As life expectancy rises, so too does the prevalence of neurodegenerative diseases. Neurodegeneration causes progressive regional brain atrophy, typically initiating prior to symptom onset. Researchers measure the impact of potential treatments on atrophy in mouse models to assess their effectiveness. This is important because treatments designed to combat neuropathology are more likely to modify the disease, per contra to symptom management. Magnetic resonance imaging, while accurate in measurement of brain region structure volumes, is prohibitively expensive. Conversely, stereological volume assessment, the process of estimating the volume of individual 3D brain regions from imaged 2D brain sections, is more commonly used. This involves manually tracing brain region(s) of interest in regularly spaced imaged cross-sections to determine their 2D area, followed by application of the Cavalieri principle to estimate the volume. The pertinent caveats of this approach are the labor-intensive manual tracing process, and potential inaccuracies that arise due to human variation. To overcome these challenges, we have created a Neuropathology Assessment Tool (NAT) to automate regional brain tracing and identification using artificial intelligence (AI) and concepts from topological data analysis. The NAT was validated by comparing manual and NAT analysis of striatal volume in Huntington disease model mice. The NAT detected striatal atrophy with higher efficiency, 93.8% agreement with manual measurements, and lower inter-group variability. The NAT will increase efficiency of preclinical neuropathology assessment, allowing for a greater number of experimental therapies to be tested and facilitating drug discovery intractable neurodegenerative diseases.

Background: Currently, there are different food classification systems in order to inform the population of the best alternatives for consumption, considering all the diseases associated with the consumption of products of low nutritional quality. Reports indicate that these forms of labelling warnings correspond to a laudable strategy for populations that do not have the knowledge to discriminate between the wide range of products offered by the food industry. However, recent publications indicate that there may be inconsistencies between the different classification guidelines, and the guidelines that nations should adopt in their food guides are still a matter of debate. In view of this, the present study aimed to evaluate the quantitative and qualitative differences that exist between the NOVA, Nutri-Score and Chilean Front-of-package (FoP) food warning label according to the Chilean basic food basket list., Method: An analytical study was carried out to classify a list of 736 foods according to three different systems, evaluating the distributions according to their methods of classifying the products. Quantitative differences were contrasted for each system, as well as between them, together with an analysis of the dimensions of each system., Results: According to the Nutri-Score classification, the most frequent category was A with 27% (high nutritional quality), followed by D with 22% (low nutritional quality) of the total. On the other hand, the NOVA classification showed that the most frequent categorization was ultra-processed food (NOVA 4) with 54%, followed by unprocessed (NOVA 1) with 19%. Regarding the FoP warning labels, 57% of the foods were categorized as free warning labels, followed by the category of foods with 3 warning labels (23%). Regarding the results of the principal component analysis, the Nutri-Score and FoP warning labels present a degree of similarity in their classification guidelines, being different than the dimension pointed out by NOVA., Conclusion: The present work managed to demonstrate that there are quantitative and qualitative differences between the classification and recommendation guidelines of the Nutri-Score, NOVA and FoP warning labels, finding concrete discrepancies between them.

Background: Daratumumab (Dara), an anti-CD38 monoclonal antibody for hematologic malignancies, interferes with routine blood bank testing, specifically affecting the antibody screen and identification panels. In 2016, the AABB recommended performing a baseline phenotype or genotype before a patient (Pt) begins taking anti-CD38 to avoid this interference and potential problems with transfusion. The objective of this study was to assess red blood cell (RBC) utilization and subsequent incidence of alloimmunization to the transfused RBCs in patients receiving Dara., Methods and Materials: We monitored 244 patients taking Dara to determine their red blood cell transfusions and incidence of clinically significant antibody formation before and following administration of Dara. Poisson generalized estimating equations with log link were used comparing the post-Dara incidence and prevalence to those prior, with significance defined as p < .05., Results: From September 1, 2015 to December 22, 2018, 244 patients on Dara were identified, of which 145 patients (59.4%) received a red blood cell transfusion. Antibody screens were performed on 97 of the 145 patients at least 2 weeks following RBC transfusion. Four of the total transfused patients (2.8% total, 4.1% patients with follow-up antibody screen testing) formed new clinically significant alloantibodies, which was not significantly different from Asare's hematologic incidence (p = .98/p = .49)., Conclusions: This study showed our patients on Dara did not form alloantibodies following RBC transfusion at a higher incidence than similar patient populations., (© 2021 AABB.)

Objective: In this cross-sectional study on family violence and resilience in a random sample of 5,149 middle school students with a mean age of 14.5 years from four EU-countries (Austria, Germany, Slovenia, and Spain) we examined the prevalence of exposure to family violence, and we worked from the premise that adolescent can be resilient to family violence. We expanded the definition of resilience to include the absence of both physical aggression and depression symptoms in adolescents who have been exposed to violence in their families and extended our understanding of resilience to include three levels which we describe as: "resilient", "near-resilient" and "non-resilient", thus responding to calls for a more fluid and paths-based understanding of resilience., Methods: Data were collected via self-administered surveys consisting of a number of subscales that investigate depression symptoms and physical aggression. The study was analyzed with a three-stage strategy using logistic regression procedures, in which regression analyses were conducted separately for girls and boys using seven steps for modeling the three resilience levels., Results: More than 30% of our respondents reported experiencing family violence. Contrary to previous research findings, our data showed that structural characteristics like country, gender, socio-economic status and migration status were minimally predictive of violence and depression resilience at any level. Overall, for both sexes, despite some small but significant sex differences, resilience is strongly linked to personal and relational characteristics and the absence of experiences that involved exposure to and direct experiences with violence. Resilience supportive factors confirmed by this study are: higher emotional self-control, talking with parents or friends about violence, seeking help to avoid violence, and not endorsing aggression supportive beliefs. Also key to resilience are irrespective of country, gender, and SES are lower levels of experience with: victimization by boys, engagement in physical altercation with boys, parental abuse, witnessing of physical spousal abuse, exposure to an authoritarian (harsh) parenting style and verbal aggression from teachers., Conclusion: From a content perspective this means that resilience is more than the absence of one or two behavioral factors. This also means that positive changes in resilience levels can be facilitated by supporting constructive personal and social relationships with family members, peers, and teachers. Acupuncture can alleviate symptoms of spinal cord injuries (SCI). The underlying mechanism, however, is unknown. We hypothesized that stem cells could be mobilized by acupuncture. Therefore, we enrolled 14 healthy study participants using acupuncture points for the treatment of SCI. The frequency of CD133 and CD34 cells in peripheral blood and the serum concentrations of matrix metalloproteinase (MMP)-9, brain-derived neurotrophic factor (BDNF), nerve growth factor (NGF), and interleukin-6 were determined before and after acupuncture (<1 hr, 24 hr, and 48 hr). CD133(+)34(-) cells were doubled 48 hr after acupuncture, with concomitant decreases in BDNF and MMP-9 levels. Interleukin-6 remained below detectable levels, eliminating a stress-induced cell release. Individuals acupunctured on control counterpoints showed no changes in CD133(+) cells. Our results indicate that acupuncture for SCI can mobilize human CD133(+)34(-) cells. (c) 2009 Wiley-Liss, Inc.

Background: Dobutamine-atropine stress echocardiography (DASE) is an established test for the diagnosis and risk stratification of patients with coronary artery disease. Atropine use to attain target heart rate prolongs test time., Hypothesis: The aim of this study was to assess the utility of isometric handgrip exercise (33% maximal voluntary contraction x 4 min) with DASE., Methods: We prospectively evaluated 131 patients undergoing DASE randomized to handgrip exercise or no handgrip. Effect of handgrip exercise on endpoints: time to target heart rate (85% maximum predicted), recovery time, total test time, mean dobutamine and atropine dosage, and the number of ischemic responses were assessed. Effect of current beta-blocker medication use was also evaluated., Results: Heart rate rose more quickly in the handgrip group. At 6-10 min (peak handgrip), mean heart rate rose 51 +/- 14 beats/min in the handgrip group compared with 38 +/- 18 beats/min in the no handgrip group (p < 0.0001). With handgrip, overall dobutamine study time was reduced by a mean of 4.3 min (16.4 +/- 6.9 vs. 20.7 +/- 8.4, p = 0.004) in all patients, and by a mean of 5.9 min in patients not on beta-blocker medication (p = 0.001). The handgrip group also had a lower mean dose of dobutamine (25.8 +/- 13.5 vs. 32.4 +/- 16.4 mg, p = 0.025). The mean atropine dose was also lower (0.2 +/- 0.4 vs. 0.4 +/- 0.5 mg, p = 0.04). Handgrip exercise, however, did not decrease endpoints in patients on beta-blocker medication., Conclusions: Use of isometric handgrip exercise with DASE decreases time to target heart rate, recovery time, overall study time, and mean dosage of dobutamine and atropine. In patients not on beta-blocker medication, handgrip exercise should be routinely incorporated into all DASE protocols.

Objectives: The purpose of this study was to determine whether the dynamic cause for mitral systolic anterior motion (SAM) is a Venturi or a flow drag (pushing) mechanism., Background: In obstructive hypertrophic cardiomyopathy (HCM), if SAM were caused by the Venturi mechanism, high flow velocity in the left ventricular outflow tract (LVOT) should be found at the time of SAM onset. However, if the velocity was found to be normal, this would support an alternative mechanism., Methods: We studied with echocardiography 25 patients with obstructive HCM who had a mean outflow tract gradient of 82 +/- 6 mm Hg. We compared mitral valve M-mode echocardiogram tracings with continuous wave (CW) and pulsed wave (PW) Doppler tracings recorded on the same study. A total of 98 M-mode, 159 CW, and 151 PW Doppler tracings were digitized and analyzed. For each patient we determined the LVOT CW velocity at the time of SAM onset. This was done by first determining the mean time interval from Q-wave to SAM onset from multiple M-mode tracings. Then, CW velocity in the outflow tract was measured at that same time interval following the Qwave., Results: Systolic anterior motion began mean 71 +/- 5 ms after Q-wave onset. Mean CW Doppler velocity in the LVOT at SAM onset was 89 +/- 8 cm/s. In 68% of cases SAM began before onset of CW and PW Doppler LV ejection., Conclusions: Systolic anterior motion begins at normal LVOT velocity. At SAM onset, though Venturi forces are present in the outflow tract, their magnitude is much smaller than previously assumed; the Venturi mechanism cannot explain SAM. These velocity data, along with shape, orientation and temporal observations in patients, indicate that drag, the pushing force of flow, is the dominant hydrodynamic force that causes SAM.

From May 1992 to August 1993, 1,196 ticks were collected by flagging vegetation and from domestic animals in Chengde, Zhangjiakou, Shijiazhuang, and Handan districts of Hebei Province. Of those ticks collected, 954 were Haemaphysalis japonicum Warburton and H. longicornis Neumann, 205 were Ixodes persulcatus Schulze, and 37 were Dermacentor silvarum Olenev. Four strains of Borrelia burgdorferi were isolated from 135 I. persulcatus collected in Weichang and Zhuolu counties. Three of 574 Haemaphysalis spp. (collected in Laishui, Pingshan, and Xinglong counties) were positive for B. burgdorferi by direct immunofluorescent assay. One of 24 H. japonicum and 1 of 1 I. persulcatus (collected in Zhuolu) were positive for B. burgdorferi by polymerase chain reaction. This is the 1st report of a B. burgdorferi isolation from H. japonicum.
