Abstract

After a part of stone walls was discovered during the construction of Takanawa Gateway station in 2019, the Takanawa Chikutei (embankment) was unearthed by the excavation in 2020. When the first Japanese railway was constructed at the beginning of the Meiji era, most of the materials necessary for the construction were imported from England, but stones and sediment for the foundation of bridges and embankments are considered to have been supplied within Japan. To understand the history of Japanese construction work, it is interesting to compare the composition of the joint material used at Takanawa Chikutei with that of joint materials made with conventional technology, and with artificial stones developed later in the Meiji era. In order to obtain indirect evidence of the use of slaked lime mixture and to calculate its composition ratio, the joint materials sampled from Takanawa Chikutei were analyzed by X-ray diffraction analysis and X-ray fluorescence analysis. As a result of the X-ray diffraction analysis, calcite was identified in 9 of 14 samples, providing the indirect evidence of the use of slaked lime mixture in the joint materials at Takanawa Chikutei. The composition ratio obtained by X-ray fluorescence analysis indicates that the content ratio of slaked lime in the joint materials at Takanawa Chikutei might be higher than that of artificial stones developed later in the Meiji era.

Abstract

Hematopoietic stem cell (HSC) function is regulated by activation of receptor tyrosine kinases (RTKs). Receptor protein tyrosine phosphatases (PTPs) counterbalance RTK signaling; however, the functions of receptor PTPs in HSCs remain incompletely understood. We found that a receptor PTP, PTPσ, was substantially overexpressed in mouse and human HSCs compared with more mature hematopoietic cells. Competitive transplantation of bone marrow cells from PTPσ-deficient mice revealed that the loss of PTPσ substantially increased long-term HSC-repopulating capacity compared with BM cells from control mice. While HSCs from PTPσ-deficient mice had no apparent alterations in cell-cycle status, apoptosis, or homing capacity, these HSCs exhibited increased levels of activated RAC1, a RhoGTPase that regulates HSC engraftment capacity. shRNA-mediated silencing of PTPσ also increased activated RAC1 levels in wild-type HSCs. Functionally, PTPσ-deficient BM cells displayed increased cobblestone area-forming cell (CAFC) capacity and augmented transendothelial migration capacity, which was abrogated by RAC inhibition. Specific selection of human cord blood [CD34.sup.+][CD38.sup.-][CD45RA.sup.-][lin.sup.-]PTP[σ.sup.-] cells substantially increased the repopulating capacity of human HSCs compared with [CD34.sup.+][CD38.sup.-][CD45RA.sup.-][lin.sup.-] cells and [CD34.sup.+][CD38.sup.-][CD45RA.sup.-] [lin.sup.-]PTP[σ.sup.+] cells. Abstract

Cutaneous syncytial myoepithelioma (CSM) is a recently recognized variant of myoepithelioma characterized by an intradermal syncytial proliferation of spindled, ovoid, and histiocytoid cells. Immunohistochemically, tumor cells usually show strong expression of S‐100 protein and epithelial membrane antigen (EMA). Here we report a case of CSM in the thigh of a 51‐year‐old Japanese woman. Histopathological findings showed a sheet‐like growth of ovoid cells and histiocytoid cells with an eosinophilic syncytial cytoplasm, and adipocytic metaplasia was widely observed in the tumor. Immunohistochemical staining revealed a diffuse, strong pattern for EMA, smooth muscle actin (SMA), and HHF35, and variable expression of S‐100 protein and p63 in ovoid and histiocytoid cells without significant mitotic figures or pleomorphism. In addition, EWSR1‐PBX3 gene fusion, which is characteristic of CSM, was observed in the tumor. Based on these findings, we diagnosed the patient as having CSM. Our case shows that CSM can exhibit extensive adipocytic metaplasia, which could make its histopathological diagnosis challenging. [ABSTRACT FROM AUTHOR]
