3. Alzheimer's diagnosis beyond cerebrospinal fluid: Probe-Free Detection of Tau Proteins using MXene based redox systems and molecularly imprinted polymers

Abstract

Phosphorylated Tau proteins are promising biomarkers for the diagnosis and prognosis of Alzheimer's disease. This study presents a novel voltametric sensor using a vanadium MXene polydopamine (VxPDA) redox active composite and a Tau-441-specific polyaniline molecularly imprinted polymer (PANI MIP) for the sensitive detection of Tau-441 in interstitial fluid (ISF) and plasma. The VxPDA/PANI MIP sensor demonstrates a broad detection range of 5 fg/mL to 5 ng/mL (122 aM/L to 122 pM/L) in ISF without the use of redox mediators, with a lower limit of detection (LOD) of 2.3 fg/mL (60 aM/L). Furthermore, a handheld device utilizing this technology successfully detects Tau-441 in artificial serum with high sensitivity (5 fg/mL to 150 fg/mL (122 aM/L to 366 aM/L)) and specificity within a clinically relevant range. The rapid detection time (∼32 min) and low cost (∼£20/device) of this sensor highlight its potential for minimally invasive, early AD diagnosis in clinical settings. This advancement aims to facilitate a transition away from invasive cerebrospinal fluid (CSF)-based diagnostic techniques for AD.

16. Left in Sabarimala: a time-marker in the political history of Kerala.

Abstract

The global reputation of the 'Kerala model' reflects a progressive dimension of the Indian state. However, discussions also involve its regressive aspects, particularly concerning caste and gender axes. Both progressive reformatory aspirations and regressive conservatism co-existed in Kerala's public sphere. This wavelength was challenged by the Sabarimala verdict and subsequent conflict in 2018 – the gender question of accessing a male space was converted to a discourse of religious rights to worship. When the government decided to implement the apex court verdict, the Hindu right wing mobilised the emotions for Ayyappa, the perennial deity. Afterwards, the Left-led regime which was enthusiastic about progressive politics began to step back from challenging the Right-wing conservative forces. This argument is informed by a political analysis of various events after the conflict where the Left-led regime and Right-wing conservatives had a standoff. Such a scenario leads Kerala's public sphere from a state of 'dialectical unity' towards a state of 'missing dialectics', which can challenge its secular fabric and communalise common sense. Abstract

Here, Nb2CTx@MoS2 composites have been synthesized and their catalytic activity towards the reduction of organic dyes was evaluated. The Nb2CTx@MoS2 composite has been synthesized in different ratios (25, 50, and 75%) and it was found that Nb2CTx@50MoS2 (50% MoS2) was the best composite, as it was able to achieve complete reduction of methylene blue and methyl orange within 3 and 4 min, with rate constant of 0.55305 min−1, and 0.371 min−1, respectively. In addition, the catalyst was able to retain 86% of activity even after the 5 consecutive cycles.

Abstract

The electrochemical detection of dopamine (DA) is of ultimate importance in the early-stage diagnosis of Parkinson's and Alzheimer's diseases. In this work, we report the development of a facile amination technique of Nb2CTx MXenes for the electrochemical sensing of DA. The amination was confirmed using fourier transform infrared spectroscopy and X-ray photoelectron spectroscopy the structural changes due to the amination process were confirmed using X-Ray diffractograms. The modified MXenes were used to modify carbon cloth electrodes, which were then applied to the electrochemical detection of DA. The developed sensor was able to detect dopamine linearly in the range 1 nM–100 μM and a limit of detection of (LOD) of 300 pM was achieved. The modified electrode was found to have a selective response to DA in the presence of other interferents in addition to being reproducible and stable. The modified electrode was also used for the detection of DA in serum samples with little difference in the sensing characteristics in phosphate buffer. This work presents a novel amination route for the surface functionalization of Nb2CTx and opens new avenues for applications of surface functionalized Nb-MXenes in electrochemical detection of neurotransmitters.

25. Reduced Effectiveness of Anti-IgE Treatment Among Adults with Severe Asthma with Older Age of Asthma Onset: Results from the CHRONICLE Study.

Abstract

Purpose: Younger age of asthma onset (AAO) has been associated with an allergic phenotype, whereas eosinophilic phenotypes have been associated with older AAO. In randomized trials, biologic efficacy among adults with severe asthma (SA) has varied by age at asthma onset. To determine whether these associations observed in trials apply to real-world outcomes, this study examined biologic effectiveness by AAO and biologic class in a large, real-world cohort. Patients and methods: CHRONICLE is an ongoing, real-world study of US adults with subspecialist-treated SA receiving biologics, maintenance corticosteroids, or who are uncontrolled on high-dosage inhaled corticosteroids with additional controllers. Patients enrolled between February 2018 and February 2022 who initiated a biologic for SA and had complete data for analysis were included. A locally estimated scatterplot smoothing (LOESS) analysis was used to plot the relationship between percentage exacerbation rate reduction and AAO by biologic class. Results: Of 578 patients with complete data, 198, 149, and 231 were diagnosed with asthma at age < 18, 18– 39, and ≥ 40 years, respectively. Across subgroups, patients were predominantly White (72– 78%), female (67– 73%), and commercially insured (54– 71%). In the LOESS analysis, exacerbation rate reductions were similar for anti-IgE and anti–IL-5/5R and anti–IL-4R subgroups with younger AAO, but the exacerbation rate reduction diminished for patients with older AAO receiving anti-IgE therapy, particularly with asthma onset age ≥ 40 years. Conclusion: Clinicians should consider age of onset in biologic treatment decisions, given reduced effectiveness of omalizumab in patients with asthma onset at age ≥ 40 years. Although asthma-related cough can be controlled by asthma-specific medications, both prescription and over-the-counter antitussives are often also necessary. The anti-immunoglobulin E monoclonal antibody omalizumab is an effective treatment for patients with moderate-to-severe asthma, but little is known about subsequent antitussive use patterns. This post hoc analysis examined data from the Phase 3 EXTRA study that included patients aged 12– 75 years with inadequately controlled moderate-to-severe asthma. Baseline antitussive use was low overall (16/427, 3.7% for omalizumab and 18/421, 4.3% for placebo). Among patients with no baseline antitussive use (n = 411 omalizumab, n = 403 placebo), most patients (88.3% omalizumab, 83.4% placebo) reported not using antitussives during the 48-week treatment period. The percentage of patients using 1 antitussive was lower for patients treated with omalizumab than placebo (7.1% vs 13.2%), although the adjusted rate of antitussive use during the treatment period was similar for omalizumab and placebo (0.22 and 0.25). Non-narcotics were used more often than narcotics. In conclusion, this analysis found low use of antitussives in patients with severe asthma and suggests that omalizumab may have the potential to decrease antitussive use.Keywords: antitussive, asthma, cough, omalizumab

Abstract

Forest Cover Density is one of the key parameters for assessing the ecological condition of a forest. The Present study estimates the change in forest cover density using remote sensing techniques for the last decade, i.e., from 2010 to 2021 of the Satpura Foothills in Nandurbar District. Remotely sensed multispectral data from Landsat-8 and Landsat-9 is highly useful in vegetation change analysis based on remote sensing indices and forest cover density parameters. NDVI (Normalized Difference Vegetation Index) and FCD (Forest cover density) relation are essential to understanding the environmental effects on the forest on a regional scale. Threshold-based classification has been used to realize forest cover change in multi-temporal studies. The result shows that forest canopy cover, as well as forest density, are radically reduced between the period 2010 and 2021. It is also stated that 119.51 km2 areas have been degraded over 11 years. Period (2010-2021) with a rate of 10.86 km2 every year is possible only due to the implementation of forest policies exclusively executing participatory or joint forest management techniques.
