Abstract

The mechanical competence and suturing ability of collagen-based membranes are paramount in Guided Bone Regeneration (GBR) therapy, to ensure damage-free implantation, fixation and space maintenance in vivo. However, contact with the biological medium can induce swelling of collagen molecules, yielding risks of membrane sinking into the bone defect, early loss of barrier function, and irreversibly compromised clinical outcomes. To address these challenges, this study investigates the effect of the crosslinked network architecture on both mechanical and suture-holding properties of a new atelocollagen (AC) membrane. UV-cured networks were obtained via either single functionalisation of AC with 4-vinylbenzyl chloride (4VBC) or sequential functionalisation of AC with both 4VBC and methacrylic anhydride (MA). The wet-state compression modulus (Ec), Atomic Force Microscopy elastic modulus (EAFM) and swelling ratio (SR) were significantly affected by the UV-cured network architecture, leading up to a three-fold reduction in SR, and about two-fold increase in both Ec and EAFM, in the sequentially functionalised, compared to the single-functionalised, samples. Electron microscopy, dimensional analysis and compression testing revealed the direct impact of the ethanol series dehydration process on membrane microstructure, yielding densification of the freshly synthesised porous samples and a pore-free microstructure with increased Ec. Noteworthy, the single-functionalised, but not the sequentially functionalised, samples displayed higher suture retention strength in both the dry state and following 1 hour in Phosphate Buffered Saline (PBS), compared to Bio-Gide(r). Abstract

Abstract The success or failure of apex carnivore reintroduction efforts can hinge on understanding and attending to diverse viewpoints of those involved in and impacted by reintroductions. Yet, viewpoints vary widely due to a suite of complex and intersecting factors, such as values, beliefs and sociocultural context. We ask, ‘what are the diverse viewpoints that exist surrounding apex carnivore recovery and what kinds of emotional, analytical and values‐based judgments might people use to construct their viewpoints?’ We used Q‐methodology to identify distinct, generalized viewpoints and areas of overlap and divergence between them, surrounding a proposal to reintroduce grizzly bears (Ursus arctos horribilis) to the North Cascades Ecosystem, USA. Q‐methodology combines qualitative and quantitative methods by asking purposefully sampled respondents to sort various statements on a given topic into an ordered grid. We found three distinct viewpoints among 67 respondents using factor analysis and responses to open‐ended questions about the sorting exercise. Two of these viewpoints represent essentially polarized perspectives corresponding to deeply normative notions about grizzly bear recovery, where one views reintroducing bears as a moral requisite, and the other views it as inappropriate and risky. These viewpoints primarily diverged on their perceptions of risk and perspectives about our collective responsibilities to and appropriate relationships with others (i.e. ‘relational values’). The third viewpoint was distinguished by its prioritization of practical considerations and views reintroducing bears as impractical and not sensible. Our analysis underscores the need to identify and attend to latent viewpoints that may be overlooked in the polarized public discourse as well as the multiple value systems and perceptions of risk that are integrated in perspectives on grizzly bear reintroduction. Additionally, our broadly defined identity groups were of very little utility in predicting viewpoints in this study, highlighting the importance of avoiding assumptions about people's views based on their identities and interests. We argue that forefronting conversations about responsibilities and appropriate relationships is critical for finding acceptable paths forward in such recovery efforts. We discuss the management implications of these findings for the North Cascades grizzly bear reintroduction, and for other large carnivore reintroductions. Abstract

Abstract Predator–prey relationships are considered a major driver for the evolution of organisms, and thus contributed to shaping morphology, ecology, and diversity. During the Late Cretaceous of North America, ammonoid cephalopods were one of the most abundant and diverse marine invertebrates. Despite frequent reports of shell breakage in ammonoids, little is known pertaining to the frequency, position, and size of the shell break through a stratigraphic succession. In this study, we analyze an extensive collection of the scaphitid ammonoid Hoploscaphites nicolletii, which exhibits shell breakage, from the Upper Cretaceous (Maastrichtian) Fox Hills Formation in South Dakota, USA. We focus on four upper Maastrichtian assemblage zones listed stratigraphically from bottom to top—the lower nicolletii Assemblage Zone (LNAZ), the Limopsis-Gervillia Assemblage Zone (LGAZ), the upper nicolletii Assemblage Zone (UNAZ), and the Protocardia-Oxytoma Assemblage Zone (POAZ). Within the collection, we observed two primary types of breakage: ventral and lateral, each displaying a relatively consistent geometry. Lateral breaks, measuring a few centimeters, represent about 20–40% of the maximum conch diameter. Ventral breaks are slightly larger, representing 30–70% of the diameter. Both breakage types occur in the body chamber at approximately 90° from the aperture extending to near the last septum. We find that the incidence of injury increased from 6.6 to 13.7% with some fluctuation across the zones. The breakage size relative to body size does not exhibit a clear change across the assemblage zones. Additionally, no significant difference is apparent in the body size between injured and uninjured specimens within each zone. A weak positive correlation between the size of lateral breaks and maximum conch diameter in LNAZ suggests a tendency for larger predators to target larger individuals. Given the consistency of geometry and size, we presume that these breaks represent lethal injuries from durophagous predators. We propose coleoid cephalopods as the likely culprits for ventral injuries, although fish and crustaceans are plausible alternatives. Concerning lateral injuries, decapod crustaceans appear to be the most probable durophagous predators.

Abstract

Clinical trials frequently include multiple end points that mature at different times. The initial report, typically based on the primary end point, may be published when key planned co-primary or secondary analyses are not yet available. Clinical Trial Updates provide an opportunity to disseminate additional results from studies, published in JCO or elsewhere, for which the primary end point has already been reported.To assess long-term risk of local tumor regrowth, we report updated organ preservation rate and oncologic outcomes of the OPRA trial (ClinicalTrials.gov identifier: NCT02008656). Patients with stage II/III rectal cancer were randomly assigned to receive induction chemotherapy followed by chemoradiation (INCT-CRT) or chemoradiation followed by consolidation chemotherapy (CRT-CNCT). Patients who achieved a complete or near-complete response after finishing treatment were offered watch-and-wait (WW). Total mesorectal excision (TME) was recommended for those who achieved an incomplete response. The primary end point was disease-free survival (DFS). The secondary end point was TME-free survival. In total, 324 patients were randomly assigned (INCT-CRT, n = 158; CRT-CNCT, n = 166). Median follow-up was 5.1 years. The 5-year DFS rates were 71% (95% CI, 64 to 79) and 69% (95% CI, 62 to 77) for INCT-CRT and CRT-CNCT, respectively (P = .68). TME-free survival was 39% (95% CI, 32 to 48) in the INCT-CRT group and 54% (95% CI, 46 to 62) in the CRT-CNCT group (P = .012). Of 81 patients with regrowth, 94% occurred within 2 years and 99% occurred within 3 years. DFS was similar for patients who underwent TME after restaging (64% [95% CI, 53 to 78]) and patients in WW who underwent TME after regrowth (64% [95% CI, 53 to 78]; P = .94). Updated analysis continues to show long-term organ preservation in half of the patients with rectal cancer treated with total neoadjuvant therapy. In patients who enter WW, most cases of tumor regrowth occur in the first 2 years.

Abstract

Abnormal expression of the transcriptional regulator and hedgehog (Hh) signaling pathway effector Gli3 is known to trigger congenital disease, most frequently affecting the central nervous system (CNS) and the limbs. Accurate delineation of the genomic cis-regulatory landscape controlling Gli3 transcription during embryonic development is critical for the interpretation of noncoding variants associated with congenital defects. Here, we employed a comparative genomic analysis on fish species with a slow rate of molecular evolution to identify seven previously unknown conserved noncoding elements (CNEs) in Gli3 intronic intervals (CNE15-21). Transgenic assays in zebrafish revealed that most of these elements drive activities in Gli3 expressing tissues, predominantly the fins, CNS, and the heart. Intersection of these CNEs with human disease associated SNPs identified CNE15 as a putative mammalian craniofacial enhancer, with conserved activity in vertebrates and potentially affected by mutation associated with human craniofacial morphology. Finally, comparative functional dissection of an appendage-specific CNE conserved in slowly evolving fish (elephant shark), but not in teleost (CNE14/hs1586) indicates co-option of limb specificity from other tissues prior to the divergence of amniotes and lobe-finned fish. These results uncover a novel subset of intronic Gli3 enhancers that arose in the common ancestor of gnathostomes and whose sequence components were likely gradually modified in other species during the process of evolutionary diversification.

Abstract

Metformin is the first-line treatment for type 2 diabetes (T2D) in youth but with limited sustained glycemic response. To identify common variants associated with metformin response, we used a genome-wide approach in 506 youth from the Treatment Options for Type 2 Diabetes in Adolescents and Youth (TODAY) study and examined the relationship between T2D partitioned polygenic scores (pPS), glycemic traits, and metformin response in these youth. Several variants met a suggestive threshold (P < 1 × 10-6), though none including published adult variants reached genome-wide significance. We pursued replication of top nine variants in three cohorts, and rs76195229 in ATRNL1 was associated with worse metformin response in the Metformin Genetics Consortium (n = 7,812), though statistically not being significant after Bonferroni correction (P = 0.06). A higher β-cell pPS was associated with a lower insulinogenic index (P = 0.02) and C-peptide (P = 0.047) at baseline and higher pPS related to two insulin resistance processes were associated with increased C-peptide at baseline (P = 0.04,0.02). Although pPS were not associated with changes in glycemic traits or metformin response, our results indicate a trend in the association of the β-cell pPS with reduced β-cell function over time. Our data show initial evidence for genetic variation associated with metformin response in youth with T2D.

18. Discovery of three new near-pristine absorption clouds at $z=2.6$-4.4

Abstract

We report the discovery of three new "near-pristine" Lyman Limit Systems (LLSs), with metallicities ~1/1000 solar, at redshifts 2.6, 3.8 and 4.0, with a targeted survey at the Keck Observatory. High resolution echelle spectra of eight candidates yielded precise column densities of hydrogen and weak, but clearly detected, metal lines in seven LLSs; we previously reported the one remaining, apparently metal-free LLS, to have metallicity <1/10000 solar. Robust photoionisation modelling provides metallicities [Si/H] = -3.05 to -2.94, with 0.26 dex uncertainties (95% confidence) for three LLSs, and [Si/H] >~ -2.5 for the remaining four. Previous simulations suggest that near-pristine LLSs could be the remnants of PopIII supernovae, so comparing their detailed metal abundances with nucleosynthetic yields from supernovae models is an important goal. Unfortunately, at most two different metals were detected in each new system, despite their neutral hydrogen column densities (10^{19.2-19.4} cm^{-2}) being two orders of magnitude larger than the two previous, serendipitously discovered near-pristine LLSs. Nevertheless, the success of this first targeted survey for near-pristine systems demonstrates the prospect that a much larger, future survey could identify clear observational signatures of PopIII stars. With a well-understood selection function, such a survey would also yield the number density of near-pristine absorbers which, via comparison to future simulations, could reveal the origin(s) of these rare systems., Comment: Accepted by MNRAS. 21 pages, 27 figures

Abstract

Recovered and recovering carnivore populations in Europe and North America can pose risks to some human livelihoods like livestock ranching. These risks can motivate wildlife managers to lethally remove carnivores—decisions that are often controversial and poorly understood. We used a 13-year dataset on gray wolves ( Canis lupus ) in the northwestern United States (Montana, Idaho, Washington, and Oregon) to analyze how social, demographic, and environmental variables influence lethal removal of wolves at the county and state levels. We found that state-level differences are a major driver of lethal removal decisions at the county level. The percentage of federally owned and protected lands was also positively correlated with lethal removal. Predation of livestock by wolves was not significantly correlated with wolf removals in Idaho, but was in Montana, Washington, and Oregon. Our results stress the need to make transparent the process by which recovering populations of carnivores are managed to enhance the legitimacy of management policies.

Abstract

Abstract Because it can lead to retaliatory killing, livestock depredation by large carnivores is among the foremost threats to carnivore conservation, and it severely impacts human well‐being worldwide. Ongoing climate change can amplify these human–wildlife conflicts, but such issues are largely unexplored, though are becoming increasingly recognized. Here, we assessed how the availability of primary resources and wild prey interact to shape large carnivore selection for livestock rather than wild prey (i.e., via prey switching or apparent competition). Specifically, we combined remotely sensed estimates of primary resources (i.e., water availability and primary productivity), wild prey movement, and 7 years (2015–2021) of reports for livestock depredation by African lions (Panthera leo) in the Makgadikgadi Pans ecosystem, Botswana. Although livestock depredation did not vary between wet versus dry seasons, analyses at finer temporal scales revealed higher incidences of livestock depredation when primary production, water availability, and wild prey availability were lower, though the effects of wild prey availability were mediated by water availability. Increased precipitation also amplified livestock depredation events despite having no influence on wild prey availability. Our results suggest that livestock depredation is influenced by the diverse responses of livestock, wild prey, and lions to primary resource availability, a driver that is largely overlooked or oversimplified in studies of human–carnivore conflict. Our findings provide insight into tailoring potential conflict mitigation strategies to fine‐scale changes in resource conditions to efficiently reduce conflict and support human livelihoods.

Abstract

Evolutionary novelties entail the origin of morphologies that enable new functions. These features can arise through changes to gene function and regulation. One key novelty is the fused rod at the end of the vertebral column in anurans, the urostyle. This feature is composed of a coccyx and a hypochord, both of which ossify during metamorphosis. To elucidate the genetic basis of these features, we used laser capture microdissection of these tissues and did RNA-seq and ATAC-seq at three developmental stages in tadpoles of Xenopus tropicalis. RNA-seq reveals that the coccyx and hypochord have two different molecular signatures. Neuronal (TUBB3) and muscle markers (MYH3) are upregulated in coccygeal tissues, whereas T-box genes (TBXT, TBXT.2), corticosteroid stress hormones (CRCH.1) and matrix metallopeptidases (MMP1, MMP8 and MMP13) are upregulated in the hypochord. ATAC-seq reveals potential regulatory regions that are observed in proximity to candidate genes that regulate ossification identified from RNA-seq. Even though an ossifying hypochord is only present in anurans, this ossification between the vertebral column and the notochord resembles a congenital vertebral anomaly seen prenatally in humans caused by an ectopic expression of the TBXT/TBXT.2 gene. This work opens the way to functional studies that can elucidate anuran bauplan evolution.

Abstract

Climate and land-use change are modifying the availability of food and water resources, which is driving more wildlife to the wildland-urban interface. For many wildlife populations, use of these areas still requires habitat connectivity both within the interface and/or to wildland habitats. However, navigating these areas can be difficult due to human development and sensory pollutants, such as artificial night light. Determining how these components of urbanization influence the behaviors and functional connectivity of species is important for managing wildlife within these mixed-use landscapes. Here we used a movescape approach based on graph theory to characterize functional uses of the landscape using metrics for behavior, connectivity, and space use intensity. We found that mule deer (Odocoileus hemionus; n = 43) functional uses of anthropogenic landscapes in the Intermountain West, USA, were dependent not only on physical barriers, terrain, and sensory factors, but also the local levels of light exposure and vegetative greenness. Remotely sensed artificial light levels had a strong influence on how mule deer used the landscape by reducing the intensity of use in the most illuminated areas given forage availability. In contrast, relatively high local light levels were associated with increased use intensity within less developed areas—highlighting the foraging tradeoffs for species using the wildland-urban interface. Corridor use was reduced in areas where road and housing density were higher, and within-corridor movement was faster when artificial light was high and vegetative greenness was low. Developing a more mechanistic understanding of how species functionally use the landscape in relation to features of urbanization can enhance conservation by delineating areas important for connectivity and foraging, while providing insights into how future development and climate change may alter movement and behavior. Spatially-explicit estimates of functional uses can directly guide management decisions to maintain species resiliency and improve land-use planning.

Abstract 1. Conserving species' ability to traverse the landscape is vital for maintaining biodiversity in the face of global change. Connectivity conservation requires identifying important pathways for species' movements and aligning societal support for conservation of those pathways. Contemporary connectivity analyses emphasize the impacts of topography, vegetation and human footprint on species' movements; but largely ignore the role that attitudes, economics and institutions play in practitioners' ability to conserve those movements. 2. We introduce implementation resistance as an analogue of biophysical resistance that combines social, economic and institutional factors that promote or impede connectivity conservation. We demonstrate the utility of integrating implementation resistance as a means of choosing between competing connectivity conservation strategies using wolves in Colorado (USA) as a case study. 3. Our analysis of five potential corridor locations based on biophysical costs revealed substantial differences in the social costs associated with implementing each corridor despite relatively minimal differences in the biophysical costs. 4. Our comparison of hypothetical interventions to reduce implementation resistance illustrates that interventions that reduce conflicts between land use and wolves may substantially reduce overall resistance, those reductions are not as well aligned with connectivity priorities as those resulting from changes in land management agency policy. 5. Our results highlight the need to design conservation interventions that fit both the social and ecological landscape and provide a framework for developing robust, interdisciplinary methods that facilitate implementable connectivity conservation. Abstract Wide-ranging carnivores experience tradeoffs between dynamic resource availabilities and heterogeneous risks from humans, with consequences for their ecological function and conservation outcomes. Yet, research investigating these tradeoffs across large carnivore distributions is rare. We assessed how resource availability and anthropogenic risks influence the strength of lion (Panthera leo) responses to disturbance using data from 31 sites across lions’ contemporary range. Lions avoided human disturbance at over two-thirds of sites, though their responses varied depending on site-level characteristics. Lions were more likely to exploit human-dominated landscapes where resources were limited, indicating that resource limitation can outweigh anthropogenic risks and might exacerbate human-carnivore conflict. Lions also avoided human impacts by increasing their nocturnal activity more often at sites with higher production of cattle. The combined effects of expanding human impacts and environmental change threaten to simultaneously downgrade the ecological function of carnivores and intensify human-carnivore conflicts, escalating extinction risks for many species.

Abstract

Drawing on quantitative and qualitative research in England, and more specifically in London, this article sheds light on trends in platform work during the COVID-19 crisis. While the number of platform workers grew, the proportion of their income it contributed to fell, making up less than a quarter of total earnings. Interviews with driving and delivery platform workers in London (Europe’s largest platform market) shed light on these puzzling trends. New recruitment by the platforms and adjustment of their algorithms during the lockdown led to downward pressure on earnings, poorer working conditions, extended waiting times, longer working hours and negative impacts on work–life balance, health and well-being. The article concludes that the pandemic provided platforms with an opportunity to consolidate their market position, but this was achieved at the cost of growing power asymmetry in the platform labour market, with workers’ attempts to organise and improve conditions undermined by over-recruitment.

BackgroundYouth overweight and obesity is a public health crisis and increases the risk of poor cardiovascular health (CVH) and chronic disease. Health care providers play a key role in weight management, yet few tools exist to support providers in delivering tailored evidence-based behavior change interventions to patients. ObjectiveThe goal of this pilot randomized feasibility study was to determine the feasibility of implementing the Patient-Centered Real-Time Intervention (PREVENT) tool in clinical settings, generate implementation data to inform scale-up, and gather preliminary effectiveness data. MethodsA pilot randomized clinical trial was conducted to examine the feasibility, implementation, and preliminary impact of PREVENT on patient knowledge, motivation, behaviors, and CVH outcomes. The study took place in a multidisciplinary obesity management clinic at a children’s hospital within an academic medical center. A total of 36 patients aged 12 to 18 years were randomized to use PREVENT during their routine visit (n=18, 50%) or usual care control (n=18, 50%). PREVENT is a digital health tool designed for use by providers to engage patients in behavior change education and goal setting and provides resources to support change. Patient electronic health record and self-report behavior data were collected at baseline and 3 months after the intervention. Implementation data were collected via PREVENT, direct observation, surveys, and interviews. We conducted quantitative, qualitative, and mixed methods analyses to evaluate pretest-posttest patient changes and implementation data. ResultsPREVENT was feasible, acceptable, easy to understand, and helpful to patients. Although not statistically significant, only PREVENT patients increased their motivation to change their behaviors as well as their knowledge of ways to improve heart health and of resources. Abstract

Spaying female and castrating male dogs, hereinafter referred to as neutering, is a US convention for the first year in the dog's life. Research on 35 breeds of dogs revealed that early neutering increases risks of joint disorders, such as hip dysplasia (HD), elbow dysplasia (ED), or cranial cruciate ligament (CCL) tear, or cancers, such as lymphosarcoma (LSA), mast cell tumor (MCT), hemangiosarcoma (has), or osteosarcoma (OSA), for some breeds. Joint disorder risks are heightened for some larger breeds and for mixed-breed dogs weighing more than 20 kg. Some breeds had elevated risks for cancers. Several other research teams have reported health complications associated with neutering. The study goal includes using the same methodology for data collection and analyses as in the study on 35 breeds for five additional dog breeds weighing at least 20 kg. The breeds were: German Short/Wirehaired Pointer, Mastiff, Newfoundland, Rhodesian Ridgeback, and Siberian Husky. Major differences among breeds appeared in vulnerability to joint disorders and cancers with early neutering: male and female Pointer breeds had elevated joint disorders and increased cancers; male Mastiff breeds had increased CCL and LSA and females had non-significant elevated CCL risks; female Newfoundland breeds had heightened risks for joint disorders and males had non-significant elevated risks; female Ridgeback breeds had heightened MCT with very early neutering; and Siberian Huskies showed no significant effects on joint disorders or cancers, but female breeds showed a non-significant but elevated CCL. Updated guidelines cover 40 dog breeds. These results further emphasize the importance of personalized decisions regarding the neutering of dogs, considering the dog's breed, sex, and context.

Abstract

The November 2018 Camp Fire, a devastating wildfire in Northern California, occurred during the peak of breeding season for field monkeys at the California National Primate Research Center (CNPRC). Effects of environmental stressors, such as wildfires, on birth outcomes in primates, and in humans, are poorly understood. Additionally, wildfires are of growing concern due to their increasing frequency and severity. The objective was to examine the impact of wildfire smoke on fertility, timing of birth, and pregnancy loss for field monkeys. A unique case-control study to investigate birth outcomes in rhesus macaques (Macaca mulatta) was conducted at the CNPRC. All females in the study were maintained in outdoor fields during a period of elevated ambient wildfire smoke from November 8-22, 2018. In addition to ambient air quality evaluations, the effects on fertility, timing to birth, and pregnancy loss were documented. Archival records of approximately 5,000 conceptions from the previous nine years served as control data. During the Camp Fire, ambient fine particulate (PM 2.5) levels exceeded the 24 -h National Ambient Air Quality Standard (35 μg/m 3) of the United States Environmental Protection Agency, reaching levels as high as 185 μg/m 3. A statistically significant association was observed between birth loss and the 2018-2019 CNPRC breeding season. As this wildfire event occurred during various stages of early pregnancy, an association can be inferred between early gestational exposure and increased risk of pregnancy loss.
