Vaccination against Anaplasma marginale has been considered an important control strategy for bovine anaplasmosis. Recently, mice immunized with rMSP1 a linked to carbon nanotubes (MWNT) showed significant immune responses, generating a new possibility for use of an inactivated vaccine. The objective of this study was to investigate the cellular and humoral responses in calves immunized with MWNT+rMSP1a , associated with inactivated vaccine of A. marginale produced in vitro, and evaluate the toxic effects of the MWNT on renal and hepatic function. rMSP1a was covalently linked to MWNT. Inactivated vaccine (AmUFMG2) was produced by cultivating A. marginale in IDE8 cells. Twenty-four Holstein calves were divided (four groups) and immunized subcutaneously with PBS and non-carboxylated MWNT (control, G1), AmUFMG2 (G2), MWNT+rMSP1a (G3), and AmUFMG2 with MWNT+rMSP1a (G4). Blood samples were collected for total leukocyte counts, biochemical profiling and evaluation of the cellular and humoral response. Immunization with MWNT+rMSP1a induced increase in the total number of leukocytes, NK cells, in the lymphocyte populations and higher levels of antibodies compared to calves immunized only with AmUFMG2. Furthermore, MWNT did not induce changes in the biochemical profile. These data indicate that MWNT+rMSP1a were able to induce the immune responses more efficiently than AmUFMG2 alone, without generating toxicity. Resumo Vacinação contra Anaplasma marginale tem sido considerada uma importante estratégia de controle da anaplasmose bovina. Recentemente, camundongos imunizados com rMSP1a funcionalizada à nanotubos de carbono (MWNT) apresentaram resposta imune significante, gerando nova possibilidade para o uso da vacina inativada. O objetivo desse estudo foi investigar a resposta celular e humoral em bezerros imunizados com MWNT+rMSP1a, associado com a vacina inativada de A. marginale produzida in vitro, e avaliar os efeitos tóxicos dos MWNT nas funções hepática e renal. rMSP1 a foi ligada covalentemente aos MWNT. Vacina inativada (AmUFMG2) foi produzida através do cultivo de A. marginale em células IDE8. Vinte e quatro bezerros Holandeses foram divididos (quatro grupos) e imunizados subcutaneamente com: PBS e MWNT não-carboxilados (controle, G1), AmUFMG2 (G2), MWNT+rMSP1 a (G3), e AmUFMG2 com MWNT+rMSP1a (G4). Amostras de sangue foram coletadas para contagem de leucócitos, perfil bioquímico e avaliação da resposta celular e humoral. Imunização com MWNT+rMSP1a induziu aumento dos leucócitos totais, células NK, na população de linfócitos e altos níveis de anticorpos comparado com animais imunizados apenas com AmUFMG2. Além disso, MWNT não induziu alterações no perfil bioquímico. Esses dados indicam que MWNT+rMSP1a foram capazes de induzir eficientemente a resposta imune comparado com AmUFMG2 sozinho, sem gerar toxicidade.

Statins are hypolipidemic drugs that cause morphological changes, mainly in the reproductive system of S. mansoni adult. The artesunate, an antimalarial drug, reduces the survival time of the worms and the elimination of eggs in vitro. It is unknow the effectiveness of these drugs in schistosomiasis hyperlipidemic mice. Female mice, Swiss Webster received a high-fat diet (29% fat) or standard diet (12% lipids). After six months were infected subcutaneously with 80 cercariae of S. mansoni (BH strain). After 9 weeks of infection, the animals were treated with simvastatin or artesunate, which resulted in the experimental groups: standard diet or a high fat diet untreated standard diet treated with simvastatin or five doses standard diet treated with artesunate or five doses high-fat diet treated with simvastatin with or five doses high-fat diet and treated with artesunate with one or five doses. Two weeks after treatment, the mice were sacrificed to collect blood (cholesterol levels), intestine (oogram) and worms (determination of worm burden reduction, morphometric analysis, morphological, confocal microscopy and scanning). Serum biochemical analysis showed that the animals that received the high-fat diet exhibited higher total cholesterol levels than controls. Treatment with simvastatin had hypolipidemic action (regardless of diet), reduced parasitic load, and caused changes in the reproductive system and especially the integument of worms recovered from dyslipidemic mice. The difference between the simvastatin doses did not appear to interfere with the observed changes. The artesunate promoved intense reduction in mean worm recovery and increase in parasitic rate of reduction in dyslipidemic group. The reproductive system and the integument showed moderate to intense change, especially when related to diet. The effect of artesunate was higher when given five doses of worms recovered dyslipidemic mice. It is concluded that the drugs used in this study had better in groups of dyslipidemic mice. A administração por longo prazo de uma dieta rica em gordura favorece o desenvolvimento de vermes adultos de Schistosoma mansoni e a infecção em camundongos. As estatinas são drogas hipolipidêmicas que promovem alterações morfológicas, principalmente no sistema reprodutivo de S. mansoni adulto. O artesunato, uma droga antimalárica, reduz o tempo de sobrevivência dos vermes e a eliminação dos ovos in vitro. Não se conhece a eficácia destes fármacos na esquistossomose de camundongos hiperlipídicos. Camundongos fêmeas, Swiss Webster receberam uma dieta hiperlipídica (29% de lipídios) ou dieta padrão (12% lipídios). Depois de seis meses foram infectados por via subcutânea com 80 cercárias de S. mansoni (cepa BH). Após 9 semanas de infecção, os animais foram tratados com sinvastatina ou artesunato, no que resultou nos grupos experimentais: dieta padrão ou dieta hiperlipídica não tratado, dieta padrão tratado com sinvastatina com uma dose durante cinco dias consecutivos, dieta padrão tratado com artesunato com uma dose ou durante cinco dias consecutivos, dieta hiperlipídica tratado com sinvastatina com uma dose ou durante cinco dias consecutivos e dieta hiperlipídica tratado com artesunato com uma dose ou durante cinco dias consecutivos. Duas semanas após o tratamento, os animais foram necropsiados para a coleta de sangue (dosagem de colesterol), intestino (oograma) e vermes (determinação da redução da carga parasitária, análise morfométrica, morfológica, microscopia confocal e de varredura). A análise bioquímica do soro mostrou que os animais que receberam a dieta hiperlipídica apresentaram níveis mais altos de colesterol total do que os controles. O tratamento com sinvastatina teve ação hipolipidêmica (independentemente da dieta), reduziu a carga parasitária, causou alterações no sistema reprodutor e no tegumento principalmente dos vermes recuperados de camundongos dislipidêmicos. A diferença entre as dosagens da sinvastatina não pareceu interferir nas alterações observadas. O artesunato promoveu intensa redução na média de recuperação dos vermes e elevação na taxa de redução parasitária no grupo dislipidêmico. O sistema reprodutor e o tegumento apresentaram alterações moderadas a intensas, principalmente quando relacionado à dieta. O efeito do artesunato foi maior quando administrado em cinco doses em vermes recuperados de camundongos dislipidêmicos. Conclui-se que os fármacos utilizados nesse experimento apresentaram maior eficácia nos grupos de camundongos dislipidêmicos.

O córrego Herivelton Martins foi escolhido para dar continuidade aos estudos com a introdução da linhagem Biomphalaria tenagophila do Taim/RS, resistente ao Schistosoma mansoni, em coleções hídricas do Município de Bananal/SP, iniciados em 2008. Oitocentos exemplares B. tenagophila adultos da linhagem Taim/RS foram marcados e introduzidos. Pelo método de marcação da concha foi estimada a densidade populacional da linhagem introduzida em relação à local. Após 15 dias da introdução 37,5% dos caramujos coletados estavam marcados. Após 4, 11 e 14 meses da introdução, caramujos jovens (≤ 5 mm de diâmetro) foram coletados e submetidos a técnica de PCR_RFLP para identificação do marcador molecular de 350pb, típico da linhagem do Taim e com caráter dominante. A proporção do marcador molecular nestes caramujos foi 37,1%, 35,7 e 60% respectivamente. Foram realizados três testes de infecção com a cepa SJ de S. mansoni com os descendentes F1 de caramujos coletados nos diferentes períodos após a introdução. Os sobreviventes foram submetidos à técnica de PCR_RFLP para verificar se havia alguma correlação entre a resistência e a presença do marcador de 350pb. No primeiro desafio com o S. mansoni 38,6% dos exemplares provenientes dos caramujos locais, coletados antes da introdução, estavam positivos, bem como 14,9% dos descendentes coletados após 4 meses da introdução. A análise molecular realizada com os caramujos positivos para S. mansoni, oriundos de exemplares pós-introdução, demonstraram que apenas 10% apresentaram o marcador de 350 pb, sendo que este marcador estava presente em 60,8% dos caramujos negativos. No segundo desafio, os descendentes de caramujos coletados antes, após 4 e 11 meses da introdução apresentaram taxas de infecção de 25%, 4,1%, 17%, respectivamente. Os caramujos positivos para S. mansoni não apresentaram o marcador molecular. A percentagem do marcador molecular nos caramujos negativos, coletados após 4 e 11 meses da introdução, foi de 66,6% e 56,2%, respectivamente. No terceiro desafio, as taxas de infecção dos descendentes foram de 26,5% antes da introdução, 6,8% após 11 meses e 2,1% após 14 meses. A análise molecular dos descendentes neste desafio está em andamento. Os dados apontam para uma significativa diminuição da suscetibilidade dos caramujos coletados após a intervenção. O marcador molecular de 350pb típico da linhagem do Taim foi detectado em proporções muito maiores nos caramujos negativos. Os resultados parciais obtidos permitem a inferência que o patrimônio genético da linhagem resistente B. tenagophila do Taim está sendo transmitido com êxito aos descendentes após intervenção tendo como consequência uma maior resistência à infecção por S. mansoni. The stream Herivelton Martins was chosen to continue the studies on introduction of Biomphalaria tenagophila from Taim/RS, resistant to Schistosoma mansoni, in hydric collections of Bananal/SP, initiated in 2008. Eight hundred adult specimens of B. tenagophila belonging to the lineage of Taim/RS were labeled and introduced. The populational density of the introduced lineage in relation to the local one was estimated using the labeling method of the shell. Fifteen days post-introduction, 37.5% of the collected snails were labeled. After 4, 11 and 14 months of the introduction, juvenile snails (≤ 5 mm diameter) were collected and submitted to PCR-RFLP technique for identification of the molecular marker 350 pb, typical of the Taim lineage and with dominant character. The proportions of the molecular marker in these snails were 37.1%, 35.7% and 60%, respectively. Three infection tests were carried out with the SJ strain of S. mansoni, using the descendants F1 of the snails collected on different periods after introduction. The surviving snails were submitted to the PCR-RFLP technique, in order to verify if there was any correlation between resistance and the presence of the marker 350 pb. In the first challenge with S. mansoni, 38.6% of the specimens belonging to the local population, and collected before introduction, were found to be positive, as well as 14.9% of the offsprings collected 4 months post-introduction. The molecular analysis performed with positive snails to S. mansoni, related to specimens after introduction, demonstrated that only 10% presented the molecular marker 350 pb, and this marker was present in 60.8% of the negative snails. In the second challenge, the offsprings of the snails collected before or 4 and 11 months post-introduction presented infection rates of 25%, 4.1% and 17%, respectively. The positive snails for S. mansoni did not present the molecular marker. The proportions of the molecular marker in the negative snails, collected 4 and 11 months after introduction, were 66.6% and 56.2%, respectively. In the third challenge, the infection rates of the descendants were 26.5% before introduction, 6.8% after 11 months and 2.1% after 14 months. The molecular analysis of the descendants related to this challenge is in progress. Data point out to a significant decrease in the susceptibility of the snails collected after intervention. The molecular marker 350 pb, typical of the Taim lineage, was detected in higher proportions in the negative snails. The partial results obtained allow to infer that the genetic heritage of the resistant B. tenagophila strain from Taim is being successfully transmitted to the descendants after intervention, showing as a result a higher resistance to S. mansoni infection.
