This study was conducted to evaluate the effects of various combinations of Lactobacillus species (L. rhamnosus, L. paracasei, and L. plantarum) on closely associated variables of production of laying hens, nitrogenous compounds in manure, the serum concentration of specific chemicals, and liver uric acid (UA) concentrations at peak lay. White Leghorns W-36 (32-week-old) were randomly assigned to five treatments for 8 weeks. Treatments were T1, the Control, a commercial feed; T2, the Control + L. paracasei + L. plantarum; T3, the Control + L. paracasei + L. rhamnosus; T4, the Control + L. plantarum + L. rhamnosus and T5, the Control + L. paracasei + L. plantarum + L. rhamnosus. Each bacterial species was included at 3.33 × 1011cfu/kg feed for a total of 6.66 x 1011 cfu/kg feed for T2–T4 and a total of 1.0 × 1012 cfu/kg feed for T5. Major effects among combinations of probiotics on production were not noted. The interaction of Probiotics by Week (Probiotics*Time) affected feed intake (P = 0.0007) and feed conversion ratio (FCR, P = 0.0049) due to fluctuation by week. Significant effects of time were also recorded for a gradual increase in body weight (BW, P = 0.0007); lowest and greatest feed intake at weeks 2 and 7, respectively (P < 0.0001); an increase in egg production (P = 0.0007) and maximum FCR at week 7 (P < 0.0001). Ammonia (NH3) concentration, ammonium nitrogen (NH4–N), total Kjeldahl nitrogen (TKN), and total nitrogen remained unaffected at P < 0.05. Although there were fluctuations, a trend emerged for the reduction of TKN. Combinations of probiotics did not affect NH3, UA, total protein (TP), albumin (ALB), creatine kinase (CK), and UA in the liver. Temporal (Time as a fixed effect) effects were noted for all nitrogenous compounds present in manure. For ammonia, temporal effects were significant due to fluctuation over time. Week 0 had the lowest value followed by weeks 4 and 8. Week 6 had the greatest value. For ammonium nitrogen, week 8 had the lowest value followed by week 0 and 4 with the next highest value. Week 6 had the greatest value. For TKN, week 4 had the lowest value followed by weeks 6 and 8. Week 0 had the greatest value. For TN, weeks 4, 6, and 8 had similar and lowest values followed by week 0 having the greatest value. However, an overall reduction in NH4-N, TKN, and TN was noted. Fluctuations in NH3 (P = 0.0033) and CK (P = 0.0085) were noted for Time. There was also a trend (P = 0.0706) for the increase of UA in serum. Two or more species of probiotics with yeast should be investigated. If the combination is applicable for increasing production measurements and reducing nitrogenous and serum compounds, the most appropriate time to feed the probiotics from day 1 to the end of production should be investigated.

Resistance to ripening in European pears depends on various preharvest and postharvest factors. Exposure to cold temperatures has been demonstrated to condition pears to produce endogenous ethylene and ripen. Mature green ‘Bartlett’ pears were exposed to 0, 5, or 10 °C for 2–14 d to induce different rates of ripening. At higher temperatures, expression of ethylene biosynthesis genes (ACS1a and ACO) and ethylene receptor genes (ETR2, ERS1a, and ETR1a) increased, while CTR1 expression decreased. Multiple linear regressions between relative expression of these genes after 0 d and fruit firmness after 6 d during ripening at 20 °C were conducted. Using K-fold cross validation and conventional validation, it appears that expression of ACO could be utilized as an early predictor of pear ripening capacity.

Assistance dogs' roles have diversified to support people with various disabilities, especially in the U.S. Data presented here are from the U.S. and Canada non-profit facilities (including both accredited and candidate members that fulfilled partial requirements: all here termed "accredited") of Assistance Dogs International (ADI) and the International Guide Dog Federation (IGDF), and from non-accredited U.S. assistance dog training facilities, on the numbers and types of dogs they placed in 2013 and 2014 with persons who have disabilities. ADI categories of assistance dogs are for guide, hearing, and service (including for assistance with mobility, autism, psychiatric, diabetes, seizure disabilities). Accredited facilities in 28 states and 3 provinces responded; accredited non-responding facilities were in 22 states and 1 province (some in states/provinces with responding accredited facilities). Non-accredited facilities in 16 states responded. U.S./Canada responding accredited facilities (55 of 96: 57%) placed 2,374 dogs; non-accredited U.S. facilities (22 of 133: 16.5%) placed 797 dogs. Accredited facilities placed similar numbers of dogs for guiding (n = 918) or mobility (n = 943), but many more facilities placed mobility service dogs than guide dogs. Autism service dogs were third most for accredited (n = 205 placements) and U.S. non-accredited (n = 72) facilities. Psychiatric service dogs were fourth most common in accredited placements (n = 119) and accounted for most placements (n = 526) in non-accredited facilities. Other accredited placements were for: hearing (n = 109); diabetic alert (n = 69), and seizure response (n = 11). Responding non-accredited facilities placed 17 hearing dogs, 30 diabetic alert dogs, and 18 seizure response dogs. Non-accredited facilities placed many dogs for psychiatric assistance, often for veterans, but ADI accreditation is required for veterans to have financial reimbursement. Twenty states and several provinces had no responding facilities; 17 of these states had no accredited facilities. In regions lacking facilities, some people with disabilities may find it inconvenient living far from any supportive facility, even if travel costs are provided. Despite accelerated U.S./Canada placements, access to well-trained assistance dogs continues to be limited and inconvenient for many people with disabilities, and the numerous sources of expensive, poorly trained dogs add confusion for potential handlers.

Sexually abused children providing essential testimony regarding crimes in forensic interviews now sometimes are provided facility dogs or therapy dogs for comfort. Facility dogs are extensively trained to work with forensic interviewers; when using therapy dogs in interviews, volunteers are the dog handlers. Interviews can impact child welfare workers' mental health causing secondary traumatic stress (STS). To investigate this stress, first data were gathered on stress retrospectively for when interviewers initially started the job prior to working with a dog, and then currently, from forensic interviewers using a facility dog, a therapy or pet dog, or no dog. These retrospective and secondary traumatic stress scale (STSS) data compared job stress among interviewers of children using: a certified, workplace facility dog (n = 16), a volunteer's trained therapy dog or the interviewer's pet dog (n = 13/3), or no dog (n = 198). Retrospective scores of therapy dog and no dog interviewers' stress were highest for the first interviewing year 1 and then declined. Extremely or very stressful retrospective scores differed among the three groups in year 1 (p < 0.038), and were significantly elevated for the therapy dog group as compared with the facility dog group (p < 0.035). All interviewing groups had elevated STSS scores; when compared with other healthcare groups that have been studied, sub-scores were especially high for Avoidance: a psychological coping mechanism to avoid dealing with a stressor. STSS scores differed among groups (p < 0.016), primarily due to Avoidance sub-scores (p < 0.009), reflecting higher Avoidance scores for therapy dog users than no dog users (p < 0.009). Facility dog users more consistently used dogs during interviews and conducted more interviews than therapy/pet dog users; both groups favored using dogs. Interviewers currently working with therapy dogs accompanied by their volunteers reported they had experienced heightened stress when they began their jobs; their high stress levels still persisted, indicating lower inherent coping skills and perhaps greater empathy among interviewers who later self-selected to work with therapy dogs. Results reveal extreme avoidant stress for interviewers witnessing children who are suffering and their differing coping approaches.

Although studies involving pet dogs and cats, and human adults and children, have been reported, the specific interactions between cats and children have not. This study sought information from parents about the cat's role in families that have at least one child 3-12 years of age and at least one cat. Demographic data on cat source, breed, gender/neuter status, was sought as well as information on adults and children in the families and on affectionate, aggressive, fearful, and playful responses of the cats to children. A convenience sample was recruited via listservs for pet owners and parents. Using a pilot tested web survey, descriptive statistics were based on 865 respondents. Multi-variate statistical analyses were conducted on data from 665 respondents with complete responses for all items, including respondents' locations and whether cats were adopted as kittens. Multi-variate analyses included consideration of demographic data, geographic region of respondents, behavioral characteristics of the cats, and responses of the children to the cats. From descriptive statistics, cats' affection was more typical with adults than young children. Neuter status or gender was unrelated to cats' aggression or affection. Being the family's only cat was associated with heightened aggression and reduced affection. Younger cats were more likely to be affectionate. Multivariate analysis revealed three primary factors accounting for children's compatibility with the specified cat: positive interactions of the cat, aggression/fearfulness of cat, and the cat's playfulness and children's reaction to the cats. Positive child-cat relationships were more typical with two or more adults and multiple cats in the home. Old cats were the least satisfactory. A breeder or shelter was a better source than as a feral, from a newspaper ad, or another source. European respondents rated their cats' interactions with children more favorably than in U.S./Canada. This difference may reflect the European adoptions more frequently being of kittens, often purebred, assuring more early handling within the family. A noteworthy finding was that all family participants, humans, and pets alike, affect the cat-child relationship, and these results reveal that many variables can play a role in achieving a desirable relationship for a cat and child.

ContextProbiotics have potential to improve health of laying hens, thus improving the overall quality of eggs. AimsA study was conducted to evaluate the use of probiotics containing Lactobacillus species to improve egg quality and serum biochemistry, and to lower the concentration of sulfur-containing gas compounds from poultry manure. MethodsNinety-six White Leghorn W-36 laying hens (32 weeks old) were randomly assigned to two feeding treatments: Control and Control + Probiotics. A combination of probiotics (Lactobacillus paracasei, L. plantarum and L. rhamnosus totalling 1 × 1012 CFU/kg feed) was provided for 8 weeks. At Weeks 0, 4 and 8, eggs were analysed for weight, shell thickness, albumen height and Haugh unit. Faecal matter was analysed for total sulfur, sulfate-sulfur and 20 reduced sulfur compounds. Blood serum was analysed for amylase, calcium, phosphate, total cholesterol and triglycerides. Key resultsNo significant differences were observed between the two treatments for any of the parameters. Five sulfide gases were detected in manure: hydrogen sulfide (H2S), dimethyl sulfide (CH3)2S, methyl mercaptan (CH3SH), carbonyl sulfide (COS) and carbon disulfide (CS2). Ratios of various detectable gases were computed. Trends for ratios H2S:(CH3)2S, H2S:CS2, and H2S:COS from manure for both treatments were >1, whereas the ratios H2S:CH3SH and (CH3)2S:CH3SH were ConclusionOverall, probiotics did not enhance production, egg quality or the serum profile. ImplicationsPossibly, the concentration of probiotics was too high, limiting bacterial colonisation and beneficial effects.

Context A practical, low-cost suggestion for industry to reduce ammonia (NH3) in layer houses is use of Lactobacillus species (L. rhamnosus, L. paracasei and L. plantarum) in drinking water or feed. Thus, we investigated their short-term (8 weeks) use in young layers. Aim A combination of species of Lactobacillus (L. rhamnosus, L. paracasei and L. plantarum at 1.0 × 1012 CFU/kg feed) was provided for laying hens in order to investigate effects on production, nitrogenous compounds in manure, serum chemistry and uric acid in the liver. Method Ninety-six White Leghorns (32 weeks old) were randomly assigned to a control diet (commercial feed) or a diet containing commercial feed + probiotics (g/kg feed: L. rhamnosus 1.667, L. paracasei 0.667 and L. plantarum 0.740) and fed for an additional 8 weeks. Key results No significant major effects were observed among diets on bodyweight, feed intake, egg production or feed conversion ratio. Numerical reductions were noted for feed intake (10%) and feed conversion ratio (9%) at Week 2 for layers receiving probiotics as compared to the control. Ammonia, ammonium-nitrogen (N), total Kjeldahl N and total N in manure were not changed significantly by probiotics, nor did probiotics significantly affect the serum profile (ammonia, uric acid, total protein, albumin and creatine kinase) or uric acid in the liver. There was a numerical but non-significant increase in creatine kinase (11%) after 8 weeks in serum of hens receiving probiotics; likewise, there was a non-significant 8% increase in uric acid concentration in the liver of hens receiving probiotics at the end of the experimental period. Conclusion The probiotics (L. rhamnosus, L. paracasei and L. plantarum at 1.0 × 1012 CFU/kg feed) used in this study did not significantly reduce N-containing compounds in manure of 32–40-week-old layers. Implications Age, different types of layers (and broilers) and mode of administration or concentration of probiotics play important roles in outcomes. Extensive collaborative studies are needed to provide definitive answers for use of probiotics in layer (as well as broiler) feed for reduction of N-containing compounds in poultry houses.

Fruit uniformity is generally regarded as an essential quality parameter in winegrape production. Deficit irrigation and cluster thinning are popular techniques to improve fruit quality, but there is little empirical information on how these practices influence fruit uniformity in the field. Deficit irrigation and targeted cluster thinning treatments were imposed in a commercial Cabernet Sauvignon vineyard in Dunnigan Hills, California, for three consecutive years. Fruit uniformity measures at three levels (blocks, vines, and clusters) were evaluated using variance components analysis and ANOVA on absolute residuals (Levene’s test). Regardless of the irrigation treatment, cluster-to-cluster variation within vines was the primary source of fruit variability each season. In general, postveraison irrigation deficits decreased cluster-to-cluster uniformity for Brix and fruit anthocyanin content. While the decrease in anthocyanin content uniformity was due to unevenness of fruit anthocyanin synthesis/loss, the decrease in Brix uniformity was due to unevenness in fruit shriveling. Cluster thinning increased Brix uniformity at veraison, but there was no effect at harvest on fruit composition and berry size uniformity. Brix uniformity was not correlated to crop load, pruning weight, or the pruning weight/crop load ratio, but an increase in Brix uniformity did occur as fruit matured.

Objective—To evaluate the thermal antinociceptive and sedative effects and duration of action of tramadol hydrochloride after oral administration to American kestrels (Falco sparverius). Animals—12 healthy 3-year-old American kestrels. Procedures—Tramadol (5, 15, and 30 mg/kg) and a control suspension were administered orally in a masked randomized crossover experimental design. Foot withdrawal response to a thermal stimulus was determined 1 hour before (baseline) and 0.5, 1.5, 3, 6, and 9 hours after treatment. Agitation-sedation scores were determined 3 to 5 minutes before each thermal stimulus test. Results—The lowest dose of tramadol evaluated (5 mg/kg) significantly increased the thermal foot withdrawal thresholds for up to 1.5 hours after administration, compared with control treatment values, and for up to 9 hours after administration, compared with baseline values. Tramadol at doses of 15 and 30 mg/kg significantly increased thermal thresholds at 0.5 hours after administration, compared with control treatment values, and up to 3 hours after administration, compared with baseline values. No significant differences in agitation-sedation scores were detected between tramadol and control treatments. Conclusions and Clinical Relevance—Results indicated oral administration of 5 mg of tramadol/kg significantly increased thermal nociception thresholds for kestrels for 1.5 hours, compared with a control treatment, and 9 hours, compared with baseline values; higher doses resulted in less pronounced antinociceptive effects. Additional studies with other types of stimulation, formulations, dosages, routes of administration, and testing times would be needed to fully evaluate the analgesic and adverse effects of tramadol in kestrels and other avian species.

Objective—To evaluate antinociceptive effects and pharmacokinetics of butorphanol tartrate after IM administration to American kestrels (Falco sparverius). Animals—Fifteen 2- to 3-year-old American kestrels (6 males and 9 females). Procedures—Butorphanol (1, 3, and 6 mg/kg) and saline (0.9% NaCl) solution were administered IM to birds in a crossover experimental design. Agitation-sedation scores and foot withdrawal response to a thermal stimulus were determined 30 to 60 minutes before (baseline) and 0.5, 1.5, 3, and 6 hours after treatment. For the pharmacokinetic analysis, butorphanol (6 mg/kg, IM) was administered in the pectoral muscles of each of 12 birds. Results—In male kestrels, butorphanol did not significantly increase thermal thresholds for foot withdrawal, compared with results for saline solution administration. However, at 1.5 hours after administration of 6 mg of butorphanol/kg, the thermal threshold was significantly decreased, compared with the baseline value. Foot withdrawal threshold for female kestrels after butorphanol administration did not differ significantly from that after saline solution administration. However, compared with the baseline value, withdrawal threshold was significantly increased for 1 mg/kg at 0.5 and 6 hours, 3 mg/kg at 6 hours, and 6 mg/kg at 3 hours. There were no significant differences in mean sedation-agitation scores, except for males at 1.5 hours after administration of 6 mg/kg. Conclusion and Clinical Relevance—Butorphanol did not cause thermal antinociception suggestive of analgesia in American kestrels. Sex-dependent responses were identified. Further studies are needed to evaluate the analgesic effects of butorphanol in raptors.

Results—Hydromorphone at 0.6 mg/kg, IM, significantly increased the thermal foot withdrawal threshold, compared with the response after administration of saline solution, for up to 3 hours, and hydromorphone at 0.1, 0.3, and 0.6 mg/kg, IM, significantly increased withdrawal responses for up to 6 hours, compared with baseline values. No significant differences in mean sedation-agitation scores were detected between hydromorphone and saline solution treatments; however, appreciable sedation was detected in 4 birds when administered 0.6 mg of hydromorphone/kg. Conclusions and Clinical Relevance—Hydromorphone at the doses evaluated significantly increased the thermal nociception threshold for American kestrels for 3 to 6 hours. Additional studies with other types of stimulation, formulations, dosages, routes of administration, and testing times are needed to fully evaluate the analgesic and adverse effects of hydromorphone in kestrels and other avian species and the use of hydromorphone in clinical settings.

Dogs’ roles to support people with disabilities are increasing. Existing U.S. laws and regulations pertaining to the use of dogs for people with disabilities are only minimally enforced. Pushback legislation against some aspects of uses of assistance dogs currently is being passed or proposed in several states. Further, the U.S. Department of the Army and the Veterans’ Administration support only dogs trained by an Assistance Dogs International (ADI) or International Guide Dog Federation (IGDF) accredited facility. Lacking a mandatory national process for screening the selection, training, and placement of assistance dogs with persons who have disabilities, the U.S. offers a creative but confusing opportunity for people to train their own dogs for any disability. While no U.S. surveillance system monitors assistance dogs, other countries generally have a legislated or regulatory process for approving assistance dogs or a cultural convention for obtaining dogs from accredited facilities. We conducted an on-line survey investigating current demographics of assistance dogs placed in 2013 and 2014 with persons who have disabilities, by facilities worldwide that are associated with ADI or IGDF, and by some non-accredited U.S. facilities. Placement data from ADI and IGDF facilities revealed that in most countries aside from the U.S., guide dogs were by far the main type of assistance dog placed. In the U.S., there were about equal numbers of mobility and guide dogs placed, including many placed by large older facilities, along with smaller numbers of other types of assistance dogs. In non-accredited U.S. facilities, psychiatric dogs accounted for most placements. Dogs for families with an autistic child were increasing in all regions around the world. Of dog breeds placed, accredited facilities usually mentioned Labrador Retrievers and Golden Retrievers, and sometimes, German Shepherd Dogs. The facilities bred their dogs in-house, or acquired them from certain breeders. Non-accredited facilities more often used dogs from shelters or assisted people in training their own dogs. Facilities in Europe and the U.S. place dogs in all roles; other parts of the world primarily focus on guide dogs. Expansion of assistance dogs in many roles is continuing, with numbers of dogs placed accelerating internationally.

The debilitating joint disorders of hip dysplasia, cranial cruciate ligament tear (CCL) and elbow dysplasia can shorten a dog's useful working life and impact its role as a family member. For this study, veterinary hospital records were examined over a 14.5-year period on 1170 intact and neutered (including spaying) German Shepherd Dogs for joint disorders and cancers previously associated with neutering. The diseases were followed through 8 years of age, with the exception of mammary cancer (MC) in females that was followed through 11 years. The cancers followed, apart from mammary, were osteosarcoma, lymphoma, hemangiosarcoma and mast cell tumour. In intact males, 7% were diagnosed with one or more joint disorders, while in males neutered prior to a year of age, a significantly higher 21% were diagnosed with one or more joint disorders. In intact females, 5% were diagnosed with one or more joint disorders, while in females neutered prior to a year of age, this measure was significantly increased to 16%. The increased joint disorder incidence mostly associated with early neutering was CCL. MC was diagnosed in 4% of intact females compared with less than 1% in females neutered before 1 year. The occurrence of the other cancers followed through 8 years of age was not higher in the neutered than in the intact dogs. Urinary incontinence, not diagnosed in intact females, was diagnosed in 7% of females neutered before 1 year, a significant difference. These findings, profiling the increase in joint disorders associated with early neutering, should help guide the timing of neutering for this breed.

The sensitivity of agricultural crops to climate change is a major area for climate impact studies. The relationship between climate and three key phenolic compounds in grape skins important to premium wine quality (anthocyanins, tannins, and total phenolics) has not been well-studied. Here we conducted a three-year field study to collect and analyze berry samples from Pinot noir vineyards in the Carneros and Sonoma Valley American Viticultural Areas of California's North Coast wine country, and correlate phenolic measurements with climate statistics derived from hourly temperature measures at each vineyard site. We used several statistical approaches to identify key phenologically-based periods influencing phenolic concentration at maturity, including classification and regression trees, factor screening, principal component analysis, and pairwise correlations. The results from these statistical models showed that cool conditions following harvest the year before maturity, warm temperatures from budburst to bloom, and cool temperatures from bloom to veraison (the onset of ripening) were positively correlated with concentrations of all three classes of phenolics, although not all trends were statistically significant. Anthocyanins were positively and significantly correlated with temperatures between 16 and 22 °C from veraison to harvest. Tannins were significantly increased by warm nights preceding budburst and warm days from budburst to bloom. We measured relatively high levels of light interception (35% of incident photosynthetically active radiation), and we found that increased light interception was significantly correlated with lower levels of all three classes of phenolic compounds in this study. For the Pinot noir sites in this study, warm temperatures from budburst to bloom appear to increase phenolic concentrations, which is likely beneficial for wine quality. However, warmer periods during the preceding fall and summer during ripening appear to offset these effects. Given projections for greater summer warming in California with climate change, the overall impact of climate change on winegrowing is likely to be negative.

To further our understanding of the response of ‘Bartlett’ pear fruit to 1-methylcyclopropene (1-MCP) and their ability to recover the capacity to ripen, ‘Bartlett’ pears were treated with 0.3 μL L −1 1-MCP for 12 h at 20 °C immediately after harvest in two seasons and to pear fruit of four maturities. 1-MCP decreased rates of softening, ethylene production, respiration, and yellow color development, and reduced incidence of scald and internal breakdown. Ripening recovery induced by cold storage of 1-MCP treated fruit depended on maturity and season and was associated with stimulated ethylene production, including 1-aminocyclopropene carboxylic acid synthase (ACS) activity, 1-aminocyclopropene carboxylic acid oxidase (ACO) activity, and transcript levels of genes associated with these enzymes.

Preharvest applications of 1-methylcyclopropene (1-MCP) were tested on California ‘Bartlett’ pears at 80 N maturity and at rates of 0, 28, and 56 mg·L−1 in 2006 and 0, 50, and 100 mg·L−1 in 2007. In 2007, a parallel experiment was conducted to compare 50 mg·L−1 1-MCP with 96 g a.i./ha 1-naphthaleneacetic acid (NAA) used commercially to control or decrease premature fruit drop. Premature fruit drop, maturity, firmness at harvest, color, softening, and ethylene production during ripening and physiological disorders were studied in fruit harvested between 7 and 21 days after 1-MCP application and either ripened at 20 °C immediately after harvest or after 3.5 to 6 months storage at –1 °C. Overall, 50 mg·L−1 1-MCP reduced the incidence of premature fruit drop when compared with the untreated fruit and fruit drop was similar to adjuvant-treated fruit and NAA-treated fruit, especially 28 days or longer after the treatment. 1-MCP was more effective in retarding color, softening, and ethylene production during ripening than delaying fruit maturation on the tree (loss of firmness), and both rates of 1-MCP tested each season yielded similar fruit responses on most evaluation times. 1-MCP's effect on ripening was lost if fruit remained on the tree 21 days or after the fruit were stored for 3.5 months in cold storage regardless of treatment concentration. A reduction of internal breakdown incidence was observed in 1-MCP-treated fruit.

The EPA regulates ambient particulate matter (PM) because substantial associations have been established between PM and health impacts. Presently, regulatory compliance involves broad control of PM emission sources based on mass concentration rather than chemical composition, although PM toxicity is likely to vary depending upon PM physicochemical properties. The overall objective of this study was to help inform source-specific PM emission control regulations. For the first time, source-oriented PM was collected from the atmosphere in Fresno, CA, onto 38 source/size substrates. Mice were exposed via oropharyngeal aspiration to equivalent mass doses [50 μg] of two size fractions: ultrafine (Dp < 0.17μm) and submicron fine (0.17 < Dp < 1 μm) during summer and winter seasons. At 24 hours post-exposure, cellular and biochemical indicators of pulmonary inflammation were evaluated in the bronchoalveolar lavage fluid. Significant inflammatory responses were elicited by vehicle, regional background, and cooking PM sources that were dependent on season and particle size. This is the first study of source-oriented toxicity of atmospheric PM and supports source-specific emissions control strategies.

Pto is a serine/threonine kinase that mediates resistance in tomato to strains of Pseudomonas syringae pv. tomato expressing the (a)virulence proteins AvrPto or AvrPtoB. DNA shuffling was used as a combinatorial in vitro genetic approach to dissect the functional regions of Pto. The Pto gene was shuffled with four of its paralogs from a resistant haplotype to create a library of recombinant products that was screened for interaction with AvrPto in yeast. All interacting clones and a representative sample of noninteracting clones were sequenced, and their ability to signal downstream was tested by the elicitation of a hypersensitive response in an AvrPto-dependent or -independent manner in planta. Eight candidate regions important for binding to AvrPto or for downstream signaling were identified by statistical correlations between individual amino acid positions and phenotype. A subset of the regions had previously been identified as important for recognition, confirming the validity of the shuffling approach. Three novel regions important for Pto function were validated by site-directed mutagenesis. Several chimeras and point mutants exhibited a differential interaction with (a)virulence proteins in the AvrPto and VirPphA family, demonstrating distinct binding requirements for different ligands. Additionally, the identification of chimeras that are both constitutively active as well as capable of binding AvrPto indicates that elicitation of downstream signaling does not involve a conformational change that precludes binding of AvrPto, as previously hypothesized. The correlations between phenotypes and variation generated by DNA shuffling paralleled natural variation observed between orthologs of Pto from Lycopersicon spp.

Disease significantly affects population abundance of Pacific herring (Clupea pallasi). Comprehensive epidemiological study of the Pacific herring population of Prince William Sound, Alaska, U.S.A., from 1994 to 2000 included complete necropsy examination of 230–500 fish each spring and 40–160 fish each fall (total n = 2983 fish). Mortality is best estimated, through modifications of an age-structured assessment model, using a disease index that combines the prevalence of viral hemorrhagic septicemia virus (VHSV) with the prevalence of ulcers. Risk factors for an epidemic include poor body condition and abundant recruitment before spawning in the spring. Prevalence of the pathogen Ichthyophonus hoferi increased as fish aged, but changes in I. hoferi prevalence were not related to changes in population abundance. Disease that caused an epidemic in 1998 (VHSV and ulcers) nearly disappeared from the population when changes in abundance were detected by traditional stock assessment methods in 1999. Disease significantly affects recruitment — the two lowest recruitment estimates on record, in 1994 and 1999, followed increased natural mortality of adults in 1993 and 1998.

Objectives To evaluate the effects of a multiple daily dose methadone regimen in pregnancy on neonatal outcomes. Methods Although methadone maintenance has been the standard for the treatment of opioid dependence in pregnancy, there is no consensus on proper dosing. Single daily dosing is the most common strategy. Because of accelerated metabolism of methadone in pregnancy, this regimen may expose mother and fetus to daily episodes of withdrawal and possibly contribute to more severe Neonatal Abstinence Syndrome (NAS). This study reports on a protocol that increased both methadone dose and dose frequency in response to maternal reports of withdrawal. Results Treatment of NAS was needed in 29% of neonates, compared to a published rate of 60% to 80%. The mean methadone dose was 152 mg at delivery, divided into 2 to 6 doses per day. Ninety-two percent of mothers were free of illicit drug use at delivery. There was no relationship between methadone dose and treatment of NAS. Female babies had a treatment rate of 16% versus 38% for male babies. Beyond abstinence symptoms, cohort outcomes in terms of gestational age, birth weight, prematurity, Caesarian sections, and breastfeeding equaled or approximated US population norms. Conclusions The protocol was associated with low rates of treatment of NAS and high rates of maternal recovery. High rates of treatment for NAS reported in methadone-exposed neonates might relate in part to iatrogenic factors and be reduced through the use of divided daily doses and protocols that minimize maternal withdrawal.

Objective: To characterize the impact of age, gender, location and individual animal variation on the composition of articular cartilage from the metacarpophalangeal joint of horses. Design: Cartilage specimens were obtained from the metacarpophalangeal joints of 28 male, female and castrated male horses ranging in age from one day to 27 years of age. Cartilage samples from the distal metacarpus, proximal first phalanx and proximal sesamoids were analyzed separately. Chondrocyte number, DNA content, proteoglycan concentration and total collagen content were determined for each animal and joint location. Results: Age and joint location had a significant effect on chondrocyte number and DNA content with higher cell counts and DNA content detected in cartilage from the youngest age groups and in cartilage from the metacarpus and proximal sesamoids. The influence of age on chondrocyte numbers was not significant in horses over two years of age. Both age and joint location also influenced total proteoglycan and collagen content. Lower proteoglycan and collagen concentrations were detected in younger horses, and cartilage from the metacarpus had lower proteoglycan and collagen concentrations than that from other joint locations. Gender did not appear to influence chondrocyte number or matrix content of equine articular cartilage. However, there was significant residual variation in cellularity, proteoglycan levels and collagen content between individual animals that could not be explained by the signalment factors considered in this study. Conclusions: Future studies examining equine articular cartilage should avoid direct morphologic comparisons between animals of different ages, and any comparisons made between individuals should be interpreted cautiously. In addition, in vitro tissue culture models should avoid the use of cartilage pooled from different animals or from different locations within the same joint.

In investigating the question of the existence of "induced demand" in connection with highway expansion projects, Hansen et al. (1993) studied eighteen California state highway segments whose capacities had been improved in the early 1970s. For the present study, these segments were paired with control segments that matched the improved segments to unimproved ones with regard to facility type, region, approximate size, and initial volumes and congestion levels. Taking annual data for average daily traffic (ADT) and design-hour-traffic-to-capacity (DTC) ratios during the 21 years 1976–1996, three approaches were used to compare growth rates between the improved and unimproved segments: overall growth comparisons for the matched pairs, repeated measures analysis, and analysis of matched mean profiles. We found the growth rates between the two types of segments to be statistically and practically indistinguishable, suggesting that the capacity expansions, in and of themselves, had a negligible effect on traffic growth over the period studied. Reasons for the differences between these results and those of aggregate cross-sectional models finding a significant induced demand effect are discussed. Our analyses suggest that the aggregate models may overestimate induced traffic due to the attribution of at least a fraction of the observed traffic growth to "induced demand" rather than to some of the confounding factors which were not controlled for in such studies. At the same time, it is noted that the traffic induced by capacity expansion may in certain circumstances be larger than that observed in the present study, with the effect of new highway construction on traffic growth being a prime candidate for scrutiny in this regard. The results of this study nonetheless suggest that, for existing facilities, the size of the induced-traffic effect that can be attributed to capacity enhancements may be sufficiently small that its detection in a case-control study would be difficult, if not impossible, without a substantially larger sample size.

Objective—To develop an objective, accurate method for quantifying forelimb ground reaction forces in horses by adapting a human in-shoe pressure measurement system and determine the reliability of the system for shod and unshod horses. Animals—6 adult Thoroughbreds. Procedure—Horses were instrumented with a human in-shoe pressure measurement system and evaluated at a trot (3 m/s) on a motorized treadmill. Maximum force, stance time, and peak contact area were evaluated for shod and unshod horses. Three trials were performed for shod and unshod horses, and differences in the measured values were examined with a mixed model ANOVA for repeated measures. Sensor accuracy was evaluated by correlating measured variables to clinically observed lameness and by a variance component analysis. Results—4 of 6 horses were determined to be lame in a forelimb on the basis of clinical examination and measured values from the system. No significant differences were observed between shod and unshod horses for maximum force and stance time. A significant decrease in peak contact area was observed for shod and unshod horses at each successive trial. Maximum force measurements provided the highest correlation for detecting lameness ( r = 0.91, shod horses; r = 1.0, unshod horses). A variance component analysis revealed that 3 trials provided a variance of 35.35 kg for maximum force (± 5.78% accuracy), 0.007 seconds for stance time (± 2.5% accuracy), and 8.58 cm2 for peak contact area (± 11.95% accuracy). Conclusions and Clinical Relevance—The in-shoe pressure measurement system provides an accurate, objective, and effective method to evaluate lameness in horses. ( Am J Vet Res 2001;62:23–28)

Objectives— To compare the biomechanical properties, in full limb preparations, of intact second phalanx and a simulated comminuted second phalangeal fracture stabilized with either two bone plates or a custom Y-plate. Study Design— In vitro biomechanical assessment of intact limbs and of paired limbs with a simulated second phalangeal fracture stabilized by one of two fixation methods. Animal Population Thirteen pairs of equine cadaveric forelimbs. Methods— A comminuted second phalangeal fracture was created in six paired cadaveric limbs. For each limb pair, the fracture was stabilized with two plates in one limb, and with a Y-plate in the contralateral limb. These limbs and seven pairs of intact limbs were subjected to axial compression in a single cycle until failure. Mechanical properties were compared with a mixed-model ANOVA and post hoc contrasts. Joint contact pressure, screw insertion torque, and final screw torque remaining after mechanical testing were also evaluated for constructs. Results— No significant differences in mechanical testing variables were detected between construct types. However, the Y-Plate construct had significantly greater yield load, yield displacement and yield energy, and failure load and stiffness values than those for intact specimens, whereas the double-plate construct only had greater stiffness than intact specimens. There were no significant differences in joint contact pressures for both constructs. The final screw torque for proximal phalangeal screws was significantly greater for the Y-plate constructs than for double-plate constructs. Conclusions— The Y-plate was as effective as the double-plate technique for stabilization of simulated comminuted second phalangeal fractures in monotonically tested equine cadaveric forelimbs. Clinical Relevance— This investigation supports evaluation of the Y-plate for repair of comminuted second phalangeal fractures in equine patients. Its specific design may facilitate repair of second phalangeal fractures, and may provide increased stability by allowing the proximal fragments of the second phalanx to be fixed with three screws placed through the plate.

AIM—To assess whether treatment of premature infants with dopamine is a risk factor for development of retinopathy of prematurity (ROP). METHODS—A retrospective case series analysis of two groups was utilised with a minimum follow up of 6 months. Clinical profiles and patient risk factors were identified along with an evaluation of ROP progression and an analysis of clinical outcome. All infants were seen in a single community neonatal intensive care unit (NICU). 41 consecutive high risk infants were identified during a 36 month period whose birth weight was less than 1000 grams and who remained in the NICU without transfer until at least 28 days of age. Dilated indirect ophthalmoscopy fundus examinations were performed on all infants to identify the degree of and progression to threshold ROP. RESULTS—18 of 41 infants were treated with dopamine for hypotension. The group of infants requiring dopamine differed statistically from the non-dopamine treated group by having a slightly higher birth weight, a greater incidence of hypotension and colloid treatment, and in manifesting more advanced respiratory disease. Within the dopamine treated group, 12 of 18 infants (67%) reached prethreshold ROP and seven infants (39%) reached threshold ROP requiring laser treatment. In contrast, only three of the infants (13%) who did not require dopamine for hypotension progressed to prethreshold (p=0.001) and only one of these infants (4%) progressed to threshold ROP (p = 0.02). Logistic regression analysis among other variables demonstrated that dopamine use and gestational age are important factors in this low birthweight population for predicting the development of threshold ROP (dopamine use: adjusted odds ratio = 119.88, p = 0.0061; gestational age: adjusted odds ratio = 0.061, p = 0.0043). CONCLUSIONS—Dopamine use in low birthweight infants may therefore be a risk factor for the development of threshold ROP. More vigilant screening of high risk infants requiring dopamine therapy for systemic hypotension may be warranted. Keywords: dopamine; retinopathy of prematurity; infants

Summary The objective of this study was to examine several simple ex vivo loading conditions for the equine metacarpus, and to evaluate their ability to reproduce the mid-diaphyseal bone surface strain distributions previously reported in vivo at the walk and trot. Distributed axial compressive loads, and 9 different axial compressive point loads at −7.5 kN and −15 kN were applied to metacarpal-distal carpal bone preparations from 6 Thoroughbred horses, aged 1–5 years. The resulting dorsal, medial, palmar, and lateral mid-diaphyseal bone surface axial and shear strains were compared with previously reported in vivo surface strain distributions using a root mean square error (RMSE) protocol. The effects of loading condition and load magnitude on RMSE were assessed with a mixed-model analysis of variance. There were significant differences between loading conditions, and, in most cases, between load magnitudes, in the fit of the ex vivo to the in vivo strain distributions. In vivo mid-diaphyseal bone surface strain distributions at the walk can be well approximated ex vivo by a distributed axial compressive load, or by a point load positioned 0.5 cm mediad to the sagittal midline, at −7.5 kN loads. In vivo mid-diaphyseal bone surface strain distributions at the trot can be well approximated by the −15 kN loads applied to the same locations. These simplified loading conditions can be used in designing biologically relevant loading protocols for ex vivo mechanical testing studies, as well as in developing boundary conditions for finite element analysis. As such, these loading conditions may be considered as tools to be used as a means of replicating in vivo loading conditions during the intial design and testing stages in the development of fracture fixation devices, as well as in the theoretical mechanical analysis of the metacarpal structure.

Social interactions in red-backed salamanders, Plethodon cinereusare complex and often depend upon the sex and reproductive status of individuals. In the population at Mountain Lake Biological Station, Virginia, adult males outnumber sexually active (gravid) females by approximately 2:1, because males court annually while females accept mates only biennially. Field observations of male P. cinereus suggest that males maintain feeding territories and allow non-gravid (not sexually active) females access to their territories for foraging, whereas they defend their territories against intrusion by males. We used a game-theoretic analysis of models of male fitness to examine conditions that would favour the evolution of this 'permissive' behaviour. Under a series of increasingly realistic behavioural assumptions, we found that the critical factor in the evolution of male permissive behaviour is female preference for permissive males. If non-gravid females are more likely to return to mate with a male that allows access to his territory, then permissive behaviour is likely to evolve. Furthermore, the evolution of permissive behaviour is facilitated when energetic losses due to territorial defence exceed those due to allowing females to forage.1997The Association for the Study of Animal Behaviour

Development of pink salmon (Oncorhynchus gorbuscha) incubating in gravel contaminated with weathered Prudhoe Bay crude oil was retarded at concentrations as low as 55.2 μg oil/g gravel. Larvae exposed to various levels of oil contamination were sampled 4 weeks before emergence, at emergence, and 13 days after emergence for histopathology (quantitative and semiquantitative) and cytochrome P4501A (CYP1A) induction (using immunohistochemical staining). A subset of postemergent fish was not fed. Hydrocarbon analysis by gas chromatography and mass spectroscopy revealed that tissue uptake of polynuclear aromatic hydrocarbons (PAH) was mediated by oil's dissolution in water, with significant biological effects when the peak total PAH concentration in water was as low as 4.4 μg/L. Oil-related effects included induction of CYP1A, development of ascites, and increased mortality. Several oil-related changes were indicative of premature emergence. Compared with control fish, for example, exposed fish of the same age and emerging on the same day had greater amounts of yolk and hepatocellular glycogen, increased apoptosis of gonadal cells and midventral skin cells, and less food in the gastrointestinal tract. Histological features were similar within groups of larvae sampled 4 weeks before and 13 days after emergence, and oil-induced changes were not affected by feeding during the first 13 days after emergence. Increased gonadal cell apoptosis may be related to later reproductive impairment documented in field studies of pink salmon up to 4 years after the Exxon Valdez oil spill.

The distribution of material properties within the equine third metacarpal bone (MC3), and its possible effect on the mechanics of the structure, was quantitatively evaluated using single-load-to-failure compressive materials testing of specimens from ten horses. Bone samples from six regions within five proximodistal levels of MC3 were milled into right cylinders and compressed at a strain rate of 0.01 s−1. Diaphyseal MC3 bone material was stiffer, stronger, deformed less to yield and failure, and absorbed more energy to yield, than metaphyseal cortical bone material. Lateral and medial MC3 cortical bone material was stiffer and deformed less to yield and failure, than dorsal and palmar material. This distribution of material properties appears to increase the structural compliance in the sagittal plane, and may serve to enhance the predictability of the strain distribution during normal locomotion, as is provided in other bones by a sagittal curvature.

The effects of weight gain and subsequent weight loss on glucose tolerance and insulin response were evaluated in 12 healthy cats. Intravenous glucose tolerance tests (IVGTT) were performed at entry into the study, after a significant gain of body weight induced by feeding palatable commercial cat food ad libitum, after a significant loss of body weight induced by feeding a poorly palatable purified diet to discourage eating and promote fasting, and after recovery from fasting when body weight had returned to pre-study values and cats were eating commercial foods. A complete physical examination with measurement of body weight was performed weekly, a CBC and serum biochemistry panel were evaluated at the time of each IVGTT, and a liver biopsy specimen obtained 2 to 4 days after each IVGTT was evaluated histologically for each cat. Mean serum glucose and insulin concentrations after glucose infusion and total amount of insulin secreted during the second 60 minutes and entire 120 minutes after glucose infusion were significantly (P < .05) increased after weight gain, as compared with baseline. At the end of weight loss, cats had hepatic lipidosis and serum biochemical abnormalities consistent with feline hepatic lipidosis. There was a significant (P < .05) increase in mean serum glucose concentration and t1/2, and a significant (P < .05) decrease in mean serum insulin concentration and the glucose disappearance coefficient (K) after glucose infusion for measurements obtained after weight loss, compared with those obtained after weight gain and at baseline. Insulin peak response, insulinogenic index, and total amount of insulin secreted during the initial 10 minutes, 20 minutes, and 60 minutes after glucose infusion were decreased markedly (P < .05), compared with measurements obtained after weight gain and at baseline. In addition, the total amount of insulin secreted for 120 minutes after glucose infusion was decreased markedly (P < .05) in measurements obtained after weight loss, compared with those obtained after weight gain. At the end of recovery, all cats were voluntarily consuming food, serum biochemical abnormalities identified after weight loss had resolved, the number and size of lipid vacuoles in hepatocytes had decreased, and results of IVGTT were similar to those obtained at baseline. These findings confirmed the reversibility of obesity-induced insulin resistance in cats, and documented initial deterioration in glucose tolerance and insulin response to glucose infusion when weight loss was caused by severe restriction of caloric intake.

Most experiments conducted to determine the optimal essential amino acid (EAA) nitrogen to total nitrogen (E:T) ratio in rats, chicks and other species have shown that weight gain and nitrogen retention are optimal when the ratio is between 0.5 and 0.65. Two experiments were conducted to determine if weight gains of kittens fed EAA as a sole source of dietary nitrogen were equivalent to those of kittens fed a control diet that contained equal amounts of EAA and dispensable amino acids (DAA). In the first experiment, kittens fed diets that contained only EAA lost weight. Amino acid analysis of plasma showed that the concentration of methionine was 9.1 times that of controls, supporting the premise that the weight loss that occurred was the result of an adverse effect of excess methionine. Also, plasma threonine and arginine were elevated whereas proline and asparagine were very low. In the second experiment, feeding lower concentrations of methionine and arginine in a diet containing only EAA resulted in weight gains and nitrogen retention that were not significantly different from results for kittens fed the control diet (E:T ratio = 0.5). The addition of proline and asparagine to this EAA diet or their removal from the control diet did not improve or reduce weight gain or nitrogen retention. It is concluded that the decreased weight gain found in kittens (and probably other species) fed only EAA is the result of an adverse effect of excesses of methionine and possibly other EAA, and not the inability to synthesize DAA. If excesses of certain EAA are avoided, near-maximal weight gain can be achieved without any DAA in the diet.

The torsional monotonic structural material properties of equine metacarpi with or without, either a 5/16 inch or 3/8 inch diameter bicortical lateromedial middiaphyseal hole were assessed to determine the effect of a hole on metacarpal strength. Torsional stiffness was not significantly effected by the presence of a bicortical hole, whereas yield and failure angles, torques and energies of metacarpi with a hole were 51% to 97% of those of intact bones. Significant differences were not apparent for yield and failure mechanical properties between metacarpi with a 5/16 inch diameter hole and metacarpi with a 3/8 inch diameter hole; however, postyield mechanical properties were lower for metacarpi with a 3/8 inch hole. Whereas some metacarpi with a 5/16 inch hole were capable of plastic deformation before failure, metacarpi with a 3/8 inch diameter hole appeared to have sufficient stress concentration to propagate complete fracture on structural yield.

In contrast to European countries, the overwhelming majority of dogs in the U.S. are neutered (including spaying), usually done before one year of age. Given the importance of gonadal hormones in growth and development, this cultural contrast invites an analysis of the multiple organ systems that may be adversely affected by neutering. Using a single breed-specific dataset, the objective was to examine the variables of gender and age at the time of neutering versus leaving dogs gonadally intact, on all diseases occurring with sufficient frequency for statistical analyses. Given its popularity and vulnerability to various cancers and joint disorders, the Golden Retriever was chosen for this study. Veterinary hospital records of 759 client-owned, intact and neutered female and male dogs, 1-8 years old, were examined for diagnoses of hip dysplasia (HD), cranial cruciate ligament tear (CCL), lymphosarcoma (LSA), hemangiosarcoma (HSA), and mast cell tumor (MCT). Patients were classified as intact, or neutered early (

Developmental morphology of the forelimb distal phalanges and lateral palmar processes of 9 Thoroughbred foals aged 3–32 weeks was assessed using radiography, microradiography and histology. For inclusion in the study, all distal phalanges had no pathologic radiographic abnormalities. Vascular channels that are characteristically found in the dorsal region of the distal phalanx were not evident radiographically in the palmar process. The proximal and distal angles of the palmar processes were separated by a lucent line continuous with the incisure in foals only through 12 weeks of age. The distal phalanges were triangular-shaped in foals 3–12 weeks of age, and were oval-shaped in older foals. The palmar aspect of the distal phalanx was the major contributor to growth of the distal phalanx in the sagittal plane, especially between 3 and 12 weeks of age. Growth of the lateral palmar process occurred through the means of endochondral ossification. The body and cortices of the lateral palmar process were composed of coarse cancellous bone. Porosity within the lateral palmar process was greater in regions sampled axial, compared to abaxial, to the parietal sulcus and did not change with age. A fracture was identified microradiographically and/or histologically in 9 of 18 (50%) and 10 of 17 (59%), respectively, of the lateral palmar processes examined. The fracture line was consistently associated with the parietal sulcus on the dorsal cortical surface and was always immediately abaxial to the deep digital flexor tendon attachment. No age-related morphological changes of the lateral palmar processes were identified with microradiography or histological examinations.

In monkeys, dogs and swine (six each) we tested the reduction of the isoflurane MAC (minimal alveolar concentration) produced by 2 mg.kg-1 morphine intravenously (i.v.) and the concurrent effect on PCO2 with spontaneous ventilation. MAC fell to a minimum of 55% of control at 53 min in monkeys, 50% at 38 min in dogs and 13% at 33 min in swine. PaCO2 rose at constant MAC with morphine to 55-60 mmHg, but did not fall over the next several hours despite the decline of plasma morphine concentration, and the resulting needed rise in isoflurane concentration to keep the anaesthesia depth at 1 MAC. After isoflurane concentration had returned to pre-morphine control levels, naloxone immediately reduced PaCO2 to or below control level. Morphine pharmacokinetics in the three species studied conformed to a two-compartment model.

Purpose To determine the effect of conjunctival incision location on the long-term efficacy of nonvalved glaucoma drainage devices. Materials and methods We conducted a retrospective review of patients ≥18 years of age with uncontrolled glaucoma [intraocular pressure (IOP) ≥18 mm Hg] who underwent glaucoma drainage device implantation. A comparison was made of a limbal-based (LB-BGI) versus fornix-based (FB-BGI) conjunctival flap during placement of a 350-mm Baerveldt glaucoma implant (AMO, Santa Ana, CA) in subjects with at least 1 year of follow-up data. The primary outcome measure was IOP; secondary outcome measures were medication burden, visual acuity, and surgical complications. Results One hundred sixty eyes of 147 glaucoma patients were included. Two years after surgery, the IOP in the LB-BGI group was 14.3±5.3 mm Hg and in the FB-BGI group 13.1±4.7 mm Hg (P=0.47). Overall success of IOP control was achieved at the final visit (range 1 to 5 y) in 90% of the LB-BGI group and 87% of the FB-BGI group (P=0.63). The medication burden of the 2 groups at 1 and 2 years after surgery was not statistically significantly different. Worsening of visual acuity by more than 2 lines was not statistically different between the groups 2 years after the surgery and at the final visit (P=0.47, P=0.60, respectively). A greater number of eyes developed endophthalmitis and were more likely to undergo subsequent tube revision in the FB-BGI group, but the differences were not significant. Conclusions Both incision techniques were equally effective in controlling IOP. Each surgical approach has its advantages and this study suggests that either technique may be used safely and effectively.

Results of recent investigations in humans and dogs indicate that gravity-independent factors may be important in determining the distribution of pulmonary blood flow. To further evaluate the role of gravity-independent factors, pulmonary blood flow distribution was examined using 15-microns radionuclide-labeled microspheres in five prone ponies over 5 h of pentobarbital sodium anesthesia. The ponies were killed, and the lungs were excised and dried by air inflation (pressure 45 cmH2O). The dry lungs were cut into transverse slices 1–2 cm thick along the dorsal-ventral axis, parallel to gravity. Radioactivity of pieces cut from alternate slices was measured with a gamma well counter. The main finding was a preferential distribution of pulmonary blood flow to dorsal-caudal regions and higher flow in the center of each lung slice when compared with the slice periphery. Flow was lowest in cranial and ventral areas. Differences of +/- 2 SD were observed between core and peripheral blood flow. No medial-lateral differences were found. Pulmonary blood flow distribution did not change over 5 h of anesthesia, and the basic flow pattern was not different in the left vs. right lung. These results suggest that in the intact prone mechanically ventilated pony (inspired O2 fraction greater than or equal to 0.95) factors other than gravity are primary determinants of pulmonary blood flow.

In many animals, exposure to cues in a natal habitat increases disperser preferences for those cues (natal habitat preference induction [NHPI]), but the proximate and ultimate bases for this phenomenon are obscure. We developed a Bayesian model to study how different types of experience in the natal habitat and survival to the age/stage of dispersal interact to affect a disperser's estimate of the quality of new natal-type habitats. The model predicts that the types of experience a disperser had before leaving its natal habitat will affect the attractiveness of cues from new natal-type habitats and that favorable experiences will increase the level of preference for natal-type habitats more than unfavorable experiences will decrease it. An experimental study of NHPI in Drosophila melanogaster provided with "good" and "bad" experiences in their natal habitats supports these predictions while also indicating that the effects of different types of natal experience on NHPI vary across genotypes. If habitat preferences are modulated by an individual's experience before dispersal as described in this study, then NHPI may have stronger effects on sympatric speciation, metapopulation dynamics, conservation biology, and pest management than previously supposed.

The present work describes a two-stage approach to analyzing combustion-generated samples for their potential to produce oxidant stress. This approach is illustrated with the two commonly encountered transition metals, copper and iron. First, their abilities to generate hydroxyl radical were measured in a cell-free, phosphate-buffered saline solution containing ascorbate and/or citrate. Second, their abilities to induce heme oxygenase-1 in cultured human epidermal keratinocytes were assessed in cell culture. Combustion-generated copper oxide nanoparticles were active in both assays and were found to be soluble in culture medium. Depletion of glutathione in the cells or loading the cells with ascorbate greatly increased heme oxygenase-1 induction in the presence of copper. By contrast, iron oxide nanoparticles were active in the phosphate buffered saline but not in cell culture, and they aggregated in culture medium. Soluble salts of copper and iron exhibited the same contrast in activities as the respective combustion-generated particles. The results suggest that the capability of combustion-generated environmental samples to produce oxidant stress can be screened effectively in a two step process, first in phosphate buffered saline with ascorbate and subsequently in epithelial cell culture for those exhibiting activity initially. The results also point to an unanticipated interaction in cells of oxidant stress-generating metals with an anti-oxidant (ascorbate) that is usually missing in culture medium formulations. Thus, ascorbate supplementation of cultured human cells is likely to improve their ability to model the in vivo effects of particulate matter containing copper and other redox-active metals.

Koi herpesvirus (KHV) has been associated with devastating losses of common carp (Cyprinus carpio carpio) and koi (Cyprinus carpio koi) in North America, Europe, Israel and Asia. A comparison of virion polypeptides and genomic restriction fragments of seven geographically diverse isolates of KHV indicated that with one exception they represented a homogeneous group. A principal environmental factor influencing the onset and severity of disease is water temperature. Optimal growth of KHV in a koi fin cell line occurred at temperatures from 15–25 °C. There was no growth or minimal growth at 4, 10, 30 or 37 °C. Experimental infections of koi with KHV at a water temperature of 23 °C resulted in a cumulative mortality of 95·2 %. Disease progressed rapidly but with lower mortality (89·4–95·2 %) at 28 °C. Mortality (85·0 %) also occurred at 18 °C but not at 13 °C. Shifting virus-exposed fish from 13–23 °C resulted in the rapid onset of mortality.

Single-fiber electromyography (SFEMG), a technique used to investigate neuromuscular transmission, has been described previously in the pelvic limb of dogs. Because preferential involvement of isolated muscle groups can occur in disorders of neuromuscular transmission, SFEMG was done in the peroneus longus (PL), extensor carpi radialis (ECR), and orbicularis oculi (OO) muscles of 10 adult, clinically normal dogs. Jitter was calculated as the mean absolute value of the consecutive differences in latency of 50 single muscle fiber action potentials after stimulation of intramuscular nerve bundles at the level of the motor point in at least 20 muscle fibers per muscle. Bilateral recordings were performed in 3 dogs. Mean jitter values were determined for each muscle, and differences among muscle groups and among dogs were compared. The upper limits of mean consecutive difference (mean plus 3 standard deviations) for the PL, ECR, and OO muscles were 21.94, 22.53, and 23.39 micros, respectively, and the upper limit of mean consecutive difference for individual muscle fibers in the respective fiber pools was 28.62, 36.39, and 35.68 micros. Jitter values for the ECR and OO were significantly higher than the jitter value for the PL muscle (P < .05). Significant differences among muscles or dogs or between sides were not observed for the ECR. Significant differences among dogs were observed for OO jitter values and were attributed to extremely low jitter values in 1 dog. Significant differences were demonstrated between sides for the PL and were attributed to small sample size. Results of this study provide normative data that can be used in the application of the stimulated SFEMG technique to dogs with suspected disorders of neuromuscular transmission.
