The presence of waste in coastal environments can lead to increased coastal damage and burden. Most of the population's activities in Bulukumba Regency are concentrated in coastal areas, thus making this region susceptible to significant pressure from waste pollution. This research aims to determine the level of coastal area sensitivity in Bulukumba towards waste pollution. The study was conducted from October to December 2022. The research location is the coastal area of Bulukumba Regency, which includes seven subdistricts: Gantarang, Ujung Bulu, Ujung Loe, Bonto Bahari, Bontotiro, Herlang, and Kajang. Primary data were obtained through interviews and direct observations at the research locations, while secondary data were collected through literature studies and relevant institutions in Bulukumba. The results of parameter weighting using the expert judgment method indicate that five important parameters are used to assess the sensitivity of the coastal environment to waste pollution. These parameters consist of current velocity (20.27%), distance of the ecosystem from the harbor (18.92%), distance of the ecosystem from settlements (18.92%), distance of the ecosystem from rivers (17.57%), and the presence of waste on the coast (17.57%). The distribution of coastal environmental sensitivity levels to waste pollution shows that the eastern coastal areas are more sensitive to waste pollution than the southern coastal areas. The current velocity is the most significant parameter influencing the coastal environment's sensitivity to waste pollution and holds the highest weight and score across all research areas.

Black band disease (BBD) is one of the oldest recognized diseases of scleractinian corals. This disease is little known on the variation of progression rates across relatively small spatial scales and how local variations in the environment can impact prevalence and spread. The purpose of this study was to explore the progression of BBD on genus Montipora in relation to spatio-temporal environmental parameters in two islands of the Seribu Islands, North Jakarta, Indonesia during dry season and rainy season. Monthly underwater pictures were taken for determining the progress of disease level. Interestingly, the progression rate of the disease recorded at Pramuka Island was higher (8.10 cm2 day-1) than the one at Pari Island (3.79 cm2 day-1). In Pramuka Island, the infected corals had almost 89% of the dead surface, compared to only 68% at Pari Island. Similar to other studies in the region, we confirmed that the disease progressed faster during the dry season, where the environmental parameters, such as temperature, light intensity, and phosphate, were starting to increase, while total organic matter, current flow rates, and turbidity were lower. Biodiversity assessment surveys are necessary for establishing conservation areas. However, such surveys are typically expensive, primarily if they cover a large area and take a long time. The survey difficulty increases when applied to cryptic, sparse, and fast-moving organisms. In addition, it requires expertise in taxonomic-biota classification. The breakthrough environmental DNA (eDNA) metabarcoding technique promises to overcome all the hurdles of assessing the potential for marine biodiversity in a non-invasive, rapid, extensive, and more effective way. We evaluated the ability of the eDNA survey to reveal the potential diversity and character of marine eukaryotes in the Lombok Island Marine Protected Area. A sampling of seawater and sediment eDNA in pore size fractions of 0.4-12 ?m and >12 ?m was carried out in the Core Zone, Non-Core Zone, and Non-Conservation Area, in east, north, and west Lombok, respectively. The detection and classification of eukaryotes using bioinformatics analysis were accomplished following extraction, amplification, and DNA sequencing. We identified 20,478 unique sequences of potential species classified in five kingdoms to 654 marine eukaryotes families. The comparison results show differences in community structure between locations, as well as differences in diversity between media and factions. Fisheries activity in Lampung Bay which has been very active in recent years is the cultivation of green mussels (P. viridis) for consumption. Located in Teluk Betung Timur sub-district, this is considered appropriate because of the environmental parameters that support the life of P. viridis. The location of P. viridis cultivation is not far from coastal settlements (±500-600 m from settlements). Anthropogenic activities of coastal residents as well as the use of plastic materials in the cultivation are concerned to be the source of microplastic contamination that found in P. viridis. This study aims to determine the presence and characteristics of microplastics in P. viridis as well as to compare the abundance of microplastics in the gills and digestive organs. Taking 30 P. viridis by purposive sampling with homogeneous size. The samples were analyzed by microplastics in chemical oceanography and biology micro Laboratory, FPIK, IPB University. P. viridis tissue was destructed by using 10% KOH solution at a ratio of 1:3 (m/v) at 60 °C for 24 hours and then filtered using a 1.6 µm Whatman GF/A filter paper. The green seaweed Caulerpa racemosa is a seaweed of high prospect that is being given emphasis by the Indonesian government. However, C. racemosa in Indonesia may include multiple species level-entity exhibiting morphological overlap and require molecular analysis for species identification. Molecular documentation of species richness of indigenous populations of C. racemosa is essential to underpin cultivar development and conservation of the species to avoid overharvesting. The present study aimed to determine the genetic diversity of C. racemosa and document the haplotype network of the specimens from four different locations (Bintan Island, Jepara, Takalar and Osi Island) using the chloroplast tufA gene. Twenty individuals from four areas were collected and amplified with the chloroplast-encoded gene tufA for species identification against publicly available data. The identification of C. racemosa based on the tufA gene showed that the species found in four locations were C. cylindracea (previously C. racemosa var. cylindracea), C. macra (previously C. racemosa var. macra), C. racemosa, and C. oligophylla (previously C. racemosa var. lamourouxii). This study records the existence of C. cylindracea in Takalar and Jepara, Indonesia for the first time. The most diverse C. racemosa species was in Osi Island, where the exploitation of this seaweed is very low. In contrast, the lowest number of C. racemosa varieties were found in Takalar, where exploitation is very high. There were only minor light variations of Caulerpa species in the tufA gene in four different sites with only four haplotypes found, and each haplotype corresponded to another species.

Lamun merupakan tumbuhan laut yang memiliki peran yang tidak kalah penting dengan ekosistem pesisir lainnya seperti terumbu karang dan mangrove baik dari segi fisik, ekologi, dan ekonomi. Indonesia memiliki 12 jenis lamun dari 58 jenis lamun di dunia. Tidak semua wilayah di Indonesia memiliki jumlah jenis yang sama karena perbedaan kondisi lingkungan atau tekanan antropogenik, sehingga perlu ada kajian keragaman jenis lamun di wilayah perairan Indonesia. Penelitian ini bertujuan untuk mengidentifikasi struktur komunitas lamun dan keterkaitan antara kerapatan lamun dan parameter lingkungan di perairan Pulau Wanci, Sulawesi Tenggara. Stasiun pengamatan berada pada daerah padang lamun yang dibagi menjadi 4 stasiun. Metode sampling yang digunakan mengacu pada McKenzie et al. (2001) menggunakan transek kuadrat 50 cm x 50 cm. Selanjutnya menghitung jumlah tegakan lamun dan mencatat jenis lamun yang ditemukan pada tiap transek kuadrat. Hasil penelitian menunjukkan bahwa ditemukan 7 jenis lamun di perairan Pulau Wanci yaitu Halophila ovalis, Halodule uninervis, Cymodocea rotundata, Thalassia hemprichii, Enhalus acroides, Thalasodendron ciliatum, dan Syringodium isoetifolium dengan kerapatan tertinggi didominasi oleh T. hemprichii, H. uninervis, dan C. rotundata dengan pola sebaran keseluruhan adalah mengelompok. Terdapat keterkaitan antara kerapatan lamun dengan parameter lingkungan, substrat berpasir memiliki korelasi positif yang tinggi terhadap kerapatan lamun sedangkan substrat liat, kecepatan arus, salinitas, dan suhu memiliki korelasi negatif yang rendah sampai tinggi terhadap kerapatan lamun di perairan Pulau Wanci.

Soft coral is one of the marine organisms that produce secondary metabolites materials and has potency as anticancer. There is much research to find bioactive compounds in the organism, but it still lacks data to find potency of bioactive in soft coral that lives under acidification pressure. The objectives of this study are to determine the characteristic of acidification in Krakatau seas and to assess the potency of cytotoxic activity from soft coral Sarcophyton. Sampling was conducted in three locations, Legon Tuo, Legon Cabe, and Umang-umang Island, to take the sample and measure water quality. Cytotoxic test using MTT method (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolinon) to MCF-7 cancer cell line to measure growth inhibition percentage. Data were analyzed using ANOVA, post hoc analysis, and multivariate analysis. Water quality in Legon Tuo and Legon Cabe have significant different to Umang-umang Island. However, Legon Tuo and Legon Cabe are suitable as acidification site, while Umang-umang Island as the reference site. The characteristic of Krakatau seas, especially in Legon Tuo and Legon Cabe study site, was categorized in low acidification. In the cytotoxic activity, soft coral Sarcophyton from Legon Cabe (acidification site) is more potential than soft coral from Umang-umang Island (reference site).

Glutathione (GSH), as a wide range of low molecular weight, which found in marine microalgae and event bacteria, are essential to prevent photooxidation and productivity loss from these Radical Oxigen Species (ROS). Symbiodinium, endo-symbiont of corals, were exposed with different UV radiation combined with irradiance treatments to explore biomass specific initial response. Intracellular glutahione was observed as potential adaptive response of Symbiodinium population under environmental specific stress. The result showed that GSH production increased significantly with increasing irradiance and/or UV levels. GSH concentration was fluctuated among populations exposed by different irradiance treatments, but not effected by UV and irradiance exposure. GSH production as a response of UV exposure was higher than irradiance treatments. Both these high correlative fluctuation of intracellular GSH production and the presence of both treatments indicated protective specific adaptation of Symbiodinium under specific environmental stress, respectively. Makroalga di daerah Manokwari memiliki keanekaragaman spesis yang tinggi, namun organisme ini sangat rentan terhadap perubahan kondisi lingkungan baik alami maupun tekanan antropogenik. Tujuan penelitian ini adalah mengkajikomposisi spesies makroalga (klasifikasi dan identifikasi) dan menganalisis struktur komunitas makroalga di perairan intertidal Manokwari Papua Barat. Penelitian pengambilan data di lapangan pada bulan Juni 2014 sampai September 2014 (periode musim timur) pada 2 daerah penelitian yaitu mainland dan outland. Komposisi spesies makroalga ditemukan 28 spesies yang diklasifikasikan ke dalam 3 divisi, 3 kelas, 11 ordo, 16 famili, dan 19 genus. Spesies alga yang di temukan tersebut dikelompokkan dalam 3 divisi utama yaitu alga hijau (Chlorophyta) 14 spesies, alga merah (Rhodophyta) 8 spesies dan alga coklat (Phaeophyta) 6 spesies. Total rata-rata kepadatan jenis makroalga daerah outland lebih tinggi dibandingkan dengan mainland. Analisis struktur komunitas makroalga daerah outland lokasi Mansinam dan Lemon memiliki indeks keanekaragaman lebih tinggi di bandingkan dengan daerah mainland lokasi Rendani, Pasir Putih dan Arfai. Indeks keseragaman pada 5 lokasi penelitian daerah mainland dan outland mendekati 1 dimana populasi individu makroalga tiap jenis menyebar merata dan tidak ada makroalga yang dominan, sedangkan indeks dominansi mendekati nol dimana komunitas makroalga penyusunnya berada dalam keadaan stabil.Kata kunci: analisis struktur komunitas, klasifikasi, Manokwari, spesies makroalga

Cembranoid content in soft coral is known as a chemotype that relate with genotype and environment. This research aimed to characterize the cembranoid Sarcophyton soft coral from the reef that acidified by CO2 volcanic vents (pHT 7.8) at Volcano Islands waters, Banda-Neira (Indonesia), as a means of predicting the future impact of ocean acidification to the genetic diversity of Sarcophyton soft coral. 30 random colonies were taken, combined, and extracted with ethanol. Cembranoid isolation and identification had been done by high performance liquid chromatography and spectroscopic techniques. Results of the study found sarcophytol derivatives (sarcophytol A, 11,12-epoxy sarcophytol A, sarcophytol B, and sarcophytol M) as the only chemotype in the sample. This may suggest low genetic diversity in the observed Sarcophyton sample. Therefore, it may suggest that even soft coral is known to be resilient to future acidification pressures, the genetic diversity or the production of diverse cytotoxic metabolite may be hampered due to ocean acidification in future climate change adaptation.

Soft coral is predicted to outcompete with hard coral in future ocean acidification scenarios. Beside the biological resilience shown in acidic conditions, soft corals ability to maintain or compete for space is shown to relate with their ability to produce cytotoxic cembranoid-type compounds. The aim of this study was to investigate composition of cytotoxic cembranoid compounds of Sarcophyton glaccum soft coral exposed to current and predicted future ocean acidification scenarios. Sarcophyton glaccum colonies were acclimated along a pH gradient to simulate predicted increases in ocean acidification: natural/current (pH 8,2), slight increase in acidification (pH 8.0 year-1 2060), and moderate increase in acidification (pH 7,8 year­-1 2100). Cembranoid composition was determined by quantitative Nuclear Magnetic Resonance spectroscopy while cytotoxic activity was determined against tumor cell lines. Results of the study showed cytotoxicity and sarcophytoxide (the most active cembranoid compound in observed Sarcophyton glaccum) were both found to be higher at pH 8,0. However, a further increase of acidification resulted on a reduction of both the cytotoxicity and sarcophytoxide production. This suggests that acidification pressures affect directly the defense system metabolism of Sarcophyton glaccum and that while they may be resilient to small decreases in pH, their ability to compete for space may be hampered by more pronounced changes. H. opuntia is one of the green algae that distributes sufficiently dominant at the coastal of West Aceh. This research aims to screen the green algae H. opuntia that is potentia as an antioxidant agent. Extraction method in this research used mono maceration. The solvents that are used including ethanol (polar), ethyl acetate (semi-polar), and n-hexane (non-polar). With those three solvents it was obtained three crude extracts including ethanol crude extract, ethyl acetate crude extract, and n-hexane crude extract. Subsequently, the crude extracts were used to conduct the assay including phytochemical assay and the antioxidant activity with DPPH method. The results of phytochemical assay showed that ethanol crude extract of H. opuntia was detected to have active compounds including alkaloids, flavonoid, phenol, tannin, and steroids whereas ethyl acetate extract of H. opuntia was detected to contain phenol compound. Total phenol content of ethanolic extract of H. opuntia = 2460.25 mg GAE/g and ethyl acetate extract = 972.68 mg GAE/g. Antioxidant activity of ethanolic extract has IC50 value = 143.63 mg/L whereas ethyl acetate extract has IC50 value = 75.51 mg/L with Vitamin C as a positive control. This results show that extract of H. opuntia possesses a vital potency as an antioxidant agent that give value added in pharmacy industries.

Acanthophyllia deshayesiana has a different habitat with Cynarina lacrymalis in the nature, but they have same character on living forms, diameter, and height of corallite. Both of these species are considered synonym, thus it needs verification study to describe whether it is synonym species or not based on morphological data. Eleven descriptive characters and seven morphometric characters were used to verify the synonym species of these coral. Descriptive data were performed by scoring method, while morphometric data were obtained from morphometric. Morphometric data were analyzed by Correspondence Analysis of Principal Coordinates (CAP) and Agglomerative Hierarchical Cluster (AHC), while descriptive data were analyzed by UPGMA (Unweight Pair Group Method with Arithmetic Mean). The result showed that both of these coral cannot differentiate based on morphometric measurement. It can differentiate significantly based on descriptive characters, so both of these coral are not synonym. Indonesian tropical soft corals are valuable resources that produce pharmacological cytotoxic cembranoids. However, the manual structure determination in these compounds requires adequate knowledge of organic chemistry. This study presents the application of Logic Structure Determination (LSD) as a freeware Computer Assisted Structure Elucidation (CASE) for automatic molecular structure determination of cembranoid compounds from soft corals species. 12 Nuclear Magnetic Resonance (NMR) datasets of cytotoxic cembranoids were used to evaluate the accuracy of LSD in generating the possible structures. The results of this study shows that LSD generated numerous possible molecular structures as the data input files were only derived from 2D-NMR HMQC (Heteronuclear Multiple-Quantum Correlation), COSY (Homonuclear Correlation Spectroscopy), and HMBC (Heteronuclear Multiple Bond Correlation). The accuracy was significantly enhanced (only 2-4 possible cembranoid structures from each NMR dataset) with the addition of H2BC (Heteronuclear 2 Bond Correlation) experimental data. This may indicate that VLRC (Very Long-Range Correlations) significantly affects LSD capability. Furthermore, LSD with direct 2 bond NMR experimental data is a reliable CASE technique for cembranoid compounds identification. In general, this freeware-CASE has the potential to be applied on other types of small molecule compounds and may serves as a solution for elucidation bottleneck step in studies on Indonesian natural products.

This study presents the relationship between benthic cover of Sarcophyton sp. living on coral reefs and their cytotoxicity (an assumption of soft coral allelochemical levels) along acidification gradients caused by shallow water volcanic vent systems. Stations with moderate acidification (pH 7.87 ± 0.04), low acidification (pH 8.01 ± 0.04), and reference conditions (pH 8.2 ± 0.02) were selected near an Indonesian CO2 seep (Minahasa, Gunung Api Island, and Mahengetang Island). Cover of the dominant soft coral species (Sarcophyton sp.) was assessed and tissue samples were collected at each site. The cytotoxicity tissue extracts were analyzed using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolinon bromide (MTT) method. Levels of cytotoxicity were strongly correlated with Sarcophyton sp. cover (p < 0.05; R2 = 0.60 at 30 ppm and 0.56 at 100 ppm), being highest at mean pH 8.01 where the soft corals were most abundant. This finding suggests that Sarcophyton sp. can be expected to survive ocean acidification near Indonesia in the coming decades. How the species might be adversely affected by further ocean acidification later in the century unless CO2 emissions are reduced remains a concern.

The anthropogenic provide a negative impact on the surrounding environment. Mangrove species, such as Sonneratia alba would get the impact of anthropogenic activities, to accumulate the pollution of heavy metals. The aim of this study were to evaluate mercury accumulation in Mangrove Apple (S. alba) and to analyze mangrove apple potency as mercury bioindicator. Samples were taken in April 2016 at Pari Island, Seribu Islands by purposive sampling. The results showed that the highest concentration of Hg in the Northern of Pari Island was found in the leaves and the lowest was in the fruit. The highest concentration of Hg in the Eastern of Pari Island was found in the leaves and lowest was in the fruit. The concentrations of Hg in the Eastern area higher the Northern area (significantly different). The accumulation of Hg mainly collected on the leaves with TF> 1, but the ability of S. alba trees absorb Hg in the environment showed a small value, namely BCF

Coral diseases that have emerged since the early 1970s have caused significant regional ecological impacts. However, there has been a paucity of research into coral disease in South-East Asia, including Indonesia. This study provides baseline coral disease data in the Kepulauan Seribu Marine National Park. In this study we show a positive correlation between overall coral cover and the dominant reef building coral Montipora spp. and found two main diseases, black band disease (BBD) and WS, were highly prevalent throughout all reefs. Based on spatial location, the highest abundance of BBD (0.08 col./m2) was found at sites nearer (zone 1) to the mainland, whilst for WS (0.05 col./m2) highest abundance was found at middle sites (zone 2). According to the temporal data, the highest abundance of BBD (0.77 col./m2) was found during the transition period (between wet and dry seasons), whereas for WS higher abundance occurred within the dry season (0.07 col./m2). There was a significant difference in disease abundance among seasons which was correlated with increasing temperature and light intensity along with variations in total organic matters, nitrite and phosphate levels. Moreover, the middle sites experienced additional stress from the waste material originating from the mainland.

Black-band disease (BBD) is a persistent disease that cause the decline of the coral reef ecosystems, which is still slightly recorded in Indonesia, including in Pari Island, Thousand Islands. The objectives of this study were to determine progression, prevalence and incidence BBD on Montipora spp in shallow water of Pari Islands. Coral cover were estimated using line intercept transect (LIT) whereas the prevalence and incidence of BBD were carried out by using belt transect method with 1 m left and right of tape as long 20 m with 3 replications recorded approximately every two months. BBD progression was documented with a digital photograph method, the photograph of affected area of each coral was taken each day for 2 weeks. BBD was found to be spread in the surveyed area. The result show that prevalence and incidence of BBD showed a positive exponential relationship with water temperature. During the observation Both prevalence and incidence increased on May transitional season (wet-dry). The highest prevalence was found at 5,96 percent and whereas the maximum disease progression found at 0,46 cm/day.

Green turtle (Chelonia mydas; Testudines) is included in the group of polyandry animals, which is single female mated with many male. DNA polymorphism method generally considered to have a high degree of accuracy as compared to other methods to elucidate polyandry phenomena on many animals. In this research, three microsatellite loci were used to identify the number and frequency of genotypes per locus, the number and frequency alleles per locus, and genotypes and number of alleles in the nest. The purpose of this research was to study the reproductive pattern of Chelonia mydas and compensation eggs of males from hatchling's population in turtle conservation area of Pangumbahan Coastal Park, West Java. The result showed that from 10 nests, we could find 37 genotypes with 11 alleles for D108 locus, 21 genotypes with 9 alleles for B103 locus, and 27 genotypes with 9 alleles for C102 locus. The alleles number of each nest was more than 5 alleles for 5 nests, and more than 4 alleles for the remaining nests. Based on the probabilities of alleles contribution of each parent, the green turtle is a polyandry animal.
