Feline infectious peritonitis (FIP) is a fatal disease of cats that currently lacks licensed and affordable vaccines or antiviral therapeutics. The disease has a spectrum of clinical presentations including an effusive ("wet") form and non-effusive ("dry") form, both of which may be complicated by neurologic or ocular involvement. The feline coronavirus (FCoV) biotype, termed feline infectious peritonitis virus (FIPV), is the etiologic agent of FIP. The objective of this study was to determine and compare the in vitro antiviral efficacies of the viral protease inhibitors GC376 and nirmatrelvir and the nucleoside analogs remdesivir (RDV), GS-441524, molnupiravir (MPV; EIDD-2801), and β-D-N4-hydroxycytidine (NHC; EIDD-1931). These antiviral agents were functionally evaluated using an optimized in vitro bioassay system. Antivirals were assessed as monotherapies against FIPV serotypes I and II and as combined anticoronaviral therapies (CACT) against FIPV serotype II, which provided evidence for synergy for selected combinations. We also determined the pharmacokinetic properties of MPV, GS-441524, and RDV after oral administration to cats in vivo as well as after intravenous administration of RDV. We established that orally administered MPV at 10 mg/kg, GS-441524 and RDV at 25 mg/kg, and intravenously administered RDV at 7 mg/kg achieves plasma levels greater than the established corresponding EC50 values, which are sustained over 24 h for GS-441514 and RDV.

Many biological high-throughput data sets, such as targeted amplicon-based and metagenomic sequencing data, are compositional in nature. A common exploratory data analysis task is to infer statistical associations between the high-dimensional microbial compositions and habitat- or host-related covariates. We propose a general robust statistical regression framework, RobRegCC (Robust Regression with Compositional Covariates), which extends the linear log-contrast model by a mean shift formulation for capturing outliers. RobRegCC includes sparsity-promoting convex and non-convex penalties for parsimonious model estimation, a data-driven robust initialization procedure, and a novel robust cross-validation model selection scheme. We show RobRegCC's ability to perform simultaneous sparse log-contrast regression and outlier detection over a wide range of simulation settings and provide theoretical non-asymptotic guarantees for the underlying estimators. To demonstrate the seamless applicability of the workflow on real data, we consider a gut microbiome data set from HIV patients and infer robust associations between a sparse set of microbial species and host immune response from soluble CD14 measurements. Astrophysics - High Energy Astrophysical Phenomena - Abstract
The Advanced LIGO and Advanced Virgo observatories recently discovered gravitational waves from a binary neutron star inspiral. A short gamma-ray burst (GRB) that followed the merger of this binary was also recorded by the Fermi Gamma-ray Burst Monitor (Fermi-GBM), and the Anticoincidence Shield for the Spectrometer for the International Gamma-Ray Astrophysics Laboratory (INTEGRAL), indicating particle acceleration by the source. The precise location of the event was determined by optical detections of emission following the merger. We searched for high-energy neutrinos from the merger in the GeV--EeV energy range using the ANTARES, IceCube, and Pierre Auger Observatories. No neutrinos directionally coincident with the source were detected within $\pm500$ s around the merger time. Additionally, no MeV neutrino burst signal was detected coincident with the merger. We further carried out an extended search in the direction of the source for high-energy neutrinos within the 14-day period following the merger, but found no evidence of emission. Objectives Interstitial lung disease (ILD) in CTDs has highly variable morphology. We aimed to identify imaging features and their impact on ILD progression, mortality, and immunosuppression response. Methods Patients with CTD-ILD had high-resolution chest CT (HRCT) reviewed by expert radiologists blinded to clinical data for overall imaging pattern [usual interstitial pneumonia (UIP); non-specific interstitial pneumonia (NSIP); organizing pneumonia (OP); fibrotic hypersensitivity pneumonitis (fHP); and other]. Transplant-free survival and change in percent-predicted forced vital capacity (FVC) were compared using Cox and linear mixed-effects models adjusted for age, sex, smoking, and baseline FVC. FVC decline after immunosuppression was compared with pre-treatment. Results Among 645 CTD-ILD patients, the most frequent CTDs were SSc (n = 215), RA (n = 127), and inflammatory myopathies (n = 100). NSIP was the most common pattern (54%), followed by UIP (20%), fHP (9%), and OP (5%). Compared with the case for patients with UIP, FVC decline was slower in patients with NSIP (by 1.1%/year, 95% CI 0.2, 1.9) or OP (by 3.5%/year, 95% CI 2.0, 4.9), and mortality was lower in patients with NSIP [hazard ratio (HR) 0.65, 95% CI 0.45, 0.93] or OP (HR 0.18, 95% CI 0.05, 0.57), but higher in fHP (HR 1.58, 95% CI 1.01, 2.40). The extent of fibrosis also predicted FVC decline and mortality. After immunosuppression, FVC decline was slower compared with pre-treatment in NSIP (by 2.1%/year, 95% CI 1.4, 2.8), with no change for UIP or fHP. Conclusion Multiple radiologic patterns are possible in CTD-ILD, including a fHP pattern. NSIP and OP were associated with better outcomes and response to immunosuppression, while fHP had worse survival compared with UIP. [ABSTRACT FROM AUTHOR]

Nine participants were interviewed about their experiences with unintentionally severe injury during engagement in nonsuicidal self-injury. Using interpretative phenomenological analysis, we identified four common themes among participants: (a) explosive affect prior to unintentional injury, (b) loss of control during unintentional injury, (c) unfamiliar method, and (d) consequences of unintentional injury. Implications for research and practice are discussed.

This article describes issues in measuring school contexts with an eye toward understanding students' experiences and outcomes. I begin with an overview of the conceptual underpinnings related to measuring contexts, briefly describe the initiatives at the National Center for Education Statistics to measure school contexts, and identify possible gaps in those initiatives that if filled could provide valuable new data for researchers. Next, I discuss new approaches and opportunities for measurement, and special considerations related to diverse populations and youth development. I conclude with recommendations for future priorities.

The accuracy of rate constants calculated using transition state theory depends crucially on the correct identification of a recrossing--free dividing surface. We show here that it is possible to define such optimal dividing surface in systems with non--Markovian friction. However, a more direct approach to rate calculation is based on invariant manifolds and avoids the use of a dividing surface altogether, Using that method we obtain an explicit expression for the rate of crossing an anharmonic potential barrier. This study is a discourse analysis of the transcribed talk recorded between tutors and students in a university peer tutoring program. We focus on STEM tutoring sessions, showing how tutors and students co-construct relational identities in and among the technical, highly task-oriented work of solving math and science problems. Using Grounded Practical Theory as a theoretical framework, we propose that understanding the co-creation of relational identities provides insight into an important component of STEM tutoring sessions. Through the process of Action Implicative Discourse Analysis, we describe three conversational practices with which tutors and students negotiate their relational identity--encouragement talk, sensemaking checks, and metadisciplinary talk. We discuss implications in terms of tutoring sessions' situated ideals and participants engaging in a community of practice.

ObjectivesMethodsResultsDiscussionSome anti-seizure medications (ASMs) are known to induce liver enzymes and impact lipid values that include total cholesterol (TC), low density lipoprotein cholesterol (LDL), high density lipoprotein cholesterol (HDL), and triglyceride (TG). In addition, use of ketogenic diet therapies, including the modified Atkins diet (MAD), has also influenced lipids. Here, we explored the combined impact of enzyme inducing ASMs (EIASMs) and MAD on lipid values in adults with epilepsy.Diet-naïve adults with epilepsy who began MAD were divided into three groups based on ASM use: EIASMs, non-EIASMs, and those on no ASMs. Demographic information, epilepsy-specific clinical history, anthropometrics and lipid values were obtained through retrospective chart review at baseline and after a minimum of 12 months of MAD use.Forty-two adults on MAD had baseline and follow up 12-month lipid outcomes. There was a significant increase in median levels of TC, LDL, non-HDL, and HDL after 12 months of MAD use. There was no change in median levels of TG. When separated according to ASM category, adults on non-EIASMs showed significant elevations in TC, HDL, and LDL after 12 months of MAD use. In contrast, adults on EIASMs only showed a significant increase in HDL after 12 months of MAD use.The increase in atherogenic cholesterol levels observed after 12 months of MAD use was most pronounced in adults with epilepsy on non-EIASMs and not observed in adults with epilepsy on EIASMs despite a higher proportion of abnormal cholesterol levels at baseline in those on EIASMs. [ABSTRACT FROM AUTHOR]

This research article employs the continuous wavelet transform analysis to identify the climatological effects among various water quality parameters to identify the successfulness of upland stream restoration on the receiving mesohaline tidal creeks. Estuaries and their corresponding tidal creeks have been impacted by human anthropogenic influences for decades, allowing a variety of restoration practices to be implemented in upland streams. In the face of climate variability and continuous human development pressures, this research performs statistical analysis and a wavelet coherence on, before, and after stream restoration for water quality changes in Chesapeake Bay's tidal tributaries in the Lower Western Shore to identify if the restoration strategies have been effective in the mesohaline tidal creeks. Statistical analysis showed that currently, the receiving tidal basins are not seeing the required positive improvements in water quality after years of upland stream restoration. Compounding this is the fact climate variability cannot be ignored. Results indicate that the North Atlantic Oscillation (NAO) has significant wavelet coherence with bottom dissolved oxygen, precipitation, and nutrients. This suggests that current restoration efforts may not be able to keep up with climate variability, and other techniques (restoration or policies) may need to be implemented. [ABSTRACT FROM AUTHOR]
