Abstract

AI is undergoing a paradigm shift with the rise of models (e.g., BERT, DALL-E, GPT-3) that are trained on broad data at scale and are adaptable to a wide range of downstream tasks. We call these models foundation models to underscore their critically central yet incomplete character. This report provides a thorough account of the opportunities and risks of foundation models, ranging from their capabilities (e.g., language, vision, robotics, reasoning, human interaction) and technical principles(e.g., model architectures, training procedures, data, systems, security, evaluation, theory) to their applications (e.g., law, healthcare, education) and societal impact (e.g., inequity, misuse, economic and environmental impact, legal and ethical considerations). Though foundation models are based on standard deep learning and transfer learning, their scale results in new emergent capabilities,and their effectiveness across so many tasks incentivizes homogenization. Homogenization provides powerful leverage but demands caution, as the defects of the foundation model are inherited by all the adapted models downstream. Despite the impending widespread deployment of foundation models, we currently lack a clear understanding of how they work, when they fail, and what they are even capable of due to their emergent properties. Abstract

Silk fibroin interactions with metallic surfaces can provide utility for medical materials and devices. Toward this goal, titanium alloy (Ti6Al4 V) was covalently grafted with polyacrylamide via electrochemically reducing 4‐nitrobenzene diazonium salt in the presence of acrylamide. Analysis of the modified surfaces with FT‐IR spectra, SEM and AFM were consistent with surface grafting. Functionalised titanium samples with a silk fibroin membrane, with and without impregnated therapeutics, were used to assess cytocompatibility and drug delivery. Initial cytocompatibility experiments using fibroblasts showed that the functionalised samples, both with and without silk fibroin coatings, supported significant increases between 72–136 % in cell metabolism, compared to the controls after 7 days. A 7‐days release profiling showed consistent bacterial inhibition through gentamicin release with average inhibition zones of 239 mm2. Over a 5‐week period, silk fibroin coated samples, both with and without growth factors, supported better human mesenchymal stem cell metabolism with increases reaching 1031 % and 388 %, respectively, compared to samples without the silk fibroin coating with. 49. THE INFLUENCE OF NITROGEN MUSTARD ON MYCOSIS FUNGOIDES

Abstract

1. Six patients with mycosis fungoides, resistant to x-radiation, have been treated with nitrogen mustard. 2. Each patient showed a striking immediate response to the drug. 3. The extent of the remissions of the disease process, during the period of study (from 4 to 7 months), varied with the nature of the lesions and the biopsy findings: (a) Three patients with histories of prolonged premycotic stages and reticulo-endothelial cell hyperplasia on biopsy showed an excellent response to the drug. (b) One patient with a history of prolonged premycotic stage and biopsy findings of a dense polymorphic infiltrate, but with sparse numbers of reticulo-endothelial cells showed a partial response to treatment. (c) Two patients whose disease was of short duration with early nodule formation and ulceration with biopsy findings of rather few endothelial cells among the infiltrate showed rapid recurrences and progression of the disease. 4. Nitrogen mustard is an effective agent in the palliative treatment of mycosis fungoides. 5. It is suggested that the point of attack of the nitrogen mustard is on the neoplastic cell arising from the reticulo-endothelial system. 6. The response of mycosis fungoides to nitrogen mustard presents additional evidence of the neoplastic nature of the disease.

50. PHENOLPHTHALEIN INTOXICATION

Abstract

Phenolphthalein has been generally looked on as a harmless drug, and because of the ease of administration it has gained very wide usage. In 1918, Abramowitz1 and Fox2 pointed out that in susceptible persons the ingestion of phenolphthalein causes a peculiar eruption of the skin. Since then, numerous reports have appeared in the literature of atypical as well as typical eruptions, and in a few instances visceral disturbances have been reported. At present the dermatosis is familiar, nineteen patients with phenolphthalein eruptions having been observed in our own clinic during the past year. The characteristic eruption is easy for the dermatologist to recognize immediately on inspection. As far as is known, only three other substances, antipyrine, arsphenamine (neoarsphenamine) or amidopyrine, are capable of provoking identical eruptions.The eruption may be spread over the entire body or may be confined to favorite sites, as the forearms, neck or thighs.
