15. Relying on a single anuran species: Feeding ecology of a snake community on kinkasan Island, miyagi prefecture, Japan

Abstract

Feeding habits of snakes on Kinkasan Island, where potential food resources for snakes is limited, was investigated based on field surveys of six years. In total 280 snakes, which consisted of five species (Elaphe quadrivirgata, E. climacophora, Euprepiophis conspicillatus, Rhabdophis tigrinus, and Gloydius blomhoffii), were collected. A total of 168 prey items were recovered from the stomach contents of these snakes. The diet of R. tigrinus and G. blomhoffii exclusively consisted of a frog, Rana tagoi, which is the only amphibian species inhabiting Kinkasan Island. Diet of El. quadrivirgata was also predominantly comprised of Ra. tagoi ( 97.7%). O nly e ndothermic animals were found in the stomach contents of El. climacophora and Eu. conspicillatus. Direct observations of feeding behavior and a broad range of body temperature of El. quadrivirgata indicated that during the breeding season of Ra. tagoi, this snake forages for frogs hiding in crevices of rocks immersed in cold running water of mountain streams. Our study demonstrated that, given the limited prey fauna on Kinkasan Island, two dietary generalists, El. quadrivirgata and G. blomhoffii, and one amphibian specialist, R. tigrinus, largely rely on the single frog species as major diet. High exploitation of Ra. tagoi by El. quadrivirgata would be attained by its unique foraging tactics that are characterized by a shuttling movement between hot basking rocks and cold streams for foraging.

17. Comprehensive clinical and molecular studies in split-hand/foot malformation: identification of two plausible candidate genes (LRP6and UBA2)

Abstract

Split-hand/foot malformation (SHFM) is a clinically and genetically heterogeneous condition. We sequentially performed screening of the previously identified Japanese founder 17p13.3 duplication/triplication involving BHLHA9, array comparative genomic hybridization, and whole exome sequencing (WES) in newly recruited 41 Japanese families with non-syndromic and syndromic SHFM. We also carried out WES in seven families with nonsyndromic and syndromic SHFM in which underlying genetic causes including pathogenic copy-number variants (CNVs) remained undetected in our previous studies of 56 families. Consequently, we identified not only known pathogenic CNVs (17p13.3 duplications/triplications [n= 21], 2q31 deletion [n= 1], and 10q24 duplications [n= 3]) and rare variants in known causative genes (TP63[n= 3], DLX5[n= 1], IGF2[n= 1], WNT10B[n= 3], WNT10B/PORCN[n= 1], and PORCN[n= 1]), but also a de novo 19q13.11 deletion disrupting UBA2(n= 1) and variants that probably affect function in LRP6(n= 1) and UBA2(n= 1). Thus, together with our previous data based on testing of 56 families, molecular studies for a total of 97 families with SHFM revealed underlying genetic causes in 75 families, and clinical studies for the 75 families indicated a certain degree of correlation between genetic causes and phenotypes. The results imply that SHFM primarily occurs as a genetic disorder with genotype–phenotype correlations. Furthermore, the results together with previous data such as the development of SHFM in Lrp6knockout mice, the presence of SHFM in two subjects with 19q13 deletions involving UBA2, and strong mouse Uba2expression in the developing limb buds, imply that LRP6and UBA2represent plausible candidate genes for SHFM.

Abstract

Background: In Japan, waist circumference (WC) percentiles to screen for childhood metabolic syndrome (MetS) are unavailable. The objectives of this study were to develop WC and WC-to-height ratio (WC/Ht) percentile curves by age and sex for Japanese children, and to test their utility in screening for MetS in children with obesity who are otherwise healthy.Methods: The WC and WC/Ht percentiles were developed using the LMS method of summarizing growth standards, which monitors changing skewness (L), medians (M), and coefficients of variation (S) in childhood distributions. A representative dataset was used, which consisted of 3,634 boys and 3,536 girls aged 4.5-12.75 years in Shizuoka prefecture, Japan, between 2010 and 2012. Children who were obese (355 boys and 230 girls) aged 6-12 years from Osaka prefecture, Japan, were screened for childhood MetS using the new percentiles and the International Diabetes Federation's (IDF's) definition of MetS.Results: The number of participants with certain metabolic abnormalities (high systolic and diastolic blood pressure, and a high level of triglycerides) was significantly higher in boys aged 10-12 years, with a WC ≥ 90th percentile, than among those with a WC < 90th percentile. None of the participants with a WC < 90th percentile exhibited two or more metabolic abnormalities, regardless of their age or sex. Among the participants aged 10-12 years, 11.4 % of boys and 4.4 % of girls with a WC ≥ 90th percentile were diagnosed with MetS.Conclusions: The new percentiles may have a certain level of potential to screen Japanese children for childhood MetS in accordance with the IDF definition. Abstract

Wage discrimination can be defined as arbitrarily assigning wages to workers based on factors unrelated to job performance or merit (Coleman, 2003). The Equal Pay Act of 1963 outlawed all forms of wage discrimination based on gender. Then came Title VII of the Civil Rights Act of 1964, which outlaws wage discrimination on the basis of race, color, sex, religion, or national origin. In 1981, the Supreme Court broadened the scope of Title VII beyond the Equal Pay Act of 1963, asserting that the latter prohibits wage discrimination even when the jobs are not the same. From a practical standpoint, however, it is worth noting that wage discrimination laws are often difficult to enforce because they are hard to prove and win in court. In recent years, enforcement by the Equal Employment Opportunity Commission (EEOC) has been lax, placing the economic burden on the employee.
