A Gram-stain-negative, aerobic, yellow-pigmented, straight rod-shaped bacterium, strain MOLA117T, was isolated from a coastal water sample from the north-western Mediterranean Sea, near Banyuls-sur-Mer, France. On the basis of phylogenetic analysis of the 16S rRNA gene sequence, strain MOLA117T was placed within the family Flavobacteriaceae , but showed less than 93 % 16S rRNA gene sequence similarity to other recognized species within the family. The most closely related genera included Arenibacter , Cellulophaga , Maribacter and Zobellia . The only isoprenoid quinone was menaquinone MK-6 and the predominant fatty acid was iso-C17 : 0 3-OH, representing over 33 % of the total fatty acids. The DNA G+C content was 36.9 mol%. Strain MOLA117T required NaCl for growth, and did not exhibit gliding motility or produce flexirubin. Based on the phenotypic and phylogenetic data, strain MOLA117T should be considered to represent a novel species of a new genus, for which the name Saonia flava gen. nov., sp. nov. is proposed. The type strain of Saonia flava is MOLA117T (=CIP 110873T=DSM 29762T).

Summary Dissolved organic matter (DOM) and heterotrophic bacteria are highly diverse components of the ocean system, and their interactions are key in regulating the biogeochemical cycles of major elements. How chemical and phylogenetic diversity are linked remains largely unexplored to date. To investigate interactions between bacterial diversity and DOM, we followed the response of natural bacterial communities to two sources of phytoplankton-derived DOM over six bacterial generation times in continuous cultures. Analyses of total hydrolysable neutral sugars and amino acids, and ultrahigh resolution mass spectrometry revealed large differences in the chemical composition of the two DOM sources. According to 454 pyrosequences of 16S ribosomal ribonucleic acid genes, diatom-derived DOM sustained higher levels of bacterial richness, evenness and phylogenetic diversity than cyanobacteria-derived DOM. These distinct community structures were, however, not associated with specific taxa. Grazing pressure affected bacterial community composition without changing the overall pattern of bacterial diversity levels set by DOM. Our results demonstrate that resource composition can shape several facets of bacterial diversity without influencing the phylogenetic composition of bacterial communities, suggesting functional redundancy at different taxonomic levels for the degradation of phytoplankton-derived DOM.

A Gram-negative, aerobic, cream-pigmented, non-motile, non-spore-forming straight rod, strain MOLA115T, was isolated from a coastal water sample from the Mediterranean Sea. On the basis of phylogenetic analysis of the 16S rRNA gene sequences, strain MOLA115T was shown to belong to the Gammaproteobacteria , adjacent to members of the genera Marinicella , Arenicella and Kangiella , sharing less than 89 % 16S rRNA gene sequence similarity with strains of all recognized species within the Gammaproteobacteria . The only isoprenoid quinone was ubiquinone-8. Polar lipids in strain MOLA115T included phosphatidylethanolamine, an aminolipid, phosphatidylglycerol and an aminophospholipid. Fatty acid analysis revealed iso-C15 : 0 and iso-C17 : 1ω9c to be the dominant components. The DNA G+C content was 44.5 mol%. Based upon the phenotypic and phylogenetic data, we propose that strain MOLA115T should be considered to represent a novel species in a new genus, for which the name Pleionea mediterranea gen. nov., sp. nov. is proposed. The type strain of Pleionea mediterranea is MOLA115T ( = CIP 110343T = DSM 25350T).

In this study, we investigated a culture supernatant of a Pseudovibrio denitrificans strain isolated from sea water. Five indole alkaloids were isolated from this culture. These include four known compounds, arundine ( 1 ), vibrindole A ( 2 ), 1,1,1-tris(indol-3-yl)methane ( 3 ), 7,7-bis(indol-3-yl)- p -cresol ( 4 ) and the new tetra(indol-3-yl)ethanone ( 5 ). Compound 5 showed interesting cytotoxic effect against L929 cells (EC 50 = 7 µM) and A549 cells (EC 50 = 8 µM).

The temporal dynamics in bulk bacterial parameters and in the richness of the total and active bacterial community, determined from CE-SSCP fingerprints of 16S rRNA genes and 16S rRNA transcripts, respectively, were followed weekly to bimonthly at an oligotrophic coastal site in the NW Mediterranean Sea. Bacterial abundance, bacterial heterotrophic production, and bacterial and community respiration determined over two seasonal cycles displayed large short-term variability and no pronounced temporal pattern was detectable for these parameters. Concentrations in inorganic nutrients, salinity, or concentrations of chlorophyll a could not significantly explain the temporal variability of the bacterial parameters determined. By contrast, bacterial respiration and the bacterial carbon demand were both negatively correlated with the richness of the active bacterial community, while the bacterial parameters determined herein were not related to the richness of the total bacterial community present. Our results indicate that a reduced number of ribotypes is active when rates of bacteria-mediated carbon processes are high. Our approach, based on fingerprints of 16S rRNA transcripts, could represent an interesting tool to investigate the relationship between the structure and function of marine bacteria, in particular, on short temporal and spatial scales.

By combining 16S rRNA gene clone libraries and MICRO-CARD-FISH we determined the dominant operational taxonomic units (OTU) and their contribution to bulk abundance and activity in the presence of buoyant water masses characterized by lower salinity (LSW, 37.9) and compared these with the winter and spring phytoplankton blooms. We demonstrate that organic matter associated with LSW markedly stimulated bacterial heterotrophic production as determined by [(3) H]-leucine incorporation. The OTUs SAR11-IA, SAR11-IIB, SAR86-I and SAR86-III were dominant in all clone libraries, while the Roseobacter clade and the Bacteroidetes OTU NorSea72 were more specific to the spring phytoplankton bloom. The relative contribution of these OTUs to leucine incorporation varied between 23% and 69% for SAR11, 2% and 17% for Roseobacter and was up to 4% for NorSea72. Together, they accounted for roughly 50% of bulk abundance and leucine incorporation during the four situations investigated. Our results suggest that a few cosmopolitan OTUs respond to different DOM sources in the NW Mediterranean Sea.

Isolates showing different and similar colony morphologies were selected from spread plates of bacteria from seawater samples taken in the northern Adriatic Sea. All isolates were characterised by restriction fragment length polymorphism (RFLP) patterns of their PCR-amplified 16S rRNA gene and by 95 physiological tests (Biolog system). Cluster analysis of both genetic and phenotypic patterns showed that different colony morphotypes were related to different species or biotypes. However, isolates belonging to the more well-defined, conspicuous colony types had a high similarity, whereas those from the less conspicuous colony morphotypes showed high genetic diversity. Although colony morphotypes clearly underestimate taxonomic diversity, they can be used combined with PCR-RFLP analysis and as a preliminary approach for ecological studies aimed at the isolation of different species. Furthermore, for some species forming very conspicuous pigmented colonies, such as some photosynthetic aerobic bacteria, colony morphology may be useful for a rapid and low-cost screening of their distribution in the natural environment, especially when combined with other molecular techniques.
