Abstract

In their comment (1), Hawreliak et al. claims that our observation of stacking fault formation and transonic dislocation propagation in diamond (2) is not valid as they interpret the observed features as cracks. In this response letter, we describe our rationale for interpreting the observed features as stacking faults. We also address other points raised in their comments, including the clarifications of how the results of Makarov et al. Abstract

We present measurements on Fe$_2$O$_3$ amorphization and melt under laser-driven shock compression up to 209(10) GPa via time-resolved in situ x-ray diffraction. At 122(3) GPa, a diffuse signal is observed indicating the presence of a non-crystalline phase. Structure factors have been extracted up to 182(6) GPa showing the presence of two well-defined peaks. A rapid change in the intensity ratio of the two peaks is identified between 145(10) and 151(10) GPa, indicative of a phase change. Present DFT+$U$ calculations of temperatures along Fe$_2$O$_3$ Hugoniot are in agreement with SESAME 7440 and indicate relatively low temperatures, below 2000 K, up to 150 GPa. The non-crystalline diffuse scattering is thus consistent with the - as yet unreported - shock amorphization of Fe$_2$O$_3$ between 122(3) and 145(10) GPa, followed by an amorphous-to-liquid transition above 151(10) GPa. Upon release, a non-crystalline phase is observed alongside crystalline $\alpha$-Fe$_2$O$_3$. Abstract

The motion of line defects (dislocations) has been studied for over 60 years but the maximum speed at which they can move is unresolved. Recent models and atomistic simulations predict the existence of a limiting velocity of dislocation motions between the transonic and subsonic ranges at which the self-energy of dislocation diverges, though they do not deny the possibility of the transonic dislocations. We use femtosecond x-ray radiography to track ultrafast dislocation motion in shock-compressed single-crystal diamond. By visualizing stacking faults extending faster than the slowest sound wave speed of diamond, we show the evidence of partial dislocations at their leading edge moving transonically. Understanding the upper limit of dislocation mobility in crystals is essential to accurately model, predict, and control the mechanical properties of materials under extreme conditions.

4. Coupling Effects in Multi-Stage Laser Wake-field Acceleration of Electrons

Abstract

Staging laser wake-field acceleration is considered as a necessary technique for developing full-optical jitter-free electron accelerators. Splitting of the acceleration length into several technical parts with their lengths smaller than the dephasing length and with independent laser drivers allows generation of stable, reproducible acceleration fields. Temporal and spatial coupling of pre-accelerated electron bunches for their injection in the acceleration phase of a successive laser pulse wake field is the key part of the staging laser-driven acceleration. Here, characterization of the coupling is performed with dense, stable, a narrow energy band <3% and energy selectable electron beams with charges ~1.6 pC and energy ~10 MeV generated from a laser plasma cathode. Cumulative focusing of electron bunches in a low density pre-plasma, exhibiting the Budker- Bennett effect, is shown to result in the efficient injection of electrons even with a long distance between the injector and the booster in the laser pulse wake. Measured characteristics of electron beams modified by the booster wake field agree well with those obtained by multidimensional particle-in-cell simulations.

7. A 226.875-Gbit/s/λ Optical Receiver Based on a High-Frequency Integration by Flip-Chip Mounting

Abstract

A high-speed optical receiver for IM-DD 226.875-Gbit/s/ $\lambda $ transmission was designed and fabricated by using high-frequency, integrated flip-chip (FC) mounting. Thanks to the developed FC-mounting technique, the optical receiver exhibited high 3-dB bandwidth and less-fluctuation optical-to-electrical (O/E) frequency response. The fabricated optical receiver demonstrated 3-dB bandwidth of 55.1 GHz with less-fluctuation O/E frequency response up to 50 GHz (i.e., Nyquist frequency under 100-Gbaud operation at 4-PAM modulation frequency). The simulated transmission performance of the optical receiver reveals that its link budget for 226.875-Gbit/s/ $\lambda $ transmission is expanded by 3.7 dB by using the developed FC-mounting technique. The fabricated optical receiver also experimentally demonstrated transmission of a 226.875-Gbit/s signal over 20-km SSMF.

Abstract

Interaction of axisymmetrically-polarized (radially or azimuthally-polarized), relativistically intense laser pulses (ALP) with under-dense plasma is shown experimentally to be different from the interaction of conventional Gaussian pulses. The difference is clearly observed in distinct spectra of scattered laser light as well as in appearance of a strong side emission of second harmonic in the vicinity of focus spot. According 3D particle-in-cell simulations, this is a result of instability in the propagation of ALP in under-dense plasma. Laser wakefield acceleration of electrons by ALP, therefore, is less efficient than that by Gaussian laser pulses but ALP may be interesting for efficient electron self-injection., Comment: 10 pages, 5 figures

21. Analgesic efficacy of continuous wound infiltration compared with continuous intravenous fentanyl after gynaecological surgery: a non‐inferiority, randomised controlled trial.

Abstract

Objective: The present trial aimed to prove the non‐inferiority of the analgesic efficacy of continuous wound infiltration (CWI) to that of continuous intravenous fentanyl (IV) and to compare the safety of the two methods. Methods: This trial was a prospective, single‐centre, two‐arm, non‐inferiority, randomised controlled trial. Patients participating in the trial were randomised to a CWI group or an IV group. The VAS (visual analogue scale), additional analgesic usage and side effects were then compared between the groups. Results: In total, 61 patients were enrolled; two in CWI were excluded, leaving 59 (30 in the CWI group and 29 in the IV group) for analysis. The difference in the VAS score at 24 h (CWI group – IV group) was −3.2 (95% confidence interval [CI] −14.7 to 8.2), which was less than the non‐inferiority margin of 15. The mean amount of total fentanyl use at postoperative hour 48 was 1395 (95% CI 886–1903) µg in the CWI group and 3186 (95% CI 2716–3658) µg in the IV group. The amount of other analgesics and the incidence of adverse effects did not differ significantly between the groups. Conclusion: CWI was non‐inferior to IV in terms of its analgesic effect, and has an opioid sparing effect in open gynaecological surgery. [ABSTRACT FROM AUTHOR]

26. Zero-Dispersion Wavelength Estimation and Optimization Method for Penalty-Free and Equalizer-Free 53-Gbps/λ IM-DD Transmission Over 100 Km

Abstract

We propose the method to estimate the zero-dispersion wavelength (ZDW) and to optimize the emission wavelength for high-speed intensity modulation and direct detection (IM-DD) transmission. The feature of this method can estimate the ZDW by using the unique notch frequency caused by chromatic dispersion from the frequency spectrum of the received signal, and reduce chromatic dispersion by tuning the emission wavelength to the estimation value of the ZDW in a configuration that using only general transmitters/receivers without additional dedicated equipment. With the proposed method, the limitation of transmission distance can be relaxed in high-speed IM-DD transmission without using a dispersion compensation device or an equalizer. We experimentally demonstrated that the notch frequency was observed at a condition of the pseudorandom binary sequence (PRBS) 215-1 and 231-1 non-return-to-zero (NRZ) signals at 53.125 Gbps for a total 80 and 100 km, and the ZDW was estimated with fast Fourier transformation sizes of 256, 512, and 1024. The results indicated that the proposed method can be estimated with an accuracy of about ±1 nm from the true value of the ZDW. In addition, to confirm the feasibility of the proposed method, we experimentally demonstrated that PRBS 215-1 and 231-1 NRZ signals at 53.125 Gbps were transmitted over 100 km by tuning the emission wavelength to the estimated ZDW value. For the ZDW with a true value of 1312.20 nm, the chromatic dispersion penalty was up to 1.5 dB under the conditions of a 53.125 Gbps NRZ signal with an emission wavelength of 1303.73 nm and a transmission distance of 100 km. Penalty-free and equalizer-free transmission was demonstrated by applying this method.

Abstract

In situ femtosecond x-ray diffraction measurements and ab initio molecular dynamics simulations were performed to study the liquid structure of tantalum shock-released from several hundred gigapascals (GPa) to the ambient condition on the nanosecond timescale. The results show that the internal negative pressure applied to the liquid tantalum reached -5.6 (0.8) GPa, suggesting the existence of a liquid-gas mixing state due to cavitation. This is the first direct evidence to prove the classical nucleation theory which predicts that liquids with high surface tension can support GPa regime tensile stress.

Abstract

Background: Isocitrate dehydrogenase (IDH)-1 and -2 are TCA cycle-involved enzymes which convert isocitrate to alpha-ketoglutarate. Mutations that alter the enzymatic activity causes accumulation of a mal-metabolite D-2-hydroxyglutarate, which results in inhibition of DNA methylation and tumorigenesis. IDH-1 and IDH-2 mutation are present in approximately 7-10% and 10% of patients with acute myeloid leukemia (AML), respectively. Recently, whole exome sequencing has been used for the next-generation sequencing of AML, and certain gene mutations have been identified in patients with AML. The treatment strategies for leukemia have undergone drastic changes with the rapid development of new drugs. However, the proper use of newly developed agents poses a major challenge in AML treatment. Genome profiling analysis can be used to select the optimal treatment for patients with newly diagnosed AML. IDH mutant-specific inhibitors such as ivosidenib and enasidenib were already approved in the US, and combination treatment with venetoclax and Azacitidine was recently approved in Japan. Methods and Results: We lunched an actionable mutation profiling multicenter study named Hematologic Malignancies (HM)-SCREEN-Japan 01 (UMIN000035233), in which a comprehensive genomic assay was performed by Foundation One Heme (F1H) panel. The primary outcome was the frequency of each genomic alteration, as determined using F1H, which is a comprehensive genome profiling test based on next-generation sequencing, in the AML specimens. The secondary outcome was the association between each genomic alteration and the clinicopathological characteristics, prognosis, and quality of specimens used in the genetic analysis. The eligibility criteria were as follows: 1) histological diagnosis of AML through bone marrow aspiration; 2) fulfillment of either of the following conditions: i) newly diagnosed AML unfit for standard treatment (ND-unfit AML) or ii) R/R-AML; 3) sufficient sample collection via bone marrow aspiration; 4) Age of participants 20 years or above during registration; 5) provision of written informed consent by participants. Paraffin-embedded bone marrow samples were gathered from 17 Japanese faculties and the F1H reports were returned to the patients. The median turnaround time was 13 days (minimum 8 days). We found 13 patients (7.3%) with IDH1 mutation and 17 patients (9.6%) with IDH2 mutation out of 177 patients who joined this study and the F1H report was successfully returned. Only one patient had both mutations, and each mutation was mutually exclusive in all the other patients (Figure 1). The major amino acid alteration of IDH1 and IDH2 were R132C/G/H/L and R140Q/W, respectively. Frequently co-occurring mutations include FLT3 (44.8%), NPM1 (34.5%), DNMT3A (31.0%) and RUNX1 mutation (20.7%). Mutations of RAS pathway-related genes (e.g., KRAS, NRAS and PTPN11) were seen in 6 patients (20.7%). Any gene alterations didn't show statistically significant co-occurrence with IDH1 and IDH2 mutation. Serial genome profiling analyses were performed to evaluate the time-dependent changes in the genome profiles of patients administered FLT3 inhibitors, gilteritinib, and quizartinib for treating FLT3-mutated AML. Also in this cohort, we are examining the properties and distribution of IDH1/2 mutations during treatment with FLT3 inhibitors. In the several patients, expansion and persistence of IDH mutated clones seemed to be cause of resistance (Figure 2 as the representative result). The detailed clinical outcomes of AML patients with IDH1/2 mutations are under investigation. Conclusions: In our evaluation of the suitability of F1H for HM-SCREEN-Japan 01, we successfully identified IDH-1/2 mutation that can be used as therapeutic targets in AML, which have rarely been identified thus far.
