Beech leaf disease (BLD), caused by the Litylenchus crenatae ssp. mccannii (Lcm) nematode, is an emerging threat to beech trees. This disease is characterized by distinct leaf symptoms, including leaf interveinal banding and thickened leaf texture, which leads to eventual tree mortality. Understanding Lcm dispersal mechanism(s) is crucial for managing BLD, yet these remain largely unknown, posing a major barrier to its effective management. This study represents a pioneering investigation into the abiotic and biotic vectors that potentially contribute to the local dispersal of Lcm in natural American beech (Fagus grandifolia) forest systems in the Northeastern United States. An experiment was set up in Stone Valley Forest, Pennsylvania (PA), using four funnel stands placed at variable distances from naturally BLD-infected beech trees. This approach enabled the recovery of active Lcm nematodes from each funnel, demonstrating their ability to naturally disperse at least 11.74 m from the nearest BLD-infected tree. The findings highlight the role of abiotic factors involved in the dispersal dynamics of Lcm, especially wind and humidity, as indicated by a generalized linear model. The current study also uncovered the incidental association of Lcm with other organisms beneath the canopy of BLD trees, including spiderwebs and caterpillars. To our knowledge, this is the first study to document the potential vectors involved in the local dispersal of Lcm, offering valuable information for the biology of this nematode, as well as insight into the development of effective BLD management strategies. A new species, Ditylenchoides anyae sp. n., recovered from soil in the headland areas of non-cultivated fields in Alborz Province, Iran, is described and illustrated based on its morphological, morphometric, and molecular characters. The new species was characterised by a slender body and length of 784 (663-925) μm (females) and 667-712 μm (males), lip region continuous, stylet length 6.4 (6.0-7.0) μm, lateral fields with four incisures, and excretory pore situated 110 (82-137) μm from the anterior end, median bulb of pharynx fusiform, non-muscular, and non-valvate, isthmus elongate, slender ending at a pyriform basal pharyngeal bulb not overlapping intestine, vulva with distinct and relatively large lateral flaps, a well-developed post-vulval uterine sac, 31.3 (27-35) μm long, a female tail elongate-conoid with pointed terminus; and male with adanal bursa and spicules 21-22 μm long. The new species was characterised using the D2-D3 expansion segments of 28S rRNA, ITS rRNA, and partial 18S rRNA gene sequences. The synaptonemal complex (SC) is a meiotic interface that assembles between parental chromosomes and is essential for gamete formation. While the dimensions and ultrastructure of the SC are conserved across eukaryotes, its protein components are highly divergent. Recently, an unexpected component of the SC has been described in the nematode Caenorhabditis elegans : the Skp1-related proteins SKR-1/2, which are components of the Skp1, Cullin, F-box (SCF) ubiquitin ligase. Here, we find that the role of SKR-1 in the SC is conserved in Pristionchus pacificus. The P. pacificus Skp1 ortholog, Ppa-SKR-1 , colocalizes with other SC proteins throughout meiotic prophase, where it occupies the middle of the SC. Like in C. elegans , the dimerization interface of Ppa-SKR-1 is required for its SC function. A dimerization mutant, ppa-skr-1F105E , fails to assemble SC and produces almost no progeny. Interestingly, the evolutionary trajectory of SKR-1 contrasts with other SC proteins. Unlike most SC proteins, SKR-1 is highly conserved in nematodes. Our results suggest that the structural role of SKR-1 in the SC has been conserved since the common ancestor of C. elegans and P. pacificus , and that rapidly evolving SC proteins have maintained the ability to interact with SKR-1 for at least 100 million years. Life history is defined by traits that reflect key components of fitness, especially those relating to reproduction and survival. Research in life history seeks to unravel the relationships among these traits and understand how life history strategies evolve to maximize fitness. As such, life history research integrates the study of the genetic and developmental mechanisms underlying trait determination with the evolutionary and ecological context of Darwinian fitness. As a leading model organism for molecular and developmental genetics, Caenorhabditis elegans is unmatched in the characterization of life history-related processes, including developmental timing and plasticity, reproductive behaviors, sex determination, stress tolerance, and aging. Building on recent studies of natural populations and ecology, the combination of C. elegans ' historical research strengths with new insights into trait variation now positions it as a uniquely valuable model for life history research. In this review, we summarize the contributions of C. elegans and related species to life history and its evolution. We begin by reviewing the key characteristics of C. elegans life history, with an emphasis on its distinctive reproductive strategies and notable life cycle plasticity. Next, we explore intraspecific variation in life history traits and its underlying genetic architecture. Finally, we provide an overview of how C. elegans has guided research on major life history transitions both within the genus Caenorhabditis and across the broader phylum Nematoda. While C. elegans is relatively new to life history research, significant progress has been made by leveraging its distinctive biological traits, establishing it as a highly cross-disciplinary system for life history studies. During challenge of Caenorhabditis elegans with human bacterial pathogens such as Pseudomonas aeruginosa and Enterococcus faecalis , the elicited host response can be damaging if not properly controlled. The activation of Nrf (nuclear factor erythroid-related factor)/CNC (Cap-n-collar) transcriptional regulators modulates the response by upregulating genes that neutralize damaging molecules and promote repair processes. Activation of the C. elegans Nrf ortholog, SKN-1 , is tightly controlled by a myriad of regulatory mechanisms, but a central feature is an activating phosphorylation accomplished by the p38 mitogen-activated kinase (MAPK) cascade. In this work, loss of CDC-48, an AAA+ ATPase, was observed to severely compromise SKN-1 activation on pathogen and we sought to understand the mechanism. CDC-48 is part of the endoplasmic reticulum (ER)-associated degradation (ERAD) complex where it functions as a remodeling chaperone enabling the translocation of proteins from the ER to the cytoplasm for degradation by the proteosome. Interestingly, one of the proteins retrotranslocated by ERAD, a process necessary for its activation, is SKN-1A, the ER isoform of SKN-1. However, we discovered that SKN-1A is not activated by pathogen exposure in marked contrast to the cytoplasmic-associated isoform SKN-1C. Rather, loss of CDC-48 blocks the antioxidant response normally orchestrated by SKN-1C by strongly inducing the unfolded protein response (UPRER). The data are consistent with the model of these 2 pathways being mutually inhibitory and support the emerging paradigm in the field of coordinated cooperation between different stress responses. The East Asian finless porpoise, Neophocaena asiaeorientalis sunameri, is an endangered species that inhabits the coastal marine environments of East Asia. In the present study, we investigated the overall infection status of anisakid nematodes in East Asian finless porpoises from three sea sectors off the Korean Peninsula. The genetic diversity and population genetic structure of the identified nematode species were evaluated. The prevalence of all stages of anisakid nematodes collected from the stomach was 57.55% (61 among the 106 porpoises examined), and 16 of the hosts were found to have adult worms. The mean number of infected adults was 211 (± 419.54, 5–1455 per host). Only one species of anisakids, Anisakis pegreffii, was identified from randomly selected worms by molecular approaches. Analysis of the mitochondrial (mt) cox2 partial gene in 50 newly generated sequences of A. pegreffii revealed 24 haplotypes, including 14 new haplotypes. We observed below-average levels of nucleotide diversity and haplotype diversity compared to other seas around the world. The mtDNA cox2 haplotypes of the species in the three Korean sea areas showed no genetic structure, suggesting well-connected gene flow within these areas. This study represents the first record of a definitive host of A. pegreffii in Korean waters, providing important information regarding anisakids genetic diversity in the cetacean species inhabiting limited regions. ABSTRACT Histopathological studies of parasitic infections in fish from the natural environment of Brazilian Amazon, are quite scarce. In this study, we investigated the histopathological changes of the proximal intestine of specimens of the Amazonian fish Hoplias malabaricus infected by the hematophagous nematode Procamallanus (Spirocamallanus) paraensis. The prevalence of the infection was 60%, with an average abundance of 1.46 and an average intensity of 2.43 parasites/fish. No significant difference was observed in the prevalence of infection and fish size or sex, but larger fish showed greater infection intensity, which was also significantly higher in male hosts. Histological sections of the proximal intestine showed reduction and loss of the epithelial lining, exposure of the lamina propria where the nematode interacts with the intestine wall, through the insertion of the buccal capsule and fish cellular debris in the intestinal lumen. In addition, areas with bleeding and inflammatory infiltrate were observed, but no changes or presence of parasite structures were observed in the other tunics of the intestinal wall. Locomotion is essential to most of the living organisms. The nematode Teratorhabditis palmarum (Gerber & Giblin-Davis, 1990), must be able to accommodate its locomotory gait to its physical environment. Particularly, when crawl on a solid surface, it shows forward movement as the contraction waves propagates from anterior to the posterior region. In the present study, we analyzed the locomotory behavior of T. palmarum on agar by using computational worm-tracking system and mathematical tools. Our tracking system was made to assist worms that travel a vast area with recurrent turns and reversals at high speeds. The locomotory parameters studied were velocity, wavelength, amplitude, angle of bending, curvature map, forward, reversal, omega and pause that was extracted from a two min video recording. Here, we examined the locomotory behaviour (movement and wave characteristic) of two, six and ten days old nematodes. In movement characteristic, except pause which increases; forward wave, backward wave and omega wave decrease significantly. In wave characteristic, wavelength and amplitude increase while average velocity and bending angle decrease significantly. Locomotion was observed between various age groups and it was concluded that as the nematodes aged, mild locomotory deficits appeared that were enough to reduce behavioral activity. Overall, our result provides the detailed analysis of locomotion and its dynamics in enabling better understand the mechanism of muscle ageing. [ABSTRACT FROM AUTHOR]

Sediment is the ultimate sink of environmental pollutants. A total of 128 surface sediment samples were collected from 8 rivers and 3 reservoirs in Maoming City, Guangdong Province. This study assessed the content and distribution of brominated flame retardants in sediments. The acute toxicity effects of tetrabromobisphenol A (TBBPA) and hexabromocyclododecane (HBCDs) in sediments were evaluated using Caenorhabditis elegans as model organisms. The concentration of TBBPA in sediments ranged from not detected (ND) to 12.59 µg/kg and was mainly distributed in the central area, which was affected by the emission of TBBPA from residential and factory. The concentration of HBCDs ranged from ND to 6.31 µg/kg, and the diastereoisomer distribution was consistent, showing a trend close to the South China Sea. The composition pattern of HBCDs in the surface sediments from rivers were 41.73%–62.33%, 7.89%–25.54%, and 18.76%–40.65% for α-, β-, and γ-HBCD, respectively, and in the sediments from reservoirs were 26.15%–45.52%, 7.44%–19.23%, and 47.04%–61.89% for α-, β-, and γ-HBCD, respectively. When the sum of concentrations of TBBPA and HBCD in sediments were above high levels, reactive oxygen species in nematodes significantly increased, resulting in an oxidative stress response. Intestinal permeability was also enhanced, causing intestinal damage. In addition, in terms of this study, TBBPA had a greater impact on biotoxicity compared to HBCDs, and more attention should be paid to the toxic effects of the river ecosystem organisms in Maoming City, Guangdong Province. This study assessed the prevalence and risk factors associated with gastrointestinal parasite infection in goats in Ayacucho, Peru. Fecal samples were collected from a total of 254 goats from four districts of Ayacucho: Ocaña (89), Colca (76), Pacaicasa (64), and Luricocha (25) during the dry season; recording the location, sex, and age of the animals. The fecal samples were analyzed using the flotation technique in salt and sugar solution, and modified McMaster egg counting techniques for eggs/oocysts (epg/opg) of gastrointestinal parasites. The associations between location, sex, and age with the prevalence of gastrointestinal parasites were analyzed using the chi-square test. Fecal samples showed an overall parasite prevalence of 87.80%. The identified parasites were Eimeria spp. (86.22%), Strongyle type eggs (STE) (65.75%), Skrjabinema spp. (7.87%), Trichuris spp. (3.15%) and Moniezia spp. (3.15%). There was a significant association between the location and the presence of parasites (p < 0.05); where STE was more prevalent in the district of Colca (98.68%); Trichuris spp. in Pacaycasa (9.38%); Skrjabinema spp. (48.00%), Moniezia spp. (12.00%) and Eimeria spp. (100%) were more prevalent in Luricocha. Sex had a significant association with the prevalence of Eimeria spp., Skrjabinema spp. and Moniezia spp. (p < 0.05); while age had a significant association with parasitosis only for Trichuris spp. and Moniezia spp. (p < 0.05). The epg/opg values revealed a higher parasite burden in goats from Colca compared to the other locations (p < 0.05). There was also no statistical relationship between fecal egg/oocyst counts and sex or age, nevertheless, there were moderate and high parasitic burdens. The high prevalence of parasites such as nematodes, cestodes, and coccidia suggests the need to implement strategic control and prevention programs in free-grazing goats. Meloidogyne incognita is one of the globally serious plant parasitic nematodes. New control measure is urgently needed to replace the common chemical control method. Ascarosides are pheromones regulating the nematodes' aggregation, avoidance, mating, dispersal and dauer recovery and formation. Ascr#9, one of the ascarosides, exhibits the potential to repel M. incognita. However, the nematode genes involved in the perception of ascr# 9 signal are totally unknown. In this study, the transcriptome of ascr#9-treated second stage M. incognita juveniles (J2s) was analyzed, 44 pathways were significantly affected, multiple ligand-receptor and mucin type O-glycan were induced, and olfactory transduction was disturbed. A total of 11 highly differentially expressed genes involved in neuroactive ligand-receptor interaction and FMRFamide-like peptide related process were identified and knocked down by RNAi. The dispersal rates of M. incognita with three knocked-down genes (flp-14, mgl-1 and ADOR-1) significantly decreased, respectively, when ascr#9 was present. The results demonstrate that flp-14, mgl-1, and ADOR-1 are involved in the dispersal behavior of M. incognita nematodes responding to ascr#9, which promotes the interaction study between ascarosides and M. incognita, and provides new ideas for the prevention and control of M. incognita by using pheromone ascarosides. Background: Fertilization practices control bacterial wilt-causing Ralstonia solanacearum by shaping the soil microbiome. This microbiome is the start of food webs, in which nematodes act as major microbiome predators. However, the multitrophic links between nematodes and the performance of R. solanacearum and plant health, and how these links are affected by fertilization practices, remain unknown. Results: Here, we performed a field experiment under no-, chemical-, and bio-organic-fertilization regimes to investigate the potential role of nematodes in suppressing tomato bacterial wilt. We found that bio-organic fertilizers changed nematode community composition and increased abundances of bacterivorous nematodes (e.g., Protorhabditis spp.). We also observed that pathogen-antagonistic bacteria, such as Bacillus spp., positively correlated with abundances of bacterivorous nematodes. In subsequent laboratory and greenhouse experiments, we demonstrated that bacterivorous nematodes preferentially preyed on non-pathogen-antagonistic bacteria over Bacillus. These changes increased the performance of pathogen-antagonistic bacteria that subsequently suppressed R. solanacearum. ABSTRACT The quarantine nematode Bursaphelenchus xylophilus is a causative agent of the pine wilt disease. In Europe, the nematode is vectored by Monochamus galloprovincialis which is also a natural vector of the native, nonpathogenic B. mucronatus. Based on the established regulations, long‐term detection surveys of pine wood and vector insects for the presence of B. xylophilus are obligatory in all EU member countries. The main objective of the presented research was to use the real‐time PCR technique for the fast and precise detection, and identification of Bursaphelenchus nematodes associated with M. galloprovincialis ssp. pistor occurring in Poland. The research conducted on beetles collected in forest stands in Poland revealed exclusive presence of the native, nonpathogenic nematode B. mucronatus. At the beginning of the beetle flight season (June–July), the use of the real‐time PCR allowed detection of B. mucronatus, in the case of its physical presence in the body of the dissected M. galloprovincialis. However, at the end of the flight season (September–October), the DNA of B. mucronatus was also found in some beetles with no physical presence of nematodes in tracheas or under elytra. The univoltine biology of the beetle, gradual decline in dispersal juveniles to zero in/on the beetle body, and positive results of real‐time PCR reaction could indicate that in some of the beetles earlier loaded with nematodes, at the end of the season, only their remains were left after the live nematodes' departure during the vector's maturation feeding and/or egg laying. The obtained results confirm extremely high sensitivity of the real‐time PCR method used in detection and identification of both nematode species which may simultaneously occur in M. galloprovincialis beetles. Background: The phylum Nematoda is incredibly diverse and includes many parasites of humans, livestock, and plants. Peptide-activated G protein-coupled receptors (GPCRs) are central to the regulation of physiology and numerous behaviors, and they represent appealing pharmacological targets for parasite control. Efforts are ongoing to characterize the functions and define the ligands of nematode GPCRs, with already most peptide GPCRs known or predicted in Caenorhabditis elegans. However, comparative analyses of peptide GPCR conservation between C. elegans and other nematode species are limited, and many nematode GPCRs remain orphan. A phylum-wide perspective on peptide GPCR profiles will benefit functional and applied studies of nematode peptide GPCRs. Results: We constructed a pan-phylum resource of C. elegans peptide GPCR orthologs in 125 nematode species using a semi-automated pipeline for analysis of predicted proteome datasets. The peptide GPCR profile varies between nematode species of different phylogenetic clades and multiple C. elegans peptide GPCRs have orthologs across the phylum Nematoda. We identified peptide ligands for two highly conserved orphan receptors, NPR-9 and NPR-16, that belong to the bilaterian galanin/allatostatin A (Gal/AstA) and somatostatin/allatostatin C (SST/AstC) receptor families. The AstA-like NLP-1 peptides activate NPR-9 in cultured cells and are cognate ligands of this receptor in vivo. In addition, we discovered an AstC-type peptide, NLP-99, that activates the AstC-type receptor NPR-16. In our pan-phylum resource, the phylum-wide representation of NPR-9 and NPR-16 resembles that of their cognate ligands more than those of allatostatin-like peptides that do not activate these receptors. Conclusions: The repertoire of C. elegans peptide GPCR orthologs varies across phylogenetic clades and several peptide GPCRs show broad conservation in the phylum Nematoda. Our work functionally characterizes the conserved receptors NPR-9 and NPR-16 as the respective GPCRs for the AstA-like NLP-1 peptides and the AstC-related peptide NLP-99. NLP-1 and NLP-99 are widely conserved in nematodes and their representation matches that of their receptor in most species. These findings demonstrate the conservation of a functional Gal/AstA and SST/AstC signaling system in nematodes. A total of 127 specimens of the "Blue Antimora" Antimora rostrata (Günther, 1878) were obtained from 2015 to 2019 as bycatch from the artisanal fishery of the Patagonian toothfish (Dissostichus eleginoides (Smitt, 1898)) at depths between 1000 and 2200 m in Northern Chile (app. 22° S 70° W). All individuals were examined for parasites. A total of seventeen parasite taxa, two Copepoda, two Monogenea, seven Digenea, three Nematoda, and three Cestoda, were found, and twelve taxa were found as adults while five taxa were found at the larval stage. Anisakis sp. (Nematoda) and Trypanorhyncha gen. sp. (Cestoda) were the predominant species with a prevalence of 53.5% and 11.8%, respectively. The high prevalence of Anisakis sp. (>50%) suggests that A. rostrata may play a significant role in the life cycle of Anisakis sp. in the southeastern Pacific Ocean. The detected parasite community, consisting predominantly of parasites from pelagic environments rather than benthopelagic, suggests that A. rostrata may fulfill a crucial role as a predator of pelagic organism communities. Faidherbia albida (Delile) A. Chev., hereafter F. albida is an important agroforestry tree species in the semi‐arid highlands of Tigray, northern Ethiopia. The paper analyses how different stand ages of the tree could influence nematodes' abundance, trophic composition and selected community index values. A total of 40 composite soil samples were collected from out‐canopies and under‐canopies of different tree stand age classes designated as young (15–20 years), medium age (35–40 years) and older (> 60 years). Nematodes were extracted following the Baermann funnel technique, identified and counted under an inverted Olympus microscope according to their trophic groups. The highest total count of nematodes and relative abundance of fungivores, omnivores and predatory nematode trophic groups were counted from soil samples influenced by older F. albida trees. The relative abundance of bacterivores and plant parasitic nematodes was low. The nematode channel ratio became lower in soils as influenced by older trees while the Wasilewska index was significantly high. The age difference of the trees influenced the abundance of the soil nematode feeding groups and their ecological index values. Moreover, the results have shown the enhancement of nematode trophic diversity. Redundancy analysis showed that tree age significantly influenced the relationships of nematode trophic groups with the studied soil bio‐physicochemical properties. In order to know whether there is a risk of anisakiasis (or anisakidosis) by consumption of fish of the genus Mullus from the western Mediterranean Sea, which are appreciated for their quality, an epidemiological survey was carried out to evaluate the occurrence of zoonotic or potentially zoonotic nematodes in M. barbatus and M. surmuletus. Although the presence of the third larval stage (L3) of anisakids (Anisakis and Contracaecum) has been previously described in these fish, the results showed the absence of anisakids and the presence, never in muscle, of L3 and L4 of raphidascaridids of the genus Hysterothylacium, molecularly identified as H. fabri. Phylogenetic analysis groups them into the Mediterranean Sea clade, far from individuals isolated in the Pacific Ocean. Prevalence was slightly higher, but not significant, in M. barbatus versus M. surmuletus (72.3% vs 60.0%), but mean intensity (MI) and mean abundance (MA) parameters were approximately twice as high in M. barbatus as in M. surmuletus (MI 5.8 vs 2.8, p =.001; MA 4.2 vs 1.7, p <.001). The presence of the parasite seems to have different effects on these two sympatric species. In M. barbatus it seems to affect their growth, as it appreciably reduces the value of allometry coefficient in infected fish (2.78 vs. 2.18). On the other hand, in M. surmuletus the infection significantly (p <.04) affects the Fulton's condition factor, an indicator of the health status of the fish. It can be concluded that the ingestion of these fish by the people poses negligible risk of anisakiasis, but the consumer should continue to be urged to follow the rules of prevention against this illness. The stubby root nematode, Paratrichodorus allius, is one of the most important plant-parasitic nematodes. Besides root feeding, P. allius also transmits the Tobacco rattle virus in potatoes, which causes corky ringspot disease. Rapid detection of P. allius is key for efficient management. This study was conducted to develop a real-time recombinase polymerase amplification (RPA) assay that is capable of detecting P. allius directly in DNA extracts from soil using a simple portable device in real time. A fluorophore-attached probe was designed to target the internal transcribed spacer (ITS)-rDNA of P. allius and was used along with primers designed previously. The real-time RPA assay had the ability to detect P. allius DNA extracted directly from infested soil with a sensitivity of one-sixteenth portion of a single nematode. This RPA assay was specific, as it did not produce positive signals from non-target nematodes tested. The real-time RPA was found to be rapid as it could even detect P. allius in as little as 7 min. Testing with 15 field soil samples validated the RPA assay developed in this study. This is the first report of P. allius detection directly from soil DNA using real-time RPA and is the fastest method for P. allius detection in soil to date. [ABSTRACT FROM AUTHOR]

Improved genetically encoded calcium indicators (GECIs) are essential for capturing intracellular dynamics of both muscle and neurons. A novel set of GECIs with ultrafast kinetics and high sensitivity was recently reported by Zhang et al. (2023). While these indicators, called jGCaMP8, were demonstrated to work in Drosophila and mice, data for Caenorhabditis elegans were not reported. Here, we present an optimized construct for C. elegans and use this to generate several strains expressing GCaMP8f (fast variant of the indicator). Utilizing the myo-2 promoter, we compare pharyngeal muscle activity measured with GCaMP7f and GCaMP8f and find that GCaMP8f is brighter upon binding to calcium, shows faster kinetics, and is not disruptive to the intrinsic contraction dynamics of the pharynx. Additionally, we validate its application for detecting neuronal activity in touch receptor neurons which reveals robust calcium transients even at small stimulus amplitudes. As such, we establish GCaMP8f as a potent tool for C. elegans research which is capable of extracting fast calcium dynamics at very low magnifications across multiple cell types. Shc (Src homologous and collagen) proteins function in many different signaling pathways where they mediate phosphorylation-dependent protein–protein interactions. These proteins are characterized by the presence of two phosphotyrosine-binding domains, an N-terminal PTB and a C-terminal SH2. We describe a previously unrecognized Caenorhabditis elegans Shc gene, shc-3 and characterize its role in stress response. Both shc-3 and shc-1 are required for long-term survival in L1 arrest and survival in heat stress, however, they do not act redundantly but rather play distinct roles in these processes. Loss of shc-3 did not further decrease survival of daf-16 mutants in L1 arrest, suggesting that like SHC-1, SHC-3 functions in the insulin-like signaling pathway. We advocate for the widespread adoption of molecular diagnostics, alongside traditional methods, to support sustainable nematode management. Tangible examples utilising molecular diagnostics as proof-of-concept studies are urgently needed, particularly those involving longitudinal multiple grazing season studies. While absolute quantification of parasite DNA in samples is a priority, it is equally important to define thresholds for treatment and to develop and optimise tools that consider the host and its specific immune response. Facilitating open dialogue between farmers and veterinarians regarding sustainable management is essential, encouraging joint planning and collaboration instead of treating them as distinct entities with separate agendas. The management of parasitic nematodes calls for a shift from conventional, indiscriminate, anthelmintic use to a more precise approach, directed by diagnostics. We should accept those parasite infection intensities that have minimal impact on production and welfare rather than attempt to eliminate them. The diagnostic toolbox for gastrointestinal nematodes (GINs) faces challenges due to anthelmintic resistance (AR), which is species-specific, drug-class-specific, and varies by region. We discuss which traditional tools may become obsolete and which tools need development to gain widespread use. Social science research highlights the need for dialogue between farmers and veterinarians that emphasises effective parasite management and upskilling the veterinary workforce for more sustainable practices centred on diagnostics to be adopted in practice by 2030. Mammalian P-glycoprotein (PGP) transports a broad range of lipophilic substrates, including the anthelmintic macrocyclic lactones (MLs), such as ivermectin (IVM). PGP controls the disposition of MLs in the host organism, and serves as a primary defender against IVM toxicity in mammals. Nematodes have several PGPs that can transport IVM, and PGP overexpression contributes to IVM resistance in parasitic nematodes. The transcription factors nuclear hormone receptor (NHR)-8 and CKY-1 are important for IVM resistance in Caenorhabditis elegans and Haemonchus contortus. Targeting NHR-8, an upstream regulator of PGPs, could enhance IVM efficacy, and delay the development of IVM resistance. P-glycoprotein (PGP) is a pivotal transmembrane transporter governing the cellular flux of diverse substances shielding mammals from toxics. It can thwart the effectiveness of medicines such as ivermectin (IVM) and other macrocyclic lactone (ML) anthelmintics, undermining therapeutic efforts. We analyze the role of PGPs in limiting the toxicity of these drugs in hosts, and their potential contribution to anthelmintic resistance in nematodes. Targeting nematode PGPs to increase drug sensitivity to MLs seems interesting, but is hampered by the lack of selective inhibitors. The nuclear hormone receptor (NHR)-8 should be seriously considered as a target because it upregulates multiple PGPs involved in anthelmintic resistance and it is specific to nematodes. Background: Iran is one of the endemic areas of Dirofilaria immitis. The modified Knott test is the common diagnostic test based on detecting and identifying microfilaria in blood samples. Still, the results may be compromised by D. immitis occult infection (D. immitis infection without microfilariae). Wolbachia pipientis in a dog's blood is an endosymbiont of D. immitis and an indicator of dirofilariasis. Objectives: This study aimed to investigate the occult infection of D. immitis in stray dogs in Guilan, Qazvin, and Mazandaran provinces, Iran, based on W. pipientis DNA tracking. Methods: Blood samples (n=138) were collected, and the presence of W. pipientis was evaluated by amplifying a partial sequence of the FtsZ gene (267 bp). Results: The results showed that 17.4% of the samples were positive by the modified Knott method, while 47.8% were positive by the molecular methods. The results of high-resolution melting (HRM) real-time PCR showed that the melting temperature of cox1 gene amplicons of D. immitis and Acanthocheilonema reconditum were obtained at 76.67±0.03 °C and 81.38±0.05 °C, respectively as well as it was achieved 80.30±0.04 °C for FtsZ gene of W. pipientis. The results of the present study showed that it is necessary to use molecular methods to diagnose dirofilariasis occult infection accurately. Conclusion: The HRM real-time PCR analysis is a simple post-PCR step that exploits the thermal characteristics of the amplicons to detect the occult infection of dirofilariasis based on W. pipientis DNA tracking and identification. Plant parasitic nematodes (PPNs), such as Meloidogyne spp., Heterodera spp. and Pratylenchus spp., are obligate parasites on a wide range of crops, causing significant agricultural production losses worldwide. These PPNs mainly feed on and within roots, impairing both the below-ground and the above-ground parts, resulting in reduced plant performance. Plants have developed a multi-component defence mechanism against diverse pathogens, including PPNs. Several natural molecules, ranging from cell wall components to secondary metabolites, have been found to protect plants from PPN attack by conferring nematode-specific resistance. Recent advances in omics analytical tools have encouraged researchers to shed light on nematode detection and the biochemical defence mechanisms of plants during nematode infection. Here, we discuss the recent progress on revealing the nematode-associated molecular patterns (NAMPs) and their receptors in plants. The biochemical defence responses of plants, comprising cell wall reinforcement; reactive oxygen species burst; receptor-like cytoplasmic kinases; mitogen-activated protein kinases; antioxidant activities; phytohormone biosynthesis and signalling; transcription factor activation; and the production of anti-PPN phytochemicals are also described. Finally, we also examine the role of epigenetics in regulating the transcriptional response to nematode attack. In order to know whether there is a risk of anisakiasis (or anisakidosis) by consumption of fish of the genus Mullus from the western Mediterranean Sea, which are appreciated for their quality, an epidemiological survey was carried out to evaluate the occurrence of zoonotic or potentially zoonotic nematodes in M. barbatus and M. surmuletus. Although the presence of the third larval stage (L3) of anisakids (Anisakis and Contracaecum) has been previously described in these fish, the results showed the absence of anisakids and the presence, never in muscle, of L3 and L4 of raphidascaridids of the genus Hysterothylacium, molecularly identified as H. fabri. Phylogenetic analysis groups them into the Mediterranean Sea clade, far from individuals isolated in the Pacific Ocean. Prevalence was slightly higher, but not significant, in M. barbatus versus M. surmuletus (72.3% vs 60.0%), but mean intensity (MI) and mean abundance (MA) parameters were approximately twice as high in M. barbatus as in M. surmuletus (MI 5.8 vs 2.8, p =.001; MA 4.2 vs 1.7, p <.001). The presence of the parasite seems to have different effects on these two sympatric species. In M. barbatus it seems to affect their growth, as it appreciably reduces the value of allometry coefficient in infected fish (2.78 vs. 2.18). On the other hand, in M. surmuletus the infection significantly (p <.04) affects the Fulton's condition factor, an indicator of the health status of the fish. It can be concluded that the ingestion of these fish by the people poses negligible risk of anisakiasis, but the consumer should continue to be urged to follow the rules of prevention against this illness. This study investigates the carbohydrate content of Schwenkiella orientalis, a nematode parasite infecting Periplaneta americana (American cockroach), focusing on its total sugar and glycogen levels. The study conducted in Bulandshahr (U.P.) and it employs the Anthrone method to quantitatively analyze carbohydrate reserves within the parasite. The findings reveal that the total sugar content increases from 42 µg/mg in 5 mg parasites to 85 µg/mg in 20 mg parasites, while glycogen levels rise from 18 µg/mg to 62 µg/mg for the same weight range. This indicates that larger parasites possess greater energy reserves. The data suggest that Schwenkiella orientalis efficiently absorbs carbohydrates from its host for immediate metabolic needs while utilizing glycogen as a crucial energy reserve during nutrient-scarce periods. This research underscores the significance of carbohydrate metabolism in parasitic nematodes, highlighting the adaptive strategies that enable their survival within host environments. This study investigates the biochemical complexity of Hammerschmidtiella diesingi, a parasitic nematode inhabiting the alimentary canal of Periplaneta americana (American cockroach). The protein composition was analyzed using Sodium Dodecyl Sulfate-Polyacrylamide Gel Electrophoresis (SDS-PAGE), revealing eight distinct protein bands ranging from 10 to 75 kDa. These bands represent a variety of proteins involved in critical physiological functions, including nutrient absorption, immune evasion and reproduction, enabling the nematode to survive in the host's digestive environment. The 75 kDa band likely corresponds to structural or enzymatic proteins, while the smaller bands (10-25 kDa) may be enzymes involved in digestive processes. The consistency in protein profiles across different sample weights indicates stable protein expression, which is crucial for the nematode's parasitic strategy. The study highlights the evolutionary adaptations of H. diesingi that allow it to grow well in a hostile environment. Future research should focus on functional assays to characterize these proteins and explore genetic mechanisms underlying their expression, which could lead to the development of novel control strategies for parasitic nematode infections in pest species. Cyathostomins are common digestive tract parasites of grazing horses that spread through contact with horse feces. Horse feces are colonized by a variety of organisms, some of which could serve to reduce parasite loads in horse pastures. Black soldier fly (Hermetia illucens L.; Diptera: Stratiomyidae) larvae (BSFL) could be an ideal candidate for biological control of cyathostomins, due to their near-global distribution, low risk of pathogen transmission, ability to develop on a variety of nutrient-poor substrates (including horse manure), and dramatic effect on microbial communities that cyathostomins depend on. Here, using controlled feeding bioassays, we evaluated the effect of BSFL on cyathostomin egg densities in horse manure while also tracking BSFL performance on manure relative to standard grain-based diets. We found that BSFL consumed less substrate, were slower to reach the prepupal stage, and ultimately yielded less biomass when reared on horse manure compared to grain-based diets. However, BSFL reduced average cyathostomin egg densities in horse manure by over 3-fold. Overall, our results suggest that despite horse manure being a poor substrate for BSFL growth, BSFL effectively reduce cyathostomin egg loads in infected horse manure, though the mechanisms by which they do this are uncertain. While BSFL are known to transform the microbial communities within a diversity of rearing substrates, their effect on larger, parasitic organisms in animal manures may be underappreciated. Kiwifruit, Actinidia spp., is an important crop worldwide and its cultivation has been increasing in Portugal. Kiwifruit plants are susceptible to plant‐parasitic nematodes (PPNs), including Meloidogyne spp. M. hapla and M. incognita have been reported to affect kiwifruit in major production regions worldwide, although information is lacking on the distribution and identity of PPNs associated with kiwifruit orchards in Portugal. Moreover, research is scarce on the drivers of PPN communities in this crop. Our primary objective was to investigate the diversity, density and distribution of PPNs within kiwifruit orchards of Portugal and to elucidate the influence of environmental factors on their community composition. Additionally, we aimed to evaluate population densities and identity of Meloidogyne spp. Nematodes were extracted from composite soil and root samples collected from orchards. Analysis of soil nematode communities suggested bacterial‐driven decomposition and high nutrient enrichment in these soils. Sixteen different genera of PPNs were detected, with Meloidogyne spp. as the dominant genus, mostly represented by a single species, M. hapla. PPN communities exhibited low richness, and β‐diversity analysis indicated a high degree of similarity among them. Additionally, their composition was significantly influenced by soil and climatic parameters. We found that 37 out of 40 kiwifruit orchards in Portugal were infested by root‐knot nematodes. The unexpectedly low species diversity, dominated by M. hapla, merits further research to establish underlying factors for its high incidence in this crop and to assess the impact of these nematodes on kiwifruit performance and productivity. Microbial and microeukaryotic communities are extremely abundant and diverse in soil habitats where they play critical roles in ecosystem functioning and services that are essential to soil health. Soil biodiversity is influenced by above‐ground (vegetation) and below‐ground factors (soil properties), which together create habitat‐specific conditions. However, the compound effects of vegetation and soil properties on soil communities are less studied or often focused on one component of the soil biota. Here, we integrate metabarcoding (16S and 18S rRNA genes) and nematode morphology to assess the effects of habitat and soil properties shaping microbial and microeukaryotic communities as well as nematode‐associated microbiomes. We show that both vegetation and soil properties (soil bulk density) were major factors structuring microbial and microeukaryotic communities in semi‐arid soil habitats. Despite having lower nutrients and lower pH, denser soils displayed significantly higher alpha diversity than less dense soils across datasets. Nematode‐associated microbiomes have lower microbial diversity, strongly differ from soil microbes and are more likely to respond to microscale variations among samples than to vegetation or soil bulk density. Consequently, different nematode lineages and trophic groups are likely to display similar associated microbiomes when sharing the same microhabitat. Different microbiome taxa were enriched within specific nematode lineages (e.g. Mycobacterium, Candidatus Cardinium) highlighting potentially new species‐specific associations that may confer benefits to their soil nematode hosts. Our findings highlight the importance of exploring above‐ and below‐ground effects to assess community structure in terrestrial habitats, and how fine‐scale analyses are critical for understanding patterns of host‐associated microbiomes. Rationale: Matrix‐assisted laser desorption/ionisation‐mass spectrometry imaging (MALDI‐MSI) is a powerful label‐free technique for biomolecule detection (e.g., lipids), within tissue sections across various biological species. However, despite its utility in many applications, the nematode Caenorhabditis elegans is not routinely used in combination with MALDI‐MSI. The lack of studies exploring spatial distribution of biomolecules in nematodes is likely due to challenges with sample preparation. Methods: This study developed a sample preparation method for whole intact nematodes, evaluated using cryosectioning of nematodes embedded in a 10% gelatine solution to obtain longitudinal cross sections. The slices were then subjected to MALDI‐MSI, using a RapifleX Tissuetyper in positive and negative polarities. Samples were also prepared for liquid chromatography‐tandem mass spectrometry (LC‐MS/MS) analysis using an Exploris 480 coupled to a HPLC Vanquish system to confirm the MALDI‐MSI results. Results: An optimised embedding method was developed for longitudinal cross‐sectioning of individual worms. To obtain longitudinal cross sections, nematodes were frozen at −80°C so that all worms were rod shaped. Then, the samples were defrosted and transferred to a 10% gelatine matrix in a cryomold; the worms aligned, and the whole cryomold submerged in liquid nitrogen. Using MALDI‐MSI, we were able to observe the distribution of lipids within C. elegans, with clear differences in their spatial distribution at a resolution of 5 μm. To confirm the lipids from MALDI‐MSI, age‐matched nematodes were subjected to LC‐MS/MS. Here, 520 lipids were identified using LC‐MS/MS, indicating overlap with MALDI‐MSI data. Conclusions: This optimised sample preparation technique enabled (un)targeted analysis of spatially distributed lipids within individual nematodes. Miscanthus × giganteus biomass plays a crucial role in producing renewable energy and bio-based products, supporting global sustainability objectives. However, its introduction into the European Union has made it susceptible to the ectoparasitic needle nematode Longidorus spp., which are known vectors of severe viral diseases. The aim of the presented research was to assess the effectiveness of the following soil amendments: vermicompost from Eisenia fetida and silver nanoparticles (Ag-NPs) applied to the soil with Miscanthus plants following artificial inoculation of Longidorus elongatus. A two-year experiment was conducted at the National Institute of Horticulture Research in Skierniewice using concrete rings filled with medium sandy soil amended with 10% peat. Treatments included: control (no amendments), vermicompost (4 L of E. fetida vermicompost), and Ag-NPs (60 mg/L soil). Each treatment was replicated four times. Application of both vermicompost and Ag-NPs positively influenced soil parameters and crop yield while suppressing nematode populations. Drug-resistant parasitic nematodes pose a grave threat to plants, animals, and humans. An innovative paradigm for treating parasitic nematodes is emphasized in this opinion. This approach relies on repurposing methuosis (a death characterized by accumulation of large vacuoles) inducing anticancer drugs as anthelmintics. We review drugs/chemicals that have shown to kill nematodes or cancerous cells by inducing multiple vacuoles that eventually coalesce and rupture. This perspective additionally offers a succinct summary on Structure–Activity Relationship (SAR) of methuosis-inducing small molecules. This strategy holds promise for the development of broad-spectrum anthelmintics, shedding light on shared molecular mechanisms between cancer and nematodes in response to these inducers, thereby potentially transforming both therapeutic domains. Forty-seven faecal samples of wolves from Bejaia were examined for intestinal helminths. Five nematodes (Ancylostoma caninum (egg), Strongyloides stercoralis (larvae L1), Toxocara canis (egg), Toxascaris leonina (egg), and Trichuris vulpis (egg)), and one cestode (Dipylidium caninum) were identified. These nematodes were detected with a prevalence of 46.8%, 23.4%, 38.2%, 25.3%, and 2.12%, respectively; and a cestode, with a prevalence of 4.3%. Our results show a high relative abundance of Strongyloides stercoralis larvae L1 (37.5%), followed by eggs of Ancylostoma caninum, whether non-embryonated (16.77%) or embryonated (14.28%). Among the ascarids, Toxocara canis was represented with 18.1%, followed by Toxascaris leonina (8.1%). Finally, Dipylidium caninum and Trichuris vulpis were only poorly represented. This is the first study on helminth parasites in wild golden wolves in Bejaia. The results indicate the presence of six gastrointestinal helminths in the wolf population. [ABSTRACT FROM AUTHOR]

The root-knot nematodes (Meloidogyne spp.) are one of the most dangerous and widespread species of nematodes affecting tomatoes. There are few methods for controlling nematodes in tomatoes. Natural resistance is important in conferring resistance against nematodes. A study was conducted to evaluate the reaction of tomato accessions to root-knot nematode Meloidogyne arenaria (Neal) Chitwood. Twenty one tomato accessions were subjected for screening. Susceptible tomato variety Ideal was used as control. The tomato plants were evaluated 60 days after inoculation on the basis of the gall index (GI), egg mass index (EMI), final populations (Pf) and reproduction factors (Rf). All the tomato accessions show varying degree of response. Most of the screened accessions were susceptible to M. arenaria, six were resistant and two accessions showed hypersensitive reaction. Nematode surveys in natural environments in the Iberian Peninsula detected three unidentified Xenocriconemella populations that closely resembled the X. macrodora-species complex, but utilization of integrative taxonomy confirmed that they comprised a new taxon described in this paper as X. andreae sp. nov. Only females were detected in the new species (considered parthenogenetic) and delineated with a bare body (274–353 µm); lip region with two annuli, continuous with body delineation; second lip annulus enclosed by the first one. Flexible and thin stylet (88.0–99.0 µm), representing 30.4–47.8% of total body length. The excretory pore is positioned 2–3 annuli posterior to the level of stylet knobs, at 101.5 (87–107) µm from the lip region. Female genital tract: monodelphic, prodelphic, large, and representing 34.4–52.4% of the body length; vagina slightly ventrally curved. The anus is located at (6–9) annuli from the rear end. Tail short, conoid, and blunt round terminus. Ribosomal and mitochondrial markers (D2-D3 expansion domains of 28S, ITS , partial 18S rRNA, and COI), as well as molecular phylogenetic analyses of sequences, confirmed this new taxon, and it was clearly delineated from X. macrodora and species within the species complex (X. costaricense, X. iberica, X. paraiberica, and X. pradense). A new belondirid, dorylaimid taxon, Oleaxonchium olearum gen. et sp. nov., collected from an olive grove in the Andalusia region of Spain, is characterised, including its morphological description, morphometrics, SEM study, and molecular (18S-, 28S-rDNA) analyses. The new genus displays a unique combination of traits that distinguishes it from its closest genera: a rectangular lip region with sclerotized margins when observed in lateral view and visibly hexagonal in face view under SEM; a comparatively long cheilostom with thickened walls at its anterior part; a short isthmus-like section separating both pharyngeal regions; a mono-opistho-ovarian didelphic female genital system without pars refringens vaginae; and a short and rounded tail. The new species is characterised by its 2.44–2.87 mm long body, lip region 7–7.5 µm wide, odontostyle 10–10.5 µm long, neck 723–973 µm long, pharyngeal expansion occupying 63–72% of the total neck length, female anterior genital branch 4–6% of body length, tripartite posterior uterus 1.9–2.6 body diameters long, with a short intermediate section bearing sclerotized elements, vulva (V = 58–61) a transverse slit, caudal region 29–35 µm long (c = 74–89, c' = 0.9–1.1), and male unknown. As derived from an integrative approach combining morphological and molecular data, the new genus is close to Metaxonchium, the polyphyly of Belondiridae is confirmed, and support is provided in favour of the monophyly of Axonchiinae. Elasmobranch populations are in steep decline mainly due to overfishing bycatch, but parasites may accelerate the collapse of vulnerable and/or highly parasitized species. We therefore studied metazoan parasites of Rajidae from the northeast Atlantic: vulnerable Leucoraja fullonica, near‐threatened Raja brachyura, Raja clavata, Raja microocellata and Raja undulata, and least‐concerned Raja montagui and Leucoraja naevus. Overall prevalence varied from 19% for R. montagui to 100% for L. fullonica. Parasite communities differed between skate species, and prevalence and abundance were higher for L. fullonica, R. microocellata, and R. undulata. We recorded 11 parasite taxa in the study: three nematodes, six cestodes, one monogenean, and one myxosporean. Whatever the skate species, the parasite component community comprised at least two nematode taxa among Phocanema spp., Proleptus sp. and Anisakis simplex. DNA‐sequencing revealed that Phocanema azarasi and Phocanema krabbei both occurred in R. microocellata and R. undulata. Phocanema spp. was first recorded in L. fullonica, L. naevus, R. microocellata, R. montagui, and R. undulata, as Proleptus sp. in L. fullonica, and A. simplex in L. fullonica and R. clavata, Rockacestus sp. and Nybelinia sp. in R. undulata, and gill‐myxosporeans on L. fullonica, L. naevus, R. microocellata, and R. undulata. The occurrence of 16 new host–parasite associations suggests potential environmental changes. Information provided by trophically transmitted helminths confirmed an opportunistic skate diet based on crustaceans and fish. This study aimed to identify parasitic species infesting marine fish collected from Lake Timsah across different seasons. A total of 480 marine fish of varying species, weights, and lengths (160 Dicentrarchus labrax, 160 Dicentrarchus punctatus, and 160 Sparus aurata) were randomly collected seasonally from September 2020 to August 2021. Most examined fish showed no pathognomonic clinical abnormalities and appeared normal. However, some D. labrax exhibited hemorrhages on the mouth, operculum, around the eyes, and at the bases of their fins, along with abdominal distension and emaciation. Infested D. punctatus showed excessive mucus and a mosaic appearance of the gills. Infested S. aurata displayed no external clinical abnormalities, except for some hemorrhagic lesions and slight abdominal distension. The total prevalence of parasitic infestation among the examined fish was 57.91%. The highest infestation rate was observed in D. punctatus (74.38%), followed by D. labrax (71.25%) and S. aurata (28.13%). The isolated parasites included nematodes (Hysterothylacium aduncum, Anisakis simplex larvae, and Contracaecum spp. larvae), digeneans (Erilepturus hamate and Tangiopsis chinensis), crustacean parasites (Lernanathropus kroyeri, Caligus minimus), and isopods (Livoneca redmanii). The total parasitic infestation rate varied significantly across different seasons (P<0.001), with the highest rate in winter (81.6%), followed by spring (70.8%), autumn (55%), and summer (24%). The nematode Caenorhabditis elegans is well known for its ability to support forward genetic screens to identify molecules involved in neuronal viability and signaling. The proteins involved in C. elegans dopamine (DA) regulation are highly conserved across evolution, with prior work demonstrating that the model can serve as an efficient platform to identify novel genes involved in disease‐associated processes. To identify novel players in DA signaling, we took advantage of a recently developed library of pre‐sequenced mutant nematodes arising from the million mutation project (MMP) to identify strains that display the DA‐dependent swimming‐induced‐paralysis phenotype (Swip). Our screen identified novel mutations in the dopamine transporter encoding gene dat‐1, whose loss was previously used to identify the Swip phenotype, as well as multiple genes with previously unknown connections to DA signaling. Here, we present our isolation and characterization of one of these genes, bbs‐1, previously linked to the function of primary cilia in worms and higher organisms, including humans, and where loss‐of‐function mutations result in a human disorder known as Bardet–Biedl syndrome. Our studies of C. elegans BBS‐1 protein, as well as other proteins that are known to be assembled into a higher order complex (the BBSome) reveal that functional or structural disruption of this complex leads to exaggerated C. elegans DA signaling to produce Swip via a cell‐autonomous mechanism. We provide evidence that not only does the proper function of cilia in C. elegans DA neurons support normal swimming behavior, but also that bbs‐1 maintains normal levels of DAT‐1 trafficking or function via a RHO‐1 and SWIP‐13/MAPK‐15 dependent pathway where mutants may contribute to Swip independent of altered ciliary function. Together, these studies demonstrate novel contributors to DA neuron function in the worm and demonstrate the utility and efficiency of forward genetic screens using the MMP library. The predominant plant-parasitic nematodes associated with Pattawia pineapple-cultivated areas were investigated using combined molecular and morphological techniques. In total, 134 soil samples were collected for nematode extraction from the main pineapple crops in 3 districts (Sam Roi Yot, Mueang and Hua Hin) of Prachuap Khiri Khan, Thailand. Based on the morphology, 6 plant-parasitic nematode genera were identified: Rotylenchulus sp., Meloidogyne sp., Helicotylenchus sp., Hoplolaimus sp., Mesocriconema sp. and Pratylenchus sp. The main plant-parasitic nematodes that occurred in Sam Roi Yot and Mueang districts were Rotylenchulus sp. (70.27 and 97.01 %, respectively), while in Hua Hin district, Helicotylenchus sp. (37.73 %) was dominant. Based on molecular identification, nucleotide sequence comparisons and the phylogenetic tree analyses based on the 18S and 28S rRNA genes revealed the species of the nematodes were R. reniformis and H. dihystera, showing 99 % identity with the nematode populations from Japan and China, respectively. Notably, the morphometric and morphological characters also identified these 2 species correctly; however, this technique required more time and may be less accurate. While R. reniformis has been identified as a common species in pineapple fields in the USA, India, Kenya, the Philippines and Nigeria, the current study was the 1st to report H. dihystera isolated from soil in Pattawia pineapple crops in Thailand. Helminths have developed intricate mechanisms to survive and evade the host's immune responses. Hence, understanding the excretory-secretory products (ESPs) by helminths is crucial for developing control tools, including drug targets, vaccines, and potential therapies for inflammatory and metabolic disorders caused by them. Proteomics, the large-scale analysis of proteins, offers a powerful approach to unravel the complex proteomes of helminths and gain insights into their biology. Proteomics, as a science that delves into the functions of proteins, has the potential to revolutionize clinical therapies against parasitic infections that have developed anthelminthic resistance. Proteomic technologies lay a framework for accompanying genomic, reverse genetics, and pharmacokinetic approaches to provide more profound or broader coverage of the cellular mechanisms that underlie the response to anthelmintics. With the development of vaccines against helminth infections, proteomics has brought a major change to parasitology. The proteome of helminths can be analyzed comprehensively, revealing the complex network of proteins that enable parasite survival and pathogenicity. Furthermore, it reveals how parasites interact with hosts' immune systems. Plants resist the invasion of plant-parasitic nematodes (PPNs) by releasing root chemical substances, activating immune responses, and utilizing plant resistance (R) genes. A balance of interaction is formed between plants and PPNs, including multilayer communications and signal exchanges. The signal network involved in plant–nematode competition includes attraction and repellency of root exudates, perception of nematode-associated signals, activation of plant signals, interference of PPNs with plant defense, and interplay of plant systemic signals. Plant-parasitic nematodes (PPNs) are widely distributed and highly adaptable. To evade the invasion and infection of PPNs, plants initiate a series of defense responses. In turn, PPNs secrete effectors into the host tissues to suppress plant defense. In this ongoing battle between PPNs and plants, complex signal transduction processes are typically involved. This article aims to review the plant signaling network involved in host perception by the nematode, nematode perception, and downstream activation of plant defense signaling and how nematodes attempt to interfere with this network. Our goal is to establish a foundation for elucidating the signaling and regulatory mechanisms of plant–nematode interactions, and to provide insights and tools for developing PPN-resistant crops and technologies. Infections with gastrointestinal nematodes on pasture have negative impact on animal health and production. Anthelmintics are used for deworming to prevent an excessive increase in parasite load and to treat parasitosis. Due to the progressive resistance of gastrointestinal nematodes to anthelmintics, their effectiveness is increasingly reduced. For this reason, alternatives are needed to combat infections with gastrointestinal nematodes. A sustainable strategy with great potential is the breeding of sheep with increased immunocompetence against gastrointestinal nematodes. Various indicators of gastrointestinal nematode infection can and should be used as selection traits for this purpose. Given the multifactorial background of infection events, the interpretation of the data and the definition of adequate breeding goals are particularly challenging. This review article deals with established and putative selection traits for breeding sheep with improved immunocompetence regarding gastrointestinal nematodes. With reference to the current state of sheep breeding in Germany, the challenges, especially with regard to the definition of breeding goals and phenotyping, are identified. Nematodes of the Onchocercidae family, such as Pelecitus spp., are filarial parasites of medical and veterinary importance. Although infections are widely distributed among avian species, only 2 cases of human Pelecitus ocular infection, both in South America, have been reported. We describe a 61-year-old man in northeast Thailand diagnosed with an ocular infection. Morphologic characteristics suggested the causative agent was a female Pelecitus nematode: coiled body, rounded anterior and posterior extremities, a distinct preesophageal cuticular ring, lateral alae, a postdeirid, and a protuberant vulva. Sequences of the 12S rDNA gene indicated 95%–96% identity and cox1 gene 92%–96% identity with published P. copsychi sequences. P-distance for cox1 sequences between the causative agent and P. copsychi was 6.71%. Phylogenetic trees of 12S rDNA and cox1 genes indicated the species differed from but is closely associated with P. copsychi. Nematode, cestode, protozoan, microsporidian, and pentastomid parasites affect domesticated and wild rabbits, hares, and jackrabbits of the genera Brachylagus, Lepus, Oryctolagus, Pentalagus, and Sylvilagus. Some endoparasite infections are of limited or no significance, whereas others have potentially profound consequences. Accurate identification of endoparasites of rabbits, hares, and jackrabbits is an important facet of the work of veterinary pathologists engaged in lagomorph pathology. Here I review endoparasites from the pathologist's perspective, focusing on pathogenesis, lesions, and implications of infection. Stomach nematodes Graphidium strigosum and Obeliscoides cuniculi are infrequently pathogenic but may cause gastritis and gastric mucosal thickening. Nematodes Passalurus ambiguus, Protostrongylus spp., Trichostrongylus spp., and Trichuris spp. are rarely associated with disease. Adult Capillaria hepatica (syn. Calodium hepaticum) nematodes and non-embryonated eggs cause granulomatous hepatitis in wild Oryctolagus cuniculus and Lepus europaeus, resulting in multifocal, off-white, hepatic lesions, which may be misdiagnosed as hepatic eimeriosis. When the rabbit is an intermediate host for carnivore cestodes, the space-occupying effects of Cysticercus pisiformis and Coenurus serialis may have pathologic consequences. Eimeria stiedai is a major cause of white-spotted liver in O. cuniculus, particularly in juveniles. Enteric coccidiosis is a noteworthy cause of unthriftiness in young animals, and frequently manifests as diarrhea with grossly appreciable multifocal off-white intestinal lesions. O. cuniculus is the natural host for the zoonotic microsporidian Encephalitozoon cuniculi. Infection may be acute and focused mainly on the kidneys, or it may follow a chronic disease course, frequently with neurologic lesions. A latent carrier status may also develop. The bear roundworm Baylisascaris transfuga has been identified in several host bears (Ursinae). However, limited genetic information is available on the bear roundworm in Japanese populations. This study evaluated the genetic composition of bear roundworms isolated from wild Japanese black bears indigenous to Lake Towada, Japan. First, we conducted genetic and/or molecular phylogenetic analyses based on cytochrome c oxidase subunit II and internal transcribed spacer 2 among Baylisascaris species. These analyses revealed that the identified roundworms were genetically B. transfuga. In addition, the average body size of the obtained roundworms in this study was almost the same as that previously reported for B. transfuga. New species of the genera Spirobolbolaimus and Ixonema (Nematoda: Microlaimidae) have been found in sediment samples collected in the South Atlantic, along the Continental Shelf break off Northeastern Brazil. Different to other Spirobolbolaimus species, S. pernambucanussp. nov. possesses six outer labial setae and four cephalic setae with approximately the same length. Ixonema gracieleaesp. nov. differs from other species of Ixonema in having somatic setae on peduncles. This is the first time that new species of these taxa have been described for the Brazilian coast. An amendment of the diagnosis and a dichotomous key are proposed for both genera. Patagonian maras, rodents endemic to South America, are classified as a near-threatened species. Various factors affect their health including parasitic diseases. The aim of this study was to perform morphometric, molecular and phylogenetic characterisation of one such parasitic disease agent, the nematode Graphidioides affinis, specimens of which were found in captive Patagonian maras. In March 2023, 18 Patagonian maras kept at the Sofia Zoo in Bulgaria were investigated with the use of coprological methods. Following the investigation, the animals were dewormed with the use of albendazole. Dead adult nematodes found in the faeces of dewormed maras were collected and preserved in 70% ethanol, and morphometrically, molecularly and phylogenetically analysed. The morphometric analyses confirmed the nematodes to be Graphidioides affinis. The partial nucleotide sequences of the small subunit ribosomal rDNA (SSU), the internal transcribe spacer 2 (ITS2) and the large subunit ribosomal DNA (LSU) of G. affinis were obtained. These are the first available nucleotide sequences of this parasite. The phylogenetic analyses of the species showed its distinctiveness in comparison to other gastrointestinal nematodes, as it was grouped separately. The Cephalobidae (Free-living nematodes) family are bacterivores that inhabit soil. Bacterial-feeding nematodes are one of the main consumers of soil bacteria. Therefore, study on this group of nematodes is essential. This molecular study was conducted in 2024 at Limpopo University to identify the free-living bacterivores nematodes from South Africa's soils, using a 28S rDNA marker. The recovered nematode was extracted using the tray method, and then its DNA was extracted using the Chelex method. The nematode was identified as Zeldia punctata. Afterwards, 28S rDNA was amplified using specific primers to identify the nematode. The Nblast analysis based on the 28S rDNA showed South African Z. punctata has 98% similarity (DQ145662) with the American population. Phylogenetic analysis using maximum likelihood placed this species with those molecularly identified as Z. punctata in the same clade with highly supported (94%) bootstrap values. In conclusion, this species was properly identified using 28S rDNA. However, other rDNA markers are recommended to better understand Zeldia phylogeny. [ABSTRACT FROM AUTHOR]
