Nowadays pulmonary embolism is the second most common cause of death for cancer patients. The reason for this is activation of the blood coagulation system, depression of the coagulation system and fibrinolysis, decrease in the linear velocity of blood flow, phlebohypertension, as well as varicose reorganization of the vascular wall, its valves and endothelial cells. However, structural changes of veins as predictors of vein thrombosis in cancer patients are not sufficiently covered. At the same time, knowledge of these changes is important for understanding pathogenesis and prevention of thromboembolic complications. Objective: to find out the peculiarities of the restructuring of the structural components of the venous wall as a source of possible primary formation of intravascular thrombosis under conditions of oncogenic pathology. Material and methods. Histologic, submicroscopic and polarization data of the study of hind limb veins of 12 sexually mature non-linear rats - males weighing 170-180 g for 30 days of chronic neoplastic intoxication and vein fragments of 12 patients with colon cancer complicated by thrombosis were analyzed. Chronic neoplastic intoxication was simulated by subcutaneously injecting 7.2 mg / kg (based on the active substance) subcutaneously into the interscapular area once a week for 30 weeks, according to the weight of the animal at the rate of 0.1 ml of dimethylhydrazine hydrochloride solution (DMH). 10 grams of the body weight of rat 1,2-DMH (from SIGMA-ALDRICH CHEMIE, made in Japan, series D161802) pre-diluted with isotonic sodium chloride solution. Results and Discussion. Morphological studies of hind limb veins in experimental neoplastic intoxication mainly concerned submicroscopic reorganization of the endothelial cells and their desquamation, as well as platelet sweetening, which may be one of the links in pathogenesis of thrombus formation. Histological and electron microscopic examination of biopsy of the veins of patients with colon cancer found disorders of laminar intima, desquamation of endothelial cells, sclerosis of all membranes of the deep vein and neoplasm of vessels in the middle membrane. Patients with cancer are considred to have dystrophic-necrotic changes of skeletal muscle, which is one of the links in pathogenesis of hemodynamic disorders in the venous system of the lower extremities. Conclusions. The features of restructuring of the structural components of the venous wall, as a source of possible primary formation of intravascular thrombosis under conditions of oncogenic pathology include focal loss of integrity of the endothelial layer, endothelial dysfunction, sweetening of platelets, fibrotic remodeling of tunica intima, tunica media, tunica adventitia, neovascularization of tunica media, dystrophic and sclerotic changes of the skeletal muscle., На сегодняшний день тромбоэмболия легочной артерии является второй по частоте причиной смерти пациентов онкологического профиля. Причиной ее является активации свертывающей потенциала крови, депрессии противосвертывающей системы и фибринолиза, снижение линейной скорости тока крови, флебогипертензии, а также варикозной реорганизации сосудистой стенки, ее клапанов и эндотелиоцитов. Однако структурные изменения вен, как предикторы тромбоза вен у больных раком, освещены недостаточно. Вместе с тем знание этих изменений важно для понимания патогенеза и предупреждения тромбоэмболических осложнений. Цель: выяснить особенности перестройки структурных компонентов венозной стенки, как источники возможного первичного формирования внутрисосудистого тромбообразования в условиях онкогенного патологии. Материал и методы. Обработано гистологические, субмикроскопические и поляризационные данные исследования вен задних конечностей 12 половозрелых нелинейных крыс-самцов массой тела 170-180 г на 30 день хронической неопластическом интоксикации и фрагменты вен 12 больных раком толстой кишки, осложненного тромбозом. Хроническую неопластическая интоксикацию моделировали путем введения подкожно в межлопаточную область в дозе 7,2 мг / кг (в расчете на действующее вещество) 1 раз в неделю в течение 30 недель, согласно массы животного из расчета 0,1 мл раствора диметилгидразин гидрохлорида (ДМГ) на 10 грамм массы тела крысы 1,2-ДМГ (фирмы SIGMA-ALDRICH CHEMIE, производства Японии, серия D161802), предварительно разведенного физиологическим раствором натрия хлорида. Результаты и обсуждение. Морфологические исследования вен задних конечностей крысы при экспериментальной неопластическая интоксикации преимущественно касались субмикроскопической реорганизации эндотелиоцитов и их десквамации, а также сладжирование тромбоцитов, может быть одним из звеньев патогенеза тромбообразования. При гистологическом и электронно исследовании биоптатов вен больных раком ободочной кишки выявлены нарушения ламинарности интимы, десквамацию эндотелиоцитов, склероз всех оболочек глубокой вены и новообразования сосудов в средней оболочке. Характерно, что у больных раком наблюдаются дистрофически-некротические изменения скелетной мускулатуры, является одним из звеньев патогенеза нарушения гемодинамики в венозной системе нижних конечностей. Выводы. К особенностям перестройки структурных компонентов венозной стенки, как источники возможного первичного формирования внутрисосудистого тромбообразования в условиях онкогенного патологии следует отнести очаговую потерю целостности эндотелиального пласта, эндотелиальную дисфункцию, сладжирование тромбоцитов, волнистые утолщение интимы, фиброзное ремоделирования tunica intima, tunica media, tunica adventitia, неоваскуляризации tunica media, дистрофические и склеротические изменения скелетной мускулатуры., На сьогоднішній день тромбоемболія легеневої артерії є другою за частотою причиною смерті пацієнтів онкологічного профілю. Причиною її є активації системи згортання потенціалу крові, депресії протизгортальної системи і фібринолізу, зниження лінійної швидкості току крові, флебогіпертензії, а також варикозної реорганізації судинної стінки, її клапанів та ендотеліоцитів. Однак структурні зміни вен, як предиктори тромбозу вен у хворих на рак, висвітлені недостатньо. Водночас знання цих змін важливе для розуміння патогенезу і попередження тромбоемболічних ускладнень. Мета: з’ясувати особливості перебудови структурних компонентів венозної стінки, як джерела можливого первинного формування внутрішньосудинного тромбоутворення за умов онкогенної патології. Матеріал та методи. Опрацьовано гістологічні, субмікроскопічні та поляризаційні дані дослідження вен задніх кінцівок 12 статевозрілих нелінійних щурів – самців масою тіла 170-180 г на 30 день хронічної неопластичної інтоксикації та фрагменти вен 12 хворих на рак товстої кишки, ускладненого флеботромбозом. Хронічну неопластичну інтоксикацію моделювали шляхом введення підшкірно в міжлопаткову ділянку в дозі 7,2 мг/кг (з розрахунку на діючу речовину) 1 раз на тиждень впродовж 30 тижнів, відповідно до маси тварини з розрахунку 0,1 мл розчину диметилгідразин гідрохлориду (ДМГ) на 10 г маси тіла щура 1,2- ДМГ (фірми SIGMA-ALDRICH CHEMIE, виробництва Японії, серія D161802), попередньо розведеного ізотонічним розчином натрію хлориду. Результати та обговорення. Морфологічні дослідження вен задніх кінцівок щура при експериментальній неопластичній інтоксикації переважно стосувалися субмікроскопічної реорганізації ендотеліоцитів та їх десквамації, а також сладжування тромбоцитів, що може бути однією із ланок патогенезу тромбоутворення. При гістологічному і електронно-мікроскопічному дослідженні біоптатів вен хворих на рак ободової кишки виявлено порушення ламінарності інтими, десквамацію ендотеліоцитів, склероз всіх оболонок глибокої вени і новоутворення судин в середній оболонці. Характерно, що у хворих на рак спостерігаються дистрофічно-некротичні зміни скелетної мускулатури, що є однією із ланок патогенезу порушення гемодинаміки у венозній системі нижніх кінцівок. Висновки. До особливостей перебудови структурних компонентів венозної стінки, як джерела можливого первинного формування внутрішньосудинного тромбоутворення за умов онкогенної патології слід віднести осередкову втрату цілісності ендотеліального пласта, ендотеліальну дисфункцію, сладжування тромбоцитів, хвилясте потовщення інтими, фіброзне ремоделювання tunica intima, tunica media, tunica adventitia, неоваскуляризацію tunica media, дистрофічні і склеротичні зміни скелетної мускулатури.

DYNAMIC OF INDICES OF LIPID PEROXIDATION AND ANTIOXIDANT PROTECTION IN MUSCULAR TISSUE AND BLOOD SERUM OF RATS WITH ACUTE ISCHEMIA-REPERFUSION A. T. Televiak, T. O. Veresiuk, P. R. Selskyy, A. S. Horbachevsky Ternopil State Medical University of the Ministry of Health of Ukraine” Abstract Acute limb ischemia occurs as a result of sudden decrease in its perfusion, which usually occurs during the obturation of the lumen of large arteries due to acute thrombosis or embolism either impaired of vascular patency caused by trauma or compression (including the application of hemostatic tourniquets). In the case of restoration of blood supply to previously ischemic tissues, there is an ischemic-reperfusion syndrome, in which one of the main pathogenetic links of tissue alteration is the activation of lipid peroxidation. The content of diene conjugates, triene conjugates, TBA-active products, superoxide dismutase and catalase in homogenate of the muscle tissue and blood serum of 30 white rats under condition of experimental acute ischemia was determined. Acute ischemia was caused by the application of SWAT (Stretch-Wrap-And-Tuck) rubber bundles on the hind limbs of the animals for 2 hours. It was experimentally found that the maximum increase in the content of diene and triene conjugates in the injured muscle tissue of the animals occurred in the early post-ischemic period on the 2nd hour of reperfusion with a decreaseing on the 1st and 7th day and the subsequent return of indicators in the late postischemic period on the 14th day to the level of control group. The increasing of TBA-active products in the muscle tissue occurred in the first three groups of the rats and peaked in the third group (reperfusion for the 1st day), after which their level decreased in the animals during the late reperfusion period, reaching in the last group on the 14th day of reperfusion indices, close to the values of the control group. The dynamics of activity of superoxide dismutase and catalase in the muscle homogenate were to a large extent similar (as indicated by the strong positive correlation (+0,99) between them), gradually increasing and reaching the maximum on 7th day of the ischemia-reperfusion symdrome. As a result of the experiment, a decreasing in the antioxidant-prooxidant index in all experimental groups of the rats was also observed in the muscle homogenate, which was the most pronounced on the 1st day of ischemia-reperfusion. The comparative analysis of the similar parameters of lipid peroxidation and antioxidant protection in the muscle homogenate and blood serum showed a direct correlation in all the studied pairs. The strong and moderate positive correlation between analogous indices of LPO and AOP in muscle homogenate and blood serum in all five studied pairs indicates similar changes of these indicators at the local and systemic levels.
