Abstract

AbstractGlycyrrhizin (GL) is a triterpenoid saponin that is commonly isolated from root of Glycyrrhiza glabra L. an indigenous medicinal and culinary herb. The present study focused on detecting and optimizing isolation method to isolate GL from successive root extracts of G. glabra L. in its purest form with high yield. The study exploited cold percolation extraction and variety of chromatographic methods such as HPTLC, HPLC and gradient column chromatography. The comparative HPTLC densitometry identified GL as a single grey color band at RF 0.36 under UV-254nm in successive extracts. The optimized extraction-isolation method isolated the in-house GL from 70% aqueous ethanol extracts using standardized eluent, i.e., ethyl acetate-methanol (7:3, v/v) and confirmed by Co-TLC. The compound obtained as colorless non sticky solid, with a yield 0.4%. Various spectroscopic tools characterized isolated GL along with HPLC peak detection, this confirmed its identity by matching with reported literature. The proposed extraction–isolation method could be utilized to enhance the efficacy of herbal pharmaceuticals, development of advanced Ayurvedic formulations and uplifting the quality of food products, and cosmeceuticals contain GL as active constituents. Additionally, this work established a simple, fast, and cost-effective HPTLC method that may also be applied to detect GL to establish genuineness of such raw drugs, identification of Ayurvedic dosages, and studying such triterpenoid saponins in other medicinal species belonging Glycyrrhiza for routine quality control. Shyonaka (Oroxylum indicumVent) is widely used in Ayurveda and in ethnomedical practice for the treatment of inflammation, pain, diarrhea, non-healing ulcers, and cancer. Owing to the high prevalence of Epstein-Barr virus (EBV) infection in Nasopharyngeal carcinoma (NPC) patients, simultaneous targeting of proteins involved in both EBV replication and NPC proliferation might help to manage the disease effectively.

22. HPTLC detection, extraction - isolation optimization and characterization of in-house isolated Wedelolactone from the whole plant extract of Eclipta prostrata (L.) L. for the development of efficacious formulations.

Abstract

In present study Wedelolactone (WL) was identified from successive extracts of whole plant Eclipta prostrata (L.) L. using HPTLC, optimised extraction/isolation method and confirmed WL structure.Extraction, isolation andidentification was performed though cold-percolation method, column chromatography and TLC respectively.WL was found abundantly in ethyl acetate and ethanol extracts at UV254 & 366 nm, respectively at R F 0.42 by using toluene:ethyl acetate: formic acid (6:3.5:0.5, v/v/v). In the Method IV got a pure pale green crystals (0.138 g, % yield of 0.23) was adopted. Isolated compound verified by TLC and the structure confirmed by various spectroscopic tools. The study proposed high efficiency extraction-isolation method for WL, which could enhance the efficacy for identification of E. prostrata, studying such coumestan in extracts and herbal formulations. Study potentially aiding in developing advance anti-cancer and hepatoprotectivedosages and quality control.
