4. Suppression of TNF‐α activity by immobilization rescues Mkx expression and attenuates tendon ossification in a mouse Achilles tenotomy model.

Abstract

Traumatic heterotopic ossification is a condition in which extraskeletal bone formation occurs in soft tissues after injury. It most commonly occurs in patients who had major orthopedic surgery and in those with severe extremity injuries. The lesion causes local pain and can impair motor function of the affected limb, but there is currently no established prophylaxis or treatment for this condition. In this study, we show that immobilization at an early stage of the inflammatory response after injury can attenuate ossification formation in a murine Achilles tenotomy model. Gene expression analysis revealed a decrease in the expression of Tnf and an increase in the expression of Mkx, which encodes one of the master regulators of tendon differentiation, Mohawk. Notably, we found that TNF‐α suppressed the expression of Mkx transcripts and accelerated the osteogenic differentiation of tendon‐derived mesenchymal stem cells (MSCs), suggesting that TNF‐α acts as a negative regulator of Mkx transcription. Consistent with these findings, pharmaceutical inhibition of TNF‐α increased the expression of Mkx transcripts and suppressed bone formation in this mouse model. These findings reveal the previously unrecognized involvement of TNF‐α in regulating tendon MSC fate through suppression of Mkx expression and suggest that TNF‐α is a potential target for preventing traumatic heterotopic ossification. Abstract

The LBX1 gene is located near a single nucleotide polymorphism that is highly associated with susceptibility to adolescent idiopathic scoliosis and is considered one of the strongest candidate genes involved in the pathogenesis of this condition. We have previously found that loss of LBX1 from skeletal muscle results not only in spinal deformity but also in lean body mass, suggesting a potential role for LBX1 in energy metabolism. The purpose of the present study was to test this hypothesis by analyzing the phenotype of mice lacking LBX1 in skeletal muscle with a focus on energy metabolism. We found that loss of LBX1 rendered mice more resistant to high-fat diet-induced obesity, despite comparable food intake between mutant and control mice. Notably, the mutant mice exhibited improved glucose tolerance, increased maximal aerobic capacity, and higher core body temperature compared to control mice. In addition, we found that overexpression of LBX1 decreased glucose uptake in cultured cells. Taken together, our data show that LBX1 functions as a negative regulator of energy metabolism and that loss of LBX1 from skeletal muscle increases systemic energy expenditure resulting in lean body mass. Abstract

LBX1 is a gene located near a single‐nucleotide polymorphism, rs11190870, which is highly associated with susceptibility to adolescent idiopathic scoliosis. However, the potential involvement of LBX1 in the etiology of this spinal deformity has not been elucidated. In this study, we aimed to determine whether the lack of LBX1 in skeletal muscle results in spinal deformities in mice. We generated mutant mice in which the Lbx1 allele was conditionally excised under the control of a human muscle actin promoter. Mice lacking LBX1 from the skeletal muscle were fertile and available. The mutant mice had hypoplastic forelimbs and weighed less than control animals, but otherwise, there were no overt anomalies. The mice did not exhibit a scoliosis‐like spinal deformity; however, they developed moderate kyphosis as they grew old. These observations indicated that LBX1 is involved in limb development and potentially in the maintenance of spinal curvature/alignment in mice. [ABSTRACT FROM AUTHOR]

Abstract

Sialic acid (Sia) is a group of acidic sugars with a 9-carbon backbone, and classified into 3 species based on the substituent group at C5 position: N -acetylneuraminic acid (Neu5Ac), N -glycolylneuraminic acid (Neu5Gc), and deaminoneuraminic acid (Kdn). In Escherichia coli , the sialate aldolase or N -acetylneuraminate aldolase (NanA) is known to catabolize these Sia species into pyruvate and the corresponding 6-carbon mannose derivatives. However, in bacteria, very little is known about the catabolism of Kdn, compared with Neu5Ac. In this study, we found a novel Kdn-specific aldolase (Kdn-aldolase), which can exclusively degrade Kdn, but not Neu5Ac or Neu5Gc, from Sphingobacterium sp. which was previously isolated from a Kdn-assimilating bacterium. Kdn-aldolase had the optimal pH and temperature at 7.0–8.0 and 50 °C, respectively. It also had the synthetic activity of Kdn from pyruvate and mannose. Site-specific mutagenesis revealed that N50 residue was important for the Kdn-specific reaction. Existence of the Kdn-aldolase suggests that Kdn-specific metabolism may play a specialized role in some bacteria. [ABSTRACT FROM AUTHOR]

33. Three Dimensional Measurement by Satellite Remote Sensing for Disaster Monitoring

Abstract

SHIKOKU Island in Japan has many slope failure disasters by the torrential rain every year. The slope failures induce other serious disaster such as landslide, mudflow and debris flow. Therefore, hidden slope failure must be found immediately. PRISM sensor mounted on ALOS has three lines scanner with 2.5m ground resolution and it can be observed at the same time from same orbit. Then, three dimensional (3D) measurements can be carried out in 2.5m spatial resolution. The 3D measurement using PRISM has a potential to detect slope failure. In the present research work, accuracy verification of 3D measurement using PRISM was carried out and the possibility of wide area monitoring of slope failure disaster was concluded. Result showed about 1 pixel error in z axis. Then, at least 5m width of slope failure might be expected to detect by PRISM using 3D perspective projection.

Abstract

Heterotopic ossification is abnormal bone formation in soft tissues that occurs primarily after injury and major surgery. This condition often causes local pain and limits joint motion in the affected limb. Currently, there is no effective treatment or prophylaxis for this condition other than surgical removal of the lesion. Recent studies suggest that retinoic acid receptor (RAR) agonists are effective in suppressing heterotopic ossification in patients with Fibrodysplasia Ossificans Progressiva, a congenital disorder characterized by progressive ossification of soft tissue, by suppressing the aberrant differentiation of mesenchymal stem cells in muscle. In this study, we aimed to elucidate the potential use of RAR agonists in suppressing injury‐induced ectopic tendon ossification using a mouse Achilles tenotomy model. Contrary to our initial hypothesis, administration of RAR agonists throughout the experimental period (5 weeks) accelerated ectopic tendon ossification in our model. Of note, in vitro differentiation experiments using tendon‐derived mesenchymal stem cells revealed that RAR agonists play opposing roles in osteogenic and chondrogenic differentiation, promoting the former and suppressing the latter. Indeed, we found that RAR agonists suppressed tendon ossification when administered before cartilage nodule formation, but promoted it when administered after. These results suggest that RAR agonists have a dual and opposing effect on tendon ectopic ossification, depending on the duration and timing of their administration. Our data may provide a basis for further investigation of the potential use of RAR agonists in the treatment of injury‐induced heterotopic ossification. [ABSTRACT FROM AUTHOR]

44. Whole-body counting of Fukushima residents after the TEPCO Fukushima Daiichi Nuclear Power Station accident

Abstract

At the request of Fukushima Prefecture, the Japan Atomic Energy Agency (JAEA) started whole-body counting of residents on July 11, 2011 to assess radiation exposure after the TEPCO Fukushima Daiichi Nuclear Power Station accident. The JAEA has examined residents in Iitate, Kawamata, Namie, and 8 other local communities. The measurement capacity of the whole-body counting device is approximately 100 persons per day and the total number of people to measure reached 9,927 by the end of January 2012. Two types of whole body counters equipped with large-size NaI(Tl) detectors were used to perform the measurements. Routinely used phantoms (Canberra RMC-II transfer phantom or water-filled block phantom developed by JAEA) were used to perform peak efficiency calibration of the counters and the results of peak efficiency calibration were verified with different-sized bottle mannequin absorber (BOMAB) phantoms imitating an adult male, a 10-year-old and a 4-year-old. As the measurement started at 4 month after the accident, short half-life radionuclides such as 131I originating from the accident were not detected in this work. Approximately 80% of the residents had levels below the minimum detectable amount (MDA). No artificial nuclides other than 134Cs and 137Cs were found in the present whole-body counting. The maximum whole-body content of radiocesium (134Cs and 137Cs together) was 2.7 kBq for children 1 mSv, and the maximum CED was 3 mSv. The extrapolated 50th percentiles (medians) of the CED for 13-17 years old and those >17 years old were 0.02 mSv and 0.025 mSv, respectively. These values represent the basic knowledge for the reconstruction of internal exposure of the entire resident population.

45. Direct measurements of employees involved in the Fukushima Daiichi nuclear power station accident for internal dose estimates: JAEA's experiences

Abstract

Japan Atomic Energy Agency (JAEA) performed internal dose measurements of employees involved in the Fukushima Daiichi nuclear power station accident. Nuclear Fuel Cycle Engineering Laboratories (NFCEL), one of the JAEA's core centers, examined 560 of these employees by direct (in vivo) measurements during the period from April 20 to August 5 in 2011. These measurements consisted of whole-body counting for radiocesium and thyroid counting for radioiodine. The whole-body counting was conducted with two types of whole-body counters (WBCs): a standing-type WBC with two large NaI(Tl) detectors (FastscanTM, Canberra Inc.) and a chair-type WBC with HPGe detectors (GC5021, Canberra Inc.) installed in a shielded chamber made of 20-cm-thick steel. The thyroid counting was mainly performed using one of the two HPGe detectors equipped with the chair-type WBC. The subjects examined in this work were divided into two groups: Group 1 was the first 39 subjects who were measured up to June 17, 2011 and Group 2 was the remaining 521 subjects who were measured on and after June 18, 2011. We validated the performance of our direct measurements by comparing measurement results of the Group 1 subjects using two different methods (e.g., the standing-type WBC vs. the chair-type WBC). Tentative internal dose estimates of the subjects of Group 1 were also performed based on the assumption of a single intake scenario on either March 12, when the first hydrogen explosion occurred at the station or the first day of work after the accident. It was found that the contribution of 131I to the total internal dose greatly exceeded those of 134Cs and 137Cs, the other major nuclides detected in the measurements. The maximum committed effective dose (CED) was found in a male subject whose thyroid content of 131I was 9760 Bq on May 23, 2011; the CED of this subject was estimated to be 600 mSv including a small contribution of 134Cs and 137Cs. The typical minimum detectable activity for 131I in the present thyroid counting was 10 Bq for a counting time of 10 min, making it difficult to identify a residual thyroid content corresponding to a CED of 20 mSv for the subjects of Group 2.

Abstract

P(論文), Pseudo-random bit sequence (PRBS) generators which can simulate the generation of the actual information signal, are installed in the transmitter side, as well as bit error rate detectors are installed in the receiver side to evaluate transmission properties of the optical communication systems. Although very expensive, these measuring instruments have been used compound semiconductor devices as GaAsMESFETs to get high-speed operation.This time, the examination circuits of the PRBS generator are applied to economic CMOSFETs which have low-power consumption and expected to have high-speed operation by shortening channel length of the CMOSFETs.Basic operations of the PRBS generators employed CMOSFETs are confirmed through logical circuit simulations and measurements of the trial manufactured circuits in this study.

Abstract

This paper describes humidity characteristics of Plasma Synthetic Jet Actuator (PSJA). PSJA is flow control device using the Electro Hydro Dynamic (EHD) effect of the glow-discharge. PSJA can be downsized, controlled easily, and has quick control response. Though the flow control around an airfoil with PSJA is mainly studied, its characteristics have not been clarified. So, it is necessary to investigate humidity characteristics of induced flow by PSJA. In this paper, the induced flow from PSJA was measured in the different relative humidity. The experimental results show that the effect of induce flow by PSJA had decreased with increase relative humidity. Therefore, when PSJA was used, relative humidity becomes an important parameter.
