To explore the feasibility of constructing a functional biomaterial complex with regenerated silk fibroin membrane and immortalized chondrocytes in vivo. Rat auricular chondrocytes (RACs) were transfected with the lentivirus vector pGC-FU-hTERT-3FLAG or pGC-FU-GFP-3FLAG, encoding the human telomerase reverse transcriptase (hTERT) or GFP gene. The effects of regenerated silk fibroin film on the adhesion, growth of immortalized chondrocytes and expression of collagen II in vitro were analyzed with immunofluorescent histochemistry. Immortalized RACs were transformed. Induction by nutrient medium promoted higher expression levels of collagen II in transformed chondrocytes. The regenerated silk fibroin film was not cytotoxic to immortalized chondrocytes and had no adverse influence on their adhesion. Collagen II expression was good in the immortalized chondrocytes in vivo. The construction of a silk-based biomaterial complex with immortalized chondrocytes may provide a feasible kind of functional biomaterial for the repair of cartilage defects in clinical applications. © 2013 Wiley Periodicals, Inc. Cisplatin (CDDP) stands as a highly effective chemotherapeutic agent; however, its ototoxicity remains a perplexing challenge in the field. Formononetin (FMNT), a potent flavonoid isolated from Astragalus membranaceus, displays a diverse range of promising pharmacological activities, encompassing antioxidant, anti-apoptotic, and anti-inflammatory effects. Nonetheless, the advantageous effects of FMNT on cisplatin-induced cochlear hair cell injury demand further investigation. This study aimed to assess the protective properties of FMNT against cisplatin-induced hair cell damage by conducting in vitro assays on explant-cultured cochlear hair cells. The findings revealed that FMNT exhibited a notable reduction in cisplatin-induced hair cell apoptosis. Also, FMNT effectively mitigated the accumulation of reactive oxygen species and mitochondrial damage in cochlear explants exposed to cisplatin, while also restoring the turnover of the reduced glutathione (GSH)/glutathione disulfide (GSSG) ratio. Furthermore, our study demonstrated that FMNT protects hair cells against CDDP injury through the activation of the PI3K/AKT-Nrf2 signaling pathway. Objectives: Temporal tenosynovial giant cell tumors (TGCTs) are often large and have invaded the middle skull base. It is difficult to protect the important neurovascular structures around the tumor and perform functional reconstruction on the basis of complete tumor resection. This study aimed at analyzing the surgical techniques and functional reconstruction during the operation of complex TGCT of temporal bone and middle skull base. Methods: Five patients with pathologically confirmed TGCT of different complex types in the temporal bone and middle skull base were treated in our hospital from December 2020 to February 2023. We collected and retrospectively analyzed their medical records, including medical imaging, surgical procedures, and follow-up data. Results: The tumors invaded beyond the temporal bone and destroyed the middle skull base in all cases, involving the intracranial space and other important neurovascular structures. The internal carotid artery, infratemporal fossa, pterygopalatine fossa, and parapharyngeal space were also involved in 1 case. All the patients' tumors were completely removed, and the operations were performed mainly via modified infratemporal fossa approach, or combined with expanded middle cranial fossa approach. All cases with temporal and skull base destruction were repaired using the temporalis muscle flap with no occurrence of intracranial complications. The dura mater, condyle of temporomandibular joint, facial nerve, and internal carotid artery were fully preserved. Normal maxillofacial morphology was also preserved. The air conductive hearing of 2 patients was preserved. Conclusions: We found and summarized some surgical techniques that can help safeguard the important structures around massive TGCTs of temporal bone and middle skull base, and reconstruct the defects after tumor resection. Background: Currently, there is no ideal material available for posterior scleral reinforcement (PSR) to prevent the progression of high myopia. In this study, we investigated robust regenerated silk fibroin (RSF) hydrogels as potential grafts for PSR in animal experiments to evaluate their safety and biological reactions. Methods: PSR surgery was performed on the right eye of twenty-eight adult New Zealand white rabbits, with the left eye serving as a self-control. Ten rabbits were observed for 3 months, while 18 rabbits were observed for 6 months. The rabbits were evaluated using intraocular pressure (IOP), anterior segment and fundus photography, A- and B-ultrasound, optical coherence tomography (OCT), histology, and biomechanical tests. Results: No complications such as significant IOP fluctuation, anterior chamber inflammation, vitreous opacity, retinal lesion, infection, or material exposure were observed. Furthermore, no evidence of pathological changes in the optic nerve and retina, or structural abnormalities on OCT, were found. The RSF grafts were appropriately located at the posterior sclera and enclosed in fibrous capsules. The scleral thickness and collagen fiber content of the treated eyes increased after surgery. The ultimate stress of the reinforced sclera increased by 30.7%, and the elastic modulus increased by 33.0% compared to those of the control eyes at 6 months after surgery. Conclusion: Robust RSF hydrogels exhibited good biocompatibility and promoted the formation of fibrous capsules at the posterior sclera in vivo . The biomechanical properties of the reinforced sclera were strengthened. Objective: To study the vertical and pyramidal segment of facial nerve canal based on three-dimensional reconstruction of temporal bone CT, and provide a quantitative data for the middle ear and mastoid surgery., Method: Reconstruction of adult axial CT images shows the vertical and pyramidal segment of facial nerve canal. Using the Able Software 3D-DOCTOR 3.5 to show and measure the angle of the vertical and pyramidal segment of the facial nerve canal and to analyze the features and regularity based on the measured data., Result: According to the angle of the vertical and pyramidal segment of facial nerve canal based on the three-dimensional reconstruction model. A certain regularity has been found in the direction and the angle of different segments of the vertical and pyramidal segment of facial nerve canal., Conclusion: The temporal bone CT three-dimensional reconstruction can accurately show the characteristic and angle of the vertical and pyramidal segment of facial nerve canal. The characteristics and quantitative angle of the vertical and pyramidal segment of facial nerve canal from the anatomical viewpoint for clinical utility could keep the vertical and pyramidal segment of facial nerve from injury in temporal bone surgery.

Objective: To explore a set of quantitative methods to determine the position of the facial nerve based on three-dimensional CT reconstruction of temporal bone structures on personal computer, which can provide a series of important parameters for ear and the lateral skull base surgery., Method: The internal structures of temporal bone were reconstructed based on a set of axial CT images of adult patients, the complicated relationship and their morphologic characteristics were clearly presented by using Able Software 3D-DOCTOR. The precise measurement of some parameters between facial nerve and its adjacent structures could easily be processed with the software. Based on all obtained data, the relationship of facial nerve and its adjacent structures were effectively summarized and analyzed., Result: Three-dimensional images, including the facial nerve, tympanic anulus, auditory ossicles, chochleariform process, pyramidal eminence, internal auditory, the cochlea, semicircular canal, jugular fossa and carotid artery in the temporal bone, were reconstructed. Some parameters obtained from measuring the distance or angle between the facial nerve and its adjacent structures in the three-dimensional models had some extent regularity, which were benefit to design surgical approach and determine the position of facial nerve during relevant operation., Conclusion: CT 3D reconstruction can accurately display the detailed internal structures anatomy of the temporal bone and their quantitative spatial relationships.

Objective: To analyze the clinical efficiency and study the possible mechanism of the carbogen combined with drugs in the treatment of sudden deafness., Method: Fifty-two patients with sudden deafness were divided into two groups: Twenty-six cases received carbogen (5% CO2 - 95% O2) combined with drug therapeutics of a course of intravenous administration of dexamethasone, vitaminB, low molecular dextran, salviae miltiorrhizae, and intake vitaminC, vitaminE, et al and the other 26 cases received the drugs only. The clinical effects were compared., Result: The total effective rate was 76.92% in the carbogen group and 50.00% in the control group. The difference was significant between the two groups. The recovery time in the carbogen group was 3-14 days while in the control group was 6-23 days., Conclusion: The efficiency of carbogen ombined with drugs is superior to drug therapeutics in the treatment of sudden deafness.
