Abstract

In Solanaceae family several plant resistant genes to pathogen (R-genes) have been mapped and cloned. Most of them encode Nucleotide Binding Site Leucine Rich Repeat domain (NBS-LRR) protein. However, little is known about the resistance genes variability pattern and the evolutionary process acting on different species belonging to the same family. The aims of the present work, was to genotype and study the evolutionary relationship of fifty wild tomato accessions using the I2 resistance gene sequences. Thirty-three new candidate homologues I2 resistance gene nucleotide sequence were obtained from wild tomato species. Nucleotide polymorphisms in I2-NBS domain was detected in wild tomato species: diversity could have accumulated over a long time and species sorting could have produced new variants. In order to study the NBS-LRR domain variability we analyzed the evolution process acting on the amino acid sequence. The FEL method (codon Model) based on dN/dS, was used to estimate the presence of positive, negative and neutral selection acting on each codon. The I2-NBS domain sequence data studied seems to be under a general purification process of evolution. However, intermittent bouts of positive selection sites were detected in high variable regions. Phylogenetic analysis conducted within the Solanaceae family shows that the Solanum genus is under a rapid adaptative divergence process and Nicotiana and Capsicum clustered separately; Solanum peruvianum, in particular, displayed to be the most polymorphic specie. These results might be important for the identification of new sources of resistance genes to tomato pathogens.

Abstract

Total ascorbic acid (total-AsA) content and percentage of reduced ascorbic acid (AsA) on total-AsA were investigated and discussed in fruits, leaves, petiole, stem and roots of the Solanum lycopersicum cv M82 and S. pennellii introgression lines IL7-3, IL10-1 and IL12-4. In fruits, total-AsA content showed to be different according to genotype analysed. Higher total-AsA accumulations were observed for IL7-3 and IL12-4 followed by M82 and IL10-1. Total-AsA was generally higher in leaves than in petioles, stems and roots of all genotypes. In roots higher total-AsA concentration and lower AsA percentage was generally observed in introgression lines than M82, in particular for IL7-3.

37. Selection for aneuploid potato hybrids combining low wild genome content and resistance traits from Solanum commersonii

Abstract

A breeding scheme based on the production of progenies with odd ploidy was followed to introduce useful genes from the wild Solanum commersonii (cmm) into S. tuberosum (tbr) genome. Hybrids from 5x x 4x crosses were characterized for traits of interest, and selection was assisted by amplified fragment length polymorphism (AFLP) analysis. As expected, most of the hybrids were aneuploids, with a trend towards a low degree of aneuploidy. Despite the fact that aneuploidy has often been associated with a reduction in male and female fertility, most of the hybrids were fertile following crosses with tbr, making it possible to produce viable offspring. A screening for resistance traits deriving from cmm was also carried out. With respect to freezing resistance, the killing temperatures of cold-acclimated genotypes were distributed between the wild and cultivated parental values, with some hybrids displaying an acclimation capacity higher than 3 degreesC. A wide variability was also found for tuber soft rot resistance, and hybrids with high levels of resistance were identified. Selection of hybrids was based on a two-stage scheme that consisted of conventional phenotypic selection followed by an estimation of the wild genome content still present in order to identify hybrids combining noteworthy traits with a low wild genome content. Previously selected cmm-specific AFLPs were used to monitor the degree of wild genome content still present in each hybrid. The percentage of cmm-specific markers ranged from 59% to 91%, with an average value of 75%. AFLP analysis was employed to assist in the selection of valuable hybrids for further breeding efforts.
