Background: Black gram (Vigna mungo) is one of the most important pulse crops grown throughout the country. It is hardy crop and fixes nitrogen in soil. Organic black gram fetch higher price in market i.e., Rs. 175 whereas other receive Rs. 120. Vermiwash, is one among the organic manures which has high potential to increase the crop growth and yield. Thus, present study aims to evaluate the influence of vermiwash prepared from different combination of organic sources on growth and yield of black gram. Methods: The field experiment was conducted during kharif, 2018 at Organic Farming Unit, Rajasthan College of Agriculture, Udaipur, with nine treatments [T1= vermiwash from 100% cow dung, T2= vermiwash from 100% buffalo dung, T3= vermiwash from 50% cow dung + 25% dry farm waste + 25% green farm waste, T4= vermiwash from 50% buffalo dung + 25% dry farm waste + 25% green farm waste, T5= vermiwash from 90% green waste incubated with 10% cow dung, T6= vermiwash from 90% green waste incubated with 10% buffalo dung, T7= vermiwash from 90% dry waste incubated with 10% cow dung, T8= vermiwash from 90% dry waste incubated with 10% buffalo dung and T9= water spray (control)] and were replicated thrice in Randomize Block Design. Result: The result of experiment revealed that the application of vermiwash from 100% cow dung significant influence towards growth parameters and yield attributes thus achieved maximum seed yield, haulm yield and B:C ratio (2.3). On the basis of field performance, it may be concluded that the treatment T1(top dressing in two equal split application at the rate of 10% at initiation of flowering and 15 days after 1st spray) may be recommended as a better organic package of blackgram. The formation of energetically favorable and metastable mineral phases within the Fe-H 2 O system controls the long-term mobility of iron complexes in natural aquifers and other environmentally and industrially relevant systems. The fundamental mechanism controlling the formation of these phases has remained enigmatic. We develop a general partial equilibrium model, leveraging recent synchrotron-based data on the time evolution of solid Fe(III) hydroxides along with aqueous complexes. We combine thermodynamic considerations and particle-morphology-dependent kinetic rate equations under full consideration of the aqueous phase in disequilibrium with one or more of the forming minerals. The new model predicts the rate of amorphous 2-line ferrihydrite precipitation, dissolution, and overall transformation to crystalline goethite. It is found that the precipitation of goethite (i) occurs from solution and (ii) is limited by the comparatively slow dissolution of the first forming amorphous phase 2-line ferrihydrite. A generalized transformation mechanism further illustrates that differences in the kinetics of Fe(III) precipitation are controlled by the coordination environment of the predominant Fe(III) hydrolysis product. The framework allows modeling of other iron(bearing) phases across a broad range of aqueous phase compositions.

In bacteria, peptidyl-tRNA hydrolase (Pth, E.C. 3.1.1.29) is a ubiquitous and essential enzyme for preventing the accumulation of peptidyl-tRNA and sequestration of tRNA. Pth is an esterase that cleaves the ester bond between peptide and tRNA. Here, we present the crystal structure of Pth from Enterococcus faecium (EfPth) at a resolution of 1.92 Å. The two molecules in the asymmetric unit differ in the orientation of sidechain of N66, a conserved residue of the catalytic site. Enzymatic hydrolysis of substrate α-N-BODIPY-lysyl-tRNA Lys (BLT) by EfPth was characterized by Michaelis-Menten parameters K M 163.5 nM and Vmax 1.9 nM/s. Compounds having pyrrolinone scaffold were tested for inhibition of Pth and one compound, 1040-C, was found to have IC 50 of 180 nM. Antimicrobial activity profiling was done for 1040-C. It exhibited equipotent activity against drug-susceptible and resistant S. aureus (MRSA and VRSA) and Enterococcus (VSE and VRE) with MICs 2-8 μg/mL. 1040-C synergized with gentamicin and the combination was effective against the gentamicin resistant S. aureus strain NRS-119. 1040-C was found to reduce biofilm mass of S. aureus to an extent similar to Vancomycin. Bacterial peptidyl tRNA hydrolase (Pth) or Pth1 emerges as a pivotal enzyme involved in the maintenance of cellular homeostasis by catalyzing the release of peptidyl moieties from peptidyl-tRNA molecules and the maintenance of a free pool of specific tRNAs. This enzyme is vital for bacterial cells and an emerging drug target for various bacterial infections. Understanding the enzymatic mechanisms and structural intricacies of bacterial Pth is pivotal in designing novel therapeutics to combat antibiotic resistance. This review provides a comprehensive analysis of the multifaceted roles of Pth in bacterial physiology, shedding light on its significance as a potential drug target. This article delves into the diverse functions of Pth, encompassing its involvement in ribosome rescue, the maintenance of a free tRNA pool in bacterial systems, the regulation of translation fidelity, and stress response pathways within bacterial systems. Moreover, it also explores the druggability of bacterial Pth, emphasizing its promise as a target for antibacterial agents and highlighting the challenges associated with developing specific inhibitors against this enzyme. Structural elucidation represents a cornerstone in unraveling the catalytic mechanisms and substrate recognition of Pth. This review encapsulates the current structural insights of Pth garnered through various biophysical techniques, such as X-ray crystallography and NMR spectroscopy, providing a detailed understanding of the enzyme's architecture and conformational dynamics. Additionally, biophysical aspects, including its interaction with ligands, inhibitors, and substrates, are discussed, elucidating the molecular basis of bacterial Pth's function and its potential use in drug design strategies. Through this review article, we aim to put together all the available information on bacterial Pth and emphasize its potential in advancing innovative therapeutic interventions and combating bacterial infections.

Excessive gingival display (EGD) is one of the most common aesthetic concerns, and its correction often presents a challenge to periodontists. It has a multifactorial etiology, and this article describes a case involving hypermobile upper lip (HUL), altered passive eruption (APE), and vertical maxillary excess (VME). Upon investigation, a positive collum angle and marked subnasal skeletal depression were observed. In this context, it is noted that during a spontaneous smile, the upper lip retracts and gets lodged in this depression. The rehabilitation plan includes included aesthetic crown lengthening via gingivectomy using Chu's proportional gauge for altered passive eruption and filling the subnasal depression by PMMA (polymethylmethacrylate) bone cement. The entire treatment plan was digitalised using cutting edge methods such as computed tomography (CT), cone beam computed tomography (CBCT), and 3D printers for virtual planning of the defect's position, size, and shape. No postoperative complications were reported. The search for ecofriendly products to reduce crop dependence on synthetic chemical fertilizers presents a new challenge. The present study aims to isolate and select efficient native PGPB that can reduce reliance on synthetic NPK fertilizers. A total of 41 bacteria were isolated from the sediment and roots of mangrove trees ( Avicennia marina ) and assessed for their PGP traits under in vitro conditions. Of them, only two compatible strains of Bacillus sp ecies were selected to be used individually and in a mix to promote tomato seedling growth. The efficiency of three inoculants applied to the soil was assessed in a pot experiment at varying rates of synthetic NPK fertilization (0, 50, and 100% NPK). The experiment was set up in a completely randomized design with three replications. Results showed that the different inoculants significantly increased almost all the studied parameters. However, their effectiveness is strongly linked to the applied rate of synthetic fertilization. Applying bacterial inoculant with only 50% NPK significantly increased the plant height (44-51%), digital biomass (60-86%), leaf area (77-87%), greenness average (29-36%), normalized difference vegetation index (29%), shoot dry weight (82-92%) and root dry weight (160-205%) compared to control plants. Concerning the photosynthetic activity, this treatment showed a positive impact on the concentrations of chlorophyll a (25-31%), chlorophyll b (34-39%), and carotenoid (45-49%). Interestingly, these increases ensured the highest values significantly similar to or higher than those of control plants given 100% NPK. Furthermore, the highest accumulation of N, P, K, Cu, Fe, Zn, and Ca in tomato shoots was recorded in plants inoculated with the bacterial mix at 50% NPK. We introduce an experimental setup to chemically image corrosion processes at metal-electrolyte interfaces under stagnant, confined conditions-relevant in a wide range of situations. The setup is based on a glass capillary, in which precipitation of corrosion products in the interfacial aqueous phase can be monitored over time with optical microscopy, and chemically and structurally characterized with microscopic synchrotron-based techniques (X-ray fluorescence, X-ray diffraction, and X-ray absorption spectroscopy). Moreover, quantification of precipitates through X-ray transmission measurements provides in-situ corrosion rates. We illustrate this setup for iron corrosion in a pH 8 electrolyte, revealing the critical role of O 2 and iron diffusion in governing the precipitation of ferrihydrite and its transformation to goethite. Corrosion and coupled reactive transport processes can thus be monitored and fundamentally investigated at the metal-electrolyte interface, with micrometer-scale resolution. How the multiple facets of soil fungal diversity vary worldwide remains virtually unknown, hindering the management of this essential species-rich group. By sequencing high-resolution DNA markers in over 4000 topsoil samples from natural and human-altered ecosystems across all continents, we illustrate the distributions and drivers of different levels of taxonomic and phylogenetic diversity of fungi and their ecological groups. We show the impact of precipitation and temperature interactions on local fungal species richness (alpha diversity) across different climates. Our findings reveal how temperature drives fungal compositional turnover (beta diversity) and phylogenetic diversity, linking them with regional species richness (gamma diversity). We integrate fungi into the principles of global biodiversity distribution and present detailed maps for biodiversity conservation and modeling of global ecological processes.

Fisheries and aquaculture output have exploded due to an alarming increase in consumption due to the global understanding of the nutritional advantages of fish. Inadvertently, the methods produce a massive amount of fish waste, posing a serious environmental threat. Recycling this waste has now become a major point of controversy that must be resolved. It is critical to emphasize the utility of discarded marine by-products for the creation of high-value commodities such as marine collagen (MC), which can be considered a sustainable solution. Because of its biocompatibility, biodegradability, safety, minimal immunogenicity, and low production costs, MC has various benefits over terrestrial collagen. Many academics have recently become interested in the use of MC as a scaffold. The transformation of 2-line ferrihydrite to goethite from supersaturated solutions at alkaline pH ≥ 13.0 was studied using a combination of benchtop and advanced synchrotron techniques such as X-ray diffraction, thermogravimetric analysis, and X-ray absorption spectroscopy. In comparison to the transformation rates at acidic to mildly alkaline environments, the half-life, t 1/2 , of 2-line ferrihydrite reduces from several months at pH = 2.0, and approximately 15 days at pH = 10.0, to just under 5 h at pH = 14.0. The calculated-first order rate constants of transformation, k , increase exponentially with respect to the pH and follow the progression log 10 k = log 10 k 0 + a ·pH 3 . Simultaneous monitoring of the aqueous Fe(III) concentration via inductively coupled plasma optical emission spectroscopy demonstrates that (i) goethite likely precipitates from solution and (ii) its formation is rate-limited by the comparatively slow redissolution of 2-line ferrihydrite. The analysis presented can be used to estimate the transformation rate of naturally occurring 2-line ferrihydrite in aqueous electrolytes characteristic to mine and radioactive waste tailings as well as the formation of corrosion products in cementitious pore solutions.

Salmonella is known to survive in raw/pasteurized milk and cause foodborne outbreaks. Lactoferrin, present in milk from all animal sources, is an iron-binding glycoprotein that limits the availability of iron to pathogenic bacteria. Despite the presence of lactoferrins, Salmonella can grow in milk obtained from different animal sources. However, the mechanism by which Salmonella overcomes iron scarcity induced by lactoferrin in milk is not evaluated yet. Salmonella employs the DNA binding transcriptional regulator Fur (ferric update regulator) to mediate iron uptake during survival in iron deplete conditions. To understand the importance of Fur in Salmonella milk growth, we profiled the growth of Salmonella Typhimurium Δfur (ST4/74Δfur) in both bovine and camel milk. ST4/74Δfur was highly inhibited in milk compared to wild-type ST4/74, confirming the importance of Fur mediated regulation of iron metabolism in Salmonella milk growth. We further studied the biology of ST4/74Δfur to understand the importance of iron metabolism in Salmonella milk survival. Using increasing concentrations of FeCl 3 , and the antibiotic streptonigrin we show that iron accumulates in the cytoplasm of ST4/74Δfur. We hypothesized that the accumulated iron could activate oxidative stress via Fenton's reaction leading to growth inhibition. However, the inhibition of ST4/74Δfur in milk was not due to Fenton's reaction, but due to the 'iron scarce' conditions of milk and microaerophilic incubation conditions which made the presence of the fur gene indispensable for Salmonella milk growth. Subsequently, survival studies of 14 other transcriptional mutants of ST4/74 in milk confirmed that RpoE-mediated response to extracytoplasmic stress is also important for the survival of Salmonella in milk. Though we have data only for fur and rpoE, many other Salmonella transcriptional factors could play important roles in the growth of Salmonella in milk, a theme for future research on Salmonella milk biology.
