Rice bean is an underutilized legume crop cultivated in Asia, and it is a good source of protein, minerals, and essential fatty acids for human consumption. Moreover, the leaves left over after harvesting rice bean seeds contain various biological constituents beneficial to humans and animals. In our study, we performed a de-novo transcriptome assembly of rice bean, characterized the WRKY transcription factors, and studied their response to aluminum stress. A total of 46.6 million clean reads, with a GC value of 43%, were generated via transcriptome sequencing. De novo assembly of the clean reads resulted in 90,933 transcripts and 74,926 unigenes, with minimum and maximum lengths of 301 bp and 24,052 bp, and N50 values of 1801 bp and 1710 bp, respectively. A total of 27,095 and 28,378 unigenes were annotated and subjected to GO and KEGG analyses. Among the unigenes, 15,593, 20,770, and 15,385 unigenes were identified in the domains of biological process, molecular function, and cellular component, respectively. A total of 16,132 unigenes were assigned to 188 pathways, including metabolic pathways (5500) and secondary metabolite biosynthesis (2858). Transcription factor analysis revealed 4860 unigenes from 98 different transcription factor families. For WRKY, a total of 95 unigenes were identified. Further analysis revealed the diverse response of WRKY transcription factors to aluminum stress. Collectively, the results of this study boost genomic resources and provide a baseline for further research on the role of WRKY transcription factors in aluminum tolerance in rice bean.

Crop yield varies considerably within agroecology depending on the genetic potential of crop cultivars and various edaphic and climatic variables. Understanding site-specific changes in crop yield and genotype × environment interaction are crucial and needs exceptional consideration in strategic breeding programs. Further, genotypic response to diverse agro-ecologies offers identification of strategic locations for evaluating traits of interest to strengthen and accelerate the national variety release program. In this study, multi-location field trial data have been used to investigate the impact of environmental conditions on crop phenological dynamics and their influence on the yield of mungbean in different agroecological regions of the Indian subcontinent. The present attempt is also intended to identify the strategic location(s) favoring higher yield and distinctiveness within mungbean genotypes. In the field trial, a total of 34 different mungbean genotypes were grown in 39 locations covering the north hill zone (n = 4), northeastern plain zone (n = 6), northwestern plain zone (n = 7), central zone (n = 11) and south zone (n = 11). The results revealed that the effect of the environment was prominent on both the phenological dynamics and productivity of the mungbean. Noticeable variations (expressed as coefficient of variation) were observed for the parameters of days to 50% flowering (13%), days to maturity (12%), reproductive period (21%), grain yield (33%), and 1000-grain weight (14%) across the environments. The genotype, environment, and genotype × environment accounted for 3.0, 54.2, and 29.7% of the total variation in mungbean yield, respectively (p < 0.001), suggesting an oversized significance of site-specific responses of the genotypes. Results demonstrated that a lower ambient temperature extended both flowering time and the crop period. Linear mixed model results revealed that the changes in phenological events (days to 50 % flowering, days to maturity, and reproductive period) with response to contrasting environments had no direct influence on crop yields (p > 0.05) for all the genotypes except PM 14-11. Results revealed that the south zone environment initiated early flowering and an extended reproductive period, thus sustaining yield with good seed size. While in low rainfall areas viz., Sriganganagar, New Delhi, Durgapura, and Sagar, the yield was comparatively low irrespective of genotypes. Correlation results and PCA indicated that rainfall during the crop season and relative humidity significantly and positively influenced grain yield. Hence, the present study suggests that the yield potential of mungbean is independent of crop phenological dynamics; rather, climatic variables like rainfall and relative humidity have considerable influence on yield. Further, HA-GGE biplot analysis identified Sagar, New Delhi, Sriganganagar, Durgapura, Warangal, Srinagar, Kanpur, and Mohanpur as the ideal testing environments, which demonstrated high efficiency in the selection of new genotypes with wider adaptability.

Black gram (Vigna mungo) is an important pulse crop of India. The leaf crinkle disease caused by the urdbean leaf crinkle virus (ULCV) is a severe threat to black gram production. Black gram plants infected by ULCV show a considerable decline in plant growth and yield. However, detailed information about the interactions between the host, black gram, and ULCV is unclear. This study investigated the responses of two cultivars VBN (Bg) 6 and CO 5 to ULCV infection by physiological, biochemical, and transcriptional analyses. Virus symptoms were mild in VBN (Bg) 6 but were serious in CO 5. Upon the viral infection, VBN (Bg) 6 exhibited a low reduction in chlorophyll content than CO 5. The levels of sugar, protein, phenol, hydrogen peroxide (H2O2), and malondialdehyde (MDA) contents were altered by a viral infection in both cultivars. Although, the activities of antioxidant enzymes [Ascorbate peroxidase (APX), superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT)] were increased by ULCV infection. Following the viral infection, the expression level of the salicylic acid (SA), jasmonic acid (JA), and ethylene (ET) defense pathway-related genes was recorded higher in the VBN (Bg) 6 than that recorded in CO 5, indicating a positive correlation between resistance and these indicative indices. This dynamic physio-biochemical and transcriptional profiles of two black gram cultivars on ULCV infection augment our understanding of the complex interaction response between this crop and its pathogen. Additionally, it offers an inventory of potential indicators for future black gram screening and breeding to enhance resistance.

Urdbean (Vigna mungo L. Hepper) is one of the important pulse crops. Its cultivation is not so popular during summer seasons because this crop is unable to withstand excessive heat stress beside lack of humidity in the atmosphere. Therefore, a panel of 97 urdbean diverse genotypes was assessed for yield under stress and non-stress conditions with an aim to identify heat tolerant genotypes. This study identified 8 highly heat tolerant and 35 highly heat sensitive genotypes based on heat susceptibility index. Further, physiological and biochemical traits-based characterization of a group of six highly heat sensitive and seven highly heat tolerant urdbean genotypes showed genotypic variability for leaf nitrogen balance index (NBI), chlorophyll (SPAD), epidermal flavnols, and anthocyanin contents under 42/25°C max/min temperature. Our results showed higher membrane stability index among heat tolerant genotypes compared to sensitive genotypes. Significant differences among genotypes for ETR at different levels of PAR irradiances and PAR × genotypes interactions indicated high photosynthetic ability of a few genotypes under heat stress. Further, the most highly sensitive genotype PKGU-1 showed a decrease in different fluorescence parameters indicating distortion of PS II. Consequently, reduction in the quantum yield of PS II was observed in a sensitive one as compared to a tolerant genotype. Fluorescence kinetics showed the delayed and fast quenching of Fm in highly heat sensitive (PKGU 1) and tolerant (UPU 85-86) genotypes, respectively. Moreover, tolerant genotype (UPU 85-86) had high antioxidant activities explaining their role for scavenging superoxide radicals (ROS) protecting delicate membranes from oxidative damage. Molecular characterization further pinpointed genetic differences between heat tolerant (UPU 85-86) and heat sensitive genotypes (PKGU 1). These findings will contribute to the breeding toward the development of heat tolerant cultivars in urdbean.

In the present study, fifty-two mungbean (Vigna radiata) genotypes were evaluated for seven morphological traits at three different environments in South Indian state Tamil Nadu, namely Virinjipuram (E1), Eachangkottai (E2), and Bhavanisagar (E3) during Kharif 2017, 2018, and 2019, respectively. The data collected were subjected to variability and correlation analyses, followed by stability analysis using additive main effects and multiplicative interaction (AMMI) model, genotype and genotype × environment interaction effects (GGE) biplot. Variablility was observed among the genotypes for the following traits viz., plant height, days to fifty per cent flowering, number of pods per plant, pod length, number of seeds per pod, hundred seed weight and grain yield. Correlation analysis showed that the trait number of pods per plant was significantly associated with grain yield. The G × E was smaller than the genetic variation of grain yield as it portrayed the maximum contribution of genotypic effects (61.07%). GGE biplot showed E3 as a highly discriminating and representative environment. It also identified environment-specific genotypes viz., EC 396111 for E1, EC 396125 for E2 and EC 396101 for E3 environments. The genotypes with minimum genotype stability index (GSI) viz., V2802BG (7), HG 22 (13), and EC 396098 (13) were observed with wide adaptation and high yields across all the three environments. In summary, we identified stable genotypes adapted across environments for grain yield. These genotypes can be used as parent/pre-breeding materials in future mungbean breeding programs.

Mungbean is a nutritionally and economically important pulse crop cultivated around Asia, mainly in India. The crop is sensitive to drought at various developmental stages of its growing period. However, there is limited or almost no research on a comparative evaluation of mung-bean plants at the flowering stage under drought conditions. Hence, the aim of this research was to impose the drought stress on two mungbean cultivars VRM (Gg) 1 and CO6 at the flowering stage and assess the physio-biochemical and transcriptional changes. After imposing the drought stress, we found that VRM (Gg) 1 exhibited a low reduction in physiological traits (Chlorophyll, relative water content, and plant dry mass) and high proline content than CO6. Additionally, VRM (Gg) 1 has a low level of H2O2 and MDA contents and higher antioxidant enzymes (SOD, POD, and CAT) activity than CO6 during drought stress. The transcriptional analysis of photosynthesis (PS II-PsbP, PS II-LHC, PS I-PsaG/PsaK, and PEPC 3), antioxidant (SOD 2, POD, CAT 2), and drought-responsive genes (HSP-90, DREB2C, NAC 3 and AREB 2) show that VRM (Gg) 1 had increased transcripts more than CO6 under drought stress. Taken together, VRM (Gg) 1 had a better photosynthetic performance which resulted in fewer reductions in chlorophyll, relative water content, and plant dry mass during drought stress. In addition, higher antioxidative enzyme activities led to lower H2O2 and MDA levels, limiting oxidative damage in VRM (Gg) 1. This was positively correlated with increased transcripts of photosynthesis and antioxidant-related genes in VRM (Gg) 1. Further, the increased transcripts of drought-responsive genes indicate that VRM (Gg) 1 has a better genetic basis against drought stress than CO6. These findings help to understand the mungbean response to drought stress and will aid in the development of genotypes with greater drought tolerance by utilizing natural genetic variants.

Yellow mosaic disease (YMD), incited by mungbean yellow mosaic virus (MYMV), is a primary viral disease that reduces mungbean production in South Asia, especially in India. There is no detailed knowledge regarding the genes and molecular mechanisms conferring resistance of mungbean to MYMV. Therefore, disclosing the genetic and molecular bases related to MYMV resistance helps to develop the mungbean genotypes with MYMV resistance. In this study, transcriptomes of mungbean genotypes, VGGRU-1 (resistant) and VRM (Gg) 1 (susceptible) infected with MYMV were compared to those of uninfected controls. The number of differentially expressed genes (DEGs) in the resistant and susceptible genotypes was 896 and 506, respectively. Among them, 275 DEGs were common between the resistant and susceptible genotypes. Functional annotation of DEGs revealed that the DEGs belonged to the following categories defense and pathogenesis, receptor-like kinases; serine/threonine protein kinases, hormone signaling, transcription factors, and chaperons, and secondary metabolites. Further, we have confirmed the expression pattern of several DEGs by quantitative real-time PCR (qRT-PCR) analysis. Collectively, the information obtained in this study unveils the new insights into characterizing the MYMV resistance and paved the way for breeding MYMV resistant mungbean in the future.

Investigations on the diversity of Odonata in and around the Agricultural College and Research Institute, Vazhavachanur, Tamil Nadu, India were studied. Eight locations were selected, of which sixteen Odonata species were recorded. In total, eleven dragonfly and five damselfly species were identified from Thiruvannamalai district, Tamil Nadu, India. Pantala flavescens, Diplacodes trivialis, Brachythemis contaminata and Ischnura aurora were recorded from all eight locations. Trithemis pallidinervis and Agriocnemis pygmaea were recorded from seven locations except from the farm pond and the open stretch area. Rhyothemis variegata was recorded only at the open stretch area. The results clearly show that, Odonates have specific habitat preferences for their growth and development. Four families Libellulidae, Gomphidae, Aeshnidae and Coenagrionidae were observed and collected during the study. Libellulidae were the most abundant family (56.25%) and comprised of 9 species, followed by Coenagrionidae (31.25%) with 5 species. Low species abundance was recorded within the Gomphidae and Aeshnidae families with 6.25% each. Simpson and Shannon Index showed maximum diversity in the open stretch area (0.8743 and 2.186, respectively) and minimum in location 8 (crop production area) and location 4 (Sathakuppam), respectively. Open stretch area showed maximum Richness Index (Margalef) (2.275) followed by farm pond (2.196) where recorded minimum in sathakuppam (1.276) location. Equitability J index, maximum species evenness (0.788) was recorded Location 6 i.e. farm pond (0.6727) followed by open stretch area 0.686.

Though crossing wild relatives to modern cultivars is a usual means to introduce alleles of stress tolerance, an alternative is de novo domesticating wild species that are already tolerant to various kinds of stresses. As a test case, we chose Vigna stipulacea Kuntze, which has fast growth, short vegetative stage, and broad resistance to pests and diseases. We developed an ethyl methanesulfonate–mutagenized population and obtained three mutants with reduced seed dormancy and one with reduced pod shattering. We crossed one of the mutants of less seed dormancy to the wild type and confirmed that the phenotype was inherited in a Mendelian manner. De novo assembly of V. stipulacea genome, and the following resequencing of the F2 progenies successfully identified a Single Nucleotide Polymorphism (SNP) associated with seed dormancy. By crossing and pyramiding the mutant phenotypes, we will be able to turn V. stipulacea into a crop which is yet primitive but can be cultivated without pesticides.

The genus Vigna (Fabaceae) consists of five subgenera, and includes more than 100 wild species. In Vigna, 10 crops have been domesticated from three subgenera, Vigna, Plectrotropis, and Ceratotropis. The habitats of wild Vigna species are so diverse that their genomes could harbor various genes responsible for environmental stress adaptation, which could lead to innovations in agriculture. Since some of the gene bank Vigna accessions were unidentified and they seemed to be novel genetic resources, these accessions were identified based on morphological traits. The phylogenetic positions were estimated based on the DNA sequences of nuclear rDNA-ITS and chloroplast atpB-rbcL spacer regions. Based on the results, the potential usefulness of the recently described species V. indica and V. sahyadriana, and some wild Vigna species, i.e., V. aconitifolia, V. dalzelliana, V. khandalensis, V. marina var. oblonga, and V. vexillata, was discussed.
