The aim of this research was to examine the relationships among Pseudomonas fluorescens (YX2) plant growth promoting rhizobacteria (PGPR), phosphorus (P) absorption by plants, and root system architecture in apple seedlings exposed to mild, moderate or severe drought stresses. All the treatments were divided into two groups: 1) inoculated with a plant rhizobacterial strain (YX2), and 2) the non-inoculated control. Under drought stress, the YX2 inoculation improved root growth, root activity by 6%, and uptake of P, thereby promoting apple seedling growth along with the dry weight of above-ground plant parts in the mild and moderate water stress regimes. Furthermore, the inoculation also promoted total P contents in plants under both mild and moderate drought stresses. Overall, application of Pseudomonas fluorescens (YX2) is a promising approach to enhance apple production in agricultural production systems.

Heavy metal (HM) accumulation in the agricultural soil and its toxicity is a major threat for plant growth and development. HMs disrupt functional integrity of the plants, induces altered phenological and physiological responses and slashes down qualitative crop yield. Chemical messengers such as phytohormones, plant growth regulators and gasotransmitters play a crucial role in regulating plant growth and development under metal toxicity in plants. Understanding the intricate network of these chemical messengers as well as interactions of genes/metabolites/proteins associated with HM toxicity in plants is necessary for deciphering insights into the regulatory circuit involved in HM tolerance. The present review describes (a) the role of chemical messengers in HM-induced toxicity mitigation, (b) possible crosstalk between phytohormones and other signaling cascades involved in plants HM tolerance and (c) the recent advancements in biotechnological interventions including genetic engineering, genome editing and omics approaches to provide a step ahead in making of improved plant against HM toxicities.

The present investigation reports antioxidant and osmotic components of Marigold at different developmental stages and also presents influence of its leachates on germination and growth of wheat (Triticum aestivum L.) seedlings. Leaves of younger plants exhibit more phenols, tannins and flavonoids as compared to root and stem. Leaves and flowers of CV. PBG exhibited greater contents of total phenols and flavonoids in comparison to CV. PNG. Leaves and flowers had greater free amino acids and free proline as compared to stem and root. Sodium was more in roots than in stem and leaves, whereas potassium and nitrogen are greater in leaves and stem of younger plants. Leaves of CV. PBG accumulated greater contents of potassium, nitrogen and calcium in comparison to those of PNG. Activity of PAL, SOD and CAT was greater in older plants and in flowers as compared to leaves. Non-enzymatic antioxidant components like ascorbic acid and reduced glutathione also exhibited the same trend. Nevertheless, APX, GPX and GR activity was less in the older plants and more in flowers than leaves. APX and GR activity was higher in CV. PBG than CV. PNG, whereas the activity of GPX and GST was more in PNG in comparison to PBG. The two cultivars (PNG and PBG) of Tagetes erecta L., their different parts and developmental stages showed considerable variation in their antioxidant and osmotic constituents. Accumulation of various antioxidant and osmotic components reported above may probably help Tagetes erecta L. adapt to varying environments. Higher concentration of leachates of leaves and flowers of marigold reduced germination percentage and seedling growth of wheat, whereas treatments of leachates of lower concentration somewhat stimulated the growth of wheat seedlings. Flower leachates showed more pronounced effects than leaf leachates. Further experimentation may lead to identification of allelochemicals in these leachates which may pave way for exploring the possibility of their use along with synthetic herbicides/weedicides in order to minimize the toxicity of soil caused by the use of synthetic compounds.

As sessile organisms, plants are unable to avoid being subjected to environmental stresses that negatively affect their growth and productivity. Instead, they utilize various mechanisms at the morphological, physiological, and biochemical levels to alleviate the deleterious effects of such stresses. Amongst these, secondary metabolites produced by plants represent an important component of the defense system. Secondary metabolites, namely phenolics, terpenes, and nitrogen-containing compounds, have been extensively demonstrated to protect plants against multiple stresses, both biotic (herbivores and pathogenic microorganisms) and abiotic (e.g. drought, salinity, and heavy metals). The regulation of secondary metabolism by beneficial elements such as silicon (Si) is an important topic. Silicon-mediated alleviation of both biotic and abiotic stresses has been well documented in numerous plant species. Recently, many studies have demonstrated the involvement of Si in strengthening stress tolerance through the modulation of secondary metabolism. In this review, we discuss Si-mediated regulation of the synthesis, metabolism, and modification of secondary metabolites that lead to enhanced stress tolerance, with a focus on physiological, biochemical, and molecular aspects. Whilst mechanisms involved in Si-mediated regulation of pathogen resistance via secondary metabolism have been established in plants, they are largely unknown in the case of abiotic stresses, thus leaving an important gap in our current knowledge.

Salinity stress reduces growth and yield productivity of most crop plants. Potentiality of kinetin (Kn) and epi-brassinolide (EBL), either individually or combinedly in preventing the salinity (100 mM NaCl) stress mediated oxidative damage and photosynthetic inhibition was studied in Solanum lycopersicum. Combined application of Kn and EBL imparted much prominent impact on the growth, photosynthesis and metabolism of antioxidants, osmolytes and secondary metabolites. Synthesis of chlorophylls and carotenoids increased and the photosynthetic parameters like stomatal conductance, intercellular CO2 concentration and net photosynthesis were significantly improved due to application of Kn and EBL. Photosystem II functioning (Fv/Fm), photochemical quenching and electron transport rate (ETR) improved significantly in Kn and EBL treated plants imparting significant decline in salinity induced non-photochemical quenching. Exogenous Kn and EBL effectively prevented the oxidative damage by significantly declining the generation of hydrogen peroxide and superoxide under saline and non-saline conditions as reflected in lowered lipid peroxidation and electrolyte leakage. Reduced oxidative damage in Kn and EBL treated plants was accompanied down-regulation of protease and lipoxygenase concomitant with up-regulation of the antioxidant system and the accumulation of compatible osmolytes. Treatment of Kn and EBL proved effective in enhancing the contents of redox homeostasis, ascorbic acid and reduced glutathione, and the secondary metabolites assisting the enzymatic antioxidant system in combating the salinity stress efficiently. Results suggest that combined application of Kn and EBL regulate growth and photosynthesis in tomato more effectively than their individual application through a probable regulatory crosstalk mechanism.

Abiotic stress causes extensive loss to agricultural yield production worldwide. Trichoderma is the most commonly used fungal biocontrol agent throughout the world. In the present study, various Trichoderma isolates were isolated from different vegetable fields. In the isolated microflora, the colony edges varied from wavy to smooth. The mycelial forms were predominantly floccose with hyaline color and conidiophores among all the strains were highly branched. Based on morphological attributes, all the isolates were identified as Trichoderma harzianum. The molecular identification using multilocus sequencing ITS, rpb2 and tef1α, genes further confirmed the morphological identification. The average chitinase activity varied from 1.13 units/mL to 3.38 units/mL among the various isolates, which increased linearly with temperature from 15 to 30 °C. There was an amplified production in the chitinase production in the presence of Mg+ and Ca2+ and Na+ metal ions, but the presence of certain ions was found to cause the down-regulated chitinase activity, i.e., Zn2+, Hg2+, Fe2+, Ag+ and K+. All the chitinase producing Trichoderma isolates inhibited the growth of tested pathogens viz., Dematophora necatrix, Fusarium solani, Fusarium oxysporum and Pythium aphanidermatum at 25% culture-free filtrate concentration under in vitro conditions. Also, under in vivo conditions, the lowest wilt incidence and highest disease control on Fusarium oxysporum was observed in isolate BT4 with mean wilt incidence and disease control of 21% and 48%, respectively. The Trichoderma harzianum identified in this study will be further used in formulation development for the management of diseases under field conditions.

Organic fertilizer usage is been introduced into agricultural practices for preventing the damaging effects of chemical fertilizers. Present study investigated the beneficial role of organic fertilizer (nano-vermicompost) on the growth, oxidative stress parameters, antioxidant and nitrogen metabolism, osmolyte accumulation and mineral elements in tomato under drought stress. Drought stress resulted in reduced growth and biomass accumulation by triggering oxidative stress due to excess accumulation of reactive oxygen species (ROS) and reduced mineral uptake. Application of nano-vermicompost proved significantly beneficial in improving growth and mitigating the drought induced growth decline. Nano-vermicompost increased growth and dry matter content and ameliorated the decline in chlorophyll contents, photosynthesis and PSII activity more significantly at higher concentration (100 mg kg−1 soil). ROS accumulation was significantly reduced by nano-vermicompost application thereby enhancing the membrane stability under normal as well as drought conditions. Furthermore, lipid peroxidation and activities of protease and lypoxygenase were significantly reduced. Drought up-regulated antioxidant system and application of nano-vermicompost further enhanced the activities of antioxidant enzymes and the contents of non-enzymatic antioxidant components. Accumulation of osmolytes including proline, glycine betaine and sugars increased significantly due to nano-vermicompost application. Besides, decline in the activity of nitrate reductase and content of essential mineral elements like nitrogen, potassium and phosphorous was also ameliorated by nano-vermicompost application.

Environmental stresses, both abiotic and biotic, affect the survival and productivity of plants. Extreme environmental conditions hamper plant metabolism by initiating excess generation of toxic molecules like reactive oxygen species (ROS), resulting in reduced growth and yield performance. Environmental stresses effect photosynthesis by influencing water and ion balance, electron transport in thylakoids and CO2 supply by affecting stomatal functioning and the carbon reduction cycle. Water stress is one of the major environmental factors the world over, particularly in arid and semi-arid areas, limiting plant growth and yield significantly. Photosynthetic regulation under salinity stress is controlled by the transcription pattern of several genes involved in chlorophyll biosynthesis, the light signalling pathway and ROS scavenging. Proline is another important osmolyte involved in growth regulation under stress. Crop plants accumulating increased proline content exhibit greater stress tolerance.

Salinity is a major cause of crop losses worldwide. Acetylcholine (ACh) can ameliorate the adverse effects of abiotic stresses on plant growth, including salinity stress; however, the underlying molecular mechanisms of this process are unclear. Here, seedlings of Nicotiana benthamiana grown under normal conditions or exposed to 150 mmol L−1 NaCl salinity stress were then treated with a root application of 10 μM ACh. Exogenous ACh application resulted in the downregulation of the activity of the antioxidant enzymes, ascorbate peroxidase, and catalase. ACh-treated plants had lower levels of reactive oxygen species, including the superoxide anion radical and hydrogen peroxide. Transcriptome analysis indicated that ACh treatment under salt stress promoted the differential expression of 658 genes in leaves of N. benthamiana (527 were upregulated and 131 were downregulated). Gene ontology enrichment and Kyoto Encyclopedia of Genes and Genomes pathway analyses revealed that exogenous ACh application was associated with a substantial increase in the transcripts of genes related to cell wall peroxidases, xyloglucan endotransglucosylases or hydrolases, and expansins, indicating that ACh activates cell wall biosynthesis in salt-stressed plants. ACh also enhanced the expression of genes associated with the auxin, gibberellin, brassinosteroid, and salicylic acid signalling pathways, indicating that ACh induces the activation of these pathways under salt stress. Collectively, these findings indicate that ACh-induced salt tolerance in N. benthamiana seedlings is mediated by the inhibition of antioxidant enzymes, activation of cell wall biosynthesis, and hormone signalling pathways. Stress-induced genes involved in osmotic regulation and oxidation resistance were induced by ACh under salt stress. The genes whose transcript levels were elevated by ACh treatment in salt-stressed N. benthamiana could be used as molecular markers of the physiological status of plants under salt stress.

The photoperiod marks a varied set of behaviors in plants, including bulbing. Bulbing is controlled by inner signals, which can be stimulated or subdued by the ecological environment. It had been broadly stated that phytohormones control the plant development, and they are considered to play a significant part in the bulb formation. The past decade has witnessed significant progress in understanding and advancement about the photoperiodic initiation of bulbing in plants. A noticeable query is to what degree the mechanisms discovered in bulb crops are also shared by other species and what other qualities are also dependent on photoperiod. The FLOWERING LOCUS T (FT) protein has a role in flowering; however, the FT genes were afterward reported to play further functions in other biological developments (e.g., bulbing). This is predominantly applicable in photoperiodic regulation, where the FT genes seem to have experienced significant development at the practical level and play a novel part in the switch of bulb formation in Alliums. The neofunctionalization of FT homologs in the photoperiodic environments detects these proteins as a new class of primary signaling mechanisms that control the growth and organogenesis in these agronomic-related species. In the present review, we report the underlying mechanisms regulating the photoperiodic-mediated bulb enlargement in Allium species. Therefore, the present review aims to systematically review the published literature on the bulbing mechanism of Allium crops in response to photoperiod. We also provide evidence showing that the bulbing transitions are controlled by phytohormones signaling and FT-like paralogues that respond to independent environmental cues (photoperiod), and we also show that an autorelay mechanism involving FT modulates the expression of the bulbing-control gene. Although a large number of studies have been conducted, several limitations and research gaps have been identified that need to be addressed in future studies.

Brassinosteroids (BRs) are an important group of plant steroidal hormones that are actively involved in a myriad of key growth and developmental processes from germination to senescence. Moreover, BRs are known for their effective role in alleviation of stress-induced changes in normal metabolism via the activation of different tolerance mechanisms. Efforts to improve plant growth through exogenous application of BRs (through different modes such as foliar spray, presowing seed treatment, or through root growing medium) have gained considerable ground world over. It has been widely demonstrated that the exogenous application of BRs to stressed plants imparts the stress tolerance mechanisms. In BR-induced regulation of physio-biochemical processes in plants, interaction (crosstalk) of BRs with other phytohormones has been reported. This crosstalk may fine-tune the effective roles of other hormones in regulating stress tolerance. The multifaceted role of BRs consolidated so far has reflected their immense potential to help plants in counteracting the stress-induced changes. The effects of introgression and up- and down-regulation of BR-related genes reported so far to improve crop productivity have been presented here. Strong evidence exists that BRs are involved in signal transduction particularly in the regulation of the mitogen-activated protein kinase (MAPK) cascade, which in turn is involved in controlled development, cell death, and the perception of pathogen-associated molecular pattern (PAMP) signaling. How far BRs are involved in signal transduction pathways operative under stressful environments has also been comprehensively discussed in this review.

Mung bean is an important pulse crop. It is highly nutritive but is vulnerable to salinity stress. Therefore, the present study was aimed to investigate the protective effect of silicon (Si) against salt stress-induced damage to mung bean plants. Mung bean plants treated with NaCl (0, 50 and 100 mM) showed considerable declines in length and dry weights of shoots and roots. Chlorophyll-a (chl-a), chl-b, total chl, carotenoids and leaf relative water content (LRWC) decreased under NaCl stress. However, supplementation with Si in the form of sodium silicate (Na2SiO3) to NaCl-stressed plants ameliorated the adverse effects of NaCl on growth, biomass, pigment synthesis and leaf relative water content (LRWC). Silicon (Si)-supplemented plants exhibited enhanced chl-fluorescence and gas exchange parameters under normal (non-stress) as well as NaCl stress conditions. Salt-induced decline in the frequency of stomata and number of leaves per plant under salt stress was significantly recovered with Si supplementation. In addition, application of Si increased the levels of proline and glycine betaine in mung bean plants. Furthermore, histochemical staining tests showed that the levels of superoxide radicals and H2O2 increased with NaCl treatments, which thereby resulted in increased lipid peroxidation (LPO) and electrolyte leakage. Contrarily, decreased levels of H2O2, lipid peroxidation (measured as MDA content), and electrolyte leakage in Si-supplemented plants under NaCl stress indicated the stress mitigating role of Si. The activities of key antioxidant enzymes (SOD, CAT, APX and GR) under NaCl stress showed an increase under the NaCl regime. However, application of Si further boosted the activities of all four antioxidant enzymes in NaCl-stressed plants. The enhanced Na+ uptake and Na+/K+ ratio in mung bean plants accompanied by decreased K+ and Ca2+ uptake under NaCl stress were reversed with Si supplementation thereby resulting in enhanced accumulation of K+ and Ca2+ and decreased Na+. In conclusion, Si supplementation mitigated the negative effects of NaCl on mung bean plants through modifications in uptake of inorganic nutrients, osmolyte production and the antioxidant defence system.

In the present study we tried to evaluate the effect of salicylic acid (SA) in alleviating the negative effects of salinity stress. NaCl stress (50 and 100 mM) declines the shoot and root length and maximum decrease was observed at 100 mM concentration of NaCl. Similarly shoot dry weight decreased by 57.14% and root dry weight by 67.24% with 100 mM NaCl stress. The pigments and leaf relative water content (LRWC) were also observed to decline with increase in NaCl concentration. However, supplementation of SA to NaCl stressed seedlings showed enhanced length and dry weight of shoot and root. The pigment and LRWC also increased by the application of SA in the present study. NaCl stress also enhanced proline and glycine betaine (GB) by 3.01 and 2.04 folds, respectively; further enhancement was recorded by the application of SA. Hydrogen peroxide (H2O2) and malondialdehyde (MDA) content also showed rise in accumulation, however, seedlings treated with SA and NaCl (100 mM + SA) declines the H2O2 accumulation to 1.90 from 2.45 folds and MDA to 1.69 from 2.34 folds over the control. Antioxidants were observed to increase with NaCl concentration and further increase was recorded by the application of SA. Indoleacetic acid (IAA) and indole butyric acid (IBA) decreased by 36.60 and 44.16%, respectively, and ABA increased by 750% with 100 mM NaCl. Addition of SA to NaCl stressed seedlings enhanced the IAA and IBA and decreased the ABA concentration to appreciable level. NaCl is also responsible for the higher accumulation of Na+ and Na+/K+ ratio and decreased uptake of Ca2+ and K+. Supplementation of SA decreased the Na+ accumulation and enhanced the uptake of Ca2+ and K+ in NaCl stressed seedlings. In conclusion, SA supplementation mitigates the negative effects of NaCl toxicity in faba bean seedlings through the modulation of different osmoprotectants, antioxidants and nutrients uptake.

We investigated the effects of exogenous application of jasmonic acid (JA) and nitric oxide (NO) on growth, antioxidant metabolism, physio-biochemical attributes and metabolite accumulation, in tomato (Solanum lycopersicum L.) plants exposed to salt stress. Treating the plants with NaCl (200 mM) resulted in considerable growth inhibition in terms of biomass, relative water content, and chlorophyll content, all of which were significantly improved upon application of JA and NO under both normal and NaCl-stress treatments. Salt treatment particularly 200 mM NaCl caused an apparent increase in electrolyte leakage, lipid peroxidation, and hydrogen peroxide production, which were reduced by exogenous application of JA and NO. Salt treatment triggered the induction of antioxidant system by enhancing the activities of antioxidant enzymes like superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), and glutathione reductase (GR). Application of JA and NO separately as well as in combination caused a significant improvement in activities of SOD, CAT, APX, and GR activities. JA and NO either applied individually or in combination boosted the flavonoid, proline and glycine betaine synthesis under NaCl treatments. In conclusion, the exogenous application of JA and NO protected tomato plants from NaCl-induced damage by up-regulating the antioxidant metabolism, osmolyte synthesis, and metabolite accumulation.

Possible involvement of calcium (Ca) and zinc (Zn) in mitigation of salt (NaCl) stress-induced oxidative damage in Brassica juncea was investigated. Salt stress (200 mM NaCl) reduced leaf pigment synthesis and some key photosynthetic attributes including stomatal conductance and internal CO2 concentration. Exogenous application of Ca and Zn resulted in enhanced growth possibly by induction of the antioxidant defense system, resulting in improved redox state thereby favoring growth improvement. Proline accumulation (3.39-fold) was stimulated by exogenous application of Zn and Ca causing improvement in growth through enhancement in relative water content (78.46%) and increased flavonoid accumulation (86.19%). NaCl stress enhanced the hydrogen peroxide (H2O2), malondialdehyde and methylglyoxal content by 3-fold, 1.51-fold, and 2.98-fold, respectively, however, supplementation of Ca and Zn individually as well as in combination reduced the accumulation to an appreciable level. Ca and Zn treatment helped Brassica juncea plants to strengthen the antioxidant system and glyoxalase system and also enzymes of ascorbate-glutathione (AsA-Glu) cycle for better protection to membranes from reactive oxygen species. Moreover, Ca and Zn supplementation reduced the salt-induced damage by maintaining Na/K ratio through improved K uptake.

Pot experiments were conducted to investigate the role of selenium in alleviating cadmium stress in Solanum lycopersicum seedlings. Cadmium (150 mg L−1) treatment caused a significant reduction in growth in terms of height and biomass accumulation and affected chlorophyll pigments, gas exchange parameters, and chlorophyll fluorescence. Selenium (10 μM) application mitigated the adverse effects of cadmium on growth, chlorophyll and carotenoid contents, leaf relative water content, and other physiological attributes. Lipid peroxidation and electrolyte leakage increased because of cadmium treatment and selenium-treated plants exhibited considerable reduction because of the decreased production of hydrogen peroxide in them. Cadmium-treated plants exhibited enhanced activity of antioxidant enzymes that protected cellular structures by neutralizing reactive free radicals. Supplementation of selenium to cadmium-treated plants (Cd + Se) further enhanced the activity of superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), and glutathione reductase (GR) by 19.69, 31.68, 33.14, and 54.47%, respectively. Osmolytes, including proline and glycine betaine, increased with selenium application, illustrating their role in improving the osmotic stability of S. lycopersicum under cadmium stress. More importantly, selenium application significantly reduced cadmium uptake. From these results, it is clear that application of selenium alleviates the negative effects of cadmium stress in S. lycopersicum through the modifications of osmolytes and antioxidant enzymes.

Plants are confronted with a variety of environmenmtal stresses resulting in enhanced production of ROS. Plants require a threshold level of ROS for vital functions and any change in their concentration alters the entire physiology of plant. Delicate balance of ROS is maintained by an efficient functioning of intriguing indigenous defence system called antioxidant system comprising enzymatic and non enzymatic components. Down regulation of antioxidant system leads to ROS induced oxidative stress causing damage to important cellular structures and hence anomalies in metabolism. Proper mineral nutrition, in addition to other agricultural practices, forms an important part for growth and hence the yield. Potassium (K) is a key macro-element regulating growth and development through alterations in physiological and biochemical attributes. K has been reported to result into accumulation of osmolytes and augmentation of antioxidant components in the plants exposed to water and salt stress. In the present review an effort has been made to revisit the old findings and the current advances in research regarding the role of optimal, suboptimal and deficient K soil status on growth under normal and stressful conditions. Effect of K deficiency and sufficiency is discussed and the information about the K mediated antioxidant regulation and plant response is highlighted.

Providing sufficient food to burgeoning population from the steadily shrinking arable land seems to be very difficult in near future and is one of the foremost challenges for plant scientists. In addition, there are several biotic and abiotic stresses which frequently encounter crop plants during various stages of life cycle, resulting in considerable yield losses. Environmental stresses, including drought, flooding, salinity, temperature (both low and high), high radiation, and xenobiotics induce toxicity, membrane damage, excessive reactive oxygen species (ROS) production, reduced photosynthesis, and altered nutrient acquisition. Several indigenous defence mechanisms (physiological and molecular) are triggered in plants on exposure to environmental cues. Enhancement of resistance of crop plants to environmental stresses has been the topic of prime interest for agriculturalists and plant scientists since long. Development of water and salinity stress-tolerant crops through genetic engineering provides an avenue towards the reclamation of farmlands that have been lost due to salinity and lack of irrigation water/rainfall. Understanding the complexity of stress tolerance mechanisms in orthodox or model plants at the genetic and molecular levels improves feasibility of enhancing tolerance of sensitive crop plants.

The effects of 24-epibrassinolide (EBL, 10−6 M) on mitigation of mercury (Hg, 15 μM and 30 μM HgCl2) toxicity on growth, physio-biochemical attributes, and antioxidant enzymes in chickpea (Cicer arietinum L.) were evaluated. Mercury (Hg)-treated plants showed reduced growth and leaf pigment content in a concentration-dependent manner, but this effect was significantly ameliorated by EBL application. Exogenously applied EBL enhanced the accumulation of proline and glycine betaine (GB) in control as well as in Hg-treated plants reflecting the positive effects of EBL on leaf’s relative water content (RWC) in the plants. EBL supplementation reduced the production of hydrogen peroxide, and hence, lipid peroxidation that ultimately provided membrane stability, thereby aiding in reducing electrolyte leakage induced by Hg toxicity. Application of EBL significantly improved the activities of antioxidant enzymes including SOD, CAT, GST, and GPX, thereby resulting in optimization of the ascorbate–glutathione pathway by improving the contents of electron donors and redox components, ascorbate (AsA), and glutathione (reduced as well as oxidized), which might have contributed to protecting the photosynthetic electron transport chain from Hg-induced oxidative stress. Considerable improvement in the uptake of essential elements was also obvious in EBL-treated plants. In conclusion, Hg stress induced adverse effects on growth and development of chickpea seedlings; however, EBL-treated seedlings showed restored growth through modulation of biochemical parameters and enzymatic and nonenzymatic antioxidants.

We examined the role of jasmonic acid (JA) in faba bean under cadmium (Cd) stress, which reduces the growth, biomass yield, leaf relative water content (LRWC) and pigment systems. Hydrogen peroxide (H2O2) and lipid peroxidation (malondialdehyde [MDA]) levels increased by 2.78 and 2.24-fold, respectively, in plants under Cd stress, resulting in enhanced electrolyte leakage. Following foliar application to Cd-treated plants, JA restored growth, biomass yield, LRWC and pigment systems to appreciable levels and reduced levels of H2O2, MDA and electrolyte leakage. Proline and glycine betaine concentrations increased by 5.73 and 2.61-fold, respectively, in faba bean under Cd stress, with even higher concentrations observed following JA application to Cd-stressed plants. Proper management techniques have been proposed to considerably lower the intensity of salinity on crop growth and productivity. Therefore experiments were conducted to assess the role of improved nitrogen (N) supplementation on the growth and salinity stress tolerance in wheat by analyzing the antioxidants, osmolytes and secondary metabolites. RESULTS: Salinity (100 mM NaCl) stress imparted deleterious effects on the chlorophyll and carotenoid synthesis as well as the photosynthetic efficiency. N supplementation resulted in increased photosynthetic rate, stomatal conductance and internal CO2 concentration with effects being much obvious in seedlings treated with higher N dose. Under non-saline conditions at both N levels, protease and lipoxygenase activity reduced significantly reflecting in reduced oxidative damage. Such effects were accompanied by reduced generation of toxic radicals like hydrogen peroxide and superoxide, and lipid peroxidation in N supplemented seedlings. Antioxidant defence system was up-regulated under saline and non-saline growth conditions due to N supplementation leading to protection of major cellular processes like photosynthesis, membrane structure and function, and mineral assimilation. Increased osmolyte and secondary metabolite accumulation, and redox components in N supplemented plants regulated the ROS metabolism and NaCl tolerance by further strengthening the antioxidant mechanisms. CONCLUSIONS: Findings of present study suggest that N availability regulated the salinity tolerance by reducing Na uptake and strengthening the key tolerance mechanisms.

The present research was performed to assess the effect of 24-epibrassinolide (EBR) on salt-stressed soybean plants. Salt stress suppressed growth, biomass yield, gas exchange parameters, pigment content, and chlorophyll fluorescence, but all these parameters were up-regulated by EBR supply. Moreover, salt stress increased hydrogen peroxide, malondialdehyde, and electrolyte leakage. EBR supplementation reduced the accumulation of oxidative stress biomarkers. The activities of superoxide dismutase and catalase, and the accumulation of proline, glycinebetaine, total phenols, and total flavonoids increased with NaCl stress, but these attributes further increased with EBR supplementation. The activities of enzymes and the levels of non-enzymatic antioxidants involved in the Asc-Glu cycle also increased with NaCl stress, and further enhancement in these attributes was recorded by EBR supplementation. Salinity elevated the methylglyoxal content, but it was decreased by the EBR supplementation accompanying with up-regulation of the glyoxalase cycle (GlyI and GlyII). Salinity enhanced the Na+ uptake in root and shoot coupled with a decrease in uptake of Ca2+, K+, and P. However, EBR supplementation declined Na+ accumulation and promoted the uptake of the aforementioned nutrients. Overall, EBR supplementation regulated the salt tolerance mechanism in soybean plants by modulating osmolytes, activities of key enzymes, and the levels of non-enzymatic antioxidants.

The present study was conducted to evaluate the effect of arsenic (As) toxicity and the mitigating role of nitric oxide (NO) donor sodium nitroprusside (SNP) on Vicia faba. Arsenics stress decreased the growth and biomass yield, and photosynthetic pigments, but it enhanced As accumulation. Supplementation of NO enhanced the afore-mentioned parameters except As accumulation which decreased in both shoot and root. Supplementation of NO enhanced the shoot tolerance index (Shoot TI%), root tolerance index (Root TI%) but it declined the As translocation factor (TF). Application of NO alleviated the As-induced decline in net assimilation rate, stomatal conductance, transpiration and leaf relative water content. The levels of proline and glycine betaine (GB) further increased due to NO application, whereas malondialdehyde (MDA), hydrogen peroxide (H2O2), electrolyte leakage (EL) and methylglyoxal (MG) declined considerably. Activities of enzymatic antioxidants such as superoxide dismutase (SOD) and catalase (CAT) increased under As stress. Supplementation of NO up-regulated the enzymes involved in Asc-Glu cycle and glyoxalase cycle under As toxicity. Another experiment was setup to authenticate whether NO was certainly able to alleviate As toxicity. For this purpose, the NO scavenger [2-(4-carboxy-2 phenyl)-4,4,5,5-tertamethylimidazoline-1-oxyl-3-oxide (cPTIO)] was added to As and NO supplemented plants. Addition of cPTIO to NO supplemented As-treated plants showed the same effect when As alone was supplied to plants. In conclusion, addition of NO to the growth medium maintained the plant performance under As toxicity through modulation of physio-biochemical attributes, antioxidant enzymes, and the Asc-Glu and glyoxalase systems.

Acetylcholine (ACh) is believed to improve plant growth. However, regulation at biochemical and molecular levels is largely unknown. The present study investigated the impact of exogenously applied ACh (10 µm) on growth and chlorophyll metabolism in hydroponically grown Nicotiana benthamiana under salt stress (150 mm NaCl). Salinity reduced root hydraulic conductivity while ACh-treated seedlings exhibited a significant increase, resulting in increased relative water content. Salinity induced a reduction in chlorophyll biosynthetic intermediates, such as protoporphyrin-IX, Mg-photoporphyrin-IX and protochlorophyllide, which were significantly ameliorated in the presence of ACh. This influence of ACh on chlorophyll synthesis was confirmed by up-regulation of HEMA1, CHLH, CAO and POR genes. Gas exchange parameters, i.e. stomatal conductance, internal CO2 concentration and transpiration rate, increased with ACh, thereby alleviating the salinity effects on photosynthesis. In addition, the salinity-induced enhancement of lipid peroxidation declined after ACh treatment through modulation of the activity of the assayed antioxidant enzymes (superoxide dismutase and peroxidase). Importantly, ACh significantly reduced the uptake of Na and increased uptake of K, resulting in a decline in the Na/K ratio. Results of the present study indicate that ACh can be effective in ameliorating NaCl-induced osmotic stress, altering chlorophyll metabolism and thus photosynthesis by maintaining ion homeostasis, hydraulic conductivity and water balance.

Influence of exogenously applied spermine (Spm) on growth and salinity stress tolerance in tomato was investigated. Salinity reduced growth, chlorophyll synthesis and mineral uptake leading to significant reduction in photosynthesis, however Spm application proved beneficial in alleviating the decline to considerable extent. Applied Spm improved nitrate reductase activity, δ-amino levulinic acid content and gas exchange parameters more apparently at 100 μM than 50 μM concentrations. Spm application enhanced the accumulation of compatible osmolytes including proline, glycine betaine and sugars leading to greater tissue water content and photosynthesis. Salinity stress induced oxidative effects were mitigated by Spm treatment reflected interms of reduced accumulation of reactive oxygen species and the activities of protease and lipoxygenase, hence leading to membrane strengthening and protection of their function. Differential influence of exogenous Spm was evident on the functioning of antioxidant system with SOD, GR and APX activities much higher in Spm treated seedlings than CAT and DHAR. Increased synthesis of GSH, AsA and tocopherol in Spm treated seedlings was obvious thereby helping in maintaining the redox homeostasis and the enzymatic antioxidant functioning. Interestingly Spm application maintained the nitric oxide levels higher than control under normal condition while as lowered its concentrations in salinity stressed seedlings depicting existence of probable interaction. Activities of polyamine metabolizing enzymes was up-regulated and the accumulation of secondary metabolites including phenols and flavonoids also increased due to Spm application. Further studies are required to understand the mechanisms clearly.

The present study reveals the effect of mercury (Hg) and sodium nitroprusside (SNP) on plant growth and metabolism in soybean cultivars (Pusa-24, Pusa-37and Pusa-40). Mercury stress decreased growth and biomass yield, and gas exchange attributes in all soybean cultivars. External supplementation of SNP mitigated Hg toxicity by improving growth and gas exchange parameters. Electrolyte leakage (EL) increased accompanied with elevated levels of malondialdehyde (MDA) and H2O2 under Hg stress, however, they were found to be reduced in all cultivars upon the exogenous application of SNP. The activities of anti-oxidative enzymes, superoxide dismutase and catalase (SOD and CAT) and those enzymes involved in the ascorbate-glutathione pathway were impaired by Hg stress, but they were regulated by the application of SNP. Accumulation of Hg and NO in the shoots and roots were also regulated by the application of NO. Although, all three cultivars were affected by Hg stress, Pusa-37 was relatively less affected. Mercury stress affected the growth and development of different soybean cultivars, but Pusa-37 being tolerant was less affected. Pusa-37 was found to be more responsive to SNP than Pusa-24, Pusa-40 under Hg toxicity. The external supplementation of SNP could be a sustainable approach to economically utilize Hg affected soils.

Abiotic stresses pose deleterious effects on plant growth and development. Impeded plant growth results from the alterations in the plant metabolism more often as a consequence of disturbance in the uptake and translocation of essential mineral nutrients. Abiotic stresses such as high salinity, heavy metals, etc. show direct competition for plant nutrients at membrane transporters therefore intensifying the ill effects of nutrient deficiency. Optimizing the mineral nutrient status under such conditions can significantly ameliorate the toxic effects of stresses by way of making the nutrients available at the transport site and hence within the plant tissue, ultimately affecting the physiological and biochemical efficiency of plants. Under suboptimal and supraoptimal occurrence of mineral nutrients certain specific signaling events are initiated to regulate the plant cellular functioning and these include both local and systemic signaling. Identifying and unraveling the role of these regulatory molecules for better understanding of the nutrient stress responsive mechanisms in plants are equally essential, and their subsequent modifications using modern technologies may open future avenues. In the present review the role of mineral nutrients in alleviation of abiotic stresses has been discussed focusing on the current understanding of nutrient signaling and crosstalk with other signaling pathways.

In general, environmental stresses cause potential damage to normal plant developmental pattern reflecting on the reduced yield. Stresses induce oxidative damage by causing abrupt increase in accumulation of reactive oxygen species, hampering the structural and functional stability of cell. For averting stress-induced damage the involvement of several key metabolites has been reported, and any degree of variation in their concentrations has deleterious effects on growth and can intensify stress as well. Key metabolites include redox components like glutathione (reduced as well as oxidized), vitamins including ascorbate and tocochromanols (vitamin E family), and compatible osmolytes like amino acids and sugars, which protect and regulate major metabolic pathways thereby affecting the growth and developmental transitions. Efforts for the dissection and improvement of the beneficial metabolites in plants using genetic and molecular approaches have been made for several crop species. Stresses bring modification in the metabolites via several enzymatic or nonenzymatic pathways, and the products of these pathways can serve as key signals for stress amelioration. In the present review the role of different metabolites in improving plant stress tolerance and their subsequent improvement by exploiting the genetic or molecular-assisted tools has been reviewed.

Plant developmental progression is regulated by a range of environmental factors. Besides the negative effects of different stresses reported, the dissection of different underlying tolerance mechanisms has also been elucidated. Physiological, biochemical, and molecular responses of plants for averting stress-triggered alterations are controlled at genetic levels. However, the key management practices such as the proper mineral nutrition and soil health amendment have also been recognized as key controlling factors, and their integration with entire physiology and biochemistry at whole plant level has been established. No doubt, different macro- and microelements promote a range of tolerance mechanisms for combating stresses efficiently. Alterations in the metabolome of a plant after exposure to particular stress and subsequent modification by the particular management practice are still in infancy. From recent past, silicon has been accepted to trigger growth-promoting effects in plants at optimal concentration and can modulate tolerance to stresses to appreciable level. This chapter addresses the involvement of silicon in plant stress tolerance with focus on the alterations in metabolism and the stress tolerance mechanisms mentioning the key hot areas for further improvement.
