Abstract

Histone deacetylase enzymes (HDACs) are potential targets for the treatment of cancer and other diseases, but it is challenging to design isoform-selective agents. In this work, we created new analogs of two established but non-selective HDAC inhibitors. We decorated the central linker chains of the molecules with specifically positioned fluorine atoms in order to control the molecular conformations. The fluorinated analogs were screened against a panel of 11 HDAC isoforms, and minor differences in isoform selectivity patterns were observed.

Abstract

How do we become the lover we want to be? The Selfsexpression Programme presented in this book is a guide to help us achieve this goal. Based on a theory of love development outlined in her previous book, The Love Path, the author offers a step-by-step approach to expanding our love consciousness. Love consciousness includes physical functioning, sexual expression, and the acquisition of various intelligences we can categorize as intellectual, emotional, social, and spiritual. Anyone, of any gender, sexual orientation, and level of experience can adapt the Selfsexpression Programme to meet their unique needs. Couples or groups may wish to undertake the programme together. The book is divided into 52 love topics – one for every week of the year. Each topic begins with a brief explanation and concludes with three optional tasks. We can choose to: (RR) reflect and record; or (PP) undertake a practical project; or (SS) engage in a scintillating sexercises. The author believes we learn better when we are having fun.

Abstract

Growing evidence implicates the brain in the regulation of both immediate fuel availability (for example, circulating glucose) and long-term energy stores (that is, adipose tissue mass). Rather than viewing the adipose tissue and glucose control systems separately, we suggest that the brain systems that control them are components of a larger, highly integrated, 'fuel homeostasis' control system. This conceptual framework, along with new insights into the organization and function of distinct neuronal systems, provides a context within which to understand how metabolic homeostasis is achieved in both basal and postprandial states. We also review evidence that dysfunction of the central fuel homeostasis system contributes to the close association between obesity and type 2 diabetes, with the goal of identifying more effective treatment options for these common metabolic disorders.

Abstract

Background Color additives requiring batch certification by the U.S. Food and Drug Administration have Code of Federal Regulations (CFR) specification limits for certain elements and are usually analyzed by X-ray fluorescence spectrometry (XRF). However, sensitivity for Hg is too low in some color additives. Objective The thermal decomposition amalgamation (TDA)-atomic absorption spectrometric (AAS) technique was investigated for providing quick and accurate determinations of Hg in certifiable color additives. Methods Tests were performed to optimize conditions and test reliability of Hg determinations at and below the CFR specification limit of 1 mg/kg. Results Sensitivity is much improved over XRF, with limits of quantitation of 0.03 mg/kg for highly homogeneous color additives. Conclusion The TDA-AAS method can be used for determining Hg concentrations at and below the CFR specification limit. The technique is effective for all color additives, including those that are difficult to analyze by XRF, but less efficient for color additives that quickly deteriorate the catalyst. Regular quality checks using certified reference materials and in-house matrix-matched check standards are essential. Highlights The TDA-AAS method is applicable for use in routine color additive batch certification. Certain matrixes (notably those that release nitrogen or sulfur oxides or halogens upon combustion) necessitate more frequent replacement of the catalyst and recalibration, impacting productivity. Color additives containing BaSO4, in color additive lakes, that are difficult to analyze by other techniques, are well suited for TDA-AAS analysis.

Abstract

On May 4, 1919, Chinese students filled the streets of Beijing to protest the Treaty of Versailles, which gave Japan claim to German concession territories in northern China. One aspect of the protests whose historical significance has often been overlooked was the involvement of female students marching, mobilizing, and getting arrested alongside their male compatriots. In a country where the cloistering of women and the separation of the sexes had long been considered to be the foundation of the social, moral, and political order, the active participation of women in public protests was a watershed moment in modern Chinese politics. While the Treaty of Versailles served as the catalyst that brought women to the streets in protest, those protests were also the consequence of several new developments in modern China: the rise of Chinese nationalism in response to Japanese imperialism, the rise of new ideas that linked women’s education to the survival of the nation, and the emergence of a critical mass of female students in Chinese cities as a result of over two decades of efforts to promote girls’ public education. Drawing upon Chinese primary source speeches and essays by female students involved in the May Fourth protests, this chapter demonstrates the transformative effects of the protests on a generation of women’s personal, political, and professional lives. Finally, the active participation of women in mass politics that began with the May Fourth protests played a critical role in helping to bring about the social and political revolutions of the twentieth century.

Abstract

Peer-delivered mental health models may hold important benefits for family members, yet their prevalence, components, and outcomes are unknown. We conducted a review of peer-delivered services for families of children and adults with mental health problems. Randomized studies of interventions published between 1990 and 2014 were included if the intervention contained a component for family members and examined familial outcomes. Of 77 studies that were assessed for their eligibility, six met criteria. Familial components included coping and parenting skills, knowledge about mental health, and emotional support. Outcomes were uneven, although significant improvements in family functioning, knowledge about mental illness, parental concerns about their child, and parenting skills were associated with the intervention. Peer-delivered services for family members may have important benefits to family members and individuals with mental health problems; however, the research base remains thin. A research agenda to develop and examine these models is discussed.

Abstract

The National Lung Screening Trial (NLST) is a randomized multicenter study comparing low-dose helical computed tomography (CT) with chest radiography in the screening of older current and former heavy smokers for early detection of lung cancer, which is the leading cause of cancer-related death in the United States. Five-year survival rates approach 70% with surgical resection of stage IA disease; however, more than 75% of individuals have incurable locally advanced or metastatic disease, the latter having a 5-year survival of less than 5%. It is plausible that treatment should be more effective and the likelihood of death decreased if asymptomatic lung cancer is detected through screening early enough in its preclinical phase. For these reasons, there is intense interest and intuitive appeal in lung cancer screening with low-dose CT. The use of survival as the determinant of screening effectiveness is, however, confounded by the well-described biases of lead time, length, and overdiagnosis. Despite previous attempts, no test has been shown to reduce lung cancer mortality, an endpoint that circumvents screening biases and provides a definitive measure of benefit when assessed in a randomized controlled trial that enables comparison of mortality rates between screened individuals and a control group that does not undergo the screening intervention of interest. The NLST is such a trial. The rationale for and design of the NLST are presented.

Abstract

The recent outbreaks of influenza A/H5N1 and ‘swine influenza’ A/H1N1 have caused global concern over the potential for a new influenza pandemic. Although it is impossible to predict when the next pandemic will occur, appropriate planning is still needed to maximize efficient use of resources and to minimize loss of life and productivity. Many tools now exist to assist countries in evaluating their plans but there is little to aid in writing of the plans. This study discusses the process of drafting a pandemic influenza preparedness plan for developing countries that conforms to the International Health Regulations of 2005 and recommendations of the World Health Organization. Stakeholders from many sectors should be involved in drafting a comprehensive pandemic influenza plan that addresses all levels of preparedness.

12. Do Tyrosine Phenolic Groups Contribute to the Alkaline Transition in the Redox Potential of Cytochrome F?

Abstract

Cytochrome f, a c-type cytochrome involved in the photosynthetic electron transport chain, has a significantly higher redox potential than most other c-type cytochromes, ranging +370 to +380 mV. Like cytochrome c, cytochrome f also exhibits an alkaline transition in which the redox potential becomes pH dependent at high pH. In the case of cytochrome c, this has been attributed to replacement of the methionine sulfur serving as the sixth iron ligand by a deprotonated amino group. This cannot be the cause for the alkaline transition in cytochrome f as there is no methionine ligand to the iron, the sixth position being occupied by the N-terminal amino group. Three tyrosine phenolic groups (Y1, Y9, and Y160) are found in close proximity to the heme in the cytochrome f structure. To explore the possibility that the ionization of one or more of these tyrosines might be responsible for the alkaline transition, we have performed site directed mutagenesis, replacing each with a phenylalanine residue which lacks an ionizible group. Each of these mutants was found to have a redox potential of 375-380 mV at pH 7.0 which became pH dependent above pH 9.0 (apparent pKa 9.3). It thus seems unlikely that any of these tyrosine residues contributes to the alkaline transition of the redox potentials in cytochrome f. Redox properties of Y160L and R156L mutants will also be described.

14. The Role of Assumptions in Service Delivery: Exploring minority student participation in educational decision-making

Abstract

In a diverse society, those who decide how government services are delivered may not be similar to the service recipients, and the assumptions underlying their policies may not apply to those recipients. Scholars have identified how various factors relate to these assumptions and affect service delivery. The first purpose of this dissertation is to explore how these factors may combine to produce inequality in the delivery of government services. The second purpose is to explore how participation by service recipients in decision-making processes can interrupt these patterns. This is done using a participatory, student-centered learning program as a case study in which teachers provide educational services to students. The results have implications for research and practice pertaining to inequality in education and delivery of other services.The first essay examines how individual, environmental, and contextual factors may lead to assumptions of service providers that result in inequality. This essay describes how factors identified by institutional theorists, representative bureaucracy scholars, and those who study organizational history can interact over time to explain how assumptions may produce inequality in service delivery. Participation by service recipients from underrepresented groups in decision-making is introduced as an intervention that potentially interrupts patterns that produce inequality.The second essay focuses on the relationship between participation by underrepresented service recipients and achievement of service delivery goals. The Math Coaching Program (MCP), a participatory, student-centered learning program, serves as a case study to explore the relationship between student participation in classroom decision-making and student test scores. Estimates using a cross-classified, multi-level model indicate that the MCP is positively related to black student test scores. The results suggest that researchers and practitioners should further explore programs that elicit participation from underrepresented groups to reduce inequality that arises as the government delivers services.The third essay explores the relationship between organizational culture and the implementation of participatory programs by analyzing the relationship between teachers’ perceived school culture and their ability to implement the MCP. Results from a survey and teachers’ responses to open-ended questions suggest that teachers who reported belonging to schools with more collaborative cultures were better able to implement the MCP. This research indicates that organizational culture may be relevant to policymakers and administrators who wish to implement participatory programs.The fourth essay constructs a systems model, based upon the Bass Diffusion Model (Bass, 1963), that can be used to understand the complexity of MCP implementation. The model is holistic and captures interdependencies among its elements, including feedback dynamics. Moreover, the model provides a space for service providers to test their assumptions regarding the best ways to implement the MCP.
