Multiple sclerosis is characterized by the destruction of myelin in the CNS. Various factors including genetics, epigenetics, and environmental factors are involved in the development of the disease. There is evidence that changes in the gut microbiome profile are associated with immune-related diseases such as MS. Probiotics can alter the composition of the gut microbiota on the mucosal surfaces by differentiating naive T cells into Th1, Th2, Th17, and Treg cells. Female C57BL/6 mice were divided into 6 groups (n = 7): Normal group, cuprizone group (gavage of cuprizone for 4 weeks), Probiotic group (gavage of probiotic for 4 weeks), Treatment1 group (Probiotic for 4 weeks and then cuprizone for 4 weeks), treatment2 group (cuprizone for 4 weeks and then probiotic for 4 weeks) and treatment3 group (cuprizone for 4 weeks and then probiotic for 4 weeks with vitamin D3). Then the expression of NLRP-1, NLRP-3, AIM2, and CYP27B1 genes were evaluated using Real-Time PCR, and serum levels of IFN-γ and IL-4 were also measured by ELISA.The results showed a significant decrease in the expression of inflammasome and CYP27B1 genes in the probiotic-treated groups compared to the cuprizone group. Also, the comparison between probiotic-treated groups and cuprizone group showed a significant decrease in the amount of IFN-γ and IL-4. Due to reduced expression of the inflammasome genes as well as the decrease in IFN-γ levels as an inflammatory cytokine, it appears that L. casei may be effective in the healing process of demyelinated mice., (Copyright © 2021 Elsevier Ltd. In this study, antibacterial, synthetic poly( Σ -caprolactone)-poly(ethylene oxide) (PCL-PEO) multilayer nanofibers were produced by an electrospinning method. The material was synthesized in 3 layers. The upper-lower protective layers were produced by PCL nanofibers and the intermediate layer was produced from PEO nanofiber containing sericin-capped silver nanoparticles (S-AgNPs). The electrospinning conditions in which nano-sized, smooth, bead-free fibers were obtained was determined to be an applied voltage of 20 kV, a flow rate of 8 μ L/min and a distance between the collector and the needle tip of 22 cm for the PCL layer (dissolved at a 12% g/mL concentration in a chloroform:methanol (3:2) solvent mixture) layer. For the S-AgNPs doped PEO layer (dissolved at a 3.5% g/mL concentration in water), the corresponding conditions were determined to be 20 kV, 15 μ L/min and 20 cm. To characterize the three-layer material that consisted of PCL and S-AgNPs doped PEO layers, FTIR and SEM analyses were performed, and the water retention capacity, in situ degradability and antibacterial activity of the material was investigated. According to SEM analysis, the fibers obtained were found to be nano-sized, smooth and bead-free and the average size of the nanofibers forming the PCL layer was 687 nm while the average size of the fibers forming the PEO layer was 98 nm. Antibacterial activity tests were performed using gram-positive ( Staphylococcus aureus ATCC 6538) and gram-negative ( Escherichia coli ATCC 25922) bacteria and the resulting biomaterial was found to have antimicrobial effect on both gram-negative and gram-positive bacteria. It was determined that the 3-layer material obtained in this study can be used as a wound dressing.

Multiple sclerosis (MS) is a complex inflammatory disease in which demyelination occurs in the central nervous system affecting approximately 2.5 million people worldwide. Recent reports have shown that the gut microbiome plays a crucial role in the functioning of the immune system in inflammatory diseases such as MS. In this study, the cuprizone-induced demyelination mouse model was used to investigate the effect of Lactobacillus casei strain T2 (IBRC-M10783) on the alleviation of these mice. Female C57BL/6 mice (8-10 weeks old) were divided into 6 groups: group 1, normal control; group 2, cuprizone control (oral administration of cuprizone 0.2% w/w for 4 weeks); group 3, probiotic control (oral administration of 1 × 10 9  CFU/ml probiotic for 4 weeks); group 4, treatment 1 (probiotic for 4 weeks then cuprizone for 4 weeks); group 5, treatment 2 (cuprizone for 4 weeks then probiotic for 4 weeks); and group 6, treatment 3 (cuprizone for 4 weeks then probiotic for 4 weeks with vitamin D 3 at a dose of 20 IU/day). Then, TGF-β and IL-17 were measured by ELISA, and the expression of miR-155, miR-25, and IDO-1 was evaluated by real-time PCR. Among the measured microRNAs, the results showed that there was a significant decrease in miR-155 expression between the treatment 1 group and the cuprizone group. In the case of IL-17, the results also showed a significant reduction between the three treatment groups and the cuprizone group. These observations suggest that L. casei can reduce proinflammatory cytokines and reduce demyelinating symptoms in the mouse model.

Hypericum perforatum oil loaded electrospun polymeric wound dressing material was produced in order to be used in wound therapy. H. perforatum oil is known to have curative effect on wound-healing process. Wound dressing material was produced in two layers, upper layer of which was made of electrospun PCL nanofibres in order to maintain membrane integrity and mechanical strength, and bottom layer that is designed to be in contact with the wound was formed by electrospraying and electrospining of PEG/H. perforatum oil and PCL polymer solutions from opposite directions (concurrently). Methods such as FTIR, optical and electron microscopy, tensile tests, gas permeability tests, contact angle and swelling tests, in vitro release tests were utilized for material characterization. Encapsulation of H. perforatum oil in PEG capsules which were hold by PEG fibres among PCL fibres was confirmed. H. perforatum oil was released in controlled manner. Antimicrobial activity tests on Staphylococcus aureus and Escherichia coli revealed that H. perforatum content exhibited antimicrobial activity on both. Material was found to be biocompatible and suitable for use as wound dressing according to the results of in vitro tests, in which L929 mouse fibroblast cell line incubated with materials for investigation of biocompatibility (WST-1) and cell-material interactions (proliferation, apoptosis/necrosis).

Background: There is an established link between obesity related metabolic derangement and colorectal cancer development. Recently, we developed a metabolic-colorectal cancer risk score. In this follow-up study, we studied its association with colorectal neoplasm by measuring two major metabolic syndrome biomarkers, leptin and adiponectin., Objectives: To evaluate the serum levels of leptin and adiponectin in patients with colorectal polyps and colorectal cancer and to determine any correlation with metabolic risk score., Results: In total, 130 individuals were studied: 30 controls without colonic pathology, 18 with colonic adenoma (CAP), and 82 with colorectal adenocarcinoma (CRC, 17 cases of T1-2 and 65 cases of T3-4). The metabolic risk scores in CAP and T1-2 CRC were higher than those in the controls and T3-4 CRC cases. There were no statistically significant differences in leptin levels among CAPs, CRCs, and controls. Both leptin and adiponectin levels reflected differences in body mass index and metabolic risk scores. Cases in the CAP group and early T-stage CRC groups had lower adiponectin levels (14.03 and 13.01 mg/ml, respectively) than the no polyps group (19.5mg/ml, p = 0.03). The average serum adiponectin level in the invasive cancer group (18.5 ng/ml) was comparable with that of the control group., Conclusions: The level of serum adiponectin was positively correlated with the metabolic risk score. Decreased serum adiponectin was significantly associated with the development of colorectal adenoma and early stage colorectal carcinoma.

Background: Many cancer patients still experience pain worldwide. There are many barriers for effective control of cancer pain and many of these are related to health care providers. There is a need for further investigation of these barriers. The aim of this study was to investigate nurse-related barriers to control of cancer pain among Iranian nurses., Materials and Methods: In this descriptive study 49 nurses from two hospitals affiliated to Tabriz and Ardebil Universities of Medical Sciences participated using a census sampling method. A demographic and profession related checklist and Barriers Questionnaire II (BQ-II) were used for data collection., Results: The results showed negative attitudes of participants regarding control of cancer pain. Participants believed that cancer pain medications do not manage cancer pain at acceptable levels; patients may become addicted by using these drugs; cancer pain medications have many uncontrollable effects; and controlling cancer pain may distract the physicians from treating disease., Conclusions: Iranian nurses have negative attitudes toward pain control in cancer patients especially about effectiveness of pain medication and their side effects. Educational intervention to reduce these misconceptions is needed.
