Efforts to reduce organic sulfur in coal are taken through biodesulfurization by using desulfurization bacteria to release covalently-bound sulfur from the coal matrix. Coal is a complex hydrocarbon material that requires collaboration from more than one type of bacteria in a consortium for desulfurization. The current study shows how the individual members of a bacterial consortium obtained directly from coal samples grew on the coal. Mineral medium containing sub-bituminous coal with a concentration of 10%, 15%, and 20% served as a carbon source and the only sulfur to support the consortium's growth. The examination included growth patterns, concentrations of dibenzothiophene as an organic sulfur representative, pH, and sulfate concentration as the sulfur product released into the medium. The growth of individual members of the consortium was observed for 336 h. The consortium grew in all three coal concentrations with slightly different cell growth patterns and the release of dibenzothiophene. Members of the consortium grew alternately and overlapped, which showed possible linkages or dependence on products and existence from the growth of other members. The existence of the primary strain Moraxella osloensis COK1 indicated that they played a role in the activities and growth of other members. The alternating growth is discussed to produce a hypothetical illustration of how several other members play in using sulfur in a well-known desulfurization pathway. In conclusion, this study provides a deeper insight into the value of consortium members individually but growing together while swarming coal as a complex resource to become low-sulfur coal.

A total of 137 strains of mesophilic and psychrotolerant hydrocarbon-oxidizing bacteria have been isolated from three regions of the Sea of Japan exposed to different levels of anthropogenic pollution. The taxonomic affiliations of the culturable bacteria from surface waters and bottom sediments that are involved in the biodegradation of petroleum hydrocarbons in these regions have been identified. The isolated microorganisms belong to the phyla Actinobacteria, Firmicutes, and Proteobacteria. The hydrocarbon-oxidizing ability of the bacteria Okibacterium sp., Lechevalieria flava, Patulibacter sp., P. minatonensis, Gracilibacillus massiliensis, Thalassobacillus sp., Virgibacillus dokdonensis, Chryseomicrobium amylolyticum, Jeotgalibacillus marinus, Moraxella osloensis, Idiomarina maritima, and I. piscisalsi has been recorded for the first time. The results we obtained extend our knowledge about the potential of biodegradation of pollutants by members of certain taxa in the marine environment.

Bacteria can establish beneficial interactions with plants by acting as growth promoters and enhancing stress tolerance during plant interactions. Likewise, bacteria can develop multispecies communities where multiple interactions are possible. In this work, we assessed the physiological effects of three bacteria isolated from an arid environment (Bacillus niacini, Bacillus megaterium, and Moraxella osloensis) applied as single species or as a consortium on oregano (Origanum vulgare L.) plants. Moreover, we assessed the quorum-sensing (QS) signaling activity to determine the molecular communication between plant-growth-promoting bacteria. The plant inoculation with B. megaterium showed a positive effect on morphometric and physiologic parameters. However, no synergistic effects were observed when a bacterial consortium was inoculated. Likewise, activation of QS signaling in biofilm assays was observed only for interspecies interaction within the Bacillus genus, not for either interaction with M. osloensis. These results suggest a neutral or antagonistic interaction for interspecific bacterial biofilm establishment, as well as for the interaction with oregano plants when bacteria were inoculated in a consortium. In conclusion, we were able to determine that the bacterial interactions are not always positive or synergistic, but they also might be neutral or antagonistic.

Background The gram-negative aerobic bacterium Moraxella osloensis is an opportunistic pathogen in brain tissues. Case summary The gram-negative aerobic bacterium Moraxella osloensis was isolated from a patient's brain tissue during a stereotactic biopsy. Conclusion This is the first report of a brain tissue infection with Moraxella osloensis possibly causing brain gliomatosis.

In Hungary, which is famous for its thermal baths, according to the regulations, waters are investigated in hygienic aspects with standard cultivation methods. In the present study, two thermal baths were investigated (the well and three different pool waters in both) using cultivation methods, taxon-specific polymerase chain reactions (PCRs), multiplex PCRs and next-generation amplicon sequencing. Mainly members of the natural microbial community of the well waters and bacteria originating from the environment were detected but several opportunistic pathogenic taxa, e.g., Pseudomonas aeruginosa, P. stutzeri, Acinetobacter johnsoni, Acinetobacter baumanni, Moraxella osloensis, Microbacterium paraoxydans, Legionella spp., Stenotrophomonas maltophilia and Staphylococcus aureus were revealed by the applied methods. Pools with charging-unloading operation had higher microscopic cell counts, colony-forming unit (CFU) counts, number of cocci, P. aeruginosa and S. aureus compared to the recirculation systems. Bacteria originating from human sources (e.g., skin) were identified in the pool waters with less than 1% relative abundance, and their presence was sporadic in the pools. Comparing the microbiological quality of the pools based on the first sampling time and the following four months' period it was revealed that recirculation operation type has better water quality than the charging-unloading pool operation from a hygienic point of view.

Two East Asian finless porpoises (Neophocaena asiaeorientalis sunameri) in Ningbo, East China Sea, China, were observed to be bacterially infected between the fat layer and muscle layer. The microbial communities in pus samples were identified by metagenome sequencing 16S in the PacBio platform and explored the relationship between emaciation and bacterial infection. The present paper was the first report on bacterial diversity in infected tissue pus of finless porpoise. In total, 101 bacterial species were identified, and the top nine species were Rhodococcus qingshengii (26.25%), Rhodococcus jialingiae (22.43%), Ralstonia pickettii (16.03%), Moraxella osloensis (9.97%), Psychrobacter cibarius (2.97%), Flavobacterium johnsoniae (2.11%), Flavobacterium chungbukense (1.80%), Stenotrophomonas maltophilia (1.73%) and Serratia marcescens (1.62%). These main bacteria could cause various diseases or metabolic disorder, such as endocarditis, meningitis, bacteraemia, septicaemia, and so on; they also involve microbiome metabolism of amino acids (12.47%), carbohydrates (11.58%), xenobiotic biodegradation (7.81%), lipids (6.01%) and energy (4.98%). Regardless of the type of disease metabolic disorder will firstly cause body weight loss. In conclusion, these bacteria could cause diseases or metabolic disorders, resulting in emaciation of East Asian finless porpoises. Admittedly, pathogenic assay is furthermore needed to determine the mechanisms regarding the pathological phenomena. The gut microbiota of insects usually plays an important role in the development and reproduction of their hosts. The fecundity of Henosepilachna vigintioctopunctata (Fabricius) varies greatly when they develop on different host plants. Whether and how the gut microbiota regulates the fecundity of H. vigintioctopunctata was unknown. To address this question, we used 16S rRNA sequencing to analyze the gut microbiomes of H. vigintioctopunctata adults fed on two host plant species (Solanum nigrum and Solanum melongena) and one artificial diet. The development of the ovaries and testes was also examined. Our results revealed that the diversity and abundance of gut microorganisms varied significantly in insects reared on different diets. The gut microbiota of H. vigintioctopunctata raised on the two host plants was similar, with Proteobacteria being the dominant phylum in both groups, whereas Firmicutes was the dominant phylum in the group reared on the artificial diet. The predominant microbiota in the S. nigrum group were Acinetobacter soli and Acinetobacter ursingii (Acinetobacter, Moraxellaceae); Moraxella osloensis (Enhydrobacter, Moraxellaceae); and Empedobacter brevis (Empedobacter, Weeksellaceae). The microbiota in this group are associated with high lipid metabolism. In addition, the beetles’ ovaries and testes were more highly developed in the S. nigrum group than in the other two groups. These findings provide valuable information for elucidating the complex roles the gut microbiota play in the fecundity of H. vigintioctopunctata, and may also contribute to developing future novel control strategies involving this economically important pest.

BackgroundThe purpose is to study the metagenomics of the microbes isolated from the lacrimal sac of patients with primary acquired nasolacrimal duct obstruction (PANDO).MethodsA prospective study was performed on ten consecutive lacrimal sac samples obtained for the metagenomic analysis from the patients with PANDO, who underwent endoscopic dacryocystorhinostomy at a tertiary care Dacryology service. The samples were collected intraoperatively soon after a full-length lacrimal sac marsupialisation and immediately transported on ice to the laboratory. Following DNA extraction and library preparation, a whole shotgun metagenome sequencing was performed on the Illumina platform. The downstream processing and bioinformatics of the samples were performed using multiple software packaged in the SqueezeMeta pipeline and MG-RAST pipeline.ResultsThe taxonomic hit distribution across the samples showed that bacteria were the most common isolates (mean—97.56%), followed by viruses (mean—0.29%), archaea (0.04%) and others. The five major phyla identified across the samples of PANDO were proteobacteria, Bacteroidetes, Fusobacteria, Actinobacteria and Firmicutes. The prevalent organisms includeAcinetobacter johnsonii, Porphyromonas catoniae, Cutibacterium acnes, Pseudomonas alcaliphila, Escherichia coli, Haemophilus influenzae, Enhydrobacter aerosaccus, Fusobacterium nucleatum, Moraxella osloensis, Butyricimonas virosaandVariovorax paradoxus,among few others. The alpha diversity of the ten sample datasets ranged from 60 to 175 species.ConclusionThis is the first whole metagenome sequencing of the lacrimal sac contents from PANDO patients. Lacrimal sacs harbour diverse microbial communities, including bacteria, viruses, and archaea. Further Lacriome studies may provide clues for a better understanding of the disease aetiopathogenesis.

Background Moraxella osloensis rarely causes infection in humans, and most of the reported cases are not fatal. It is often difficult to identify M. osloensis using conventional biochemical methods. Methods Here, we report a bacteremia case caused by M. osloensis in a patient with advanced lung cancer who initially presented symptoms of fever. Results Blood culture revealed growth of a gram-negative bacterium, which was identified as M. osloensis through 16S rRNA gene sequencing and MALDI-TOF analyses. The patient could not recover from sepsis with empirical treatment. Conclusions As M. osloensis can cause serious infections in immunocompromised patients, its prompt identification is important.

As commodity virtual reality (VR) systems become more common, they are rapidly gaining popularity for entertainment, education, and training purposes. VR utilizes headsets which come in contact with or close proximity to the user’s eyes, nose, and forehead. In this study, the potential for these headsets to become contaminated with bacteria was analyzed. To the best of our knowledge, this study is the first to address the potential for microorganisms to be transmitted via VR headsets. The data discussed herein were collected roughly one year prior to the outbreak of the COVID-19 pandemic in the United States. We feel it is important to be clear that this study focuses exclusively on bacteria, as opposed to viruses like those responsible for the present pandemic. The nosepieces and foreheads of two HTC Vive headsets were sampled over the course of a seven-week period in a VR software development course. Serial dilutions were performed, and samples were plated on various culture media. Following incubation, counts of bacteria were determined. DNA was extracted from bacterial colonies and the 16S rRNA gene was sequenced to identify bacterial contaminates present on the headsets. Chief among these contaminates was Staphylococcus aureus. The results of these tests indicated that the Staphylococcus aureus strains isolated from the headsets possessed high levels of antibiotic resistance. Other notable bacterial isolates included Moraxella osloensis, the bacteria responsible for foul odors in laundry and, Micrococcus luteus, a communalistic bacterial species capable of causing opportunistic infections. Other bacterial isolates were detected in variable amounts throughout the trial.

Moraxella osloensis has been reported in the literature as a human pathogen, particularly among immunocompromised adults. In contrast to the adult population, most pediatric cases are among patients with no underlying immunological defect; however, no patient underwent further investigation and no data about the long-term follow-up are available. We report the case of a 2-month-old previously healthy girl infected with Moraxella osloensis. Here, we review case reports and case series of children and adults with Moraxella osloensis infection and compare them with our experience. On the basis of our findings, we recommend further investigations (immunological or other underlying diseases) when a child is found to be infected with these bacteria.

Soleha S, Retnaningrum E. 2020. Screening and molecular identification of lipolytic bacteria from spent bleaching earth. Biodiversitas 21: 4155-4161. Lipolytic bacteria can produce nanobioremediation materials that encourage researchers to explore the diversity of these bacteria. Lipolytic bacteria were isolated from spent bleaching earth (SBE). Bacteria isolates were screened for their lipolytic index and crude lipase activity. Strain with the highest lipolytic index and crude lipase activity were identified using the 16S rRNA gene as genetic markers. The maximum likelihood method in MEGA6 software has been used to analyze the relationship of the isolate. The results showed that strain SBE01 had the highest lipolytic index and crude lipase activity of 8.8 ± 0.7 mm and 5.7 U/mL, respectively. Based on 16S rRNA gene analysis, the isolate was identified as Moraxella sp. Phylogenetic tree was shown that SBE01 isolate was closely related to Moraxella osloensis. Moraxella sp. has the potential to be used as a bioremediation agent from oil-contaminated environment.

We report the complete genome sequence of Moraxella osloensis strain YV1, which was isolated from a wastewater treatment plant in Australia. The YV1 genome comprises a 2,615,801-bp chromosome and four plasmids. Moraxella osloensis strain YV1 displays the distinctive morphology of Eikelboom morphotype 1863.

Objectives To describe the bacterial and fungal microbiota of the conjunctiva and factors influencing these microbiota of healthy cats. To evaluate the antimicrobial resistance profile and discuss the use of appropriate antimicrobial drugs. Animals studied One hundred and twenty healthy cats PROCEDURES: Conjunctival samples with dry or premoistened swabs were obtained from both eyes and cultured for aerobic and anaerobic bacteria and fungi. In vitro bacterial, susceptibility testing was performed. The effects of age, sex, breed, season, living environment, and sample collection technique on the frequency and composition of microbial isolation were evaluated. Results In 49 of 120 cats (40.8%) and 73 of 240 swabs (30.4%), microorganisms were isolated. Of the isolates, 71% (61/86) were Gram-positive bacteria, 26% (22/86) were Gram-negative bacteria, and 3% (3/86) were fungi. Staphylococcus felis (17/86; 19.8%) was the most commonly isolated species. Moraxella osloensis (5/86; 5.8%) was the most frequent Gram-negative species. The season had a statistically significant influence (P = 0.04) on the frequency of isolation. The use of premoistened swabs increased the rate of Gram-positive bacterial detection significantly (P = 0.03). The in vitro susceptibility testing showed high efficacy of chloramphenicol, gentamicin, pradofloxacin, and enrofloxacin. Conclusions The isolated microbiota aligns with previous studies performed in other countries, although the microbiota of cats in the present study showed wider bacterial diversity, and more Gram-negative species were isolated. Swab sampling should be performed with premoistened swabs. The topical antimicrobials gentamicin and chloramphenicol are suitable therapeutics for first-line treatment.

Objectives To determine the occurrence of mcr-1 and mcr-2 genes in Gram-negative bacteria isolated from healthy pigs in Great Britain. Methods Gram-negative bacteria (n = 657) isolated from pigs between 2014 and 2015 were examined by WGS. Results Variants of mcr-1 and mcr-2 were identified in Moraxella spp. isolated from pooled caecal contents of healthy pigs at slaughter collected from six farms in Great Britain. Other bacteria, including Escherichia coli from the same farms, were not detected harbouring mcr-1 or mcr-2. A Moraxella porci-like isolate, MSG13-C03, harboured MCR-1.10 with 98.7% identity to MCR-1, and a Moraxella pluranimalium-like isolate, MSG47-C17, harboured an MCR-2.2 variant with 87.9% identity to MCR-2, from E. coli; the isolates had colistin MICs of 1-2 mg/L. No intact insertion elements were identified in either MSG13-C03 or MSG47-C17, although MSG13-C03 harboured the conserved nucleotides abutting the ISApl1 composite transposon found in E. coli plasmids and the intervening ∼2.6 kb fragment showed 97% identity. Six Moraxella osloensis isolates were positive for phosphoethanolamine transferase (EptA). They shared 62%-64.5% identity to MCR-1 and MCR-2, with colistin MICs from 2 to 4 mg/L. Phylogenetic analysis indicated that MCR and EptA have evolved from a common ancestor. In addition to mcr, the β-lactamase gene, blaBRO-1, was found in both isolates, whilst the tetracycline resistance gene, tetL, was found in MSG47-C17. Conclusions Our results add further evidence for the mobilization of the mcr-pap2 unit from Moraxella via composite transposons leading to its global dissemination. The presence of mcr-pap2 from recent Moraxella isolates indicates they may comprise a reservoir for mcr.

application/pdf, A three-year-old spayed domestic short-haired cat presented for evaluation of weight loss, cardiomegaly and pleural effusion. Echocardiographic examination demonstrated a thickened pericardium with mild pericardial effusion and a large volume of pleural effusion characterized by exudate. Although the cat was treated with antibiotics, the clinical symptoms did not improve. The cat developed dyspnea and died on day 7. Necropsy revealed a large amount of modified transudates ascites, pleural effusion and markedly dilated pericardium. Histopathological examination revealed severe exudation of fibrin and granulation tissue in a thick layer of the epicardium. The cat was diagnosed with fibrinous pericarditis secondary to bacterial infection.

Peritonitis is a very serious complication encountered in patients undergoing peritoneal dialysis and healthcare providers involved in the management should be very vigilant. Gram-positive organisms are the frequent cause of peritonitis compared to gram-negative organisms. There has been recognition of peritonitis caused by uncommon organisms because of improved microbiological detection techniques. We report a case of peritonitis caused by Moraxella osloensis (M. osloensis), which is an unusual cause of infections in humans. A 68-year-old male, who has been on peritoneal dialysis for 2 years, presented with abdominal pain and cloudy effluent. Peritoneal fluid analysis was consistent with peritonitis and peritoneal fluid culture grew gram-negative bacteria. M. osloensis was identified by 16 S PCR phenotypic and sequencing techniques. Patient responded well to the treatment, with intraperitoneal cephalosporin, and repeat peritoneal fluid culture yielded no growth. M. osloensis rarely causes infection in humans and responds well to treatment, as reported in literature.

Moraxella osloensis has been reported in the literature as a rare cause of sepsis, central nervous system infection, chest infection, and endophthalmitis. In the present case, the organism was isolated from the blood of an 8-year-old immunocompetent boy. It could not be identified with VITEK-2 and API-20NE (bioMerieux SA, Marcy-l'Etoile, France). Latex-based bacterial antigen test (BD Directigen Meningitis Combo Test; Becton, Dickinson and Company, Sparks, Maryland, United States) was positive for Neisseria meningitidis A/Y. It was identified as M. osloensis using matrix-associated laser desorption/ionization-time of flight mass spectrometry (MALDI-TOF) (Vitek MS, bioMerieux, Marcy-l'Etoile) (confidence level 99.9%) and 16S rRNA gene sequencing (99% sequence identity). The child responded well to ampicillin, and was discharged home after 4 days, on oral amoxicillin–clavulanic acid for the next 7 days. M. osloensis is a rare cause of bacteremia. It may masquerade as N. meningitidis in the routine tests performed in the routine microbiology laboratories. It is imperative to confirm the identification with MALDI-TOF or a molecular method to remove the false positive diagnosis of N. meningitidis, and to avoid the unnecessary use of prophylaxis with rifampicin or ciprofloxacin.

OBJECTIVE To compare the microbial flora of cholesteatoma and normal middle ears using gene-based sequencing analysis. STUDY DESIGN Controlled ex vivo human study. SETTING Academic, tertiary medical center. SUBJECTS AND METHODS Brush, swab, and tissue samples were each taken from cholesteatoma matrix and uninvolved tissue in patients with previously untreated, acquired cholesteatoma (n = 19) or middle ear mucosa from patients undergoing cochlear implantation with no history of cholesteatoma or previous middle ear surgery (control; n = 12). DNA was isolated from specimens then 16S rRNA gene sequencing was performed. RESULTS There was no difference in microbial yield between the sampling methods. The mouth is an important niche for bacterial colonization. Previous research used mouth microbiota to predict diseases like colon cancer and inflammatory bowel disease (IBD). It is still unclear how the sampling methodology influences microbial characterization. Our aim was to determine if the sampling methods, e.g., cotton swab or tissue biopsy, and the age influence the oral microbial composition of mice. Microbial DNA was extracted using a commercial kit and characterized targeting the 16s rRNA gene from mouth swabs and tissue biopsies from 2 and 15 months old C57BL/6 male mice kept in the same SPF facility. Our results show statistical different microbial community of the different ages, type of sampling, and the two fixed factors age x type of sample (p-value < 0.05). At the genus level, we identified that the genera Actinobacillus, Neisseria, Staphylococcus, and Streptococcus either increase or decrease in abundance depending on sampling and age. Additionally, the abundance of Streptococcus danieliae, Moraxella osloensis, and some unclassified Streptococcus was affected by the sampling method. While swab and tissue biopsies both identified the common colonizers of oral microbiota, cotton swabbing is a low-cost and practical method, validating the use of the swab as the preferred oral sampling approach.

Background Etiological diagnosis is a key to therapeutic adaptation and improved prognosis, particularly for infections such as endocarditis. In blood culture–negative endocarditis (BCNE), 22% of cases remain undiagnosed despite an updated comprehensive syndromic approach. This prompted us to develop a new diagnostic approach. Methods Eleven valves from 10 BCNE patients were analyzed using a method that combines human RNA bait-depletion with phi29 DNA polymerase-based multiple displacement amplification and shotgun DNA sequencing. An additional case in which a microbe was serendipitously visualized by immunofluorescence was analyzed using the same method, but after laser capture microdissection. Results Background DNA prevented any diagnosis in cases analyzed without microdissection because the majority of sequences were contaminants. Moraxella sequences were dramatically enriched in the stained microdissected region of the additional case. A consensus genome sequence of 2.4 Mbp covering more than 94% of the Moraxella osloensis KSH reference genome was reconstructed with 234X average coverage. Several antibiotic-resistance genes were observed. Etiological diagnosis was confirmed using Western blot and specific polymerase chain reaction with sequencing on a different valve sample. Conclusions Microdissection could be a key to the metagenomic diagnosis of infectious diseases when a microbe is visualized but remains unidentified despite an updated optimal approach. Moraxella osloensis should be tested in blood culture–negative endocarditis.

Attachment of psychrotrophic meat spoilage organisms to longissimus dorsi (1. dorsi) muscle placed in an attachment bath was studied. The numbers of bacteria (log10) attaching to 10.0 g of 1. dorsi muscle during a 20-min incubation period in the attachment bath ranged from 3.88 for Acinetobacter LD-2 to 5.66 for Pseudomonas fluorescens . The lowest attachment values were recorded with two non-motile bacteria, Acinetobacter LD-2 and Moraxella osloensis ; the highest, with two motile fluorescent pseudomonads, Pseudomonas putida and P. fluorescens . Attachment strength values (S values) ranged from 0.19 for P. putida to 0.70 for Acinetobacter LD-2. In general, the non-motile bacteria possessed higher S-values than the motile organisms. When two different microorganisms were in contact with the 1. dorsi muscle in the attachment bath, minimal competition for attachment to the meat occurred.

Herein we report the case of a 10-year-old boy with an autosomal mosaic mutation who developed bacteremia. The causative agent was identified as Moraxella osloensis by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry and 16S rRNA gene sequencing. In the pediatric population, there have been 13 case reports of infection attributed to M. osloensis and this is the fifth reported case of pediatric bacteremia due to M. osloensis. After Moraxella species infection was confirmed, the patient recovered with appropriate antimicrobial therapy. It is important to consider that M. osloensis can cause serious infections, such as bacteremia, in otherwise healthy children. Nematodes of the genus Phasmarhabditis (Nematoda: Rhabditidae) are world-wide distributed molluscs´ parasites. Species P. hermaphrodita is the only commercially produced bio-agent for slug control. The aim of this thesis is to evaluate some ecologic characteristics of three newly described species of the genus Phasmarhabditis (P. bohemica, P. bonaquaense a P. apuliae) in polyxenic and monoxenic cultures. Three bacterial isolates, Pseudomonas sp., Acinetobacter sp. and Flavobacterium sp. were chosen for testing the monoxenic cultures. The growing ability of tested nematodes, the effect on the quality of progeny and the rate of development were examined on various organic substrates (compost, faeces of Deroceras slugs, homogenized Deroceras slugs, homogenized larvae of Galleria mellonella and homogenized pig kidney). Subsequently, the growth of nematodes was tested in solid and liquid media. The last experiment was focused on evaluating the impact of polyxenic and monoxenic nematode cultures on the mortality and feeding activity of Deroceras reticulatum and Cepaea hortensis. As is assumed in the results chapter, all examined species of nematodes are lethal facultative parasites of Deroceras spp. They are able to grow in broad spectre of organic substrates and bacteria which influence the production of the progeny, but the qualitative parameters of dauer juveniles remain more or less unaffected.

There is an abundance of evidence that chronic inflammation is linked to prostate cancer (PC), and pathogen infection is considered to be one of the possible causes for PC. Prostate infections by bacteria or viruses have been widely investigated. However, most of these studies have showed conflicting and controversial results. Most studies have focused on the PCR-based targeted detection of bacteria or viruses, and pathogen for PC remains to be found. To identify pathogen for PC, we investigated the transcriptomes of human prostate samples using whole-genome next-generation sequencing. The prostate specimens obtained by radical prostatectomy from 20 PC patients were sliced and divided into four sections. High-quality RNAs were extracted from the sections including PC. RNA-seq libraries were constructed according to the Illumina protocol and next-generation sequenced on a HiSeq 2000 instrument. To explore the potential of other pathogens being associated with PC, we performed de novo assembly of unmapped reads from the RNA-seq data from 20 PC samples. RNA-seq analysis identified four bacteria as candidate pathogens, which included Cutibacterium acnes, uncultured Chroococcidiopsis, Micrococcus luteus, and Moraxella osloensis. Among these candidate pathogens, Cutibacterium acnes was detected in 19 of 20 PC tissue samples by immunohistochemistry. We also performed PCR to detect other candidate pathogens (uncultured Chroococcidiopsis, Micrococcus luteus, and Moraxella osloensis) in PC samples for validation. Our findings provide further evidence of Cutibacterium acnes infections in human PCs as a promising pathogen for PC, and suggest that uncultured Chroococcidiopsis, Micrococcus luteus, and Moraxella osloensis could be novel candidate pathogens for PC. Further research is required and ongoing to clarify the molecular mechanisms underlying bacterial pathogenesis of PC. Rushan cheese, an essential part of the Bai culture, has been produced and consumed for centuries by the Bai people living mostly in Yunnan province of China, however, studies on the naturally occurring microbial communities of Rushan cheese are lacking. In this study, we applied high throughput sequencing technique to analyze the microbial compositions of Rushan cheese samples from three different geographical origins (i.e., Weishan, Eryuan, and Jianchuan). The microbiota in Weishan, Eryuan and Jianchuan Rushan cheese samples were distinct in terms of taxonomic composition and abundance. Linear discriminant analysis (LDA) of effect size (LEfSe) analysis found the characteristic taxonomic species in Weishan Rushan cheese samples were Lactobacillus pentosus, Lactobacillus crustorum, Lactobacillus brevis, Leuconostoc mesenteroides, and Pediococcus pentosaceus; the representing taxonomic species in Eryuan Rushan cheese samples were Lactobacillus kefiranofaciens, Lactococcus lactis, Acetobacter pasteurianus and Moraxella osloensis; by comparison, Acinetobacter was enriched in Jianchuan Rushan cheese samples. Characterization of the microbial diversity in Rushan cheese samples from different geographical origins will contribute to the understanding of microorganisms responsible for the Rushan cheese fermentation, and enable us to develop bioresources derived from Rushan cheese in the future.
