Protein kinases (PKs) are involved in many intracellular signal transduction pathways through phosphorylation cascades and have become intensely investigated pharmaceutical targets over the past two decades. Inhibition of PKs using small-molecular inhibitors is a premier strategy for the treatment of diseases in different therapeutic areas that are caused by uncontrolled PK-mediated phosphorylation and aberrant signaling. Most PK inhibitors (PKIs) are directed against the ATP cofactor binding site that is largely conserved across the human kinome comprising 518 wild-type PKs (and many mutant forms). Hence, these PKIs often have varying degrees of multi-PK activity (promiscuity) that is also influenced by factors such as single-site mutations in the cofactor binding region, compound binding kinetics, and residence times. The promiscuity of PKIs is often—but not always—critically important for therapeutic efficacy through polypharmacology. Various in vitro and in vivo studies have also indicated that PKIs have the potential of interacting with additional targets other than PKs, and different secondary cellular targets of individual PKIs have been identified on a case-by-case basis. Given the strong interest in PKs as drug targets, a wealth of PKIs from medicinal chemistry and their activity data from many assays and biological screens have become publicly available over the years. On the basis of these data, for the first time, we conducted a systematic search for non-PK targets of PKIs across the human kinome. Starting from a pool of more than 155,000 curated human PKIs, our large-scale analysis confirmed secondary targets from diverse protein classes for 447 PKIs on the basis of high-confidence activity data. These PKIs were active against 390 human PKs, covering all kinase groups of the kinome and 210 non-PK targets, which included other popular pharmaceutical targets as well as currently unclassified proteins. The target distribution and promiscuity of the 447 PKIs were determined, and different interaction profiles with PK and non-PK targets were identified. Nonthermal plasma (NTP) is an interesting technique regarding improvements of surface properties of polymeric materials. Three polymers are considered in this study, namely polypropylene (PP), polycarbonate (PC) and polyurethane (PU) chosen for their diversity of use in technical applications. An atmospheric pressure plasma jet (APPJ) using air as working gas is constructed for the treatment of polymer specimens with moderate thermal load for different treatment time intervals and at different nozzle-specimen inter distances. PC specimen is found to suffer most from temperature increase, followed by PU then PP which withstand better the heat generated by plasma. After plasma treatment, surface properties of the considered polymers are investigated using water drop analysis, attenuated total reflectance Fourier transform infrared spectroscopy, surface tester profilometer, and reflectance spectrophotometer. Plasma improves surface properties as wettability, work of energy, surface energy, roughness, and dyeability of treated polymer surfaces. Those properties are found to increase with plasma exposure time; showing more pronounced effects on PU followed by PC and PP. This study can guide the decision of using a certain polymer type treated by plasma for achieving specific applications demanding higher performance of the polymeric material to face challenging practical conditions on wettability and dyeability. Nonthermal plasma discharge is used for the decolourization of dyes used in textile industry. Two dyes were considered, namely methylene blue and malachite green in aqueous solution. Plasma was generated by a Pin-water surface DBD between a metal pin electrode placed in air and the surface of an aqueous dye solution filling a glass container. Active radicals, especially hydroxyl radicals, generated by plasma in air penetrated the aqueous dye solution and induced the oxidizing reactions leading to the dyeing material disintegration. The measurements of solution parameters, including pH, oxidation-reduction potential, and conductivity can help to optimize the plasma decolourization efficiency of the two dye solutions. It was found that the pH values of the two dye solutions decrease with the increasing plasma treatment time. This is accompanied by an increase of the oxidation-reduction potentials and conductivities. The concentration of hydrogen peroxide formed in the two dye solutions during plasma treatment was found to increase with the plasma treatment time. The decolourization efficiencies of the two dye solutions increase with plasma treatment time and can be related to the solution parameters, including reduction of the pH values, increase in the oxidation-reduction potentials as well as solution conductivities and increase of hydrogen peroxide during the plasma treatment time.
