Cowpea planting season is crucial for high yield and should comprise the period of the year that warrants the best climatic conditions for cowpea cultivation. Thus, the objective of this study was to evaluate the influence of water availability and temperature on the performance of cowpea cultivars. A greenhouse experiment was conducted using a 4 × 2 × 5 factorial arrangement, with four replications. Factors included four levels of soil moisture (25, 50, 75, and 100% of water holding capacity), two growing seasons (mild and hot), and five cowpea cultivars (Carijó, Itaim, Pujante, Rouxinol, and Tapahium). The number of pods and seeds per plant, seed production, water use efficiency, shoot dry mass, root dry mass, and physiological parameters were evaluated. Seed production was higher during the mild season than during the hot season and increased linearly with increasing soil water availability. Photosynthetic activity and transpiration were higher during the hot season than during the mild season, with their reduction under a water availability of 25% regardless of the growing season. Total chlorophyll content decreased with excess water. Regardless of water availability, temperature was the most limiting climatic factor for cowpea performance. Triploid watermelon is highly appreciated by the most demanding markets, and due to its small size, it is ideal for consumption by small families. With the growth in areas cultivated with seedless watermelon worldwide, there is a demand for the development of tetraploid germplasm to obtain triploid hybrids with better agronomic performance. This study performed two tests to induce polyploidy in the powdery mildew–resistant line developed by Embrapa Semi-Arid, LDRO, under different colchicine concentrations and application methods. In Experiment 1, the seeds were treated with colchicine (0.0%, 0.1%, and 0.2%) for 24 h and 48 h. In Experiment 2, 0.2% colchicine was applied by different methods: (a) directly on the seed (MDS) with and without scarification, (b) on seeds with radicle emission (MER), (c) at the insertion point between the hypocotyl and the root (MIHR), (d) at the seedling apex (MAP), and (e) in the inverted hypocotyl (MHI). Chromosome count (cytogenetic analysis), number of chloroplasts per pair of guard cells, number of stomata, seedling height, and hypocotyl diameter were measured. In the LDRO line, chromosomal duplication occurred in some plant cells, but it was not possible to obtain 100% tetraploid plants. Colchicine (0.2%) for 48 h without mechanical scarification induced chromosomal duplication in watermelon. The analysis of the number of chloroplasts identified the level of ploidy early, reducing the number of plants needed to be evaluated by cytogenetics, which allowed us to more accurately identify the different levels of ploidy of the plant. RESUMO A melancia triploide é muito apreciada pelos mercados mais exigentes, e, devido ao seu tamanho reduzido, torna-se ideal para consumo em famílias pequenas. Com o incremento de áreas cultivadas com melancia sem sementes no mundo, há demanda pelo desenvolvimento de germoplasma tetraploides para obtenção de híbridos triploides de melhor desempenho agronômico. Foram realizados dois ensaios com o objetivo de induzir poliploidia na Linhagem resistente ao oídio desenvolvida pela Embrapa Semiárido, LDRO, sob diferentes concentrações de colchicina e métodos de aplicação. As sementes foram tratadas com colchicina (0,0%; 0,1% e 0,2%) por 24h e 48h (Experimento 1) e sob diferentes métodos de aplicação de colchicina a 0,2% a) direto na semente (MDS) com e sem escarificação, b) sementes com emissão da radícula (MER), c) no ponto de inserção entre o hipocótilo e a raiz (MIHR), d) no ápice da plântula (MAP) e e) no hipocótilo invertido (MHI) (Experimento 2). Foi realizada a análise citogenética (contagem do número cromossômico), do número de cloroplastos por pares de células-guardas, número de estômatos, altura da plântula e diâmetro do hipocótilo. Na Linhagem LDRO, a duplicação cromossômica ocorreu em algumas células das plantas, mas não foi possível obter plantas 100% tetraploides. A colchicina a 0,2% por 48h sem escarificação mecânica induz a duplicação cromossômica em melancia. A análise do número de cloroplastos identifica os níveis de ploidia, de forma precoce, reduzindo o número de plantas, que serão avaliadas em citogenética, sendo que esta permite identificar com maior precisão os diferentes níveis de ploidia da planta.

Water deficit and high temperature limit agricultural productivity, affecting plant growth, development, and metabolism. Thus, the objective of this study was to evaluate the impact of the interaction between water deficit and temperature increase on cowpea production. The experiment was carried out with the cultivar BRS Rouxinol, in growth chambers, in a 4 × 2 factorial arrangement, being four soil water availability (25; 50; 75 and 100% of field capacity) and two daily temperature regimes (T°1: 20-26-33 °C and T°2: 24.8-30.8-37.8 °C). The increase in water availability favored an increase in shoot and root dry mass. The vegetative stage was prolonged in plants maintained at a temperature regime of 24.8-30.8-37.8 °C. The cowpea yield increased with higher water availability. The water availability of 90% was the one that provided the highest water use efficiency. The 4.8°C increase in air temperature promotes an increase in oxidative stress with lack of synchrony in the antioxidative defense system. The cultivar Rouxinol is sensitive to the increase in air temperature and water deficit. However, even with 100% water availability, there was no seed production with an increase in air temperature. Temperature is one of the climatic elements that affect most the germination process and seedling development. Thus, the objective of this study was to evaluate the germination and seedling development of cowpea cultivars under increasing temperature. The seeds of the cultivars Acauã, Carijó, Guariba, Gurguéia, Itaim, Juruá, Pajeú, Potengi, Pujante, Rouxinol, Tapahium, and Tumucumaque were kept in germination chamber at temperatures of 20, 25, 30, 35 and 40°C, with photoperiod of 12 h. Cowpea seeds have optimal germination performance and seedling development at temperatures in the range of 30.49 - 35.48°C. The cultivars presented germination percentage above 91% under 40°C, however, the seedling were abnormal. The temperature of 20°C provided germination above 96%, but with a longer period to germinate. The temperature range between 30 - 35°C favored the germination speed index, average time, and seed germination speed, with optimum temperatures varying between the cultivars. In micropropagation, potassium nitrate (KNO3), an ACS reagent grade chemical, used in the preparation of growing mediums is expensive and its procurement depends on bureaucratic procedures, as it is controlled by the Brazilian Army. This research to assessed the effect of replacing the ACS KNO3 for a commercially available fertilizer (KNO3- based) on the micropropagation of the prickly pear cactus (Opuntia stricta (Haw.) Haw. cv. Elephant Ear. Treatments used six different fertilizer concentrations (0, 0.5, 1, 1.5, 2 and 2.5 g L-1) and a control consisting of 1.9 g L-1 KNO3 , as shown in the MS salts. The survival, size and number of sprouts and the value of fresh biomass were evaluated. After seedling acclimation, we assessed the survival, number of sprouts, length, and number of roots, racket formation, average fresh biomass mass, macronutrient absorption and morphological changes of the seedlings. Explants inoculated with fertilizers at concentrations of 0.0; 2.0 and 2.5 g L-1 did not grow. The response of explants at concentrations of 0.5 and 1.5 g L-1 of the fertilizer were the same as those developed in a KNO3 medium, and at a concentration of 1.0 g L-1, in all variables, the means were higher than those of the control medium. Therefore, it showed the feasibility of using fertilizers in the in vitro cultivation of the prickly pear cactus, which may remove bureaucratic barriers and reduce product costs by 99.12%. Adenia is an Old World genus of Passifloroideae closely related to Passiflora. The two genera comprise the large majority of Passifloroideae species, although most studies are concentrated on Passiflora. Cytological analyses reveal that changes in chromosome numbers played an important role in the evolution of Passiflora, whereas in the remaining genera little is known, hindering the identification of the base number of the family. Here we analyzed the chromosome number and the 35S rDNA sites of three species of Adenia and reevaluated the base number (x) of the subfamily Passifloroideae and the family Passifloraceae, including chromosome data for Turneroideae and Malesherbioideae. The chromosome number of Adenia species seemed to be stable with 2n = 24 or 48 and one or two pairs of rDNA sites, very similar to Passiflora subgenus Astrophea, suggesting a common ancestral karyotype with x = 12. Differently, Turneroideae and Malesherbioideae present x = 7. A whole genomic duplication detected after the separation of Passifl oroideae and Malesherbioideae suggests that the base number of Passifl oraceae most probably was x = 7, which by dysploidy and polyploidy generated x = 12 for the subfamily Passifl oroideae. Callus induction is one of the pathways required for haploid plant regeneration through anther culture. Pollen viability, as well as the effect of growth regulators and cold pretreatment on anthers of two watermelon lines (Smile and Sugar Baby) to induce callus formation were herein evaluated. Pollen viability was estimated through the staining technique using 2% acetic carmine. Male flower buds were collected and disinfested to allow removal anthers. These anthers were placed on Murashige and Skoog medium, which was supplemented with 2,4-dichlorophenoxyacetic (2,4-D) at 0.0, 0.5, 1.0, 2.0 or 5.0 ?M or with 6-benzylaminopurine at 0.0, 0.5, 1.0, 1.5, or 2.0 ?M, in combination with 2.0 ?M of 2,4-dichlorophenoxyacetic. Anthers were pretreated at 4 °C, for two days and then placed in vitro. Both watermelon lines provided high pollen viability rates (from 93 to 98%). The 2.0 and 5.0 ?M concentrations of 2,4-D stimulated higher friable callus formation. The optimal concentration of 2,4-D was estimated at 3.78 ?M and 4.17 ?M, which had callus induction rates of 64% and 52%, respectively. The combination of 2.0 ?M of 2,4-D and 6-benzylaminopurine did not lead to increased anther response to callus induction. Water deficit and high temperatures are abiotic factors that most limit plant growth and development. However, its effects depend on crop development stage and on stress duration and intensity. Thus, the objective of was to evaluate the development of cowpea subjected to water restriction in different phenological stages and to increase in air temperature. The experiment was conducted with the cultivar ?Carijó?, in growth chambers, in a 4 x 3 x 2 factorial arrangement, corresponding to levels of water availability (25, 50, 75, and 100%,), phenological stages (vegetative, flowering and pod filling) and temperature regimes (T°1: 20-26-33 °C e T°2: 24.8-30.8-37.8 °C), respectively. Reduction of water availability in the vegetative and flowering stages caused decrease in grain production. The percentage of aborted flowers was higher in plants maintained under an increased temperature of +4.8 °C, with consequent reduction in grain production. Higher water availability values favored shoot and root dry mass production. Increase of 4.8 °C did not affect shoot and root dry mass but reduced water use efficiency by about 83%. The highest enzymatic activities of CAT, GPX and SOD were found in plants subjected to the temperature of +4.8 °C. Only APX showed lower enzymatic activity with increasing temperature. Vigorous germination and uniform seedling establishment are determinant and dependent on the performance of cultivars in the face of rising air temperatures. Thus, the objective was to evaluate the initial development of different cultivars of cowpea against the increase of 4.8 ° C in the daily temperature regime. The experiment was carried out in growth chambers, in a 5x2 factorial scheme (cultivars x temperature regimes). It was evaluated: germination percentage, germination speed index, average germination time, average germination speed, average germination speed, the initial seedling length and the initial seedling dry weight. The increase of 4.8 ° C in the temperature did not affect the germination percentage of the seeds, being above 90%. The germination speed index and the average germination speed were higher at temperatures of 24.8-30.8-37.8 ° C. For the germination percentage, the cultivars Pujante, Rouxinol and Tapahium presented the highest indexes, above 90%. The germination speed index and the average germination speed were higher for cv. For Rouxinol, consequently, the average germination time was shorter. The initial seedling length did not differ between cultivars, with an average value of 17.73 cm. The cv. Pujante differed among the other cultivars in terms of seedling dry mass. The initial growth and dry mass of seedlings were higher in the environment with a temperature of 24.8-30.8-37.8 ° C The 4.8 ° C increase in air temperature does not affect the initial phase of cowpea. Seed quality is determined by the environmental conditions to which they were exposed during their formation. Thus, the objective of this study was to evaluate the seed viability and seedling formation of cowpea cultivars produced in a high-temperature environment. Seeds of the cowpea cultivars Carijó, Itaim and Tapahium produced in growth chambers under temperature regimes T°1: 20-26-33 °C and T°2: 24.8-30.8-37.8 °C were used. The experimental design was completely randomized, with four replicates of 25 seeds, placed to germinate at a temperature of 25 °C in B.O.D chambers. Seed sizeis an important agronomic trait and is applicable todifferent abilities. Small seeds guarantee the greater use of the pulp, while larger seeds facilitate sowing. However, there is little work on the genetic control of this characteristicin watermelon. The objective of this work was to study the seed size inheritance in watermelon populations by crossing contrasting genotypes, seeking to gain information to provide technical support during the selection of seed size for the development of new watermelon genotypes. The seed lengths of six populations, P1, P2, F1,F2,BC1and BC2, were measured using the GENES software segregating and nonsegregating generations procedure. This trait is controlled by two genes with incomplete dominance. In addition, depending on the populations studied, inheritance for the characteristicin question may behave differently. Nevertheless, the selection of superior individuals within populations can be performed based on this phenotype, which allows the exploitation of these individuals within breeding programs to develop lines or hybrids. Traits related to fruit yield and physical and chemical fruit quality are being evaluated by Embrapa pumpkin breeding program. Path and Pearson's correlation analysis are necessary to allow the knowledge of the relation between the variables of interest for this program. The objective of the present study was to determine the genetic correlations between yield- and sensorial and nutritional quality attributes-associated variables in pumpkin progenies to support the selection process. Twelve genotypes were evaluated for 10 yield-related traits, and 8 sensory and nutritional quality traits of fruit, including the commercial cultivar Jacarezinho. A path analysis considered the fruit weight (FrW) was as basic variable and the others as explanatory. The FrW had 98% of its total variation available in a function of the explanatory variables. The highest variations were observed in the fruit-shape related traits. In addition, the variables internal median cavity diameter (IDmed), equatorial pulp thickness (EPT), apical pulp thickness (APT), and smallest fruit diameter (Dsm) are indicated for a direct selection of progenies targeting a higher FrW. The correlation analysis highlighted that color-related variables have the possibility of indirect selection aiming to increase total carotenoid content (TCC) and ?-carotene content (?-car). The evaluated progenies showed notable soluble solids content (SSC), TCC and ?-car contents compared to the commercial cultivar. The estimation of genetic parameters allows for the selection of superior individuals for important traits in plant breeding. The objectives of the present study were to estimate and predict the gains in genetic parameters, as well as to select superior individuals for morphoagronomic and pulp quality characteristics in C. moschata. Ten progenies derived from accessions collected in the northeast of Brazil were evaluated for 17 morphoagronomic characteristics and chemical quality of the pulp, including total carotenoids and β-carotene. The low values of additive genetic variance denote the need for additional selection cycles for the evaluated characteristics. Heritability greater than 30% was observed for 13 of the 17 variables analysed, indicating success in selection. Likewise, accuracy values between 74 and 93% were obtained for 15 of the evaluated variables, demonstrating the existence of high genetic variance. Individual ranking was conducted for the variables fruit weight (FRW), soluble solid content (SSC), titratable acidity (TA), total carotenoid content (CBT) and β-carotene (β-Car), identifying five individuals of progeny 10 that were promising for advancement in selection for SSC, CBT and β-car. In ranking the simultaneous evaluations for all the characteristics and taking into account the formats piriform and 'moranga', two individuals of progeny 10, which coincided with the top ranking for SSC, CBT and β-car, were identified. This identification of more than one promising individual enhances the potential of the progenies evaluated for the development of commercial and productive lines in semi-arid conditions. Umbu plants are drought resistant trees which are able to store water and several other substances into its adapted root, named xylopodium. The exsudate from xilopodium is a natural solution rich in salts, sugars and a little concentration of proteins. In this work, we report the purification of a peroxidase (POX) from umbu xilopodium exsudate by direct extraction from polyacrylamide electrophoresis gels. Umbu POX showed optimum activity at pHs around 6.0 to 7.0 and high thermal resistance after incubation at 7 0°C for 6 min. POX activity present in crude extracts was more heat - resistant than in its purified form. When assayed with metal ions, umbu POX activity was shown to be inhibited by Mn 2+ and stimulated by Ca 2+ and Mg 2+ ; it was also inhibited by sodium azid e (concentrations higher then 1 mM) and not inhibited by either EDTA or tropolone. POX Km values for guaiacol and methylcatechol substrates were 6.83 and 22.25, respectively. The enzyme is proposed to be a guaiacol peroxidase and was seen to be located at higher concentrations in the outermost layers of xylopodium tissue, such as the endoderm. Schinopsis brasiliensis is an endangered tree species found in the Caatinga biome. It presents a characteristic slow development and difficult propagation, although it has been traditionally exploited in the region. Application of arbuscular mycorrhizal fungi (AMF) and phosphorus (P) fertilization may be beneficial to S. brasiliensis development at the seedling stage, which at the same time may help species conservation and the recovery of degraded areas in the Caatinga biome. We assessed the response of S. brasiliensis to AMF inoculation (Claroideoglomus etunicatum and Acaulospora longula) and P fertilization (0, 12, 24, and 48 mg dm −3 addition of P 2 O 5 ). S. brasiliensis responded positively to both AMF inoculation and P fertilization. At low P concentrations, the inoculated plants showed higher leaf area and enhanced vegetative development, nutrient content and biomass production compared with non-inoculated plants. Conversely, increasing levels of P fertilization decreased the level of mycorrhizal colonization, plant responsiveness to inoculation, and spore production in C. etunicatum . Thus, P concentrations were able to influence the response of S. brasiliensis to mycorrhization and responsiveness to increased mycorrhization with the decrease in P availability. These results showed that mycorrhizal symbiosis plays an essential role in the development of S. brasiliensis.

The aim of this study was to evaluate three plant species native to Caatinga (Aspidosperma pyrifolium, Ziziphus joazeiro and Pseudobombax simplicifolium) in response to inoculation with two arbuscular mycorrhizal fungi (autoctone: Claroideoglomus etunicatum or exotic: Acaulospora longula) and phosphate fertilization. The experiment was conducted in a greenhouse with a completely randomized design involving a factorial arrangement of three inoculation treatments control, C. etunicatum or A. longula ) and 4 levels of single superphosphate (0 - no added P 2 O 5 , 12, 24 and 48 mg dm -3 ) in 10 repetitions. Under the conditions of this study, the inoculum of C. etunicatum , autoctone to the semi-arid region, showed great compatibility and efficiency in promoting plant growth and nutrient intake when compared to the exotic inoculum of A. longula. Plants of A. pyrifolium were not responsive to mycorrhization and hosphate fertilization. In soil with low phosph orus content, mycorrhized plants of Z. joazeiro and P. simplicifolium presented better vegetative development with high shoot dry weight. Mycorrhized plants of Z. joazeiro had a proportional increase in the levels of macro- an d micronutrients at the phosphorus doses applied. Mycorrhizal symbiosis may have a fundamental role in the development of these species from the Caatinga.

The aim of this study was to identify parents and promising hybrid combinations for the improvement of forage watermelon. Five parents were evaluated: BGCIA 996 (1), BGCIA 997 (2), BGCIA 998 (3), BGCIA 228 (4), Jojoba (5) and ten F1 hybrids, which were obtained from balanced diallel crosses. The experimental design was in a complete randomized block, with three replications. The morphoagronomic and bromatological traits were evaluated. The highlights were the progenitors BGCIA 997, GCIA 998, BGCIA 228 and Jojoba for protein content, fruit yield, in vitrodigestibility of dry matter and number of seeds, respectively. The hybrid 1x4 stood out for fruit length, seed number, and ethereal extract. The hybrid 2x3 stood out for rind and pulp thickness while the hybrids 3x4, 3x5 and 4x5 had exceptional digestibility, protein content and fruit yield, respectively. The analysis of the standard deviation of the SCA estimates of both SD (Sij-Sik) and SD (Sij-Skl) indicated that no hybrids were found that had SCA estimates twice that of SD (Sij-Sik) or SD (Sij-Skl), except for the hybrid 1x4 for the number of seeds per fruit. These results suggest that the parents were more promising than the hybrids. Similarly, the GCA estimates favor the intrapopulation method, which will promote greater efficiency in selection for genetic gains

The traditional method of propagating Etlingera elatior (Jack) R.M. Sm. promotes the proliferation of pathogens, reducing the quality of its output. However, as an alternative, one can use in vitro culture, making it possible to obtain plants free of pathogens on a large scale. Moreover, inoculation of plants with arbuscular mycorrhizal fungi (AMF) can be critical to ensure greater survival and development of these plants under ex vitro conditions. Thus, the present study examined the effect of AMF inoculation (Gigaspora albida and Claroideoglomus etunicatum) on success in the establishment and development of E. elatior on the acclimatization phase. Plants were inoculated with G. albida, C. etunicatum or MIX (both AMF isolates) and after cultivation in a greenhouse for 60 dayswere evaluated for survival percentage, height, leaf area, biomass production of plants, spores number and mycorrhizal colonization. The effect of AMF on plant survival could be observed from 30 days after inoculation; and by 60 days, there was an increase of up to 50% in survival compared to non-inoculated plants. In addition, mycorrhizal inoculation also provided better performance in plant growth, compared to those not inoculated. Finally, different effects were observed between the AMF species studied (Gigaspora albida and Claroideoglomus etunicatum). It follows from this work that plants of E. elatior respond differently to inoculation with different AMF species and show better development when acclimatized with G. albida. A aplicação de fungos micorrízicos arbusculares (FMA) em plantas forrageiras pode contribuir para maior produção de biomassa porém seu uso deve preceder o conhecimento sobre a formação de simbiose na planta de interesse. Desta forma objetivou- se, neste estudo, verificar se plantas de B. cheilantha formavam simbiose micorrízica no campo e se havia variação entre os períodos seco e chuvoso. Após a confirmação da formação de simbiose micorrízica em plantas de B. cheilantha testou-se, em casa de vegetação, o efeito da inoculação com o isolado Claroideoglomus etunicatum sobre o crescimento inicial de B. cheilantha em dois tipos de solo. Em campo, foram observados maior colonização micorrízica no período chuvoso e mais espécies do gênero Acaulospora. Verificou-se, em casa de vegetação, maior desenvolvimento de B. cheilantha micorrizadas em Neossolo quartzarênico e maior esporulação de Claroideoglomus etunicatum em Argissolo acizentado. Conclui-se que no período de maior pluviosidade a colonização micorrízica em B. cheilantha é mais intensa e seu crescimento inicial é favorecido pela micorrização em Neossolo quartzarênico.

The genus Passiflora L. consists of approximately 530 widely distributed species, including Passiflora edulis, which has drawn interest because of its commercial and agronomic value. Passiflora cincinnata is another important species owing to its long flowering period and resistance or tolerance to diseases and pests. In the present study, the meiotic segregation and pollen viability of an interspecific hybrid (P. edulis × P. cincinnata) and its parents were analyzed. The genomic contents were characterized using chromomycin A3 (CMA3)/4′-6′-diamidino-2-phenylindole (DAPI) staining, fluorescent in situ hybridization with 5S/45S ribosomal DNA (rDNA), genomic in situ hybridization (GISH), and inter-simple sequence repeat (ISSR) markers. The results indicated the diploid chromosome number for the parents and interspecific hybrid was 2n = 18. We also observed regular meiosis, one pair of 5S rDNA sites, and two pairs of 45S rDNA sites that co-localized with two pairs of CMA3 +/DAPI− bands. The GISH data revealed three distinct chromosomal groups in the hybrid. The genetic origins of the interspecific hybrid, and its relationship with its parents were also confirmed using ISSR markers.

The fruit production for export is an economically significant activity in the Valley of São Francisco River, especially in the irrigated lands of Petrolina-PE/Juazeiro-BA, Brazil. The development of new genetic material most suitable to the tropical climate and the demands of the consumer market have led to the selection of new seedless grapes cultivars. In this case, the use of the embryo rescue technique has produced satisfactory results for obtaining such materials, especially in the semiarid region. This study aimed to evaluate the in vitro development of intraspecific hybrids of grapevine (Vitis vinifera L.), derived from the rescue of immature embryos resultant from the crossing of Superior Seedless and Thompson Seedless Brazilian clones. To establish and develop the cultivation, the culture media was supplemented with 30 g/L sucrose, 0.1 g/L myo-inositol, 0.002 g/L glycine, 0.1 mg/L indoleacetic acid (IAA), 6.5 g/L of agar, adjusted pH to 5.7. The experiment was evaluated after 90 days. The variables measured were: number of nodes, number of leaves, plant height (cm), number of roots and length (cm) of the root system and internodes. The period of 60 days of in vitro culture of ovules resulted in the highest values of embryos (about 50%), as well as better characterized developmental stages with higher germination (47.3%). The three types of hybrid grapes evaluated in micropropagation showed very similar values of the measured parameters, even having originated from embryos of different developmental stages.

No Brasil, Meloidogyne enterolobii vem causando perdas significativas na produção de goiabeiras e, no Submédio do Vale do São Francisco, por exemplo, o impacto negativo decorrente da infecção e morte de goiabeiras tem refletido diretamente na qualidade de vida dos agricultores. Até o momento, não existem métodos de controle efetivos: os nematicidas avaliados experimentalmente não têm sido eficientes e não há produtos registrados no Ministério da Agricultura, Pecuária e Abastecimento para aplicação em goiabeiras. Assim, há necessidade de realizar pesquisa básica nesta área. A prospecção de proteínas relacionadas com a resistência em espécies resistentes pode ser uma alternativa para o desenvolvimento de medidas para seu controle. Para isso, foram realizados ensaios biológicos de toxicidade para a análise do efeito de extratos proteicos de amendoim sobre o desenvolvimento, a capacidade infectiva e atividade de enzimas proteolíticas de M. enterolobii visando à identificação de proteínas com potencial para controle desse nematoide. Os resultados obtidos indicaram que o extrato proteico total de amostras de raízes de plantas de amendoim inoculadas não tem efeito sobre as três características supracitadas. Sendo assim, são necessários estudos dessa natureza com outras espécies resistentes ao patógeno visando à identificação de proteínas que apresentem potencial para o seu controle.

O cancro bacteriano da videira é causado por Xanthomonas campestris pv. viticola (Xcv). Visando à limpeza clonal de mudas de ?Red Globe?, foram estudados: tamanho ideal de ápices e gemas axilares para cultivo em meio de Galzy modificado (MGM); efeito da termoterapia (38ºC/30 dias); e ação de antibióticos na eliminação de Xcv em videiras infectadas. Os percentuais de contaminação por Xcv e de regeneração foram analisados, e as plantas obtidas foram indexadas em meio ágar nutritivo-dextrose-extrato de levedura-ampicilina (NYDAM), seguindo-se teste de patogenicidade. O cultivo de explantes com 3 mm possibilitou a obtenção de plantas livres da bactéria, com regeneração 14,3 vezes maior que explantes com 1 mm. A termoterapia de mudas infectadas, associada ao cultivo in vitro, não eliminou o patógeno. O cultivo de explantes com 10 mm, durante 40 dias em MGM + cefotaxima (300 mg L-1), proporcionou limpeza clonal das mudas. A indexação de plantas de videira regeneradas in vitro, quanto à infecção por Xcv utilizando NYDAM, seguida de teste de patogenicidade, é uma alternativa econômica e eficiente para produção de mudas de alta qualidade fitossanitária.

Tecidos de plantas infectados constituem uma importante fonte de inóculo primário para fitobacterioses. O objetivo deste trabalho foi investigar a sobrevivência de Xanthomonas campestris pv. viticola (Xcv), agente do cancro bacteriano da videira, em tecidos infectados na superfície do solo e durante a compostagem de restos de poda. Mudas de videira ?Festival? foram inoculadas com mutante resistente à rifampicina Xcv2Rif e mantidas em casa de vegetação até apresentarem alta severidade da doença. No primeiro experimento, ramos fragmentados e folhas inteiras destas plantas foram acondicionados em bolsas de malha plástica e colocados na superfície de microparcelas. No segundo experimento, ramos e folhas fragmentados foram acondicionados em bolsas plásticas e depositados no interior de pilhas de compostagem de restos de poda de videira. A sobrevivência de Xcv2Rif em tecidos infectados foi monitorada a intervalos de 8 e 10 dias a partir do início do primeiro e segundo experimentos, respectivamente. No primeiro experimento, foi também avaliada a decomposição de tecidos, e no segundo, as curvas de temperatura das pilhas, conteúdo de fenóis e microbiota antagonista a Xcv2Rif. O patógeno sobreviveu em altas populações até 80 dias em tecidos infectados na superfície do solo. A compostagem eliminou Xcv2Rif de restos culturais em 10 dias, devido às altas temperaturas, liberação de compostos fenólicos e antagonismo microbiano. Concluindo, em tecidos infectados de videira Xcv sobrevive na superfície do solo por, pelo menos, 80 dias mas é eliminada pela compostagem em 10 dias.

Apesar de oferecer uma série de vantagens, quanto à qualidade do produto final, a técnica de cultivo in vitro de plantas ainda é considerada dispendiosa, por causa, dentre outros fatores, da utilização de reagentes com alto grau de pureza no preparo de meios nutritivos. Entre as alternativas que podem ser adotadas para a redução dos custos relacionados com a produção de mudas, apresenta potencial a substituição de produtos PA por aqueles de menor custo. Baseado nestas informações, o objetivo deste estudo foi avaliar o efeito do melado de cana-de-açúcar sobre o desenvolvimento in vitro de bananeira, cv. Maçã. Para tal propósito, plantas de bananeira foram inoculadas em meios nutritivos, formulados à base de melado de cana-de-açúcar, com quatro concentrações distintas (brix de 1,5; 3,0; 4,5 e 6,0), e os dados obtidos foram comparados com aqueles das plantas cultivadas em meio MS (controle), perfazendo um total de cinco tratamentos. Apesar de terem-se desenvolvido em todos os tratamentos testados, observou-se que, para as variáveis número médio de folhas e biomassa da matéria fresca, as plantas cultivadas no meio MS (controle) apresentaram valores superiores. Entretanto, para o número médio de raízes, não houve diferença estatística entre os tratamentos utilizados. O melado de cana-de-açúcar não favorece o desenvolvimento de plântulas de bananeira, mas pode ser utilizado para o enraizamento dessas plantas in vitro.

In this work, the cytotaxonomic implications of the chromosomal characterization of cultivated and native Zephyranthes species described in northeastern Brazil were studied. All individuals had karyotype formed by a set of metacentric chromosomes, in addition to submetacentric and acrocentric chromosomes. In Zephyranthes robusta, 2n = 12 was observed and karyotype with formula 4M ? 2SM in somatic cells, representing the most symmetric karyotype among the investigated species. Z. sylvatica showed three different chromosome complement numbers: 2n = 12 with formula 1M ? 5SM, 2n = 12 ? 1B with1M ? 5SM ? (1B), and 2n = 18 formed by cracks. The cultivated species Z. rosea Lindl. presented 2n = 24 with 4M ? 7SM ? 1A, however Z. grandiflora Lindl. showed the same chromosome number with 2M ? 5SM ? 5A. Zephyranthes aff. rosea Lindl. presented 2n = 25 with one small metacentric forming a crack in the fourth metacentric pair. Z. brachyandra has 2n = 24 ? (1B) and formula 4M ? 3SM ? 5A ? (1B). Z. candida Herb. presented 2n = 38 and karyotype formula 9M ? 10SM. In Habranthus itaobinus numerical variation was observed, with the majority of populations showing a chromosome complement composed of 2n = 44 ? 1B with 5M ? 12SM ? 5A ? (1B), or 2n = 44 ? 3B in a single population. Mechanisms involved in the formation of these karyotypes from chromosomal imbalance data are discussed. Taken together, data from this study only partially confirm previous counts for epithets and further enhance the cytological variability data previously reported for the genus.

Species belonging to the Amaryllidaceae (Zephyranthes and Habranthus) were analyzed by banding with chromomycin A3 (CMA)/4,6-diamidino-2-phenylindole (DAPI) fluorochromes. The patterns of bands were studied in seven species of Zephyranthes Herb. and one of Habranthus Herb. Subterminal and interstitial DAPI+ bands were observed in Z. robusta 2n = 12 and Z. brachyandra 2n = 24. Other species showed no AT-rich heterochromatin. In species with 2n = 12, CMA+ bands were observed on one chromosome pair of Z. robusta and Zephyranthes sp., while in Z. sylvatica an additional small terminal band in the fifth chromosome pair was observed. Z. rosea and Z. grandiflora presented with 2n = 24 and had four CMA+ bands, while in Z. brachyandra, with 2n = 24 + 1B, there were eight interstitial dot bands and a larger terminal band in the short arm of the B chromosome. Z. candida with 2n = 38 presented CMA+ heterochromatin blocks on the long arms of five metacentric pairs and in the short arm of one of the submetacentric pairs; in addition a terminal band was observed on the long arm of one of the homologues of a larger submetacentric pair. H. itaobinus showed a heterozygous pair revealing a strong CMA+ band in only one of the homologues, likely a nucleolus organizing region. Taxonomic implications and karyotype evolution of this group are discussed and correlated with previous data from the literature.

Euchroma giganteawas karyotypically studied using conventional staining, C-banding, silver nitrate staining and ribosomal fluorescent in situ hybridization (rDNA FISH). Broad wide autosomal polymorphism and a complex sex determination system were found in this beetle. Karyotype complements ranging from 2n = 32, X1X2X3Y1Y2Y3 to 2n = 36,X1X2X3Y1Y2Y3 were detected in the sample analyzed. Punctiform suernumerary chromosomes were present in the different karyotypes. The karyotypic evolution of Brazilian E. giganteamay have taken two directions, reduction in the diploid number of 2n = 36 to 24 through centric fusions or 2n = 24 to 36 due to chromosomal fissions. In addition, pericentric inversions were also involved. The complex multiple sex mechanism of this species seems to be old and well established since it is found in specimens from different populations. Small pericentromeric blocks of constitutive heterochromatin were located on the autosomes and terminal blocks were also found on some small pairs. The sex chromosomes showed larger constitutive heterochromatin blocks. Silver nitrate staining during prophase I of meiosis showed labeling of the sex chromosome chain. However, the rDNA sites could only be precisely determined by FISH, which permitted the identification of these ribosomal sites on chromosomes X1 and X2 of this species. Zephyranthes Herb. is a taxonomically complex and cytologically variable group, withabout 65 species of Neotropical distribution. Chromosome number variability in 32 individuals of a Zephyranthes sylvaticapopulation from Northeast Brazil was investigated. Three cytotypes were found: 2n = 12 (one metacentric, four submetacentricand one acrocentric pairs), in 24 individuals; 2n = 12 + 1B, in five and three individuals with 2n = 18, a triploid cytotype.All diploid individuals showed chromosomes with polymorphism in pair one and two, while in triploids this polymorphismwas observed in all chromosome triplets, generally with two homomorphic chromosomes and a higher or lower heteromorphicchromosome. All individuals had reticulated interfasic nucleus and a slightly asymmetric chromosome complement, withone metacentric chromosome pair and the others more submetacentric to acrocentric. These data confirm the cytologicalvariability previously registered for the genus. Mechanisms involved in karyotypic evolution in this population are discussed. A manga do Vale do São Francisco é uma das principais espécies da fruticultura brasileira.

A cultura da goiabeira, especialmente da variedade Paluma, é de grande importância para a economia da região do Submédio do São Francisco. No entanto, perdas significativas estão ocorrendo nas plantações de goiaba em virtude da infecção pelo nematóide Meloidogyne mayaguensis. Sendo assim, a produção in vitro de goiabeira desta variedade é uma alternativa indicada para obtenção de mudas de alta qualidade sanitária. Entretanto, são encontrados na literatura diversos protocolos com muitas variações, tanto nas concentrações quanto nos tipos de reguladores de crescimento responsáveis por determinados padrões de desenvolvimento. Além disso, estudos realizados demonstraram a ocorrência da variação de protocolos de micropropagação da goiabeira entre as diferentes variedades. Chromosome analyses were performed in two grasshopper species of the genus Schistocerca, S. pallens and S. flavofasciata. Both species shared the same diploid number (2n = 23, X in males; 2n = 24, XX in females); and a con- served karyotype composed exclusively of acrocentric chromosomes but differed in their distribution patterns of constitutive heterochromatin and nucleolus organizer regions (NORs). Constitutive heterochromatin was located in the pericentromeric region of alI chromosomes in both species. S. flavofasciata presented an additional C-band on the distal region of the long arm of a small autosome pair (S9). Nucleolus organizer regions (NORs), revealed by silver nitrate staining (Ag-NORs), were observed on a medium autosome pair (M5) in both species. Fungos micorrízico arbusculares e vermicomposto na aclimatação de Alpinia purpurata (Viell.) Schum e Zingiber spectabile Griff. (Zingiberaceae)). Visando selecionar fungos micorrízicos arbusculares (FMA) e substratos para produção de mudas micropropagadas de alpinia (Alpinia purpurata) e sorvetão (Zingiber spectabile), conhecidas espécies ornamentais tropicais, foram inoculadas mudas com 1.500 propágulos infectivos de Gigaspora albina, Glomus etunicatum e Acaulospora longula, em recipientes contendo: (a) solo; (b) solo e vermicomposto (3:1) e (c) solo, areia e vermicomposto (2:2:1). O delineamento foi inteiramente casualizado em fatorial de: quatro tratamentos de inoculação (três com FMA e controle não inoculado), três substratos, oito repetições, para cada vegetal. Após 70 (Z. spectabile) e 100 (A. purpurata) dias da inoculação, parâmetros de crescimento da planta foram avaliados tais como altura, número de folhas e perfilhos, massa seca da parte aérea e subterrânea, área foliar, e ainda a colonização micorrízica e a esporulação dos FMA. Não houve interações significativas entre os FMA e os substratos testados. Em A. purpurata os dois substratos com vermicomposto promoveram maior desenvolvimento; em Z. spectabile, apenas o substrato com maior proporção do fertilizante orgânico induziu crescimento diferenciado. A aplicação de adubo reduziu a colonização micorrízica dos dois hospedeiros e a esporulação de A. longula. Na fase de aclimatação, o uso de substratos orgânicos favorece a formação de mudas vigorosas de alpinia e sorvetão, mas pode ser prejudicial para a comunidade de FMA. Ancorados em pesquisas realizadas em parceria com a Embrapa, os estudos mostram a viabilidade do novo cultivo, desde que submetido a um manejo especial que considere as condições de clima e solo do semi-árido. A expectativa é que as laranjas, pomelos, tangerinas e limões devem liderar a expansão da citricultura nos sertões de Pernambuco e Bahia.
