Antioxidants (AO) inhibiting unspecific oxygen reactions are added to most susceptible industrial products, including liquid hydrocarbon-based aviation fuels. Regulatory approved jet fuel, whether synthetic or crude oil-based, requires the addition of performance antioxidants to inhibit radical-induced oxidation during storage and transport. While presently used antioxidants are petroleum-based, there have been no sustainable, less-toxic, bio-based equivalents reported so far. This study addresses the initial evaluation of carotenoid-based antioxidants as a substitution for current alkyl-phenolic antioxidants to kerosene. Inspired from natural products of Brevibacterium and Synechococcus genus synthesized analogues of candidate aromatic carotenoid cleavage products were evaluated for their effects on combustion characteristics of jet fuel, comparative radical scavenging, and endothelial cell-culture cytotoxicity. These bioinspired antioxidants show no adverse effect on jet fuel bulk properties and combustion chemistry. The radical scavenging properties of the evaluated phenolic compounds are superior to non-aromatic β-carotene cleavage products and in range with current alkyl-phenolic additives. Cytological assays demonstrated low toxicity towards human endothelial cells similar to butylated hydroxytoluene (BHT), one of the approved alkyl-phenol based additives to jet fuel, food, and cosmetics. The initial data set suggests, that the evaluated bioinspired compounds do not interfere with normal operations of jet engines and human fuel handling. Further developments towards the sustainable, biotechnological production and application of aromatic carotenoid cleavage products as antioxidants could contribute to reduced non-renewable consumption and possibly being an environmentally more compatible alternative due to their biosynthetic origin.

Background: The task of the Competence Centers for vocational training (KW) is to increase the attractiveness and quality of vocational (=post-graduate) training in general practice. For this purpose, they offer, among other things, a structured seminar program for post-graduate trainees in general practice (GP-trainees). During the Covid-19 pandemic the seminar program of the KWBW-Verbundweiterbildungplus® in Baden-Württemberg was converted to digital formats. The goal of the paper is to evaluate the acceptance by the GP-trainees and lecturers, to describe experiences with the conversion to e-learning and to derive recommendations with regard to the future orientation of seminar programs in post-graduate as well as continuing medical education. The implementation was based on a modified Kern-cycle and aimed at offering eight teaching units of 45 minutes each to a large number of GP-trainees. It tried to maintain the high quality of content and education as well as the interactive character of the previous seminars. For this purpose, the events were designed as synchronous webinars (six units) with asynchronous preparation and post-processing (two units) according to the flipped classroom method. The evaluation by the participating GP-trainees and lecturers was performed online using a multi-center developed and pre-piloted questionnaire. Results and discussion: N=101 GP-trainees participated in the evaluation of five individual seminar days in the second quarter of 2020 (response rate 97%). 58% (N=59) of the trainees were satisfied or very satisfied with the implementation. 82% (n=83) rated pre-tasks as helpful. 99% (n=100) would participate in an online seminar again. For 52% (n=53) of the trainees, the attitude towards e-learning had changed positively. The main advantages mentioned were no travel, save in time and costs as well as increased flexibility. The main disadvantages mentioned were less personal interaction and technical obstacles. The high acceptance of the new digital format showed the fundamental potential of e-learning in continuing medical education. The experiences can be a source of inspiration for other departments and KW. However, it also shows that important goals of KW, such as the personal interaction of the peer group, could not be achieved. In the future, it is important to develop a suitable mix of presence and digital formats with the aim to improve the attractiveness as well as sustainability of continuing medical education., Hintergrund: Um die Attraktivität und Qualität der Weiterbildung in der Allgemeinmedizin zu steigern, bieten die Kompetenzzentren Weiterbildung (KW) u.a. ein strukturiertes Seminarprogramm für Ärztinnen und Ärzte in Weiterbildung (ÄiW) an. Im Zuge der Covid-19-Pandemie wurde das Seminarprogramm der KWBW-Verbundweiterbildungplus® in Baden-Württemberg auf digitale Formate umgestellt. Ziel der Arbeit ist es, die Akzeptanz bei ÄiW und Dozierenden zu evaluieren, die Erfahrungen bei der Umstellung auf E-Learning zu beschreiben und daraus Empfehlungen abzuleiten im Hinblick auf die zukünftige Ausrichtung von Seminarprogrammen in der Facharztweiterbildung. Die Umsetzung orientierte sich an einem modifizierten Kern-Zyklus und hatte zum Ziel, einer großen Zahl von ÄiW weiterhin acht Unterrichtseinheiten (UE) à 45 min anbieten zu können und dabei die hohe inhaltliche und didaktische Qualität sowie den interaktiven Charakter der bisherigen Seminare beizubehalten. Dazu wurden die Veranstaltungen als synchrone Webinare (sechs UE) mit asynchroner Vor- und Nachbereitung (zwei UE) im Sinne der flipped classroom Methode konzipiert. Die Evaluation durch die teilnehmenden ÄiW und Dozierenden erfolgte online mittels eines multizentrisch entwickelten und vorab pilotierten Fragebogens. Ergebnisse und Diskussion: An der Evaluation der fünf Einzelseminartage im zweiten Quartal 2020 nahmen N=101 ÄiW teil (Rücklauf 97%). 58% (N=59) der ÄiW waren mit der Umsetzung zufrieden oder sehr zufrieden. 82% (N=83) empfanden die Vorbereitungsaufgaben als hilfreich. 99% (N=100) würden erneut an einem Online-Seminar teilnehmen. Bei 52% (N=53) der ÄiW hat sich die Einstellung gegenüber E-Learning positiv geändert. Vorteile ergäben sich v.a. durch den Wegfall der Anreise, Zeit- und Kostenersparnisse sowie eine erhöhte Flexibilität. Als nachteilig wurden insbesondere der geringere persönliche Austausch und technische Hürden genannt. Die hohe Akzeptanz des neuen digitalen Formats zeigt das grundsätzliche Potenzial von E-Learning in der ärztlichen Weiterbildung. Die Erfahrungen können Impulsgeber für andere Fachbereiche und KW sein. Allerdings zeigt sich auch, dass wichtige Ziele der KW, wie der persönliche kollegiale Austausch, damit nicht erreicht werden können. A holistic bio-process based on the microbial upcycling of low-value feedstocks and leading to chemo-enzymatically derived, functionalized taxanoids was established. The upcycling of biotechnological by-product streams glycerol and corn steep liquor by an engineered Escherichia coli strain that constitutively expressed bottleneck enzymes within the MEP-pathway led to the formation of 364.4 ± 10.7 mg L−1 taxadiene within 44 h. In contrast to standard inducible systems, our constitutive microbial production system provided concomitant growth and taxadiene formation. This strategy is the basis for subsequent continuous taxadiene production processes, which is favorable under economic constraints. The growth dependent taxadiene production showed improved yields, reproducibility and transferability at any scale compared to inducible taxadiene production platforms examined in this study. Additionally, we developed new taxadiene isolation and purification strategies. To that end, these new downstream processing strategies display efficient alternatives to conventional biphasic, in situ extraction and purification procedures. Specifically, we developed a rapid and easy two-step extraction procedure followed by centrifugal partition chromatography purification. This process strategy provided 249.0 ± 11.1 mg L−1 taxadiene with 95% purity from E. coli high cell-density cultures at a liter scale. To functionalize taxadiene, a mild lipase mediated epoxidation reaction was devised. Optimization of this biology-oriented synthesis strategy led to a quantitative conversion of taxadiene to taxa-4(5),11(12)-bisepoxide with an enantiomeric excess of over 83%. The holistic process strategy afforded in excess of 215 mg taxa-4(5),11(12)-bisepoxide by upcycling the low-value feedstocks glycerol (300 g) and corn steep liquor (25 g).

The consumption of energy, water and detergents during the washing process of laundry in private households in Germany is remarkably high, with over 5.5 billion kWh of electricity and 380 million m3 of water used every year. In this paper, the work of the research team Simulation Waschepflege (“Laundry care simulation”) is presented. This includes insights into the research steps simulation, experimental work and physical textile testing. The main focus is on results from a consumer survey conducted to gain a better understanding of the education level of consumers with respect to laundry care. The washing behavior of 840 consumers in Germany was analyzed in 2012. Consumers were asked questions about themselves and their households, washing machines and practices, laundry sorting and ideas for future washing technologies.

With over 50.000 identified compounds terpenes are the largest and most structurally diverse group of natural products. They are ubiquitous in bacteria, plants, animals and fungi, conducting several biological functions such as cell wall components or defense mechanisms. Industrial applications entail among others pharmaceuticals, food additives, vitamins, fragrances, fuels and fuel additives. Central building blocks of all terpenes are the isoprenoid compounds isopentenyl diphosphate and dimethylallyl diphosphate. Bacteria like Escherichia coli harbor a native metabolic pathway for these isoprenoids that is quite amenable for genetic engineering. Together with recombinant terpene biosynthesis modules, they are very suitable hosts for heterologous production of high value terpenes. Yet, in contrast to the number of extracted and characterized terpenes, little is known about the specific biosynthetic enzymes that are involved especially in the formation of highly functionalized compounds. Novel approaches discussed in this review include metabolic engineering as well as site-directed mutagenesis to expand the natural terpene landscape. Focusing mainly on the validation of successful integration of engineered biosynthetic pathways into optimized terpene producing Escherichia coli, this review shall give an insight in recent progresses regarding manipulation of mostly diterpene synthases.

Background Therapeutic fasting may improve health-related quality of life (HRQoL) and sleep but is not applicable for everyone. Time-restricted eating (TRE) offers a low threshold alternative but research on associations with HRQoL and sleep is rare. Methods We conducted a secondary analysis of two pilot studies in a pre-post design, which examined TRE in healthy employees at the Ulm University and in abdominal obese patients in a general practitioners office. Participants reported their HRQoL (EQ-5D visual analogue scale) before and after 3 months of restricting their daily eating to 8–9 h. They kept a diary to protocol timing of first and last meal, sleep quality (analogue scale) and duration. Pearson’s correlation coefficient was applied to test bivariate correlations between continuous variables and linear regression analyses were conducted to identify associated factors with the pre-post differences in HRQoL and the differences in sleep quality. Results Ninety-nine participants (aged aged 48.9 ± 1.1, 83.8% female) reached the fasting target of 15–16 h on average on 77.2 ± 18.7% of all recorded days. HRQoL increased by 7.8 ± 12.6 and sleep quality by 9.6 ± 13.9 points, but sleep duration was not extended. Regression analysis revealed mean fasting duration and baseline sleep quality as significant factors associated with changes in HRQoL. Improvements in sleep quality correlated with baseline sleep quality and HRQoL at follow-up but not with fasting. Changes in anthropometry did not correlate with the HRQoL or sleep quality. Conclusions TRE correlates with increased HRQoL and sleep quality independent from weight loss. TRE is easily applicable with or without medical supervision. The potential effects of TRE on health and sleep should be further investigated in larger randomized trials. Trial registration German Register for Clinical Trials (DRKS), DRKS-ID: DRKS00015057. Leishmaniaspecies cause a spectrum of human diseases in tropical and subtropical regions of the world. We have sequenced the 36 chromosomes of the 32.8-megabase haploid genome ofLeishmania major(Friedlin strain) and predict 911 RNA genes, 39 pseudogenes, and 8272 protein-coding genes, of which 36% can be ascribed a putative function. These include genes involved in host-pathogen interactions, such as proteolytic enzymes, and extensive machinery for synthesis of complex surface glycoconjugates. The organization of protein-coding genes into long, strand-specific, polycistronic clusters and lack of general transcription factors in theL. major, Trypanosoma brucei, andTrypanosoma cruzi(Tritryp) genomes suggest that the mechanisms regulating RNA polymerase II–directed transcription are distinct from those operating in other eukaryotes, although the trypanosomatids appear capable of chromatin remodeling. Abundant RNA-binding proteins are encoded in the Tritryp genomes, consistent with active posttranscriptional regulation of gene expression.

The parallel density functional program PARAGAUSS has been extended by a tool for computing solvent effects based on the conductor-like screening model (COSMO). The molecular cavity in the solvent is constructed as a set of overlapping spheres according to the GEPOL algorithm. The cavity tessellation scheme and the resulting set of point charges on the cavity surface comply with the point group symmetry of the solute. Symmetry is exploited to reduce the computational effort of the solvent model. To allow an automatic geometry optimization including solvent effects, care has been taken to avoid discontinuities due to the discretization (weights of tesserae, number of spheres created by GEPOL). In this context, an alternative definition for the grid points representing the tesserae is introduced. In addition to the COSMO model, short-range solvent effects are taken into account via a force field. We apply the solvent module to all-electron scalar-relativistic density functional calculations on uranyl, UO22+, and its aquo complexes in aqueous solution. Solvent effects on the geometry are very small. Based on the model [UO2(H2O)5]2+, the solvation energy of uranyl is estimated to be about −400 kcal/mol, in agreement with the range of experimental data. The major part of the solvation energy, about −250 kcal/mol, is due to a donor–acceptor interaction associated with a coordination shell of five water ligands. One can interpret this large solvation energy also as a compounded effect of an effective reduction of the uranyl moiety plus a solvent polarization. A calculation of the optical conductivity and related optical properties of liquid silicon is presented. Starting from the results of a single superchain/effective medium approximation for the electronic properties, the frequency dependent conductivity is evaluated in terms of the Kubo-Greenwood formula. For a fast computation of the three and four centre terms therein the effective handling of the angular dependences of the Green's functions known from the sp³-fluid model is generalized to the current matrix elements. A good agreement of our results with experimental data for liquid silicon is found. At higher frequencies (>5 eV) and in the fitted Drude parameters, however, there are discrepancies due to the limitations of the sp³-model.

In order to combine the advantages of tight-binding integral equation methods with basis sets not restricted to the lowest two orbital quantum numbers we extend the molecular fluid approach for the sp3 model of Lomba et al to arbitrarily high orbital quantum numbers. As an example, we present a single-superchain/effective-medium calculation of the electronic density of states for liquid silicon within a tight-binding approximation that includes, besides the 3s and 3p orbitals, also the 3d orbitals. Comparison with results from the sp3 model and different molecular dynamics results shows that within this sp3 d5 model very good agreement with plane-wave calculations is found whereas the sp3 model leads to unsatisfactory results.

By Thiophenols Mediated Conversion of Semisquaric Hydrazides to Squaric-1,2-Diamides In the presence of thiophenols semisquaric hydrazides (4) are converted into squaric-1,2-diamides (6) upon heating in alcoholic solution. Proof is given that thiosquaric-S-ester-amides (8) are intermediates in this process.

The reaction of semisquaric hydrazides 5a-f, with thiols 6a-j, 12, 14 has been investigated. Depending upon the substitution pattern of the educts, the solvent, and the reaction temperature the S-alkyl and S-aryl thiosquarate amides 8a-m, 11a-g, 13, 15 were obtained

In the present study, we report for the first time on the detection of bovine herpesvirus type 1 (BHV-1) in whole-blood samples derived from naturally infected cattle. Sensitive PCR assays specific for glycoprotein B (gB), gC, and gE of BHV-1 allow the detection of one BHV-1 DNA copy in 10 5 to 10 7 peripheral blood leukocytes (PBLs). The incidence of BHV-1-positive PBLs in naturally infected cattle appears to be quite high (92.2% positive PBLs among all samples tested), although in most cases only between 10 −5 and 10 −7 positive leukocytes were present. The results demonstrate that the viral DNA is detectable not only in the peripheral blood of acutely infected animals but, more importantly, also in the peripheral blood of subclinically infected cattle. The gE-specific PCR described in the report allows discrimination between wild-type (WT) virus-infected and vaccinated animals, which is of importance for control programs that use the recently introduced vaccination strategy with a gE-negative virus. The results further show that doubtful serological results can be verified or falsified and that individual animals can be monitored for the presence or absence of WT BHV-1 or gE-negative virus in cattle herds. The PCR protocols allow the detection of BHV-1 prior to seroconversion or in BHV-1-seronegative cattle. Finally, the results indicate the simultaneous presence of WT and gE-negative vaccine virus in the PBLs of several cattle. Therefore, investigations of viremia in naturally and experimentally infected cattle and on the identification of infected cell types of bovine PBLs can be now performed.
