Introduction. The article covers the issue of professional selection and determination of the set of qualities that should be possessed by the specialists of “person – machines” field, in particular, workers of locomotive brigades. The issue of assessing the professional suitability of operators of locomotives in Russia was raised in the late XIX century by railway engineers-administrators. However, the study of professionally important qualities of the members of the locomotive crews proves to be relevant today. The practical importance of corresponding research is related to ensuring the safety of trains movement. Materials and Methods. The study incorporated a combination of methods: literature analysis, testing (methods for diagnostics of level of personal risk readiness “PSK” R. Schubert, test “A study of volitional self-regulation” A. V. Zverkova, E. V. Eydman, methodology for perceptual assessment of stress), the survey (questionnaire). Spearman rank correlation method was used for statistical analysis of empirical data. Results. The article describes the results of empirical research of peculiarities of professionally important qualities of locomotive brigade workers. In particular, the authors introduce the data of diagnostics of volitional self-regulation and stress resistance of locomotive drivers and assistant drivers, and also results of measuring their risk tolerance, considered as an anti-PVK. It was revealed that the majority of the tested workers of locomotive brigades demonstrate low and medium levels of personal readiness to take risks, they are not inclined to act rashly and randomly. The majority of drivers and their assistants have a high level of volitional self-regulation development, i.e., they are emotionally mature, active, independent and self-reliant. Three quarters of the surveyed workers from the locomotive crews demonstrated a high level of perseverance and an overwhelming majority showed highly developed self-control. More than a half of the sample turned out to be representatives of the B type stress, i. e., stress-resistant people. However, more than a third of the members of locomotive crews tend to exhibit A type stress. They showed manifestations of aggressiveness, impatience, and intolerance to stress. The article contains the analysis of the survey of the locomotive staff members concerning the qualities required for the representatives of their profession. Most frequent feedback among the responses was about volitional traits expressing attitude to work and to other people. The survey revealed the particular importance of fostering some psychological qualities of the locomotive drivers and their assistants: patience, attentiveness, responsibility, determination, self-confidence, vigilance, diligence. Correlation analysis of empirical data using Spearman’s rank correlation determined the presence of statistically significant connection of volitional self-regulation and self-control with the types of stress resistance. There is an average force feedback between the indicated qualities of workers of locomotive brigades. The data of correlation analysis and test logic, used to measure stress resistance, suggest that higher level of volitional self-regulation development and self-control as its component inevitably boost better stress resistance of locomotive crew members. Discussion and Conclusion. Efficiency of professional activities of locomotive brigade members of a depot can be bettered by means of training exercises, carried out by a specialist of psychological profile, involving locomotive drivers and their assistants. The exercises should be targeted at the development of volitional self-regulation, and its element self-control, thus contributing to stress resistance improvement.

Introduction: the main purpose of this paper is to present the theoretical and practical aspects of professional and personal self-development of future professionals, the construction of the structural model and the determination of levels of self-development. The study of personal and professional self-development attributes of future professionals is of special interest to modern researchers, as well as for practicing psychologists studying the problem of self-realization and self-improvement in their professional fields. This article discusses the theoretical approaches to the study of the model and structure of personal and professional self- development. It analyses the results of the comparative empirical study of the features of students self-development with various levels of formation of motivational-semantic sphere. The authors carried out the detailed theoretical analysis of the problem of personal and professional self-development. They offer the structural model of the process, including the set of interrelated value-semantic, reflexive and regulatory components. The study revealed that the personal and professional self-development is a comprehensive system based on the active personal transformation being carried out in the course of behavior and activities self-regulation of their behavior, designed to achieve personal and professional goals. Materials and Methods: diagnostic methods were used to identify the level of self-development and personal qualities of students. Processing of the data was carried out by means of correlation, factor and cluster analysis. Results: this article considers the results of the empirical study conducted with the help of psycho-diagnostic methods aimed at identifying the level of self-development of future professionals, its structure and features. The scientifically valid results are obtained, allowing to identify the levels of self-development of students as well as to identify factors af fecting the process of self-development of future social sphere specialists. Discussion and Conclusions: the structural model of the personal-professional self-development includes a set of interconnected components: value-semantic, reflexive and regulative. The main factor influencing the process of personal and professional self- development of future professionals is the indicator of the meaningfulness of life, which is significantly associated with readiness to self-knowledge and selfdevelopment and affect the level of self-development. The desire for self-development can be considered as one of the personal and professional values, which give future professionals the opportunity to feel the meaningfulness of life, satisfaction with the process and its results, and achieve their personal and professional goals. The results of this study have several important implication for practicing psychologists who can broaden the scope of research of self-development and self- identity processes and determine the basic ways of self-realization of the young people potential in their prof essional career.

Background: Poly(hydroxyalkanoates) (PHA) have recently attracted increasing attention due to their biodegradability and high biocompatibility, which makes them suitable for the development of new prolong drug formulations. Objective: This study was conducted to develop new prolong paclitaxel (PTX) formulation based on poly(3- hydroxybutyrate) (PHB) microparticles. Method: PHB microparticles loaded with antitumor cytostatic drug PTX were obtained by spray-drying method using Nano Spray Dryer B-90. The PTX release kinetics in vitro from PHB microparticles and their cytotoxity on murine hepatoma cell line MH-22a were studied. Microparticles antitumor activity in vivo was studied using intraperitoneally (i.p.) transplanted tumor models: murine Lewis lung carcinoma and xenografts of human breast cancer RMG1. Results: Uniform PTX release from PHB-microparticles during 2 months was observed. PTX-loaded PHB microparticles have demonstrated a significant antitumor activity versus pure drug both in vitro in murine hepatoma cells and in vivo when administered i.p. to mice with murine Lewis lung carcinoma and xenografts of human breast cancer RMG1. Conclusion: The developed technique of PTX sustained delivery from PHB-microparticles has therapeutic potential as prolong anticancer drug formulation.

Ribonucleases (RNases) are a non-mutagenic alternative to harmful DNA-damaging anticancer drugs. Targeting of RNases with antibodies to surface antigens that are selectively expressed on tumor cells endows specificity to the cytotoxic actions of RNases. Barnase, a ribonuclease from Bacillus amyloliquefaciens, is a promising candidate for targeted delivery to cancer cells because of its insusceptibility to the ubiquitous cytoplasmic ribonuclease inhibitor, and its high stability and catalytic activity. Here, we characterized in vitro and in vivo an immunoRNase, scFv 4D5-dibarnase, which consists of two barnase molecules that are fused serially to the single-chain variable fragment (scFv) of humanized 4D5 antibody. The latter is directed against the extracellular domain of human epidermal growth factor receptor 2 (HER2), a cancer marker that is overexpressed in many human carcinomas. The scFv 4D5-dibarnase exerted a specific cytotoxic effect on HER2-overexpressing SKBR-3 and BT-474 human breast carcinoma cells (IC(50) = 4.1 and 2.4 nM, respectively) via induction of apoptosis. Ten doses of 0.7 mg/kg scFv 4D5-dibarnase to BALB/c nude mice that bore SKBR-3 human breast cancer xenografts resulted in a 76% reduction in tumor growth. A single injection of scFv 4D5-dibarnase at a total course dose of 7 mg/kg did not cause severe side effects in BALB/c nude or BDF1 mice. The cytotoxicity and selectivity of scFv 4D5-dibarnase merit consideration of this immunoRNase as a potent anticancer agent.
