DNA ladder is commonly used to determine the size of DNA fragments by electrophoresis in routine molecular biology laboratories. In this study, we report a new procedure to prepare a DNA ladder that consists of 10 fragments from 100 to 1000 bp. This protocol is a combination of routinely employed methods: cloning, PCR, and partial digestion with restriction enzymes. DNA fragments of 100 bp with unique restriction site at both ends were self-ligated to create a tandem repeat. Once being cloned, the tandem repeat was rapidly amplified by PCR and partially digested by restriction enzymes to produce a ladder containing multimers of the repeated DNA fragments. Our procedure for production of DNA ladder could be simple, time saving, and inexpensive in comparison with current ones widely used in most laboratories.

Với mục tiêu cung cấp một thước đo tương tự cho Việt Nam, đặc biệt trong bối cảnh nền kinh tế và thị trường tài chính trong nước đang hội nhập sâu rộng với các thị trường khu vực và toàn cầu, thông qua ứng dụng phương pháp nghiên cứu tổng hợp, phân tích, so sánh kinh nghiệm xây dựng FCI ở một số khu vực và quốc gia trên thế giới, bài viết này đã nhấn mạnh tầm quan trọng của tính linh hoạt của FCI, sự phù hợp của các biến số cũng như tỷ trọng của các biến số trong việc xây dựng FCI Việt Nam trong tương lai.

In this study, we evaluated the protective effects of Panax bipinnatifidus Seem. extracts (PSE) on hypoxia/reoxygenation (HR)-treated rat H9C2 cardiomyocytes. Aims. The purpose of this study was to evaluate the protective effect of liquiritin (LIQ) from Radix Glycyrrhizae on cardiac mitochondria against hypoxia/reoxygenation (HR) injury. Methods. H9C2 cells were subject to the HR model. LIQ purified from Radix Glycyrrhizae (purity > 95%) was administrated to reoxygenation period. Abstract. The significant differences in DNA methylation that are considered to be a biomarker for the diagnosis of cancer are a barrier to the application of biomarkers in the clinical field. In the present study, new primers were designed and further standard controls were set up to validate the accuracy of the methylation-specific PCR (MSP), a method widely used to analyze DNA methylation. By analyzing the methylation pattern of breast cancer 1 (BRCA1) and estrogen receptor (ER) in 60 patients with breast cancer, the number of cases of methylated BRCA1 and ER detected by the primer was 7/60 and 21/60, respectively, whereas that detected by the previous widely used primers was 25/60 and 47/60, respectively. Sequencing of the MSP products indicated that the 18 and 26 false-positive methylations of BRCA1 and ER, respectively, were due to insufficient validation of the previously used primers. Nghiên cứu được thực hiện nhằm xác định tỷ lệ thay thế thức ăn viên bằng cà rốt (Daucus carota) thích hợp cho tăng trưởng và chất lượng của tôm thẻ chân trắng (Liotopenaeus vannamei) (TTCT) nuôi theo công nghệ biofloc. Thí nghiệm được bố trí ngẫu nhiên với các mức thay thế cà rốt khác nhau gồm: (i) 100% thức ăn viên (đối chứng); (ii) thay thế 10%; (iii) 20% và (iv) 30% thức ăn viên bằng cà rốt. Tôm có khối lượng ban đầu 0,37±0,09 g và chiều dài 3,49±0,32 cm được nuôi trong hệ thống biofloc với tỉ lệ C:N=15:1, độ mặn 15‰­­và mật độ nuôi 150 con/m3. Sau 60 ngày nuôi, khối lượng tôm ở các nghiệm thức dao động 8,2-9,0 g và khác biệt không có ý nghĩa thống kê (p>0,05). Tỷ lệ sống và sinh khối của tôm ở nghiệm thay thế 30% đạt cao nhất (86,7% và 1,1 kg/m3), khác biệt có ý nghĩa so nghiệm thức đối chứng (56,3% và 0,8 kg/m3), nhưng không khác biệt so với hai nghiệm thức còn lại. Tương tự, chi phí thức ăn cho 1 kg tôm thương phẩm thấp nhất ở nghiệm thức thay thế 30% (49.702 đồng/kg), khác biệt có ý nghĩa (p0,05) so với nghiệm thức thay thế 10% và 20%. Bên cạnh đó, màu sắc tôm nuôi ở các nghiệm thức có bổ sung cà rốt đậm hơn so với nghiệm thức đối chứng, nhưng thành phần sinh hóa của tôm nuôi khác nhau không có ý nghĩa thống kê (p>0,05) giữa các nghiệm thức.

Nghiên cứu nhằm đánh giá ảnh hưởng của việc bổ sung cà rốt làm thức ăn cho tôm thẻ chân trắng lên sinh trưởng và chất lượng của tôm thẻ chân trắng. Thí nghiệm được bố trí hoàn toàn ngẫu nhiên với 4 nghiệm thức gồm: (i) 100% thức ăn viên; (ii) bổ sung 10% cà rốt; (iii) 20% cà rốt và (iv) 30% cà rốt. Tôm được nuôi theo công nghệ biofloc (C:N=15:1), độ mặn 15o/oo­­ và mật độ nuôi 150 con/m3. Tôm có khối lượng ban đầu là 0,37±0,09 g. Sau 60 ngày nuôi, tôm nuôi ở nghiệm thức không bổ sung cà rốt (đối chứng) có khối lượng nhỏ nhất (8,95 g) và khác biệt có ý nghĩa (p0,05). Kết quả biểu thị bổ sung 10% lượng cà rốt làm thức ăn trong nuôi tôm thẻ chân trắng đã cải thiện tăng trưởng, tỷ lệ sống, sinh khối, màu sắc của tôm hay chi phí thức ăn.
