Las enfermedades causadas por Fusarium en Agave aumentan, lo cual significa importantes pérdidas económicas. Es necesario entonces buscar alternativas en la prevención y control de pató­genos para reducir el uso de fungicidas químicos, y así promo­ver un tipo de control de enfermedades que sea amigable con el ambiente. Los aceites y extractos de hojas de algunas plantas son eficientes en el control de plagas y enfermedades. Piper auritum (“hierba santa”) contiene aceites esenciales que cumplen funcio­nes ecológicas, como las de atraer polinizadores y causar efectos alelopáticos. Baccharis glutinosa (“jarilla”) también se usa en el tratamiento de diversas enfermedades infecciosas. Hay diferentes extractos de plantas que tienen una actividad biológica contra microorganismos que afectan cultivos importantes. El objetivo de esta investigación fue evaluar la actividad antimicrobiana del aceite esencial y extractos de dos plantas con uso tradicional en el estado de Chiapas sobre los hongos fitopatógenos (Fusarium spp.) de Agave spp. Los extractos de P. auritum y B. glutinosa se probaron contra los hongos fitopatógenos, como parte de un di­seño experimental completamente aleatorizado. Un ANOVA se realizó con seis tratamientos y tres repeticiones por tratamiento, utilizados en tres especies de Fusarium. El aceite esencial extraí­do de las hojas de Piper auritum tuvo el mayor efecto inhibi­dor significativo (96 h) sobre Fusarium oxysporum f. sp. comiteca (75.32%), Fusarium oxysporum f. sp. tequilana (86.57%) y Fu­sarium solani f. sp. comiteca (63.36%). Por lo tanto, P. auritum mostró un claro efecto fungistático.

Plant growth regulators naphthaleneacetic acid (NAA), 2,4-dichlorophenoxyacetic acid (2,4-D), 6-Benzylamino purine (BAP) were evaluated at different concentrations (0, 0.5, 1.0, and 2.0 mgL-l) in three different explants (leaf, stem, and root) on the induction of responses morphogenic. In the suspension cell culture establishment, callus obtained from the stem were used and two concentrations of coconut water (20 and 40% v/v) were evaluated to increase the disintegration of the cell suspension. Bonediol was quantified using the HPLC technique in callus obtained from different explants (leaf, stem, and root) and suspension cell cultures. Higher callus formation was obtained in stem explants, with BAP at 1.0 mgL-l indirect organogenesis was obtained and with 2,4-D at 0.5 and 1.0 mgL-l somatic embryogenesis. Bonediol concentration detected was 49.57 mg/g callus, derived from stem explant. During the cultivation of cells in suspension, greater disintegration of the compact calli was observed with 20% coconut water. Bonediol concentration in suspension cell cultures was 24.44 mg/g dry cell. The objective of this work was to evaluate the effect of growth regulators on callus formation as well as the quantification and determination of bonediol content in callus and suspension cells of Bonellia macrocarpa.

Moringa oleifera is a highly versatile plant with potential use in the agro-food and biochemical industry. The goals of this study were to evaluate the effect of chemical fertilization and vermicompost on plant growth, and to analyze the metabolomic profile of M. oleifera crops cultivated over agricultural and native soils. The extracts were obtained from 90-day-old leaves via extraction with a hydroalcoholic mixture. Multivariate data analyses, such as principal component analysis (PCA) and partial least square-discriminant analysis (PLS-DA), were used to differentiate the distribution of leaf metabolites according to the soils or types of fertilizers used for the cultivation of Moringa oleifera. The results indicated that there was no significant effect on parameters such as plant height, root length and dry weight of leaves (p &lt, 0.05). UPLC-ESI-MS/MS analysis of leaf extracts revealed a wide range of flavonoids, alkaloids and organic acids. The results of PCA and PLS-DA confirmed that the type of fertilizer had an effect on the metabolomic profile of M. oleifera leaves. The application of vermicompost induced changes in the metabolomic profile, but not in the morphometric variables of Moringa oleifera. These results are important for metabolite production via organic cultures and over different soil types in the industrialization of Moringa.

La quitinasa y B-1,3-glucanasa desempeñan una función importante en la reacción de defensa contra patógenos fúngicos. En Agave americana existe poca información sobre el comportamiento de estas proteínas en respuesta a la infección de Fusarium oxysporum. En este estudio, se realizó un análisis de microscopía electrónica de barrido (MEB) en raíces de A. americana infectadas con F. oxysporum y se evaluó la actividad de quitinasa y B-1,3-glucanasa en raíces de A. americana a 0, 7, 15 y 30 días después de inoculación con F. oxysporum. MEB reveló que 15 días después de la inoculación (DDI) son suficientes para que el hongo infecte las raíces de A. americana. La actividad de quitinasa en plantas infectadas incrementó significativamente a los 15 DDI con respecto a las plantas no infectadas, mientras que la actividad de B-1,3 glucanasa no presentó diferencia estadística significativa con plantas testigo. Los resultados sugieren que, en respuesta a la infección por F. oxysporum, A. americana activa proteínas PRs de tipo quitinasas.

Fusarium are considered as the major plant pathogen fungi, that cause the majority of soil-borne diseases to more than 100 plant species in the world, including maize. Thus, there are emerging demands of biocontrol reagents, and Crotalaria longirostrata showed fungicidal activity. The C. longirostrata branch extract was phytochemically characterized and evaluated for efficacy for the control of Fusarium wilt in maize. The application of the extract reduced the percentage of disease incidence significantly caused by Fusarium verticillioides from 70.4% to 40.12% as compared to non-treated plants, and evenly the disease severity was reduced from 40.15% to 29.46%. The phytochemical components of the extract were cinnamic acids (caffeic acid and ferulic acid) and phenolic acid (gallic acid). Furthermore, multiple structures were detected through mass spectrometry such as: phenols, alkaloids, esters, terpene, ketones, and amides. The bioautography assay showed that to separate the compounds of C. longirostrata branch extract causes it&rsquo, s the loss of fungicidal activity. This is due to the synergy or additive interactions of secondary metabolites present in the raw extract. Our results suggest that the application of C. longirostrata branch extract is a promising strategy to be applied to the soil as a preventive treatment.

Las plantas utilizan diversos mecanismos para defenderse del ataque de hongos patogenos. Algunos de estos mecanismos incluyen el aumento de metabolitos secundarios, antioxidantes, enzimas hidroliticas y polisacaridos que causan la muerte o reducen la progresion de hongos patogenos. Hemileia vastatrix (roya del cafe) es un hongo biotrofo que causa graves danos a las plantaciones de cafe. Ocasionalmente, se pueden observar plantas con ausencia de sintomas. El estudio de estas plantas asintomaticas es de suma importancia para la conservacion y mejoramiento del germoplasma resistente a H. vastatrix . Para el estudio, aquellas plantas que durante la incidencia de la enfermedad no presentaban sintomas fueron denominadas no infectadas (Ni) y las que si los presentaban como infectadas (i). Se evaluaron dos periodos: antes de la infeccion por roya y durante esta, en plantas de Coffea arabica Ni e i cultivadas en campo bajo un sistema de produccion organico. Olive trees are one of the most important oil crops in the world due to the sensorial and nutritional characteristics of olive oil, such as lipid composition and antioxidant content, and the medicinal properties of its leaves. In this paper, callus formation was induced using nodal segments of olive tree (Olea europaea cv. cornicabra) as explants. Fatty acid profile, total phenolic compounds and total flavonoid compounds were determined in callus culture after 15 weeks and compared with leaf and nodal segments tissues. There was no statistical difference in phenolic compounds among leaf, nodal segments and callus culture, whereas flavonoid compounds were higher in leaf. Fatty acid profile was similar in leaf, nodal segments and callus culture and was constituted by hexadecanoic acid, octadecanoic acid, cis-9-octadecenoic acid, cis-9,12-octadecadienoic acid, cis-9,12,15-octadecatrienoic acid. Hexadecanoic acid was the main fatty acid in callus, leaf and nodal segments with 35.0, 39.0 and 40.0% (w/w), of the lipid composition, respectively. With this paper, it is being reported for the first time the capacity of callus culture to accumulate fatty acids. Our results could serve to continue studying the production of fatty acids in callus cultivation as a biotechnological tool to improve different olive cultivars.

K+ is widely used by plant cells, whereas Na+ can easily reach toxic levels during plant growth, which typically occurs in saline environments; however, the effects and functions in the chloroplast have been only roughly estimated. Traditionally, the occurrence of ionic fluxes across the chloroplast envelope or the thylakoid membranes has been mostly deduced from physiological measurements or from knowledge of chloroplast metabolism. However, many of the proteins involved in these fluxes have not yet been characterized. Based on genomic and RNA sequencing (RNA-seq) analyses, we present a comprehensive compilation of genes encoding putative ion transporters and channels expressed in the chloroplasts of the moss Physcomitrella patens, with a special emphasis on those related to Na+ and K+ fluxes. Based on the functional characterization of nhad mutants, we also discuss the putative role of NHAD transporters in Na+ homeostasis and osmoregulation of this organelle and the putative contribution of chloroplasts to salt tolerance in this moss. We demonstrate that NaCl does not affect the chloroplast functionality in Physcomitrella despite significantly modifying expression of ionic transporters and cellular morphology, specifically the chloroplast ultrastructure, revealing a high starch accumulation. Additionally, NHAD transporters apparently do not play any essential roles in salt tolerance.

The essential oils of plants of the genus Piper have secondary metabolites that have antimicrobial activity related to their chemical composition. The objective of our work was to determine the chemical composition and evaluate the antifungal activity of the aerial part essential oil of P. auritum obtained by hydrodistillation on Fusarium&nbsp, oxysporum and Fusarium equiseti isolated from Capsicum chinense. The antifungal activity was evaluated by direct contact and poisoned food tests, and the minimum inhibitory concentration (MIC50) and maximum radial growth inhibition (MGI) were determined. The identification of oil metabolites was carried out by direct analysis in real time mass spectrometry (DART-MS). By direct contact, the essential oil reached an inhibition of over 40% on Fusarium spp. The 8.4 mg/mL concentration showed the highest inhibition on F. oxysporum (40–60%) and F. equiseti (&gt, 50%). The MIC50 was 6 mg/mL for F. oxysporum FCHA-T7 and 9 mg/mL for F. oxysporum FCHJ-T6 and F. equiseti FCHE-T8. DART-MS chemical analysis of the essential oil showed [2M-H]− and [M-H]− adducts of high relative intensity that were mainly attributed to eugenol and thymol/p-cimen-8-ol. The findings found in this study show a fungistatic effect of the essential oil of P. auritum on Fusarium spp.

The aim of this study was analyze the effect of jasmonic acid (JA) and abscisic acid (ABA) as elicitors on fatty acids profile (FAP), phenolic compounds (PC) and antioxidant capacity (AC) in callus of Thevetia peruviana. Schenk & Hildebrandt (SH) medium, supplemented with 2 mg/L 2, 4-dichlorophenoxyacetic (2, 4-D) and 0.5 mg/L kinetin (KIN) was used for callus induction. The effect of JA (50, 75 and 100 μM) and ABA (10, 55 and 100 μM) on FAP, PC and AC were analyzed using a response surface design. A maximum of 2.8 mg/g of TPC was obtained with 100 plus 10 µM JA and ABA, respectively, whereas AC maximum (2.17 μg/mL) was obtained with 75 plus 100 µM JA and ABA, respectively. The FAP was affected for JA but not for ABA. JA increased cis-9, cis-12-octadecadienoic acid and decreased dodecanoic acid. Eight fatty acids were identified by GC-MS analysis and cis-9-octadecenoic acid (18:1) was the principal fatty acid reaching 76 % in treatment with 50 μM JA plus 55 μM ABA. In conclusion, JA may be used in T. peruviana callus culture for obtain oil with different fatty acids profile.

The objective of this study was to evaluate the changes of green metallic nanoparticles on biochemical and physiological effects in Annona muricata L., plants. Changes in proline, phenolic compounds, antioxidants enzyme, phenylalanine ammonia lyase (PAL) activity and phytohormones in A. muricata leaves exposed to Ag, Cu and Cu/Ag phytonanoparticles were evaluated up to 120 h. The results showed that the foliar application of green metallic nanoparticles at 250, 500, 750 and 1000 ppm caused changes significantly in flavonoids, anthocyanins, phenylalanine ammonia lyase and peroxidase activities in A. muricata leaves after 120 h exposure. The foliar application of green nanoparticles stimulates the enhancement of the antioxidant protection system that protect at A. muricata against oxidative stress caused by reactive oxygen species (ROS). Our results showed that values of gibberelins, auxins and proline depended of nanoparticle concentration and composition.

The use of microbial inoculants is an effective biotechnological alternative to increase crop productivity. In this work we studied the diversity of the Plant Growth Promoting Bacteria (PGPB) associated with pioneer plants that grow at the surroundings of the crater-lake of El Chichon volcano. Cheilanthes aemula (Pteridaceae) and Andropogon glomeratus (Poaceae) plants were dominant in this area. A total of 80 endophytes and 128 strains from the rhizosphere were isolated using selective culture media, and then characterized phenotypically. Isolates were tested for growth in high salt concentrations, at different pH and in moderate concentrations of heavy metals. The genetic diversity and identity of the strains were revealed by BOX_PCR and by sequencing of 16S rRNA gene. A. glomeratus isolates were classified within the genera Brevibacillus, Sinomonas, Sphingobium, Chryseobacterium, Pantoea, Ralstonia, Burkholderia, Serratia, Bacillus, Sphingomonas and Methylobacterium using 16S rDNA gene sequence analysis. C. aemula isolates were grouped into the genera Dyella, Burkholderia, Microbacterium, Enterobacter, Exiguobacterium, Pandoraea, Methylobacterium, Kocuria and Agrobacterium. The rhizospheric and endophytic strains were distinguished by their ability to produce different extracellular enzymes. The extremophile isolates had the ability to produce indole compounds (IAA), solubilize phosphate, synthesize siderophores, ACC deaminase and nitrogenase and showed a positive effect on the growth of Phaseolus vulgaris and upon the total content of nitrogen and phosphorus of plants. The diversity of bacteria associated to the pioneer plants had multifunctional qualities as PGPB that may contribute to their adaptation and proliferation at this active volcano and be used efficiently as biofertilizers to promote plant growth in organic farming systems

The genus Crotalaria comprises about 600 species that are distributed throughout the tropics and subtropical regions of the world; they are antagonistic to nematodes in sustainable crop production systems, and have also shown antimicrobial capacity. Chipilin (C. longirostrata), which belongs to this genus, is a wild plant that grows in the state of Chiapas (Mexico) and is traditionally is used as food. Its leaves also have medicinal properties and are used as hypnotics and narcotics; however, the plant has received little research attention to date. In the experimental part of this study, dried leaves were macerated by ethanol. The extract obtained was fractionated with ethyl ether, dichloromethane, ethyl acetate, 2-propanone, and water. The extracts were evaluated against three bacteria—namely, Escherichia coli (Ec), Citrobacter freundii (Cf), and Staphylococcus epidermidis (Se)—and three fungi—Fusarium oxysporum A. comiteca (FoC), Fusarium oxysporum A. tequilana (FoT), and Fusarium solani A. comiteca (FSC). During this preliminary study, a statistical analysis of the data showed that there is a significant difference between the control ciprofloxacin (antibacterial), the antifungal activity experiments (water was used as a negative control), and the fractions used. The aqueous fraction (WF) was the most active against FoC, FsC, and FoT (30.65, 20.61, and 27.36% at 96 h, respectively) and the ethyl ether fraction (EEF) was the most active against Se (26.62% at 48 h).

High-affinity K+ (HAK) transporters are encoded by a large family of genes and are ubiquitous in the plant kingdom. These HAK-type transporters participate in low- and high-affinity potassium (K+) uptake and are crucial for the maintenance of K+ homeostasis under hostile conditions. In this study, the full-length cDNA of CcHAK1 gene was isolated from roots of the habanero pepper (Capsicum chinense). CcHAK1 expression was positively regulated by K+ starvation in roots and was not inhibited in the presence of NaCl. Phylogenetic analysis placed the CcHAK1 transporter in group I of the HAK K+ transporters, showing that it is closely related to Capsicum annuum CaHAK1 and Solanum lycopersicum LeHAK5. Characterization of the protein in a yeast mutant deficient in high-affinity K+ uptake (WΔ3) suggested that CcHAK1 function is associated with high-affinity K+ uptake, with Km and Vmax for Rb of 50 μM and 0.52 nmol mg−1 min−1, respectively. K+ uptake in yeast expressing the CcHAK1 transporter was inhibited by millimolar concentrations of the cations ammonium (NH4+) and cesium (Cs+) but not by sodium (Na+). The results presented in this study suggest that the CcHAK1 transporter may contribute to the maintenance of K+ homeostasis in root cells in C. chinense plants undergoing K+-deficiency and salt stress.

Rex, Chichen-Itza and Naranja and Capsicum annuum L. cv. Padron), considering primary root growth and viability of the post-stressed seedlings. NaCl-induced K+ and H+ efflux in roots was also studied by ion-selective microelectrodes under application of pharmacological agents. In these pepper varieties, the magnitude of the K+ leakage in the roots positively correlated with growth inhibition of the primary root in the presence of NaCl, with Rex variety showing a higher level of tolerance than Chichen-Itza. The K+ leakage and the activity of the H+ pump in the roots were dependent on the NaCl concentration. Pharmacological analysis indicated that the NaCl-induced K+ leakage was mediated by TEA+-sensitive KOR channels but not by NSCC channels. In addition, we present evidence for the possible participation of proline, and a Na+-insensitive HAK K+ transporter expressed in habanero pepper roots for maintaining K+ homeostasis under salt stress conditions.

The pungency of chili peppers is conferred by compounds called capsaicinoids that are produced only in the fruits of the Capsicum genus. Accumulation of capsaicinoids in these fruits may be affected by environmental conditions such as water and nutrient stresses, although these effects may vary even among genotypes within a species. The Habanero pepper (Capsicum chinense Jacq.), grown in the Yucatán, is in especially high demand as a result of its unique flavor, aroma, and pungency and is the second most important commercial crop in the state after the tomato. Although the Habanero pepper is a significant economic resource for the region, few studies have investigated the effects of abiotic stresses on capsaicinoid production. In this study, the effects of water stress on plant growth, capsaicinoid accumulation, and capsaicin synthase activity were evaluated. Habanero pepper plants under water stress had a lower height, root dry weight, and root/shoot relation than control plants, which were irrigated daily. However, fruit growth and production were unaffected by water stress. Capsaicin and dihydrocapsaicin concentrations increased in fruits of stressed plants compared with control plants, and this effect was correlated with fruit age. However, capsaicin synthase activity was reduced in response to water stress, and this effect depended on both stress severity and fruit age. These results provide new information on the regulation of capsaicinoid metabolism in response to abiotic stress from the fruit of a highly pungent chili pepper.

Habanero pepper (Capsicum chinense Jacq.) is a very important crop in Mexico and demand for it is increasing in national and international markets. The habanero pepper produced on the Yucatan Peninsula is considered of superior quality to that grown in the rest of the world as a result of its shelf life and pungency. Despite its importance, little research has been done on cultivation conditions that may affect its productivity and fruit quality. The effect of N or K fertilization on habanero pepper development and fruit pungency was evaluated. Plants under fertilization stress (control) had high capsaicin content. Nitrogen fertilization significantly increased plant growth and fruit while maintaining high capsaicin levels. Optimum response was produced with 15 mm urea as the N source. Potassium fertilization had no positive effects on growth or productivity. The N treatments modified endogenous K levels in the pepper plants and vice versa. The K : N ratio (specifically in leaves and roots) varied between treatments with values greater than 1 in the K treatments, near 0.5 in the control, and less than 0.5 in the N treatments. This parameter may be an important indicator of habanero pepper productivity and requires study under different fertilization regimes.

Dynamics and distribution of K, Ca, Mg, P, and Na were studied in leaves of three cultivars of habanero pepper (Capsicum chinense Jacq.) at the time of transplanting and 8 and 14 weeks afterward. Most nutrients analyzed were mobilized from younger to mature leaves, except for P, which occurred in the opposite direction, probably due to its role in the synthesis of nucleic acids, which is more active in young tissues. Information about mineral distribution in leaves during the fi rst 14 weeks after transplantation could be used to indicate plant nutritional status and fertilizer requirements. Peppers (Capsicum spp.) are agricultural products of great demand in international markets, either as fresh or dried fruit, or as an industrial supply to obtain a variety of end-products. Habanero pepper has acquired considerable importance due to its great genetic diversity and the high pungency of its fruit. As in other peppers, pungency in the habanero de- pends on environmental factors, including soil properties such as texture, osmotic properties, and nutrient availability, which had been poorly studied (Soria-Fregoso et al., 2002). Nutrient mobilization rates have been cal- culated for many species of conifers and for an extensive group of perennial trees (Chapin and Kedrowski, 1983; Helmisaari, 1992; Kill- ingbeck, 1996; Loneragan, 1997; Nambir and Fife, 1987; Wendler et al., 1995). Nevertheless, there is insuffi cient comparable data for the Capsicum genus. There are just two reports on mineral supplementation in Capsicum an- nun—one on improving Jalapeno pungency level, plant growth, and pot yield (Johnson and Decoteau, 1996) and the other on study- ing the effects of fertilizer supplementation on fruit development and pungency in Padron peppers (Estrada et al., 1998)—and only one on C. annum in which the authors reported that sweet pepper possesses characteristics for well-controlled recirculation of sodium (Blom-Zandstra et al., 1998). Capsaicinoids, the chemical compounds that confer the pungency trait to peppers, are accumulated at different levels in all species of the genus Capsicum. There is much evidence suggesting that the synthesis of capsaicinoids occurs in the placenta interlocular septum of pepper fruits; however, the exact localization of the capsaicinoids biosynthesis accumulation pathways is still under debate. Thus, the aim of the present work was to evaluate whether pepper plants synthesize or accumulate capsaicinoids in vegetative organs as an indirect way to elucidate the systemic regulation of the capsaicinoid biosynthesis. For that purpose, we studied habanero pepper grown in the Yucatan Peninsula, which is among the hottest pepper worldwide. Our results, obtained by chromatographic and enzymatic measurements, provide solid evidence that habanero pepper plants do not accumulate capsaicinoids in the vegetative organs analyzed, even under water stress conditions. Thus, it is probable that the accumulation of capsaicinoids is restricted to reproductive organs.
