Abstract

The International Fusion Materials Irradiation Facility (IFMIF), presently in its Engineering Validation and Engineering Design Activities (EVEDA) phase under the frame of the Broader Approach Agreement between Europe and Japan, accomplished in summer 2013, on schedule, its EDA phase with the release of the engineering design report of the IFMIF plant, which is here described. Many improvements of the design from former phases are implemented, particularly a reduction of beam losses and operational costs thanks to the superconducting accelerator concept, the re-location of the quench tank outside the test cell (TC) with a reduction of tritium inventory and a simplification on its replacement in case of failure, the separation of the irradiation modules from the shielding block gaining irradiation flexibility and enhancement of the remote handling equipment reliability and cost reduction, and the water cooling of the liner and biological shielding of the TC, enhancing the efficiency and economy of the related sub-systems. In addition, the maintenance strategy has been modified to allow a shorter yearly stop of the irradiation operations and a more careful management of the irradiated samples. The design of the IFMIF plant is intimately linked with the EVA phase carried out since the entry into force of IFMIF/EVEDA in June 2007.

Abstract

The ITER equatorial visible and infrared Wide Angle Viewing System (WAVS) will play a major role in the protection of plasma facing components by providing surface temperature measurements of these components. It will also image the plasma emission in the visible range. The WAVS is composed of 15 lines of sight located in four Equatorial Ports (EPs) 3, 9, 12, and 17. Its development is being carried out by the Consortium constituted by CEA, CIEMAT, INTA, and Bertin Technologies, within grant contracts financed by F4E. In the EP12, the in-vessel and ex-vessel components of the WAVS are at their final design phase. This article presents an overview of the opto-mechanical design of the WAVS in the EP12 presented at the final design review. [ABSTRACT FROM AUTHOR]

Abstract

The variational grid generation method is a powerful tool for generating structured convex grids on irregular simply connected domains whose boundary is a polygonal Jordan curve. Several examples that show the accuracy of a difference approximation to the solution of a Poisson equation using these kind of structured grids have been recently reported. In this paper, we compare the accuracy of the numerical solution calculated by applying those structured grids with finite differences against the the solution obtained with Delaunay-like triangulations on irregular regions.

20. The discovery of small molecule modulators of soluble guanylate cyclase aided by surface plasmon resonance

Abstract

Soluble guanylate cyclase is a multidimeric enzyme that regulates cardiovascular homeostasis and is the receptor for nitric oxide in the brain. The enzyme is the known target for a new agonist drug used for the treatment of pulmonary hypertension. Whilst drug discovery has been successful for the finding of small molecules that activate the enzyme, the currently available inhibitors lack selectivity as they act through oxidation of a heme prosthetic group in the enzyme, which is conserved amongst other hemeproteins. Nonetheless, it has been suggested that inhibition of soluble guanylate cyclase by small molecules could be useful in the treatment of neurological conditions such as Parkinson’s Disease. In this thesis, new activators of soluble guanylate cyclase have been identified by virtual screening, and a new class of inhibitors has been designed and synthesised. The synthetic routes developed are efficient and take advantage of microwave-assisted organic synthesis. The drug-target interaction was characterised using a biophysical technique based on surface plasmon resonance, which allows the detection of label-free binding between small molecules and biological macromolecules. The biophysical assay has been developed using different constructs of soluble guanylate cyclase and validated through binding of the natural ligands ATP and GTP. The instrument and assay design were validated using the well-defined interaction between natriuretic peptides and the extracellular domain of natriuretic peptide type-C. Additional biochemical characterisation of the ligands allowed discrimination between activators and inhibitors. This combination of biophysical and biochemical techniques allowed the identification of the catalytic domain of soluble guanylate cyclase as the target for binding of the new class of synthesised inhibitors and has given insight into the functional groups necessary for activity and binding to the enzyme.
