of serum complement. In addition, rAil1-vaccinated fish displayed the higher ROS and acid phosphatase activity of macrophages. Adaptive management on rainfed agricultural ecosystem is crucial to improve field productivity and soil carbon sequestration under climate change, particularly in semiarid east African Plateau (EAP). We first introduced ridge-furrow (RF) farming system with plastic mulching and balanced fertilization into EAP to explore its effects on maize growth, yield formation, water use and soil carbon sequestration in a typical semiarid site of EAP in long but cool (2012) and short but warm (2013) rainy seasons. Local maize ( Zea mays L.) hybrid, KCB was used in field experiments including three farming patterns: 1) conventional flat planting (FP); 2) RF with transparent polyethylene film (RFT) and 3) RF with black polyethylene film (RFB), in which each pattern was exposed to high fertilization (compound fertilizer) (375 kg ha −1 ), conventional fertilization (225 kg ha −1 ) and zero fertilization respectively. The results indicated that RFT and RFB treatments significantly increased soil water storage in the depth of 0–120 cm, aboveground biomass, grain yield and water use efficiency over two growing seasons. On average, fertilization treatments further increased biomass accumulation, grain yield, leaf area index and WUE by 24.3%, 13.1%, 18.7% and 9.6%, respectively on the basis of sole RFM treatment. Across two seasons, RF mulching with fertilization averagely increased soil organic carbon and C/N ratio (soil organic carbon/soil total nitrogen) by 5.1% and 10.7% respectively. Critically, increased root biomass resulted in more root residue import into soil system due to improved hydro-thermal balance, which may account for biological carbon sequestration effects in this system. In addition, our field demonstration suggested that local farmers benefited from higher economic return from this integrated farming system, showing a high acceptance extent. Taken together, this system could serve as a promising adaptive management on increasing agricultural carbon fixation and maize productivity to cope with food security and climate change in semiarid EAP and other similar areas of Africa. Two experiments were conducted to investigate how linguistic information influences attention allocation in visual search and memory for words. In Experiment 1, participants searched for the synonym of a cue word among five words. The distractors included one antonym and three unrelated words. In Experiment 2, participants were asked to judge whether the five words presented on the screen comprise a valid sentence. The relationships among words were sentential, semantically related or unrelated. A memory recognition task followed. Results in both experiments showed that linguistically related words produced better memory performance. We also found that there were significant interactions between linguistic relation conditions and memorization on eye-movement measures, indicating that good memory for words relied on frequent and long fixations during search in the unrelated condition but to a much lesser extent in linguistically related conditions. We conclude that semantic and syntactic associations attenuate the link between overt attention allocation and subsequent memory performance, suggesting that linguistic relatedness can somewhat compensate for a relative lack of attention during word search. Complement C1q domain containing protein (C1qDC) is a vital recognition molecule and has an important effect on immunity. The C1qDCs exhibit opsonic activity in fish, while the mechanisms of C1qDCs in activation complement still remain unclear. This study explored immunological characteristics of a C1qDC from Japanese flounder (Paralichthys olivaceus) (PoC1qDC). PoC1qDC consists of 296 amino acid residues, possessing a collagen domain and a C1q domain. According to our results, PoC1qDC was expressed in 9 diverse tissue samples and showed up-regulation after bacterial challenge. Recombinant PoC1qDC (rPoC1qDC) activated normal serum bactericidal and hemolytic activities by interaction with Japanese flounder IgM, but not enhanced the complement activity of C3-depeleted serum. rPoC1qDC was significantly bound to various bacterial species and agglutination activity against Edwardsiella piscicida and Streptococcus iniae. Furthermore, rPoC1qDC showed direct interaction with peripheral blood leucocytes while enhancing phagocytic and chemotactic activity. When PoC1qDC was overexpressed in Japanese flounder before E. piscicida infection, bacterial replication was significantly inhibited in fish tissues. Consistently, when PoC1qDC expression in Japanese flounder was knocked down, bacterial replication was significantly enhanced. CD59 is a complement regulatory protein that inhibits the formation of membrane attack complex of complement. In this study, we examined the expression and activity of tongue sole ( Cynoglossus semilaevis ) CD59 (CsCD59). CsCD59 possesses the conserved structural features of CD59 and shares 33%–46% sequence identities with other fish CD59. Expression of CsCD59 was high in liver, spleen, and muscle, and was stimulated by infection of bacterial pathogens. Recombinant CsCD59 (rCsCD59) exhibited an apparent inhibition effect on the activation of tongue sole serum complement. ELISA and microscopy detected binding of rCsCD59 to a number of Gram-negative and Gram-positive bacteria. Interaction with rCsCD59 did not affect bacterial viability but significantly enhanced bacterial resistance against the killing effect of fish serum. In semiarid Kenya, field productivity of maize has been at a low integrity level due to insufficient use of rainwater use. From 2012 to 2013, an innovative ridge-furrow mulching system (RFMs) was tested using local maize ( Zea mays L.) hybrid, KCB in KARI-Katumani Farm, Kenya in long and short rainy seasons. Field experiments were conducted in a randomized complete block design with four treatments: 1) RFMs with transparent polyethylene film (RFT), 2) RFMs with black polyethylene film (RFB), 3) RFMs with grass straw mulching (RFS), and 4) RFMs without mulching (CK). Soil moisture & temperature, grain yield, water use and economic benefit were determined and analyzed. The results indicated that both RFT and RFB treatments significantly increased soil water storage amount in the depth of 0–60 cm. Grain yield and water use efficiency (WUE) in both treatments were increased by 66.5–349.9% and 72.9–382% respectively, compared with those of CK over two growing seasons. In addition, grain yield and WUE in RFS treatment were only increased by from 4.2–127.1% compared with those of CK. Particularly, two types of plastic films displayed different effects on modifying topsoil temperature. Transparent film mulching significantly increased topsoil temperature by 1.3 °C ( p < 0.05) higher than CK, to facilitate growth and grain formation in long (but cool) growing season. In contrast, black film mulching lowered soil temperature by 0.3 °C lower than CK in short (but warm) growing season, which led to better soil thermal balance. Overall, RFMs with film mulching could serve as an effective solution to increase maize productivity, and hence a promising strategy to cope with food security under climate change in semiarid Kenya. Micro-field rain-harvesting farming system (MRFS) has demonstrated great potentials to enhance field productivity and profitability of dryland wheat ( Triticum aestivum L.) in semiarid eastern Asia, yet little is known whether this system results in desired effects in semiarid Africa such as Kenya. A two-year field experiment was conducted during 2012 and 2013 growing seasons to evaluate the effects of introduced MRFS on water availability, field productivity and economic benefits using a local wheat ( T. aestivum L.) cultivar DUMA in a semiarid site of Kenya. Five treatments were designed as: 1) ridge and furrow with transparent plastic mulching (RFT); 2) ridge and furrow with black plastic mulching (RFB); 3) ridge and furrow with grass straw mulching (RFS); 4) ridge and furrow without mulching (RF); and 5) traditional flat planting (CK). The results showed that mulching treatments (RFT, RFB and RFS) significantly decreased the evapotranspiration (ET) by 11.4–88.5 mm, increased wheat grain yield by 60%–163%, above-ground biomass by 58%–104% and water use efficiency for grain by 68%–271%, compared with CK over two growing seasons. RFT and RFB treatments resulted in maximal soil water storage at 1-m depth and the greatest harvest index among all treatments. Linkage analyses indicated that grain yield showed significantly positive correlation with plant height, leaf area and major spike components ( P ≤ 0.05), suggesting that plant type of wheat was altered for better yield production as a result of MRFS operation. More importantly, economic ratios of output to input were also calculated and compared. The average ratio of output to input for CK was 3.86, slightly lower than 4.28, 4.06, 5.86 and 5.34 for RFT, RFB, RFS and RF, respectively across two growing seasons. In particular, net incomes in MRFS (RFT, RFB, RFS and RF) were increased by 145%, 128%, 117% and 82% respectively, compared with that of CK. In conclusion, on-field rain-harvesting farming system provides an innovative management to boost the productivity and profitability of dryland wheat, and a potential solution to cope with food security in semiarid Kenya. The observed historical changes in crop phenological events are not only as a sensitive indicator to temperature variations but also the consequence of agronomic management such as adjustment of sowing date, cultivar selection and innovative farming practices. Clarifying how these factors influence crop phenology is of paramount importance in understanding/implementing adaptation strategies to future climate change. Changes in phenophases of dryland spring-sown wheat and maize were investigated using the observed phenological data from 1992 to 2013 across the Loess Plateau. Using a series of phenological growth models, we illustrated that once the varietal effect was fixed, rising temperature alone produced a general advancement on dates of heading and maturity, leading to a significant shortening of the vegetative phase (5.2 days decade −1 in wheat and 1.1 days decade −1 in maize), a shortened duration of reproductive phase (0.9 and 2.0 days decade −1 respectively) and the length of whole growing season (6.1 days decade −1 in wheat and 3.0 days decade −1 in maize). In contrast, cultivar shifts prolonged the reproductive duration and the growing season length by 2.2 and 4.7 days decade −1 for spring wheat, and 3.3 and 1.5 days decade −1 for maize, respectively. The continuous update of cultivars during the past two decades has offset 63.5% of the shortening effect of growing season length caused by rising temperature for both crops. Our data indicate that adjusting sowing date and shifts to longer season cultivars are the critical adaptive strategies to cope with temperature increases in dryland spring-sown wheat and maize in the Loess Plateau or regions with similar environments. Complement plays an important role in the innate immune system, and it comprises about 35 individual proteins. In mammals, complement is activated via three different pathways, the classical pathway, the alternative pathway, and the lectin pathway. All three activation pathways produce C3-convertase in different forms. C3-convertase cleaves C3 to C3a and C3b and initiates a cascade of cleavage and activation, eventually resulting in the formation of the membrane attack complex. Complement activation results in the generation of activated fragments that are involved in microbial killing, phagocytosis, inflammatory reactions, immune complex clearance, and antibody production. Although the complement system has been studied extensively in mammals, complement is less well understood in teleosts. This review summarizes the current knowledge of the teleost complement components involved in phagocytosis, chemotaxis, and cell lysis. We report the characterized complement components in various teleost species. In addition, we provide a comprehensive compilation of complement regulators, and this information is used to analyze the role of complement regulators in pathogen infection. The influence of complement receptors on the immune responses of teleosts is reviewed. Finally, we propose directions for future study of the molecular evolution, structure, and function of complement components in teleosts. Benefiting from excellent electroconductivity and unique two-dimensional (2D) nanosheet structure, the optimized electrode (NiCoSe/G-10) exhibits a high specific capacity of 421.3 C/g (1 A/g) and an excellent cycling performance (after 5000 cycles) of 84.6 % capacity retention. Activation of the complement system leads to the cleavage of component factor C5 into C5a and C5b. C5a can induce chemotaxis and inflammatory responses in mammals. CsC5 is composed of 1683 amino acid residues that include an anaphylatoxin homologous domain. Expression of CsC5 could be detected in a variety of tissues and was up-regulated by bacterial or viral pathogen infection. Purified recombinant CsC5a (rCsC5a) could bind to peripheral blood leukocytes (PBL) and stimulate PBL chemotaxis, proliferation, respiratory burst, acid phosphatase activity, and phagocytosis. Tongue sole administered rCsC5a exhibited enhanced resistance against bacterial and viral infections. Tumor necrosis factor (TNF) is one of the most important cytokines involved in inflammation, apoptosis, cell proliferation, and stimulation of the immune system. The TNF gene has been cloned in teleost fish; however, the in vivo function of fish TNF is essentially unknown. In this study, we report the identification of a TNF homologue, CsTNF1, from tongue sole ( Cynoglossus semilaevis ) and analysis of its expression and biological effect. CsTNF1 is composed of 242 amino acid residues and possesses a TNF domain and conserved receptor binding sites. Expression of CsTNF1 was detected in a wide range of tissues and up-regulated in a time-dependent manner by experimental challenge with bacterial and viral pathogens. Bacterial infection of peripheral blood leukocytes (PBL) caused extracellular secretion of CsTNF1. Purified recombinant CsTNF1 (rCsTNF1) was able to bind to PBL and stimulate the respiratory burst activity of PBL. In contrast, rCsTNF1M1 and rCsTNF1M2, the mutant CsTNF1 bearing substitutions at the receptor binding site, failed to activate PBL. Fish administered with rCsTNF1, but not with rCsTNF1M1 and rCsTNF1M2, exhibited enhanced expression of IL-1, IL-6, IL-8, IL-27, TLR9 and G3BP in a time-dependent manner and augmented resistance against bacterial and viral infection. These results provide the first evidence that the receptor binding sites are essential to a fish TNF, and that CsTNF1 is involved in the innate immune defense of fish against microbial pathogens. Chemokines are a large, diverse group of small cytokines that can be classified into several families, including the CC chemokine family, which plays a pivotal role in host defense by inducing leukocyte chemotaxis under physiological and inflammatory conditions. Here we studied 9 CC chemokines from half-smooth tongue sole ( Cynoglossus semilaevis ). Phylogenetic analysis divided these chemokines into four groups. The tissue specific expression patterns of the 9 chemokines under normal physiological conditions varied much, with most chemokines highly expressed in immune organs, while some other chemokines showing high expression levels in non-immune organs. In addition, the 9 chemokines exhibited similar or distinctly different expression profiles in response to the challenge of virus and intracellular and extracellular bacterial pathogens. Megalocytivirus, a DNA virus belonging to the Iridoviridae family, is a severe pathogen to a wide range of marine and freshwater fish. In this study, using turbot ( Scophthalmus maximus ) as a host model, we examined the immunoprotective property of one megalocytivirus gene, ORF75, in the form of DNA vaccine (named pORF75). Immunofluorescence microscopy and RT-PCR analysis showed that P444, the protein encoded by ORF75, was naturally produced in the tissues of turbot during megalocytivirus infection, and that the vaccine gene in pORF75 was expressed in fish cells transfected with pORF75 and in the tissues of turbot immunized with pORF75. Following vaccination of turbot with pORF75, a high level of survival (73%) was observed against a lethal megalocytivirus challenge. Consistently, viral replication in the vaccinated fish was significantly inhibited. Immune response analysis showed that pORF75-vaccinated fish (i) exhibited upregulated expression of the genes involved in innate and adaptive immunity, (ii) possessed specific memory immune cells that showed significant response to secondary antigen stimulation, and (iii) produced specific serum antibodies which, when co-introduced into turbot with megalocytivirus, blocked viral replication. Furthermore, whole-genome transcriptome analysis revealed that ORF75 knockdown altered the transcription of 43 viral genes. Gas diffusivity in shales is significantly affected by water. However studies on the model of diffusion coefficient for water-bearing shales are very limited. In this work, we proposed a model to calculate the Knudsen gas diffusion coefficient at the presence of water in shale nanopores. This model was used to study the gas diffusivity in a real shale pore structure composed of both organic pores and inorganic pores at different water saturations. The model was also utilized to analyze the sensitivity of the Knudsen diffusion coefficient on a variety of factors. Results indicate that the existence of water drastically reduces the Knudsen diffusion in shales. When water saturation increases from 10% to 50%, gas concentration in inorganic pores rises by 53.7%. This effect can happen both in organic pores and inorganic pores due to pore connectivity. Water in inorganic pores can transport into organic pores and hinders gas diffusion in organic pores. The sensitivity study demonstrates that the increase of temperature, pore radius and porosity can lead to the rise of Knudsen diffusion coefficient. Inversely, the increase of tortuosity induces the decrease of the Knudsen diffusion coefficient. The pore wall roughness also affects the Knudsen diffusion coefficient. In mammals, CD83 is a surface marker on mature dendritic cells and vital to lymphocyte activation. In teleost, studies on the function of CD83 are very limited. In this study, we examined the potential involvement of turbot ( Scophthalmus maximus ) CD83, SmCD83, in vaccine-induced immunity. For this purpose, turbot were immunized with pORF75, a DNA vaccine against megalocytivirus, in the presence or absence of pSmCD83, a plasmid that constitutively expresses SmCD83. Immune response and protection analysis showed that the presence of pSmCD83 significantly (i) enhanced the activation of head kidney macrophages (HKM) and immune gene expression, (ii) inhibited viral replication in fish tissues following megalocytivirus challenge and increased the survival of the vaccinated fish, and (iii) stimulated production of specific serum antibody and the cytotoxicity of peripheral blood leukocytes. To further examine the effect of SmCD83, pORF75 was administered into turbot in which SmCD83 was knocked down. Subsequent analysis showed that in fish with SmCD83 knockdown, vaccine-induced HKM activation and antibody production were severely reduced, and, consistently, the protectivity of pORF75 was drastically decreased. The antibacterial and anti-inflammatory activities, and protective effects of extracts (flavonoid glycosides) of Polygonum capitatum were investigated to detect the evidence for the utilization of the herb in the clinical therapy of gastritis caused by H. pylori. A mouse gastritis model was established using H. pylori. According to treating methods, model mice were random assigned into a model group (MG group), a triple antibiotics group (TG group, clarithromycin, omeprazole and amoxicillin), low/middle/high concentrations of flavonoid glycosides groups (LF, MF and HF groups) and low/middle/high concentrations of flavonoid glycosides and amoxicillin groups (LFA, MFA and HFA groups). A group with pathogen-free mice was regarded as a control group (CG group). The eradicate rates of H. pylori were 100%, 93%, 89% in TG, MFA and HF groups. The serum levels of IFN-gamma and gastrin were higher in a MG group than those from all other groups (P < 0.05). The serum levels of IFN-gamma and gastrin were reduced significantly in LF, MF and HF groups (P < 0.05) while little changes were observed in LFA, MFA and HFA groups. In contrast, the serum levels of IL-4 were lower and higher in MG and CG groups compared with other groups (P<0.05). The serum levels of IL-4 were increased significantly in LF, MF and HF groups (P < 0.05) while little changes were found in LFA, MFA and HFA groups. According to pathological scores, flavonoid glycosides therapy showed better protection for gastric injuries than the combination of flavonoid glycoside and amoxicillin (P < 0.05). The results suggested that flavonoid glycoside has repairing functions for gastric injuries. The results suggest that the plant can treat gastritis and protect against gastric injuries. Edwardsiella tarda is a Gram-negative bacterium with a broad host range that includes a wide variety of farmed fish as well as humans. E. tarda has long been known to be able to survive in host serum, but the relevant mechanism is unclear. In this study, we investigated the fundamental question, i.e. whether E. tarda activated serum complement or not. We found that (i) when incubated with flounder serum, E. tarda exhibited a high survival rate (87.6%), which was slightly but significantly reduced in the presence of Mg 2+ ; (ii) E. tarda -incubated serum possessed strong hemolytic activity and bactericidal activity, (iii) compared to the serum incubated with a complement-sensitive laboratory Escherichia coli strain, E. tarda -incubated serum exhibited much less chemotactic activity, (iv) in contrast to the serum incubated with live E. tarda , the serum incubated with heat-inactivated E. tarda exhibited no apparent hemolytic capacity. Taken together, these results indicate for the first time that E. tarda circumvents serum attack by preventing, to a large extent, complement activation via the alternative pathway, and that heat-labile surface structures likely play an essential role in the complement evasion of E. tarda . Cadmium is highly toxic and can cause oxidative damage, metabolic disorders, and reduced lifespan and fertility in animals. In this study, we investigated the effects of cadmium in Drosophila melanogaster , performing transcriptome analysis by using tag-based digital gene expression (DGE) profiling. Among 1970 candidate genes, 1443 were up-regulated and 527 were down-regulated following cadmium exposure. Using Gene Ontology analysis, we found that cadmium stress affects three processes: transferase activity, stress response, and the cell cycle. Furthermore, we identified five differentially expressed genes (confirmed by real-time PCR) involved in all three processes: Ald , Cdc2 , skpA , tefu , and Pvr . Pathway analysis revealed that these genes were involved in the cell cycle pathway and fat digestion and absorption pathway. This study reveals the gene expression response to cadmium stress in Drosophila , it provides insights into the mechanisms of this response, and it could contribute to our understanding of cadmium toxicity in humans. NK-lysin is an antimicrobial protein produced by cytotoxic T lymphocytes and natural killer cells. In this study, we examined the biological property of a peptide, NKLP27, derived from tongue sole (Cynoglossus semilaevis) NK-lysin. NKLP27 is composed of 27 amino acids and shares little sequence identity with known NK-lysin peptides. NKLP27 possesses bactericidal activity against both Gram-negative and Gram-positive bacteria including common aquaculture pathogens. The bactericidal activity of NKLP27 was dependent on the C-terminal five residues, deletion of which dramatically reduced the activity of NKLP27. During its interaction with the target bacterial cells, NKLP27 destroyed cell membrane integrity, penetrated into the cytoplasm, and induced degradation of genomic DNA. In vivo study showed that administration of tongue sole with NKLP27 before bacterial and viral infection significantly reduced pathogen dissemination and replication in tissues. Further study revealed that fish administered with NKLP27 exhibited significantly upregulated expression of the immune genes including those that are known to be involved in antibacterial and antiviral defense. These results indicate that NKLP27 is a novel antimicrobial against bacterial and viral pathogens, and that the observed effect of NKLP27 on bacterial DNA and host gene expression adds new insights to the action mechanism of fish antimicrobial peptides. Streptococcus iniae is a Gram-positive bacterium and a severe pathogen to a wide range of economically important fish species. In addition, S. iniae is also a zoonotic pathogen and can cause serious infections in humans. In this study, we identified from a pathogenic S. iniae strain a putative bacteriocin, Sil, and examined its biological activity. Sil is composed of 101 amino acid residues and shares 35.6% overall sequence identity with the lactococcin 972 of Lactococcus lactis. Immunoblot analysis showed that Sil was secreted by S. iniae into the extracellular milieu. Purified recombinant Sil (rSil) exhibited a dose-dependent inhibitory effect on the growth of Bacillus subtilis but had no impact on the growths of other 16 Gram-positive bacteria and 10 Gram-negative bacteria representing 23 different bacterial species. Treatment of rSil by heating at 50°C abolished the activity of rSil. rSil bound to the surface of B. subtilis but induced no killing of the target cells. Cellular study revealed that rSil interacted with turbot (Scophthalmus maximus) head kidney monocytes and inhibited the innate immune response of the cells, which led to enhanced cellular infection of S. iniae. Antibody blocking of the extracellular Sil produced by S. iniae significantly attenuated the infectivity of S. iniae. Consistent with these in vitro observations, in vivo study showed that administration of turbot with rSil prior to S. iniae infection significantly increased bacterial dissemination and colonization in fish tissues. Taken together, these results indicate that Sil is a novel virulence-associated bacteriostatic and an immunoregulator that promotes S. iniae infection by impairing the immune defense of host fish. Abstract: Thioredoxin (Trx) is a redox protein that plays an important role in anti-oxidative stress and redox signaling. In a recent report, we observed that the Trx of tongue sole (Cynoglossus semilaevis), CsTrx1, is a reductase and an antioxidant with immunostimulatory property. In the current work, we studied the biological effect of a truncated CsTrx1, CsTrx80, which is composed of the N-terminal 80 residues of CsTrx1. We found that recombinant CsTrx80 (rCsTrx80) purified from Escherichia coli exhibited no insulin disulfide reductase activity; however, unlike rCsTrx1, which showed no apparent chemotactic activity, rCsTrx80 was able to bind to the head kidney monocytes of tongue sole and caused cellular migration in a dose-dependent manner. Mutation of the two cysteine residues in the highly conserved CXXC motif had no effect on the chemotactic activity of rCsTrx80. Like rCsTrx1, rCsTrx80 induced the expression of immune relevant genes in the target cells, but the induction patterns differed from that induced by rCsTrx1. In vivo study showed that when tongue sole were administered with rCsTrx80 before being infected with a bacterial pathogen, bacterial dissemination and colonization in fish tissues were significantly reduced compared to control fish. Taken together, these results indicate for the first time that a Trx80 derived from fish Trx possesses chemotactic and immunoregulatory properties which promote host resistance against bacterial infection. Background: Natural products are often favored in the study of crop pests and diseases. Previous studies have shown that citronellal has a strong inhibition effect on Magnaporthe oryzae. The objective of this study was to clarify its mechanism of action against M. oryzae. Results: Firstly, the biological activity of citronellal against M. oryzae was determined by direct and indirect methods, and the results show that citronellal had a strong inhibition effect on M. oryzae with EC50 values of 134.00 mg/L and 70.48 μL/L air, respectively. Additionally, a preliminary study on its mechanism of action was studied. After citronellal treatment, electron microscopy revealed that the mycelium became thin and broken; scanning electron microscopy revealed that the mycelium was wrinkled and distorted; and transmission electron microscopy revealed that the mycelium cell wall was invaginated, the mass wall of mycelium was separated, and the organelles were blurred. The mycelium was further stained with CFW, and the nodes were blurred, while the mycelium was almost non-fluorescent after PI staining, and there was no significant difference in the relative conductivity of mycelium. In addition, chitinase was significantly enhanced, and the expression of chitin synthesis-related genes was 17.47-fold upregulated. Finally, we found that the efficacy of citronellal against the rice blast was as high as 82.14% according to indoor efficacy tests. Conclusion: These results indicate that citronellal can affect the synthesis of chitin in M. oryzae and damage its cell wall, thereby inhibiting the growth of mycelium and effectively protecting rice from rice blasts. Abstract: Pentraxins are a family of evolutionarily conserved proteins that play an important part in innate immunity. C-reactive protein (CRP) is a member of the pentraxin family and in humans is known to be the major acute phase protein. In this work, we report the identification and analysis of a CRP, CsCRP, from half-smooth tongue sole (Cynoglossus semilaevis). CsCRP is composed of 228 amino acid residues and possesses a Pentraxin/CRP domain. Expression of CsCRP occurred in a wide range of tissues and was upregulated by pathogen infection in kidney, spleen, blood, and, in particular, liver. Following bacterial infection, CsCRP level in blood rose rapidly within 12 h and was approximately 3.8 fold of that of the basal level. Purified recombinant CsCRP (rCsCRP) was able to interact with Gram-negative and Gram-positive bacteria including those of pathogenic nature in a dose-dependent manner. When peripheral blood leukocytes (PBL) were infected with bacterial pathogen in the presence of rCsCRP, the respiratory burst and phagocytic capacity of the cells were increased to significant extents.
