Pathogenesis-related (PR) genes are useful molecular markers, and gene expressions are activated in the reaction of plant-parasitic nematodes. Determining the PR gene expression and the resistance reaction of Phaseolus vulgaris L. genotypes against root-knot nematode are valuable for understanding plant–nematode interactions. However, resistance reactions of P. vulgaris genotypes and the gene expression of host–nematode interactions are not fully understood. Therefore, this study aimed to determine the expression of PR genes during the infection of Meloidogyne incognita on resistant and susceptible bean varieties at 1, 3, 5, and 7 days postinfection (dpi). The results revealed that one genotype gave a reaction as “immune”, and numerous genotypes were susceptible. The expression of PvPR1 and PvPR2 genes reached the highest peak at 1 dpi in nematode-infected immune plants, and the expression of PvPR1 reached 5 dpi in susceptible plants. In conclusion, PR genes involved in nematode resistance and parasitism and the immune common bean genotype have great potential in breeding programs and understanding the exact location of resistance on the gene map would be advantageous as future work.

Son yıllarda, Avrupa Gül Böceği, Cetonia aurata (Linné, 1761) (Coleoptera: Scarabaeidae)’nın meyve yetiştiriciliği yapılan alanlarda ekonomik düzeyde zarara neden olduğu sık sık rapor edilmiştir. Ülkemizde bu zararlıya karşı ruhsatlı bir insektisitin bulunmaması, üreticileri zor durumda bırakmaktadır. Bu çalışmada, elma yetiştiriciliği yapılan alanlardan toplanan 2.dönem C. aurata larvalarına karşı Heterorhabditis bacteriophora‘nın biyokontrol potansiyeli farklı dozlarda (50, 100, 200 IJ/larva) laboratuvar şartlarında araştırılmıştır. Uygulamadan sonraki 10. Gündeki ölüm oranları %25 ile 65 arasında değişmiştir. En yüksek ölüm oranı (%82,5) 200 IJ/larva dozunda uygulamadan sonraki 15. günde elde edilmiştir. Sonuçlar, H. bacteriophora‘nın C. aurata larvalarına karşı yüksek bir biyokontrol potansiteline sahip olduğunu göstermektedir.

Meloidogyne chitwoodi Golden, O’Bannon, Santo & Finley, 1980 and M. hapla Chitwood, 1949 are important pests of potato in Turkey. This study was conducted in three experimental areas where conventional agronomic practices have been done in Nigde (Center district) and Nevsehir (Center and Derinkuyu districts) provinces. Nematodes were extracted by modified Oostenbrink dish from soil samples taken from 0 to 30 cm depths every month between 2014 and 2017 to assess the population fluctuations of these root-knot nematodes under field conditions. The whole suspension was checked for Meloidogyne juveniles which were counted under an inverted microscope. Morphological and molecular identification results showed that two experimental areas were infested by M. chitwoodi while one experimental area (Derinkuyu) was infested with both M. chitwoodi and M. hapla. Our study showed that M. chitwoodi population level decreased during wheat cultivation and were maintained under rye cultivation, whereas M. chitwoodi and M. hapla population levels increased rapidly under potato and bean cultivation. It was found that M. chitwoodi population increased more when potatoes were planted, while M. hapla increased more in bean. The best sampling time for M. chitwoodi and M. hapla was observed to be right after harvest, when the population was highest, but both could be detected in soil samples even when the ambient temperature was below zero. These results demonstrated that farmers should be careful in their choice of rotational crop with potatoes in areas infested with M. chitwoodi and M. hapla.

Plant-parasitic nematodes (PPNs) are one of the major biotic factors that cause significant yield losses in wheat-growing areas worldwide. The major PPN groups causing significant economic losses in wheat quantity and quality are cereal cyst nematodes (CCNs) and root-lesion nematodes (RLNs). Based on their wide distribution, pathogenicity, high occurrence in wheat cultivated areas, they are considered major threats to the global food supply. The economic loss caused by these destructive pathogens ranges from 10 to 100% depending on different agro-ecological conditions such as drought, heat stress, and cold stress. Multidisciplinary management practices are being implemented to manage cereal nematodes (CNs) that range from cultural to molecular strategies. Integration of wheat resistant varieties with appropriate agronomic practices is recognized as the safest and most practical, effective, and applicable management strategy. Nine resistance genes (Cre1–Cre9) to CCN are well- documented in the literature. CreR, CreV and CreZ genes are relatively recently characterized from wheat and confer resistance to CCNs. On the other hand, Rlnn1 is the only resistance gene characterized from wheat that is known to confer resistance towards RLNs. However, breeding for resistance to PPN has numerous challenges that originate from the narrow genetic diversity and difficulty in the process of transferring resistance gene(s) from the source to the target variety. A unique opportunity for wheat genetic improvement was provided due to the availability of genomic resources and the wheat worldwide germplasm collection which includes wild wheat germplasm. Moreover, the presence of several genome-wide association studies and genome editing technologies could also help for further improvement to enhance CNs resistance in wheat. This article provides the latest information regarding the progress made in the identification and characterization of resistance genes from different sources and its utility against both CCNs and RLNs, which will attract the attention of the scientific community and other relevant stakeholders.

Root lesion nematodes (RLNs), Pratylenchus thornei Sher & Allen, 1953 and Pratylenchus neglectus (Rensch, 1924) (Tylenchida: Pratylenchidae) are important plant-parasitic nematodes that cause economic yield losses in wheat cropping systems worldwide. The use of resistant and tolerant cultivars is the most effective method to control these nematodes in wheat. There are currently no commercial wheat cultivars identified as completely resistant to the RLN species. The aim of this research was to evaluate 19 Turkish spring wheat cultivars for reaction to P. thornei and P. neglectus under in-vitro conditions over 16 weeks in 2019. In the result of the study, nine wheat cultivars (Adana99, Ata89, Bürküt, Cumhuriyet75, Gönen98, Marmara86, Meta2002, Troya and Uludağ) were found to have moderate resistance against P. thornei, whereas five wheat cultivars (Adana99, Alibey, Ata89, Ceyhan99 and Uludağ) were moderately resistant to P. neglectus. The study also showed that Adana99, Ata89 and Uludağ are resistant to both nematode species, and these cultivars, thus, are considered to be excellent sources of genes for further development RLN resistant commercial wheat cultivars., Kök yara nematodları (RLN), Pratylenchus thornei Sher & Allen, 1953 ve Pratylenchus neglectus (Rensch, 1924) (Tylenchida: Pratylenchidae) dünya genelinde buğday yetiştiriciliğinde ekonomik ürün kayıplarına neden olan önemli bitki paraziti nematodlarıdır. Dayanıklı ve tolerant çeşit kullanımı bu nematodlar ile mücadelede en etkin metot olarak bilinmektedir. Günümüzde, RLN türlerine karşı tamamen dayanıklı ticari herhangi bir çeşit yoktur. Bu çalışmada 19 adet yazlık buğday çeşidinin kontrollü koşullarda 16 hafta süreyle P. thornei ve P. neglectus’a karşı reaksiyonları 2019 yılında değerlendirilmiştir. Çalışma sonucunda Adana99, Ata89, Bürküt, Cumhuriyet75, Gönen98, Marmara86, Meta2002, Troya ve Uludağ’ın içinde olduğu dokuz buğday çeşidi P. thornei’ye karşı orta dayanıklı, buna karşın Adana99, Alibey, Ata89, Ceyhan99 ve Uludağ’ın içinde olduğu beş buğday çeşidi P. neglectus’a karşı orta dayanıklı bulunmuştur. Ayrıca çalışmada Adana99, Ata89 ve Uludağ'ın her iki nematod türüne dayanıklılık gösterdiği ve bu çeşitlerin RLN'e karşı dayanıklı ticari buğday çeşitleri geliştirmek için oldukça iyi dayanıklılık gen kaynakları olduğu düşünülmektedir.

Summary Pratylenchus thornei and P. neglectus attack common bean and cause economic yield losses throughout cultivated areas in Turkey. The most effective management strategy for the P. thornei and P. neglectus infections is crop rotation with non-host crops and breeding resistant/tolerant varieties. However, parent bean genotypes immune to P. thornei and P. neglectus are not available for breeding programmes; thus, resistant varieties are commonly incorporated as parents. In the present research, a total of 36 common bean varieties were tested in a growth room for their host response to these two nematode pests. The reproduction factor and the population density of both nematode species were calculated. All tested bean varieties showed varying levels of resistance and susceptibility to P. thornei and P. neglectus. Among the dry bean varieties, nine were found to be resistant to P. thornei and three to P. neglectus, with three varieties (‘Kantar-05’, ‘Önceler-98’ and ‘Karacasehir-90’) resistant to both species. Among fresh bean varieties, ten were found to be resistant to P. thornei and four to P. neglectus, with four varieties (‘Helda’, ‘Gina’, ‘Gelincik’ and ‘Bourgondia’) resistant to both species. The resistant common bean varieties identified in this study are a valuable untapped genetic pool that will offer improved resistance levels to P. thornei and P. neglectus, especially ‘Gina’ and ‘Önceler-98’, which seem to possess a great source of resistance to P. thornei and P. neglectus, respectively, and can be used in breeding programmes in the near future.

Several pathogens and pests damage tomato plants, and only one and/or more pathogens and pests can coexist in the same plant at the same time. As several numerous pathogens are found in the same plant, the damage to the tomato plants is higher. Pathogens such as nematodes, viruses, viroids, bacteria, and insects adversely affect the growth and development of tomato plants. They may infect roots or upper part of the plant and can cause not only slow down the growth of plants, but also crop losses and their death. Damaging of plant caused by pathogens and pests reduces the market value of plant products. Those pathogens and pests are also called biotic stress agents. The damage, mode of infection, and the mechanism of infection in each tomato plant and pathogens might be different. This situation is crucially important to understand plant pathogen relationship in detail in terms of controlling pests and pathogen. The effect of each pest/pathogen on tomato plants during the cultivation, the type of damage, and new developments and perspectives on morphological and molecular aspects in tomato-pathogen interactions will be discussed in this chapter.

Yield losses because of cereal cyst nematodes could be as high as 92%, causing a bottleneck for wheat production. An integrated approach (application of pesticides, crop rotation, and use of host resistance) is needed to manage this devastating pathogen where resistant cultivars are considered most effective. This necessitates the identification of nematode-resistant sources in the available germplasm. Here, we report on the genetic mapping of nematode resistance in 255 diverse prebreeding lines (PBLs) employing an association mapping strategy. Altogether, seven additive quantitative trait loci (QTL) were identified on chromosomes 1A, 2A, 2B, 2D, 3A, 6B, and 6D explaining a maximum of 9.42% phenotypic variation where at least five QTL (on chromosomes 2A, 2B, 2D, 6B, and 6D) are located on the same chromosomes that harbor the already known nematode resistance genes. Resistant PBLs carried Aegilops squarrosa (436) in their pedigree which could be the possible source of positive alleles. To add to it, better yield performance of the identified nematode-resistant lines under stress conditions indicates that the germplasm can provide both nematode resistance and high-yielding cultivars.

The golden nematode, Globodera rostochiensis (Wollenweber, 1923) Skarbilovich, 1959 (Tylenchida: Heteroderidae) is one of the most economically important pests of potato in the world. Tests for this nematode are routinely performed for outbreaks and densities in potato growing areas. The morphological and molecular analyses for precise determination of the nematode are employed to establish appropriate management strategies. This study showed that 96% of soil samples obtained from Bozdağ and Ödemiş Districts of İzmir Province, during 2017 and 2018 potato growing seasons, were positive for G. rostochiensis. The mean number of cysts ranged from 0.01 to 3.70 cysts g-1 soil in the fields examined. The examination of the morphological and morphometric features of the second-stage juveniles and cysts of the G. rostochiensis revealed slight differences among the populations obtained from Bozdağ and Ödemis. To assess the accuracy of the identification, partial sequences of ribosomal DNA for all populations were amplified, sequenced, and deposited in GenBank. The comparisons of the sequences with those of corresponding G. rostochiensis populations available in GenBank showed 99.89-100% nucleotide similarity. The results of this study will help to better understand the physiology, ecology and biology of the nematode to quarantine this pest more effectively., Altın nematod, Globodera rostochiensis (Wollenweber, 1923) Skarbilovich, 1959 (Tylenchida: Heteroderidae) dünyada ekonomik açıdan en önemli patates zararlılarından birisidir. Genellikle patates yetiştirilen alanlarındaki dağılımları ve yoğunluklarını belirlemek amacıyla bu nematod için rutin olarak testler gerçekleştirilmektedir. Morfolojik ve moleküler analizler bu nematodun kesin tanısını yapmak ve mücadele stratejilerini oluşturmak amacıyla kullanılmaktadır. Bu çalışma, İzmir İli’nin Bozdağ ve Ödemiş ilçelerinden 2017 ve 2018 yılları patates yetiştiriciliği sezonlarında alınan toprak örneklerinin %96'sının G. rostochiensis ile enfekte olduğunu göstermiştir. Bulaşık alanlardaki ortalama kist sayısı, 0.01 ila 3.70 kist g-1 toprak arasında değişmiştir. Globodera rostochiensis'in ikinci dönem larva ve kistlerinin morfolojik ve morfometrik karakterlerinin incelenmesi, Bozdağ ve Ödemiş’ten elde edilen popülasyonlarda arasındaki hafif farklılıkları ortaya koymuştur. Tanımlamanın doğruluğunu değerlendirmek için, tüm popülasyonlar için kısmi ribozomal DNA sekansları amplifiye edilmiş, sekanslanmış ve GenBank veritabanına kaydedilmiştir. Morfolojik ölçümler ve filogenetik analizler sonucunda Bozdağ ve Ödemiş'ten elde edilen popülasyonlar arasında küçük farklılıkların olduğu belirlenmiştir. Sekansların GenBank'ta mevcut karşılık gelen G. rostochiensis popülasyonlarıyla karşılaştırılması sonucunda nükleotid benzerliği %99.89-100 oranında görülmüştür. Bu çalışmanın sonuçları, bu zararlıyı daha etkili bir şekilde karantinaya almak için nematodun fizyolojisini, ekolojisini ve biyolojisini daha iyi anlamaya yardımcı olacaktır.

Summary The cereal cyst nematode, Heterodera latipons, is an important plant parasite causing substantial yield losses in wheat throughout the world. This study aimed to determine genetic and pathogenic variation in H. latipons populations obtained from the southern part of Turkey. The populations were identified as H. latipons by sequencing the ITS-rDNA region and further sequence analysis showed an intraspecific genetic variation in H. latipons populations, which were clustered into different groups. The International Test Assortment materials were used to determine pathogenic variation (pathotypes) in these populations. The results showed that ‘Ortolan’, ‘Morocco’, ‘KVL191’, ‘Bajo Aragon 1-1’, ‘Herta’, ‘Martin 403-2’, ‘Sun II’ and ‘Pusa Hybrid Bsi’ cultivars were resistant or moderately resistant to the tested nematode populations. ‘Emir’, ‘Dalmatische’ and ‘Capa’ were susceptible to H. latipons populations. The Hatay population of H. latipons was detected as the most virulent nematode population because ten out of 20 cultivars were susceptible or moderately susceptible to this population. The least virulent population was the Kilis population, which caused susceptible reaction on six out of all cultivars with different levels. Based on this scheme, the Turkish populations were in the Ha1 group: the reactions of barley, oats and wheat classified them as either Ha41 or Ha51. Barley ‘KVL191’ was resistant to all nematode populations but susceptible to Ha51, and the reactions of the other barley cultivars were also consistent with the Turkish populations being Ha51. ‘AUS10894’ was susceptible to three nematode populations but resistant to Ha41, and the reaction of ‘Capa’ was also consistent with the Turkish populations being Ha51. However, the degree of susceptibility of all wheat differentials distinguishes the Turkish populations from other pathotypes in the Ha1 group.

The fall webworm (FWW), Hyphantria cunea Drury (Lepidoptera: Erebidae), is an invasive and polyphagous insect pest of many economically important crops such as hazelnuts, apple, and mulberry. Recently, there have been an increasing number of reports about the damaging activities of FWW from hazelnut growing areas of Turkey indicating that currently existing control methods fail to satisfy the expectations of growers. Entomo- pathogenic nematodes (EPNs) in the Steinernematidae and Heterorhabditidae (Nematoda: Rhabditida) families and the symbiotic bacteria they carry in their intestine have a great potential for the management of many agriculturally important pests. In this study, the symbiotic bacteria of local EPN species (Heterorhabditis bac- teriophora AVB-15, Steinernema feltiae KCS-4S, and Steinernema bicornotum MGZ-4S) recovered from the central Anatolia region was characterized using recA gene region as Photorhabdus luminescens, Xenorhabdus bovienii and Xenorhabdus budapestensis. The contact (25, 50, 100, 200 IJs/Petri) and oral efficacies of the infective juveniles (IJs) (25, 50, 100, 200 IJs/leaf) of these EPN isolates determined on 3rd/4th instar larvae, and cell-free su- pernatants from the identified symbiotic bacteria were evaluated separately on the 3rd and 4th larval instars of FWW in Petri dish environment under laboratory conditions (25 ± 1 ◦C, 60% of RH). In the Petri dish bioassays of EPN species, the most pathogenic isolate at the 1st DAT and 4th DAT was S. feltiae which caused 50% mortality at the highest concentration (200 IJs/Petri) and the highest mortality rate (97.5%) were achieved at 4th DAT by H. bacteriophora AVB-15 isolate. Surprisingly, the mortality rates were generally higher at the lowest concen- trations and 82.5% mortality were reached 4th DAT by S. bicornotum at the lowest concentration (25 IJs/leaf) in the leaf bioassays. Mortality rates were higher in both Petri dish and filter paper efficacies of cell-free super- natants at the 2nd DAT and the highest mortality (87.5%) was reached in the contact efficacy studies when applied X. bovienii KCS-4S strain. The results suggest that the tested EPN species and CFSs have good potential for biological control of the larvae of FWW and can contribute to the IPM programs of FWW. However, the efficacy of both IJs of EPNs and CFSs of their symbiotic bacteria on larvae of FWW requires further studies to verify their efficiency in the field. WOS:000867343700007

AimsCitrus plants are hosts of numerous plant-parasitic nematodes (PPNs), which are microscopic organisms and hidden enemies for plants. Among the PPNs, the citrus root nematode, Tylenchulus semipenetrans, causes significant damage to citrus plantations in the world. Understanding the citrus nematode population, precise nematode identification, host preference among the citrus species, and damage threshold level are crucially important to control T. semipenetrans. Although some studies have been conducted, minutiae about citrus plant-nematode interactions, nematodes density, and molecular nematode identification have not been well understood. MethodsThis study aimed to reveal nematode identification using molecular and morphological methods, host-nematode interactions, host (citrus species) preference, damage of economic threshold (ET), and economic-injury level (EIL). Therefore, citrus plantations of different provinces located in the Mediterranean region of Turkey were investigated.ResultsITS sequences revealed that samples were infested with the citrus root nematode Tylenchulus semipenetrans. The lowest nematode density was obtained from Mersin (80-second-stage juveniles(J2s)/150g-soil), while the highest density was obtained from Adana (18260-J2s/150g-soil). Out of the 73 samples collected from the Mediterranean region, 21.9% have passed the EIL, while 8.2% are on the ET. Citrus nematode reproduced utmost level on roots of Citrus reticulata, followed by C.sinensis, C.limon, and C.parasidisi, respectively. ConclusionThis study revealed that the Citrus nematode is more common than it was thought and that the population fluctuations change according to the citrus plant species. It is of great importance to investigate the possible genetic changes in nematode populations caused by global warming and its negative impact on citrus yield.

The aim of this study was to identified cyst nematodes in the cabbage production areas in Niğde Province by molecular methods to underpin decision making for field control. The sequences of ribosomal DNA region (rDNA-ITS) and cytochrome oxidase subunit 1 (mtDNA-COI) were used for the first time for identification Heterodera cruciferae (cabbage cyst nematode) in Turkey.Heterodera cruciferae populations extracted from cabbage growing areas of Niğde Province and investigated using sequences of rDNA-ITS and mtDNA-COI. Similarities and differences between 13 geographic populations of H. cruciferae were detected in surveys during 2020-2021. DNA from single cysts was successfully amplified, and genetic variability was revealed within nematode populations. Based on these results, H. cruciferae is reported for the first time from Niğde Province, Turkey. This study showed clear discrimination among the sampled populations of H. cruciferae.This finding is important for control and managing cabbage cyst nematode in cabbage fields in Turkey as more than one Heterodera spp. can occur. Future studies should investigate the population dynamics and control of H. cruciferae in fields in the sampled districts.

Entomopathogenic nematodes (EPNs) in the families of Steinernematidae and Heterorhabditidae are known as an important biocontrol agent to many insects including BCW. In 2020 and 2021, surveys of EPNs were conducted in major potato-growing areas of seven provinces of Turkey, including Afyonkarahisar, Konya, İzmir, Sivas, Bolu, Kayseri, and Niğde, in the sake to find a new sustainable biological control option for BCW. Out of the 400 collected soil samples, 48 samples (12%) consisted of EPNs. Based on the morphological, morphometric, and sequence analysis of the internal transcribed spacer (ITS), 34 isolates were identified as Steinernema feltiae Filipjev, 1934 (Rhabditida: Steinernematidae), 13 isolates as Heterorhabditis bacteriophora Poinar, 1976 (Rhabditida: Heterorhabditidae), 1 one isolate as Oscheius tipulae Lam and Webster, 1971 (Rhabditida: Rhabditidae). The virulence of EPN isolates was assessed on the larvae of Galleria mellonella L. (Lepidoptera: Pyralidae), and the most pathogenic isolates were further tested for their biocontrol potential against the 3rd/4th larval instar of BCW larvae at two concentrations of 50 and 100 IJs/cm2 under laboratory conditions. All selected isolates were highly effective against the larvae of BCW, and larval mortality ranged between 70 and 100% for both concentrations after 3 days of exposure time. The maximum mortality (100%) was achieved only by two isolates (H. bacteriophora AF-12 and S. feltiae KAY-4) at the higher concentration of 100 IJs/cm2. The results indicate that EPNs are abundantly present in potato-growing areas of Turkey and have the potential to be used in the biocontrol of BCW. To our knowledge, this is the first report of the presence of Oscheius tipulae in Turkey.

WOS:000529950700012 The Mediterranean cereal cyst nematode, Heterodera latipons Franklin, 1969 (Tylenchida: Heteroderidae) is a destructive plant-parasitic nematode on cereal crops in particularly wheat and barley. It has a global distribution with a severe negative impact on yield quantity. In this study, a survey was conducted to identify plant-parasitic nematodes in cereal-growing areas in Cyprus. Forty-five samples including roots and soil from the root zone of plants were collected from cereal fields located in Gazimagusa, Gime, Guzelyurt and Lefkosa Provinces before crop harvesting from late-May and early-June in 2017. Cyst-forming nematodes were determined by Fenwick's flotation and decanting techniques from 37 soil samples (82%). The internal transcribed spacer (ITS) regions of the ribosomal DNA of isolates were amplified and sequenced and subjected to a BLASTn search of the NCBI database for species identification, and the analyses showed that all samples were identified as H. latipons. Phylogenetic analyses based on ITS sequences revealed that H. latipons isolates from Northern Cyprus were closely related to isolates obtained from Morocco, Russia, Syria and Turkey. Data of this study demonstrated for the first time the presence of H. latipons in the cereal fields of Gazimagusa, Gime, Guzelyurt and Lefkosa Provinces, where the nematode most likely causes serious economic problems in the cereal production. These results were the most up-to-dated analyses on the occurrence of H. latipons in cereal fields of Northern Cyprus and provided basic data for breeding programs to improve the resistant levels in the local cultivars. Bu çalışmada, Kıbrıs’ın tahıl alanlarındaki patojen nematodları belirlemek için bir sürvey yapılmıştır. Gazimağusa, Girne, Güzelyurt ve Lefkoşa tahıl üretim alanlarından toplam 45 adet toprak ve kök örneği 2017 yılının mayıs ayı sonu-haziran ayı başı arasında tahıl hasadı öncesinde alınmıştır. Toprak ve kök örneklerinden Fenwick yöntemii kullanılarak kistler toplanılmış, 37 adet örnekte (%82) kiste rastlanılmış olup, tüm örnekler ribozomal DNA’nın ITS bölgesi kullanılarak, tahıl kist nematodu H. latipons moleküler düzeyde tanımlanarak belirlenmiştir. Çalışmada Kuzey Kıbrıs arpa ve buğday alanlarından elde edilen H. latipons örneklerine ait ITS bölgesi sekans dizilerinin Fas, Rusya, Suriye ve Türkiye’den elde edilen izolatlara oldukça benzerlik gösterdiği tespit edilmiştir. Kuzey Kıbrıs'ın Gazimağusa, Girne, Güzelyurt ve Lefkoşa illeri tahıl alanlarında H. latipons'un ilk olarak tespit edildiği bu çalışma, nematodun yaygınlığını ortaya koyan ve zararlının mücadelesinde başta dayanıklı çeşit kullanımı olmak üzere diğer mücadele yöntemlerinin uygulanmasında temel oluşturacak verileri içermektedir.

Plant-parasitic nematodes (PPNs) are considered serious damaging on the global cereals production systems. The current study was conducted to evaluate the incidence of PPNs in the main cereal-growing areas in northern Kazakhstan. PPNs were detected in about 90% of 78 soil samples and thirteen genera were identified, including Pratylenchus, Heterodera, Geocenamus, Ditylenchus, Helicotylenchus, Rotylenchus, Pratylenchoides, and Tylenchorhynchus. Out of the 78 samples, 32 samples were found infested by Heterodera filipjevi based on the morphological and molecular analysis. To our knowledge, this is the first report on this cereal cyst nematode species in northern Kazakhstan. During the morphological and molecular assays, intraspecific polymorphism was observed within H. filipjevi populations and the populations divided into at least two groups. The highest frequency of infestation of H. filipjevi (76%) was recorded from Kokshetau Province when compared to other provinces: Astana (50%), Petropavl (37%), and Kostanay (16%). The highest number of cysts (30.4) was found among Astana samples while the lowest number of cysts (18.2) was recorded from Kostanay samples. Cyst nematodes can maintain their population above the economic threshold as stimulated by the cereal monoculture system (mainly wheat) which is similar to the cereal production systems of northern Kazakhstan.

The Mi gene is known to involve in the resistance response to some Root knot nematodes, Meloidogyne spp. in tomato plants Lycopersicon esculentum. Root knot nematodes cause significant damages to almost all crops in the world. Among Meloidogyne species, Meloidogyne incognita is one of the most damages species causing a vast amount of crop loss including tomato plants. The resistance gene may be found in some tomato genotypes or cultivars. Determining the resistance gene, Mi, in tomato cultivars is very important to control root knot nematodes. For this purpose, this study was conducted in 99 tomato genotypes that were screened for resistance against Meloidogyne incognita in molecular and screening assays. Results revealed that Mi gene was only determined in one genotype (Tom113). The result of the screening assay indicated that one of the genotypes (Tom113) showed an immune reaction against the nematode; two genotypes (Tom146, Tom141) were found intermediate; 17 genotypes moderately susceptible, 43 genotypes susceptible and 36 genotypes found highly susceptible. Results of the screening assay confirmed the presence of Mi gene in a tomato genotype, which accompanied with a decreased ability for the nematode in causing the infection. Immune and some promising genotypes are thought as important genetic tools for breeding studies for future works.

The insecticidal effect of native entomopathogenic nematode (EPN) isolates (Steinernema feltiae UTP-5 isolate, Heterorhabditis bacteriophora UMK-7 isolate, S. feltiae DDKY-11 isolate and H. bacteriophora AVB-15 isolate) recovered from the Cappadocia Region of Turkey were investigated on the adults of S. granarius and R. dominica and the larvae of E. kuehniella at different temperatures (15 degrees C, 20 degrees C and 25 degrees C) and concentrations (250, 500 and 1000 IJs/adult for S. granarius and R. dominica; 10, 50 and 100 IJs/larvae for E. kuehniella) under controlled conditions. Insect mortality was recorded after 4th, 6th and 8th day of exposure time for S. granarius and R. dominica, and 2nd and 4th day for E. kuehniella. The results showed that the efficacy of the isolates at 25 degrees C was generally higher than the ones at 15 degrees C and 20 degrees C. Steinernema feltiae DDKB-17 isolate caused the highest mortality (86%) against S. granarius adults at all temperatures tested. The virulence of the tested isolates on R. dominica adults did not exceed 44% at all temperatures and concentrations tested. UMK-7 and AVB-15 isolates were the most virulent isolates against E. kuehniella larvae with a mortality rate of 100%. Based on the results obtained from the laboratory tests, it appeared that the isolates tested have a good potential for the management of S. granarius, R. dominica and E. kuehniella. TheMediterranaen cereal cyst nematode, Heteroderalatipons, causing serious yield losses in protected cultivation cerealcropping areas in Turkey. An accurate and reliable identification is primarilyneeded to establish effective, sustainable and environmentally safe controlmeasures to any cyst nematode species. In this study, twenty-five populations ofHeterodera latipons collectedfrom cereal areas in East-Mediterranean and South-East Anatolian regions wereidentified using species-specific primers (SCAR). TheSCAR primer pair, H-latac F and H-latact R, defined species-specific primerswere used to identify Heteroderapopulations. In this study stated that H-latac F and H-latact R primers foridentifying of H. latipons can beefficient tools to identify the Turkish Mediterranean cereal cyst nematodepopulations. Moreover, the results indicated that H. latipons was the prominent cereal cyst nematode species in theEast Mediterranean and South East Anatolian regions of Turkey. Ayrıca, çalışmada kullanılan türeözgü primerler Heterodera latipons popülasyonlarının kolay ve hızlıtanımlanmasında kullanılabileceği sonucuna varılmıştır.

Potato (Solanum tuberosum L.) cyst nematodes (Globodera rostochiensis (Wollenweber) and Globodera pallida (Stone) cause significant yield losses and have been listed as quarantine pests for many countries in the world including Turkey, and the European and Mediterranean Plant Protection Organization (EPPO, A2). In Turkey, the current potato production area is decreasing due to contamination by quarantine agents, and new fields are under threat of invasion by new quarantine organisms. In this study, samples were collected from Nevşehir province, Turkey, in potato growing fields during the spring growing season in 2014-2015. Soil samples were washed using Fenwick’s can and cysts were extracted and identified using molecular markers. The species-specific molecular markers generated a 435 bp using PITSr3, PITSp4 and ITS5 primers. The potato golden cyst nematode, G. rostochiensis was identified in Nevşehir potato production areas and this is the first report for the Central Anatolia Region. In addition, G. rostochiensis was found in two samples of the same field in the surveys. The average cyst nematode density was determined as 60 cysts in 250 g soil. Patates (Solanum tuberosum L.) kist nematodları (Globodera rostochiensis (Wollenweber) ve Globodera pallida (Stone) önemli verim kayıplarına neden olmakta, Türkiye de dahil olmak üzere dünyadaki birçok ülke, Avrupa ve Akdeniz Bitki Koruma Örgütü’nde (EPPO, A2) karantina zararlıları olarak listelenmiştir. Türkiye'de mevcut patates üretim alanı karantina zararlısı olarak bulaşmaya bağlı olarak azalmakta ve yeni alanlar yeni karantina organizmaları tarafından istila tehdidi altında bulunmaktadır. Bu çalışmada, Türkiye, Nevşehir ilinden 2014-2015 yıllarında bahar mevsiminde patates yetiştirme alanlarında örnekler toplanmıştır. Toprak örnekleri Fenwick aletinde yıkanarak bulaşık örneklerdeki kistler elde edilmiş ve kist popülasyonlarının moleküler olarak teşhisleri yapılmıştır. Kist popülasyonlarının moleküler düzeyde türe özgü PITSr3, PITSp4 ve ITS5 primerleri kullanılarak yapılan teşhiste söz konusu primerlerin 435 bp de bant oluşturduğu tespit edilmiştir. Bu çalışma sonucunda Nevşehir ili patates ekiliş alanlarında patates kist nematodu G. rostochiensis saptanmış olup, bu sonuç İç Anadolu Bölgesi için ilk kayıt olma özelliğini taşımaktadır. Ayrıca, sürveylerde aynı tarlaya ait iki örnekte G. rostochiensis’e rastlanılmış, 250 g toprakta ortalama 60 kist yoğunluğunda olduğu tespit edilmiştir.

Fusarium crown rot of wheat is an economically important disease that leads to significant yield and quality losses, especially in many arid and semi-arid wheat-growing areas worldwide. In June 2020, winter wheat (Triticum aestivum L.) plants exhibiting crown rot symptoms were identified in a commercial field located in the Tokbay location (43.033719°N, 74.325623°E), Chuy Province, Kyrgyzstan. The diseased plants were stunted and had brown discoloration on internodes of the stem bases and roots. Disease incidence was about 3%. A total of 10 plants were sampled at the ripening stage from the field to identify the causal agent. Symptomatic tissues were excised, surface disinfected with 1% NaOCl, rinsed three times with distilled water, and placed on one-fifth strength potato dextrose agar (PDA) followed by incubation at 23°C in the dark for 5 days. A total of 8 Fusarium isolates were recovered from tissues and purified by the hyphal tips method onto fresh PDA and Spezieller-Nährstoffarmer agar (SNA) plates (Leslie and Summerell 2006). Sequence analysis of the translation elongation factor 1α (TEF1) and the RNA polymerase II beta subunit (RPB2) genes were performed with primers EF1 and EF2 (O'Donnell et al. 1998), and 5f2 (Reeb et al. 2004) and 7cr (Liu et al. 1999), respectively. The sequences of three isolates showed 100% identities with the corresponding sequences of the strain NRRL 66652 of Fusarium algeriense LarabaO'Donnell (TEF1: MF120515 and RPB2: MF120504), and the sequences of a representative isolate (KyrFa01) were deposited in GenBank (TEF1: OM135603 and RPB2: OM135604). On PDA, fungal colonies were initially yellowish-white but gradually turned yellowish-brown. Ellipsoidal microconidia produced in false heads on monophialides were usually aseptate (8.30 ± 1.17 μm, n = 50) and occasionally one-septate (21.89 ± 2.01 μm, n = 50). Sporodochial macroconidia were mostly 3-4 septate measuring 43.41 ± 2.83 μm (n = 50), slightly curved and formed generally on monophialides on SNA. No chlamydospores formation was detected after 15 days on SNA or PDA. Morphological characteristics described above were consistent with the morphology of F. algeriense, as reported by Laraba et al. (2017). To confirm pathogenicity, seeds of wheat cultivar Seri 82, Fusarium crown rot susceptible, were treated in 1% NaOCl for 2 min, rinsed twice, and placed in plates containing a piece of sterile filter paper saturated with water to induce germination for 3 days. Five pregerminated seeds were placed on the soil surface for each 9-cm-diameter pot, which was filled with a sterile potting mix containing peat, vermiculite, and soil (1:1:1 by v/v/v). A 1-cm-diameter mycelial plug taken from the margin of actively growing colonies (PDA) of the representative isolate KyrFa01 was contacted with each seed, and then seeds were covered with the same potting mix. The seeds in control pots were treated with sterile PDA plugs. The experiment was conducted in a growth chamber in a completely randomized design with five replicated pots at 23°C with a 12-h photoperiod. Disease assessment was made after 4 weeks of fungal inoculation. The isolate KyrFa01 induced discoloration on the crown and root tissues of inoculated plants similar to those observed in the field-grown plants, whereas no symptoms were observed on plants grown in the control pots. The pathogen was successfully reisolated from the symptomatic tissues, confirming Koch's postulates. To the best of our knowledge, this is the first report of crown rot caused by F. algeriense on wheat in Kyrgyzstan. Fusarium algeriense was firstly described within the Fusarium burgessii species complex by Laraba et al. (2017) as a crown rot pathogen of wheat in Algeria. The pathogen was secondly reported from wheat-growing areas in Azerbaijan (Özer et al. 2020a) and thirdly from Kyrgyzstan in this report. Özer et al. (2020b) confirmed the coexistence of this pathogen with other Fusarium species. The result warrants the need to further investigate the potential of this species in the Fusarium crown rot complex of wheat.

Kazakhstan is one of the biggest wheat producers, however, its wheat production is far below the average international wheat production standard due to biotic and abiotic stressors. Plant-parasitic nematodes are devastating for cereal production systems worldwide. A comprehensive survey was conducted in 2019 to identify plant-parasitic nematodes associated with wheat in different locations of central, eastern, and southeastern Kazakhstan. The results revealed 33 root-lesion and 27 cyst nematode populations from the 77 localities sampled. These two genera occurred in separate or in mixed populations. The root-lesion populations were identified as Pratylenchus neglectus and P. thornei while all cyst nematodes were identified as Heterodera filipjevi. The identification of nematodes was firstly performed based on morphological and morphometric features and confirmed by BLAST and phylogenetic analyses based on the internal transcribed spacer and the D2-D3 expansion located in the 28S gene of ribosomal DNA for CCN and RLN populations, respectively. Pratylenchus neglectus and P. thornei populations from Kazakhstan showed a high similarity with the American, European, and Asian populations. Heterodera filipjevi populations formed a well-supported cluster with the corresponding populations from different countries and showed a slightly intraspecific polymorphism. Kazakhstan populations of H. filipjevi may have multiple introductions in Kazakhstan due to the divergence among them. The results of this study are of great importance for breeding programs and will enable awareness for extension advisors to develop measures to control these nematodes in cereal cropping areas in Kazakhstan. Triticale (×Triticosecale Wittmack) is obtained from wheat × rye crossing. It is positioned between wheat and rye in terms of resistance to soilborne pathogens including Gaeumannomyces graminis var. tritici, Fusarium culmorum, F. avenaceum, and Bipolaris sorokiniana (Arseniuk and Goral 2015). In 2019, seven triticale fields were surveyed in Almaty Province, Kazakhstan to examine soil-borne fungal pathogens. A total of 28 symptomatic plants with stunting, rot or discolored root were collected to identify causal agents. The overall disease incidence was approximately 8 to 10% in the fields. Fungi were isolated from 3-5 mm pieces excised from symptomatic tissues. The pieces were exposed to surface disinfection in 1% sodium hypochlorite solution for 2 min, rinsed three times with sterile distilled water, blotted dry, and plated on 1/5 strength potato dextrose agar (PDA) amended with 0.01% streptomycin. Plates were left in the dark at 23°C for 7 days. A total of 34 fungal colonies were isolated of which nineteen isolates, originally from six fields showed the cultural characteristics of B. sorokiniana. This species was previously reported to cause common root rot on triticale in Kazakhstan (Ozer et al. 2020). Ten isolates from four fields produced pale orange and cottony mycelium with red pigmentation on the agar, which is typical of Fusarium-like growth. The remaining isolates (n=5) from two fields produced salmon-colored and scarce aerial mycelium with no soluble pigmentation, similar to Microdochium spp. Fusarium isolates produced thick-walled and curved macroconidia with 3-4 septa (n=50, 25.7 to 37.6 × 4.1 to 7.3 μm in size) and notched basal cell on PDA, but microconidia were absent, which matches the description of F. culmorum (Wm.G. Sm.) Sacc. (Leslie and Summerell 2006). Microdochium isolates produced swollen, brown, and thick-walled chlamydospores and hyaline, one-celled, and thin-walled conidia (n=50, 5.4 to 9.3 × 1.5 to 3.0 μm in size) formed on ampullate and cylindrical conidiogenous cells on oatmeal agar (OA). These morphological features are consistent with previous observations for Microdochium bolleyi (R. Sprague) de Hoog & Herm.-Nijh. (Hong et al. 2008). To confirm morphological preliminary identifications, the portion of the translation elongation factor 1-alpha (EF1-α) gene was amplified with EF1/EF2 primers (O'Donnell et al. 1998) for representative Fusarium isolates (n=4) for each field. Additionally, the internal transcribed spacer (ITS) of ribosomal DNA was amplified with ITS1/ITS4 primers (White et al. 1990) for representative Microdochium isolates (n=2) for each field. BLASTn queries against NCBI GenBank revealed that the EF1-α sequences of Fusarium isolates (MW311081-MW311084) shared 100% identity with F. culmorum strain CBS 110262 (KT008433). The ITS sequences of M. bolleyi isolates (MW301448-MW301449) matched that of M. bolleyi strain CBS 137.64 (AM502264) with 100% sequence similarity. Pathogenicity test was conducted on pregerminated seeds of triticale cv. Balausa. A plastic pot (17 cm height, 9 cm in diam) was filled with a sterile mixture of vermiculite, peat, and soil (1:1:1, v/v/v). Mycelial plugs (1 cm in diam) were cut from the margin of a growing culture of representative isolates (Kaz_Fus123 and Kaz_Mb01) and placed onto the mixture in the pot. A sterile agar plug was employed as a control treatment. One pregerminated seed was put on the plug and covered with the mixture. The pots were transferred to a growth chamber set at 23 ± 2°C and a photoperiod of 14 hours. The experiment was performed twice using 5 replication pots per isolate. Four weeks after inoculation, discoloration of the crown was observed on all the inoculated roots, whereas no symptoms were observed on the control plants. Koch's postulates were fulfilled by reisolating and identifying the pathogen based on the morphology described above. This is the first report of M. bolleyi and F. culmorum causing root rot on triticale in Kazakhstan. Although B. sorokiniana is the most primary pathogen that may limit yield in the production of triticale in Kazakhstan, F. culmorum and M. bolleyi have been found to be less frequent and less aggressive pathogens, respectively. Further studies are needed to better understand the potential distribution and impact of these pathogens on triticale.

Plant parasitic nematodes are harmful organisms that cause economic losses in cultivated plants. This study was carried out to determine the plant parasitic nematode species found in potato cultivation areas of Talas and Tokmok Regions located in the North of Kyrgyzstan. For this purpose, 15 plant and soil samples were taken from potato cultivation areas during the survey studies conducted in the summer of 2016. The identification of the plant parasitic nematode species obtained from the samples was made by classical methods using morphological and morphometric measurements. As a result of the diagnostic studies carried out, 4 families belonging to the Tylenchida order and 6 species belonging to 6 genera were determined. These species are Bitylenchus goffarti (Sturhan, 1966), Ditylenchus destructor (Thorne, 1945), Filenchus cylindricus (Thorne Malek, 1968), Helicotylenchus vulgaris (Yuen, 1964), Rotylenchus buxophilus and Scutylenchus rugosus (Sıddoqii, 1963). The most common species identified in the study were recorded as F. cylindricus and B. goffarti. All the species obtained are new records for Kyrgyzstan and D. destructor (potato tuber nematode) is a pest causing significant losses in the potato plant., Bitki paraziti nematodlar kültür bitkilerinde ekonomik kayıplara neden olan zararlı organizmalardır. Bu çalışma Kırgızistan’ın kuzeyinde yer alan Talas ve Tokmok Bölgeleri patates ekiliş alanlarında bulunan bitki paraziti nematod türlerini saptamak amacıyla yürütülmüştür. Bu amaçla 2016 yılı yaz aylarında yapılan survey çalışmalarında patates ekiliş alanlarından 15 adet bitki ve toprak örneği alınmıştır. Örneklerden elde edilen bitki paraziti nematod türlerinin teşhisleri morfolojik ve morfometrik ölçümler yapılarak klasik yöntemlerle belirlenmiştir. Yürütülen teşhis çalışmaları sonucunda Tylenchida takımına ait 4 familya ve 6 cinse bağlı 6 tür tespit edilmiştir. Bu türler Bitylenchus goffarti (Sturhan, 1966), Ditylenchus destructor (Thorne, 1945), Filenchus cylindricus (Thorne Malek, 1968), Helicotylenchus vulgaris (Yuen, 1964), Rotylenchus buxophilus ve Scutylenchus rugosus (Sıddoqii, 1963)’dur. Çalışmada saptanan en yaygın türler ise F. cylindricus ve B. goffarti olarak kaydedilmiştir. Elde edilen tüm türler Kırgızistan için yeni kayıt niteliğinde olup D. destructor (patates çürüklük nematodu) patates bitkisinde önemli kayıplara yol açan bir zararlıdır.
