This study was designed to observe the effects of temperature, impregnation time, and stirring rate on the structure of potassium ion-impregnated calcium oxide prepared from potassium hydroxide and waste eggshell-derived calcium oxide. A total of 18 reactions were carried out in different environments to impregnate K+. The catalyst products were analyzed using Infrared spectroscopy (IR), X-ray diffraction (XRD), Field emission scanning electron microscopy (FE-SEM), and Energy dispersive X-ray spectroscopy (EDX). The XRD and EDX results indicated that at higher temperatures calcium hydroxide tends to form as well a greater incorporation of K+ ions is achieved. Moreover, stirring speed was observed to affect the growth of specific phases in both calcium oxide and calcium hydroxide crystals. However, the impregnation time variation did not show any significant effect. The prepared catalysts were applied to the transesterification reaction using waste cooking oil as the fatty acid methyl ester (FAME) source, achieving a maximum of 98.28 % conversion to FAME calculated from 1H-NMR spectra. This indicates that tailoring of the catalyst may be carried out by changing these parameters in order to achieve a desired catalyst or sorbent structure which in turn may significantly improve catalytic efficiency.

Objectives: Lead (Pb), a toxic heavy metal, is a serious concern for poultry that negatively affects their productivity and health. To combat those issues efficiently, it is necessary to include feed supplements that have rich antioxidant properties for satisfactory health and productivity. Spirulina platensis (Sp), a microalgae, is a compound that provides several health benefits for humans and animals. This study explores that supplementation of Sp in diet as well as in water reduces the burden of Pb in different tissues, improves hematology, and improves the productive performance of advanced-age laying hens. Materials and methods: Forty birds were separated into four groups: the control (C), Spirulina (Sp), Pb, and (Pb + Sp) groups. The Pb group was given Pb acetate at a dose of 2 gm/l in water ad libitum for 4 weeks. Sp group was fed Sp at a dose of 4 gm/kg feed. The Pb + Sp group was given Pb and Sp as in the previous groups. Results: Productive performance and hematology such as hemoglobin (Hb), packed cell volume, red blood cell, mean corpuscular volume, mean corpuscular Hb (MCH) concentration, and MCH were significantly (p < 0.05) decreased in Pb-treated groups compared to controls. The distribution of Pb concentration was highest in the bones and lowest in the gizzard. However, Sp treatment significantly (p < 0.05) increased the productive performance and the hematological parameters. Moreover, Pb concentration in different organs significantly decreased in the group treated with Sp. Conclusion: This study indicates that Sp can possibly be used as a natural and powerful dietary additive to mitigate heavy metal intoxication in chickens, thereby being efficient and effective for production. Escherichia coli is one of the major pathogens causing mastitis in lactating mammals. We hypothesized that E. coli from the gut and mammary glands may have similar genomic characteristics in the causation of mastitis. To test this hypothesis, we used whole genome sequencing to analyze two multidrug resistant E. coli strains isolated from mammary tissue (G2M6U) and fecal sample (G6M1F) of experimentally induced mastitis mice. Both strains showed resistance to multiple (>7) antibiotics such as oxacillin, aztreonam, nalidixic acid, streptomycin, gentamicin, cefoxitin, ampicillin, tetracycline, azithromycin and nitrofurantoin. The genome of E. coli G2M6U had 59 antimicrobial resistance genes (ARGs) and 159 virulence factor genes (VFGs), while the E. coli G6M1F genome possessed 77 ARGs and 178 VFGs. Both strains were found to be genetically related to many E. coli strains causing mastitis and enteric diseases originating from different hosts and regions. The G6M1F had several unique ARGs (e.g., QnrS1, sul2, tetA, tetR, emrK, blaTEM-1/105, and aph(6)-Id, aph(3″)-Ib) conferring resistance to certain antibiotics, whereas G2M6U had a unique heat-stable enterotoxin gene (astA) and 7192 single nucleotide polymorphisms. Furthermore, there were 43 and 111 unique genes identified in G2M6U and G6M1F genomes, respectively. These results indicate distinct differences in the genomic characteristics of E. coli strain G2M6U and G6M1F that might have important implications in the pathophysiology of mammalian mastitis, and treatment strategies for mastitis in dairy animals.

Salt stress is one of the major impairments to agricultural soil that significantly reduces growth and productivity in rice (Oryza sativa L.) and other crop plants. The proteomic mechanisms underlying salt stress tolerance in rice have not been well established. Therefore, a comprehensive understanding of molecular mechanisms associated with salt signaling, salt-toxicity detoxification, and other metabolic mechanisms is essential for elucidating salt tolerance mechanisms in rice as well as ensuring global food security. Recent proteome studies have provided a global proteomic signature of rice cultivars. Integrative studies of proteomic, physiological, and molecular responses under salt stress have provided detailed mechanisms associated with salt stress tolerance in rice. This review explores the proteomic mechanisms with finely-tuned salt-responsive networks in this cereal. Several proteomic processes, including salt sensing and signaling, scavenging of reactive oxygen species (ROS) and stress defense, salt compartmentalization and homeostasis, alterations of cell wall components, modulation of the cytoskeleton, regulation of salt-responsive genes, transcription factors, and protein synthesis, protein folding and processing, protein degradation, and strategies of carbohydrate and energy metabolism for organ development, elucidate extensive molecular mechanisms linked to salt stress responses and tolerance in rice. Further, it is updated the prospects of salt stress tolerance in rice using multi-omics and CRISPR/Cas approaches. These finely-tuned molecular insights will be beneficial to rice breeders and farmers for developing high-yielding, salt-tolerant rice cultivars to achieve global food security.

The erosion of riverbanks has a negative impact on many nations across the world, costing them land, buildings, food, fish, and other living things, which forces people to relocate. Both the frequency and severity of riverbank erosion are alarming in Bangladesh. In Zajira Upazila, a baseline investigation was carried out to evaluate the effects of river erosion on the local availability of food. The results show how erosion threatens the majority of residents in the research territory. Additionally, as a consequence of the depletion of farmland brought on by the disaster and its effects, crop production is steadily dropping. Occasionally, people lose whole properties, leaving them with few employment options and little spending power. As a result, they are always in danger of experiencing a shortage of food. The suffering people use various coping strategies to deal with these challenging circumstances, such as shifting to cheaper or less appealing eating options. Despite the fact that local governing bodies have put in place various programs to help them, including food assistance and social protection programs, these are inadequate because of the government’s constrained organizational assets and competence The study’s findings will aid scientists and decision-makers in Bangladesh and abroad in better comprehending the requirements of vulnerable riverine populations and in designing programs that would increase those societies’ food safety Therefore, these results imply that developing and implementing efficient rural development strategies could increase the food security of those residing in Bangladeshi regions threatened by riverbank erosion.

Objectives: Recent clinical studies suggest that oxidative stress is one of the key players in the pathogenesis of coronavirus disease 2019 (COVID-19), and N-acetylcysteine (NAC), a potent anti¬oxidant, has been shown to improve clinical outcomes in COVID-19 patients. We conducted a systematic review and meta-analysis of the literature published on the therapeutic intervention of NAC on COVID-19 infection. Methods: We searched PubMed, Google Scholar, and Science Direct. We identified and screened eight studies with 20,503 participants, including 2,852 in the NAC-treated group and 17,651 in the placebo group, which reported the effect of NAC on COVID-19 infection. A meta-analysis was performed using forest plots under fixed effect estimates based on the standardized mean difference (SMD) and risk ratio (RR). Results: Pooled analysis showed that NAC was associated with lower mortality in patients with COVID-19 compared with the placebo group [RR, 0.65; (95% CI: 0.56 to 0.75); p < 0.0001]. Similarly, C-reactive protein (CRP) [SMD, −0.32; (95% CI: −56 to −0.09); p = 0.0070] and D-dimer [SMD, −0.35, (95% CI: −0.59 to −0.10; p = 0.0062] levels were significantly decreased, and the oxygenation marker, PaO2/FiO2 ratio, was increased in the NAC-treated group compared with the placebo group [SMD, 0.76; (95% CI: 0.48 to 1.03); p < 0.0001]. Conclusion: Although the number of included studies was minimal, this meta-analysis suggests that NAC may have a positive effect on COVID-19 outcomes, specifically, a significant decrease in CRP and D-dimer levels and a significant increase in oxygen saturation, which decreased mortality. Taro is a plant in the Araceae family, and its leafstalk possesses significant botanical and culinary value owing to its noteworthy medicinal and nutritional attributes. Leafstalk colour is an essential attribute that significantly influences its desirability and appeal to both breeders and consumers. However, limited information is available about the underlying mechanism responsible for the taro plant’s colouration. Thus, the purpose of the current study was to elucidate the information on purple leafstalks in taro through comprehensive metabolome and transcriptome analysis. In total, 187 flavonoids, including 10 anthocyanins, were identified. Among the various compounds analysed, it was observed that the concentrations of five anthocyanins (keracyanin chloride (cyanidin 3-O-rutinoside chloride), cyanidin 3-O-glucoside, tulipanin (delphinidin 3-rutinoside chloride), idaein chloride (cyanidin 3-O-galactoside), and cyanidin chloride) were found to be higher in purple taro leafstalk compared to green taro leafstalk. Furthermore, a total of 3330 differentially expressed genes (DEGs) were identified by transcriptome analysis. Subsequently, the correlation network analysis was performed to investigate the relationship between the expression levels of these differentially expressed genes and the content of anthocyanin. There were 18 DEGs encoding nine enzymes detected as the fundamental structural genes contributing to anthocyanin biosynthesis, along with seven transcription factors (3 MYB and 4 bHLH) that may be promising candidate modulators of the anthocyanin biosynthesis process in purple taro leafstalk. The findings of the current investigation not only provide a comprehensive transcriptional code, but also give information on anthocyanin metabolites as well as beneficial insights into the colour mechanism of purple taro leafstalk.

The purpose of this research is to investigate the impact of human resource management on organizational innovation based on knowledge management. This research is descriptive and correlational. The statistical population includes employees and managers of the headquarters centers for the provision and development of welfare services in Tehran. According to Cochran's formula, the sample size was 109 people. The researcher used a simple random sampling method to distribute the questionnaires. In order to obtain and collect information, Nonaka and Takeuchi's (1995) knowledge management questionnaires, Bosley and Weil's (2002) human resource management, and Mirmiran model’s organizational innovation (2013) questionnaires were used. The reliability of the questionnaires was obtained using Cronbach's alpha method, 0.95, 0.91, and 0.97, respectively. Also, in order to test the hypotheses, the structural equation modeling method was used, and for descriptive data analysis and factor analysis, 22 spss and 3smart pls software were used. The results indicate that human resource management combined with knowledge management has been effective in the organizational innovation of Tehran welfare development and supply centers. Also, human resource management has had a significant impact on organizational innovation and knowledge management. This research, by increasing and applying knowledge by human resources management in Tehran welfare, has promoted organizational innovation and as a result, improved the effectiveness of services to the society, and the integration of knowledge in performance has been recommended by the management of human resources in Tehran welfare.

Abstract This study investigates the possibility of supporting tourists in a foreign land intelligently by using the Tourism Cloud Management System (TCMS) to enhance and better their tourism experience. Some technologies allow tourists to highlight popular tourist routes and circuits through the visualisation of data and sensor clustering approaches. With this, a tourist can access the shared data on a specific location to know the sites of famous local attractions, how other tourists feel about them, and how to participate in local festivities through a smart tourism model. This study surveyed the potential of smart tourism among tourists and how such technologies have developed over time while proposing a TCMS. Its goals were to make physical/paper tickets redundant via the introduction of a mobile app with eTickets that can be validated using camera and QR code technologies and to enhance the transport network using Bluetooth and GPS for real-time identification of tourists’ presence. The results show that a significant number of participants engage in tourist travels, hence the need for smart tourism and tourist management. It was concluded that smart tourism is very appealing to tourists and can improve the appeal of the destination if smart solutions are implemented. This study gives a first-hand review of the preference of tourists and the potential of smart tourism.

Drought is one of the most significant abiotic stress threatening to crop production worldwide. Soybean is a major legume crop with immense economic significance, but its production is highly dependent on optimum rainfall or abundant irrigation. As the global climate changes, it is more important to find solutions to make plants more resilient to drought. The prime aimed of the study is to investigate the effect of melatonin on drought tolerance in soybean and its potential mechanisms. Soybean seedlings were treated with 20% polyethylene glycol 6000 (PEG 6000) and subjected to osmotic stress (14 days) with or without 100 μM melatonin treatment. Our results revealed that melatonin supplementation significantly mitigated PEG-induced growth retardation and increased water absorption ability. Foliar application of melatonin also increased gas exchange and the chlorophyll fluorescence attributes by the mitigation of the osmotic-induced reduction of the reaction activity of photosystems I and II, net photosynthetic rate (Pn), stomatal conductance (Gs), transpiration rate (Tr), electron transport activity, and photosynthetic efficiency. In addition, PEG-induced elevated production of reactive oxygen species (ROS) and malondialdehyde (MDA) content were significantly reversed by melatonin treatment. Equally important, melatonin boosted the antioxidant activities of soybean plants. Moreover, osmotic stress substantially increased abscisic acid (ABA) accumulation in roots and leaves, while melatonin-received plant leaves accumulated less ABA but roots content higher ABA. Similarly, melatonin significantly suppressed ABA biosynthesis and signaling gene expression in soybean exposed to drought stress. Furthermore, osmotic stress significantly suppressed plasmalemma (GmPIPs) and tonoplast aquaporin (GmTIPs) genes expression, and their transcript abundance was up-regulated by melatonin co-addition. Taken together, our results indicated that melatonin potentially improves drought tolerance of soybean through the regulation of ABA and aquaporin gene expression, increasing photosynthetic efficiency as well as enhancing water uptake efficiency.

Pueraria thomsonii and Pueraria lobata are important medicinal plants with unique chemical compositions that are widely used in traditional Chinese medicine. To compare the nutritional and medicinal profiles of these two species, we analyzed the flavonoid, dietary fiber, total starch, and crude protein contents of one P. lobata and three P. thomsonii varieties using ultra-performance liquid chromatography-tandem mass spectrometry, enzyme weight, acid hydrolysis, and Kjeldahl methods. Furthermore, we used principal component analysis and hierarchical clustering heatmap analysis to separate the data obtained from the P. thomsonii and P. lobata samples. We detected 279 flavonoid compounds in the two Pueraria species, including 90 isoflavones and 78 flavonoids. A large proportion of isoflavones and flavonoids were more abundant in P. lobata than in P. thomsonii. The total starch content was significantly higher in P. thomsonii than in P. lobata. By contrast, the soluble dietary fiber, insoluble dietary fiber, and crude protein contents were substantially lower in P. thomsonii than in P. lobata. Taken together, our results demonstrate that P. lobata is better suited for use as a medicine, whereas P. thomsonii is better suited as an edible food, and provide a theoretical foundation for developing P. thomsonii and P. lobata germplasm resources.

Background The origins of sex differences in cardiovascular diseases are not well understood. We examined the contribution of childhood risk factors to sex differences in adult carotid artery plaques and intima‐media thickness (carotid IMT). Methods and Results Children in the 1985 Australian Schools Health and Fitness Survey were followed up when they were aged 36 to 49 years (2014–19, n=1085–1281). Log binomial and linear regression examined sex differences in adult carotid plaques (n=1089) or carotid IMT (n=1283). Childhood sociodemographic, psychosocial, and biomedical risk factors that might contribute to sex differences in carotid IMT/plaques were examined using purposeful model building with additional adjustment for equivalent adult risk factors in sensitivity analyses. Women less often had carotid plaques (10%) than men (17%). The sex difference in the prevalence of plaques (relative risk [RR] unadjusted 0.59 [95% CI, 0.43 to 0.80]) was reduced by adjustment for childhood school achievement and systolic blood pressure (RR adjusted 0.65 [95% CI, 0.47 to 0.90]). Additional adjustment for adult education and systolic blood pressure further reduced sex difference (RR adjusted 0.72 [95% CI, 0.49 to 1.06]). Women (mean±SD 0.61±0.07) had thinner carotid IMT than men (mean±SD 0.66±0.09). The sex difference in carotid IMT (β unadjusted −0.051 [95% CI, −0.061 to −0.042]) reduced with adjustment for childhood waist circumference and systolic blood pressure (β adjusted −0.047 [95% CI, −0.057 to −0.037]) and further reduced with adjustment for adult waist circumference and systolic blood pressure (β adjusted −0.034 [95% CI, −0.048 to −0.019]). Conclusions Some childhood factors contributed to adult sex differences in plaques and carotid IMT. Prevention strategies across the life course are important to reduce adult sex differences in cardiovascular diseases.

The Omicron, the latest variant of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), was first detected in November 2021 in Botswana, South Africa. Compared to other variants of SARS-CoV-2, the Omicron is the most highly mutated, with 50 mutations throughout the genome, most of which are in the spike (S) protein. These mutations may help the Omicron to evade host immunity against the vaccine. Epidemiological studies suggest that Omicron is highly infectious and spreads rapidly, but causes significantly less severe disease than the wild‐type strain and the other variants of SARS-CoV-2. With the increased transmissibility and a higher rate of re-infection, Omicron has now become a dominant variant worldwide and is predicted to be able to evade vaccine-induced immunity. Several clinical studies using plasma samples from individuals receiving two doses of US Food and Drugs Administration (FDA)-approved COVID-19 vaccines have shown reduced humoral immune response against Omicron infection, but T cell-mediated immunity was well preserved. In fact, T cell-mediated immunity protects against severe disease, and thus the disease caused by Omicron remains mild. In this review, I surveyed the current status of Omicron variant mutations and mechanisms of immune response in the context of immune escape from COVID-19 vaccines. I also discuss the potential implications of therapeutic opportunities that are independent of SARS-CoV-2 variants, including Omicron. A better understanding of vaccine-induced immune responses and variant-independent therapeutic interventions that include potent antiviral, antioxidant, and anti-cytokine activities may pave the way to reducing Omicron-related COVID-19 complications, severity, and mortality. Collectively, these insights point to potential research gaps and will aid in the development of new-generation COVID-19 vaccines and antiviral drugs to combat Omicron, its sublineages, or upcoming new variants of SARS-CoV-2.

In recent times, climate change has become a serious threat to crop production in many parts of the globe. Particularly, in Bangladesh, climate-smart agriculture practices (CSAPs) are promoted as a practical tool for mitigating and adapting to climate change in an effort to enhance productivity. Despite myriad studies on determinants of CSAPs adoption, complements and substitutes among CSAPs are seldom emphasized and recognized, thus up till now policy has not been cogently informed on how to satisfactorily promote adoption of multiple CSAPs in salinity risk hotspots. Therefore, the aim of this present study is to examine the factors that affect the likelihood and intensity of adoption of multiple CSAPs in Bangladesh with special emphasis on compliments and substitutes. To this end, survey and plots level observation data were collected from 600 farm households and 1260 plots in flood prone areas, respectively. Also, five CSAPs are under consideration – use of stress tolerant varieties (STV), intercropping with legumes (ICL), minimum tillage (MT), crop rotation with legumes (CRL) and furrow irrigated bed planting (FIBP). Multivariate and ordered Probit models were applied to achieve the objectives of the study. In the interest of effective climate action, we find two sets of complementary CSAPs (STV × ICL × MT or STV × ICL × FIBP) and substitutes (CRL with ICL and MT or FIBP with MT). Key determinants of complements are sources of information, training on CSA, extension service, and government support. Therefore, the study strongly recommends effectively coordinating the functions of multiple stakeholders such as farmers, agricultural institutions, agricultural service providers, and concerned government departments at the local level to enhance successful adoption of multiple CSAPs (compliments). This is pivotal especially against the background of the sustainable development goals on climate action as it could potentially recuperate the dynamism of agriculture in salinity risk hotspots of Bangladesh.

Members of the SNF1-related protein kinase 2 (SnRK2) family are plant-specific serine or threonine kinases that play a pivotal role in the response of plants to abiotic stresses. Members of this plant-specific kinase family have included a critical regulator (SnRK2) of abscisic acid (ABA) response in plants. Plant organ development is governed substantially by the interaction of the SnRK2 and the phytohormone abscisic acid (ABA). Recent research has revealed a synergistic link between SnRK2 and ABA signaling in a plant’s response to stress such as drought and shoot growth. SnRK2 kinases play a dual role in the control of SnRK1 and the development of a plant. The dual role of SnRK2 kinases promotes plant growth under optimal conditions and in the absence of ABA while inhibiting the growth of plants in response to ABA. In this review, we have uncovered the roles of ABA-activated SnRK2 kinases in plants, as well as their physiological mechanisms.

The post-annealing treatment condition has a great impact on the orientation and growth of perovskite phases. In this study, the formation of CH3NH3PbI3 films was controlled using different post-annealing processes, including a modified N, N-dimethylformamide (DMF) vapor treatment. Field-emission scanning electron microscopy (FE-SEM), and X-ray diffraction (XRD) were used to characterize the microstructure of the perovskite films. An appropriate amount of DMF with a modified post-annealing setup dramatically improves the properties of the perovskite films, whereas an excess amount of DMF solvent leads to poor coverage and degraded films. The average grain size of the modified setup solvent-annealed perovskite film with 100 μL of DMF at 100 °C for 10 min reached ≈1.84 μm, almost five times larger than the mere 0.427 μm of the only thermally annealed at 100 °C for 10 min CH3NH3PbI3 film. The devices fabricated with the films with modified DMF vapor treatment showed improved photovoltaic performance, providing insight into the formation mechanism, and an effective method to improve the film quality of perovskite for photovoltaic application.

Maize-soybean intercropping is practiced worldwide because of some of the anticipated advantages such as high crop yield and better utilization of resources (i.e., water, light, nutrients and land). However, the shade of the maize crop has a detrimental effect on the growth and yield of soybean under the maize-soybean intercropping system. Hence, this experiment was conducted to improve the shade tolerance of such soybean crops with optimal nitrogen (N) fertilization combined with foliar application of iron (Fe) and molybdenum (Mo). The treatments comprised five (5) maize-soybean intercropping practices: without fertilizer application (F0), with N fertilizer application (F1), with N fertilizer combined with foliar application of Fe (F2), with N fertilizer coupled with foliar application of Mo (F3) and with N fertilizer combined with foliar application of Fe and Mo (F4). The findings of this study showed that maize-soybean intercropping under F4 treatment had significantly (p< 0.05) increased growth indices such as leaf area (cm2), plant height (cm), stem diameter (mm), stem strength (g pot-1), and internode length (cm) and yield indices (i.e., No of pods plant-1, grain yield (g plant-1), 100-grain weight (g), and biomass dry matter (g plant-1)) of the soybean crop. Moreover, intercropping under F4 treatment enhanced the chlorophyll SPAD values by 26% and photosynthetic activities such as Pn by 30%, gs by 28%, and Tr by 28% of the soybean crops, but reduced its CO2 by 11%. Furthermore, maize-soybean intercropping under F4 treatment showed improved efficiency of leaf chlorophyll florescence parameters of soybean crops such as Fv/Fm (26%), qp (17%), ϕPSII (20%), and ETR (17%), but reduced NPQ (12%). In addition, the rubisco activity and soluble protein content of the soybean crop increased by 18% in maize-soybean intercropping under F4 treatment. Thus, this suggested that intercropping under optimal N fertilization combined with foliar application of Fe and Mo can improve the shade tolerance of soybean crops by regulating their chlorophyll content, photosynthetic activities, and the associated enzymes, thereby enhancing their yield and yield traits.

This research work aims to isolate endophytic bacteria from the Ajuga turkestanica plant that have plant growth promoting properties. The microscopic, enzymatic and biochemical properties of the isolated endophytic bacteria are described. Twelve isolates of endophytic bacteria were isolated by standard microbiological methods. Isolated endophytic bacteria were identified by (MALDI-TOF MS). Endophytic bacteria isolated from the plant were detected in MALDI-TOF MS. Bacterial isolates were dominated by endophytic bacteria belonging to the genus Bacillus, Pseudomonas, Siccibacter. When the growth-enhancing properties of cultivated plants used in sustainable agriculture were studied, the isolated bacterial strains showed positive results in siderophor, ammonia, indole acetic acid production, and nitrogen fixation properties. Numerous endophytic bacteria have been found to have the enzymatic activity of oxidase, catalase, amylase, lecithinase and lipase. The results showed that endophytic bacteria isolated from the plant A. turkestanica are have plant stimulating properties which can be shown as a candidate for the production of bioinoculants individually or as part of a consortium.

Previous studies have shown that spermidine (Spd) can improve tolerance to high temperature stress in tomato seedlings. To further understand how Spd regulates the molecular components of high temperature stress signaling pathways, we performed a genome-wide transcriptome analysis in tomato seedlings treated with high temperature and/or exogenous Spd. The results demonstrate that, under high temperature conditions, Spd significantly alleviated the inhibition of plant growth, as well as improving the net photosynthetic rate and pigment contents. The transcriptome analysis revealed thousands of differentially expressed genes (DEGs) in response to high temperature with or without Spd treatment. Half of the genes were induced by high temperature, part of the genes were induced by high temperature with exogenous Spd, and some were induced by the coordinated effect of high temperature and Spd. A GO analysis indicated that genes involved in cellular processes, metabolic processes, and nucleotide binding in the sample were subjected to high temperature. Some DEGs were also involved in plant physiological processes. These results suggest potential genes and molecular pathways were involved in the exogenous Spd-mediated tolerance to high temperature stress in tomato plants. A JA signaling test was designed, which indicated that MYC2 and JAS1 in heat-resistant materials were both increased, through quantitative RT-PCR.

Background and Aim: The emergence of antimicrobial resistance (AMR) in commensal organism, such as Escherichia coli of food animals, is an alarming issue for global health. It increases the possibility of transmitting AMR determinant(s) to human bacterial pathogens by transferable genetic materials, particularly by plasmids. Hence, it is important to know which resistant genes are being carried by commensal organisms in food chain in a country and their level of temporal loads. As a result, pre-emptive measures can be advocated with an aim to reduce their risks in their primary source of circulation which consequently would benefit the public health. Materials and Methods: Commensal E. coli strains from broiler chickens on randomly selected 30 farms and the farm environments were examined for the frequencies of isolation of resistant strains to oxytetracycline and ciprofloxacin. Five birds were randomly selected from each farm to collect cloacal swab samples (total of 150 samples). Furthermore, a total of 150 environmental samples comprising one each from feed, water, soil, litter, and litter damping site of each farm were screened for the isolation of commensal E. coli strains. Strains thus obtained were initially tested for their resistance to oxytetracycline and ciprofloxacin by Kirby–Bauer disk diffusion method. Oxytetracycline-resistant strains were further screened for the presence of resistance determining genes, namely, tetA, tetB, and tetC by uniplex polymerase chain reactions. Risks associated with the isolation frequency of oxytetracycline- and ciprofloxacin-resistant E. coli were also assessed by univariable logistic regression analysis. Results: The results revealed that all E. coli isolates, regardless of the source of origin, were resistant to oxytetracycline, while 78.4% (95% confidence interval [CI] 69.1-85.5%) showed resistance to ciprofloxacin. All the randomly selected (20) oxytetracycline-resistant strains harbored the tetA gene, whereas tetB and tetC were reported in three and two isolates, respectively. After univariable analysis, only one variable, that is, strain 1 of broiler chickens compared to two other strains was found to be positively associated with the isolation of ciprofloxacin-resistant E. coli (odds ratio 12.75 [95% CI 1.0- 157.1], p=0.047). Conclusion: Resistance emerged against oxytetracycline and ciprofloxacin in commensal E. coli strains circulating in live poultry and farm environments in Bangladesh seems to be very high. Thus, human infection with drug-resistant E. coli strains through food chain will critically compromise the therapeutic measures currently available.

Background and Aim: Highly pathogenic avian influenza (HPAI) is a deadly virus of zoonotic potential. The study mainly aims to determine the risk pathways (RPs) for the probable incursion of HPAI virus (HPAIV) in backyard poultry in Bangladesh. Materials and Methods: The study involves expert elicitation technique. The concept map determines the possible RPs. The map consists of 16 concepts, each with nodes from which probabilities of an event originates. These probabilities are described by qualitative descriptors ranging from negligible to high. Risk assessment has been performed using the subjective risk assessment tool. Results: The tool demonstrates positive correlation among groups of experts in the level of agreement by scoring RP; however, the level of agreement varies from 71% to 93% among group of experts. The median risk score of viral incursion through the "Exposure of backyard poultry with farm poultry in the trading market" was 11 and ranked as top, followed by "Contaminated live bird market environment" and "Sharing common scavenging space with migratory birds" (median risk score, 10.5; rank, 2), and "Scavenging of infected slaughtered poultry remnants by backyard poultry" (median risk score, 5.3; rank, 3) when no control options were applied along with the RPs. After applying or considering control option along with contaminated live bird market environment, the median risk score was reduced to 5.0. Applying a specific control option along with each RP reduced estimated median risk scores for HPAIV incursions. Conclusion: This study provides an insight into the incursion risks of HPAIV through various RPs in backyard poultry in Bangladesh.

Abstract Background Curculigo recurvata (C. recurvata) is an enthnomedicinally important herb reported to have significant medicinal values. The present study aimed to explore the in vivo and in silico anti‐nociceptive and anti‐diarrheal effects of a C. recurvate rhizome methanol extract (Me‐RCR). Methods The analgesic effects of Me‐RCR were assessed using acetic acid‐induced writhing and the formalin‐induced flicking test. The drugs were administered intraperitoneally (IP) at doses of 200 and 400 mg/kg body weight (bw). Anti‐diarrheal activity was evaluated by assessing intestinal motility, hypersecretion, and fecal score in mice at oral doses of 200 and 400 mg/kg·bw. Immediate after the official declaration of COVID-19 in Bangladesh on 8 March 2020, it has created public panic which results in price plummeting of the capital market and price hike of many essential commodities. Worldwide, the outbreak of COVID-19 has declared a pandemic. In response, the Government of Bangladesh has initiated some strict measures such as stopping the entry of passengers from Europe, stopping on-arrival visas and self-quarantine for 2 weeks for all passengers return from abroad. Still, many loopholes exist at the entry points of Bangladesh. Most of the people of Bangladesh are yet to aware of the consequences of COVID-19. In this backdrop, this article has attempted to create public awareness about COVID-19, providing some guidelines to restrict this deadly disease, enlisting current challenges of this disease in Bangladesh. This article attempts to make benefit from the concepts of nostalgia to educate the concepts of poem and also will study the Nostalgias in the Persian poems. The term of Nostalgia has been interpreted in Persian language as the sorrow of loneliness, feeling longing, sweet sadness and remorse for the bygone. Although this term has recently entered literature, its examples are seen in Persian literature from distant past. Gholam Reza Barusan is amongst the poets whose poems are replete with nostalgia and its various examples. The poems of him are tough grasp by themselves, however using the concept of Nostalgia they can be educated. Meanwhile introducing the poems by Gholam Reza Barusan, the present article investigated the various manifestations of nostalgia in his poems, including thinking about death, loneliness, loss of dears, sorrow of loneliness and paling of the human values. The current study has been carried out based on a library research and sought identifying the factors influencing the poet’s thought and mind and investigating the method by which nostalgia cases have been reflected in his poems.

Abstract Background Melatonin is a pleiotropic signaling molecule that plays multifarious roles in plants stress tolerance. The polyamine (PAs) metabolic pathway has been suggested to eliminate the effects of environmental stresses. However, the underlying mechanism of how melatonin and PAs function together under heat stress largely remains unknown. In this study, we investigated the potential role of melatonin in regulating PAs and nitric oxide (NO) biosynthesis, and counterbalancing oxidative damage induced by heat stress in tomato seedlings. Results Heat stress enhanced the overproduction of reactive oxygen species (ROS) and damaged inherent defense system, thus reduced plant growth. However, pretreatment with 100 μM melatonin (7 days) followed by exposure to heat stress (24 h) effectively reduced the oxidative stress by controlling the overaccumulation of superoxide (O2 •−) and hydrogen peroxide (H2O2), lowering the lipid peroxidation content (as inferred based on malondialdehyde content) and less membrane injury index (MII). This was associated with increased the enzymatic and non-enzymatic antioxidants activities by regulating their related gene expression and modulating the ascorbate–glutathione cycle. The presence of melatonin induced respiratory burst oxidase (RBOH), heat shock transcription factors A2 (HsfA2), heat shock protein 90 (HSP90), and delta 1-pyrroline-5-carboxylate synthetase (P5CS) gene expression, which helped detoxify excess ROS via the hydrogen peroxide-mediated signaling pathway. In addition, heat stress boosted the endogenous levels of putrescine, spermidine and spermine, and increased the PAs contents, indicating higher metabolic gene expression. Moreover, melatonin-pretreated seedlings had further increased PAs levels and upregulated transcript abundance, which coincided with suppression of catabolic-related genes expression. Under heat stress, exogenous melatonin increased endogenous NO content along with nitrate reductase- and NO synthase-related activities, and expression of their related genes were also elevated. Conclusions Melatonin pretreatment positively increased the heat tolerance of tomato seedlings by improving their antioxidant defense mechanism, inducing ascorbate–glutathione cycle, and reprogramming the PAs metabolic and NO biosynthesis pathways. These attributes facilitated the scavenging of excess ROS and increased stability of the cellular membrane, which mitigated heat-induced oxidative stress.

Palm grass (Curculigo recurvata) is an ethnomedicinally important herb reported to have significant medicinal values. The present study aimed to evaluate the antidepressant and anxiolytic activities of a methanol extract of C. recurvata rhizome (Me-RCR) through different approaches. The antidepressant and anxiolytic properties of Me-RCR were assessed by using elevated plus maze (EPM), hole-board (HBT), tail suspension (TST), and forced swimming (FST) tests in Swiss Albino mice. The in-depth antioxidative potential of Me-RCR was also evaluated through DPPH radical scavenging activity, ferric-reducing power capacity, total phenolic, flavonoid, flavonol, and antioxidant content analysis. Computational investigations were performed using computer-aided methods for screening the anxiolytic, antidepressant, and antioxidative activities of the selected lead molecules. Treatment with Me-RCR (200 and 400 mg/kg, b.w.) notably increased the number of open arm entries and the time spent in the EPM test. In the HBT, Me-RCR exhibited significant anxiolytic activity at a dose of 200 mg/kg, whereas similar activity was observed at 400 mg/kg in the EPM test. Me-RCR significantly decreased the immobility time in a dose-dependent manner in both TST and FST. The IC50 for DPPH and reducing power capacity assay were found to be 18.56 and 193 μg/mL, respectively. Promising outcomes were noted for the determination of total phenolics, flavonoids, flavonols, and antioxidant capacity. In the case of computer-aided studies, nyasicoside showed promising binding energy for antidepressant and anxiolytic activities, whereas isocurculigine demonstrated promising effects as an antioxidant. Overall, these findings suggest that Me-RCR could be a favourable therapeutic candidate for the treatment of mental and psychiatric disorders, as well as a good source of antioxidants.

Polyamine oxidases (PAOs) are key enzymes in polyamine metabolism and are related to the tolerance of plants to abiotic stresses. In this study, overexpression of cucumber (Cucumis sativus L.) PAO2 (CsPAO2) in Arabidopsis resulted in increased activity of the antioxidant enzyme and accelerated conversion from Put to Spd and Spm, while malondialdehyde content (MDA) and electrolyte leakage (EL) was decreased when compared with wild type, leading to enhanced plant growth under salt stress. Photosystem Ⅰ assembly 3 in cucumber (CsPSA3) was revealed as an interacting protein of CsPAO2 by screening yeast two-hybrid library combined with in vitro and in vivo methods. Then, CsPAO2 and CsPSA3 were silenced in cucumber via virus-mediated gene silencing (VIGS) with pV190 as the empty vector. Under salt stress, net photosynthetic rate (Pn) and transpiration rate (Tr) of CsPAO2-silencing plants were lower than pV190-silencing plants, and EL in root was higher than pV190-silencing plants, indicating that CsPAO2-silencing plants suffered more serious salt stress damage. However, photosynthetic parameters of CsPSA3-silencing plants were all higher than those of CsPAO2 and pV190-silencing plants, thereby enhancing the photosynthesis process. Moreover, CsPSA3 silencing reduced the EL in both leaves and roots when compared with CsPAO2-silencing plants, but the EL only in leaves was significantly lower than the other two gene-silencing plants, and conversion from Put to Spd and Spm in leaf was also promoted, suggesting that CsPSA3 interacts with CsPAO2 in leaves to participate in the regulation of salt tolerance through photosynthesis and polyamine conversion.

Active transposable elements (TEs) have drawn more attention as they continue to create new insertions and contribute to genetic diversity of the genome. However, only a few have been discovered in rice up to now, and their activities are mostly induced by artificial treatments (e.g., tissue culture, hybridization etc.) rather than under normal growth conditions. To systematically survey the current activity of TEs in natural rice accessions and identify rice accessions carrying highly active TEs, the transposon insertion polymorphisms (TIPs) profile was used to identify singleton insertions, which were unique to a single accession and represented the new insertion of TEs in the genome. As a result, 10,924 high-confidence singletons from 251 TE families were obtained, covering all investigated TE types. The number of singletons varied substantially among different superfamilies/families, perhaps reflecting distinct current activity. Particularly, eight TE families maintained potentially higher activity in 3,000 natural rice accessions. Sixty percent of rice accessions were detected to contain singletons, indicating the extensive activity of TEs in natural rice accessions. Thirty-five TE families exhibited potentially high activity in at least one rice accession, and the majority of them showed variable activity among different rice groups/subgroups. These naturally active TEs would be ideal candidates for elucidating the molecular mechanisms underlying the transposition and activation of TEs, as well as investigating the interactions between TEs and the host genome.

Antimony trisulfide (Sb2Se3), a non-toxic and accessible substance, has possibilities as a material for use in solar cells. The current study numerically analyses Sb2Se3 solar cells through the program Solar Cell Capacitance Simulator (SCAPS). A detailed simulation and analysis of the influence of the Sb2Se3 layer’s thickness, defect density, band gap, energy level, and carrier concentration on the devices’ performance are carried out. The results indicate that a good device performance is guaranteed with the following values in the Sb2Se3 layer: an 800 optimal thickness for the Sb2Se3 absorber; less than 1015 cm−3 for the absorber defect density; a 1.2 eV optimum band gap; a 0.1 eV energy level (above the valence band); and a 1014 cm−3 carrier concentration. The highest efficiency of 30% can be attained following optimization of diverse parameters. The simulation outcomes offer beneficial insights and directions for designing and engineering Sb2Se3 solar cells.

The present study evaluated the effect of different colored plastic mulching viz. olive polythene film (OPF), silver polythene film (SPF), white polythene film (WPF), black polythene film (BPF) and blue polythene film (BLPF) on soil properties, growth, physiology, biochemical and mineral nutrient composition in onion bulbs. Soil properties at 0–10 cm depth are variably affected by colored polythene film mulches. The highest values of soil moisture and pH were found when covered with a silver film, whereas black film provided the higher temperature among other types of film. The contents of nitrogen, phosphorus, potassium, and sulfur in the soils were slightly enhanced by mulching. Fresh biomass of shoot, bulb, and root was enhanced by silver colored mulching due to suitable moisture conditions, temperature and nutrients in the root zone. Plant height, number of leaves, bulb diameter and bulb length, were also remarkably increased after mulching where also the utmost values of dry biomass of shoot, bulb, and root were found. Under different colored polythene film mulches, a significant difference was found only for blue-yellow chromaticity (b*) while lightness (L*) and green-red chromaticity (a*) of onion skin did not affect significantly. With the mulching of olive colored polythene film, the skin of onion bulbs exhibited maximum b* values in compare to all mulches and control. Sugars, pyruvate and mineral nutrient concentration in onion bulbs also increased in all colored mulches whereas silver film provided the utmost value almost in all. Comparing all results, silver color polythene mulching is found to be the best choice for better onion growth, physiology, and quality. Nitrogen (N) supply and seed rate (SR) are two essential factors that affect the accumulation and partitioning of N and dry matter (DM) and, therefore, grain yield (GY) and N use efficiency (NUE). The objective of this experiment was to optimize N application and SR to regulate wheat growth and increase both GY and NUE. The results revealed that net photosynthetic rate (Pn), stomatal conductance (Gs), chlorophyll content, and activities of metabolic enzymes (NR and GS) significantly increased with increasing of N levels while decreasing SR. Plant tillers, GY, DM before anthesis, and N translocation, N agronomic efficiency (NAE), N recovery efficiency (NRE), and N uptake efficiency (NUPE) were highest in a combined treatment of N235 and SR180. However, N levels beyond 235 kg ha−1 significantly decreased NAE, NRE, and NUPE. By increasing SR from 135 to 180 kg ha−1 an increase of 12.9 % and 9.1% GY and NUPE, respectively, was observed. Based on this result, we estimate that 1 kg N ha−1 might be replaced by an increase of approximately 0.6 kg ha−1 SR. Our study suggested that using a combination of N and SR (N235 + SR180) could attain maximum GY and improve NUE parameters.

Soil salinization is the leading environmental factor that restricts crop growth. This study studied the effects of Bacillus cereus (B. cereus) on growth, photosynthesis, and antioxidant metabolism in salt stressed-cucumber seedlings. The results showed that B. cereus could maintain high activity in the high salt environment (4% NaCl). B. cereus significantly increased plant height, stem diameter, fresh weight, and dry weight of cucumber seedlings under salt stress, and increased root vitality, net photosynthetic rate (Pn), stomatal conductance (Gs), intercellular CO2 concentration (Ci), and transpiration rate (Tr) of cucumber seedlings under salt stress. B. cereus significantly increased the maximum photochemical quantum yield of photosystem II (Fv/Fm), the actual photochemical quantum yield (ΦPSII), and the quantum yield of regulatory energy dissipation Y (NPQ) under salt stress, which were 9.31%, 20.44%, and 5.22% higher than those under salt stress, respectively. The quantum yield of non-regulatory energy dissipation Y (NO) was reduced by 19.81%. Superoxidase (SOD), peroxidase (POD), and catalase (CAT) activities in leaves and roots of cucumber seedlings were significantly increased by B. cereus under salt stress. Compared with salt stress, SOD activities in leaves were significantly increased by 1.70% and 6.32% on the first and third days after treatment. At 1 d, 3 d, and 5 d after treatment, SOD activity in roots increased by 3.06%, 11.24%, and 3.00%, POD activity in leaves increased by 113.38%, 38.81%, and 52.89%, respectively. The POD activity in roots increased by 56.79% and 10.92% on the third and fifth days after treatment, the CAT activity in leaves increased by 8.50% and 25.55%, and the CAT activity in roots increased by 30.59% and 84.45%. Under salt stress, the H2O2 and MDA contents of seedlings treated with B. cereus decreased significantly. Compared with salt stress, the proline content in leaves decreased by 12.69%, 3.90%, and 13.12% at 1 d, 3 d, and 5 d, respectively, while the proline content in roots decreased by 44.94% and 60.08% at 3 d and 5 d, respectively. These results indicated that B. cereus could alleviate salt-induced inhibition of growth and photosynthesis by regulating antioxidant metabolism of cucumber seedlings and thus enhancing salt tolerance of cucumber seedlings.

Crops around the world are facing a diversity of environmental problems, of which high temperatures are proving to be the most serious threat to crops. Polyamine putrescine (Put) acts as a master growth regulator that contributes to optimal plant growth and development and increased stress tolerance. Here, the current study aimed to elucidate how Put functions in regulating chlorophyll (Chl) metabolism, oxidative stress, and antioxidant defense, as well as to characterize the expression of genes related to heat stress in tomato seedlings under such stress. The results revealed that Put treatment significantly attenuates heat-induced damage by promoting biomass production, increasing photosynthetic efficiency, and inhibiting excessive production of oxidative stress markers. Heat stress markedly decreased the Chl content in the tomato leaf and accelerated the leaf yellowing process. However, Put-treated tomato seedlings showed a higher Chl content, which could be associated with the functions of Put in elevating PBGD activity (Chl biosynthesis enzyme) and suppressing the activity of the Chl catabolic enzyme (Chlase and MDCase). Under high-temperature stress, the expression levels of the gene encoding factors involved in Chl biosynthesis and Chl catabolism were significantly down- and upregulated, respectively, and this trend was reversed in Put-treated heat-stressed seedlings. In addition, exogenous application of Put boosted the activity of antioxidant enzymes, along with the levels of expression of their encoding genes, only in plants that were heat stressed. Furthermore, the expression levels of heat-shock-related genes (HSP90, HSP70, and HsfA1) were elevated in Put-treated, high-temperature-stressed tomato seedlings. Taken together, our results indicate that Put treatment significantly increases the heat tolerance of tomato seedlings, by elevating Chl concentrations and suppressing Chl catabolic enzyme activity, modulating endogenous free PA content, increasing antioxidant defense efficiency, and upregulating the expression of heat-shock-related genes.

Salinity stress is one of the most prominent abiotic stresses that negatively affect crop production. Transcription factors (TFs) are involved in the absorption, transport, or compartmentation of sodium (Na+) or potassium (K+) to resist salt stress. The basic helix–loop–helix (bHLH) is a TF gene family critical for plant growth and stress responses, including salinity. Herein, we used the CRISPR/Cas9 strategy to generate the gene editing mutant to investigate the role of OsbHLH024 in rice under salt stress. The A nucleotide base deletion was identified in the osbhlh024 mutant (A91). Exposure of the A91 under salt stress resulted in a significant increase in the shoot weight, the total chlorophyll content, and the chlorophyll fluorescence. Moreover, high antioxidant activities coincided with less reactive oxygen species (ROS) and stabilized levels of MDA in the A91. This better control of oxidative stress was accompanied by fewer Na+ but more K+, and a balanced level of Ca2+, Zn2+, and Mg2+ in the shoot and root of the A91, allowing it to withstand salt stress. Furthermore, the A91 also presented a significantly up-regulated expression of the ion transporter genes (OsHKT1;3, OsHAK7, and OsSOS1) in the shoot when exposed to salt stress. These findings imply that the OsbHLH024 might play the role of a negative regulator of salt stress, which will help to understand better the molecular basis of rice production improvement under salt stress.

Di (n-butyl) phthalate (DBP), a potential endocrine disruptor, adversely affects male reproductive function via activation of oxidative stress. This study was conducted to investigate the protective effects of vitamin E (Vit E) and selenium (Se) on DBP-induced testicular damage. Prepubertal rats were given 500 mg DBP/kg alone and in combination with Vit E (250 mg/kg) & Se (1 mg/kg) by oral gavage for 7 days and sacrificed at day 1 (D1), D30 and D45 after administration. Testicular section of DBP-treated rats showed markedly distorted seminiferous tubules, no spermatids and a reduction in the thickness of their epithelial lining, compared to the control. In contrast, co-administration of Vit E & Se protected the seminiferous tubules and regained its normal architecture to the control level. To clarify whether spermatogenic cells differentiate into mature spermatids in the treated testes at the end of first wave of spermatogenesis, immunostaining for Hsc 70t, a specific marker for spermatids, was carried out. As a result, the increase in maturation of spermatids in Vit E & Se+DBP-treated testes, compared to the DBP-treated, was demonstrated. For example, the most advanced spermatids in the tubules from rats in the DBP-treated groups were steps 8-9 at D45 of recovery, while those of the DBP+Vit E & Se-treated were steps 14-19 that were more or less similar to the control group. These results show for the first time that prepubertal administration of Vit E & Se have protective effects on DBP-induced testicular damage and restoration of normal spermatogenesis. The condition of soil and microclimate are prerequisite for proper growth and development of a plant. The environment may be mediated by artificial filming with different colored polyethylene. The mechanism of lettuce growing under colored plastic film remains largely unknown. Thus, to investigate the impact of different plastic film on the growth environment, yield parameters and phytochemical properties of lettuce, the present study was performed. Five different colored polyethylene films viz. black, white, blue, olive and silver were used as a treatment factor, in filed condition. The results demonstrated that the soil pH, temperature and overall performance of lettuce, as well as the phytochemical composition, varied within the use of different films. The maximum soil pH and temperature were found under black filming plots. Additionally, the black colored plastic film exhibited the best values for most of the yield parameters studied over other plastic films, whereas the field with non-plastic film condition showed the worst data in the experiment. Phytochemical concentrations and nutritive value of lettuce were also significantly affected by the colored plastic treatments. Furthermore, the chlorophyll, anthocyanin, phenols and carotenoids contents were increased in plants grown in soil covered with black polyethylene. Vitamin C content was of 2.5 fold higher in the plants grown in the black film compared to control conditions. The application of plastic film led to elevated concentrations of macro and micronutrients in lettuce because of the direct effect of modified radiation under the plastic film. Taken together, it can be concluded that plastic filming could be used to enhance the yield and nutritional status of lettuce in natural plant production facilities.

Background and Aim: Newcastle disease is one of the most common diseases affecting poultry in Bangladesh. The disease can cause up to 100% mortality but is preventable if birds are timely and properly vaccinated with a vaccine of standard virus titer. Different live vaccines are commercially available in the country - most, if not all, are produced using lentogenic strains of the virus with variable virulence. One of the disadvantages of these vaccines is that they are not stable at high environmental temperature, and therefore, a proper cold chain must be maintained during transportation and storage. Information on how long these vaccine viruses can withstand environmental temperature, which is near the vicinity of 37°C in the summer season in Bangladesh, is scanty. The aim of this research was to measure the effect of temperature on virus titer of live ND virus vaccines and to develop a real-time reverse transcription polymerase chain reaction (rRT-PCR) standard curve to indirectly determine hemagglutination (HA) titer of virus by this highly sensitive method. Materials and Methods: In this study, thermostability of five commercial live vaccines containing LaSota, F, Clone 30, and B1 type LaSota strains was observed for up to 35 days keeping them at 37°C. From the most thermostability yielding sample, two rRT-PCR standard curves were developed: (1) By plotting the cycle threshold (CT) values as obtained from 10-fold serial dilutions up to 10-3 against their corresponding log (to the base 10) dilutions and (2) by plotting the CT values obtained from serial HA dilutions up to 2-4 against their corresponding HA titer dilutions. The PCR efficiencies based on which the graphs were fitted were also evaluated. Results: The vaccine from the LaSota strain withstood 37°C for 35 days with a gradual declination of HA titer over time, and this vaccine also had the highest initial HA titer, which was 211. The vaccine made from F strain was inactivated quickly, and it had the lowest HA titer at the beginning of the study. The first standard curve developed can be used to assess the level of virus titer in a diluted sample compared with the titer in the original undiluted vaccine preparation by plotting the CT value obtained from the dilution by rRT-PCR. The second standard curve can be used to calculate the HA titer of a vaccine dilution by plotting the CT value as obtained from the dilution by rRT-PCR. Conclusion: The regression equations for the first and second graphs were y=–3.535x+14.365 and y=–1.081x+13.703, respectively, suggesting that, for every 3.53 cycles, the PCR product would have increased 10 times and 2 times for every 1.08 cycles, respectively, indicating nearly (but not exactly) 100% PCR efficiency.

Heat stress and abscisic acid (ABA) induce leaf senescence, whereas melatonin (MT) and gibberellins (GA) play critical roles in inhibiting leaf senescence. Recent research findings confirm that plant tolerance to diverse stresses is closely associated with foliage lifespan. However, the molecular mechanism underlying the signaling interaction of MT with GA and ABA regarding heat-induced leaf senescence largely remains undetermined. Herein, we investigated putative functions of melatonin in suppressing heat-induced leaf senescence in tomato and how ABA and GA coordinate with each other in the presence of MT. Tomato seedlings were pretreated with 100 μM MT or water and exposed to high temperature (38/28°C) for 5 days (d). Heat stress significantly accelerated senescence, damage to the photosystem and upregulation of reactive oxygen species (ROS), generating RBOH gene expression. Melatonin treatment markedly attenuated heat-induced leaf senescence, as reflected by reduced leaf yellowing, an increased Fv/Fm ratio, and reduced ROS production. The Rbohs gene, chlorophyll catabolic genes, and senescence-associated gene expression levels were significantly suppressed by MT addition. Exogenous application of MT elevated the endogenous MT and GA contents but reduced the ABA content in high-temperature-exposed plants. However, the GA and ABA contents were inhibited by paclobutrazol (PCB, a GA biosynthesis inhibitor) and sodium tungstate (ST, an ABA biosynthesis inhibitor) treatment. MT-induced heat tolerance was compromised in both inhibitor-treated plants. The transcript abundance of ABA biosynthesis and signaling genes was repressed; however, the biosynthesis genes MT and GA were upregulated in MT-treated plants. Moreover, GA signaling suppressor and catabolic gene expression was inhibited, while ABA catabolic gene expression was upregulated by MT application. Taken together, MT-mediated suppression of heat-induced leaf senescence has collaborated with the activation of MT and GA biosynthesis and inhibition of ABA biosynthesis pathways in tomato.

BackgroundWith the proposed pathophysiologic mechanism of neurologic injury by SARS CoV-2, the frequency of stroke and henceforth the related hospital admissions were expected to rise. This paper investigated this presumption by comparing the frequency of admissions of stroke cases in Bangladesh before and during the pandemic.MethodsThis is a retrospective analysis of stroke admissions in a 100-bed stroke unit at the National Institute of Neurosciences and Hospital (NINS&H) which is considerably a large stroke unit. All the admitted cases from 1 January to 30 June 2020 were considered. Poisson regression models were used to determine whether statistically significant changes in admission rates can be found before and after 25 March since when there is a surge in COVID-19 infections.ResultsA total of 1394 stroke patients took admission in the stroke unit during the study period. Half of the patients were older than 60 years, whereas only 2.6% were 30 years old or younger. The male to female ratio is 1.06:1. From January to March 2020, the mean rate of admission was 302.3 cases per month, which dropped to 162.3 cases per month from April to June, with an overall reduction of 46.3% in acute stroke admission per month. In those two periods, reductions in average admission per month for ischemic stroke (IST), intracerebral hemorrhage (ICH), subarachnoid hemorrhage (SAH) and venous stroke (VS) were 45.5%, 37.2%, 71.4% and 39.0%, respectively. Based on weekly data, results of Poisson regressions confirm that the average number of admissions per week dropped significantly during the last three months of the sample period. Further, in the first three months, a total of 22 cases of hyperacute stroke management were done, whereas, in the last three months, there was an 86.4% reduction in the number of hyperacute stroke patients getting reperfusion treatment. Only 38 patients (2.7%) were later found to be RT-PCR SARS Cov-2 positive based on nasal swab testing.ConclusionThis study revealed a more than fifty percent reduction in acute stroke admission during the COVID-19 pandemic. Whether the reduction is related to the fear of getting infected by COVID-19 from hospitalization or the overall restriction on public movement or stay-home measures remains unknown.

Bangladesh is rich in wetland biodiversity with aquatic plants, fishes, and birds. Mohanganj Upazila is known as the capital of lower Bangladesh. The present study focuses on the changes of land use and land cover (LULC) with a diversity of species that are being least concerned (LC), vulnerable (VU), and endangered (EN). Over the last two decades, the wetland species of Mohanganj were gradually declined. Our results showed that 19 fish, 4 aquatic plants, and 7 bird species were LC in 2015. Among the fish and aquatic plant species, 6 fish species (Wallago attu, Ompok pabda, Channa punctate, Chitala chitala, Salmostoma phulo, and Corica soborna) and 2 aquatic plant species (Nymphaea nouchali and Nymphaea lotus) were VU during the dry and rainy season of 2017 and 2019, respectively. In the dry season of 2019, 4 fish species (W. attu, O. pabda, C. punctate, and Ch. chitala), 2 aquatic plant species (N. nouchali and N. lotus), and 7 bird species (Anas platyrhynchos, Ardeola grayii, Gyps bengalensis, Alcedo atthis, Phalacrocorax fuscicollis, Porphyrio porphyria, and Larus ridibundus) were EN. Among the species, W. attu, N. nouchaii, G. bengalensis, P. porphyria, and L. ridibundus were extremely endangered categories. Changes in LULC, the establishment of settlements for the increasing population, indiscriminate use of pesticides, environmental pollutions, and climate change are the potential reasons for declining trends of wetland biodiversity. Stern actions on land use policy, expansion of organic agriculture, bioremediation of industrial effluents, and adoption of sustainable environmental policies should be taken by the Government of Bangladesh for immediate conservation of wetland biodiversity.

Introduction: The ability to stop the execution of a movement in response to an external cue requires intact executive function. The effect of psychotropic drugs on movement inhibition is largely unknown. Movement stopping can be estimated by the Stop Signal Reaction Time (SSRT). In a recent publication, we validated an improved measure of SSRT (optimum combination SSRT, ocSSRT). Here we explored how diazepam, which enhances transmission at GABAA receptors, affects ocSSRT.Methods: Nine healthy individuals were randomized to receive placebo, 5 mg or 10 mg doses of diazepam. Each participant received both the dosage of drug and placebo orally on separate days with adequate washout. The ocSSRT and simple reaction time (RT) were estimated through a stop-signal task delivered via a battery-operated box incorporating green (Go) and red (Stop) light-emitting diodes. The task was performed just before and 1 h after dosing.Result: The mean change in ocSSRT after 10 mg diazepam was significantly higher (+27 ms) than for placebo (−1 ms; p = 0.012). By contrast, the mean change in simple response time remained comparable in all three dosing groups (p = 0.419).Conclusion: Our results confirm that a single therapeutic adult dose of diazepam can alter motor inhibition in drug naïve healthy individuals. The selective effect of diazepam on ocSSRT but not simple RT suggests that GABAergic neurons may play a critical role in movement-stopping.

Tomato is an important vegetable that is highly sensitive to drought (DR) stress which impairs the development of tomato seedlings. Recently, melatonin (ME) has emerged as a nontoxic, regulatory biomolecule that regulates plant growth and enhances the DR tolerance mechanism in plants. The present study was conducted to examine the defensive role of ME in photosynthesis, root architecture, and the antioxidant enzymes’ activities of tomato seedlings subjected to DR stress. Our results indicated that DR stress strongly suppressed growth and biomass production, inhibited photosynthesis, negatively affected root morphology, and reduced photosynthetic pigments in tomato seedlings. Per contra, soluble sugars, proline, and ROS (reactive oxygen species) were suggested to be improved in seedlings under DR stress. Conversely, ME (100 µM) pretreatment improved the detrimental-effect of DR by restoring chlorophyll content, root architecture, gas exchange parameters and plant growth attributes compared with DR-group only. Moreover, ME supplementation also mitigated the antioxidant enzymes [APX (ascorbate peroxidase), CAT (catalase), DHAR (dehydroascorbate reductase), GST (glutathione S-transferase), GR (glutathione reductase), MDHAR (monodehydroascorbate reductase), POD (peroxidase), and SOD (superoxide dismutase)], non-enzymatic antioxidant [AsA (ascorbate), DHA (dehydroascorbic acid), GSH (glutathione), and GSSG, (oxidized glutathione)] activities, reduced oxidative damage [EL (electrolyte leakage), H2O2 (hydrogen peroxide), MDA (malondialdehyde), and O2•− (superoxide ion)] and osmoregulation (soluble sugars and proline) of tomato seedlings, by regulating gene expression for SOD, CAT, APX, GR, POD, GST, DHAR, and MDHAR. These findings determine that ME pretreatment could efficiently improve the seedlings growth, root characteristics, leaf photosynthesis and antioxidant machinery under DR stress and thereby increasing the seedlings’ adaptability to DR stress.

The strawberry (Fragaria × ananassa Duch.) is an important fruit crop cultivated worldwide for its unique taste and nutritional properties. One of the major risks associated with strawberry production is cold damage. Recently, melatonin has emerged as a multifunctional signaling molecule that influences plant growth and development and reduces adverse consequences of cold stress. The present study was conducted to investigate the defensive role of melatonin and its potential interrelation with abscisic acid (ABA) in strawberry plants under cold stress. The results demonstrate that melatonin application conferred improved cold tolerance on strawberry seedlings by reducing malondialdehyde and hydrogen peroxide contents under cold stress. Conversely, pretreatment of strawberry plants with 100 μM melatonin increased soluble sugar contents and different antioxidant enzyme activities (ascorbate peroxidase, catalase, and peroxidase) and non-enzymatic antioxidant (ascorbate and glutathione) activities under cold stress. Furthermore, exogenous melatonin treatment stimulated the expression of the DREB/CBF—COR pathways’ downstream genes. Interestingly, ABA treatment did not change the expression of the DREB/CBF—COR pathway. These findings imply that the DREB/CBF-COR pathway confers cold tolerance on strawberry seedlings through exogenous melatonin application. Taken together, our results reveal that melatonin (100 μM) pretreatment protects strawberry plants from the damages induced by cold stress through enhanced antioxidant defense potential and modulating the DREB/CBF—COR pathway.

Light and temperature are related to the growth and development of plants as well as their energy consumption in plant factories. However, most of the studies to date have focused on light and temperature extremes, while the adaptive responses and underlying mechanisms of plants to non-stress light intensity (LI) and night temperature (NT) largely remain elusive. Here, we investigated the growth and physiological responses of tomatoes grown under three LI regimes of 250 (LL), 300 (LM), and 350 (LH) μmol m−2 s−1, respectively, combined with two NT conditions. The results revealed that increased LI comprehensively improved tomato growth and physiological status at lower NT levels, but the growth stimulations induced by increasing LI were limited by higher NT. In addition, the lower NT at LM and LH conferred a relatively better endogenous physiological condition and significantly promoted tomato growth, but the higher NT significantly accelerated shoot growth at LL, indicating a compensation of higher NT for low light induced growth restriction. Taken together, the current study suggests that the adaptation mechanism of tomato plants to higher NT varied with LI levels, and higher LI plus lower NT would be an effective strategy to improve tomato growth.

The functionality of the Internet is continually changing from the Internet of Computers (IoC) to the “Internet of Things (IoT)”. Most connected systems, called Cyber-Physical Systems (CPS), are formed from the integration of numerous features such as humans and the physical environment, smart objects, and embedded devices and infrastructure. There are a few critical problems, such as security risks and ethical issues that could affect the IoT and CPS. When every piece of data and device is connected and obtainable on the network, hackers can obtain it and utilise it for different scams. In medical healthcare IoT-CPS, everyday medical and physical data of a patient may be gathered through wearable sensors. This paper proposes an AI-enabled IoT-CPS which doctors can utilise to discover diseases in patients based on AI. AI was created to find a few disorders such as Diabetes, Heart disease and Gait disturbances. Each disease has various symptoms among patients or elderly. Dataset is retrieved from the Kaggle repository to execute AI-enabled IoT-CPS technology. For the classification, AI-enabled IoT-CPS Algorithm is used to discover diseases. The experimental results demonstrate that compared with existing algorithms, the proposed AI-enabled IoT-CPS algorithm detects patient diseases and fall events in elderly more efficiently in terms of Accuracy, Precision, Recall and F-measure.
