An unprecedented extension of life expectancy observed during the past century drastically increased the number of patients diagnosed with Parkinson's diseases (PD) worldwide. Estimated costs of PD alone reached $52 billion per year, making effective neuroprotective treatments an urgent and unmet need. Current treatments of both AD and PD focus on mitigating the symptoms associated with these pathologies and are not neuroprotective. In this review, we discuss the most advanced therapeutic strategies that can be used to treat PD. We also critically review the shift of the therapeutic paradigm from a small molecule-based inhibition of protein aggregation to the utilization of natural degradation pathways and immune cells that are capable of degrading toxic amyloid deposits in the brain of PD patients. Advancements in chimeric antigen receptor engineered T-cell (CAR-T) therapy have revolutionized treatment for several cancer types over the past decade. Despite this success, obstacles including the high price tag, manufacturing complexity, and treatment-associated toxicities have limited the broad application of this therapy. Chimeric antigen receptor engineered natural killer cell (CAR-NK) therapy offers a potential opportunity for a simpler and more affordable “off-the-shelf” treatment, likely with fewer toxicities. Unlike CAR-T, CAR-NK therapies are still in early development, with few clinical trials yet reported. Given the challenges experienced through the development of CART therapies, this review explores what lessons we can apply to build better CARNK therapies. In particular, we explore the importance of optimizing the immunochemical properties of the CAR construct, understanding factors leading to cell product persistence, enhancing trafficking of transferred cells to the tumor, ensuring the metabolic fitness of the transferred product, and strategies to avoid tumor escape through antigen loss. We also review trogocytosis, an important emerging challenge that likely equally applies to CAR-T and CAR-NK cells. Finally, we discuss how these limitations are already being addressed in CAR-NK therapies, and what future directions may be possible. [ABSTRACT FROM AUTHOR]

To explore ability of Fe3O4-PEG-CD56/Avastin @ Ce6 targeted probe to bind to NK92 cells and perform MRI to cells in vitro. The nanoprobes of Fe3O4-PEG-CD56/Avastin@Ce6 were prepared and were characterized. Then the cytotoxicity of this nano-materials to NK92 was determined by apoptotic kit and the cellular uptake of nanomaterials to NK92 cells was analyzed by flow cytometry. Finally, MRI was used to research the signal intensity of cells. The nanoprobes with good biocompatibility were synthesized successfully. The cytotoxicity of the nanoprobe was low and the apoptosis levels were basically the same at different concentrations for NK92 cells. Flow cytometry showed that the nanomaterials bound to NK92 cells increased with the raised concentration. MRI showed that the signal of T2-weighted image (T2WI) was declined significantly with the rise of Fe3O4-PEG-CD56/Avastin@Ce6 concentrations. The Fe3O4-PEG-CD56/Avastin@Ce6 nanoprobe has potential to targeting NK92 cells and can monitor using the 3.0T MR scanner in vitro. [ABSTRACT FROM AUTHOR]

The liver is considered a preferential tissue for NK cells residency. In humans, almost 50% of all intrahepatic lymphocytes are NK cells that are strongly imprinted in a liver-specific manner and show a broad spectrum of cellular heterogeneity. Hepatic NK (he-NK) cells play key roles in tuning liver immune response in both physiological and pathological conditions. Therefore, there is a pressing need to comprehensively characterize human he-NK cells to better understand the related mechanisms regulating their effector-functions within the dynamic balance between immune-tolerance and immune-surveillance. This is of particular relevance in the liver that is the only solid organ whose parenchyma is constantly challenged on daily basis by millions of foreign antigens drained from the gut. Therefore, the present review summarizes our current knowledge on he-NK cells in the light of the latest discoveries in the field of NK cell biology and clinical relevance.
