Abstract

Abstract Synthetic Notch (synNotch) receptors are genetically encoded, modular synthetic receptors that enable mammalian cells to detect environmental signals and respond by activating user-prescribed transcriptional programs. Although some materials have been modified to present synNotch ligands with coarse spatial control, applications in tissue engineering generally require extracellular matrix (ECM)-derived scaffolds and/or finer spatial positioning of multiple ligands. Thus, we develop here a suite of materials that activate synNotch receptors for generalizable engineering of material-to-cell signaling. We genetically and chemically fuse functional synNotch ligands to ECM proteins and ECM-derived materials. We also generate tissues with microscale precision over four distinct reporter phenotypes by culturing cells with two orthogonal synNotch programs on surfaces microcontact-printed with two synNotch ligands. Finally, we showcase applications in tissue engineering by co-transdifferentiating fibroblasts into skeletal muscle or endothelial cell precursors in user-defined micropatterns. These technologies provide avenues for spatially controlling cellular phenotypes in mammalian tissues.

Abstract

International audience; Six candidate gene SNP assays and 12 new microsatellites were developed in the European flounder (Platichthys flesus), in order to provide new tools to (1) further investigate signatures of selection suspected on candidate genes, and (2) better depict the neutral evolutionary background shaping the differentiation of populations. All markers were successfully tested in two populations from contrasted estuaries, and should be helpful to better understand the possible adaptive responses of flounder populations to anthropogenic pressures (e.g. global warming, eutrophication, pollutions), over the whole distribution range of the species.

Abstract

International audience; In the context of global change, ectotherms are increasingly impacted by abiotic perturbations. Along the distribution area of a species, the populations at low latitudes are particularly exposed to temperature increase and hypoxic events. In this study, we have compared the proteomic responses in the liver of European flounder populations, by using 2-D electrophoresis. One southern peripheral population from Portugal vs two northern core populations from France, were reared in a common garden experiment. Most of the proteomic differences were observed between the two experimental conditions, a cold vs a warm and hypoxic conditions. Consistent differentiations between populations were observed in accumulation of proteins involved in the bioenergetics- and methionine-metabolisms, fatty acids transport, and amino-acid catabolism. The specific regulation of crucial enzymes like ATP-synthase and G6PDH, in the liver of the southern population, could be related to a possible local adaptation. This southern peripheral population is spatially distant from northern core populations and has experienced dissimilar ecological conditions; thus it may contain genotypes that confer resilience to climate changes.

Abstract

WOS:000407981500023; A two months common garden experiment was carried out to explore the potential differences of energy metabolism in northern core (France, 50 degrees N and 47 degrees N)vs southern peripheral (Portugal, 41 degrees N) populations of European flounder Platichthys flesus, submitted to cold condition (CC: water temperature = 10 degrees C) and to warm and hypoxic condition (WHC: water temperature = 22 degrees C, and moderate hypoxia with O-2 saturation = 40% during the last 6 days). Convergent growth rates (in length) were observed in the different populations and conditions, when the southern peripheral population of Portugal did not grow under cold conditions. A general reduction in liver lipid storage was observed in all populations subjected to WHC when compared to CC, whereas muscle lipid storage was unaffected. The thermal and hypoxia treatment induced changes in muscle phospholipids (PL) ratios: phosphatidylserine/PL, phosphatidylinositol/PL, between northern and southern populations. Fish from northern estuaries displayed marked anaerobiosis in WHC (increased liver LDH activity) vs marked aerobiosis under CC (higher muscle CS and CCO activities). Contrariwise, fish from the southern estuary displayed equilibrium between anaerobiosis and aerobiosis activities in WHC. Flounders from the southern population exhibited generally lower G6PDH activity (proxy for anabolism and for defense against oxidative damage), tissue-specific anaerobiosis response (muscle LDH activity) and lower CS and CCO muscle activities (aerobiosis markers) when compared to northern populations. Globally, these inter-population differences in bioenergetics suggest that southern peripheral vs northern core populations have developed differential capacity to cope with interacting stressors and that much of this variation is more likely due to local adaptation.

Abstract

Juvenile flounders (Platichthys flesus), collected in two estuaries with similar temperature regimes (the heavily polluted Seine and the moderately contaminated Vilaine), were submitted to a common garden experiment. After an acclimation period, both populations were challenged by a thermal stress (9-24 degrees C for 15 days, then maintenance at 24 degrees C for 19 days). The condition factor of the Vilaine fish increased in both conditions, while it decreased for the heated Seine flounders after 34 days. The expression of genes related to the energetic metabolism was measured in the liver. The expression levels for ATP-F0 and COII were significantly reduced for heated vs. standard fish from both estuaries, while a decrease of the 12S expression was detected only in heated vs. standard fish from the Seine estuary. Thus, it is suggested that highly contaminated fish from Seine could display a lower tolerance to thermal stress, compared to moderately contaminated fish from Vilaine.

Abstract

In the multilayered aquifer system of North Aquitania in South-West France, the Oligocene and Eocene aquifers are pumped intensively for 50 years, mainly for domestic water. The huge pumping, which reaches 290 million m3 per year, causes a steady decline in groundwater levels. In particular, given the low recharge by rainfall, the Eocene confined aquifer collapsed with a water level decrease exceeding 30 m near the Bordeaux city. The resource is threatened, both in quantity, since the pumping exceeds the renewal and in quality, due to the reversal of flow gradients, with risk of brackish water invasion from the Gironde estuarine area in which the Eocene aquifer outcrops. The Oligocene is also subject to an increased vulnerability to surface pollution. A mathematical model simulating groundwater flows in this multilayer system has been implemented to quantify the water savings to be achieved, and simulate scenarios trend (population growth, industry and agriculture development) and combined scenarios (savings and substitutions). It is a regional multilayer water resources management model with 15 aquifer layers starting from the Plio-quaternaire aquifer at the top, down to the Bajocian aquifer at the bottom. The model, which has a spatial extension varying from 10,000 to 25,000 km2 according of the aquifer, takes into account 67,000 square cells of size 2 km. It incorporates, at an annual time step, pumping in 3,250 wells and climatic data in five meteorological stations. It is calibrated in transient state using 380 observed time series of water level. The model was used to analyze the ­possibilities of restoring a balanced and safe state, using simulation of pumping scenarios. These simulations include scenarios incorporating trend forecasts of ­population growth, scenarios of economy needs, scenarios of pumping alternative aquifers and climate change scenarios. The climate change scenario selected is the moderate IPCC Arpege A1B scenario from Meteo – France.

Tras el golpe de Estado de junio de 1954, en Guatemala se produjo un asilo político en masa. El gobierno argentino recibió cerca de doscientas personas, entre las que se encontraban importantes figuras del Partido Guatemalteco del Trabajo. No obstante, a los pocos días de haber llegado al país, un grupo de más de treinta guatemaltecos fueron detenidos y apresados en la cárcel de Villa Devoto. El artículo tiene por objetivo abordar tres dimensiones, desde la perspectiva de la sociología histórica, que permiten dar cuenta de las condiciones de posibilidad de este acontecimiento: la ideológica, la normativa y la política exterior del gobierno de Juan Domingo Perón. El trabajo nos permitirá adentrarnos en la complejidad y ambigüedad que caracterizó a los populismos clásicos de América Latina y resultará una excusa para reponer la década del cincuenta, generalmente opacada por las décadas del sesenta y setenta, como coyuntura crítica para explicar las condiciones sociohistóricas de la violencia política en América Latina. After the coup of June 1954, in Guatemala there was a mass political asylum. The Argentine government received about two hundred people, including leading figures of the Guatemalan Labor Party. However, a few days after arriving in the country, a group of over thirty Guatemalans were arrested and imprisoned in the prison of Villa Devoto. The article seeks to address three dimensions, from the perspective of historical sociology, which allow to account for the conditions of possibility of this event: ideology, law and foreign policy of the government of Juan Domingo Perón. The work will allow us to explore the complexity and ambiguity that characterized the classical populism in Latin America and will be an excuse to relocate the fifties, usually overshadowed by the sixties and seventies, as critical juncture to explain the historical conditions of the political violence in Latin America.
